Pearl culture in Fiji relies on a supply of pearl oysters obtained through the strategic deployment of spat collectors. Although the design and deployment of spat collectors aim to maximize the recruitment of pearl oysters (Pinctada and Pterin spp.), this activity incidentally captures many other bivalves as bycatch. This study explored whether tropical rock oysters (specifically Magallana bilineata and Saecostrea spp.) were present and sufficiently abundant as bycatch from pearl oyster spat collectors, to support additional commercial opportunities. The results showed that the absolute (0.7 [+ or -] 2.1 spat collector-1) and proportional (2.9% of all bivalves) abundance of these taxa was very low, which would limit oyster harvest in meaningful quantities. Although the prospect of creating additional revenue streams from bycatch could be seen as an incentive to on-grow and sell tropical rock oysters despite their low abundance, it is argued that greater benefit would come from prioritizing activities related to pearl oysters and associated downstream products such as mabe pearl, shellcraft, and pearl-meat production. Ultimately, existing spat collection activities within the cultured pearl sector did not have the anticipated potential to support supplemental aquaculture of tropical rock oysters. This study elucidates to generate a linkage map for Portuguese oysters, Crassostrea angulata, and recognise potential markers linked to economical traits in C. angulata. Genome-wide association studies (GWAS) using 19,475 single nucleotide polymorphisms (SNPs) generated by a genotyping-by-sequencing approach consisting of 647 individuals in two selection generations of C. angulata, consisting of 57 full-sib family of the first generation and 33 full-sib family of the second generation. GWAS identified six significant SNPs linked to whole weight (P < 10−4). These six SNPs explained 10.2% of the total genetic variance of whole weight. Additionally, three SNPs were identified linked to meat yield (i.e., soft tissue weight) and contributed to 9.2% of the total genetic variance for this trait. By linkage analysis on the raw data, a total of 19,475 SNP markers were mapped to 10 linkage groups, with an average density of 19.1 SNPs per cM. This linkage map was developed and potential markers identified for whole weight and soft tissue weight provide a novel resource to characterise the genetic structure of economic traits in C. angulata. The Suminoe oyster Crassostrea ariakensis (Fujita, 1913) is one of the most important ecological and fishery bivalve mollusks with a worldwide distribution. Here, we reported an improved high-quality chromosomal-level genome assembly of C. ariakensis inhabiting the South China Sea, using Nanopore technology, Illumina sequencing, and high-throughput chromosomal conformation capture analysis. The assembled genome size is 631.73 Mb, with contig N50 length of 5.36 Mb and scaffold N50 length of 61.15 Mb, and is assigned to 10 chromosomes. A total of 29,357 protein-coding genes are predicted, 96.68% of which are functionally annotated. The genome contains 347.11 Mb (54.94%) of repetitive elements and 1130 noncoding RNAs. This improved genome assembly of south C. ariakensis is an important resource for understanding oyster diversity and evolution, and provides insights into genetic improvement, protection and management of oyster resource. Molluscan shellfish aquaculture contributes to 42.6% of global aquaculture production. With a continued increase in shellfish production, disposal of shell waste during processing is emerging as an environmental and financial concern. Whilst major commercial species such as Crassostrea spp. has been extensively investigated on usage of its shell, with information that are crucial for valorisation, e.g. safety and crystal polymorphs, evaluated. There is currently little understanding of utilisation opportunities of shell in several uprising Australian commercially harvested species including Akoya Oyster (Pinctada fucata), Roe's Abalone (Haliotis roei) and Greenlip Abalone (Haliotis levigata), making it challenging to identify ideal usages based on evidence-based information. Therefore, in this study, an interdisciplinary approach was employed to characterise the shells, and thereafter suggest some potential utilisation opportunities. This characterisation included crude mineral content, elemental profiling and food safety evaluation. As well, physical, chemical, and thermal stability of the shell products was assessed. TGA result suggests that all shells investigated have high thermal stability, suggesting the possibility of utilisation as a functional filler in engineering applications. Subsequent FTIR, SEM and XRD analyses identified that CaCO3 was the main compositions with up to 77.6% of it found to be aragonite. The spectacular high aragonite content compared to well-investigated Crassostrea spp. suggested an opportunity for the utilisation of refined abalone shell as a source of biomedical engineering due to its potent biocompatibility. Additionally, safety evaluations on whole shell also outlined that all investigated samples were safe when utilised as a crude calcium supplement for populations > 11 years old, which could be another viable options of utilisation. Oysters, particularly Portuguese oyster (Crassostrea angulata), are highly valued for their nutritional and flavor qualities, making them important in global aquaculture. Triploid oysters have gained attention for maintaining higher meat quality year-round compared to diploids, but there is limited research on how ploidy affects their biochemical and flavor profiles. This study uses a non-targeted metabolomics approach, including gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-mass spectrometry (LC-MS), to investigate flavor substance differences between triploid and diploid C. angulata. A total of 13 volatile compounds were identified in diploid oysters, while 28 were found in triploids. Significant upregulation of inosine, guanosine, L-aspartic acid, and taurine in triploids contributes to their enhanced flavor profile. Additionally, triploids showed higher nicotinamide concentrations, while diploids had increased 25-hydroxycholesterol. These findings highlight the advantages of triploid oysters in aquaculture for improved flavor and nutrition, supporting their potential for year-round production. [ABSTRACT FROM AUTHOR]

In order to create new value in biomass production using sterile Ulva with high production efficiency, we selected sterile Ulva with high culture stability from the coast of Imari City in Japan and investigated the conditions for generating high concentrations of dissolved oxygen through photosynthesis using the sterile Ulva. In the cultivation of sterile Ulva, we investigated the conditions for generating high concentrations of dissolved oxygen (DO) by controlling conditions such as aeration, NO3‐N concentration, and population density. As a result, we found that under the cultivation control conditions we found, DO reached about 3.8 times the saturation concentration after 3 h. It was found that this high level of DO can be maintained if the supersaturated DO seawater obtained here is properly sealed. Seawater containing high levels of DO is essential for the cultivation of oysters and shellfish, which also prefer a growth temperature of 20°C−25°C. Therefore, from the perspective of creating new value in biomass production, we developed a new system for simultaneously cultivating sterile Ulva and cultivating oysters. [ABSTRACT FROM AUTHOR]

Arcobacter-related species are considered emerging food-borne and waterborne pathogens, with shellfish being a suggested reservoir. In a published study that investigated 204 shellfish samples and 476 isolates, the species Arcobacter butzleri (now known as Aliarcobacter butzleri) and Arcobacter molluscorum (now known as Malaciobacter molluscorum) have been isolated as the most dominant species. However, the efficiency of depuration for eliminating A. butzleri and M. molluscorum in comparison with Escherichia coli from mussels and oysters is unknown and is therefore the objective of this investigation. The shellfish depuration process was evaluated in the laboratory, in summer and winter, using mussels and oysters collected from the Ebro Delta harvesting areas after performing a natural contamination and an artificial contamination using the same conditions for both mollusk and seasons. The natural contamination was performed by exposing the shellfish to a freshwater channel that receives untreated sewage from the village of Poble Nou (PNC) and that had a salinity of 10.7–16.8‰. The artificial contamination exposed the shellfish to A. butzleri and E. coli (in one tank) and to M. molluscorum and E. coli in another tank under controlled conditions of salinity (34.5‰) and temperature (20 °C summer and 14 °C winter). When evaluating the reduction in the bacteria load (every 24 h) throughout 120 h, the