2. Disturbance history alters the development of the HPA axis in altricial nestling birds

Abstract

Glucocorticoid (GC) hormones regulate the vertebrate stress response and are secreted by the hypothalamic–pituitary–adrenal (HPA) axis. Acute elevation of GCs is thought be adaptive because it promotes physiological and behavioural changes that allow animals to cope with disturbances. In contrast, chronic elevation of GCs is associated with reduced body condition, immune function, reproductive success, and survival. In adult birds, the effects of chronic stress have been well documented, including human-related disturbances. In contrast, the effects of chronic stress on nestlings have rarely been addressed. This is of interest, as many ecological or monitoring studies of wild birds require nestlings to be regularly handled. However, the consequences of repeated handling of nestlings on HPA axis function and body mass in wild birds remain poorly quantified. We examined whether daily exposure to handling stress increased corticosterone (the dominant avian glucocorticoid) secretion and reduced pre-fledging body mass relative to undisturbed control nestlings of the noisy miner (Manorina melanocephala), a native Australian passerine bird. Daily handling resulted in an elevated baseline and attenuated stress-induced corticosterone levels in disturbed 14-day nestlings, in comparison with control nestlings handled for the first time. Despite this, disturbed and control nestlings fledged at a similar body mass. Baseline and stress-induced corticosterone increased with nestling age but remained independent of nestling sex and hatching order. Our findings are some of the first to suggest that chronic handling stress causes physiological alterations to the development of the HPA axis in nestling birds, and our data suggest that researchers should minimise or account for handling stress in their experiments. These data also raise the possibility that other chronic stressors may have long term physiological consequences for the development of the HPA axis in nestling birds.

6. On K3-fibred LARGE Volume Scenario with de Sitter vacua from anti-D3-branes

Abstract

In the context of type IIB superstring compactifications on K3-fibred (weak) Swiss-cheese Calabi Yau (CY) orientifolds, we consider the realisation of de Sitter vacua obtained through the introduction of an anti-D3-brane at the tip of a highly warped throat of Klebanov-Strassler type. Aiming to have a concrete global realisation, we perform a systematic search for the CY threefolds with $2 < h^{1,1} < 5$ arising from the Kreuzer-Skarke database, which satisfy the minimal requirements of being K3-fibred and suitable for moduli stabilisation within the LARGE Volume Scenario (LVS). In this context, after scanning the set of K3-fibred CY threefolds with a so-called diagonal del-Pezzo divisor needed for LVS, we realise that one of the main challenging requirements for having anti-D3-brane uplifting is to find a suitable orientifold involution which can simultaneously result in a sufficient large $D3$ tadpole charge along with the presence of suitable $O3$-planes. In our detailed analysis (limited to) using the CY threefolds with small $h^{1,1}$, we observe that these topological requirements rule out most of the CY geometries leading to only few possibly suitable candidates for the purpose of anti-D3-brane uplifting. Subsequently, we present a global model using one such explicit K3-fibred CY threefold with $h^{1,1}=4$ in which all the moduli can be consistently stabilised in a de Sitter minimum of the scalar potential., Comment: v2: 40 pages, 1 figure, improved version to appear in JHEP; v1: 37 pages, 1 figure

7. A Database of Calabi-Yau Orientifolds and the Size of D3-Tadpoles

Abstract

The classification of 4D reflexive polytopes by Kreuzer and Skarke allows for a systematic construction of Calabi-Yau hypersurfaces as fine, regular, star triangulations (FRSTs). Until now, the vastness of this geometric landscape remains largely unexplored. In this paper, we construct Calabi-Yau orientifolds from holomorphic reflection involutions of such hypersurfaces with Hodge numbers $h^{1,1}\leq 12$. In particular, we compute orientifold configurations for all favourable FRSTs for $h^{1,1}\leq 7$, while randomly sampling triangulations for each pair of Hodge numbers up to $h^{1,1}=12$. We find explicit string compactifications on these orientifolded Calabi-Yaus for which the D3-charge contribution coming from O$p$-planes grows linearly with the number of complex structure and K\"ahler moduli. We further consider non-local D7-tadpole cancellation through Whitney branes. We argue that this leads to a significant enhancement of the total D3-tadpole as compared to conventional $\mathrm{SO}(8)$ stacks with $(4+4)$ D7-branes on top of O7-planes. In particular, before turning-on worldvolume fluxes, we find that the largest D3-tadpole in this class occurs for Calabi-Yau threefolds with $(h^{1,1}_{+},h^{1,2}_{-})=(11,491)$ with D3-brane charges $|Q_{\text{D3}}|=504$ for the local D7 case and $|Q_{\text{D3}}|=6,664$ for the non-local Whitney branes case, which appears to be large enough to cancel tadpoles and allow fluxes to stabilise all complex structure moduli. Abstract

There has been continuous progress in diabetes management over the last few decades, not least due to the widespread dissemination of continuous glucose monitoring (CGM) and automated insulin delivery systems. These technological advances have radically changed the daily lives of people living with diabetes, improving the quality of life of both children and their families. Despite this, hypoglycemia remains the primary side-effect of insulin therapy. Based on a systematic review of the available scientific evidence, this paper aims to provide evidence-based recommendations for recognizing, risk stratifying, treating, and managing patients with hypoglycemia. The objective of these recommendations is to unify the behavior of pediatric diabetologists with respect to the timely recognition and prevention of hypoglycemic episodes and the correct treatment of hypoglycemia, especially in patients using CGM or advanced hybrid closed-loop systems. All authors have long experience in the specialty and are members of the Italian Society of Pediatric Endocrinology and Diabetology. The goal of treating hypoglycemia is to raise blood glucose above 70 mg/dL (3.9 mmol/L) and to prevent further decreases. Oral glucose at a dose of 0.3 g/kg (0.1 g/kg for children using “smart pumps” or hybrid closed loop systems in automated mode) is the preferred treatment for the conscious individual with blood glucose

