Si bien la investigación en Ciencias Sociales conlleva ciertas dificultades propias del mismo proceso de investigación, como la recopilación de entrevistas o el trabajo de campo, también hay dificultades dentro de la misma academia. Existen ciertas barreras para la plena participación de las mujeres tanto en la misma investigación como en puestos de dirección y como lideresas, lo cual a veces se supera con el reconocimiento a la trayectoria de las investigadoras. Al respecto, hay notables casos. Sin embargo, en la misma academia, a las mujeres todavía se les considera a través de perspectivas distintas con relación a los hombres: desde la forma de obtener su plaza o adscripción, sobre su desenvolvimiento en la docencia, hasta la crítica hacia sus ascensos o sus distintas producciones, no sólo por parte de los compañeros, sino también de las compañeras. El covid-19, en todo el tiempo de pandemia, ha exacerbado muchas de estas dificultades, sin mencionar lo que sucede en la vida de la investigadora en el ámbito privado. Por ello, en el artículo se presentan las múltiples dificultades a las que se enfrentan las mujeres investigadoras, pero también se proponen formas para que logren una mayor visibilidad e incorporación dentro de la investigación, tal como el acompañamiento a jóvenes investigadoras, para así lograr una mayor cantidad, pero también calidad, en torno a la investigación hecha por mujeres

This article delves into the abuses stemming from the Nigerian state forces and their failure to protect Nigerian citizens as illustrated in Yejide Kilanko’s Daughters Who Walk this Path (2012). The novel narrates the struggles undergone by young Morayo as she is repeatedly abused as a child by an elder cousin, Bros T. Here, I seek to trace a parallelism between the instances of physical and affective violence against Morayo and the episodes of “intra-societal violence” (Hill 2012: 15) occurring in Nigeria from the 1980s to the mid-1990s, when the country’s sociopolitical sphere was marked by the social chaos resulting from armed robberies, military coups, rigged elections, and instances of police brutality towards women. I shall analyze such episodes as instances of ‘forced intimacy’ within the public and private spheres, which translates into the impositions of negative forms of affect upon personal and collective development. In this context, the physical and psychological abuses suffered by Morayo will be presented as shaping what Ahmed refers to as one’s “biographies of violence” (2017: 23). My ultimate aim is to trace Morayo’s development of what I will describe as ‘affective resilience’ against the affective forces of the state.

The bildungsroman has dominated the Nigerian literary landscape since the 1990s with novels such as Chimamanda N. Adichie’s Purple Hibiscus (2004), Unoma Azuah’s Sky-High Flames (2005), Chika Unigwe’s Night Dancer (2012), and Chinelo Okparanta’s Under the Udala Trees (2015). These coming-of-age stories place a special emphasis on the national evolution of Nigeria. Thus, by delving into such interrelation of the national and personal domains, I propose to theorize Under the Udala Trees as a “bildungsromance,” or novel of affective and romantic development. I use the term “bildungsromance” to describe the growth of Okparanta’s protagonist, Ijeoma, as intrinsically associated with the experiences gathered from her affective attachments both in the public and the private spheres. I will analyze Ijeoma’s sentimental relationships as she discovers and naturalizes her queer identity. For this, I will delve into the negative affects − in the form of fear, shame and guilt (Braidotti 2009, 50) − that influence Ijeoma’s experience as a result of her non-conforming to the dictums of Nigerian customary laws. Ironically, such negative affects will be introduced as tightly associated with “the promise of happiness” (Ahmed 2010, 14). In turn, I shall underline the role of Ijeoma’s intimate and romantic relationships in prompting positive forms of affect such as self-love and pride. These affective experiences allow Ijeoma to acquire a critical perspective towards Nigerian customary laws and their impact on identity formation and on what has been referred to as the “democratisation of the private sphere” (Giddens 1992, 184). Ultimately, I will hinge upon Ijeoma’s awareness of her own affective advancements as a key element in her bildungsromance.

Resumen: El Programa de Apoyo a los Estudiantes de Medicina de la Facultad de Medicina de la Universidad de Vic-Universidad Central de Cataluña (UVic-UCC) ofrece un acompañamiento a lo largo de todo el grado. Este programa tiene por objetivo identificar y atender las necesidades del estudiante que incluyen cualquier condición con impacto sobre su desarrollo académico. En cuanto se identifican dichas necesidades de apoyo se valora con un enfoque multidimensional (situación académica, psicoemocional y personal) con el fin de determinar un plan de actuación individualizado. Los servicios ofrecidos por el Programa de Apoyo a los Estudiantes de Medicina incluyen 3 abordajes: 1) optimización del rendimiento académico: tutorías grupales e individuales de orientación (metodología de estudio y técnica de examen); 2) atención psicológica individualizada, ya sea relacionada con dificultades psicológicas, de aprendizaje o bien con diversidades funcionales y 3) Mentorización: seguimiento y orientación en el itinerario curricular y en su desarrollo personal y profesional. El programa proporciona información que permite conocer más profundamente la realidad del estudiantado y posibilita la implementación de cambios docentes que tienen un impacto en la mejora del aprendizaje y también en la salud mental de los estudiantes. Abstract: The Support Program for Medical Students of the Faculty of Medicine, University of the Vic-Universitat Central de Catalunya (UVic-UCC) offers students support throughout the entire Degree. This program aims to identify and address the student's needs which include any condition that impacts their academic development. As soon as these support needs are identified, they are assessed with a multidimensional approach -academic, psycho-emotional and personal situation- in order to determine an individualized action plan. The services offered by the Support Program for Medical Students include 3 approaches: 1) Optimization of the academic performance: group and individual orientation tutorials (study strategies and exam techniques); 2) Individualized psychological care, whether related to psychological or learning difficulties, or functional diversity; 3) Mentoring: monitoring and guidance in the curricular itinerary and in their personal and professional development. Th program provides information that allows for a much deeper understanding of the reality of the students and makes it possible to implement teaching changes that have an impact on improving learning and, also, mental health of the students.

ABSTRACTConsidering the more frequent and longer drought events due to climate change, improving plant drought tolerance became a priority. The search for plant growth promoting rhizobacteria (PGPR) able to improve plant drought tolerance has been long addressed, but with inconsistent results. Here, we summarize the PGPR mechanisms that improve plant drought tolerance, identify the pitfalls in current PGPR isolation and selection routines, and discuss the key points to define new strategies to get optimal PGPR for plant drought tolerance. Drought and host genotype impact rhizo-communities, and host-mediated selection strategies may be used to obtain a drought-adapted rhizomicrobiome that can be a source for PGPR isolation. Alternatively, an integrated omics-level analysis can improve our knowledge on the mechanisms of rhizomicrobiome construction, and a targeted approach can be designed, which will be focused on key PGP traits. New strategies to build PGPR consortia for improvement of plant drought tolerance are also suggested.

Salinity is one of the main factors causing soil deterioration, making it unsuitable for agriculture. It is well documented that the application of halotolerant and halophilic plant growth-promoting bacteria (PGPR: plant growth-promoting rhizobacteria) with biological control activities as an inoculant of cultivated plants offers a biological alternative to the use of agrochemicals, particularly when subjected to salt stress. From this perspective, 70 bacterial strains were isolated from saline soils (sebkha) in arid and semi-arid areas of Eastern Algeria. Three isolates were selected based on their ability to produce bioactive molecules allowing them to promote plant growth, such as hydrolytic enzymes, indole acetic acid (auxin-phytohormone), HCN, NH3, etc. Two of these isolates belonged to the genus Serratia and the third was a halophilic Halomonas bacteria. These bacteria were identified based on their 16S rDNA sequences. Antagonism tests against phytopathogenic fungi were carried out. The identification of the antifungal molecules produced by these bacteria was determined using high-performance liquid chromatography. These bacteria can inhibit mycelial development against phytopathogenic fungi with rates reaching 80.67% against Botrytis cinerea, 76.22% against Aspergillus niger, and 66.67% against Fusarium culmorum for Serratia sp. The strain Halomonas sp. inhibited mycelial growth through the production of volatile substances of Aspergillus niger at 71.29%, Aspergillus flavus at 75.49%, and Penicillium glabrum at a rate of 72.22%. The identification of the antifungal molecules produced by these three bacteria using HPLC revealed that they were polyphenols, which makes these strains the first rhizobacteria capable of producing phenolic compounds. Finally, pot tests to determine the effectiveness of these strains in promoting wheat growth under salinity stress (125 mM, 150 mM, and 200 mM) was carried out. The results revealed that a consortium of two isolates (Serratia sp. and Halomonas sp.) performed best at 125 mM. However, at higher concentrations, it was the halophilic bacteria Halomonas sp. that gave the best result. In all cases, there was a significant improvement in the growth of wheat seedlings inoculated with the bacteria compared to non-inoculated controls.

