Bjerkandera adusta can decompose polycyclic aromatic hydrocarbons including cellulose and lignin, but its roles in inhibiting plant pathogens are unclear. Here, the confrontation culture and greenhouse pot experiments were employed to study the control effect of B. adusta M1 on Fusarium graminearum and wheat scab. The results showed that B. adusta M1 fermentation broth (FB) inhibited the growth of F. graminearum, with an inhibition rate of 52.7-89.17%. FB had a significant control effect (72.14 ± 1.42%) on wheat scab, which was slightly lower than that of the chemical fungicide carbendazim (77.34 ± 1.76%). The growth rate was significantly higher in B. adusta M1 than in F. graminearum, indicating a strong competitiveness by B. adusta M1. The images from a scanning electron microscope showed substantial deformations of the hyphae of F. graminearum being penetrated by the hyphae of B. adusta M1, indicating a strong mycoparasitism by B. adusta M1. In addition, FB increased the activity of catalase, peroxidase, and phenylalanine ammonia-lyase in wheat leaves related to disease resistance and decreased the malondialdehyde production and cell membrane permeability. We conclude that B. adusta M1 is a promising fungal agent to control the detriment of F. graminearum to cereal growth in the field.

Background: Fomitopsis officinalis (Vill. ex Fr. Bond. et Sing) is a medicinal mushroom, commonly called ‘Agarikon’; it has traditionally been used to treat cough and asthma in the Mongolian population. Objective: The objective of the study was to examine the significance of biological activity of F. officinalis and evaluation of the antioxidant activity and anticancer activity of six fractions of F. officinalis residues (Fo1-powder form dissolved in ethanol, Fo2-petroleum ether residue, Fo3-chloroformic, Fo4-ethylacetate, Fo5-buthanolic, and Fo6-waterethanolic) against hepatocellular carcinoma cells. Methods: We performed in vitro studies of cell proliferation and viability assay, annexin V-FITC/Propidium Iodide assay, and NF-kB signaling pathway by immunoblot analysis. Results: Our findings revealed that all six fractions/extracts have antioxidant activity, and somehow, they exert anticancerous effects against cancer cells. In cancerous cell lines (HepG2 and LO2), Fo3 chloroformic extract promoted the cancer cell apoptosis and cell viability, activated G2/M-phase cell cycle, and selectively induced NF-kB proteins, revealing as a novel antitumor extract. Conclusion: This study reports that Fo3-chloroformic extract is rich in antitumor activity, which was previously not investigated in cancer. To develop the impact of F. officinalis among natural products to treat/prevent oxidative stress disorders or cancers, further examinations of F. officinalis are needed to develop new natural drugs to treat cancer. However, this study assessed only one extract, Fo3-chloroformic, which has a significant impact against cancer cell lines.

Creosote oil, a byproduct of coal distillation, is primarily composed of aromatic compounds that are difficult to degrade, such as polycyclic aromatic hydrocarbons, phenolic compounds, and N-, S-, and O-heterocyclic compounds. Despite its toxicity and carcinogenicity, it is still often used to impregnate wood, which has a particularly negative impact on the condition of the soil in plants that impregnate wooden materials. Endoglucanases from glycoside hydrolase family 5 (GH5) are the key enzymes in degradation of diverse plant polysaccharides. Present study reports purification, characterization and partial sequencing of novel thermostable GH5 family endoglucanase from a newly isolated brown rot fungi Fomitopsis meliae CFA 2. Endoglucanase was purified 34.18 fold with a specific activity of 302.90 U/mg. The molecular weight of the endoglucanase was 37.87 kDa as determined by SDS PAGE. LC MS/MS analysis identified the protein to be a member of GH5_5 family. The temperature and pH optima for endoglucanase activity were 70 °C and 4.8, respectively. The enzyme catalyzed the hydrolysis of carboxymethyl-cellulose with a Km of 12.0 mg/ml, Vmax of 556.58 μmol/min/mg and Kcat of 129.41/sec. The enzyme was stimulated by Zn+2 and K+ metal ions and DTT. Half-life (t1/2) for endoglucanase was found to be 11.36 h with decimal reduction time (D) of 37.75 h at 70 °C. The activation energy for endoglucanase was found to be 30.76 kJ/mol (50 °C–70 °C). Looking at the results, the endoglucanase from Fomitopsis meliae CFA 2 seems to be a promising thermostable enzyme which may be applicable in applications like biomass hydrolysis.

