Chronic traumatic encephalopathy (CTE) is a neurodegenerative disorder that is associated with repetitive head impacts. Neuropathologically, it is defined by the presence of perivascular hyperphosphorylated tau aggregates in cortical tissue (McKee et al., 2016, Acta Neuropathologica, 131, 75–86). Although many pathological and assumed clinical correlates of CTE have been well characterized, its effects on cortical dendritic arbors are still unknown. Here, we quantified dendrites and dendritic spines of supragranular pyramidal neurons in tissue from human frontal and occipital lobes, in 11 cases with (M(age) = 79 ± 7 years) and 5 cases without (M(age) = 76 ± 11 years) CTE. Tissue was stained with a modified rapid Golgi technique. Dendritic systems of 20 neurons per region in each brain (N = 640 neurons) were quantified using computer-assisted morphometry. One key finding was that CTE neurons exhibited increased variability and distributional changes across six of the eight dendritic system measures, presumably due to ongoing degeneration and compensatory reorganization of dendritic systems. However, despite heightened variation among CTE neurons, CTE cases exhibited lower mean values than Control cases in seven of the eight dendritic system measures. These dendritic alterations may represent a new pathological marker of CTE, and further examination of dendritic changes could contribute to both mechanistic and functional understandings of the disease.

Long bones are subjected to mechanical loads during locomotion that will influence their biomechanical properties through a feedback mechanism (the bone mechanostat). This mechanism adapts the spatial distribution of the mineralized tissue to resist compression, bending and torsion. Among vertebrates, anurans represent an excellent group to study long bone properties because they vary widely in locomotormodes and habitat use, which enforce different skeletal loadings. In this study, we hypothesized that (a) the cortical bone mass, density and design of anuran femur and tibiofibula would reflect the mechanical influences of the different locomotor modes and habitat use, and (b) the relationships between the architectural efficiency of cortical design (cross-sectional moments of inertia) and the intrinsic stiffness of corti-cal tissue [cortical mineral density; the ´distribution/quality´ (d/q) relationship] would describe some inter-specific differences in the efficiency of the bone mechanostat to improve bone design under different mechanical loads. To test this hypothesis, we determined tomographic (peripheral quantitative computed tomography) indica-tors of bone mass, mineralization, and design along the femur and tibiofibula of four anuran species with different modes of locomotion and use of habitat. We found inter-specific differences in all measures between the distal and proximal ends and mid-diaphysis of the bones. In general, terrestrial-hopper species had the highest values. Arboreal-walker species had the lowest values for all variables except forcortical bone mineral density, which was lowest in aquatic-swimmer species. The d/q relationships showed similar responses of bone modeling as a function of cortical stiffness for aquatic and arboreal species, whereas terrestrial-hoppers had higher values for moments of inertia regardless of the tissue compliance to be deformed.These results provide new evidence regarding the significant role of movement and habitat use in addition to the biomechanical properties of long bones within a morpho-functional and comparative context in anuran species. This study aims to investigate the effect of melatonin on activation, growth and morphology of bovine primordial follicles, as well as on stromal cells density in ovarian tissues after in vitro culture. Ovarian fragments were cultured in α-MEM+ alone or supplemented with melatonin (250, 500, 1,000 or 2,000 pM) for a period of six days. Non-cultured and cultured tissues were processed for histological analysis; according to developmental stages, follicles were classified as primordial or growing follicles. These follicles were further classified as morphologically normal or degenerated. Ovarian stromal cell density was also evaluated. The percentages of primordial and developing follicles, as well as those classified of normal follicles, were compared by Fisher's exact test, and the differences were considered significant when p < .05. The results showed that the presence of 1,000 and 2,000 pM melatonin in culture medium promoted a reduction in the percentage of primordial follicles and an increase in the percentage of development follicles, when compared to follicles cultured in control medium. On the other hand, the presence of 250 or 500 pM melatonin did not show a significant effect on the percentage of primordial and developing follicles. Besides that, the presence of 500, 1,000 and 2,000 pM melatonin maintained the percentage of normal follicles similar to those seen uncultured control. Moreover, tissues cultured in presence of 1,000 pM melatonin showed a higher percentage of normal follicles when compared to follicles cultured in the presence of 250 pM melatonin. It was observed a similar profile of stromal density in both uncultured tissues and those cultured in vitro in the presence of melatonin. In conclusion, melatonin (1,000 and 2,000 pM) promotes bovine primordial follicles activation and maintains the stromal cell density during in vitro culture of ovarian cortical tissue.