The addition of biostimulants to nutrient solutions of hydroponically grown crops to speed up plant growth and improve plant yield and quality has been attracting more and more attention. This study investigated the effects of wood distillate (WD) addition to hydroponically grown lettuce (Lactuca sativa L.) plants. Two concentrations of WD, 0.2% and 0.5%, were added to the nutrient solution, and biometric (i.e., leaf fresh weight, root fresh weight, root length and root surface area), photosynthetic (i.e., chlorophyll a, chlorophyll b, and carotenoid content) and biochemical (i.e., electrolyte leakage, total polyphenols, total flavonoids, and total antioxidant power content) parameters were evaluated. The effects of WD were hormetic, as the 0.2% concentration stimulated biometric and biochemical parameters, while the 0.5% concentration inhibited plant growth. Based on these results, it can be suggested that the addition of 0.2% WD to the nutrient solution has a stimulating effect on the growth of lettuce plants, and could be a successful strategy to boost the yield of crops grown hydroponically.

Seed priming with synthetic chemicals may be harmful to the environment and human health. Their replacement with bio-based compounds may overcome these concerns. In this study, we assessed the effectiveness of wood distillate (WD) in enhancing in vitro germination of crop plants using basil, chickpea, and lettuce as case studies. Seeds of the three species were soaked for 24 h in 0.25% and 0.17% WD solutions and then left to germinate for 7 days at 20 °C in a dark germination chamber. Seed pre-treatment with 0.25% WD enhanced germination in all tested species, while 0.17% WD stimulated germination in lettuce and chickpea, but not in basil. For lettuce, 0.17% WD worked better than 0.25% WD. Radicle length of basil and chickpea increased following pre-treatment with 0.25% WD, while in lettuce, it increased after pre-treatment with 0.17% WD. Treating seeds with appropriate WD solutions is a potential strategy to improve germination of crop plants.

Lichens have long been considered as composite organisms composed of algae and/or cyanobacteria hosted by a fungus in a mutualistic relationship. Other organisms have been gradually discovered within the lichen thalli, such as multiple algal species, yeasts, or even viruses. Of pivotal relevance is the existence of the lichen microbiome, which is a community of microorganisms that can be found living together on the lichen surface. This community performs a growing number of functions. In this entry, we explore the journey of lichens being considered from a dual partnership to a multi-species symbiotic relationship.

Desde o início do século XXI que se percebe a necessidade de uma resposta única e singular à criança e à infância. Até então, o contexto sociocultural regia-se por uma lógica adultocentrada, não sendo reconhecido por parte do adulto a incapacidade de adotar a perspetiva da criança. Tal acentua-se com a Contemporaneidade, a sociedade do conhecimento e a vulgarização das TIC a enaltecerem o papel das crianças “nativo digitais”. O hiato geracional confonta pais/cuidadores, docentes, profissionais que atuam na infância e comunidade em geral com a criança competente e de direito. O desafio envolve educadores qualificados, orientações educativas específicas e contextos consagrados às crianças dos 0-3 anos, verdadeiros “lugares de infância”. Através da metodologia de position paper, pretende este artigo sustentar a posição de que esta é a conjuntura ideal para a “emergência” de um novo, revigorado e inclusivo “Sistema Educativo”, aquele que abraça, considera e destaca a CRECHE.

Abstract The state of the art of earthen architecture and vernacular built heritage comprises a complex set of issues that range from fundamental problematic recognition to anthropological and cultural studies and, more recently, to technological and experimental analyses. This paper addresses the development of the field, following the milestones of the international literature and pursuing a reflective-theory approach within a historical framework. It aims to explore the main contributions that have enhanced vernacular heritage and earthen architecture as specific domains, from pioneering public awareness essays to institutional expertise guidelines. Finally, in addition to the literature review process, this paper considers the recent corpus of recommendations from conservation management reference institutions, the updating of the operative problematic of earthen vernacular built heritage, and the relevance of local community involvement in facing increasing challenges.

Dextro-transposition of the great arteries (D-TGA) is a congenital heart disease (CHD) classically palliated with atrial switch (ATR-S) and nowadays corrected with arterial switch (ART-S). Our aim was to observe a group of D-TGA patients followed in an adult CHD Outpatient Clinic. We analyzed a group of D-TGA patients born between 1974 and 2001. Adverse events were defined as a composite of death, stroke, myocardial infarction or coronary revascularization, arrhythmia, and ventricular, baffle or significative valvular dysfunction. A total of 79 patients were enrolled, 46% female, mean follow-up of 27±6 years after surgery. ATR-S was performed in 54% and ART-S in 46%; median age at procedure was 13 months and 10 days, respectively. During follow-up, almost all ART-S remained in sinus rhythm versus 64% of ATR-S (p=0.002). The latter group had a higher incidence of arrhythmias (41% versus 3%, p

High salt levels in soil can severely limit plant development and diminish the positive effect of plant-growth-promoting rhizobacteria (PGPR). However, extracts of organisms adapted to high salinity, such as Opuntia ficus-indica (OFI) and Enteromorpha intestinalis (EI), can restore the growth of PGPR. Therefore, we used OFI or EI extracts and their combination with the PGPR Achromobacter xylosoxidans BOA4 to evaluate salt stress relief in tomato (Solanum lycopersicum). The experimental setup consisted of a plant pot trial under greenhouse conditions with 12 treatments: control, irrigation with OFI extract; EI extract; BOA4-inoculated plus OFI extract and BOA4-inoculated plus EI extract under no salinity or salinity conditions (150 mM NaCl). The percentage of germination, and plant’s fresh and dry weight were registered 30 and 46 days after sowing. At 46 days, the ratio between proline and glutamic acid concentration (PR/GA) was determined, expecting high PR/GA ratios in plants more responsive to salt stress since proline is an osmolyte mainly synthesized from glutamate. The results showed that 52% of the control seeds under salt stress germinated, a figure that was increased to 92% in OFI-treated seeds. Tomato plants were shown to be very sensitive to salt stress since the dry weight was ca. one fourth that of the plants grown without salinity. However, EI or BOA4 plus EI stimulated plant biomass by ca. 3 times compared to the control with salt, restoring plant biomass to values comparable to those of control plants grown without salinity. The joint treatments with BOA4 and EI or OFI caused distinct PR/GA levels in plant tissues. An inverse relationship between the sum of relative shoot proline and glutamic acid contents and shoot biomass accumulation was observed, namely in treatments accumulating more biomass under no salinity and salinity conditions. This indicates that the proline/glutamate pathway represents a carbon sink that is needed to fight stress and is competing with the carbon flow used for growth.