The fruiting bodies of bracket fungi are a specific microhabitat colonized by various invertebrates of which mites (Acari) are rarely studied, and if they are, the study is usually faunistic. The aim of the research was to determine whether the diversification of mite assemblages (Mesostigmata, Oribatida) inhabiting the fruiting bodies of Fomitopsis pinicola (Sw.) P. Karst. (Polyporales) are connected with the character of the forests and/or the degree of decay (DD) of the fruiting bodies. The research was conducted at Białowieża National Park (BNP), in forests close to natural ones and in Karkonosze National Park (KNP) which was affected by a large-scale forest dieback in the 1980s. Eighty fruiting bodies (40 at each study site) of F. pinicola belonging to four DD categories were collected. In total, 4,345 individuals of 120 mite species were recorded at BNP, and 13,912 individuals of 96 species were recorded at KNP. Analyses revealed that the sample dispersion at each study site was comparable, nevertheless the samples from each study site were clearly grouped into slightly overlapping sets which allow observation of the differences between them. In the less decayed fungi (DD 1 and 2) there were fewer mite species and individual mites than in the more decayed samples (DD 3 and 4). There were also significant differences between the fauna of the fungi in each particular DD: the fauna of DD 1 differed from all others, whereas the fauna of heavily decayed fungi (DD 3 and 4) was more comparable. Endogenous cytokinins in mycelia of medicinal mushrooms Hericium coralloides and Fomitopsis officinalis grown in vitro were identified using high-performance liquid chromatography coupled with mass spectrometry. High amounts of zeatin-type cytokinins and isopentenyladenine were found. The qualitative composition and quantitative content of cytokinins were species-specific traits of mushrooms. Optical microscopy was used to perform a comparison analysis of the influence of crude extracts and purified cytokinin fractions from both species' mycelial biomass on HepG2 tumor cell growth in vitro and morphology. The results showed that purified cytokinin fractions from H. coralloides and F. officinalis mycelia demonstrated a cytotoxic effect on HepG2 cells, unlike crude extracts. Under the influence of all mushroom extracts, similar patterns of changes in HepG2 cell morphology were observed, but they were more pronounced for H. coralloides compared with F. officinalis. Purified fractions of both mushroom species caused an increased level of apoptosis compared to crude extracts. Some increase in glucose uptake by cultured cells was found in all investigated samples wherein the influence of H. coralloides extracts was approximately twice the effect of the corresponding F. officinalis extracts. The data obtained confirm the assumption that cytokinins are involved in the expression of therapeutic effects of medicinal mushrooms and indicate the need to take into consideration the methods of cytokinin extraction when preparing pharmacologically active drugs based on fungal raw materials. Thus, extracts from H. coralloides and F. officinalis mycelial biomass are promising in the search for anticancer agents.

Next to cellulose, chitin is the most abundant biopolymer on Earth and participates, in particular, in the assembly of the fungal cell walls. Easy monitoring of the amount of chitinous polymers could be advantageous during biotechnological cultivation and mushroom farming in order to control and optimize the production process. In this study, a modified colorimetric glucosamine (GlcN) assay was validated using high-performance anion exchange chromatography (HPAE) coupled with pulsed amperometric detection (PAD) following acid hydrolysis of the Fomes fomentarius cell walls. The average data obtained from both methods were well comparable. The HPAE-PAD analysis was suggested to be preferable because it had less data scattering, but it could be replaced with the simplified colorimetric GlcN screening in some cases such as low budgetary resources or an absence of suitable equipment, materials, or highly skilled staff. The robust, cost-effective colorimetric GlcN assay presented here can be readily requested, considering the rapidly growing market as well as the scientific and industrial potential of gourmet and medicinal mushrooms.

Fomitopsis officinalis is a popular species of therapeutic arboreal mushroom with a documented history of use in traditional European and Asian medicine. It was chosen as a research material not only due to its anti-inflammatory, antimicrobial, and immunostimulatory properties but also because it is endangered in Poland. The aim of this study was to determine the effects of supplementing inorganic zinc and magnesium salts to the culture medium in which F. officinalis was multiplied in vitro in 10-L bioreactors equipped with an aeration and CO2 removal system. The influence of the addition of zinc and magnesium salts to the culture medium on the quantity of bioelements and organic compounds (indole, phenolic compounds, and L-phenylalanine), as well as their bioavailability to the human body (extraction to artificial digestive juices), was determined. The lowest amount of magnesium and zinc was found in the fruiting bodies of F. officinalis (132.1 and 15.3 mg/100 g dry weight [DW], respectively), whereas the highest amount was found in the mycelium obtained on the medium enriched with sulfate salts (1261 and 182.8 mg/100 g DW, respectively). Enrichment of the medium increased the content of the studied bioelements and organic compounds in the F. officinalis mycelium. The results showed that the applied method allowed obtaining fortified mycelium of the tested species as a natural therapeutic material that can supplement the deficiency of bioelements, phenolic compounds, L-phenylalanine, and indole compounds.

