Aim of this study was to analyse the genetic background of milk Fourier transform infrared (FTIR) spectra in dairy sheep. Individual milk FTIR spectra, with 1060 wavenumbers each, were available for 793 adult Sarda breed ewes genotyped at 45,813 SNP. The absorbance values of each wavenumber was analysed using a linear mixed model that included dim class, parity and lambing month as fixed effects and flock-test date and animal as random effects. The model was applied to estimate variance components and heritability and to perform a genome-wide association study for each wavenumber. Average h 2 of wavenumbers absorbance was 0.13 ± 0.08, with the largest values observed in the regions associated with the characteristic bonds of carbonylic and methylenic groups of milk fat (h 2  = 0.57 at 1724-1728 cm -1 ; and h 2  = 0.34 at 2811-2834 cm -1 , respectively). The absorbance values of wavenumbers were moderately correlated with the estimated heritabilities. After the Bonferroni correction, a total of nine markers were found to be significantly associated with 32 different wavenumbers. Of particular interest was the SNP s63269.1, mapped on chromosome 2, that was found to be associated with 27 wavenumbers. Genes previously found to be related to traits of interest (e.g. disease resistance, milk yield and quality, cheese firmness) are located close to the significant SNP. The aim of this study was to monitor the behaviour of purebred and crossbred beef cattle reared in the same optimal environmental conditions according to Classyfarm ® . Thirty-yearling beef 11.5 months old, including 10 Limousines (LMS), 10 Sardo-Bruna (SRB), and 10 crossbred Limousine × Sardo-Bruna (LMS × SRB), balanced for sex and body weight, were used. Animals were evaluated for five months by two trained operators by SCAN ("sternal resting", "lateral resting", " central or peripheral position in the pen", standing", "walking", "feeding", "drinking", and "ruminating) and FOCUS ("displacement for space", "displacement for feed or water", "play-fighting", "self-grooming", "allo-grooming", "stereotyping", and "mounting") protocols. Feeding behaviour was monitored by a CCTV system. The application of the SCAN sampling evidenced that SRB animals preferred the "standing" activity over the LMS animals, while the LMS × SRB did not differ from them. The "standing" and " ruminating "activities were observed mostly in females than males ( p < 0.05). For behaviour parameters assessed by the FOCUS methodology, the n-events of "allo-grooming" were higher ( p < 0.05) in SRB than in LMS and LMS × SRB genetic types. Males showed higher ( p < 0.05) n-events than females for "play-fighting". For feeding behaviour, the "eating concentrate" activity (expressed as n-events) was higher ( p < 0.05) in SRB than LMS × SRB and LMS being intermediate ( p < 0.05). The duration of "eating concentrate" (expressed in minutes) was higher ( p < 0.05) in females than males. In conclusion, behaviour indicators of animal welfare did not evidence substantial differences among genetic types and between sexes reared in the same "optimal" environmental conditions. Female beef and the autochthon's cattle breed of Sardinia, although typically hardy, showed a wide behavioural repertoire.

Individual dry matter intake (DMI) is a relevant factor for evaluating feed efficiency in livestock. However, the measurement of this trait on a large scale is difficult and expensive. DMI, as well as other phenotypes, can be predicted from milk spectra. The aim of this work was to predict DMI from the milk spectra of 24 lactating Sarda dairy sheep ewes. Three models (Principal Component Regression, Partial Least Squares Regression, and Stepwise Regression) were iteratively applied to three validation schemes: records, ewes, and days. DMI was moderately correlated with the wavenumbers of the milk spectra: the largest correlations (around ±0.30) were observed at ~1100-1330 cm -1 and ~2800-3000 cm -1 . The average correlations between real and predicted DMI were 0.33 (validation on records), 0.32 (validation on ewes), and 0.23 (validation on days). The results of this preliminary study, even if based on a small number of animals, demonstrate that DMI can be routinely estimated from the milk spectra.

