Genetic alterations in signaling pathways that control cell-cycle progression, apoptosis, and cell growth are common hallmarks of cancer, but the extent, mechanisms, and co-occurrence of alterations in these pathways differ between individual tumors and tumor types. Using mutations, copy-number changes, mRNA expression, gene fusions and DNA methylation in 9,125 tumors profiled by The Cancer Genome Atlas (TCGA), we analyzed the mechanisms and patterns of somatic alterations in ten canonical pathways: cell cycle, Hippo, Myc, Notch, Nrf2, PI-3-Kinase/Akt, RTK-RAS, TGFβ signaling, p53 and β-catenin/Wnt. We charted the detailed landscape of pathway alterations in 33 cancer types, stratified into 64 subtypes, and identified patterns of co-occurrence and mutual exclusivity. Eighty-nine percent of tumors had at least one driver alteration in these pathways, and 57% percent of tumors had at least one alteration potentially targetable by currently available drugs. Thirty percent of tumors had multiple targetable alterations, indicating opportunities for combination therapy. An integrated analysis of genetic alterations in 10 signaling pathways in >9,000 tumors profiled by TCGA highlights significant representation of individual and co-occurring actionable alterations in these pathways, suggesting opportunities for targeted and combination therapies. Objectives: We sought to document the prevalence and nature of sleep-disordered breathing (SDB) in individuals with Friedreich ataxia (FRDA) as well as establish the relationship, if any, between SDB and clinical parameters of FRDA. Method(s): Eighty-two individuals with FRDA were administered the Epworth Sleepiness Scale on an annual basis for up to 3 years. Individuals were referred for a sleep study if they had an Epworth Sleepiness Scale score .8 or had clinical symptoms suggestive of SDB. Result(s): From this cohort, 21 individuals underwent a sleep study and 17 were diagnosed with obstructive sleep apnea syndrome, giving a minimum prevalence of 21%, which is greater than that found in the general population (3%-7%).Moreover, the presence of obstructive sleep apnea was significantly correlated with the duration of disease and clinical severity of FRDA. Conclusion(s): It is recommended that individuals with FRDA undergo regular screening for obstructive sleep apnea to identify the need for a sleep study and subsequent treatment if SDB is diagnosed. © 2013 American Academy of Neurology.

- Abstract
Background: Obstructive sleep apnoea (OSA) may increase energy expenditure (EE) during sleep via increased frequency of arousal, work of breathing, or sympathetic activity. Actigraphy is inaccurate at estimating EE from body movement counts alone. The SenseWear armband however incorporates biaxial accelerometry with multiple physiologic sensors to improveEEestimationandcan record during both wake and sleep. We aimed to compare the SenseWear with polysomnography (PSG) for determination of sleep and wake, and to test the hypothesis that OSA is associated with increased EE during sleep. Method(s): We studied 50 adults (34 males) referred for routine overnight PSG. In addition to PSG, the SenseWear Pro3 ArmbandTM (Bodymedia Inc.) was placed on the upper right arm and analysed using SenseWear Professional Software, Version 6.1. Epoch-by-epoch agreement rate (AR) between the measures of sleep versus wake was calculated. Cohen's kappa coefficient (K), predictive value for sleep (PVS) and wakefulness (PVW), sensitivity and specificity were calculated. Linear regression analyses were performed for EE against apnea hypopnea index (AHI), 3% oxygen desaturation index (ODI), body mass index (BMI), waist-hip ratio (WHR), gender, age and average heart rate (HR) during sleep. Result(s): A total of 23 758 mins of sleep and wake were analysed. Whilst the epoch-by-epoch AR was high (79.9 +/- 1.6%), Cohen's K revealed only fair agreement (0.36 +/- 0.03). The ability of the Sensewear to estimate sleep was very good (PVS: 85.8 +/- 1.8%. Sensitivity: 88.7 +/- 1.5%), however it was less accurate in determining wake (PVW: 55.8 +/- 2.9%, specificity 49.9 +/- 3.6%). Sleep EE was significantly associated with AHI (r = 0.54), 3% ODI (r = 0.51), BMI (r = 0.68), WHR (r = 0.52) and male gender (r = 0.58, P < 0.001 for all). 21. The effect of continuous positive airway pressure (CPAP) on energy expenditure in obstructive sleep apnoea - Preliminary findings.