A recent field study indicated that thalli of the beard lichen Ramalina usnea undergo diffuse (intercalary) growth throughout their length. We examined thallus sections with TEM to better understand how the highly thickened cell walls of the prosoplectenchymatous cortex behave under conditions of continued expansion. Cell protoplasts were surrounded by massive accumulations of structured electron-dense wall layers interspersed with amorphous, electron-transparent substances, visible as concentric rings in transverse section. Nearest the protoplast, electron-dense wall layers were distinct and more or less alternated with irregular deposits of electron-transparent material. With increasing distance from the protoplast, the electron-dense wall layers were increasingly disrupted and intermixed among the electron-transparent materials. New cell branches grew through the accumulated wall materials, interrupting the layers they penetrated while producing their own concentric wall layers. The differing amounts of cell wall material accumulated was further indication of the different relative ages of such neighbouring cells. These observations suggest that cell walls are disrupted by diffuse tissue expansion and continually replaced by new walls and wall materials deposited to their interior at the interface with the protoplast. This pattern of development, documented previously in R. menziesii and U. longissima, suggests that component cells of lichen prosoplectenchyma behave quite differently from those of diffusely expanding filaments studied in non-lichen-forming fungi, where a single, discrete cell wall is maintained throughout growth., Funding was provided by the Spanish Ministry of Economy and Competitiveness (Grant CTM2015-64728-C2-2-R)

Electrocorticogram (ECoG), obtained by low-pass filtering the brain signal recorded from a macroelectrode placed on the cortex, is extensively used to find the seizure focus in drug-resistant epilepsy and is of growing importance in cognitive and brain–machine-interfacing studies. To accurately estimate the epileptogenic cortex or to make inferences about cognitive processes, it is important to determine the “spatial spread” of ECoG (i.e., the extent of cortical tissue that contributes to its activity). However, the ECoG spread is currently unknown; even the spread of local field potential (LFP) obtained from microelectrodes is debated, with estimates ranging from a few hundred micrometers to several millimeters. Spatial spread can be estimated by measuring the receptive field (RF) and multiplying by the cortical magnification factor, but this method overestimates the spread because RF size gets inflated due to several factors. This issue can be partially addressed using a model that compares the RFs of two measures, such as LFP and multi-unit activity (MUA). To use this approach for ECoG, we designed a customized array containing both microelectrodes and ECoG electrodes to simultaneously map MUA, LFP, and ECoG RFs from the primary visual cortex of awake monkeys (three femaleMacaca radiata). The spatial spread of ECoG was surprisingly local (diameter ∼3 mm), only 3 times that of the LFP. Similar results were obtained using a model to simulate ECoG as a sum of LFPs of varying electrode sizes. Our results further validate the use of ECoG in clinical and basic cognitive research.SIGNIFICANCE STATEMENTBrains signals capture different attributes of the neural network depending on the size and location of the recording electrode. Electrocorticogram (ECoG), obtained by placing macroelectrodes (typically 2–3 mm diameter) on the exposed surface of the cortex, is widely used by neurosurgeons to identify the source of seizures in drug-resistant epileptic patients. The brain area responsible for seizures is subsequently surgically removed. Accurate estimation of the epileptogenic cortex and its removal requires the estimation of spatial spread of ECoG. Here, we estimated the spatial spread of ECoG in five behaving monkeys using two different approaches. Our results suggest that ECoG is a local signal (diameter of ∼3 mm), which can provide a useful tool for clinical, cognitive neuroscience, and brain–machine-interfacing applications.