naturally contaminated shellfish at the PNC showed a higher reduction than the shellfish contaminated at the laboratory, with the exception of M. molluscorum, that at 24 h could not be detected in summer, neither in mussels nor in oysters. This may be attributed to the fact that the bacteria from the PNC were less adapted to the conditions of high salinity (34.5‰) in which the depuration process was performed. Although temperature did not statistically make a difference in depuration, at 20 °C a higher elimination of all bacteria was recorded relative to 14 °C. In general, E. coli survived more in mussels than in oysters, and M. molluscorum suffered in both mollusks a higher reduction than A. butzleri. Biofloc technology is instrumental in improving growth performance and yield in shellfish aquaculture, while leading to enhanced water quality through maintaining the nutrients level within a safe range. More specifically, toxic nitrogenous wastes are converted into beneficial microbial biomass known as "biofloc", contributing to improve shellfish immune system. Among the various parameters affecting the efficiency of the process is the carbon source and C:N ratio. In light of these, the present work critically reviews the effects of biofloc systems on growth performance, immunity and diseases resistance in shellfish production. Moreover, it scrutinizes the microbial diversity and nutritional composition of biofloc. Then, the application of the technology in various shellfish cultures, including shrimp, freshwater prawn, crabs, crayfish, clam, and oyster, is presented. Overall, biofloc systems contribute to enhanced shellfish survival rate to the highest value of 96–100% for marine shrimp, 95–99% for freshwater prawn, 70–83% for crayfish, 83–100% for oyster, Crassostrea sp. and up to 2% for mud crabs larvae through substantially reducing the ammonia level in the culture (summarized in Table 1 – Table 5). Finally, the main challenges in utilizing biofloc systems, i.e., suitable aeration and mixing and microbial mechanisms involved are also explained to shed light on future research directions in the field. Their draft genomes will play an important role in the application of genomic methods such as RNAseq, population-based genomic scans aiming at addressing expression responses to pollution stress, adaptation to salinity and temperature variation, and will also permit investigating the genetic bases and enable marker-assisted selection of economically important traits like shell and mantle coloration and resistance to temperature and disease. Data description The draft assembly size of Crassostrea gasar is 506 Mbp, and of Crassostrea rhizophorae is 584 Mbp with scaffolds N50 of 11,3 Mbp and 4,9 Mbp, respectively. The general masked bases by RepeatMasker in both genomes were highly similar using different datasets. The masked bases varied from 9.41% in C. gasar to 10.05% in C. rhizophorae and 42.85% in C. gasar to 44.44% in C. rhizophorae using Dfam and RepeatModeler datasets, respectively. Functional annotation with eggNog resulted in 34,693 annotated proteins in C. rhizophorae and 26,328 in C. gasar. BUSCO analysis shows that almost 99% of genes (5,295) are complete in relation to the mollusk orthologous genes dataset (mollusca_odb10).

Oyster cultivation is a pivotal economic industry in Hainan Island, South China, where a high oyster species diversity is shown. However, the specific distribution and biodiversity of oyster resources in the island have remained unclear. To elucidate the diversity of oyster species and their distribution there, 307 oyster samples were collected from 29 sites in the intertidal zone around the island, and were identified using both morphological and molecular approaches. A minimum of 12 oyster species were identified in taxonomy, including Crassostrea species (C. gigas angulata, C. sikamea, C. iredalei, C. dianbaiensis, C. talonata, C. ariakensis, and C. hongkongensis), and Saccostrea species (S. malabonensis, S. mordax, S. echinata, S. circumsuta, and S. mordoides). The results revealed a remarkable diversity of oyster species along the coast of the island. Particularly noteworthy are that S. malabonensis and S. mordax constituted 36% and 22% of the collected specimens, respectively. This study provided a comprehensive overview on current state of oyster biodiversity in Hainan, serving as a valuable reference for conservation and research on species distribution and resource dynamics in oyster populations. Fossil oysters are commonly found in Rio Grande do Sul Coastal Plain (RSCP), where the main species are Ostrea cf. puelchana d'Orbigny, 1841 and Crassostrea praia Ihering, 1907. These fossils are transported towards the beach by storm events throughout the year, being available in the swash zone. The objective of this article is to reconstruct the Holocene palaeoenvironment by analysing these fossils. A total of 1898 valves, collected along Cassino and Hermenegildo beach, both in southernmost Brazil, were described. It was observed a prevalence of C. praia at Cassino beach (39.20%) and O. cf. puelchana at Hermenegildo beach (89.02%). Hermenegildo beach sample were statistically greater for C. praia height (p -value: 0.1736) and length (p -value: 0.4604) and for O. cf. puelchana height (p -value: 0.0602) and length (p -value: 0.07394). The predominance for northern and southern regions for each species indicates a palaeoenvironment setting that favoured these organisms, probably related to temperatures and salinity variation. The most common ichnogenera found in the oysters were Caulostrepsis and Entobia present in both sites, observed in over 40% of internal and external sides of the samples' shell. According to radiocarbon modelled age, the studied samples coexisted during the Mid-Holocene (8.290–8.130 cal. yr. BP), with the subsequent sea-level rise and fall the shells became bioavailable. The lack of samples with closed valves and high percentage of bioerosion in the inner side of the valve, indicates that the palaeoenvironment in both Cassino and Hermenegildo beaches gradually changed, not as result of an extreme event. Maintaining genetic diversity in cultured shellfish can be challenging due to high variance in individual reproductive success, founder effects, and rapid genetic drift, but is important to retain adaptive potential and avoid inbreeding depression. To support broodstock management and selective breeding in cultured Pacific oysters (Crassostrea (Magallana) gigas), we developed an amplicon panel targeting 592 genomic regions and SNP variants with an average of 50 amplicons per chromosome. Target SNPs were selected based on elevated observed heterozygosity or differentiation in Pacific oyster populations in British Columbia, Canada. The use of the panel for parentage applications was evaluated using multiple generations of oysters from a breeding program on Vancouver Island, Canada (n = 181) and families selected for Ostreid herpesvirus-1 resistance from the Molluscan Broodstock Program in Oregon, USA (n = 136). Population characterization was evaluated using wild, naturalized, farmed, or hatchery oysters sampled throughout the Northern Hemisphere (n = 189). Technical replicates showed high genotype concordance (97.5%; n = 68 replicates). Parentage analysis found suspected pedigree and sample handling errors, demonstrating the panel's value for quality control in breeding programs. Suspected null alleles were identified and found to be largely population dependent, suggesting population-specific variation impacting target amplification. Null alleles were identified using existing data without the need for pedigree information, and once they were removed, assignment rates increased to 93.0 and 86.0% of possible assignments in the two breeding program datasets. Superheated steam (SS) is promising for the thermal shucking application of oysters due to its high energy. Tropical oysters Crassostrea iredalei was probed with K-type thermocouples connected to a datalogger to monitor and record process temperatures during superheated steam pretreatment at different temperatures (170, 210, and 250 °C) and holding time (4, 5, 6 min) followed by rapid ice immersion. The combination of heating and rapid chilling on the release of oyster meat during shucking was evaluated for muscle relaxation, muscle release, meat quality, texture profile analysis, and color and optimized for maximum muscle release and meat quality. The thermal profiles of SS revealed rapid temperature increment of both muscle and oyster shell compared to conventional steam, resulting in increased relaxation of adductor muscles and degree of meat release with an increase in temperature and time. Texture of the oyster meat retained the hardness, gumminess, and chewiness of the fresh oyster at low superheated steam temperature and short exposure time. Sufficient meat detachment of more than 85% was achieved at 170 °C and 5 min with minimum meat quality changes which were