Wild mushrooms and truffles (MT) are important resources, which can contribute to the socioeconomic sustainability of forestry ecosystems. However, not all wild MT are edible. Fast, cheap, and reliable methods that distinguish wild MT species (including the deadly ones) can contribute to valuing these important forest resources. Here, we tested if wild MT species, and their edibility, could be distinguished based on their aroma profiles (i.e., smellprints). For that, we combined the use of the electronic nose with classification models (linear discriminant analysis (LDA) and partial least squares discriminant analysis (PLS-DA)) to distinguish between 14 wild MT species (including edible and non-edible species) collected in Portugal. The 14 wild MT species could be accurately distinguished using LDA (93% accuracy), while the edible and non-edible species could be accurately distinguished using both LDA and PLS-DA (97% and 99% accuracy, respectively). Keeping in mind that our methodological design’s feasibility was verified using a small sample, the data show the potential of the combined use of the electronic nose with discriminant analysis to distinguish wild MT species and their edibility based on their aromatic profile. Although a larger dataset will be necessary to develop a quick and reliable identification method, it shows potential to be as accurate as the identification performed by mycologists and molecular biology, yet requiring less technical training, and the analyses are cheaper and faster.

The overuse of chemical pesticides and fertilizers in crop farming has led to a decrease in crop quality and negative impacts on soil and the environment. It is crucial to adopt alternative strategies to maintain soil and environmental quality while enhancing crop growth and yield. To explore this, a study was conducted under greenhouse conditions to investigate the effect of Rhizobium tropici CIAT 899 alone, as well as in association with mycorrhizae (Rhizophagus irregularis) and endophytic fungus (Serendipita indica), on the growth, yield, and nutrient status of snap bean plants. At harvest, the rhizobial strain CIAT 899 demonstrated the highest effectiveness. It significantly increased the number of nodules in both Contender and Garrafal Enana varieties by 6.97% and 14.81%, respectively, compared with the control without inoculation. Furthermore, the results indicated that co-inoculation of Rhizobium and symbiotic fungi had positive effects on nitrogen content, phosphorus availability, and overall plant growth. Regardless of the variety, plants inoculated with R. tropici CIAT 899 and Serendipita indica exhibited the highest values for plant growth parameters. This combination resulted in 168% and 135% increases in root dry biomass, as well as 140% and 225% increases in the number of pods for Contender and Garrafal Enana, respectively, compared with the control at harvest. Additionally, this study highlights the potential benefits of combining R. tropici with either Serendipita indica or Rhizophagus irregularis in terms of nitrogen and phosphorus uptake. These symbiotic microorganisms demonstrated synergistic interactions with snap bean plants, leading to improved mineral nutrition and enhanced growth. Overall, these findings suggest that utilizing these symbiotic microorganisms can effectively enhance the mineral nutrition and growth of snap bean plants.

Terfezia arenaria is a desert truffle native to the Mediterranean Basin region, highly appreciated for its nutritional and aromatic properties. Despite the increasing interest in this desert truffle, T. arenaria is not listed as an edible truffle authorized for trade in the European Union. Therefore, our objective was to showcase T. arenaria’s nutritional and chemical composition and volatile profile. The nutritional analysis showed that T. arenaria is a good source of carbohydrates (67%), proteins (14%), and dietary fibre (10%), resulting in a Nutri-Score A. The truffle’s volatile profile was dominated by eight-carbon volatile compounds, with 1-octen-3-ol being the most abundant (64%), and 29 compounds were reported for the first time for T. arenaria. T. arenaria’s nutritional and chemical compositions were similar to those of four commercial mushroom and truffle species, while the aromatic profile was not. An electronic nose corroborated that T. arenaria‘s aromatic profile differs from that of the other four tested mushroom and truffle species. Our data showed that T. arenaria is a valuable food resource with a unique aroma and an analogous composition to meat, which makes it an ideal source for plant-based meat products. Our findings could help promote a sustainable future exploitation of T. arenaria and ensure the quality and authenticity of this delicacy.

BackgroundCoastal areas in Guinea-Bissau and elsewhere in West Africa are bordered by mangrove forests. In several of these places, swaths of mangrove forest have been removed and the landscape has been technologically adapted for the production of mangrove rice-a regionally important staple. However, the effects of global warming, in particular sea-level rise, pose challenges to these socioecological environments. In this context, knowledge appears as an important resource and knowing what knowledge has been produced and which perspectives have guided that production may inform future responses to climate change. We have developed a systematic literature review protocol focusing on the main question: "How have mangrove forest and mangrove rice spaces been represented in the literature on Guinea-Bissau?" The main hypothesis is that although they occupy contiguous, interrelated and interactant spaces in coastal environments, mangrove forests and mangrove rice have been studied and analyzed independently in the literature.MethodsThis is a protocol for conducting a systematic review that will include academic and non-academic literature in Portuguese, English and French. The academic literature will be retrieved from both Web of Science and Scopus using Boolean expressions. The non-academic literature will be accessed from relevant institutions, specialized libraries, and reference lists of previously selected items. Data extraction will follow a standard procedure based on an information sheet. Our analysis will be both qualitative (inductive and deductive coding, content analysis) and quantitative (word clouds, descriptive statistics and statistical testing).DiscussionThis systematic review will provide information about the conceptual framework that has been produced through research, policymaking, and conservation and development programs in the management of coastal areas. This study will identify the limitations of previous approaches and contribute to both future research and strategies for planning adaptation to climate change. Finally, the outputs will add to broader debates about people-nature coexistence and climate change adaptation and mitigation.

COVID-19 pandemic has unquestionably influenced care of acute myocardial infarction (AMI). Still, its impact on patients (pts) characteristics, presentation, treatment, and outcomes remains not well established in late pandemic times. To address this issue, we performed a prospective study of type-1 AMI pts admitted in a tertiary care hospital. Pts were enrolled during 6-months in 2019 (n=122; pre-COVID-19 (PC) group) and in 2021 (n=196; late-COVID-19 (C) group). Data was based on pts interview and review of medical records. Age and gender distribution, as well as ST/Non-ST-Elevation Myocardial Infarction (STEMI/NSTEMI) proportion and access to coronariography and revascularization were similar between groups. C pts presented more pre-existing established cardiovascular disease (CVD) (43% vs 30%; p=0,03); more frequent description of typical chest pain (94% vs 84%; p=0,002); higher levels of pain intensity, in a 0-10 scale (8±2 vs 7±2; p=0,02); higher frequencies of AMI complications (27% vs 15%; p=0,01) and worse Killip (K) class evolution (K≥2 in 22% C vs 13% PC pts; p=0,05). In conclusion, late pandemic AMI pts presented worse in-hospital outcomes in our study, though pre-hospital and hospital care were comparable to pre-pandemic times. Covid pts had a higher burden of pre-existing established CVD and a more typical and intense symptom presentation. Therefore, it can be hypothesized that "sicker" pts continued to look for help when presenting AMI symptoms, while "less sick" pts and the ones with less typical and intense symptoms possibly avoided contact with health care services during late pandemic period.

In recent decades, Higher Education has undergone a gradual but progressive transformation worldwide. Globalisation has promoted convergence mechanisms between states to achieve common standards in Higher Education; among them, those who seek to guarantee the quality of university teaching. The aim of this research is to analyse the most recent studies carried out on the impact that new teaching evaluation systems have on academics. For this purpose, a systematic review of the literature has been developed in which three different databases have been used (Wos, Scopus and ERIC). After searching and applying the inclusion / exclusion criteria, a total of 15 documents were selected. The articles were subjected to a thematic analysis and also of their characteristics. Among the most outstanding results, the development of fragmented academic identities that have an impact on the work and the wellbeing of the academics is detected. Also, the presence of contradictory opinions on teaching evaluation. Likewise, possible alternatives on teaching evaluation systems are mentioned. To conclude, a series of future lines of research are outlined, such as the consideration of more varied samples that allow more detailed comparisons between various variables, the development of longitudinal studies, and the promotion of alternatives to the current systems of evaluation.