Uptake, distribution and speciation of arsenic (As) were determined in the bracket fungus Fomitopsis betulina (previously Piptoporus betulinus), commonly known as the birch polypore, collected from a woodland adjacent to a highly contaminated former mine in the Southwest UK and at an uncontaminated site in Quebec, Canada, with no past or present mining activity. The fruiting body was divided into cap, centre and pores representing the top, middle and underside to identify trends in the distribution and transformation of As. Total As, determined by inductively coupled plasma–mass spectrometry (ICP–MS), was approximately tenfold higher in the mushroom from the contaminated compared to the uncontaminated site. Overall, accumulation of As was low relative to values reported for some soil-dwelling species, with maximum levels of 1.6 mg/kg at the contaminated site. Arsenic speciation was performed on aqueous extracts via both anion and cation high-performance liquid chromatography–ICP–MS (HPLC–ICP–MS) and on whole dried samples using X-ray absorption near edge structure (XANES) analysis. Seven As species were detected in F. betulina from the contaminated site by HPLC–ICP–MS: arsenite (AsIII), arsenate (AsV), dimethylarsinate (DMAV), methylarsonate (MAV), trimethylarsine oxide (TMAO), tetramethylarsonium ion (Tetra) and trace levels of arsenobetaine (AB). The same As species were observed at the uncontaminated site with the exception of TMAO and Tetra. Arsenic species were localized throughout the fruiting body at the contaminated site, with the cap and pores containing a majority of AsV, only the cap containing TMAO, and the pores containing higher concentrations of DMAV and MAV as well as tetra and a trace of AB. XANES analysis demonstrated that the predominant form of As at the contaminated site was inorganic AsIII coordinated with sulphur or oxygen and AsV coordinated with oxygen. This is the first account of arsenic speciation in F. betulina or any fungi of the family Fomitopsidaceae.

This study examined biological characteristics, liquid fermentation, and cultivation of Fomitopsis pinicola. A single-factor test concluded that the optimal carbon and nitrogen sources for mycelial growth were soluble starch and yeast paste; the optimal culture temperature was 31°C, and the optimal pH was 6.0. The orthogonal experiment indicated that the optimal formula for mycelial culture was 25 g soluble starch, 2 g yeast extract, 1 g KH2PO4, and 1.5 g MgSO4 added to 1 L water. The optimal conditions for liquid fermentation culture consisted of the following: a loading volume 90 mL, inoculation volume 30 mL, and rotation speed 160 rpm. The optimal substrate formula for domestic culture was 20% corn cob, 30% sawdust, 20% wheat bran, 25% cotton seed shell, 3% corn meal, 1% gypsum, and 1% lime, which produced the highest yield of fruiting bodies. The results provided basic data for deep liquid fermentation culture and recommendations for the further development and utilization of F. pinicola.

The antidiabetic effect of different doses of water extract (WE) and ethanol extract (EE) was tested on a high-fat diet and streptozotocin (STZ) induced diabetic rats. Parameters were evaluated with normal control (NC), diabetes mellitus control (DM), and metformin (M) groups. In the experiment, nine groups were used with eight rats in each group and three doses of each WE and EE were used, with low, medium, and high doses. The results revealed that the DM group lost a significant amount of weight, whereas the NC group's weight increased throughout the experiment. After treatment with Fomitopsis pinicola, the EE group's weight increased gradually. Liver, kidney, and pancreas weight decreased after STZ injection and returned to normal in EE treated groups. Fasting blood glucose (FBG) levels were observed to be significantly lower after F. pinicola treatment. Serum insulin levels were also restored to normal after mushroom extracts supplementation. Specifically, STZ-induced hyperglycemia was inhibited by high dose EE administration. The biochemical analysis revealed that high-dose EE treatment increased HDL-C and decreased TC, TG, and LDL-C. Results demonstrated that high-dose EE administration protected the organ tissues from oxidative stress by normalizing the antioxidant levels, and CAT, SOD, and GSH-Px suppressed the lethal effect of MDA. The study concluded that F. pinicola EE at the dose 300 mg/kg has a more hypoglycemic, hyperinsulinemic, antioxidant, and antihyperlipidemic effect than NC, DM, and M, and regulates hyperglycemia by increasing insulin secretion.

To efficiently decompose biomass, fungi have developed various enzymatic and non-enzymatic strategies and are a source of versatile biocatalysts. The endoglucanases in glycosyl hydrolase CAZy family 45 (GH45) are known for their small size, a high thermostability and a broad substrate specificity that has been employed in textile and detergent industries. Here we report the heterologous expression and characterisation of an GH45 endoglucanase from the brown rot Fomitopsis pinicola and its direct comparison to an already characterised GH45 from the white rot Phanerochaete chrysosporium. Both enzymes were recombinantly expressed in Pichia pastoris and purified by two chromatographic steps. The biochemical characterisation highlighted the acidophilic character, with an optimal pH of 4, and a preference for amorphous substrates as carboxymethyl cellulose (CMC) and substrates containing β-1,4-glucans rather than the previously reported β-1,3/1,4-glucans lichenan and β-glucan. The dominating products from β-1,4-glucans were C3-C6 oligosaccharides, whereas from β-1,3/1,4-glucans glucose was the main reaction product. From the characterisation no differences between the brown rot and the white rot GH45 was evident.

