Around 4.5 million tons of leaves are generated each year as a by-product of the olive and olive-oil production. These olive leaves must be removed from the fields and olive-mills. Usually they are burned, or else used as fuels or for animal feed. However, olive leaf is extremely rich in bio-resources, being estimated that around 1 million tons of bioactive compounds, 1 million tons of cellulose and 1.5 million tons of lignin are currently underexploited [1]. In particular, the extraction and isolation of high-added value bioactive compounds in olive leaf (e.g. polyphenols, triterpenoids, essential oils, lipids, lignocellulose) is especially appealing due to their high-market potential in the food, feed, chemical, nutraceutical, cosmetic and pharmaceutical sectors We acknowledge the support through the OLEAF4VALUE project. Polyphenols have been attracting a growing interest by pharmaceutical, biomedical, cosmetic and food industries due to their proven beneficial effects on human health (e.g. antioxidant and anti-inflammatory effects with positive impact on protection of the cardiovascular system, cancer prevention or anti-aging actions). However, an effective bioavailability is an important practical issue. Failing to achieve it leads to no visible benefits of their use. Indeed, fast degradation in common conditions (owing to high sensitivity to temperature, pH and light), poor solubility in biological fluids and high metabolic and secretion rates are some well-known processes limiting polyphenols impact [1]. Thus, the development of vehicles aiming at stabilizing and protecting polyphenols from degradation, also allowing their sustained release, is a key issue to improve the effectiveness of such compounds. In this research, we explore RAFT polymerization to develop tailored materials useful as vehicles for the uptake and release of polyphenols. The preparation and testing of polymersomes based on RAFT-synthetized amphiphilic block copolymers and the generation of hybrid natural/synthetic materials, namely through the RAFT-mediated grafting of synthetic polymers on cellulose, are two different approaches here reported. This research is devoted to the design and synthesis of amphiphilic/stimuli-responsive block copolymers for encapsulation of polyphenols in polymersomes. The Reversible Addition- Fragmentation Chain Transfer (RAFT) polymerization technique is used to get tailored amphiphilic block copolymers. Multiple combinations between hydrophilic/hydrophobic monomers, RAFT agents and initial compositions are the main design variables of the synthesis runs. Experimental studies are guided by modeling tools to aid in the design of the materials (e.g. to specify the sizes of the two blocks, pH sensitivity). Isolated polyphenols (e.g. resveratrol, quercetin) and more complex extracts from different kinds of plants are afterwards encapsulated in polymersomes, aiming at the development of carriers for protection and controlled release of these bioactive compounds. It is shown that the produced aggregates have a long colloidal stability and are promising vehicles for the stimulatedrelease of polyphenols. Owing to their potential beneficial effects on human health, polyphenols (e.g. resveratrol, polydatin, catechin, etc) have been considered in many research works and also for applications in pharmaceutic, biomedical, cosmetic or food industries. The development of carriers allowing to stabilize and protect polyphenols from degradation, thus preserving biological activity and enlarging their bioavailability, is a key issue to achieve the effective benefits of such compounds. On other hand, the use of cellulose materials in many application fields (including the aforementioned industrial areas) is also nowadays an important theme due to the abundance, sustainability and biocompatibility of this natural polymer. Supercritical fluid technology, namely with carbon dioxide, often avoids the use unwanted organic solvents (enabling a more “green” approach) and combines good solubility conditions (associated to liquids) with high diffusion rates (intrinsic to gases). These issues are especially relevant for the transport of high size molecules (as many polyphenols) in complex matrices (such cellulose polymers). Our research combines these three working lines through the synthesis of cellulose-based hydrogels, the supercritical CO2 uploading of polyphenols in the produced materials and the posterior assessment of their liberation for controlled-release purposes (see Figure 1). Chemical crosslinking of cellulose with epichlorohydrin is used to prepare the cellulose-based hydrogels and physically crosslinked alginate hydrogels are considered for comparison purposes. Amphiphilic materials are also synthesized through esterification of OH-groups with a RAFT agent, followed by grafting of synthetic polymer chains. Molecularly imprinted polymer (MIP) particles are here developed to target phenolic compounds present in mixtures resulting from supercritical extraction processes. Engineered MIP particles are synthetized considering different polymerization processes