Abstract The tumor suppressor FANCD1/BRCA2 is crucial for DNA homologous recombination repair (HRR). BRCA2 biallelic pathogenic variants result in a severe form of Fanconi anemia (FA) syndrome, whereas monoallelic pathogenic variants cause mainly hereditary breast and ovarian cancer predisposition. For decades, the co-occurrence in trans with a clearly pathogenic variant led to assume that the other allele was benign. However, here we show a patient with biallelic BRCA2 (c.1813dup and c.7796 A > G) diagnosed at age 33 with FA after a hypertoxic reaction to chemotherapy during breast cancer treatment. After DNA damage, patient cells displayed intermediate chromosome fragility, reduced survival, cell cycle defects, and significantly decreased RAD51 foci formation. With a newly developed cell-based flow cytometric assay, we measured single BRCA2 allele contributions to HRR, and found that expression of the missense allele in a BRCA2 KO cellular background partially recovered HRR activity. Our data suggest that a hypomorphic BRCA2 allele retaining 37–54% of normal HRR function can prevent FA clinical phenotype, but not the early onset of breast cancer and severe hypersensitivity to chemotherapy.

(1) Background: Increasing salinity, further potentiated by climate change and soil degradation, will jeopardize food security even more. Therefore, there is an urgent need for sustainable agricultural practices capable of maintaining high crop yields despite adverse conditions. Here, we tested if wheat, a salt-sensitive crop, could be a good reservoir for halotolerant bacteria with plant growth-promoting (PGP) capabilities. (2) Methods: We used two agricultural soils from Algeria, which differ in salinity but are both used to grow wheat. Soil halotolerant bacterial strains were isolated and screened for 12 PGP traits related to phytohormone production, improved nitrogen and phosphorus availability, nutrient cycling, and plant defence. The four ‘most promising’ halotolerant PGPB strains were tested hydroponically on wheat by measuring their effect on germination, survival, and biomass along a salinity gradient. (3) Results: Two halotolerant bacterial strains with PGP traits were isolated from the non-saline soil and were identified as Bacillus subtilis and Pseudomonas fluorescens, and another two halotolerant bacterial strains with PGP traits were isolated from the saline soil and identified as B. megaterium. When grown under 250 mM of NaCl, only the inoculated wheat seedlings survived. The halotolerant bacterial strain that displayed all 12 PGP traits and promoted seed germination and plant growth the most was one of the B. megaterium strains isolated from the saline soil. Although they both belonged to the B. megaterium clade and displayed a remarkable halotolerance, the two bacterial strains isolated from the saline soil differed in two PGP traits and had different effects on plant performance, which clearly shows that PGP potential is not phylogenetically determined. (4) Conclusions: Our data highlight that salt-sensitive plants and non-saline soils can be reservoirs for halotolerant microbes with the potential to become effective and sustainable strategies to improve plant tolerance to salinity. However, these strains need to be tested under field conditions and with more crops before being considered biofertilizer candidates.

Eimeriid coccidians represent one of the largest groups of parasitic unicellular organisms and comprise many species of veterinary and medical importance. The aim of this work is to provide information, as complete as possible, concerning the oocyst phase of the species of coccidians (Apicomplexa: Eimeriorina) with four sporocysts, which were reported in marine fish. For each species, the accepted scientific name and respective authorities, its synonyms, reported hosts, site of infection (organ), and geographic distribution have been assembled. Available information on morphology (oocyst, sporocyst, and sporozoite) and GenBank accession numbers were also compiled. A total of 100 species of coccidians were described and reported from 60 families of marine fishes. Most species have been described from marine teleosts, with only 14 of the species described from marine elasmobranchs. Most of the species reported in marine fish belong to the genera Eimeria and Goussia, and only a handful belong to the genera Epieimeria, Calyptospora, and Crystallospora. Although marine coccidians have began to be described for more than one century, the collection of genetic data on marine fish coccidians only started in the 2010s and remains largely disconnected from the morphological analysis of specimens, which is available for only six formally described species.

Grapevine is one of the most important fruit crops worldwide, being Portugal one of the top wine producers. It is well established that wine sensory characteristics from a particular region are defined by the physiological responses of the grapevine to its environment and thus, the concept of terroir in viticulture was established. Among all the factors that contribute to terroir definition, soil microorganisms play a major role from nutrient recycling to a drastic influence on plant fitness (growth and protection) and of course wine production. Soil microbiome from four different terroirs in Quinta dos Murças vineyard was analysed through long-read Oxford Nanopore sequencing. We have developed an analytical pipeline that allows the identification of function, ecologies, and indicator species based on long read sequencing data. The Douro vineyard was used as a case study, and we were able to establish microbiome signatures of each terroir.

Food safety is a significant challenge worldwide, from plantation to cultivation, especially for perishable products such as tomatoes. New eco-friendly strategies are needed, and beneficial microorganisms might be a sustainable solution. This study demonstrates bacteria activity in the tomato plant rhizosphere. Further, it investigates the rhizobacteria’s structure, function, and diversity in soil. Rhizobacteria that promote the growth and development of tomato plants are referred to as plant growth-promoting bacteria (PGPR). They form a series of associations with plants and other organisms in the soil through a mutualistic relationship where both parties benefit from living together. It implies the antagonistic activities of the rhizobacteria to deter pathogens from invading tomato plants through their roots. Some PGPR are regarded as biological control agents that hinder the development of spoilage organisms and can act as an alternative for agricultural chemicals that may be detrimental to the health of humans, animals, and some of the beneficial microbes in the rhizosphere soil. These bacteria also help tomato plants acquire essential nutrients like potassium (K), magnesium (Mg), phosphorus (P), and nitrogen (N). Some rhizobacteria may offer a solution to low tomato production and help tackle food insecurity and farming problems. In this review, an overview of soil-inhabiting rhizobacteria focused on improving the sustainable production of Solanum lycopersicum.

La Covid-19 generó un importante impacto en la salud mental de los estudiantes universitarios. En este contexto, surge una doble necesidad: analizar los efectos de este impacto y trabajar la educación emocional con este sector educativo. Este estudio forma parte de un proyecto más amplio de innovación educativa centrado en trabajar la educación emocional de los estudiantes universitarios. En este caso, el principal objetivo del presente estudio fue analizar las experiencias de los estudiantes universitarios (N=6) durante la pandemia a través de Digital Storytellings. Los resultados muestran aspectos reveladores sobre la influencia que tuvo la pandemia en los estudiantes: las carencias de la virtualidad en la enseñanza, el desarrollo de estrategias de afrontamiento, las lecciones aprendidas de la pandemia, etc. Por último, se exponen una serie de implicaciones para la educación superior, como la promoción de planes de educación emocional, la optimización de los recursos educativos en línea y el aumento de la formación del profesorado en temas digitales y de apoyo emocional.

Resumo: Nos últimos anos, tem sido crescente o reconhecimento das causas genéticas das doenças cardiovasculares resultado dos significativos progressos das técnicas laboratoriais. Este conhecimento tem permitido a identificação de «novos» fenótipos e a subclassificação das síndromes clínicas, tendo impacto nas decisões terapêuticas e no aconselhamento genético que é facultado às famílias.No presente documento descreve‐se o «estado da arte» relativamente às principais recomendações para testes genéticos nas doenças cardiovasculares, pretendendo‐se providenciar uma ferramenta útil de consulta para cardiologistas e outros profissionais envolvidos na prestação nos cuidados de saúde a doentes com cardiopatias hereditárias e respetivas famílias. Abstract: In recent years, the importance of genetic causes of cardiovascular diseases has been increasingly recognized, as the result of significant advances in molecular diagnosis techniques. This growing knowledge has enabled the identification of new phenotypes and the subclassification of clinical syndromes, impacting the therapeutic approach and genetic counseling offered to affected families.This paper describes the state of the art of genetic testing in the main cardiovascular diseases, aiming to provide a useful tool to help cardiologists and other health professionals involved in the care of individuals with hereditary heart diseases and their families.