Bioactive complexes of medicinal mushrooms have become attractive as complementary anticancer rem-edies. Our in vitro study focused on the cytotoxicity of the polyphenol-reach and beta-glucan-containing aqueous alkali extract from Fomes fomentarius fruiting bodies (FFE) using murine fibroblasts (L929), human colon adenocarcinoma cells (Caco-2), and cutaneous melanoma cells (COLO-818). Dose-dependent FFE cytotoxicity with an half maximal inhibitory concentration of 0.44 mg/mL was observed for L929 cells upon analysis of the total number of adherent cells, degree of cell viability, cell morphology, and mitochondrial metabolic activity. Cytotoxic effects on cancer cells tested using cell impedance were dependent on FFE concentration, type of cells, and their density. As a routine in vitro model for predicting human intestinal absorption, Caco-2 cells did not react on FFE, which can indirectly support its safety for the human intestinal epithelium. Melanoma cells were affected in a dose-dependent manner, even at low FFE concentrations (0.01-0.05 mg/mL). The confluent cell layer, which resembles a fully formed tumor, was much more resistant than the incompletely formed, subconfluent cell layer, simulating tumor formation. FFE applied topically could be a promising candidate to prevent melanoma development in its early stages.

In our ongoing process of discovering bioactive macromolecules, a homogeneous polysaccharide (FOP80-1) was first purified from Fomes officinalis. FOP80-1 with molecular weight of 4560 Da was mainly composed of →3)-d-Galp-(1→, →4)-β-d-Manp-(1→, →6)-α-d-Glcp-(1→, →3,6)-d-Glcp-(1→, and t--d-Glcp. Besides the structure features, the anti-tumor activity and potential mechanism of FOP80-1 were also investigated. The cellular and zebrafish experiments revealed that FOP80-1 inhibited tumor proliferation, invasion, and metastasis by increasing ROS, arresting cell cycle, inducing apoptosis, and suppressing angiogenesis. Corresponding to the inhibition of angiogenesis, the surface plasmon resonance (SPR) experiments revealed that FOP80-1 had good affinity with VEGF, a crucial protein to regulate angiogenesis. Molecular docking indicated that FOP80-1 could interact with the protein VEGF.

The aim of this study was to evaluate the bioremoval of anthracycline antibiotics (daunomycin-DNR, doxorubicin–DOX, and mitoxantrone-MTX) by immobilized mycelium of B. adusta CCBAS 930. The activity of oxidoreductases: versatile peroxidases (VP), superoxide dismutase (SOD), catalase (CAT), and glucose oxidase (GOX), and the levels of phenolic compounds (PhC) and free radicals (SOR) were determined during the biotransformation of anthracyclines by B. adusta strain CCBAS 930. Moreover, the phytotoxicity (Lepidium sativum L.), biotoxicity (MARA assay), and genotoxicity of anthracyclines were evaluated after biological treatment. After 120 h, more than 90% of anthracyclines were removed by the immobilized mycelium of B. adusta CCBAS 930. The effective biotransformation of anthracyclines was correlated with detoxification and reduced genotoxicity.

Response surface methodology (central composite design of experiments) was employed to simultaneously optimize enzyme production and productivities of two ligninolytic enzymes produced by Ceriporiopsis subvermispora. Concentrations of glucose, ammonium tartrate and Polysorbate 80 were varied to establish the optimal composition of liquid media (OLM), where the highest experimentally obtained activities and productivities were 41 U L$^{−1}$ and 16 U L$^{−1}$ day$^{−1}$ for laccase (Lac), and 193 U L$^{−1}$ and 80 U L$^{−1}$ day$^{−1}$ for manganese peroxidase (MnP). Considering culture growth in OLM on various types of immobilization support, the best results were obtained with 1 cm beech wood cubes (BWCM). Enzyme activities in culture filtrate were 152 U L$^{−1}$ for Lac and 58 U L$^{−1}$ for MnP, since the chemical composition of this immobilization material induced higher Lac activity. Lower enzyme activities were obtained with polyurethane foam. Culture filtrates of OLM and BWCM were applied for dye decolorization. Remazol Brilliant Blue R (RBBR) was decolorized faster and more efficiently than Copper(II)phthalocyanine (CuP) with BWCM (80% and 60%), since Lac played a crucial role. Decolorization of CuP was initially faster than that of RBBR, due to higher MnP activities in OLM. The extent of decolorization after 14 h was 60% for both dyes.

In folklore, Heritiera fomes (H. fomes) has been extensively used in treatment of various ailments such as diabetes, cardiac and hepatic disorders. The present study aimed to elucidate the antidiabetic actions of hot water extract of H. fomes (HWHF), including effects on insulin release from BRIN BD11 cells and isolated mouse islets as well as glucose homeostasis in high-fat-fed rats. Molecular mechanisms underlying anti-diabetic activity along with isolation of active compounds were also evaluated. Non-toxic concentrations of HWHF stimulated concentration-dependent insulin release from isolated mouse islets and clonal pancreatic β-cells. The stimulatory effect was potentiated by glucose and isobutyl methylxanthine (IBMX), persisted in presence of tolbutamide or a depolarizing concentration of KCl but was attenuated by established inhibitors of insulin release such as diazoxide, verapamil, and Ca2+ chelation. HWHF caused depolarization of the β-cell membrane and increased intracellular Ca2+. The extract also enhanced glucose uptake and insulin action in 3T3-L1 differentiated adipocytes cells and significantly inhibited in a dose-dependent manner starch digestion, protein glycation, DPP-IV enzyme activity, and glucose diffusion in vitro. Oral administration of HWHF (250 mg/5ml/kg b.w.) to high-fat fed rats significantly improved glucose tolerance and plasma insulin responses and it inhibited plasma DPP-IV activity. HWHF also decreased in vivo glucose absorption and intestinal disaccharidase activity while increasing gastrointestinal motility and unabsorbed sucrose transit. Compounds were isolated from HWHF with similar molecular weights to quercitrin (C21 H20 O11) ranging from 447.9 to 449.9 Da which stimulated the insulin release in vitro and improved both glucose tolerance and plasma insulin responses in mice. In conclusion, H. fomes and its water-soluble phytochemicals such as quercitrin may exert antidiabetic actions mediated through a variety of mechanisms which might be useful as dietary adjunct in the management of type 2 diabetes.