also lower in microbial load (4.1 × 103 CFU/g) and TVB-N (11.7 mg/100 g) with higher final pH (6.3) compared to untreated [microbial load (5.2 × 103 CFU/g), TVB-N (15.7 mg/100 g), final pH (5.9)] and microwave-treated [microbial load (4.3 × 103 CFU/g), TVB-N (21.0 mg/100 g), final pH (6.0)] samples. Morphological alterations of the gill and digestive gland were studied in Magallana gigas from areas with different degrees of human activity over one year. For that, the height of the plicas and filaments of the gill and the mean epithelial thickness were measured, the ratio of cell type and tissue composition analyzed of the digestive gland as well as alterations in the pattern of structural organization in these organs were assessed. The specimens were collected in November 2021, and February, May and August 2022 from two representative sampling sites of distinct anthropic pressures in the coastal area of Buenos Aires Province: Arroyo Parejas (AP, 38° 55′ S, 62° 04′ W) and Pehuén-Có (PC, 39° 01′ S; 61° 32' W). The results of the morphological parameters, ratio of cell-type, tissue composition, and histological alterations of M. gigas showed differences between the studied areas and seasons, suggesting a negative relationship between human activity and the health of the gills and digestive gland of the oyster. Understanding the gill and digestive gland morphology of oysters, as well as their response to varying levels of human activity in the study region, could help mitigate the impacts of this non-native species. Majority of macrozooplankton have a wider dietary niche breadth and utilize small invertebrates, microzooplankton and mesozooplankton, so effect on primary production might be through trophic cascading effect. To better understand the ecosystem structure of benthic oyster-macroalgae reefs, we analyzed zooplankton community structure before (July 2016) and after (from September 2016 to October 2017) the construction of benthic reefs in the 2 km2 sea ranch area in Xiangyun Cove, Tangshan, China. We identified 57 zooplankton species, including the 12 cnidarian (e.g., Clytia hemisphaerica Linnaeus and Eirene ceylonensis Browne), 1 ctenopharyngodon (Pleurobrachia globosa Moser), 24 crustacean (e.g., Calanus sinicus Brodsky, Paracalanus parvus Claus, Labibocera euchaeta Glesbrecht, Labibocera bipinnata Tanaka, Calanopia thompsoni Scott, and Centropages dorsispinatus Thompson), 1 chaetognath (Sagitta crassa Tokioka), 1 urochordate species (Oikopleura dioica Fol), and 18 species of planktonic polychaete and gastropod larvae. The zooplankton density and biomass values before reef construction were 266.14 ind/m3 and 2.72 mg/m3, respectively, and those after reef construction were 138.06 ind/m3 and 32.91 mg/m3, respectively. The biomass trend was as follow: October 2017 (89.08 mg/m3) > August 2017 (70.97) > September 2016 (3.17) > July 2016 (2.72) > June 2017 (0.86) > May 2017 (0.44). The common dominant organisms were crustaceans and chaetognaths. According to the RDA ranking results, water temperature was positively correlated with the Shannon-Wiener diversity index and Margalef's richness indexes. With the increasement of Margalef's richness index, the value of dissolved oxygen content showed a significant negative correlation with zooplankton abundance. The results of this study are applicable to sustainable development and management strategies of coastal reef ecosystems and provide a basis for further surveys of secondary productivity in the sea ranch area. Dakhla Bay, situated on the African Atlantic coast, has witnessed sporadic harmful algal blooms (HABs) caused by toxic dinoflagellate species over the past two decades. In this study, we investigated the distribution, abundance, and diversity of dinoflagellate cysts, with a focus on potentially toxic species that develop in this ecosystem where such data are lacking. Sediment samples were collected in April 2018 through coring at 49 stations distributed across the bay. The highest abundance of dinoflagellate cysts was recorded at 304 cysts/g dry sediment, observed at the inner part of the bay, indicating that this area is the preferential zone for cyst accumulation. Pearson's tests revealed significant positive correlations (P < 0.05) between cyst abundance and the water content, organic matter, and fine fraction (< 63 μm) of the sediment. Cyst morphotypes of potentially toxic dinoflagellate species known to produce saxitoxins, such as Alexandrium minutum, Alexandrium tamarense species complex, Gymnodinium catenatum, and yessotoxins, such as Lingulodinium polyedrum and Gonyaulax cf. spinifera, were identified in the sediment of Dakhla Bay. These findings were further supported by our long-term monitoring period (2005–2018), underscoring the presence of these HAB species in Dakhla Bay. During our survey, sporadic mollusk intoxication events were recorded at station PK25 for the grooved razor shell Solen marginatus and at station Boutelha for the oyster Crassostrea gigas. Paralytic shellfish toxin concentrations exceeded the sanitary threshold (80 μg STX di-HCl eq/100 g of shellfish meat) only twice, in December 2006 and January 2007 at station PK25. Contamination by amnesic shellfish toxins occurred sporadically but never reached the sanitary threshold of 20 µg/g of shellfish meat. Lipophilic shellfish intoxication occurred multiple times in the two investigated areas. These observations suggest that the cysts of the identified HAB species germinated and inoculated the water column, resulting in the observed intoxication events. Relatively low levels of intoxication could be attributed to the moderate abundances of cysts, which may reduce the seeding capacity. This could be explained by the significant interaction of Dakhla Bay with the Atlantic Ocean, characterized by hydrological dynamics that impede the deposition and accumulation of cysts in the bay's sediments. Microplastic ingestion by marine organisms presents a challenge to both ecosystem functioning and human health. We characterized microplastic abundance, shape, size, and polymer types ingested by the West African mangrove oyster, Crassostrea tulipa (Lamarck, 1819) sampled from estuaries and lagoons from the Gambia, Sierra Leone, Ghana, Benin, and Nigeria using optical microscopy and Fourier transform infrared (FTIR) techniques. A total of 780 microplastics were isolated in the whole tissues of the 250 oysters (n = 50 oysters per country). The abundance and distribution of microplastics in the oysters followed the pattern: the Gambia > Ghana > Sierra Leone > Nigeria > Benin. The Tanbi wetlands in the Gambia recorded the highest average of 10.50 ± 6.69 per oyster while the Ouidah lagoon in Benin recorded the lowest average of 1.80 ± 1.90 per oyster. Overall, microplastic numbers varied significantly (p < 0.05) among the five countries. Microfibers, particularly those within 1001–5000 μm size, dominated the total microplastic count with a few fragments and films. No spherical microplastics were isolated in the oysters. In the Sierra Leone and Benin oysters, fragments and films were absent in the samples. Microplastic between the 1001 and 5000 μm size class dominated the counts, followed by 501–1000 μm, 101–500 μm, and 51–100 μm. Five polymer groups namely polyethylene, polyester, nylon, polypropylene, and polyamide were identified across the five countries, with polyethylene occurring in oysters from all five countries and polyester occurring in all but the oysters from Nigeria. This diversity of polymers suggests varied sources of microplastics ingested by the studied oysters. Four species of shellfish, blue mussel (Mytilus galloprovincialis), Pacific abalone (Haliotis discus hannai), zhikong scallops (Chlamys farreri), and Pacific oyster (Crassostrea gigas), were exposed to decoupled carbonate system variables to investigate the impacts of different seawater carbonate parameters on the CO2 excretion process of mariculture shellfish. Six experimental groups with two levels of seawater pH (pH 8.1 and pH 7.7) and three levels of total alkalinity (TA = 1000, 2300, and 3600 μmol/kg, respectively) were established, while pH 8.1 and TA = 2300 μmol/kg was taken as control. Results showed that the CO2 excretion rates of these tested shellfish were significantly affected by the change in carbonate chemistry (P < 0.05). At the same TA level, animals incubated in the acidified group (pH 7.7) had a lower CO2 excretion rate than those in the control group (pH 8.1). In comparison, at the same pH level, the CO2 excretion rate increased when seawater TA level was elevated. No significant correlation between the CO2 excretion rate and seawater pCO2 levels (P > 0.05) was found; however, a significant correlation (P < 0.05) between CO2 excretion rate and TA-DIC (the difference between total alkalinity and dissolved inorganic carbon) was observed. Blue mussel has a significantly higher CO2 excretion rate than the other three species in the CO2 excretions per unit mass of soft