Organisations are increasingly adopting sustainability performance assessment tools. However, these formal organisational sustainability assessments are typically managed and prepared by technical staff. There is a lack of research on approaches that enable a stakeholder-driven performance assessment. This paper develops a framework of informal/complementary stakeholder-driven sustainability performance assessment, from the perspective of employee voluntary collaboration. The framework composes a checklist of questions covering the main sustainability domains: perceptions, individual practices, and voluntary monitoring indicators. In an exploratory case study in a public organisation, the checklist was evaluated by employees in a participatory workshop. The evaluation criteria of understanding and usefulness were rated more positively than reliability. This paper shows a novel way of integrating employee inputs for informal sustainability assessment and supports the importance of empowering public organisations, thereby increasing their understanding of sustainability management frameworks. Understanding the mechanisms of vaccine-elicited protection contributes to the development of new vaccines. The emerging field of systems vaccinology provides detailed information on host responses to vaccination and has been successfully applied to study the molecular mechanisms of several vaccines. Long noncoding RNAs (lncRNAs) are crucially involved in multiple biological processes, but their role in vaccine-induced immunity has not been explored. We performed an analysis of over 2,000 blood transcriptome samples from 17 vaccine cohorts to identify lncRNAs potentially involved with antibody responses to influenza and yellow fever vaccines. We have created an online database where all results from this analysis can be accessed easily. We found that lncRNAs participate in distinct immunological pathways related to vaccine-elicited responses. Among them, we showed that the expression of lncRNA FAM30A was high in B cells and correlates with the expression of immunoglobulin genes located in its genomic vicinity. We also identified altered expression of these lncRNAs in RNA-sequencing (RNA-seq) data from a cohort of children following immunization with intranasal live attenuated influenza vaccine, suggesting a common role across several diverse vaccines. Taken together, these findings provide evidence that lncRNAs have a significant impact on immune responses induced by vaccination.

We have recently found that the silicon trivacancy (V3) is a bistable defect that can occur in fourfold coordinated and (110) planar configurations for both the neutral and singly negative charge states [V. P. Markevich et al., Phys. Rev. B 80, 235207 (2009)]. Acceptor levels ofV3 in both these configurations have been determined. It has also been shown that at T>200 8C, the interaction of mobile trivacancies with interstitial oxygen atoms results in the formation ofV3Ocomplex with the first and second acceptor levels at Ec~0.46 and~0.34 eV. In the present work we identify donor levels arising from V3 and V3O complexes by means of deep level transient spectroscopy (DLTS) and high-resolution Laplace DLTS on n+p silicon structures irradiated with 6MeV electrons, combined with density functional modelling studies. It is found that both defects possess two donor levels in the (110) planar configurations. First donor levels at Ev +0.19 and +0.235 eV, and the second donor levels at Ev +0.105 and +0.12 eV are found for the V3 and V3O complexes, respectively.

O Pênfigo Vulgar é uma doença auto-imune rara na infância,com evolução crónica e mortalidade significativa. A doença manifesta-se por lesões vesiculares, semelhantes a infecções benignas, mas que são refractárias ao tratamento sintomático e persistem após a fase aguda. Apresenta-se o caso de uma criança de três anos com lesões muco-cutâneas orais, persistentes. A biópsia permitiu o diagnóstico de Pênfigo Vulgar, tendo sido iniciada terapia corticosteróide sistémica. A dificuldade no controlo da doença levou à introdução de outros fármacos imunossupressores, até se atingir a estabilidade clínica. Todas as opções de tratamento do Pênfigo apresentam efeitos iatrogénicos, sendo imprescindível um equilíbrio entre controlo da doença e qualidade de vida do doente.

Marsupials have considerable merits as models for studying the developmental dynamics of the mammalian immune system, but until recently there has been a conspicuous lack of specific immune probes to facilitate such studies. To begin a precise study of the ontogeny of the marsupial Didelphis albiventris we have used cross-reactive polyclonal antibodies raised against evolutionarily highly conserved peptides which form part of the antigen specific receptor complexes of human differentiated lymphocytes. Moreover, because of antigen receptor conservation, the antibodies also recognise specifically the immunocompetent T and B lymphocytes of other species including those in the organs of the opossum. Use of the antipeptide antibodies together with other cross-reacting antibodies has allowed us to study the cellular immunology of T and B cells and antigen presenting cells (APC) during the development of thymus, skin, lymph nodes and spleen in the Brazilian white-belly opossum. The molecular nature and identity of the T cell antigens detected in opossum tissues were confirmed by immunoblotting. These findings indicate that it is now possible to exploit these antibody probes for comparative mammalian studies, and indeed to investigate interesting features of the opossum, such as reaction of the immature immune system of the pouch young to antigenic stimulation.

Background: BET (bromodomain and extraterminal) epigenetic reader proteins, in particular BRD4 (bromodomain-containing protein 4), have emerged as potential therapeutic targets in a number of pathological conditions, including cancer and cardiovascular disease. Small-molecule BET protein inhibitors such as JQ1 have demonstrated efficacy in reversing cardiac hypertrophy and heart failure in preclinical models. Yet, genetic studies elucidating the biology of BET proteins in the heart have not been conducted to validate pharmacological findings and to unveil potential pharmacological side effects. Methods: By engineering a cardiomyocyte-specific BRD4 knockout mouse, we investigated the role of BRD4 in cardiac pathophysiology. We performed functional, transcriptomic, and mitochondrial analyses to evaluate BRD4 function in developing and mature hearts. Results: Unlike pharmacological inhibition, loss of BRD4 protein triggered progressive declines in myocardial function, culminating in dilated cardiomyopathy. Transcriptome analysis of BRD4 knockout mouse heart tissue identified early and specific disruption of genes essential to mitochondrial energy production and homeostasis. Functional analysis of isolated mitochondria from these hearts confirmed that BRD4 ablation triggered significant changes in mitochondrial electron transport chain protein expression and activity. Computational analysis identified candidate transcription factors participating in the BRD4-regulated transcriptome. In particular, estrogen-related receptor α, a key nuclear receptor in metabolic gene regulation, was enriched in promoters of BRD4-regulated mitochondrial genes. Conclusions: In aggregate, we describe a previously unrecognized role for BRD4 in regulating cardiomyocyte mitochondrial homeostasis, observing that its function is indispensable to the maintenance of normal cardiac function.