Novel α-(1 → 3)-glucooligosaccharides (α-(1 → 3)-GOS) were prepared by acid hydrolysis of α-(1→ 3)-glucan isolated from Fomitopsis betulina fruiting bodies and characterized. Their anti-cancer potential was evaluated in in vitro assays in a colon cancer cell model. The tested α-(1 → 3)-GOS showed antiproliferative (MTT assay) and pro-apoptotic (Annexin V-FITC and PI technique) features against colon cancer but not against normal epithelial colon cells. Additionally, we did not observe cytotoxic activity (neutral red and lactate dehydrogenase assays) of α-(1 → 3)-GOS against several types of normal cell lines. In the present study, we demonstrated the anticancer potential of α-(1 → 3)-GOS in a colon carcinoma model. The anti-tumour effect of α-(1 → 3)-GOS is related with induction of apoptosis. Based on these results, we conclude that α-(1 → 3)-GOS may be considered as a dietary or therapeutic agent with an ability to inhibit the growth of cancer cells.

Ceriporiopsis subvermispora (C. subvermispora) is a selective degrader of lignin in the woody biomass. Glutathione S-transferases (GSTs) are multifunctional enzymes that play important roles in cellular detoxification and metabolism. The crystal structures of a GST of C. subvermispora, CsGST83044, in GSH-free and -bound forms were solved at 1.95 and 2.19 A resolution, respectively. The structure of the GSH-bound form revealed that CsGST83044 can be categorized as an atypical-type of GST. In the GSH-bound form of CsGST83044, Asn22, Asn24, and Tyr46 are located closest to the sulfur atom and form hydrogen bonds with the thiol group. The functional mutagenesis indicated that they are critical for the enzymatic activities of CsGST83044. The critical residues of an atypical-type GST belonging to the GSTFuA class were revealed for the first time. A previous study indicated that CsGST83044 and another GST, CsGST63524, differ in substrate preference; CsGST83044 prefers smaller substrates than CsGST63524 for its esterase activity. The GSH-bound pocket of CsGST83044 turns out to be small, which may explain the preference for smaller substrates. Protein engineering of GSTs of C. subvermispora in the light of the obtained insight may pave a path in the future for utilization of the woody biomass.

Ceriporiopsis subvermispora was used to modify corn stover for improving the biodegradability and biomethane yield. Corn stover was incubated with C. subvermispora for 5–90 days then anaerobically digested. It was found that the corn stover modified for 15 days achieved the highest biomethane yield of 235 mL·g−1 VS, which was an increase of 15.2% over that of the non-modified one. The mechanism analyses indicated that the improvement resulted from the combined roles of degradation selectivity, destruction of lignocellulosic structures, and linkages. The analyses showed that C. subvermispora has a high relative selectivity of lignin degradation. The structure of the lignin and the linkages among lignin and hemicellulose and cellulose were broken obviously by acetyl group removal, and the enzymatic hydrolysis of cellulose was increased by 35.61%. The finding indicated that C. subvermispora modification is one of the effective methods for enhancing biomethane yield of corn stover.

Brown-rot fungi are the wood-decay basidiomycetes and have ability to break down plant cell wall carbohydrates. It has been suggested that degradation of pectin is important for the initial stages of brown rot. We purified an endo-polygalacturonase (FpPG28A) from the brown-rot fungus Fomitopsis palustris, analysis of the predicted amino acid sequence indicated that FpPG28A belongs to GH family 28. The highest activity of purified FpPG28A was observed at 60 °C in 50 mM sodium acetate buffer (pH 5.0); this activity was highly specific for polygalacturonic acid chains. However, calcium polygalacturonate gel was not degraded by FpPG28A under those optimal conditions. We observed that calcium polygalacturonate gel was readily degraded by the enzyme in the oxalate buffer. Furthermore, the thermostability of FpPG28A was elevated in oxalate buffer at pH 3.0. These results indicated that oxalate has an important role in the degradation of woody pectin by FpPG28A.

