Abstract

International audience; Bioregenerative life support systems (BLSS) are the foundation stone to self-sustainable manned space missions. The MELiSSA is a BLSS concept that has evolved through a mechanistic engineering approach designed to acquire both theoretical and technical knowledge on each subsystem independently and, therefore, produces the necessary knowledge and experience needed to co-integrate all the subsystems together with a high level of control. One of the subsystems is a photobioreactor colonized by an axenic culture of the cyanobacterium Limnospira indica PCC8005 for revitalizing the air for the crew. This subsystem was extensively studied, and a mass balanced mechanistic model was developed to describe, predict, and control the photobioreactor. The model was based on a light transfer limitation model coupled to a kinetic model for the cyanobacteria growth through a Linear Thermodynamics of Irreversible Processes (LTIP) approach, including substrate limitation. The model was integrated into several hydrodynamic models adapted to several photobioreactors design and experiments, from a 100 L airlift pilot scale ground photobioreactor to a 50 ml membrane photobioreactor for ISS flight. Through this article we detail the principles of this mechanistic model and their application to different photobioreactor scales for predictive and descriptive simulations.

15. Limnospira indicaPCC8005 growth in photobioreactor: model and simulation of the ISS and ground experiments

Abstract

The Arthrospira-B experiment is the first experiment in space ever allowing the online measurements of both oxygen production rate and growth rate of Limnospira indica PCC8005in batch photobioreactors running on-board ISS. Four bioreactors were integrated in the ISS Biolab facility. Each reactor was composed of two chambers (gas and liquid) separated by a PTFE membrane and was run in batch conditions. Oxygen production was monitored by online measurement of the total pressure increase in the gas chamber. The experiments are composed of several successive batch cultures for each reactor, performed in parallel on ISS and on ground. In this work, a model for the growth of the cyanobacterium Limnospira indicaPCC8005 (also known as Arthrospiraor spirulina) in these space membrane photobioreactors was proposed and the simulation results obtained are compared to the experimental results gathered in space and on ground.

Abstract

National audience; Ce travail caractérise un dispositif expérimental conçu pour étudier les effets des ondes électro magnétiques sur des cellules eucaryotes (Saccharomyces cerevisiae) cultivées en bioréacteur tout en permettant les régulations de température, de pH, de pO2, d’agitation et d’aération influençant la croissance des cellules. Les simulations numériques réalisées montrent que l’atténuation des ondes électromagnétiques au sein du milieu de culture est importante et que les modifications des propriétés diélectriques du milieu liées à la fréquence des ondes incidentes et aux variations de composition du milieu au cours de la culture peuvent avoir une influence sur le niveau d’exposition des cellules aux champs électromagnétiques. Les essais réalisés en environnements anéchoïque et réverbérant ne mettent pas en évidence d’effet sur le taux de croissance et les rendements de production des différents métabolites de la levure soumise aux champs électromagnétiques.

Abstract

For long-term manned missions, higher plants cultivation is mandatory for food supply. MELiSSA project (Micro-Ecological Life Support System Alternative) that is developed by ESA includes a higher plants compartment. In order to satisfy the general specification of any MELiSSA compartment, plants’ behavior must be understood and predicted using a knowledge-based and multi-parameter model. This paper concerns the presentation of the building blocks of a structured model, including the main plant processes: plant growth and development, plant architecture and morphology, plant physiology and metabolism for the principal organs (roots, stems, leaves, storage organs e.g. fruits, seeds, tubers…), depending on physical rate limiting processes: light interception, gas transfer, water and nutrients uptake, sap diffusion. Each of these different submodels is described. Light interception depends on the incident light intensity, properties of leaves and the canopy architecture. It permits calculation of the available light energy in order to perform photosynthesis. Gas exchange depends on the atmosphere composition, the stomata processes, the diffusion and dissolution rates, the leaf cells concentration of each exchanged gas and rate of use or production. Sap diffusion depends on the stem morphology, and the metabolic and physical processes of sap movement. Root uptake depends on the nutrient solution concentration and the metabolic and physical processes of nutrients and water absorption. The plant metabolism is used to predict the final composition of the plant, the growth rate and the available energy for the non-photosynthetic organs. Mass and energy balances give the structure of the model of the environmental variables, e.g. atmosphere composition, incident light, temperature, humidity, nutrient solution composition. The morphology and architecture model gives the plant dimensions e.g. stem length, leaf area or root surface, and the development model predicts evolution with time and environment. The general model will permit to predict the CO 2 and nutrient solution consumption, and oxygen, clean water and food production from the plant depending on the plant compartment conditions, using mass balance analysis of the system.