The continuous increase in the cost of feeds and the need to improve the sustainability of animal production require the identification of alternative feeds, such as those derived from the agro-industrial sector, that can be effectively used for animal nutrition. Since these by-products (BP) are sources of bioactive substances, especially polyphenols, they may play an important role as a new resource for improving the nutritional value of animal-derived products, being effective in the modulation of the biohydrogenation process in the rumen, and, hence, in the composition of milk fatty acids (FA). The main objective of this work was to evaluate if the inclusion of BP in the diets of dairy ruminants, as a partial replacement of concentrates, could improve the nutritional quality of dairy products without having negative effects on animal production traits. To meet this goal, we summarized the effects of widespread agro-industrial by-products such as grape pomace or grape marc, pomegranate, olive cake, and tomato pomace on milk production, milk composition, and FA profile in dairy cows, sheep, and goats. The results evidenced that substitution of part of the ratio ingredients, mainly concentrates, in general, does not affect milk production and its main components, but at the highest tested doses, it can depress the yield within the range of 10-12%. However, the general positive effect on milk FA profile was evident by using almost all BP at different tested doses. The inclusion of these BP in the ration, from 5% up to 40% of dry matter (DM), did not depress milk yield, fat, or protein production, demonstrating positive features in terms of both economic and environmental sustainability and the reduction of human-animal competition for food. The general improvement of the nutritional quality of milk fat related to the inclusion of these BP in dairy ruminant diets is an important advantage for the commercial promotion of dairy products resulting from the recycling of agro-industrial by-products.

The aim of the study was to compare odd and branched-chain fatty acids (OBCFA) of milk from sheep, goat, cow, buffalo, donkey, human, and formula milk. Ruminant, monogastric, and human milks have different concentrations of these fatty acids (FA). To highlight the differences on OBCFA, a total of 282 individual milk samples were analyzed by gas chromatography. The OBCFA were found higher in ruminant than non-ruminant milks (p < 0.05). Among ruminants, sheep milk had the highest OBCFA (4.5 g/100 g of total FAME), whereases the lowest values were found in formula milk (0.18 g/100 g of total FAME). Regarding individual linear odd-chain FA (linear-OCFA), C11:0 was found higher in donkey milk than others, while sheep and buffalo milks had the greatest concentration of C15:0. Among BCFA, the iso-BCFA were higher than anteiso-BCFA in all considered milks. The isoC17:0 showed the highest concentration in all milks except for donkey and buffalo, which showed higher concentration of isoC16:0 than others. In conclusion, ruminant milks are different in terms of these FA compared to human milk and its substitutes. However, the greatest differences were found with formula milk, suggesting that this product needs the implementation of these FA to be more similar to human milk composition.

The aim of this study was to evaluate the effects of two commercially available fish- (FH) and chicken (CH)-based diets and the same diets combined with antioxidant compounds, e.g., FH-AOX and CH-AOX, respectively, on biochemical parameters and blood fatty acid (FA) profile. For this purpose, 28 dogs were allocated to four groups and fed the four diets for six months. Blood samples were taken before starting the experimental period and at the end of the trial. Concerning the biochemical parameters, the animals fed CH-AOX reached the highest values for ALB compared to the animals fed CH or FH-AOX; however, the values were within the reference values for old dogs. Triglycerides and urea were significantly higher in FH compared to the CH diet; however, both FH-AOX and CH-AOX showed a marked decrease in the TRI and urea concentration with respect to FH and CH. Moreover, CH-AOX and FH-AOX increased glucose values, linoleic acid content, and polyunsaturated FA n-3 with respect to FH and CH. On the contrary, arachidonic acid was lower in the CH-AOX and FH-AOX groups. In conclusion, FH led to a better FA profile than that of the CH diet, while CH-AOX and FH-AOX improved the FA profile regardless of the basal diet.