parts, with no significant difference observed among these three species. However, in terms of CO2 excretion rate per unit mass of gills, abalone has the highest CO2 excretion rate, while significant differences were found between each species. Our studies indicate that the CO2 buffering capacity impacts the CO2 excretion rate of four shellfish species largely independent of pCO2. Since CO2 excretion is related to acid–base balancing, the results imply that the effects of other carbonate parameters, particularly the CO2 buffering capacity, should be studied to fully understand the mechanism of how acidification affects shellfish. The chloride channel 7 gene (CLC7) of the Hong Kong oyster Crassostrea hongkongensis was cloned and named ChCLC7. The cDNA was 2 572 bp in length, with a 5′ non-coding region containing 25 bp, a 3′ non-coding region containing 327 bp, and an open reading frame of 2 298 bp. ChCLC7 has 96.8% and 92.1% homology with CLC7 of Crassostrea gigas and Crassostrea virginica, respectively, and it was clustered with CLC7 of C. gigas and C. virginica. QRT-PCR showed that ChCLC7 was expressed in all eight tissues, with the highest in adductor muscle and second in gill. The ChCLC7 expression pattern in gill was altered significantly under high salinity stress with an overall upward and then downward trend. After RNA interference, the expression of ChCLC7 and survival rate of oyster under high salinity stress was reduced significantly, and so did the concentration of hemolymph chloride ion in 48–96 h after RNA interference. We believed that ChCLC7 could play an important role in osmoregulation of C. hongkongensis by regulating Cl− transport. Marine molluscan cell lines, required for virus screening and cultivation, form essential tools for developing health management strategies for these animals in the blue economy. Moreover, they are also crucial to develop cultivated seafood. As there is no valid marine molluscan cell line, primary cell cultures are relied upon for all investigations. A sound protocol for generating primary cell cultures from molluscs is entailed, but existing protocols often involve heavy antibiotic usage and depuration that invariably affect gene expression and cell health. This work presents an easy‐to‐adopt, time‐saving protocol using non‐depurated mollusc Crassostrea madrasensis, which requires only initial antibiotic treatment and minimal exposure or no use of antibiotics in the cell culture medium. The important experimental considerations for arriving at this protocol have been elucidated. Accordingly, sodium hypochlorite and neomycin sulfate were chosen for disinfecting tissues. The study is the first to use shrimp cell culture medium (SCCM) as a cell culture medium for molluscan cell culture. Despite being osmoconformers, the oysters exhibited stable intracellular osmotic conditions and pH, which, when provided in vitro, promoted effective cardiomyocyte formation. The cell viability could be enhanced using 10% fetal bovine serum (FBS), but healthy cell culture could also be obtained using SCCM without FBS. The optimized culture conditions allowed for regular beating cardiomyocyte clusters that could be retained for a month. Limited cell proliferation, as shown by the BrdU assay, demands further interventions, such as possibly producing induced pluripotent stem cells. Native Olympia oysters (Ostrea lurida) occur in remnant natural populations throughout San Francisco Bay (SFB), California, United States, where managers have been working to restore oysters since 2002. Environmental conditions vary substantially along the estuarine gradient in the bay, such that oysters are exposed to different stressors in different locations and years. We monitored oysters that naturally recruited to experimental restoration substrates over 10 years at two sites on this gradient. We found that recruitment rates and adult densities varied over time and had different trajectories at the study locations. We found gradients in predation, space competition, and physical stressors between sites and along a tidal elevation span that corresponded with differences in adult densities. We documented mass mortality resulting from an extreme low‐salinity event at one site and subsequent recovery later that year. Our results, combined with data from natural populations at multiple sites, suggest that restoration outcomes within SFB could be improved by building and strengthening a connected portfolio of sites that are not simultaneously exposed to the same stressors. We recommend siting restoration projects across locations that represent a range of environmental conditions but that are sufficiently close for larval exchange. Unlike typical restoration approaches that use site selection to identify the best locations, the portfolio approach, just like the economic investment principle, would employ a diversity of environmental conditions. Suspended oysters employ physiological strategies to adjust their metabolic needs with the available food resources. Using the biodeposition method, the feeding and processing behavior of Pacific oysters (Crassostrea gigas) was investigated with a field study comparing four periods (May, July, October, and December 2016) with different upwelling intensities in the coastal lagoon of San Quintin Bay (Mexico). We calculated physiological feeding responses throughout the culture cycle, including the clearance rate (CR), filtration rate (FR), net organic ingestion rate (NOIR), net organic absorption rate (NOAR), net organic selection efficiency, net absorption efficiency, and the ammonium excretion rate (AER). The dietary quality predictors showed large fluctuations in terms of total particulate material, organic fraction of seston, and chlorophyll concentration. Unlike the pumping activity, FR, NOIR, and NOAR were related to upwelling conditions, and C. gigas removed twofold, ingested fourfold, and assimilated fivefold more of the organic suspended material during the upwelling season compared with periods of weak upwelling. C. gigas showed the potential of depositing nearly twice the organic biodeposits to the sediments during the intense upwelling events. The highest AER was recorded in July and October, suggesting that seasonal temperature variation is the most important exogenous factor regulating nitrogen metabolism, even in a subtropical environment. Also, mechanistic models incorporating dietary quality predictors to the feeding and processing response functions of C. gigas were performed. We conclude that coastal upwelling plays an important bottom-up control on oysters' feeding and processing activity, and our results facilitate further studies of the carrying capacity of embayments influenced by eastern boundary current systems. The content of trace elements from Crassostrea tulipa (Ostreidae, Bivalvia) from four localities (Pra Estuary, Densu Estuary, Kpone Beach, and Anyanui Creek) along the coast of Ghana has been used as a bioindicator of environmental pollution. The lowest values of heavy metals correspond to the Crassostrea shells from Densu Estuary and Anyanui Creek, whereas the highest contents are recorded in the shells from Pra Estuary and Kpone Beach. Densu Estuary and Anyanui Creek are located in areas not affected by major industries, and only Densu is located in a densely urbanized setting. Crassostrea shells from Densu Estuary are only enriched in Ni. However, the Total Organic Carbon content of the sediments of the Densu Estuary indicates anthropogenic organic pollution not evidenced by the composition of Crassostrea shells. The Crassostrea shells from the Pra Estuary show enrichment in most of the analysed trace elements (Li, Be, V, Cu, Cr, Zn, Ga, Y, Sn, Pb, U, Th, ∑REE, and As) compared with other studied sites. This enrichment is related to heavy metals influx to the Pra River by the mining activities, extensive forest clearance, and loss of soil minerals. The Crassostrea shells from Kpone Beach present comparatively high content of some trace elements (Be, Ba, Cu, Zn, Nb, Pb, and REE). The metal pollutants are related to the input of the Heavy Industrial Area of Tema (located around 3.5 km west of Kpone Beach) and urban effluences from the Greater Accra Region. The high content of Ba, Cu, Zn, and Pb in Crassostrea is directly related to high phytoplankton productivity (mainly for diatoms) linked to the input of trace metals transferred into the marine food webs. In addition, some analyses of gastropod shells from Kpone Beach indicate that Cerithium and Nerita are more sensitive to accumulating Ni and As than Crassostrea. The values of trace metals from the Crassostrea shells of the Densu Estuary and Anyanui Creek are comparatively low. Considering the role of Crassostrea shells as bioindicators for heavy metals pollution, the less polluted localities are Anyanui Creek and Densu Estuary, whereas Pra Estuary and Kpone Beach are affected by anthropic activities such as mining, urban effluents, and industry.