Intelligent behavior and cognitive functions in mammals depend on cortical microcircuits made up of a variety of excitatory and inhibitory cells that form a forest-like complex across six layers. Mechanistic understanding of cortical microcircuits requires both manipulation and monitoring of multiple layers and interactions between them. However, existing techniques are limited as to simultaneous monitoring and stimulation at different depths without damaging a large volume of cortical tissue. Here, we present a relatively simple and versatile method for delivering light to any two cortical layers simultaneously. The method uses a tiny optical probe consisting of two micro-prisms mounted on a single shaft. We demonstrate the versatility of the probe in three sets of experiments: first, two distinct cortical layers were optogenetically and independently manipulated; second, one layer was stimulated while the activity of another layer was monitored; third, the activity of thalamic axons distributed in two distinct cortical layers were simultaneously monitored in awake mice. Its simple-design, versatility, small-size and low-cost allow the probe to be applied widely to address important biological questions.

IntroductionThe dynamic nature and complexity of the cellular events that take place during the last trimester of pregnancy make the developing cortex particularly vulnerable to perturbations. Abrupt interruption to normal gestation can lead to significant deviations to many of these processes, resulting in atypical trajectory of cortical maturation in preterm birth survivors.MethodsWe sought to first map typical cortical micro and macrostructure development using invivo MRI in a large sample of healthy term-born infants scanned after birth (n=270). Then we offer a comprehensive characterisation of the cortical consequences of preterm birth in 78 preterm infants scanned at term-equivalent age (37-44 weeks postmenstrual age). We describe the group-average atypicality, the heterogeneity across individual preterm infants, and relate individual deviations from normative development to age at birth and neurodevelopment at 18 months.ResultsIn the term-born neonatal brain, we observed regionally specific age-associated changes in cortical morphology and microstructure, including rapid surface expansion, cortical thickness increase, reduction in cortical anisotropy and increase in neurite orientation dispersion. By term-equivalent age, preterm infants had on average increased cortical tissue water content and reduced neurite density index in the posterior parts of the cortex, and greater cortical thickness anteriorly compared to term-born infants. While individual preterm infants were more likely to show extreme deviations (over 3.1 standard deviations) from normative cortical maturation compared to term-born infants, these extreme deviations were highly variable and showed very little spatial overlap between individuals. Measures of regional cortical development were associated with age at birth, but not with neurodevelopment at 18 months.ConclusionWe showed that preterm birth alters cortical micro and macrostructural maturation near the time of fullterm birth. Deviations from normative development were highly variable between individual preterm infants.

Development of cortical tissue during infancy is critical for the emergence of typical brain functions in cortex. However, how cortical microstructure develops during infancy remains unknown. We measured the longitudinal development of cortex from birth to six months of age using multimodal quantitative imaging of cortical microstructure. Here we show that infants’ cortex undergoes profound microstructural tissue growth during the first six months of human life. Comparison of postnatal to prenatal transcriptomic gene expression data demonstrates that myelination and synaptic processes are dominant contributors to this postnatal microstructural tissue growth. Using visual cortex as a model system, we find hierarchical microstructural growth: higher-level visual areas have less mature tissue at birth than earlier visual areas but grow at faster rates. This overturns the prominent view that visual areas that are most mature at birth develop fastest. Together, in vivo, longitudinal, and quantitative measurements, which we validated with ex vivo transcriptomic data, shed light on the rate, sequence, and biological mechanisms of developing cortical systems during early infancy. Interictal epileptiform discharges (IEDs), also known as interictal spikes, are large intermittent electrophysiological events observed between seizures in patients with epilepsy. While seizures are infrequent and unpredictable, IEDs are far more common, often occurring several times per minute. Yet despite the abundance of IEDs, it remains unknown how they relate to seizures. To better understand this relationship, we examined multi-day recordings of 96-channel microelectrode arrays implanted in human epilepsy patients. These recordings (spanning single cell action potentials to population field potentials) allowed us to study the microscale spatiotemporal organization of over 45,000 IEDs across 10 participants from 2 surgical centers. These recordings showed that the majority of IEDs propagate across neocortex as traveling waves. While all of these traveling wave distributions exhibited a predominant, consistent direction, the majority also exhibited a second, auxiliary, direction. Clustering the IED distributions revealed that their predominant and auxiliary distributions were antipodal, mimicking the spatial microstructure of seizure discharges (SDs) that we have previously reported. We thus compared spatial features of IED sub-distributions to those for SDs, showing a correspondence between ictal and interictal spatial properties in participants whose microelectrode arrays were recruited into the seizure from adjacent cortical tissue. These results reveal fundamental relationships between IEDs and seizures and suggest how IEDs could be used to infer spatial features of seizures.