Natural adaptation and artificial selection have shaped the genome of modern livestock breeds. Among SNP-based metrics that are used to detect signatures of selection at genome-wide level, runs of homozygosity (ROH) are getting increasing popularity. In this paper, ROH distribution and features of a sample of 823 Sarda breed ewes farmed at different levels of altitude are analysed to investigate the effect of the environment on the patterns of homozygosity. A total of 46,829 (33,087 unique) ROH were detected. OAR2 exhibited the largest average number of ROH per animal. The most frequent ROH (OAR27, 38.9-44.2 Mb) was shared by 327. ROH length was statistically affected (p < 0.001) by both the altitude and temperature of the place where the flock was located. The highest probability of a SNP falling in a ROH was observed for hill ewes, whereas the smallest one for mountain. A total of 457 SNP exceeded the 99th percentile of the ROH count per SNP distribution and were considered significant. These markers mapped in eight chromosomes and they clustered into 17 ROH islands, where 80 candidate genes were mapped. A multivariate analysis was used to investigate the fatty acid (FA) profile in three different Italian sheep breeds: Comisana, Massese, and Sarda. A sample of 852 animals was considered: 118 Massese, 303 Comisana, 431 Sarda. Sarda sheep were divided into two groups, based on their breeding origin (298 and 133 reared in Sardinia and Tuscany, respectively). Sarda sheep, bred both in Sardinia and in Tuscany, were considered in different groups, both because in these two regions most of the sheep of this breed are reared, and because they differ in geographical characteristics and in the farming system. The individual milk FA composition of dairy ewes was analyzed with multivariate factor analysis. The extracted factors were representative of the following eight groups of fatty acids or functions: factor 1 (odd branched fatty acids and long-chain fatty acids), factor 2 (sn3&#95;position), factor 3 (alternative biohydrogenation), factor 4 (SCD&#95;1), factor 5 (SCD&#95;2), factor 6 (SCD&#95;3), factor 7 (fat secretion) and factor 8 (omega-3). A factor analysis suggested the presence of different metabolic pathways for de novo short- and medium-chain fatty acids and Δ9-desaturase products. The ANOVA of factor scores highlighted the significant effects of the breed. The results of the present study showed that breed is an important factor in defining the fatty acid profile of milk, combined with the effect of the diet. Breeds reared in the same farming system (Comisana, Massese and Sarda reared in Tuscany) showed significant differences for all the factors extracted. At the same time, we found differences between the Sarda sheep reared in Sardinia and Tuscany, two different regions of Italy.

Genomic information allows for a more accurate calculation of relationships among animals than the pedigree information, leading to an increase in accuracy of breeding values. Here, we used pedigree-based and single-step genomic approaches to estimate variance components and breeding values for β-hydroxybutyrate milk content (BHB). Additionally, we performed a genome-wide association study (GWAS) to depict its genetic architecture. BHB concentrations within the first 90 days of lactation, estimated from milk medium infrared spectra, were available for 30,461 cows (70,984 records). Genotypes at 42,152 loci were available for 9,123 animals. Low heritabilities were found for BHB using pedigree-based (0.09 ± 0.01) and genomic (0.10 ± 0.01) approaches. Genetic correlation between BHB and milk traits ranged from -0.27 ± 0.06 (BHB and protein percentage) to 0.13 ± 0.07 (BHB and fat-to-protein ratio) using pedigree and from -0.26 ± 0.05 (BHB and protein percentage) to 0.13 ± 0.06 (BHB and fat-to-protein ratio) using genomics. Breeding values were validated for 344 genotyped cows using linear regression method. The genomic EBV (GEBV) had greater accuracy (0.51 vs. 0.45) and regression coefficient (0.98 vs. 0.95) compared to EBV. The correlation between two subsequent evaluations, without and with phenotypes for validation cows, was 0.85 for GEBV and 0.82 for EBV. Predictive ability (correlation between (G)EBV and adjusted phenotypes) was greater when genomic information was used (0.38) than in the pedigree-based approach (0.31). Validation statistics in the pairwise two-trait models (milk yield, fat and protein percentage, urea, fat/protein ratio, lactose and logarithmic transformation of somatic cells count) were very similar to the ones highlighted for the single-trait model. The GWAS allowed discovering four significant markers located on BTA20 (57.5-58.2 Mb), where the ANKH gene is mapped. This gene has been associated with lactose, alpha-lactalbumin and BHB. Results of this study confirmed the usefulness of genomic information to provide more accurate variance components and breeding values, and important insights about the genomic determination of BHB milk content., (© 2021 The Authors. This bibliographic review presents and discusses the nutritional strategies able to increase the concentration of beneficial fatty acids (FA) in sheep and goat milk, and dairy products, with a particular focus on the polyunsaturated FA (PUFA), and highlights differences between the two species. In fact, by adopting appropriate feeding strategies, it is possible to markedly vary the concentration of fat in milk and improve its FA composition. These strategies are based mostly on the utilization of herbage rich in PUFA, or on the inclusion of vegetable, marine, or essential oils in the diet of lactating animals. Sheep respond more effectively than goats to the utilization of fresh herbage and to nutritional approaches that improve the milk concentration of c9,t11-conjugated linoleic acid (c9,t11-CLA) and α-linolenic acid. Dietary polyphenols can influence milk FA profile, reducing or inhibiting the activity and growth of some strains of rumen microbes involved in the biohydrogenation of PUFA. Although the effectiveness of plant secondary compounds in improving milk FA composition is still controversial, an overall positive effect has been observed on the concentration of PUFA and RA, without marked differences between sheep and goats. On the other hand, the positive effect of dietary polyphenols on the oxidative stability of milk fat appears to be more consistent.