Oysters are notably susceptible to accumulating substantial amounts of cadmium (Cd), leading to food safety concerns, yet the molecular mechanisms are not fully understood. Previous study identified a significant correlation between polymorphisms in the Zip1 and Zip3 of Fujian oyster (Crassostrea angulata) and the levels of Cd accumulation. Zip1 and Zip3, belonging to the Zip family, are primarily responsible for the cellular transport of zinc and other metal ions. To delve deeper into their roles in Cd transport within oysters, this research cloned and analyzed the sequences of Fujian oyster Zip1 and Zip3. The open reading frames for Zip1 and Zip3 were 960 and 978 bp, respectively, encoding proteins of 319 and 325 amino acids. Following Cd exposure, the expression levels of these genes in the mantle, gills, and other tissues initially increased significantly before decreasing, demonstrating a dose-dependent response. The lowest expression levels were observed in the adductor muscle, with Zip1 reaching its highest expression in the mantle and Zip3 in the visceral mass. In vitro RNAi experiments revealed that silencing Zip1 led to a 1.26-fold decrease in new 113Cd accumulation in the mantle, whereas silencing Zip3 resulted in a 1.29-fold increase. Immunohistochemistry showed weak expression of Zip1 and Zip3 proteins in the connective tissue of the mantle. Simple Summary: As a filter-feeding and sessile invertebrate living in estuaries and intertidal areas, Crassostrea gigas must cope with and adapt to a dynamic and changeable environment. Vibrio alginolyticus is a Gram-negative bacterium that is widespread in oceans and estuaries, and is one of the main Vibrio species that cause oyster disease. In this study, we used transcriptome sequencing to help us better understand how the giant oyster adapts to pathogen-rich environments. By focusing on the gills, which play a crucial role in the immune response, we aimed to shed light on the molecular processes underlying the interaction between the oyster and the pathogen. Over the years, oysters have faced recurring mass mortality issues during the summer breeding season, with Vibrio infection emerging as a significant contributing factor. Tubules of gill filaments were confirmed to be in the hematopoietic position in Crassostrea gigas, which produce hemocytes with immune defense capabilities. Additionally, the epithelial cells of oyster gills produce immune effectors to defend against pathogens. In light of this, we performed a transcriptome analysis of gill tissues obtained from C. gigas infected with Vibrio alginolyticus for 12 h and 48 h. Through this analysis, we identified 1024 differentially expressed genes (DEGs) at 12 h post-injection and 1079 DEGs at 48 h post-injection. Enrichment analysis of these DEGs revealed a significant association with immune-related Gene Ontology (GO) terms and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. To further investigate the immune response, we constructed a protein–protein interaction (PPI) network using the DEGs enriched in immune-associated KEGG pathways. This network provided insights into the interactions and relationships among these genes, shedding light on the underlying mechanisms of the innate immune defense mechanism in oyster gills. To ensure the accuracy of our findings, we validated 16 key genes using quantitative RT-PCR. Overall, this study represents the first exploration of the innate immune defense mechanism in oyster gills using a PPI network approach. This study explored the potential of three local microalgal isolates as feed for oyster larvae in laboratory‐rearing experiments, towards the optimisation of seed production of Crassostrea tulipa to support its large‐scale farming along the West African coast. Three species of local microalgae—Rhodomonas sp., Nannochloropsis sp. and Pseudanabaena sp.—were isolated from waters off the coast of Ghana, West Africa, following a serial dilution technique. The growth performance of the isolates was assessed in the laboratory through daily estimation of cell density until the stationary phase was observed. Characterisation of the microalgal isolates was carried out by estimation of their biovolume, carbon content and energy content. Biovolumes of the microalgae were calculated from the estimated equivalent spherical diameters using proposed geometric shapes and formulae. Carbon weight and carbon energy content were subsequently calculated using derived conversions. The three microalgal isolates showed potential for large‐scale cultivation in the laboratory with marked differences in daily increases in cell densities. Nannochloropsis sp. and Rhodomonas sp. recorded the highest and the lowest peak densities of 2.4 × 105 and 1.5 × 105 cell mL−1, respectively, from an initial inoculating cell density of 1.05 × 105 cell mL−1. The estimated mean biovolumes of Rhodomonas sp., Nannochloropsis sp. and Pseudanabaena sp. were 238.9, 8.182 and 42.42 µm3, respectively, and the corresponding derived carbon energy contents were 1.7 × 10−6, 7.13 × 10−8 and 1.05 × 10−7 J, respectively. Results from a laboratory rearing experiment indicated that the individual microalgal isolates supported the growth and survival of oyster larvae at different scales, but a mixed diet of the three promoted superior growth and survival of C. tulipa larvae. The three local microalgal isolates‐ Rhodomonas sp., Nannochloropsis sp., and Pseudanabaena sp.‐ were well adapted to laboratory culture conditions, and the observed differences in growth and survival of the oyster larvae fed on these algal diets could be due to the differences in diet properties and biochemical compositions of the different species. A combination of the three algal diets, however, provided complementaery nutrients for the optimal growth and survival of C. tulipa larvae. The outcome of this study shows that local microalgal isolates have the potential to support hatchery rearing of C. tulipa, which is essential for the development of commercial mangrove oyster aquaculture in West Africa. Settlement is a critical period in the life cycle of marine invertebrates with a planktonic larval stage. For reef-building invertebrates such as oysters and corals, settlement rates are predictive for long-term reef survival. Increasing evidence suggests that marine invertebrates use information from ocean soundscapes to inform settlement decisions. Sessile marine invertebrates with a planktonic stage are particularly reliant on environmental cues to direct them to ideal habitats. As gregarious settlers, oysters prefer to settle amongst members of the same species. It has been hypothesized that oyster larvae from species Crassostrea virginica and Ostrea angasi use distinct conspecific oyster reef sounds to navigate to ideal habitats. In controlled laboratory experiments we exposed Pacific Oyster Magallana gigas larvae to anthropogenic sounds from conspecific oyster reefs, vessels, combined reef-vessel sounds as well as off-reef and no speaker controls. Our findings show that sounds recorded at conspecific reefs induced higher percentages of settlement by about 1.44 and 1.64 times compared to off-reef and no speaker controls, respectively. In contrast, the settlement increase compared to the no speaker control was non-significant for vessel sounds (1.21 fold), combined reef-vessel sounds (1.30 fold), and off-reef sounds (1.18 fold). Coastal biogenic structures, formed by ecosystem engineering species, often feature rough surfaces characterized by intricate topographies and highly threedimensional reliefs. Their surfaces are shaped by waves and tidal currents and reciprocally influence the ambient hydrodynamics, reflecting an equilibrium. Despite their significance, the impact of these surfaces on the ambient hydrodynamics remains underexplored due to limited knowledge of accurately replicating their