Abstract Few studies have tested how plant quality and the presence of competitors interact in determining how herbivores choose between different leaves within a plant. We investigated this in two herbivorous spider mites sharing tomato plants: Tetranychus urticae, which generally induces plant defenses, and Tetranychus evansi, which suppresses them, creating asymmetrical effects on coinfesting competitors. On uninfested plants, both herbivore species preferred young leaves, coinciding with increased mite performance. On plants with heterospecifics, the mites did not prefer leaves on which they had a better performance. In particular, T. urticae avoided leaves infested with T. evansi, which is in agreement with T. urticae being outcompeted by T. evansi. In contrast, T. evansi did not avoid leaves with the other species, but distributed itself evenly over plants infested with heterospecifics. We hypothesize that this behavior of T. evansi may prevent further spread of T. urticae over the shared plant. Our results indicate that leaf age determines within‐plant distribution of herbivores only in absence of competitors. Moreover, they show that this distribution depends on the order of arrival of competitors and on their effects on each other, with herbivores showing differences in behavior within the plant as a possible response to the outcome of those interactions.

The effect of a soil thermophilic bacteria (STB), Ureibacillus sp. 18UE/10 on the status of wheat plants was evaluated. A greenhouse assay was performed, mimicking scenarios of soil impoverishment and aridity, which included Rhizophagus irregularis, a crop enhancer AMF, for effect comparison. Treatments with strain 18, R. irregularis or both had no significant effect on biomass production, however affected plant physiology. A different partition in biomass, nitrogen and carbon content were observed, resulting in a decreased C/N ratio. Elemental analysis showed an increase in N and P content in shoots, and for treatments containing STB a decrease in the content of several toxic metals. Strain 18 had a distinct δ13C isotopic signature translating an increased stomatal conductance. ATR-IR spectroscopy revealed that root exudate influenced STB cell wall structure and increased the bacterial survival rate at 25°C. These findings show that STB can interact with a plant partner under rhizospheric conditions.

It is known that ectomycorrhizal (ECM) fungi can modulate below and aboveground communities. They are a key part of belowground communication as they produce a vast array of metabolites, including volatile organic compounds (VOCs) such as 1-octen-3-ol. Here, we tested if the VOC 1-octen-3-ol may be involved in the ECM fungal mechanisms that modulate below and aboveground communities. For that, we conducted three in vitro assays with ECM fungi and the 1-octen-3-ol volatile to (i) explore the effects of mycelium growth of three ECM species, (ii) investigate the impact on the germination of six host Cistaceae species, and (iii) study the impact on host plant traits. The effects of 1-octen-3-ol on mycelium growth of the three ECM species depended on the dose and species: Boletus reticulatus was the most sensitive species to the low (VOC) dose, while T. leptoderma was the most tolerant. In general, the presence of the ECM fungi resulted in higher seed germination, while 1-octen-3-ol resulted in lower seed germination. The combined application of the ECM fungus and the volatile further inhibited seed germination, possibly due to the accumulation of 1-octen-3-ol above the plant species’ threshold. Seed germination and plant development of Cistaceae species were influenced by ECM fungal volatiles, suggesting that 1-octen-3-ol may mediate changes in below and aboveground communities.

Drought is a worldwide problem, especially in arid and semi-arid regions. Detection of drought stress at the initial stages, before visible signs, to adequately manage irrigation and crop fertilization to avoid crop yield loss, is a desire of most farmers. Here, we evaluated the response of tomato plants to water scarcity, through changes in leaf reflectance due to modification in leaf pigments’ content, which translates into differences in spectral reflectance indices (SRI) values. Our methodology is innovative, as we were able to easily calculate and identify several SRIs for the early detection of drought stress “invisible” responses. We used a handheld spectro-radiometer to obtain SRI values from leaves of tomato plants growing under two different water regimes for 37 days. In an ensemble of 25 SRIs, we identified 12 that showed a consistent trend of significant differences between treatments along the experiment and within these, NDVI, SR, ZMI, Ctr2, GM1, and GM2 were already significantly different between treatments at day 7 after the start of the experiment and Ctr1 at day 9; although, no signs of damage were visible. Therefore, our results pinpoint these SRIs as promising proxies for the early detection of “invisible” responses to drought onset. We also analyzed the relationship between the monitored SRIs and plant morphological parameters measured during the experiment, highlighting a relationship between GM1 and plant height and leaf number. Finally, we observed a high abundance of putative beneficial bacteria among isolates collected from the tomato water-limited rhizo-environment at the terminus of the experiment, suggesting the active recruitment or selection of Plant Growth Promoting Rhizobacteria by tomato roots as a response to drought. Our work may be adapted into an easy protocol, of rapid execution, to be used in small-scale fields for early drought stress detection.

Plants and their associated microbiota share ecological and evolutionary traits that are considered to be inseparably woven. Their coexistence foresees the use of similar metabolic pathways, leading to the generation of molecules that can cross-regulate each other’s metabolism and ultimately influence plant phenotype. However, the extent to which the microbiota contributes to the overall plant metabolic landscape remains largely unexplored. Due to their early presence in the seed, seed-borne endophytic bacteria can intimately colonize the plant’s endosphere while conferring a series of phytobeneficial services to their host. Understanding the dynamics of these endophytic communities is a crucial step toward the formulation of microbial inoculants that can modulate the functionality of the plant-associated microbiota for improved plant fitness. In this work, wheat (Triticum aestivum) roots non-inoculated and inoculated with the bacterium Herbaspirillum seropedicae strain RAM10 were analyzed to explore the impact of inoculant–endophyte–wheat interrelationships on the regulation of tryptophan (Trp) metabolism in the endosphere environment. Root inoculation with H. seropedicae led to phylum-specific changes in the cultivable seed-borne endophytic community. This modulation shifted the metabolic potential of the community in light of its capacity to modulate the levels of key Trp-related metabolites involved in both indole-3-acetic acid (IAA) biosynthesis and in the kynurenine pathway. Our results support a mode of action of H. seropedicae relying on a shift in both the composition and functionality of the seed-borne endophytic community, which may govern important processes such as root growth. We finally provide a conceptual framework illustrating that interactions among roots, inoculants, and seed-borne endophytes are critical to fine-tuning the levels of IAA in the endosphere. Understanding the outcomes of these interactions is a crucial step toward the formulation of microbial inoculants based on their joint action with seed-borne endophytic communities to promote crop growth and health in a sustainable manner.

Woody invasive species pose a big threat to ecosystems worldwide. Among them, Acacia longifolia is especially aggressive, fundamentally changing ecosystem structure through massive biomass input. This biomass is rarely harvested for usage; thus, these plants constitute a nuisance for stakeholders who invest time and money for control without monetary return. Simultaneously, there is an increased effort to valorise its biomass, e.g., for compost, growth substrate or as biofuel. However, to incentivise A. longifolia harvest and usage, stakeholders need to be able to estimate what can be obtained from management actions. Thus, the total biomass and its quality (C/N ratio) need to be predicted to perform cost–benefit analyses for usage and determine the level of invasion that has already occurred. Here, we report allometric biomass models for major biomass pools, as well as give an overview of biomass quality. Subsequently, we derive a simplified volume-based model (BM ~ 6.297 + 0.982 × Vol; BM = total dry biomass and Vol = plant volume), which can be applied to remote sensing data or with in situ manual measurements. This toolkit will help local stakeholders, forest managers or municipalities to predict the impact and valorisation potential of this invasive species and could ultimately encourage its management.

Atmospheric nitrogen (N) inputs in the Mediterranean Basin are projected to increase due to fossil fuel combustion, fertilizer use, and the exacerbation of agricultural production processes. Although increasing N deposition is recognized as a major threat to ecosystem functioning, little is known about how local environmental conditions modulate ecosystem function response to N addition, particularly in the context of Mediterranean-Basin ecosystems. Here, we assess how N addition affects important ecosystem properties associated with litter decomposition, soil physical-chemical properties, soil extracellular enzymatic activity and microbial abundance across three long-term N addition experimental sites in the Mediterranean Basin. Sites were located in El Regajal (Madrid, Spain), Capo Caccia (Alghero, Italy), and Arrábida (Lisbon, Portugal) and are all representative of Mediterranean shrublands. No common pattern for litter decomposition process or other studied variables emerged among the control plots of the studied sites. Nitrogen supply only affected soil pH, a major driver of decomposition, in two out of three experimental sites. Moreover, when we explored the role of N addition and soil pH in controlling litter decay, we found that the effects of these factors were site-dependent. Our results point out to local ecosystem features modulating N addition effects in controlling litter decomposition rates in Mediterranean ecosystems, suggesting that the responses of soil functioning to N deposition are site-dependent. These findings provide further knowledge to understand contrasting ecosystem responses to N additions based on a single field experiments.

