Our study aimed to investigate the association between polymorphism of three core genes belonging to NER pathway and preeclampsia (PE) risk in Chinese Han Women. The DNA used in our experiment was extracted from 1004 cases and 1274 normal pregnant women. All single nucleotide polymorphisms (SNPs) were analyzed with TaqMan allelic discrimination real-time PCR. Our findings showed the genotype and allele frequency of XPA rs1800975, XPF rs1799801, XPG rs17655 in case group has no significant differences compared with control group (all P 0.05). However, we found the genotype of rs1799801 in the control group was significantly different from severe PE group in cases (P 0.05). Moreover, the genotype frequency of rs1800975 and rs1799801in the early-onset PE were significantly different from control group (all P 0.05). These results indicated that the polymorphism of the three SNPs had no significant correlation with risk of PE as a whole. However, we found XPA rs1800975 played a role in the development of early-onset PE, and XPF rs1799801 was associated with severe PE and early-onset PE. So we may need to continue to increase the sample size to clarify their relationship.

Purpose: Preeclampsia (PE) is a common complication during pregnancy that seriously affects the health of both the mother and child due to its insidious onset and delayed diagnosis. Therefore, a retrospective evaluation is necessary for developing a clinical nomogram for predicting prognosis among PE patients. Methods: We developed a nomogram to predict PE prognosis based on 5058 patients recruited from Shandong Province, China between January 2012 and November 2017; then, we externally validated the nomogram in a separate cohort of 1947 patients across China by testing the calibration and discrimination of the nomogram in predicting the delivery time among PE patients. Results: According to forward stepwise linear regression analyses, the time of hypertension onset (TOH) is the most predictive factor in the prognosis of patients with PE, followed by the urine protein, serum creatinine, and alkaline phosphatase levels and ten other clinical parameters. Then, we developed a prognostic visualized nomogram and a risk assessment table to predict the outcome of PE patients and found that PE patients with high scores may have an earlier delivery time; this finding was validated by performing calibration and discrimination analyses, and the agreement between the nomogram predictions and actual delivery times was excellent in both the primary and validation cohorts. In addition, this nomogram could estimate the fetal prognosis. Conclusion: We established and validated a nomogram that can predict the prognosis of PE patients. This practical prognostic PE model may be helpful for clinicians in making clinical decisions and providing treatment. Objective: Previous studies have been indicated that catechol-O-methyltransferase gene (COMT) might play a significant role in the development of preeclampsia (PE). Our study aims to investigate the association between polymorphism in COMT with the susceptibility to PE in Chinese Han women. Method: A total of 1028 PE patients and 1399 normal pregnant women were enrolled. We detected the genotyping of COMT Val158Met loci by the TaqMan allelic discrimination real-time PCR . Results: No significant difference in the genotypic and allelic distribution was found between the two groups (genotype: X2 = 0.583, p = 0.747; allele:X2 = 0.526, p = 0.468). Conclusion: The COMT Val158Met polymorphism might not be associated with PE in Chinese women.