The application of solid-state fermentation offers an alternative to conventional, submerged approaches for a variety of bioconversion processes, including animal feeds, biofuels and fungal bioproducts. Optimizing solid-state fermentation under low moisture conditions could significantly impact the proportion of dry biomass that could be processed and improve the commercial viability of this approach, because of reduced input costs and higher yields of final products. Pleurotus erygnii that appeared to show tolerance to low moisture conditions was grown on saturated and desaturated wheat straw. Pleurotus erygnii showed insignificant fibre degradation although showed significantly lower biomass decomposition on desaturated wheat straw. Fibre decomposition by the fungus on wheat straw containing wheat bran showed marginally higher decomposition when saturated although there was no difference in biomass decomposition. The levels of delignification achieved were similar under different saturation conditions. It would appear that the fungus effectively decomposed fibre under low moisture conditions often resulting in lower biomass losses. SIGNIFICANCE AND IMPACT OF THE STUDY: In this study, a white rot fungus, Pleurotus erygnii, effectively decomposed fibre under low moisture conditions when grown on wheat straw at similar levels under higher moisture conditions. However, the addition of wheat bran to wheat straw created a heterogeneous system that appeared to allow P. erygnii to thrive under much lower moisture conditions although lower levels of fibre decomposition was obtained. These factors could influence the preparation of solid-state fermentation.

White rot mushroom Fomes fomentarius is a medicinal fungus with great potential to be explored. This work focused on the chemical composition of a basic aqueous extract from F. fomentarius fruiting bodies. The extract was mostly composed of phenolics, carbohydrates, minerals, and crude fat with a low amount of proteins and chitin. One-third of the total carbohydrates were in the form of beta-glucans with minor amounts of alpha-glucans. The most valuable essential part of the extract was composed of an acid-resistant ultraviolet (UV)-absorbing mixture of phenolic compounds such as melanins, lignins, and humic acids. These compounds, also referred to as melanin-like pigments, provided for the high antioxidant activity of the extract measured in vitro. Moderate sun-protective capacity was observed with regard to UVB rays and also expected in the UVA range. Quantification of melanin-like pigments in the F. fomentarius extract was possible either gravimetrically as acid-insoluble residue or spectrophotometrically in the UV region. Melanin estimation, based on nitrogen measurements, offered misleading results due to the presence of nitrogen-free melanins along with other nitrogen-containing compounds such as proteins and chitin. F. fomentarius water-soluble basic extract, containing beta-glucans and rich in melanin-like substances, could be used, for example, for topical skin application to prevent cell damage caused by excessive UV exposure or cytotoxic free radicals. The bioactive potential, safety, and further applications of the F. fomentarius extract are currently being investigated.

Laccase is predominantly found in lignin degrading filamentous white rot fungi, where it is involved in the oxidative degradation of this recalcitrant heteropolymer. In brown rot fungi it is much less prevalent: laccases from only a few brown rots have been detected and only two have been characterized. This study tries to understand the role of this ligninolytic enzyme in brown rots by investigating the catalytic properties of laccases secreted by Fomitopsis pinicola FP58527 SS1. When grown on either poplar or spruce wood blocks, several laccases were detected in the secretome. Two of them (FpLcc1 and FpLcc2) were heterologously produced using Trichoderma reesei QM9414 Δxyr1 as expression host and purified to homogeneity by consecutive steps of hydrophobic interaction, anion exchange and size exclusion chromatography. With the substrates 2,2-azino-bis(3-ethylthiazoline-6-sulfonate) (ABTS), 2,6-dimethoxyphenol (2,6-DMP) and guaiacol both laccases showed similar, low pH-optima below 3 for ABTS and 2,6-DMP and at pH 3.5 for guaiacol which is at the acidic end of laccases isolated from white rot fungi. The determined KM values were low while kcat values measured at acidic conditions were comparable to those reported for other laccases from white rot fungi. While both enzymes showed a moderate decrease in activity in the presence of oxalic and citric acid FpLcc2 was activated by acetic acid up to 3.7 times. This activation effect is much more pronounced at pH 5.0 compared to pH 3.0 and could already be observed at a concentration of 1 mM acetic acid.

The indigenous people of the United States and Canada long have used forest fungi for food, tinder, medicine, paint, and many other cultural uses. New information about historical uses of fungi continues to be discovered from museums as accessions of fungi and objects made from fungi collected over the last 150+ years are examined and identified. Two textiles thought to be made from fungal mats are located in the Hood Museum of Art, Dartmouth College, and the Oakland Museum of California. Scanning electron microscopy and DNA sequencing were used to attempt to identify the fungus that produced the mats. Although DNA sequencing failed to yield a taxonomic identification, microscopy and characteristics of the mycelial mats suggest that the mats were produced by Laricifomes officinalis. This first report of fungal mats used for textile by indigenous people of North America will help to alert museum curators and conservators as well as mycological researchers to their existence and hopefully lead to more items being discovered that have been made from fungal fabric.

The aim of this study was to evaluate the bioremoval mechanism of anthracycline antibiotics by the white-rot fungus B. adusta CCBAS 930. The activity of oxidoreductases and levels of phenolic compounds and free radicals were determined during the biotransformation of anthraquinone antibiotics: daunomycin (DNR) and doxorubicin (DOX) by B. adusta strain CCBAS 930. Moreover, phytotoxicity (Lepidium sativum L.), ecotoxicity (Vibrio fischeri), genotoxicity and cytotoxicity of anthraquinone dyes were evaluated before and after biological treatment. More than 80% and 90% of DNR and DOX were removed by biodegradation (decolorization). Initial solutions of DNR and DOX were characterized by eco-, phyto-, geno- and cytotoxicity. Despite efficient decolorization, secondary metabolites, toxic to bacteria, formed during biotransformation of anthracycline antibiotics in B. adusta CCBAS 930 cultures. DNR and DOX metabolites did not increase reactive oxygen species (ROS) production in human fibroblasts and resazurin reduction. DNR metabolites did not change caspase-3 activity.