to tailor products morphology (e.g. precipitation or inverse-suspension polymerization to obtain micro-particles [1]) and also diverse functional monomers to explore preferential interactions with the template polyphenols (e.g. polydatin, resveratrol, etc). Moreover, MIP particles with surface grafted functional polymer chains (e.g. using RAFT polymerization [2,3]) are produced to assess the improvement of the selectivity of MIPs towards the target polyphenols, namely through the tuning of the hydrophilic/hydrophobic effects (amphiphilic materials are generated). The produced MIP particles are applied for the identification, separation and concentration of phenolic compounds present in vegetable extracts. Different plants abundant in the Trás-os-Montes and Alto Douro region (e.g. vineyard, chestnut tree, olive tree, cherry tree, etc) are considered as potential sources of phenolic compounds. Supercritical extraction with CO2 is used to obtain the vegetable extracts (see Figure 1) and the effects of the operation conditions (temperature, pressure, vegetable used, etc) on extract composition is also assessed. Molecular recognition capabilities of the MIPs synthetized towards the polyphenols are evidenced (e.g. packing the particles for chromatography) but hydrophilic/hydrophobic interactions are unavoidable and a solvent gradient is needed. info:eu-repo/semantics/publishedVersion

The grafting of functional brushes on the surface of molecularly imprinted polymer (MIP). particles hás been explored in the last few years to synthesize materiais combining high molecular recognition capabilities and stimulation triggered by changes in the surrounding environment [1, 2]. In the present work, MIP particles for 5-fluorouracil (a drug used in câncer treatment) were produced by precipitation polymerization in acetonitrile, using either MAA or HEMA as imprinting fünctional monomers, and m the presence of different kinds of RAFT agents. In a second step, taking advantage of the RAFT groups present in the surface of the particles, different kinds of fiinctional polymer brushes were grafted on the MIPs considering a "grafting from" process in the presence of a RAFT agent. Lithium-ion (Li-ion) batteries are in the front edge of recent achievements concerning energy storage. However, Li-ion devices are reaching their maximum regarding energy density storage which restricts their appiicatíon in systems with large power needs, such as electric vehicles. Driven by this shortcoming, in the last few years, Lithium-Sulfur (Li-S) batteries are being considered as an alternative for the exploitatíon of energy storage and conversion systems with improved performance. Indeed, to the S cathodes is associated a theoretical specifíc capacity of 1672 mA h g-1 and a specifíc energy of 2600 W h kg-\ which are several times higher than the correspondent to other possible systems. The relative low atomic weight of S in comparison with other elements (e.g. cobalt) and the multí-electron transfer reactíons in the pair Li/S are at the source ofthis superior theoretícal performance of Li-S batteries. Population balances of polymer species in terms 'of discrete transforms with respect to counts of groups lead to tractable first order partial differential equations when ali rate constants are independent of chain length and loop formation is negligible [l]. Average molecular weights in the absence ofgelation are long known to be readily found through integration of an initial value problem. The extension to size distribution prediction is also feasible, but its performance is often lower to the one provided by methods based upon real chain length domain [2]. Moreover, the absence ofagood starting procedure and a higher numerical sensitivity hás decisively impaired its application to non-linear reversibly deactivated polymerizations, namely NMRP [3]. BACKGROUND Stimuli responsive imprinted hydrogel micro-particles were prepared using reversible addition-fragmentation chain transfer polymerization for targeting genotoxic impurity aminopyridine in aqueous environment using a continuous flow micro-reactor. RESULTS The feasibility of operation with a continuous flow micro-reactor for particles production was demonstrated. A comparative evaluation was carried out between batch and micro-reactor produced imprinted and non-imprinted hydrogels. Experimental results proved that molecular imprints generated by free radical polymerization and controlled radical polymerization showed outstanding performance in adsorption behavior: the q value estimate was about 1000 times higher than the value presented by other researchers. Solid phase extraction results further evidenced the promising imprinting with hydrogels using free radical polymerization and controlled radical polymerization by retaining c. 100% of 3-aminopyridine. The imprinting factor of 4.3 presented in this research appears to be the best value shown so far. CONCLUSION The imprinted materials were successfully prepared both in batch and with a continuous flow micro-reactor. The inclusion of a reversible addition-fragmentation chain