complex topographies in experimental setups. The recent advent of advanced digital manufacturing presents an efficient means to manufacture highly complex, three-dimensional surrogate models for experimental modeling. This work explores the accurate replication of rough coastal biogenic structures for experimental modeling on the examples of an oyster reef and a mussel bed, utilizing a flexible design methodology and, for the first time, particle bed 3D printing with Selective Cement Activation (SCA) as a fabrication and manufacturing method. A workflow is proposed, which includes an iterative surrogate model development based on in-situ topographical features, requirements of the experimental setup, and parameters of the particle bed 3D printer with SCA. The results demonstrate the effectiveness of the methodology in achieving highly accurate surrogate surfaces of complex coastal biogenic structures by validation against a set of topographical features relevant to hydraulic roughness. Particle bed 3D printing with SCA proved to be a suitable method to manufacture complex surrogate surfaces for experimental modeling, offering advantages such as independence of production time from surface complexity. However, challenges persist in achieving exact comparability between the manufactured surrogate surface and the real coastal biogenic structures, particularly for surfaces with very high complexity. Nonetheless, the manufactured generic surrogate surfaces enable detailed investigations into the influence of complex coastal biogenic structures on the ambient hydrodynamics, thereby enhancing the understanding of the processes governing wave energy dissipation attenuation, turbulence production, and vertical mixing - critical for efficient application as a nature-based solution on coastal protection or restoration efforts. The increase of the frequency and severity of marine diseases affecting farmed marine mollusks are currently threatening the sustainability of this aquaculture sector, with few available prophylactic or therapeutic solutions. Recent advances have shown that the innate immune system of invertebrates can develop memory mechanisms allowing for efficient protection against pathogens. These properties have been called innate immune memory, immune priming or trained immunity. Previous results demonstrated the possibility to elicit antiviral immune priming to protect Pacific oysters against the ostreid herpes virus 1 (OsHV-1), currently plaguing M. gigas production worldwide. Here, we demonstrate that UV-inactivated OsHV-1 is also a potent elicitor of immune priming. Previous exposure to the inactivated virus was able to efficiently protect oysters against OsHV-1, significantly increasing oyster survival. We demonstrate that this exposure blocked viral replication and was able to induce antiviral gene expression potentially involved in controlling the infection. Finally, we show that this phenomenon can persist for at least 3 months, suggesting the induction of innate immune memory mechanisms. This study unravels new ways to train the Pacific oyster immune system that could represent an opportunity to develop new prophylactic strategies to improve health and to sustain the development of marine mollusk aquaculture. Crassostrea is the most farmed oyster genus worldwide and has significant economic and social impacts with environmental benefits. Hatchery oyster seed production is a highly costly phase, and a fluidized nursery system can help reduce this cost and reduce seed production time. The present study evaluated the survival and growth of two oyster species (Crassostrea gasar and Crassostrea gigas) in a fluidized bed bottle nursery system. With C. gasar, two experiments were performed; one tested three stocking densities and the other three bialgae diets. With C. gigas, one experiment with a bialgae and monoalgae in an initial bottle occupation of 8.8% produced more seeds per bottle, but an initial bottle occupation of 2.2% produced bigger seeds. Also, the experiment with C. gasar and with C. gigas tested diets did not affect seed survival, but the diets with bialgae I. galbana and N. oculate promoted more seed growth. Caspase-8, a member of the caspase family, is an initiating caspase and plays a crucial role in apoptosis. In this study, the full-length cDNA of caspase8-like (CASP8-like) was isolated from Crassostrea hongkongensis (C. hongkongensis) by RACE-PCR. ChCASP8-like contained a 1599-bp open reading frame (ORF) encoding 533 amino acids with two conserved death effector domains (DEDs) and a cysteine aspartase cysteine structural domain (CASc). Amino acid sequence comparison showed that ChCASP8-like shared the highest identity (85.4%) with CASP8-like of C. angulata. The tissue expression profile showed that ChCASP8-like was constitutively expressed in gills, hepatopancreas, mantle, adductor muscle, hemocytes and gonads, and was significantly upregulated in hemocytes, hepatopancreas and gills under hyper-salinity stress. The apoptosis-related genes, including ATR, CHK1, BCL-XL, CASP8-like, CASP9 and CASP3, were significantly activated by hyper-salinity stress, but were remarkably inhibited by ChCASP8-like silencing. The caspase 8 activity was increased by 1.7-fold after hyper-salinity stress, and was inhibited by 9.4% by ChCASP8-like silencing. Moreover, ChCASP8-like silencing clearly alleviated the apoptosis resulting from hyper-salinity stress. In the Hardy–Weinberg equilibrium test, for the seven populations and ten microsatellites, in 10 of the 70 groups, the segregation distortion was significant. These results suggest that a stocking density of 20 oysters/substrate can promote the shell height, shell width and total weight of "South China No. 1". Fangchenggang is a suitable place to cultivate the "South China No. 1" breed according to the total weight and sum of all algal genus abundances. This study aims to explore the effects of stocking density on the growth performance of "South China No. 1", compared their growth performance and genetic diversity to C. hongkongensis, and found the best place breeding site for "South China No. 1" in Beibu Gulf. The results showed that stocking a density of 20 oysters/substrate can significantly increase the shell height, shell width, total weight, survival rate, daily shell height gain and daily body mass gain. It was found that the shell height and total weight of "South China No. 1" cultured in Fangchenggang were significantly higher than that of those in Beihai and Qinzhou from September 2018 to November 2018. Similarly, the shell width of oysters in Fangchenggang and Qinzhou was also significantly higher in September 2018, and the interaction between site and stocking density had significant effects on the shell width in March 2018 and November 2018. In addition, the shell height and shell width of "South China No. 1" were significantly higher than that of C. hongkongensis in all three sites. At all three sites, the phytoplankton community structure was mostly dominated by Bacillariophyta. Paralytic shellfish poisoning is an important concern for mollusk fisheries, aquaculture, and public health. In Galicia, NW Iberian Peninsula, such toxicity has been monitored for a long time using mouse bioassay. Therefore, little information exists about the precise toxin analogues and their possible transformations in diverse mollusk species and environments. After the change in the European PSP reference method, a refinement of the Lawrence method was developed, achieving a 75% reduction in chromatogram run time. Since the beginning of 2021, when this refinement Lawrence method was accredited under the norm UNE-EN ISO/IEC 17025, it has been used in the area to determine the toxin profiles and to estimate PSP toxicity in more than 4500 samples. In this study, we have summarized three years of monitoring results, including