Twelve undescribed lanostane-type triterpenes, and twenty-two known triterpenes were isolated and identified from a medicinal bracket fungus Fomitopsis pinicola (Sw.) P. Karst. The structures of these compounds were determined by spectroscopic and spectrometric analyses. The antiinflammatory potential of thirty-two triterpene compounds was evaluated using neutrophils as an assay model, and pinicolasin J was the most potent inhibitor of superoxide anion generation and elastase release, with IC50 values of 1.81 ± 0.44 and 2.50 ± 0.64 μM, respectively. This study provides scientific insight into the nutritional supplement value and medicinal development of Fomitopsis pinicola.

Significant concentrations of pharmaceuticals and personal care products (PPCPs) have been detected in aquatic environment. Fungal enzymatic processes can oxidize these persistent PPCPs; thus, these processes have attracted considerable attention from the scientific community. Here, we evaluated the efficacy of the removal of PPCPs using native fungal enzymes derived from Bjerkandera spp. TBB-03 under various conditions. Among the eight lignocellulosic substrates, ash, which showed the highest laccase production, was selected as the sole enzyme inducer. TBB-03 laccase was found to exhibit remarkable stability under varied pH and temperature conditions. Acetaminophen and bisphenol A were effectively removed by TBB-03 laccase under various conditions, except at pH 8. Although TBB-03 laccase could not efficiently remove single-state sulfamethoxazole directly, a 22% of improvement in sulfamethoxazole removal was observed in the presence of acetaminophen. Overall, our proposed approach showed that Bjerkandera adusta TBB-03 can be potentially applied for further research regarding PPCP remediation.

The contamination of the environment with anticancer drugs, which show recalcitrance to conventional wastewater treatment, has become a significant ecological threat. Fungi represent a promising non-conventional biological alternative for water conditioning. The aim of this work was to evaluate the efficacy of five white-rot fungi (Fomes fomentarius (CB13), Hypholoma fasciculare (CB15), Phyllotopsis nidulans (CB14), Pleurotus ostreatus (BWPH) and Trametes versicolor (CB8)) in the removal of bleomycin and vincristine. The removal capacity was measured at 0, 4, 9, and 14 days of incubation using SPE-UPLC-MS. The enzymatic profiles of laccase, manganese, and lignin peroxidases and wide range of eco- and cytotoxicity, assays of the post-process samples were also conducted. We observed >94% vincristine elimination by F. fomentarius, H. fasciculare and T. versicolor after only 4 days. Bleomycin removal occurred after a minimum of 9 days and only when the drug was incubated with T. versicolor (36%) and H. fasciculare (25%). The removal of both cytostatics was associated with laccase production, and the loss of eco- and cytotoxicity, especially in regard to viability of Lemna minor and Daphnia magna, as well as fibroblasts morphology.

Fungal mycelia can eliminate almost all cocultured cyanobacterial cells within a short time. However, molecular mechanisms of algicidal fungi are poorly understood. In this study, a time‐course transcriptomic analysis of algicidal fungus Bjerkandera adusta T1 was applied to investigate gene expression and regulation. A total of 132, 300, 422, and 823 differentially expressed genes (DEGs) were identified at 6, 12, 24, and 48 hr, respectively. Most DEGs exhibited high endopeptidase activity, cellulose catabolic process, and transmembrane transporter activity by using Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses. Many decomposition genes encoding endopeptidases were induced a little later in B. adusta T1 when compared with previously investigated algicidal fungus Trametes versicolor F21a. Besides, the accumulated expression of Polysaccharide lyases8 (PL8) gene with peptidoglycan and alginate decomposition abilities was greatly delayed in B. adusta T1 relative to T. versicolor F21a. It was implied that endopeptidases and enzymes of PL8 might be responsible for the strong algicidal ability of B. adusta T1 as well as T. versicolor F21a., Algicidal process of fungus Bjerkandera adusta T1 was investigated by a time‐course transcriptomic analysis. The results showed that most differentially expressed genes exhibited high endopeptidase activity, cellulose catabolic process, and transmembrane transporter activity. Comparison of gene expression between B. adusta T1 and Trametes versicolor F21a during the algicidal process suggested that endopeptidases together with enzymes of the polysaccharide lyases family 8 might be responsible for the strong algicidal activity of B. adusta T1 as well as T. versicolor F21a.