Abstract

We consider de Sitter vacua realised in concrete type IIB Calabi-Yau compactifications with an anti D3-brane at the tip of a warped throat of Klebanov-Strassler type. The K\"ahler moduli are stabilised together with the complex structure modulus of the warped throat. The volume is exponentially large as in the large volume scenario (LVS). We analyse the conditions on the parameters of the EFT such that they are in the region of validity of our approximations, there are no runaway problems and the vacua satisfy all consistency constraints, such as tadpole cancellation. We illustrate our results with an explicit Calabi-Yau orientifold with two K\"ahler moduli and one antibrane on top of an O3-plane in a warped throat, that has the goldstino as its only massless state. The moduli are stabilised with $g_s\sim 0.2$ and volume $\mathcal{V}\sim 10^4$ in string units, justifying the approximation used to derive the corresponding EFT. Abstract

We analyze the semiclassical and quantum behavior of the Bianchi IX Universe in the Polymer Quantum Mechanics framework, applied to the isotropic Misner variable, linked to the space volume of the model. The study is performed both in the Hamiltonian and field equations approaches, leading to the surprising result of a still singular and chaotic cosmology, whose Poincar\'e return map asymptotically overlaps the standard Belinskii-Khalatnikov-Lifshitz one. In the quantum sector, we reproduce the original analysis due to Misner, within the revised Polymer approach and we arrive to demonstrate that the quantum numbers of the point-Universe still remain constants of motion. This issue confirms the possibility to have quasi-classical states up to the initial singularity. The present study clearly demonstrates that the asymptotic behavior of the Bianchi IX Universe towards the singularity is not significantly affected by the Polymer reformulation of the spatial volume dynamics both on a pure quantum and a semiclassical level., Comment: 25 pages, 10 figures. v2: more discussions/clarification added

Abstract

Objective: Brain somatic variants in SLC35A2 were recently identified as a genetic marker for mild malformations of cortical development with oligodendroglial hyperplasia in epilepsy (MOGHE). The role of SLC35A2 in cortical development and the contributions of abnormal neurons and oligodendrocytes to seizure activity in MOGHE remain largely unexplored. Methods: Here, we generated a novel Slc35a2 floxed allele, which we used to develop two Slc35a2 conditional knockout mouse lines targeting (1) the Emx1 dorsal telencephalic lineage (excitatory neurons and glia) and (2) the Olig2 lineage (oligodendrocytes). We examined brain structure, behavior, and seizure activity. Results: Knockout of Slc35a2 from the Emx1 lineage, which targets both cortical neurons and oligodendrocytes, resulted in early lethality and caused abnormal cortical development, increased oligodendroglial cell density, early onset seizures, and developmental delays akin to what is observed in patients with MOGHE. By tracing neuronal development with 5‐Ethynyl‐2'‐deoxyuridine (EdU) birthdating experiments, we found that Slc35a2 deficiency disrupts corticogenesis by delaying radial migration of neurons from the subventricular zone. To discern the contributions of oligodendrocytes to these phenotypes, we knocked out Slc35a2 from the Olig2 lineage. This recapitulated the increased oligodendroglial cell density and resulted in abnormal electroencephalographic activity, but without a clear seizure phenotype, suggesting Slc35a2 deficiency in neurons is required for epileptogenesis. Significance: This study presents two novel Slc35a2 conditional knockout mouse models and characterizes the effects on brain development, behavior, and epileptogenesis. Together, these results demonstrate a direct causal role for SLC35A2 in MOGHE‐like phenotypes, including a critical role in neuronal migration during brain development, and identify neurons as key contributors to SLC35A2‐related epileptogenesis. Abstract

ObjectiveSomatic variants are a recognized cause of epilepsy-associated focal malformations of cortical development (MCD). We hypothesized that somatic variants may underlie a wider range of focal epilepsy, including nonlesional focal epilepsy (NLFE). Through genetic analysis of brain tissue, we evaluated the role of somatic variation in focal epilepsy with and without MCD.MethodsWe identified somatic variants through high-depth exome and ultra-high-depth candidate gene sequencing of DNA from epilepsy surgery specimens and leukocytes from 18 individuals with NLFE and 38 with focal MCD.ResultsWe observed somatic variants in 5 cases in SLC35A2, a gene associated with glycosylation defects and rare X-linked epileptic encephalopathies. Nonsynonymous variants in SLC35A2 were detected in resected brain, and absent from leukocytes, in 3 of 18 individuals (17%) with NLFE, 1 female and 2 males, with variant allele frequencies (VAFs) in brain-derived DNA of 2 to 14%. Pathologic evaluation revealed focal cortical dysplasia type Ia (FCD1a) in 2 of the 3 NLFE cases. In the MCD cohort, nonsynonymous variants in SCL35A2 were detected in the brains of 2 males with intractable epilepsy, developmental delay, and magnetic resonance imaging suggesting FCD, with VAFs of 19 to 53%; Evidence for FCD was not observed in either brain tissue specimen.InterpretationWe report somatic variants in SLC35A2 as an explanation for a substantial fraction of NLFE, a largely unexplained condition, as well as focal MCD, previously shown to result from somatic mutation but until now only in PI3K-AKT-mTOR pathway genes.
