1. Availability and evaluation of European forest soil monitoring data in the study on the effects of air pollution on forests

Abstract

In the study of air pollution effects on forest ecosystems, solid soil data such as cation exchange capacity, base saturation and other exchangeable cation fractions, soil texture, soil moisture, soil weathering rates, C/N ratio and other variables form an important information base for many air pollution impact models. This paper shows some of the possibilities and the limitations of the soil data that European countries collected on the systematic Level I and on the intensive and permanent Level II monitoring plots within the ICP Forests programme. The soil data date from a first inventory in the 1990s and from a second inventory more than 10 years later. Both surveys were conducted following a common manual on sampling and analysis of soil. An example of the changes in pH(CaCl2) and base saturation in the forest floor and mineral soil on more than 2000 plots till a depth of 80 cm between the two surveys is presented. In this period the pH(CaCl2) significantly increased in the very acid forest soils [with pH(CaCl2) below 4.0] but further decreased in forest soils with pH(CaCl2) above 4.0. Following the trend in pH, the base saturation increased in soils with a very low buffering capacity (soils with a base saturation below 20% in the first inventory) and decreased in forest soils with reference base saturation values above 20%. There is both a decrease of soil pH and base saturation in the forest floor of the Arenosols and Podzols. In the Podzols this decreasing trend could not be established in the mineral soil, though this decreasing trend persisted in a number of mineral soil layers of the Arenosols. The only consistent increasing trend of pH and base saturation when stratifying according to the WRB reference soil groups was seen in the forest floor of the Luvisols and Cambisols.

Abstract

To detect changes in forest ecosystems at the European scale samplings and measurements in the field and in the laboratories have to be done in some homogeneous way over decades and across countries. In many of the surveys of the ICP Forests program samples are taken and analysed in laboratories: -measurement of atmospheric deposition -measurement of soil solution -measurement of changes in soil and humus layers -measurement of tree nutrition Therefore the evaluation and improvement of the quality in laboratories area key issue. This presentation will show the structure and development of the quality control program for laboratories as an example for the quality assurance within the ICP Forests program.

Abstract

As age increases, immune responses and consequently protection following vaccination to seasonal influenza is commonly believed to decrease. Possible drivers of this immune dysfunction include immunosenescence, repeated exposure to the same seasonal influenza antigens, and prior infection with cytomegalovirus (CMV). Here, to determine immune parameters distinguishing vaccine humoral responders (R) from non-responders (NR) following vaccination, we surveyed broad peripheral blood “cellular immune correlates” of older adults vaccinated with Fluad® (an adjuvanted subunit influenza vaccine containing strains H1N1, H3N2 and B). Phenotyping included αβ-T-cells, γδ-T-cells, B-cells and myeloid cells. The frequencies of most of these lymphocyte phenotypes were found to be similar in R and NR, although perhaps counterintuitively, one of the few differences seen between the two groups was higher frequencies of regulatory T-cells in R. These differences were more prominent for responses to the vaccine strains H1N1 and H3N2 than to the B strain, and in CMV-seropositive than CMV-seronegative elderly. Further, frequencies of early-differentiated CD4+ T-cells tended to be higher and frequencies of memory CD4+ T-cells tended to be lower in R than NR. There were also differences in B-cells, with higher frequencies in R compared to NR. To the best of our knowledge, these results are the first to report such differences in elderly people responding or failing to respond to adjuvanted seasonal influenza vaccination. © 2017 Elsevier Ltd

Contains fulltext : 191000.pdf (publisher's version ) (Open Access), Cellular therapies with CD4+ T regulatory cells (Tregs) hold promise of efficacious treatment for the variety of autoimmune and allergic diseases as well as posttransplant complications. Nevertheless, current manufacturing of Tregs as a cellular medicinal product varies between different laboratories, which in turn hampers precise comparisons of the results between the studies performed. While the number of clinical trials testing Tregs is already substantial, it seems to be crucial to provide some standardized characteristics of Treg products in order to minimize the problem. We have previously developed reporting guidelines called minimum information about tolerogenic antigen-presenting cells, which allows the comparison between different preparations of tolerance-inducing antigen-presenting cells. Having this experience, here we describe another minimum information about Tregs (MITREG). It is important to note that MITREG does not dictate how investigators should generate or characterize Tregs, but it does require investigators to report their Treg data in a consistent and transparent manner. We hope this will, therefore, be a useful tool facilitating standardized reporting on the manufacturing of Tregs, either for research purposes or for clinical application. This way MITREG might also be an important step toward more standardized and reproducible testing of the Tregs preparations in clinical applications.

The methodologies for generating tolAPC vary greatly between different laboratories, making it difficult to compare data from different studies; thus constituting a major hurdle for the development of standardised tolAPC therapeutic products. Here we describe an initiative by members of the tolAPC field to generate a minimum information model for tolAPC (MITAP), providing a reporting framework that will make differences and similarities between tolAPC products transparent. In this way, MITAP constitutes a first but important step towards the production of standardised and reproducible tolAPC for clinical application.