Background: Obesity is thought to play a role in the pathogenesis of obstructive sleep apnoea (OSA). Additionally, OSA itself may contribute to obesity as many patients report substantial weight gain after the onset of sleep symptoms. Treating OSA with continuous positive airway pressure (CPAP) is often recommended as an adjunct to assist with weight loss. Although some patients lose weight after commencement of CPAP, other patients remain unchanged or even gain weight. This study aims to determine the pattern of physical activity and energy expenditure (EE) in free-living, obese individuals with OSA, and the subsequent effect of treatment with CPAP. Method(s): Prior to CPAP treatment, obese patients with OSA undergo a Dual Energy X-ray Absorptiometry (DEXA) scan to determine body composition and to enable correction of EE for fat free mass (FFM). Patients then wear the SenseWear Pro3 ArmbandTM (Bodymedia Inc.) for 1 week placed on the upper right arm as well as completing a daily sleep diary. This armband is a portable, combined physical activity and EE monitor which incorporates biaxial accelerometry with multiple physiologic sensors to estimate EE during both wake and sleep. Result(s): We present pilot data from a 62 year old obese male (weight: 93.7 kg, fat mass: 27.4%) with moderate OSA as documented by polysomnography (apnoea hypopnoea index: 21.5 events per hour, 3% oxygen desaturation index: 15.7 events per hour, Epworth sleepiness score: 12/24). Prior to CPAP treatment, the mean (SD) daytime EE/ FFM was 2.46 (0.32) kcal h-1 kg-1. Mean sleep efficiency was 73.3 (6.7) % and EE/FFM during the night-time sleep period was 1.16 (0.01) kcal h-1 kg-1. Data will be collected after 1 month of CPAP treatment to examine changes in the pattern of physical activity and energy expenditure. Patient recruitment is ongoing. 22. The 2007 AASM recommendations for EEG electrode placement in polysomnography: The impact on sleep and arousal scoring.

Introduction: The 2007 AASM Manual for the Scoring of Sleep and Associated Events1 recommends the use of 3 standard EEG derivations for scoring of sleep; including frontal, central and occipital derivations. This is contrast to the Rechtschaffen & Kales2 manual which recommended a minimum of one central EEG derivation. The new recommendation is partly based on observations that not all sleep scoring features (K-complexes, sleep spindles, slow waves, alpha activity) are recorded optimally in the central EEG derivation and therefore that sleep scoring using a single EEG may be inaccurate. There is little data, however, to demonstrate how using an increased number of EEG channels may impact on sleep and arousal scoring. Objective(s): (i) To assess the impact of using 3 EEG derivations (frontal, central and occipital) vs. 1 central EEG on sleep and arousal scoring parameters, in a cohort of patients presenting for diagnosis or exclusion of obstructive sleep apnoea. (ii) To assess the impact of using 3 vs. 1 EEG on intra- and inter-scorer reliability (Kappa) of sleep and arousal scoring. Method(s): (i) The impact on scoring parameters was assessed using 30 consecutive PSGs, scored twice by one of 3 scorers from different Australian sleep laboratories, once using each method. (ii) The impact on reliability was assessed using a subset of 10 PSGs scored 4 times, twice using each method, by all 3 scorers. All PSGs and versions were de-identified and scored in random order according to the 2007 manual1. Result(s): (i) There was a mean (95%confidence interval) reduction in N1 of 9.6 min (1.7, 17.5) and an increase in N3 of 10.6 min (4.8, 16.4) when scoring with 3 compared to 1 EEG (both P < 0.05). No other significant differences were found in any sleep or arousal scoring parameters. Peroxisome proliferator-activated receptor gamma (PPARγ) agonists demonstrate antitumor activity likely through transactivating genes that regulate cell proliferation, apoptosis, and differentiation. The PAX8/PPARγ fusion oncogene, which is common in human follicular thyroid carcinomas appears to act via dominant negative suppression of wild-type PPARγ, suggesting that it may be a tumor suppressor gene in thyroid cells. We have identified a novel high-affinity PPARγ agonist (RS5444) that is dependent upon PPARγ for its biological activity. This is the first report of this molecule and its antitumor activity. In vitro, the IC50 for growth inhibition is ∼0.8 nM while anaplastic thyroid carcinoma (ATC) tumor growth was inhibited three- to fourfold in nude mice. siRNA against PPARγ and a pharmacological antagonist demonstrated that functional PPARγ was required for growth inhibitory activity of RS5444. RS5444 upregulated the cell cycle kinase inhibitor, p21WAF1/CIP1. Silencing p21WAF1/CIP1 rendered cells insensitive to RS5444. RS5444 plus paclitaxel demonstrated additive antiproliferative