Abstract

An analysis of edge localised mode-free (quiescent) H-mode discharges exhibiting edge harmonic magnetoydrodynamic activity in the JET-carbon wall machine is presented. It is observed that the otherwise quiescent pulses with multiple-n harmonic oscillations are sustained until a threshold in pedestal electron density and collisionality is crossed. The macroscopic pedestal parameters associated with the quiescent phase are compared with those of a database of JET-ELMy discharges with both carbon and ITER-like wall (ILW). This comparison provides the identification of the existence regions in the relevant pedestal and global plasma parameters for edge harmonic oscillations (EHOs) in JET plasmas. Although the ELMy database scans pedestal collisionality and β values typical of ET-carbon quiescent operation, shaping and current are not simultaneously compatible with EHO existence. Nevertheless, ILW operation with JET-carbon quiescent-like parameters could in principle be achieved, and improved pedestal performance could be observed in more recent JET-ILW pulses.

Abstract

We propose a novel interpretation of the Edge Harmonic Oscillation (EHO), which is seen as part of quiescent high-confinement (QH) mode, and compare its predictions to JET data. This new model for the EHOs will improve our ability to predict the QH state. Unlike the standard high confinement (Hmode) regime, QH plasmas avoid violent and periodic plasma eruptions known as Edge Localised Modes (ELMs) [1]. ELMs deposit unacceptable peak heat loads causing a severe deterioration of the plasma facing components. This poses a serious operational threat to reactor relevant plasma scenarios. Thus, the development of naturally ELM-free regimes has become of crucial importance [2]. One such regime is the so called QH mode. In QH plasmas, ELMs are replaced by dominantly low- steady mild MHD EHOs [3]. EHOs have been observed in DIII-D [3], ASDEX-U [4], JET [5], JT60 [6] and NSTX [7] at low edge collisionality over a fairly broad range in [3]. EHOs, which are typically characterised by multiple rotating toroidal harmonics localised in the pedestal region [8], enhance particle transport allowing density control and potentially ash removal without the impulsive heat load caused by ELMs [9]. A successful access to and control of this favourable regime, still requires a deeper understanding of its mechanisms in terms of basic stability concept. We thus propose, within the ideal MHD linear stability framework, a novel interpretation for the EHO onset mechanism. Provided by the possibility of edge infernal-type instabilities in QH plasmas [10], we show that the interplay of poloidal flows (MHD and diamagnetic) allows the suppression of short wavelength modes so that low- oscillations, namely EHOs, can emerge [11]. Our model retrieves several features measured experimentally such as mode rotation frequencies, radial struture [8], and amplitude of the critical shearing rate [12]. The theoretical understanding of the EHO mode onset is then applied to the interpretation of JET-C discharges exhibiting Outer Mode activity [5] which are subsequently compared with the JET-ILW database. Abstract

SP142 programmed cell death ligand 1 (PD-L1) status predicts response to atezolizumab in triple-negative breast carcinoma (TNBC). Prevalence of VENTANA PD-L1 (SP142) Assay positivity, concordance with the VENTANA PD-L1 (SP263) Assay and Dako PD-L1 IHC 22C3 pharmDx assay, and association with clinicopathologic features were assessed in 447 TNBCs. SP142 PD-L1 intraobserver and interobserver agreement was investigated in a subset of 60 TNBCs, with scores enriched around the 1% cutoff. The effect of a 1-hour training video on pretraining and posttraining scores was ascertained. At a 1% cutoff, 34.2% of tumors were SP142 PD-L1 positive. SP142 PD-L1 positivity was significantly associated with tumor-infiltrating lymphocytes (P <0.01), and node negativity (P=0.02), but not with tumor grade (P=0.35), tumor size (P=0.58), or BRCA mutation (P=0.53). Overall percentage agreement (OPA) for intraobserver and interobserver agreement was 95.0% and 93.7%, respectively, among 5 pathologists trained in TNBC SP142 PD-L1 scoring. In 5 TNBC SP142 PD-L1-naive pathologists, significantly higher OPA to the reference score was achieved after video training (posttraining OPA 85.7%, pretraining OPA 81.5%, P<0.05). PD-L1 status at a 1% cutoff was assessed by SP142 and SP263 in 420 cases, and by SP142 and 22C3 in 423 cases, with OPA of 88.1% and 85.8%, respectively. The VENTANA PD-L1 (SP142) Assay is reproducible for classifying TNBC PD-L1 status by trained observers; however, it is not analytically equivalent to the VENTANA PD-L1 (SP263) Assay and Dako PD-L1 IHC 22C3 pharmDx assay. Abstract

The aim of the present study is to analyze the stability of the pressure gradient driven modes (PM) and Alfvén eigenmodes (AE) in the large helical device (LHD) plasma if the rotational transform profile is modified by the current drive of the tangential neutral beam injectors (NBI). This study forms a basic search for optimized operation scenarios with reduced mode activity. The analysis is performed using the code FAR3d which solves the reduced MHD equations describing the linear evolution of the poloidal flux and the toroidal component of the vorticity in a full 3D system, coupled with equations for density and parallel velocity moments of the energetic particle (EP) species, including the effect of the acoustic modes. The Landau damping and resonant destabilization effects are added via the closure relation. On-axis and off-axis NBI current drive modifies the rotational transform which becomes strongly distorted as the intensity of the neutral beam current drive (NBCD) increases, leading to wider continuum gaps and modifying the magnetic shear. The simulations with on-axis NBI injection show that a counter (ctr-) NBCD in inward shifted and default configurations leads to a lower growth rate of the PM, although strong n = 1 and 2 AEs can be destabilized. For the outward shifted configurations, a co-NBCD improves the AEs stability but the PM are further destabilized if the co-NBCD intensity is 30 kA T−1. If the NBI injection is off-axis, the plasma stability is not significantly improved due to the further destabilization of the AE and energetic particle modes (EPM) in the middle and outer plasma region., source:J. Abstract