Recently, the interest in industrial by-products produced at the local level in Mediterranean areas, resulting from fruit and vegetable processes, has increased because of their considerable amounts of bioactive compounds, including polyphenols. In this review, we analyze the most recent scientific results concerning the use of agro-industrial by-products, naturally rich in polyphenols (BPRP), in the diets of small dairy ruminants. Effects on milk production, milk and rumen liquor fatty acid profile, metabolic parameters, and methane production are reviewed. The feed intake and digestibility coefficients were generally depressed by BPRP, even though they were not always reflected in the milk yield. The main observed positive effects of BPRP were on quality of the milk's FA profile, antioxidant activity in milk and blood, a reduction of rumen ammonia, and, consequently, a reduction of milk and blood urea. The expected beneficial effects of dietary polyphenols in small ruminants were not always observed because of their complex and variable matrices. However, owing to the large quantities of these products available at low prices, the use of BPRB in small ruminant nutrition offers a convenient solution to the valorization of residues arising from agricultural activities, reducing feed costs for farmers and conferring added value to dairy products at the local level, in a sustainable way., Competing Interests: The authors declare no conflict of interest.

The objectives of this study were (i) the prediction of sheep milk coagulation properties (MCP) and individual laboratory cheese yield (ILCY) from mid-infrared (MIR) spectra by using partial least squares (PLS) regression, and (ii) the comparison of different data pre-treatments on prediction accuracy. Individual milk samples of 970 Sarda breed ewes were analyzed for rennet coagulation time (RCT), curd-firming time (k20), and curd firmness (a30) using the Formagraph instrument; ILCY was measured by micro-manufacturing assays. An Furier-transform Infrared (FTIR) milk-analyzer was used for the estimation of the milk gross composition and the recording of MIR spectrum. The dataset (n = 859, after the exclusion of 111 noncoagulating samples) was divided into two sub-datasets: the data of 700 ewes were used to estimate prediction model parameters, and the data of 159 ewes were used to validate the model. Four prediction scenarios were compared in the validation, differing for the use of whole or reduced MIR spectrum and the use of raw or corrected data (locally weighted scatterplot smoothing). PLS prediction statistics were moderate. The use of the reduced MIR spectrum yielded the best results for the considered traits, whereas the data correction improved the prediction ability only when the whole MIR spectrum was used. In conclusion, PLS achieves good accuracy of prediction, in particular for ILCY and RCT, and it may enable increasing the number of traits to be included in breeding programs for dairy sheep without additional costs and logistics.

The fatty acid (FA), polyphenol content and evaluation of the antioxidant capacity of exhausted Myrtus communis berries (EMB) resulting from the production of myrtle liqueur were assessed. All parts of the exhausted berries exhibited high concentrations of carbohydrates, proteins, lipids and phenolic compounds. The lipid fraction contained a high amount of poly unsaturated fatty acids (PUFA), mainly represented by linoleic acid (>70%). Of the phenolic acids evaluated by liquid chromatography/mass spectrometry, ellagic acid was the most predominant (>50%), followed by gallic and quinic acids. Quercetin and quercetin3-O-rhamnoside were the most abundant flavonoids. The seed extracts showed a higher antioxidant potential than the pericarp extracts; the same trend was observed for total phenolic compounds evaluated by spectrophotometric assay. The overall high content of bioactive compounds and the high antioxidant potential of this byproduct sustain its suitability for a number of industrial applications, such as a food ingredient in novel foods, an additive in cosmetic formulations or a component of animal feed formulations., Competing Interests: The authors declare no conflict of interest.

The present work aimed to investigate the milk oxidative stability when the sheep diet includes a source of polyphenols (grape seed, (GS)) and a source of polyunsaturated fatty acids (linseed, (LIN)), alone or in combination (MIX) compared to a control group (CON). For this purpose light-induced oxidation in milk was studied. After 24 h of light exposure the lipid hydroperoxides increased in milk in the LIN and MIX groups. The calculated ratio between the level of lipid hydroperoxides and unsaturated fatty acids (UFAs) in milk was lower in the GS and MIX than in the LIN group. At the same time the level of the ratio between hexanal/linoleic acid in milk was lower in the GS and MIX than in the CON group. Although the dietary inclusion of grape seed did not reduce the level of lipid oxidation products in sheep milk, it effectively reduced the extent of oxidation of UFA.