activity in cell culture and minimal ATC tumor growth in vivo. RS5444 did not induce apoptosis but combined with paclitaxel, doubled the apoptotic index compared to that of paclitaxel. Our data indicate that functional PPARγ is a molecular target for therapy in ATC. The behavioral responses of the parasitoid Psyllaephagus pistaciae, the major biocontrol agent of the common pistachio psylla, Agonoscena pistaciae, to volatiles emanating from its host plant and host honeydew, were examined using a four-arm airflow olfactometer. In addition, the arrestment behavior of this parasitoid on clean and honeydew-treated leaves of the pistachio, Pistacia vera, was monitored. The infested pistachio leaves were the most favored source of the volatile attracting the parasitoids. The parasitoid clearly distinguished and responded to infochemicals emitted by psyllid honeydew but at a lower level than to the volatiles from infested host plants. However, the searching time, locomotory behavior, antennal drumming and ovipositor probing were all affected when they encountered honeydew-contaminated zones on pistachio leaves. These findings suggest that the psyllid honeydew releases kairomones that stimulate the parasitoids to greater searching activity, as well as providing a directional cue. The intensive searching activities in the presence of the volatiles tested were very similar to responses by the parasitoid females when encountering patches treated with psyllid honeydew. Such behavior could retain the parasitoid in a favorable area, thereby increasing the probability of additional host encounters. [ABSTRACT FROM AUTHOR]

Objective: Our goal was to demonstrate expression and functionality of oxytocin receptors in a human endometrial cell line. This cell line could then be used for further investigation of the role of oxytocin in reproductive function at the cellular level.Study Design: Oxytocin receptor messenger ribonucleic acid expression was determined by reverse transcriptase-polymerase chain reaction deoxyribonucleic acid amplification with ribonucleic acid from confluent Ishikawa cells. Ligand binding to whole cells was evaluated by nonlinear regression analysis with an iodinated oxytocin antagonist. The coupling of the oxytocin receptor to signaling pathways was evaluated by measuring oxytocin-stimulated increases in intracellular calcium concentration, phosphorylation of ERK2 (extracellular-regulated protein kinase 2) mitogen-activated protein kinase, and prostaglandin E(2) release.Results: Polyacrylamide gel electrophoresis of the reverse transcriptase-polymerase chain reaction products demonstrated the presence of oxytocin receptor messenger ribonucleic acid in Ishikawa cells. Ligand-binding analysis of these cells demonstrated a single class of noninteracting sites, with a B(max) (maximal number of binding sites) of 77.7 fmol/mg deoxyribonucleic acid and an apparent dissociation constant of 8.3 x 10(-11) mol/L. Stimulation with 100-nmol/L oxytocin caused a rapid transient increase in intracellular free calcium concentration, which was blocked by 1-micromol/L oxytocin antagonist. Treatment of cells with oxytocin for 10 minutes resulted in a marked increase in the phosphorylation of ERK2, as determined by Western blot analysis, and a 5-fold increase in prostaglandin E(2) release.Conclusion: This study is the first to demonstrate functional oxytocin receptors in an established human endometrial cell line. The histopathology of Myxidium giardi infections in wild and cultured elvers, fingerlings and adult eels is described. The lesions in the target organs of the kidney and gills were minimal. Marked granulomatous changes were evident in the spleen, peritoneal fat and, to a lesser extent, the liver and gas gland. The granulomatous reaction consisted of a mononuclear infiltration into the spore mass and proliferative fibrosis of the surrounding connective tissue. Histological evidence of an immunological response is described. The types of M. giardi cysts are described a primary interstitial cyst of the kidney and gill, a secondary cyst of the gills and a glomerular spore mass. Primary interstitial cysts in the kidney were observed to rupture and formed spore masses in the interstitial tissue. Evidence of vascular spread and the presence of spore emboli is presented. It is suggested that primary multiplication occurs in the primary interstitial cyst of the kidney and gills, and secondary multiplication in the secondary gill cyst and glomerular spore mass from which mature spores are released to the environment. The difference between the two descriptions is based on the shape of the polar capsule and a refractile fold found on the spore of Myxobolus anguillae. The article reports the presence of Myxobolus dermatobia in wild Anguilla auguilla in England and describes the lesions and spores.