FACS has many challenges that limit comprehensive cell purification from the brain, leading to incomplete molecular characterization. Astrocytes connect the vasculature to neurons and mediate the supply of nutrients and biochemicals. They also remove metabolites from the neurons and extracellular environment. They are involved in a growing number of physiological and pathophysiological processes. Understanding the biophysical, physiological, and molecular interactions in this neuro-glia-vascular ensemble (NGV) and how they support brain function is severely restricted by the lack of detailed cytoarchitecture. To address this problem, we used data from multiple sources to create a data-driven digital reconstruction of the NGV at micrometer anatomical resolution. We reconstructed 0.2 mm3 of rat somatosensory cortical tissue with approximately 16000 morphologically detailed neurons, its microvasculature, and approximately 2500 morphologically detailed protoplasmic astrocytes. The consistency of the reconstruction with a wide array of experimental measurements allows novel predictions of the numbers and locations of astrocytes and astrocytic processes that support different types of neurons. This allows anatomical reconstruction of the spatial microdomains of astrocytes and their overlapping regions. The number and locations of end-feet connecting each astrocyte to the vasculature can be determined as well as the extent to which they cover the microvasculature. The structural analysis of the NGV circuit showed that astrocytic shape and numbers are constrained by vasculature’s spatial occupancy and their functional role to form NGV connections. Synapse density is reduced in postmortem cortical tissue from schizophrenia patients, which is suggestive of increased synapse elimination. Using a reprogrammed in vitro model of microglia-mediated synapse engulfment, we demonstrate increased synapse elimination in patient-derived neural cultures and isolated synaptosomes. This excessive synaptic pruning reflects abnormalities in both microglia-like cells and synaptic structures. Further, we find that schizophrenia risk-associated variants within the human complement component 4 locus are associated with increased neuronal complement deposition and synapse uptake; however, they do not fully explain the observed increase in synapse uptake. Finally, we demonstrate that the antibiotic minocycline reduces microglia-mediated synapse uptake in vitro and its use is associated with a modest decrease in incident schizophrenia risk compared to other antibiotics in a cohort of young adults drawn from electronic health records. These findings point to excessive pruning as a potential target for delaying or preventing the onset of schizophrenia in high-risk individuals.

There are differences in renal biopsy yield related to on-site evaluation, tissue division, and operator, among others. To understand these variations, we collected adequacy-associated data (%cortex, glomeruli, arteries, length) from consecutive native and allograft kidney biopsies over a 22-month period. In total, 1332 biopsies (native: 873, allograft: 459) were included, 617 obtained by nephrologists, 663 by radiologists, and 559 with access to on-site division. Proceduralists with access to on-site evaluation had significantly lower inadequacy rates and better division of tissue for light microscopy (LM), immunofluorescence, and electron microscopy than those without access to on-site evaluation. Radiologists in our region were significantly less likely to have access to on-site evaluation than nephrologists. On multivariate analysis for native kidney biopsies, the effect of having a radiologist perform the biopsy and having access to on-site division were both significant predictors of obtaining greater calculated amount of cortex for LM. Despite the trend for radiologists to obtain more tissue in general, biopsies from nephrologists contained a greater percentage of cortex and were more likely to be considered adequate for LM (native kidney inadequacy rate for LM: 1.11% vs. 5.41%, P=0.0086). Biopsies in which inadequate or marginal cortical tissue was submitted for LM had only minor decreases in the amount of cortex submitted for immunofluorescence and electron microscopy, revealing an opportunity for improved specimen triaging when limited tissue is obtained. In conclusion, both on-site evaluation/division and proceduralist significantly affect quantitative kidney biopsy metrics, which in turn affects the pathologist's ability to render an accurate diagnosis with appropriate prognostic information for the patient and treating nephrologist.

