Abstract

Abstract Background Down syndrome (DS), caused by trisomy of human chromosome 21 (HSA21), is the most common genetic birth defect. Congenital heart defects (CHD) are seen in 40% of DS children, and >50% of all atrioventricular canal defects in infancy are caused by trisomy 21, but the causative genes remain unknown. Results Here we show that aberrant adhesion and proliferation of DS cells can be reproduced using a transchromosomic model of DS (mouse fibroblasts bearing supernumerary HSA21). We also demonstrate a deacrease of cell migration in transchromosomic cells independently of their adhesion properties. We show that cell-autonomous proteome response to the presence of Collagen VI in extracellular matrix is strongly affected by trisomy 21. Conclusion This set of experiments establishes a new model system for genetic dissection of the specific HSA21 gene-overdose contributions to aberrant cell migration, adhesion, proliferation and specific proteome response to collagen VI, cellular phenotypes linked to the pathogenesis of CHD.

Abstract

Mycosis fungoides is a T cell lymphoma of the skin, a malignant condition that also involves the lymph nodes and internal organs. The disease does not appear to be caused by infectious agents or environmental factors, but may respond to treatment with the antifungal agent, griseofulvin. Most cases of mycosis fungoides occur in middle-aged and older adults. A case is described of a nine-year-old boy who developed mycosis fungoides at the same site of a previous benign skin lesion. At almost five years of age, the patient was seen for skin lesion on his left finger, which was swollen and scaly. The lesion was successfully treated with corticosteroids but recurred a year later and was partially removed. The following year, the patient developed a rash on his trunk, which was successfully treated with coal tar, and a tumor in the palm of the left hand, which was removed. Two years later, the swelling and scaling of the left hand recurred with abnormal nail changes. Skin abnormalities developed on his upper arms, and right hand and knee. Microscopic examination of the skin lesion from the swollen left finger suggested a diagnosis of mycosis fungoides. The patient was treated with photochemotherapy, or the use of ultraviolet radiation, and the skin lesions improved. (Consumer Summary produced by Reliance Medical Information, Inc.)