Salinity causes significant agricultural losses in many areas in the world. Plant growth promoting bacteria (PGPB) are a promising solution to enhance plant growth and productivity under such stress conditions by different mechanisms, mainly phosphorous solubilization. This study aims to improve wheat seedling growth under salt stress by a halotolerant phosphorous-solubilizing bacterial strain. Soil sample was collected in the south of Algeria (Ghardaia), and bacterial isolation was carried out on nutrient agar (NA) at different NaCl concentrations (300; 600 and 900 mM). The ability of the halotolerant isolates to solubilize inorganic phosphorous at 0; 300; 600 and 900 mM NaCl was determined. The isolate that showed the highest solubilization indexes was selected and identified as Pseudomonas azotoformans. Sterile wheat (Triticum aestivum) seeds were inoculated by this strain and then sown in soil at different NaCl concentrations (0; 100; 200; 300 mM). Different growth parameters were measured after 15 days. The strain showed its highest capacity for phosphorous solubilization (255.152 ± 0.01 µg/mL) at 300 mM NaCl, and for phytate mineralization (0.168 ± 0.002 U/mL) at 100 mM NaCl. The highest amount of soluble phosphorous in the soil was 49.42 ± 0.36 ppm obtained at 100 mM NaCl. Seed germination percentage, shoot and root length and fresh and dry weights were found to be higher in bacterial inoculated seedlings compared to uninoculated ones. According to this study, the use of plant growth promoting bacteria represents an important biotechnological approach to restore phosphorous levels in saline soils and to promote plant growth in salt-affected agricultural land.

Mankind’s strong dependence on nitrogen (N) began when we started farming and, ever since, we have depended on nitrogen in the soil for plant production. More than a century has passed since the discovery of N as an element until the advent of synthetic fertilizers. Today, after a century of Haber–Bosch innovation, many other endeavors and challenges can be launched to understand how the effects of N in the environment can be perceived as ‘good’ or ‘bad’. All this knowledge evolution was truly dependent on the scientific advances, both technological and methodological, and particularly on the approaches at the micro and macro level. As with nearly everything in our lives (e.g., events, people, food, decisions, world history), we tend to use the dichotomy ‘good or bad’ to categorize, and scientific advances are no exception. The integration of scientific and technological advances allows us to move beyond this simple dichotomy ‘good or bad’ and to make choices. Here, we review the main marks in understanding plant nutrition throughout time, with special emphasis on N, from the Greeks to the most recent trends in the 21st century. Since improving plant N use efficiency is a main avenue to meet several Sustainable Developmental Goals (e.g., SDG2 zero hunger, SDG12 responsible production and consumption, SDG15 life on land), the European Green Deal, and The Farm to Fork strategy, we propose that the ecological principles must be integrated in agro-ecosystem management. During the last 40 years, our research group has contributed to: (i) the clarification of the so-called dichotomy of choices when it comes to the environmental effects of N; and (ii) fetching natural solutions for N manmade problems. This was based on the knowledge that life is a continuous symbiotic interplay between mutualism and parasitism depending on environmental conditions and that there is a need for feeding people, assuring food quality and diminishing environmental impacts. We argue that, as a society, we have the scientific and technological means to learn from nature and to apply the ecological rules in agro-ecosystems. However, this is a choice we must make as individuals and as a society.

Dès la première version du programme de formation de la licence en animation socioculturelle de l’École Supérieure d’Éducation de Lisbonne en 2006, l’équipe enseignante a jugé bon d’y intégrer une composante de sciences physiques et naturelles. En effet, dans les sociétés contemporaines comprises comme des sociétés du risque, la réflexivité est centrale et la science et les scientifiques deviennent de plus en plus nécessaires, mais sans qu’on les sépare des populations, au contraire. Ce besoin de réflexivité sur les risques apporte des défis supplémentaires à la formation et à l’intervention des professionnels en animation socioculturelle. Une enquête auprès des étudiants nous montre quelles sont leurs représentations de sciences physiques et naturelles et quelle importance ils attribuent à ces connaissances en tant que citoyens et animateurs socioculturels. Une approche transdisciplinaire, incluant les sciences physiques et naturelles, semble offrir des outils utiles pour la difficile tâche des animateurs qui est de trouver des moyens pour faire face à une réalité pleine de complexités et d’ambiguïtés.

C’est à la suite des enseignements découlant de conférences prononcées par Jean-Claude Gillet à Lisbonne que la formation en animation offerte à l’École Supérieure d’Éducation de l’Institut Polytechnique a été recadrée autour, d’une part, de la relation complexe que les animateurs tissent dans leur activité professionnelle avec les décideurs, les élus et les employeurs et, d’autre part, de la centralité de l’agir professionnel permettant à la fois la production de savoirs et le développement de compétences pour décider et agir. Le texte qui suit évoque les origines et la pérennité de ce legs tenace.

Previous works indicate that data fusion, compared to single data modelling can improve the assessment of soil attributes using spectroscopy. In this work, two different kinds of proximal soil sensing techniques i.e., mid-infrared (MIR) and X-ray fluorescence (XRF) spectroscopy were evaluated, for assessment of seven fertility attributes. These soil attributes include pH, organic carbon (OC), phosphorous (P), potassium (K), magnesium (Mg), calcium (Ca) and moisture contents (MC). Three kinds of spectra fusion (SF) (spectra concatenation) approaches of MIR and XRF spectra were compared, namely, spectra fusion-Partial least square (SF-PLS), spectra fusion-Sequential Orthogonalized Partial least square (SF-SOPLS) and spectra fusion-Variable Importance Projection-Sequential Orthogonalized Partial least square (SF-VIP-SOPLS). Furthermore, the performance of SF models was compared with the developed single sensor model (based on individual spectra of MIR and XRF). Compared with the results obtained from single sensor model, SF models showed improvement in the prediction performance for all studied attributes, except for OC, Mg, and K prediction. More specifically, the highest improvement was observed with SF-SOPLS model for pH [R2p = 0.90, root mean square error prediction (RMSEP) = 0.15, residual prediction deviation (RPD) = 3.30, and ratio of performance inter-quantile (RPIQ) = 3.59], successively followed by P (R2p = 0.91, RMSEP = 4.45 mg/100 g, RPD = 3.53, and RPIQ = 4.90), Ca (R2p = 0.92, RMSEP = 177.11 mg/100 g, RPD = 3.66, and RPIQ = 3.22) and MC (R2p = 0.80, RMSEP = 1.91%, RPD = 2.31, RPIQ = 2.62). Overall the study concluded that SF approach with SOPLS attained better performance over the traditional model developed with the single sensor spectra, hence, SF is recommended as the best SF method for improving the prediction accuracy of studied soil attributes. Moreover, the multi-sensor spectra fusion approach is not limited for only MIR and XRF data but in general can be extended for complementary information fusion in order to improve the model performance in precision agriculture (PA) applications.