Microglia are the tissue-resident macrophages of the central nervous system (CNS). Recent studies based on bulk and single-cell RNA sequencing in mice indicate high relevance of microglia with respect to risk genes and neuro-inflammation in Alzheimer’s disease. Here, we investigated microglia transcriptomes at bulk and single cell level in non-demented elderly and AD donors using acute human post-mortem cortical brain samples. We identified 9 human microglial subpopulations with heterogeneity in gene expression. Notably, gene expression profiles and subcluster composition of microglia did not differ between AD donors and non-demented elderly in bulk RNA sequencing nor in single-cell sequencing.

Two largely distinct bodies of research have demonstrated age-related alterations and disease-specific aberrations in both local gamma oscillations and patterns of cortical thickness. However, seldom has the relationship between gamma activity and cortical thickness been investigated. Herein, we combine the spatiotemporal precision of magnetoencephalography (MEG) with high-resolution magnetic resonance imaging and surface-based morphometry to characterize the relationships between somatosensory gamma oscillations and the thickness of the cortical tissue generating the oscillations in 94 healthy adults (age range: 22-72). Specifically, a series of regressions were computed to assess the relationships between thickness of the primary somatosensory cortex (S1), S1 gamma response power, peak gamma frequency, and somatosensory gating of identical stimuli. Our results indicated that increased S1 thickness significantly predicted greater S1 gamma response power, reduced peak gamma frequency, and improved somatosensory gating. Furthermore, peak gamma frequency significantly and partially mediated the relationship between S1 thickness and the magnitude of the S1 gamma response. Finally, advancing age significantly predicted reduced S1 thickness and decreased gating of redundant somatosensory stimuli. Notably, this is the first study to directly link somatosensory gamma oscillations to local cortical thickness. Our results demonstrate a multi-faceted relationship between structure and function, and have important implications for understanding age- and disease-related deficits in basic sensory processing and higher-order inhibitory function.

Transcranial magnetic stimulation (TMS) involves the application of time-pulsed magnetic fields to cortical tissue through a coil positioned near the head. TMS is widely used for studying the mechanisms underlying perception and behaviour and is considered a potential therapeutic technique for various conditions. However, the application of TMS has been hindered by the lack of understanding of its mechanism of action. Here we studied the effects of three repetitive TMS (rTMS) paradigms on intra-cortical neuronal responses to oriented gratings in cat area 18. Each of stimulation protocols, including continuous theta-burst, intermittent theta-burst, and 15 Hz rTMS, consisted of 600 pulses. Application of continuous theta-burst and 15 Hz rTMS suppressed the action potential response to oriented grating stimuli for 1.5-6 minutes. In contrast, application of intermittent theta-burst stimulation was associated with enhancement of the action potential response ∼15 minutes following TMS. Neuronal assemblies that were more responsive before the application of rTMS, were affected more than neurons that were less responsive before rTMS. In spite of these changes, the preferred orientation, the orientation tuning of the multi-unit activity and the spatial pattern of the responses recorded from assemblies neurons remained unaffected. Our findings demonstrate that rTMS does not modify the functional selectivities of ensembles of neurons; rather, it has a linear gain effect on their responses.

Access to deep-seated intraventricular lesions frequently requires a focal corticotomy to allow adequate exposure of the affected area. This is necessary to create a surgical corridor to the lesion. However, this may lead to iatrogenic trauma of healthy cortical tissue, subcortical white matter tracts, cortical and subependymal veins. A sulcal approach minimises length of the trajectory through healthy brain tissue but still involves substantial destruction of grey and white matter structures. The conventional corticotomy involves coagulation or suctioning of considerable volume of tissue until the lesion is reached. To minimise damage to the normal brain tissue during ventricular access, the authors describe a technique using Dandy cannula and a finger of an ordinary surgical rubber glove to access ventricle and to obtain the much-desired radial exposure through a tubular corridor. In addition, the Dandy cannula can be linked to frameless neuronavigation to facilitate accurate and shortest trajectory to the lesion.