transfer agent in controlled radical polymerization was important in optimizing the experimental conditions in the continuous microfluidic approach. Though the reversible addition-fragmentation chain transfer agent was very useful in controlling the reaction kinetics, imprinted micro-particles showed the existence of both non-specific and imprinted sites. The feasibility of making polymer crosslinked materials with applications in improving energy efficiency is here demonstrated. The encapsulation of phase change materials (PCM), for thermal energy storage/release, and sulphur inverse-vulcanization (producing materials with useful electrochemical properties) are here used as case studies. Effective thermal properties of the produced encapsulated PCM materials were measured through TG/DSC analysis (see graphical abstract). Designing tools allowing the synthesis of materials with tailored thermal properties were exploited (e.g. use of controlled radical polymerization). Sulphur-rich networks are synthetized using different operation conditions (e.g. considering RAFT/NMRP). Testing of these materials in Li-S batteries to assess the impact of the synthesis conditions in their electrochemical properties is ongoing work. Different classes of stimuli-responsive smart hydrogels (SRSH) were synthesized in order to assess the usefulness of molecular imprinting and generation of interpenetrating polymer networks to obtain advanced materials with tailored properties/performance. Reversible Addition-Fragmentation Chain-Transfer (RAFT) polymerisation was exploited as an additional tool to increase the control on the formation process of these materials. Batch adsorption and frontal analysis (e.g. for 3-aminopyridine (3AMP) as depicted in the graphical abstract) techniques were used to quantify the affinity of different drugs with the produced SRSH. Stimulated drug release (e.g. due to temperature/pH changes) and protein immobilisation/release were also tested. Results obtained show that molecular imprinting and generation of interpenetrating networks are effective routes to obtain tailored materials with a particular affinity to selected template molecules. Experimental and theoretical studies concerning the RAFT copolymerization of styrene (S) and commercial divinylbenzene (DVB) were performed in order to assess the use of controlled radical polymerization (CRP) for the production of nonconventional polymer networks. Aqueous suspension polymerizations were carried out at a 1 L scale in a stirred batch reactor and changes of a few operation parameters were tried in order to assess their effect on key product properties. The reaction temperature was kept in the range 80 to 150º C (pressurization used when needed) and different initial ratios RAFT agent/initiator were considered (2 to 10 mol/mol). Monomer dilution in the organic phase was changed between 20 and 50% (v/v) and crosslinker content in the monomer mixture between O and 100% (DVB homopolymerization). Polymerizations were extended beyond the gel point in order to synthesize gel beads. The effect of different commercially available RAFT agents (e.g. S-thiobenzoyl thioglycolic acid and different trithiocarbonates) on network process formation was also studied. Dynamics of gel formation was measured using size exclusion chromatography with refractive index and light scattering detection. Iodine chloride titration yielded pendant double bonds concentration. Network particles morphology was observed by SEM. A general kinetic aproach (Chem. Eng. Sci. 2005, 60, 423) was used for modeling studies, helping in the design of tailored networks. Some rules useful for production engineering could be established . Three modeling approaches for the NMRP of slyrene-divinylbenzene were used for describing experiments performed at different initial conditions: a rigorous approach (model A) based on balance of species and method of characteristics in terms of generating functions and two simplified approaches based upon mass and moment balances, numerical fractionation and pseudo-kinetic method (models B and C). Balances of paths connecting groups were used for tackling loop formation reactions. A critical comparison of the effect of underlying assumptions of lhe three approaches was carried out. Simulations making use of the concept of path balances presented good results for the predictions of pendant double bonds (PDB) concentration. Numerical fractionation can easily allow for empirical reactivity changes with average molecular size (Through the generation index). Thus, simultaneous agreement for predictions of sol weigh-average molecular weigh (Mw) and gel fraction (Wg) could be found with experiments at 90º C. Experiments at 130 º C implied a higher crosslinking reactivity. which might be related lo a higher mobility of polymer chains. This work reports polymer reaction engineering studies on the synthesis process of different classes of smart hydrogels (e.g. super-absorbents, pH/temperature-sensitive). Stimuli-responsive gel beads were obtained using inverse suspension polymerization. Dynamics of product formation was