The effect of seed coating salt-stressed tomato with the bacterium Achromobacter xylosoxidans BOA4 and/or irrigation with an extract of the marine algae Enteromorpha intestinalis (EI) is herein evaluated. The plant shoots and roots were harvested separately on day 50, following extensive saline stress. The addition of BOA4 and/or EI extract resulted in an average increase of 33% in plant shoot DW, but an averaged decrease of 44% in the root to shoot biomass ratio. Anthocyanin content increased by over 34% and 44% with EI and BOA4 plus EI treatments, respectively. Since enhanced protein tyrosine nitration (PTN) is a known plant response to salt stress, the PTN level was inspected through 3-nitrotyrosine content determination. This was drastically increased by salt stress; however, BOA4, EI or both caused an averaged PTN decrease of 30% in stressed roots or shoots. This PTN response could be associated with tomato phenotypic characteristics and is postulated to be inversely correlated to cytokinin contents in stressed plants, namely cis-zeatin-type-cis-zeatin (cZ) plus cis-zeatin riboside (cZR), and isopentenyladenine (iP). The latter showed a drastic average increase by 3.6-fold following BOA4 and/or EI treatments of salinized tomato. This increment could be related to cytokinin biosynthesis induced by the applied bio-stimulants; IP and derivatives are the main cytokinins in seaweeds, and Achromobacter xylosoxidans BOA4 was shown to produce up to 17.5 pmol mL−1 of isopentenyladenine. This work is the first report on the influence of bio-stimulants, used to improve salt stress tolerance, on plant PTN levels; BOA4 and/or EI treatments decreased PTN, while increasing cis-zeatin-type and iP cytokinins in tomato, the latter showed an enhanced tolerance to salt stress.

Soil salinization has become a major problem for agriculture worldwide, especially because this phenomenon is continuously expanding in different regions of the world. Salinity is a complex mechanism, and in the soil ecosystem, it affects both microorganisms and plants, some of which have developed efficient strategies to alleviate salt stress conditions. Currently, various methods can be used to reduce the negative effects of this problem. However, the use of biological methods, such as plant-growth-promoting bacteria (PGPB), phytoremediation, and amendment, seems to be very advantageous and promising as a remedy for sustainable and ecological agriculture. Other approaches aim to combine different techniques, as well as the utilization of genetic engineering methods. These techniques alone or combined can effectively contribute to the development of sustainable and eco-friendly agriculture.

Climate change and atmospheric nitrogen (N) deposition on drylands are greatly threatening these especially vulnerable areas. Soil biocrust-forming lichens in drylands can provide early indicators of these disturbances and play a pivotal role, as they contribute to key ecosystem services. In this study, we explored the effects of different long-term water availability regimes simulating climate changes and their interaction with N addition on the physiological response of the soil lichen Cladonia rangiferina. Three sets of this lichen were subjected to control, reduced watering, and reduced watering and N addition (40 kg NH4NO3 ha−1 year−1) treatments for 16 months. Finally, all samples were subjected to daily hydration cycles with N-enriched water at two levels (40 and 80 kg NH4NO3 ha−1 year−1) for 23 days. We found that reduced watering significantly decreased the vitality of this lichen, whereas N addition unexpectedly helped lichens subjected to reduced watering to cope with stress produced by high temperatures. We also found that long-term exposure to N addition contributed to the acclimation to higher N availability. Overall, our data suggest that the interactions between reduced watering and increased N supply and temperature have an important potential to reduce the physiological performance of this soil lichen.

Serendipita indica (former Piriformospora indica) is a non-obligate endophytic fungus and generally a plant growth and defence promoter with high potential to be used in agriculture. However, S. indica may switch from biotrophy to saprotrophy losing its plant growth promoting traits. Our aim was to understand if the free-living stage growth conditions (namely C availability) regulate S. indica’s phenotype, and its potential as plant-growth-promoting-microbe (PGPM). We grew S. indica in its free-living stage under increasing C availabilities (2–20 g L–1 of glucose or sucrose). We first characterised the effect of C availability during free-living stage growth on fungal phenotype: colonies growth and physiology (plasma membrane proton pumps, stable isotopic signatures, and potential extracellular decomposing enzymes). The effect of the C availability during the free-living stage of the PGPM was evaluated on wheat. We observed that C availability during the free-living stage regulated S. indica’s growth, ultrastructure and physiology, resulting in two distinct colony phenotypes: compact and explorer. The compact phenotype developed at low C, used peptone as the major C and N source, and displayed higher decomposing potential for C providing substrates; while the explorer phenotype developed at high C, used glucose and sucrose as major C sources and casein and yeast extract as major N sources, and displayed higher decomposing potential for N and P providing substrates. The C availability, or the C/N ratio, during the free-living stage left a legacy to the symbiosis stage, regulating S. indica’s potential to promote plant growth: wheat growth promotion by the explorer phenotype was ± 40% higher than that by the compact phenotype. Our study highlights the importance of considering microbial ecology in designing PGPM/biofertilizers. Further studies are needed to test the phenotypes under more extreme conditions, and to understand if the in vitro acquired characteristics persist under field conditions.

Hydrogen peroxide (H2O2) functions as an important signaling molecule in plants during biotic interactions. However, the extent to which H2O2 accumulates during these interactions and its implications in the development of disease symptoms is unclear. In this work, we provide a step-by-step optimized protocol for in situ quantification of relative H2O2 concentrations in wheat leaves infected with the pathogenic bacterium Pseudomonas syringae pv. atrofaciens (Psa), either alone or in the presence of the beneficial bacterium Herbaspirillum seropedicae (RAM10). This protocol involved the use of 3-3′diaminobenzidine (DAB) staining method combined with image processing to conduct deconvolution and downstream analysis of the digitalized leaf image. The application of a linear regression model allowed to relate the intensity of the pixels resulting from DAB staining with a given concentration of H2O2. Decreasing H2O2 accumulation patterns were detected at increasing distances from the site of pathogen infection, and H2O2 concentrations were different depending on the bacterial combinations tested. Notably, Psa-challenged plants in presence of RAM10 accumulated less H2O2 in the leaf and showed reduced necrotic symptoms, pointing to a potential role of RAM10 in reducing pathogen-triggered H2O2 levels in young wheat plants.

Nitrification inhibitors are commonly used to prevent nitrate leaching. However, the use of nitrification inhibitors is not free of side-effects. Some may be absorbed by the plant and cause phytotoxicity or even affect the food chain. Therefore, a solution that limits the absorption of nitrification inhibitors and its accumulation by the plant may mitigate health and environmental issues potentially associated with high levels of nitrification inhibitors. This solution may relay in the modulation of the plant’s metabolism through the interaction with specific fungal partners. This work tested the hypothesis that the symbiotic interaction between fungi and plant roots can reduce the destructive effects of the nitrification inhibitor Dicyandiamide (DCD) in plants by reducing the uptake of nitrification inhibitors. A greenhouse experiment was conducted, using a complete randomized block design, to test the effect of symbiotic fungi (plants inoculated with Piriformospora indica, Glomus etunicatum, and Glomus mosseae and noninoculated) on the phytotoxicity of DCD applied at four concentrations (0, 5, 50, and 100 mg kg−1 soil). Latuca sativa, cultivar Siyahoo, was selected for this experiment due to its economic value all over the world. The use of high DCD concentrations (100 mg kg−1 soil) affected the leaf chlorophyll content and plant growth in a manner that was significantly mitigated by the symbiosis of the plant with the fungal partner. These results highlight the benefits of using symbiotic fungal inoculants as plant protectors against the phytotoxic effects of DCD.

Abstract Poly(ADP‐ribose) polymerase (PARP) inhibitors (PARPi) are effective in cancers with defective homologous recombination DNA repair (HRR), including BRCA1/2‐related cancers. A test to identify additional HRR‐deficient tumors will help to extend their use in new indications. We evaluated the activity of the PARPi olaparib in patient‐derived tumor xenografts (PDXs) from breast cancer (BC) patients and investigated mechanisms of sensitivity through exome sequencing, BRCA1 promoter methylation analysis, and immunostaining of HRR proteins, including RAD51 nuclear foci. In an independent BC PDX panel, the predictive capacity of the RAD51 score and the homologous recombination deficiency (HRD) score were compared. To examine the clinical feasibility of the RAD51 assay, we scored archival breast tumor samples, including PALB2‐related hereditary cancers. The RAD51 score was highly discriminative of PARPi sensitivity versus PARPi resistance in BC PDXs and outperformed the genomic test. In clinical samples, all PALB2‐related tumors were classified as HRR‐deficient by the RAD51 score. The functional biomarker RAD51 enables the identification of PARPi‐sensitive BC and broadens the population who may benefit from this therapy beyond BRCA1/2‐related cancers.