The data of root anatomical structure and the formation of aerenchyma tissues of five varieties of tobacco under waterlogging stress were obtained by modified paraffin method. Each tobacco varieties performed distinct anatomical adaptation response, including changes of cortical tissue, stele diameter, xylem diameter and the formation of aerenchyma under periodic waterlogging stress. We present a highly scalable assay for whole-genome methylation profiling of single cells. We use our approach, single-cell combinatorial indexing for methylation analysis (sci-MET), to produce 3,282 single-cell bisulfite sequencing libraries and achieve read alignment rates of 68 ± 8%. We apply sci-MET to discriminate the cellular identity of a mixture of three human cell lines and to identify excitatory and inhibitory neuronal populations from mouse cortical tissue.

INTRODUCTION: The dynamic nature and complexity of the cellular events that take place during the last trimester of pregnancy make the developing cortex particularly vulnerable to perturbations. Abrupt interruption to normal gestation can lead to significant deviations to many of these processes, resulting in atypical trajectory of cortical maturation in preterm birth survivors. METHODS: We sought to first map typical cortical micro- and macrostructure development using invivo MRI in a large sample of healthy term-born infants scanned after birth (n = 259). Then we offer a comprehensive characterization of the cortical consequences of preterm birth in 76 preterm infants scanned at term-equivalent age (37-44 weeks postmenstrual age). We describe the group-average atypicality, the heterogeneity across individual preterm infants, and relate individual deviations from normative development to age at birth and neurodevelopment at 18 months. RESULTS: In the term-born neonatal brain, we observed heterogeneous and regionally specific associations between age at scan and measures of cortical morphology and microstructure, including rapid surface expansion, greater cortical thickness, lower cortical anisotropy and higher neurite orientation dispersion. By term-equivalent age, preterm infants had on average increased cortical tissue water content and reduced neurite density index in the posterior parts of the cortex, and greater cortical thickness anteriorly compared to term-born infants. While individual preterm infants were more likely to show extreme deviations (over 3.1 standard deviations) from normative cortical maturation compared to term-born infants, these extreme deviations were highly variable and showed very little spatial overlap between individuals. Measures of regional cortical development were associated with age at birth, but not with neurodevelopment at 18 months. CONCLUSION: We showed that preterm birth alters cortical micro- and macrostructural maturation near the time of full-term birth. Despite all scientific studies, researches are continuing to increase outcomes of freezing ovarian tissue. The objective was to evaluate whether follicular perforation could improve the survival of antral follicles after vitrification. The present study was conducted on 20 bovine ovaries collected at slaughterhouse and a total of 344 antral follicles (less than 4 mm in diameter). For each ovary, three slices (approximately 1 × 1 cm) were separated from the cortex of each ovary and randomly were assigned in three groups, control (without freezing, fixed in 10% buffered formalin), non­perforated vitrified and perforated vitrified (after exposure to equilibration solution, antral follicles were immediately perforated) groups. After thawing, the tissue slices were evaluated histopathologically. Our histological evaluation showed that vitrification in group V1 did not significantly influence the degree of follicle damage compared to the follicles in fresh group as 69.8% of vitrified follicles were appeared as normal. Considering the limited effectiveness of drugs treatments in cognitive disorders, the emergence of noninvasive techniques to modify brain function is very interesting. Among these techniques, repetitive transcranial magnetic stimulation (rTMS) can modulate cortical excitability and have potential therapeutic effects on cognition and behaviour. These effects are due to physiological modifications in the stimulated cortical tissue and their associated circuits, which depend on the parameters of stimulation. The objective of this article is to specify current knowledge and efficacy of rTMS in cognitive disorders. Previous studies found very encouraging results with significant improvement of higher brain functions. Nevertheless, these few studies have limits: a few patients were enrolled, the lack of control of the mechanisms of action by brain imaging, insufficiently formalized technique and variability of cognitive tests. It is therefore necessary to perform more studies, which identify statistical significant improvement and to specify underlying mechanisms of action and the parameters of use of the rTMS to offer rTMS as a routine therapy for cognitive dysfunction.

Swyer syndrome is a 46 XY pure gonadal dysgenesis. Affected individuals are phenotypically female with external female genitalia, hypoplastic Mullerian structures, and the presence of streak gonads. Gonadectomy is indicated due to increased risk of development of malignancy in streak gonads. We hereby report a case of 15-year-old patient with Swyer syndrome who underwent laparoscopic gonadectomy. Histopathological examination showed streak gonads with the presence of accessory adrenal cortical tissue. The patient was put on hormonal replacement therapy postsurgery. Heterotopic adrenal rest may be an incidental finding in normal ovary and testis. Its presence in streak gonads has never been reported, and we propose a theory of increased risk of gonadal and adrenal tumors, possibly because of mutations in disorders of sex differentiation.