interspecific, seasonal, and geographical variability of PSP toxicity and toxin profile. PSP was detected in more than half of the samples analyzed (55%), but only 4.4% of the determinations were above the EU regulatory limit. GTX1,4 was the pair of STX analogs that produced the highest toxicities, but GTX2,3 was found in most samples, mainly due to the reduction of GTX1,4 but also by the higher sensitivity of the method for this pair of analogs. STX seems to be mainly a product of biotransformation from GTX2,3. The studied species (twelve bivalves and one gastropod) accumulated and transformed PSP toxins to a different extent, with most of them showing similar profiles except for Spisula solida and Haliotis tuberculata. Two seasonal peaks of toxicity were found: one in spring-early summer and another in autumn, with slightly different toxin profiles during outbreaks in relation to the toxicity during valleys. In general, both the total toxicity and toxin profiles of the southernmost locations were different from those in the northern part of the Atlantic coast and the Cantabrian Sea, but this general pattern is modified by the PSP history of some specific locations. Microplastics (MPs) pollution has wreaked havoc on biodiversity and food safety globally. The false ingestion of MPs causes harmful effects on organisms, resulting in a decline in biodiversity. The present review comprehended the current knowledge of MP contamination in Crustacea and Mollusca from 75 peer‐reviewed articles published in Asia between 2015 and 2023. A total of 79 species (27 Crustacea and 52 Mollusca) have been recorded to be contaminated with MPs. Out of the total 27 species of Crustacea, Metopograpsus quadridentatus (327.56 MPs/individual) and Balanus albicostatus (0.42 MPs/individual) showed the highest and lowest contamination, respectively. Out of the total 52 species of Mollusca, Dolabella auricularia (2325 MPs/individual) and Crassostrea gigas and Mytilus edulis (0.2 MPs/individual) showed the highest and lowest contamination, respectively. In terms of country‐wise MP contamination, China has the highest number of contaminated species in both phylums among Asia. Findings of pollution indices revealed a very high risk of MP contamination in all the countries. Fiber was reported predominantly in both groups. Blue and black‐colored MPs having <500 μm and <500 μm–1 mm size were found dominantly in Crustacea and Mollusca, respectively. Polypropylene was recorded as the dominant plastic polymer in both Crustacea and Mollusca. In essence, this review has provided a comprehensive insight into MP concentration in Crustacea and Mollusca of Asia, highlighting variations among species and geographic locations. This understanding is crucial for tackling urgent environmental challenges, safeguarding human health, and promoting global sustainability initiatives amid the escalating issue of plastic pollution. Crassostrea ariakensis (Fujita, 1913) is one of the most important economic and ecological oysters that is naturally distributed along the coast of Asia, separated by the Yangtze River estuary. They are usually compared as different populations, while there is no consensus on whether C. ariakensis in northern and southern areas should be considered as two species or subspecies. Here, we analyzed morphological characteristics, COI, 16s rRNA, mitogenome sequences, and species delimitation analysis (ASAP and PTP) to resolve the intraspecific taxonomic status of the C. ariakensis. Phylogenetic and ASAP analysis highlight that C. ariakensis was divided into N-type and S-type. PTP was unable to differentiate between the two types of C. ariakensis. The divergence time of N-type and S-type C. ariakinsis is estimated to be 1.6 Mya, using the relaxed uncorrelated lognormal clock method. Additionally, significant morphological differences exist between the two groups in terms of the adductor muscle scar color. Despite these differences, the COI (0.6%) and 16S rRNA (0.6%) genetic distance differences between N-type and S-type C. ariakensis has not yet reached the interspecific level. These results suggest that N-type and S-type C. ariakensis should be treated as different subspecies and renamed as C. ariakensis ariakensis subsp. nov and C. ariakensis meridioyangtzensis subsp. nov. Shell‐closing strength (SCS) is commonly used to assess oyster vigor and quality as an important physiological stress indicator of the oyster organism. In this study, a quality decision support system based on flexible wireless sensor network and web services was designed and developed to explore the application of SCS in the rapid assessment of vitality and quality. Based on physiological and environmental information collected by flexible wireless sensor networks, this study proposes a novel quantitative vitality assessment method to identify oyster life decline cycles and processes. Meanwhile, TVB‐N, TVC, and pH were selected as quality indicators, and a back‐propagation artificial neural network (BP‐ANN) was used to predict and evaluate oyster quality indicators. The results showed that the root mean square errors (RMSE) for TVB‐N, TVC, and pH were 0.4624, 0.8827, and 0.1941; the coefficients of determination (R2) were 0.8919, 0.8578, and 0.6249. Therefore, the rapid assessment of oyster vitality and quality based on a flexible wireless sensor network is a reliable and effective means. Practical applications: This study aims to explore the application of live oyster SCS for rapid quality evaluation. In this article, a quality decision support system based on a flexible wireless sensor network (WSN) and web services is developed to improve the quality and health of live oysters in the supply chain. The use of the multi‐sensor system enables the monitoring and collection of environmental and physiological signals of live oysters throughout the supply chain process. The evaluation of the quality of live oysters using physiological and environmental information collected by flexible wireless sensor networks is a reliable and efficient method. Squid (Loligo vulgaris) is commonly prone to spoilage, leading to a short shelf-life. High-pressure processing (HPP) can play a role in maintaining the quality and freshness of squid. Along with HPP, food preservatives from natural sources such as mint extract (ME), which are effective, safe, available, and cost-effective, are required. The present study aimed to investigate the combined effect of ME and HPP on the quality of refrigerated squid mantle cuts (SMC) over a period of 15 days. The time-kill profiles of ME and planktonic cell inactivation by HPP were assessed. ME (400 mg/L) inhibited bacterial growth, while planktonic cells treated with HPP (400 MPa) exhibited a reduction at 5 min. Physicochemical and microbial qualities of SMC treated with ME (0, 200, 400 mg/L) followed by HPP (0.1, 200, 400 MPa) for 5 min were monitored during refrigerated storage. Samples treated with ME (400 mg/L) and HPP (400 MPa) exhibited lower weight loss, cooking loss, pH changes, volatile base content, microbial counts, and higher textural properties than other samples. Based on next-generation sequencing results, Brochothrix campestris from family Listeriaceae was the predominant spoilage bacteria in treated sample after 12 days of storage. The Pacific oyster Crassostrea gigas is one of the most commercially important aquaculture species worldwide. The occurrence of inbreeding is common in the breeding practice of oysters due to limited population size, successive high selection pressure, and genetic drift. To determine the potential effects of inbreeding on the potential of genetic improvement for fast growth of oysters, genetic parameters of growth traits in the 7th to 9th generation of the orange-shell strain (a typical inbred population) were estimated. At the grow-out stage, the average genetic gain (GG), selection response (SR), and realized heritability ( h R 2 ) for shell height were 6.06–7.06%, 0.43–0.50, and 0.24–0.27, respectively. Meanwhile, the average GG, SR, and h R 2 for body weight were 6.03–7.36%, 0.38–0.41, and 0.21–0.22. There were no significant differences in the corresponding genetic parameters among the three generations (P > 0.05). Genetic parameters were estimated for an unselected population of C. gigas that served as a control group and were significantly higher than those of the orange-shell strain (P < 0.05). The results of this study suggest that inbreeding reduced the potential for genetic improvement of the rapid growth of oysters. However, some easy-to-operate and low-cost measures including the large size of broodstock, balanced sex ratio, and artificial spawning have been applied to the mass selection of the orange-shell strain, which prevents the situation from getting worse. These results will contribute to a better understanding of the negative effects of inbreeding on C. gigas and the management of inbreeding in shellfish breeding programs. [ABSTRACT FROM AUTHOR]