Abstract

Purpose/Objective: Dendritic cells (DC) are widely known as professional antigen-presenting cells and provide an important link to the adaptive immune system where they regulate the balance between immunity and tolerance. Alternations in the DC compartment can ultimately lead to the induction or perpetuation of autoimmune diseases such as multiple sclerosis (MS). This study aims to identify alterations in DC phenotype and functionality in MS. Moreover, the contribution of genetic risk factors to DC alterations was determined. Materials and methods: An ex vivo analysis of myeloid (mDC) and plasmacytoid DC (pDC) was carried out on peripheral blood of MS patients(n=104) and age- and gender-matched healthy controls(HC, n=112). Frequencies and expressions of costimulatory (CD80 and CD86) and migratory molecules (CD62L, CCR5 and CCR7) were investigated. Interleukin (IL)-12p70 and interferon (IFN)-a secretion was measured following Toll-like receptor (TLR) challenge. Study subjects were genotyped for HLA-DRB1*1501 and IL-7R α. Results: A significant decrease of circulating pDC was found in peripheral blood of patients with chronic progressive MS (CPMS) compared to relapsing-remitting (RR) MS and HC. No differences in blood frequencies of mDC were found between different study groups. Both mDC and pDC of MS patients show shifts in the expression of CD86, CCR5 and CCR7 indicating that activation and migratory patterns of DC change during MS. Moreover, RRMS patients showed a reduced upregulation of CD86 on pDC and enhanced IL-12 production by mDC and pDC. HLA-DRB1*1501 carriers have reduced frequencies of circulating mDC as compared to non-HLA-DRB1*1501 carriers. Moreover, patients not carrying the protective IL-7Ra haplotype 2 have lower frequencies of pDC in the peripheral blood, indicating that genetic risk factors may impact the DC compartment of MS patients. Conclusion: Our data indicate that circulating DC subsets undergo changes in phenotype and functionality during MS disease. This studie further provides evidence that MS-associated genetic risk factors such as HLA-DRB1-1501 and absence of IL-7Ra haplotype 2 have an impact on the DC compartment and thereby may contribute to the induction and/or maintenance of autoimmune responses. Key message Exceedance of critical limits in soil solution samples was more frequent in intensively monitored forest plots across Europe with critical loads for acidity and eutrophication exceeded compared to other plots from the same network. Elevated inorganic nitrogen concentrations in soil solution tended to be related to less favourable nutritional status. Context Forests have been exposed to elevated atmospheric deposition of acidifying and eutrophying sulphur and nitrogen compounds for decades. Critical loads have been identified, below which damage due to acidification and eutrophication are not expected to occur. Aims We explored the relationship between the exceedance of critical loads and inorganic nitrogen concentration, the base cation to aluminium ratio in soil solutions, as well as the nutritional status of trees. Methods We used recent data describing deposition, elemental concentrations in soil solution and foliage, as well as the level of damage to foliage recorded at forest plots of the ICP Forests intensive monitoring network across Europe. Results Critical loads for inorganic nitrogen deposition were exceeded on about a third to half of the forest plots. Elevated inorganic nitrogen concentrations in soil solution occurred more frequently among these plots. Indications of nutrient imbalances, such as low magnesium concentration in foliage or discolouration of needles and leaves, were seldom but appeared more frequently on plots where the critical limits for soil solution were exceeded. Conclusion The findings support the hypothesis that elevated nitrogen and sulphur deposition can lead to imbalances in tree nutrition. © 2015, The Author(s).

Contains fulltext : 88044.pdf (publisher's version ) (Closed access), Active immunization using tumor antigen-loaded dendritic cells holds promise for the adjuvant treatment of cancer to eradicate or control residual disease, but so far, most dendritic cell trials have been performed in end-stage cancer patients with high tumor loads. Here, in a phase I/II trial, we investigated the effect of autologous dendritic cell vaccination in 10 patients with acute myeloid leukemia (AML). The Wilms' tumor 1 protein (WT1), a nearly universal tumor antigen, was chosen as an immunotherapeutic target because of its established role in leukemogenesis and superior immunogenic characteristics. Two patients in partial remission after chemotherapy were brought into complete remission after intradermal administration of full-length WT1 mRNA-electroporated dendritic cells. In these two patients and three other patients who were in complete remission, the AML-associated tumor marker returned to normal after dendritic cell vaccination, compatible with the induction of molecular remission. Clinical responses were correlated with vaccine-associated increases in WT1-specific CD8+ T cell frequencies, as detected by peptide/HLA-A*0201 tetramer staining, and elevated levels of activated natural killer cells postvaccination. Furthermore, vaccinated patients showed increased levels of WT1-specific IFN-gamma-producing CD8+ T cells and features of general immune activation. These data support the further development of vaccination with WT1 mRNA-loaded dendritic cells as a postremission treatment to prevent full relapse in AML patients.