Fermentation is a metabolic process that converts sugars into acids, gases, or alcohol. This process occurs in yeasts and bacteria, as well as in muscle cells when faced with a lack of oxygen. In this paper, isolation, culture, purification and extracellular polysaccharides of strain Fomes fomentarius were studied. Extraction of polysaccharides from a culture based on F. fomentarius extracellular polysaccharides, extracellular polysaccharides fermentation experiments was optimized and compared, the optimal fermentation method was obtained; extracellular polysaccharides were sulfated, phosphorylated experiments, selenium acidified, discussed the preparation of derivative polysaccharides and microscopic detection, and finally studied extracellular polysaccharides on DPPH, The scavenging ability, superoxide anion radical and hydroxyl radical scavenging ability of the derived polysaccharides were compared. The results showed that the extracellular polysaccharide and derivatized polysaccharide of F. fomentarius had certain antioxidant activity.

Medicinal mushrooms of the order Polyporales have a long history of use, which is evidenced by the finding of dissected fruiting bodies with Ötzi, who lived over 5000 years ago. Because of its valuable biological properties and its use in 18th and 19th-century pharmacy, Fomitopsis officinalis used to be mass-collected. Moreover, the large demand for larch wood and non-wood materials (resin) caused an excessive exploitation of larch forests, which directly contributed to the disappearance of F. officinalis from its natural environment. The qualities of medicinal preparations obtained from the F. officinalis fruiting bodies are determined by the unique composition of its bioactive compounds, such as: triterpenoids, polysaccharides, organic acids, coumarins and phenolic compounds. It has been proved that both crude extracts and the compounds isolated from F. officinalis have a wide spectrum of therapeutic effects, including anti-inflammatory, cytotoxic, and antimicrobial effects.

Several factors affect the vegetative growth of fungi, such as temperature, pH, and culture medium. In addition to mycelial growth, these factors affect metabolite production. There are limited studies that have identified the metabolites produced by the fungus

Fusarium wilt is caused by Fusarium oxysporum f. sp. elaeidis, and constitutes a severe threat to the oil palm industry in Africa. This study is aimed at surveying, identifying the secreted effector genes responsible for virulence during pathogenesis, and investigating the level of genetic diversity and cluster resolutions of alleles accountable for virulence in pathogenic strains of F. oxysporum f.sp. elaeidis from African countries. Fifty-eight fungal strains were isolated from acute and chronic Fusarium wilt diseased oil palms in Nigeria, Ghana and Cameroon. Morphological and sequencing analysis of the Internal Transcribed Spacer (ITS) region grouped all strains into nine dominant strains with a majority (41.37%) belonging to F. oxysporum, followed by F. solani (20.68%), F. equiseti (20.68%), F. verticilliodes (5.17%), F. proliferatum (3.44%), F. chlamydosporum (3.44%), F. nelsonii (1.72%), Fomes fomentarius, and Penicillium simplicissimum (1.72%). Disease incidence and severity showed varying levels of virulence with some Fusarium strains causing severe symptoms while others exhibited slight symptoms. ISSR evaluation disclosed a considerable level of genetic diversity among pathogenic F. oxysporum f.sp. elaeidis strains. Molecular characterization using defense gene primers revealed that the oil palm genotypes screened did not amplify defense genes. During pathogenesis, Fusarium strains produced GMC oxidoreductases, hypothetical proteins, FOIG 16629, FOXG 14258, and Pyranose dehydrogenase 3-like proteins using virulent effector gene primers. Polymerase Chain Reaction analysis using specific gene primers revealed that PRK02106, beta and BetA effector genes were secreted explicitly by F. oxysporum f.sp. elaeidis (4) and F. oxysporum f.sp. elaeidis (CRT) strains while screening tolerant oil palm genotypes. During screening susceptible oil palm genotypes, F. oxysporum f.sp. elaeidis (4) and F. oxysporum f.sp. elaeidis (CRT) strains produced FGGY_L-XK1, PRK10939, FGGY_N1, XylB1, XylB2, FGGY_L-XK2, XylB3, FGGY_N2, and XylB4 effector genes. Identifying these effector genes will provide the platform to study the basis of pathogenesis which will help breeders to modify breeding techniques for the improvement of oil palm genotypes in order to reduce oil palm loss in plantations and enhance food security.

The present study offers an alternative method for green synthesis of the formation of two types of nanoparticles (NPs). These NPs, titanium oxide and silver NPs (TiO2 and Ag NPs, respectively), were obtained from the amalgamation of intracellular extract of a wild mushroom, Fomes fomentarius, with aqueous solutions of titanium isopropoxide and silver nitrate, respectively. F. fomentarius was identified phenotypically and by 18S ribosomal RNA gene sequencing (Gene accession no: MK635351). The biosynthesis of TiO2 and Ag NPs was studied and characterized by X-ray diffraction (XRD), diffuse reflectance UV-Visible spectroscopy (DR-UV), fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM) and transmission electron microscope (TEM). Success was achieved in obtaining NPs of differing sizes and shapes. The antibacterial and anticancer activity of the NPs was significant with morphological damage being caused by both, although Ag NPs (10–20 nm) were found to have profound effects on bacterial and cancer cells in comparison to TiO2 NPs (100–120 nm). These metal NPs, synthesized using wild mushrooms, hold a great potential in biomedicinedue to an effective enzyme combination, which permits them to modify different chemical compounds to less toxic forms, which is required for ecofriendly and safe biomaterials.