The Quiescent High confinement operational mode (QH-mode) offers an excellent alterna- tive to the well known H-mode due the absence of ELMs while keeping similar plasma performance. A feature of QH-mode is that a continuous long wavelength perturbation called Edge Harmonic Oscillation (EHO) saturates non-linearly, enhancing particle transport in the edge thus avoiding impurity accumulation in the core plasma. Given the importance of EHO in QH-mode, it could be useful for control and reproduction of robust QH-mode scenarios to assess how non-axisymmetric resonant magnetic perturbations couple with EHO. This could help for example to modify the main characteristics of QH-mode discharges while keeping constant the key elements needed to achieve QH-mode. With that in mind, the effect of global toroidal mode seeding in the vacuum magnetic field is investigated.

Tokamak high-confinement regimes are normally affected by the presence of often violent edge localised modes (ELMs). ELMs deposit unacceptable peak heat loads causing a severe deterioration of the plasma facing components. Although several ELM control/suppression techniques have been developed, they may not be applicable to future reactors such as ITER and DEMO. Thus a vivid interest has arisen on the development of naturally ELM-free regimes. One of the most promising of such regimes is the so called quiescent high-confinement (QH) mode. In QH plasmas, ELM activity is replaced by dominantly low-n steady mild rotating MHD edge harmonic oscillations (EHOs) localised in the pedestal region. EHOs have been observed in DIII-D, ASDEX-U, JET and JT60 [1]. In JET such a MHD activity is referred to as Outer Modes (OMs) [2].

Abstract

Hedgehog (Hh) signaling regulates cell fate and self-renewal in development and cancer. Canonical Hh signaling is mediated by Hh ligand binding to the receptor Patched (Ptch), which in turn activates Gli-mediated transcription through Smoothened (Smo), the molecular target of the Hh pathway inhibitors used as cancer therapeutics. Small cell lung cancer (SCLC) is a common, aggressive malignancy with universally poor prognosis. Although preclinical studies have shown that Hh inhibitors block the self-renewal capacity of SCLC cells, the lack of activating pathway mutations have cast doubt over the significance of these observations. In particular, the existence of autocrine, ligand-dependent Hh signaling in SCLC has been disputed. In a conditional Tp53;Rb1 mutant mouse model of SCLC, we now demonstrate a requirement for the Hh ligand Sonic Hedgehog (Shh) for the progression of SCLC. Conversely, we show that conditional Shh overexpression activates canonical Hh signaling in SCLC cells, and markedly accelerates tumor progression. When compared to mouse SCLC tumors expressing an activating, ligand-independent Smo mutant, tumors overexpressing Shh exhibited marked chromosomal instability and Smoothened-independent upregulation of Cyclin B1, a putative non-canonical arm of the Hh pathway. In turn, we show that overexpression of Cyclin B1 induces chromosomal instability in mouse embryonic fibroblasts lacking both Tp53 and Rb1. These results provide strong support for an autocrine, ligand-dependent model of Hh signaling in SCLC pathogenesis, and reveal a novel role for non-canonical Hh signaling through the induction of chromosomal instability.Copyright © The Autor(s) 2017.

Abstract

Since the identification of anaplastic lymphoma kinase ( ALK) gene rearrangements in non-small cell lung cancer (NSCLC) in 2005, the treatment of ALK-rearranged NSCLC ( ALK+ NSCLC) has evolved at a rapid pace. This molecularly distinct subset of NSCLC has uniquely important biology, clinicopathologic features and mechanisms of drug resistance which impact on the choice of treatment for a patient with this disease. There are multiple ALK tyrosine kinase inhibitors now available in clinical practice with efficacy data continuing to emerge and guide the optimal treatment algorithm. A detailed search of medical databases and clinical trial registries was conducted to capture all relevant articles on this topic enabling an updated detailed overview of the landscape of management of ALK-rearranged NSCLC. Abstract

The collisionless confinement of all reflected particles with orbits that star within the inner half of the plasma of a stellarator with the poloidal direction of the contours of the magnetic field strenght B is shown computationally to be possible. It is also demonstrated that the confinement achieved is well compatible with the fulfilment of Mercier stability criterion.

Abstract

The concept of disjunct eddy sampling (DES) for use in measuring ecosystem-level micrometeorological fluxes is re-examined. The governing equations are discussed as well as other practical considerations and guidelines concerning this sampling method as it is applied to either the disjunct eddy covariance (DEC) or disjunct eddy accumulation (DEA) techniques. A disjunct eddy sampling system was constructed that could either be combined with relatively slow sensors (response time of 2 to 40 s) to measure fluxes using DEC, or could also be used to accumulate samples in stable reservoirs for later laboratory analysis (DEA technique). Both the DEC and DEA modes of this sampler were tested against conventional eddy covariance (EC) for fluxes of either CO2 (DEC) or isoprene (DEA). Good agreement in both modes was observed relative to the EC systems. However, the uncertainty in a single DEA flux measurement was considerable (40%) due to both the reduced statistical sampling and the analytical precision of the concentration difference measurements. We have also re-investigated the effects of nonzero mean vertical wind velocity on accumulation techniques as it relates to our DEA measurements. Despite the higher uncertainty, disjunct eddy sampling can provide an alternative technique to eddy covariance for determining ecosystem-level fluxes for species where fast sensors do not currently exist.