Microglia are the tissue-resident macrophages of the central nervous system (CNS). Recent studies based on bulk and single-cell RNA sequencing in mice indicate high relevance of microglia with respect to risk genes and neuro-inflammation in Alzheimer's disease (AD). Here, we investigated microglia transcriptomes at bulk and single-cell levels in non-demented elderly and AD donors using acute human postmortem cortical brain samples. We identified seven human microglial subpopulations with heterogeneity in gene expression. Notably, gene expression profiles and subcluster composition of microglia did not differ between AD donors and non-demented elderly in bulk RNA sequencing nor in single-cell sequencing.

Silicon is a promising material for tissue engineering since it allows to produce micropatterned scaffolding structures resembling biological tissues. Using specific fabrication methods, it is possible to build aligned 3D network-like structures. In the present study, we exploited vertically-aligned silicon micropillar arrays as culture systems for human iPSC-derived cortical progenitors. In particular, our aim was to mimic the radially-oriented cortical radial glia fibres that during embryonic development play key roles in controlling the expansion, radial migration and differentiation of cortical progenitors, which are, in turn, pivotal to the establishment of the correct multilayered cerebral cortex structure. Here we show that silicon vertical micropillar arrays efficiently promote expansion and stemness preservation of human cortical progenitors when compared to standard monolayer growth conditions. Furthermore, the vertically-oriented micropillars allow the radial migration distinctive of cortical progenitors in vivo. These results indicate that vertical silicon micropillar arrays can offer an optimal system for human cortical progenitors&rsquo, growth and migration. Furthermore, similar structures present an attractive platform for cortical tissue engineering.

Advances in computation and neuronal modeling have enabled the study of entire neural tissue systems with an impressive degree of biological realism. These efforts have focused largely on modeling dendrites and somas while largely neglecting axons. The need for biologically realistic explicit axonal models is particularly clear for applications involving clinical and therapeutic electrical stimulation because axons are generally more excitable than other neuroanatomical subunits. While many modeling efforts can rely on existing repositories of reconstructed dendritic/somatic morphologies to study real cells or to estimate parameters for a generative model, such datasets for axons are scarce and incomplete. Those that do exist may still be insufficient to build accurate models because the increased geometric variability of axons demands a proportional increase in data. To address this need, a Ruled-Optimum Ordered Tree System (ROOTS) was developed that extends the capability of neuronal morphology generative methods to include highly branched cortical axon terminal arbors. Further, this study presents and explores a clear use-case for such models in the prediction of cortical tissue response to externally applied electric fields. The results presented herein comprise (i) a quantitative and qualitative analysis of the generative algorithm proposed, (ii) a comparison of generated fibers with those observed in histological studies, (iii) a study of the requisite spatial and morphological complexity of axonal arbors for accurate prediction of neuronal response to extracellular electrical stimulation, and (iv) an extracellular electrical stimulation strength-duration analysis to explore probable thresholds of excitation of the dentate perforant path under controlled conditions. ROOTS demonstrates a superior ability to capture biological realism in model fibers, allowing improved accuracy in predicting the impact that microscale structures and branching patterns have on spatiotemporal patterns of activity in the presence of extracellular electric fields.

Fully-implantable, bi-directional brain-computer interfaces (BCIs) necessitate simultaneous cortical recording and stimulation. This is challenging since electrostimulation of cortical tissue typically causes strong artifacts that may saturate ultra-low power (ULP) analog front-ends of fully-implantable BCIs. To address this problem, we propose an efficient hardware-based method for artifact suppression that employs an auxiliary stimulator with polarity opposite to that of the primary stimulator. The feasibility of this method was explored first in simulations, and then experimentally with brain phantom tissue and electrocorticogram (ECoG) electrode grids. We find that the canceling stimulator can reduce stimulation artifacts below the saturation limit of a typical ULP front-end, while delivering only ~10% of the primary stimulator’s voltage.