measured using different techniques (e.g. FTIR-ATR, SEC/RI/MALLS). End-use properties of final networks were tested and the morphology of the beads was observed by SEM. A general kinetic approach was exploited to describe the formation of these hydrogels. Temperature/pH stimuli-responsive hydrogel particles were synthesized using inverse-suspension polymerization in batch stirred reactor. Different water soluble comonomers were present in the initial mixture (namely N-isopropylacrylamide and acrylic acid) as well as crosslinkers with different functionalities (bi-, tri- and tetrafunctional) so that their effect on the network crosslinking density could be seen. Different operating conditions such as polymerization temperature (in the range 20 to 70 °C), monomers dilution, neutralization and the inilial ratios 01 co-monomers and monomers/crosslinker were also tried. Classical Iree-radical polymerization (FRP) and RAFT polymerization (e.g. using 4-cyano-4-phenylcarbonothioylthio-pentanoic acid) were compared in order to put into evidence the impact of the polymerization mechanism on the hydrogel molecular architecture. Sampling at different polymerization limes allowed the study of the kinetics of gel formalin through the analysis by SEC of the soluble phase. RI, LALLS, Intrinsic Viscosity and UV signals were simultaneously measured using a tetra-detector array, thus yielding absolute molecular weight, branching factors, hydrodynamic radius and radius 01 gyration. The performance 01 hydrogel beads was assessed through drug delivery tests triggered by changes in the environmental temperature/pH. Aiming at the development of tools hopefully useful for the design of such advanced materials, a general kinetic approach (Chem. Eng. Sci. 2005, 60, 423) was used to carry out modeling studies including consideration of finite loop formation reactions. A kinetic model including the cyclic propagation (cyclization) is proposed for the nitroxide-mediated radical copolymerization of styrene–divinylbenzene. The method involves a balance of sequences of units, which connect a radical center and a pendant double bond present in the same polymer chain. The rate constant for cyclization was considered a function of the sequence length. Good agreement between the model predictions and experimental data for solution and suspension controlled copolymerizations was found. The rate constant of cyclization for the smallest ring (3 monomeric units) was estimated to be 700 s−1 at 90 °C, and the activation energy was estimated to be 32 500 cal mol−1 Marie Curie Initial Training Network “Nanopoly”:. Hydrogels find important applications in drug and gene delivery, tissue engineering, bio-separations and different fields of biomedicine.1 End-use properties of such polymer networks are dependent on their structural topology which is affected by the production process used. Formation of non-ideal networks due to reduced reactivity of pendant double bonds and/or cyclization are some mechanisms hindering the prediction of physical properties of hydrogels from their synthesis conditions, which is an important open problem in this research field. This work reports the synthesis/characterization of different classes of hydrogels and also the kinetic modeling of their formation process. Time evolution in batch reactor of key properties of the gelation process was measured, namely monomer conversion, weight fraction of gel, molecular weights and average radius of gyration of the soluble phase. The performance of the resulting hydrogels was also assessed (e.g. swelling ratio in different environmental conditions) in order to link the synthesis conditions with the end-use properties of such materials. A general kinetic approach is used to predict the dynamics of hydrogel formation. Comparison between experimental measurements and these predictions allows the build-up of a computational tool useful in the design of processes for production of tailored hydrogels. Kinetics of gelation was experimentally and theoretically studied for different polymerization systems leading to the formation of pH/temperature-sensitive hydrogels. Acrylamide (AAm), N-isopropylacrylamide (NIPA), N,Ndimethylacrylamide (DMA), acrylic acid (AA), methacrylic acid (MAA) were used as main monomers, and N,N’- methylenebisacrylamide (MBAm) as crosslinker. The influence of the synthesis conditions on the end use properties of SAPs was experimentally observed. Initial proportion crosslinker/acrylic acid and degree of neutralization are examples of parameters with strong influence in the performance of these materials. The kinetic model developed is able to capture the main features of SAP production. Ongoing research should allow the prediction of elastic properties of this class of materials. Different samples of pH-sensitive hydrogel particles were produced using inverse-suspension, leading to the identification of the influence on their end-use properties of the degree of neutralization of AA monomer and the initial composition of the terpolymerization system which are therefore the key parameters of the synthesis.