The clinical signs and histopathology of Saprolegnia sp. [related to S. diclina Humphrey (syn. S. parasitica Coker)] are described in cultured Anguilla anguilla L. elvers under an intensive production system. The main lesions observed were loss of epithelium leading to ulceration, oedema and myofibrillar degenerative changes of the muscle masse The oedema resulted in swelling of the inter-myotomal connective tissue, loss of nuclei and minor host reaction. Traumatic lesions with subsequent bacteria! infections had a mononuclear inflammatory response. It is suggested that the rapid extension of the lesion is due to the loss of integrity of the integument and the widespread oedema, which altered the viability of tissues and assisted the Saprolegnia infection. Myxidium giardi was found in wild and cultured eels. The prevalence rate in elvers was 77% in the River Torridge (Devon), 80% in the River Parrett (Somerset) and 90% in the River Ouse (Yorkshire). Adult eels from the River Ouse had a prevalence rate of 53%. Cultured fingerlings and adults had a prevalence rate of 60% and 7%, respectively. There was an apparent age-related pattern in the organ location of the histozoic trophozoite: an initially high prevalence of kidney trophozoites which gradually decreased with age; in the gills of elvers there was a low prevalence of histozoic trophozoites which increased in the adult stage. Histozoic trophozoites were also found in the stomach and intestine of elvers and fingerlings. A qualitative estimate of trophozoite numbers in each organ of elvers is given. - Abstract
The acute change in pulmonary artery pressure in response to oxygen may have prognostic value for patients with chronic obstructive pulmonary disease treated with long term domiciliary oxygen. A study was carried out to elucidate the mechanism of the acute cardio-respiratory response to oxygen in such patients and to determine whether it can be quantified non-invasively. The effects of acute oxygen administration (100% for 20 minutes and 28% oxygen for 24 hours) were assessed by non-invasive means and right heart catheterisation in 17 patients with severe stable hypoxaemic chronic obstructive pulmonary disease. Measurements included change in the ratio of dead space to tidal volume (VD/VT), effective pulmonary capillary blood flow (by rebreathing and single breath soluble gas uptake: QRB, QSB), left ventricular ejection fraction (radionuclide ventriculography), and M mode echocardiographic estimates of ventricular diameters and fractional shortening. These values were compared with those obtained from right heart catheter measurements of pulmonary artery pressure, cardiac index (thermodilution and direct carbon dioxide Fick: QTD, QFICK), and pulmonary vascular resistance. Oxygen administration resulted in a significant fall in pulmonary artery pressure, QTD, and QRB and a significant increase in VD/VT. The fall in QRB after 100% oxygen breathing for 20 minutes correlated strongly with the fall in pulmonary artery pressure (r = 0.86). There was no correlation between the fall in pulmonary artery pressure and the fall in QSB or the risen in VD/VT. Left ventricular ejection fraction did not change significantly. Echocardiography was technically unsatisfactory because of lung hyperinflation. Apart from a possible relation between VO2max and fall in pulmonary artery pressure after 24 hours of 28% oxygen breathing (r = 0.49, p less than 0.1) none of the baseline respiratory function measurements predicted the fall in pulmonary artery pressure or QRB. It is concluded that the cardiopulmonary response to acute oxygen breathing in patients with hypoxic chronic obstructive pulmonary disease includes a reduction in pulmonary artery pressure and cardiac output and a redistribution of pulmonary blood flow, and that rebreathing measurements of effective pulmonary blood flow can be used to quantify this response non-invasively. Eleven freshwater salmonid hatcheries in. southern Australia were surveyed for bacterial pathogens and diseases between 1981 and 1985. Twenty-five populations of fish were examined in the study, representing a total of 2755 fish, from which kidney, liver, spleen, and in some cases peritoneum. blood and faeces were cultured. Bacteria of pathogenic significance isolated included Aeromonas hvdrophila, Streptococcus sp., Lactobacillus piscicola. Yersinia ruckeri, Mycobacterium fortuitum, M. chelonai and a filamentous acid-fast organism of uncertain taxonomic position. Lactobacillus piscicola and Streptococcus sp. were associated with clinical and subclinical peritonitis. Mycobacterium chelonei was isolated from visceral granulomas in an externally normal fish. A erotnonas salmonicida, Renihacteriwn salmon marum and Edwardsiella tarda were not isolated, indicating that the diseases furunculosis, bacterial kidney disease and edwardsiellosis are exotic to Australian salmonids. Similarly. while Y. ruckeri was isolated. enteric redmouth disease had not been recorded and is considered an exotic disease. [ABSTRACT FROM AUTHOR]

Eleven freshwater salmonid hatcheries in southern Australia were surveyed for the presence of viruses and viral diseases. Tissues from a total of 6025 fish of five salmonid species were cultured for viruses on RTG-2, BF-2 and FHM cell lines. No viruses were detected. The study strongly suggests that Australian salmonid populations are free of the cytopathic saimonid viruses. This virus-free status is of economic significance for the production and export of ova and other salmonid products. and deserves stringent quarantine protection. [ABSTRACT FROM AUTHOR]