Crassostrea gasar is a bivalve mollusk of economic interest in the Amazon estuary, because it displays good zootechnical performance and nutritional value. However, there is still a gap in the literature regarding studies on basic biology, especially spermatogenesis at the ultrastructural level. This study aimed to describe for the first time the male gonad development. Gonad fragments were analyzed using light microscopy and transmission and scanning electron microscopy. The ultrastructural analysis of the gonadal tubule showed two distinct cell populations: support cells and the cells of the spermatogenic lineage. Regarding the ultrastructural aspects, support cells were characterized by an irregular shape, sparse electron-dense cytoplasm, and a large nucleus with condensed chromatin; and two types of spermatogonia were seen (A and B); spermatocytes I where it was possible to characterize four stages of prophase I, namely leptotene, zygotene, pachytene, diplotene; spermatocytes II showing a reduction in cytoplasmic and condensed chromatin. Spermatids were classified into three stages of development: early, middle, and late. Mature spermatozoa are of the primitive type and have a cone-shaped head with a discoidal acrosome, a midpiece formed by four mitochondria, and a long flagellum. This information can help establish phylogenetic relationships between species of the genus Crassostrea. In addition, these findings can support conservation biology by developing and implementing biotechnological approaches, such as cryopreservation, and contributing to commercial-scale cultivation in the Amazon region. The mangrove oyster Crassostrea rhizophorae is identified as a potentially valuable species for tropical aquaculture, however, information on the physiological mechanisms of reproduction under laboratory conditions for this species is limited. This study investigated the effects of salinity at different concentrations (15, 20, 25, 30, 35, and 40 g/L) on the induction of germinal vesicle breakdown (GVBD) of oocytes obtained through stripping, the release of polar bodies (PB1 and PB2), and the larval development of the mangrove oyster. The results revealed a relationship between salinity and the percentage of GVBD, with the most effective range being 30–40 g/L within the hydration time frame between 70 and 120 min. The release of 50 % of PB1 was detected within this salinity range, while for the release of 50 % of PB2, the saline treatments of 35 and 40 g/L showed the best results. Overall, the salinity range of 30–40 g/L is suggested as the most suitable of polyploidy induction methodologies through the retention of PB1 or PB2. Regarding larval hatching, while salinities between 25 and 40 g/L presented similar percentages, at 15 g/L no hatching was observed. This study demonstrated that salinity is a key factor in early pre- and post-fertilization stages for the successful reproduction of mangrove oyster in hatcheries and that the percentages of oocyte maturation and artificial fertilization can be optimized by adjusting salinity. • Salinity significantly influenced the timing of germinal vesicle breakdown (GVBD). • Salinity influences the process of polar bodies (PB1 and PB2) release synchronization. • Hydrated oocytes at 35 and 40 g L exhibited higher percentages of GVBD, PB1 and PB2. • Higher salinities are crucial for the early pre- and post-fertilization stages, but not at later stages (larval hatching). This study aimed to compare the growth and survival of Crassostrea gigas spat at four sites (Sambaqui (SBQ), Paraíso das Ostras (PAR), Atlâtico Sul (ATL), and Cia das Ostras (CDO)) in Brazil. Batches of spat were reared in the summer and winter of 2018 and the summer of 2019 for two month periods. SBQ site had higher total particulate matter and particulate inorganic matter values than those observed at the other sites. The salinity at the PAR site was higher than that found at the other sites, but the temperature was lower. No differences in survival were observed among the sites in the summer or winter of 2018; however, in the summer of 2019, there were significant differences. Shell height was higher during warmer seasons than during winter. In conclusion, the SBQ and ATL sites were found to have better conditions for spat cultivation than the PAR and CDO sites. Contaminants compounds like Heavy Metals and Polycyclic Aromatic Hydrocarbons (PAH's) are primarily sourced from industrial and urban areas. As pollutants, these chemical elements and compounds are potentially toxic and present the capacity to negatively impact the ecosystem, especially the most sensitive ones such as estuaries. Bivalves' species have been successfully utilized as monitoring tools to evaluate pollutants bioaccessibility around the globe. The present study assessed heavy metals (Zn, Cd, Ni, Fe, Cr, Cu and Pb), As and PAH's levels in two mollusk species (Perna perna and Crassostrea rhizophorae) from Santos Estuarine System, in São Paulo State, in order to compare the effectiveness of each species in the biomonitoring process. According to the obtained data, Perna perna mussels may be better indicators of environmental pollution than Crassostrea rhizophorae oysters for some metals (Iron and Nickel) and hydrocarbons. The glycoprotein-N-acetylgalactosamine β1,3-galactosyltransferase, known as T-synthase (EC 2.4.1.122), plays a crucial role in the synthesis of the T-antigen, which is the core 1 O-glycan structure. This enzyme transfers galactose from UDP-Gal to GalNAc-Ser/Thr. The T-antigen has significant functions in animal development, immune response, and recognition processes. Molluscs are a successful group of animals that inhabit various environments, such as freshwater, marine, and terrestrial habitats. They serve important roles in ecosystems as filter feeders and decomposers but can also be pests in agriculture and intermediate hosts for human and cattle parasites. The identification and characterization of novel carbohydrate active enzymes, such as T-synthase, can aid in the understanding of molluscan glycosylation abilities and their adaptation and survival abilities. Here, the T-synthase enzymes from the snail Pomacea canaliculata and the oyster Crassostrea gigas are identified, cloned, expressed, and characterized, with a focus on structural elucidation. The synthesized enzymes display core 1 β1,3-galactosyltransferase activity using pNP-α-GalNAc as substrate and exhibit similar biochemical parameters as previously characterised T-synthases from other species. While the enzyme from C. gigas shares the same structural parameters with the other enzymes characterised so far, the T-synthase from P. canaliculata lacks the consensus sequence CCSD, which was previously considered indispensable.