Introduction and Objectives: Aortic dilatation can develop late after tetralogy of Fallot repair. Its extension beyond the aortic root is not clearly understood. We aimed to assess the prevalence and predictors of ascending aorta dilatation to set up an imaging protocol. Methods: In this prospective study including adult patients after tetralogy of Fallot repair followed at a referral center, we assessed the aorta by cardiovascular magnetic resonance and defined ascending aorta dilatation as an observed-to-expected ratio >1.5. Results: We included 78 adults (mean age 31±10 years; 56% female), with a mean follow-up of 23±7 years since tetralogy of Fallot repair. The prevalence of ascending aorta dilatation was 11.5%. The ascending aorta was larger than the sinuses of Valsalva in 12.8% of cases. Patients with ascending aorta dilatation were older, predominantly male, with later repair and larger left ventricular mass and volumes. By multivariate analysis left ventricular mass index (LVMI) was the only factor independently associated with ascending aorta dilatation (odds ratio 1.10, 95% confidence interval 1.01-1.20, p=0.03). A cut-off value of ≥57.9 g/m2 for LVMI had 89% sensitivity and 71% specificity for ascending aorta dilatation. Conclusions: Ascending aorta assessment as part of a routine cardiovascular magnetic resonance study after tetralogy of Fallot repair is recommended to screen for future aortic complications, particularly in males and older patients, and those with later repair and larger left ventricles. LVMI assessment has potential as a screening tool for ascending aorta dilatation with future clinical implications. Pretendemos avaliar a prevalência e os preditores de dilatação da aorta ascendente para elaborar um protocolo imagiológico. Métodos: Estudo prospetivo com adultos operados a tetralogia de Fallot seguidos num centro de referência. Estudamos a aorta por ressonância magnética cardiovascular e definimos dilatação da aorta ascendente pelo rácio observado-esperado > 1,5. Resultados: Incluímos 78 adultos (idade média 31 ± 10 anos; 56% mulheres); seguimento médio de 23 ± 7 anos desde a cirurgia. A prevalência de dilatação da aorta ascendente foi 11,5%. A aorta ascendente era maior do que os seios de Valsalva em 12,8% dos casos. Os doentes com dilatação da aorta ascendente eram mais velhos, maioritariamente homens, operados mais tarde, com massa e volumes ventriculares esquerdos maiores. Na análise multivariada a massa ventricular esquerda indexada foi a única variável independente associada a dilatação da aorta ascendente (odds ratio 1,10; intervalo de confiança de 95% 1,01-1,20; p = 0,03). A massa ventricular esquerda indexada ≥ 57,9 g/m2 apresentou uma sensibilidade de 89% e uma especificidade de 71% para dilatação da aorta ascendente. Conclusões: Recomendamos a inclusão da aorta ascendente na avaliação por ressonância magnética da tetralogia de Fallot operada, para prevenir complicações aórticas futuras, em particular em homens, doentes mais velhos, operados mais tarde e com ventrículos esquerdos maiores. A massa ventricular esquerda indexada tem potencial para ser usada no rastreio da dilatação da aorta ascendente com implicações clínicas futuras. Introduction and Aims: Intrinsic aortopathy can lead to dilatation late after tetralogy of Fallot (TOF) repair. Its extent and prevalence are not known. We aimed to assess aortic dimensions and elasticity and to find predictors of aortic dilatation. Methods: A total of 126 adults were prospectively included after TOF repair and compared to 63 gender- and age-matched controls. Transthoracic echocardiography was used to assess aortic diameters at the level of the sinuses of Valsalva and ascending aorta and aortic dilatation was defined as z-score >+2. M-mode parameters of the ascending aorta were used to calculate strain, distensibility and stiffness index. Results: TOF patients (mean age 30±9 years; 52% male) had a complete repair at a median age of five (2-49) years; mean follow-up time since repair was 23±7 years. The prevalence of aortic dilatation at the sinuses of Valsalva and ascending aorta was 29% and 24%, respectively. Compared to controls, TOF patients had a higher ascending aorta z-score, lower strain (6.4% [0.0-61.5] vs. 15.2% [0.0-45.0], p +2. Resultados: Doentes com tetralogia de Fallot (idade média 30 ± 9 anos; 52% homens) foram operados com uma idade mediana de 5 (2-49) anos; tempo médio de seguimento desde a cirurgia 23 ± 7 anos. A prevalência de dilatação dos seios de Valsalva e da aorta ascendente foi 29% e 24%, respetivamente. Comparado aos controlos, os doentes com tetralogia de Fallot apresentaram maior z-score da aorta ascendente, menor strain (6,4 [0,0-61,5] versus 15,2 [0,0-45,0]%; p < 0,01) e maior índice de rigidez (7,3 [0,8-23,6] versus 3,1 [0,9-14,1]; p < 0,01). Na análise multivariada o sexo masculino associou-se significativamente à dilatação dos seios de Valsalva (odds ratio 6,3, intervalo de confiança de 95% 1,5-26,3; p = 0,01). Conclusões: Na tetralogia de Fallot operada há uma prevalência significativa de dilatação tardia da aorta, com a aorta ascendente maior e mais rígida. O sexo masculino parece influenciar a dilatação da raiz da aorta. Esta aortopatia requer um seguimento cuidadoso para evitar complicações futuras. The aim of our study was to apply a biomimetic approach, inspired by the Ammophila arenaria. This organism possesses a reversible leaf opening and closing mechanism that responds to water and salt stress (hydronastic movement). We adopted a problem-based biomimetic methodology in three stages: (i) two observation studies; (ii) how to abstract and develop a parametric model to simulate the leaf movement; and (iii) experiments with bimetal, a smart material that curls up when heated. We added creases to the bimetal active layer in analogy to the position of bulliform cells. These cells determine the leaf-closing pattern. The experiments demonstrated that creases influence and can change the direction of the bimetal natural movement. Thus, it is possible to replicate the Ammophila arenaria leaf-rolling mechanism in response to temperature variation and solar radiation in the bimetal. In future works, we will be able to propose responsive facade solutions based on these results.

It was hypothesized that under induced lipid malabsorption/maldigestion conditions, an enriched sn-1(3)-monoacylglycerol (MAG) oil may be a better carrier for n-3 long-chain PUFAs (LC-PUFAs) compared with triacylglycerol (TAG) from fish oil. This monocentric double blinded clinical trial examined the accretion of EPA (500 mg/day) and DHA (300 mg/day) when consumed as TAG or MAG, into the erythrocytes, plasma, and chylomicrons of 45 obese (BMI ≥30 kg/m2 and ≤40 kg/m2) volunteers who were and were not administered Orlistat, an inhibitor of pancreatic lipases. Intake of MAG-enriched oil resulted in higher accretion of LC-PUFAs than with TAG, the concentrations of EPA and DHA in erythrocytes being, respectively, 72 and 24% higher at 21 days (P < 0.001). In addition, MAG increased the plasma concentration of EPA by 56% (P < 0.001) as compared with TAG. In chylomicrons, MAG intake yielded higher levels of EPA with the area under the curve (0–10 h) of EPA being 55% greater (P = 0.012). In conclusion, in obese human subjects with Orlistat-induced lipid maldigestion/malabsorption conditions, LC-PUFA MAG oil increased LC-PUFA levels in erythrocytes, plasma, and chylomicrons to a greater extent than TAG. These results indicate that MAG oil might require minimal enzymatic digestion prior to intestinal uptake and transfer across the epithelial barrier.
