Abstract

Soft robots promise improved safety and capability over rigid robots when deployed in complex, delicate, and dynamic environments. However, the infinite degrees of freedom and highly nonlinear dynamics of these systems severely complicate their modeling and control. As a step toward addressing this open challenge, we apply the data-driven, Hankel Dynamic Mode Decomposition (HDMD) with time delay observables to the model identification of a highly inertial, helical soft robotic arm with a high number of underactuated degrees of freedom. The resulting model is linear and hence amenable to control via a Linear Quadratic Regulator (LQR). Using our test bed device, a dynamic, lightweight pneumatic fabric arm with an inertial mass at the tip, we show that the combination of HDMD and LQR allows us to command our robot to achieve arbitrary poses using only open loop control. We further show that Koopman spectral analysis gives us a dimensionally reduced basis of modes which decreases computational complexity without sacrificing predictive power., Comment: Submitted to IEEE International Conference on Robotics and Automation, 2021

5. Upstream CtrA-binding sites both induce and repress pilin gene expression in Caulobacter crescentus.

Abstract

Pili are bacterial surface structures important for surface adhesion. In the alphaproteobacterium Caulobacter crescentus, the global regulator CtrA activates transcription of roughly 100 genes, including pilA which codes for the pilin monomer that makes up the pilus filament. While most CtrA-activated promoters have a single CtrA-binding site at the − 35 position and are induced at the early to mid-predivisional cell stage, the pilA promoter has 3 additional upstream CtrA-binding sites and it is induced at the late predivisional cell stage. Reporter constructs where these additional sites were disrupted by deletion or mutation led to increased activity compared to the WT promoter. In synchronized cultures, these mutations caused pilA transcription to occur approximately 20 min earlier than WT. The results suggested that the site overlapping the − 35 position drives pilA gene expression while the other upstream CtrA-binding sites serve to reduce and delay expression. EMSA experiments showed that the − 35 Site has lower affinity for CtrA∼P compared to the other sites, suggesting binding site affinity may be involved in the delay mechanism. Mutating the upstream inhibitory CtrA-binding sites in the pilA promoter caused significantly higher numbers of pre-divisional cells to express pili, and phage survival assays showed this strain to be significantly more sensitive to pilitropic phage. These results suggest that pilA regulation evolved in C. crescentus to provide an ecological advantage within the context of phage infection. [ABSTRACT FROM AUTHOR]

7. Design of a Soft Continuum Sensor Utilizing Macrobend Losses of Optical Fibers

Abstract

When compared with traditional robotic manipulators that use discreterigid links and discrete joints, the sensing of soft robotic manipulators isinaccurate and their control is clumsy. This is because that the orientationof a traditional robotic arm can be known sufficiently by measuring relativerotation and translation of the links and joints subject to some initialcondition. This could be done by implementing rotary encoders at everyjoint in a traditional rigid manipulator. Because of their continuum nature,a soft robotic arm’s orientation can only be known exactly if an infinitenumber were to be used. Therfore, the orientation of soft robotic manipulatorscan only be practically described when applying a set of simplifyingassumptions such as piecewise-constant curvature. There are many waysto obtain information that can be used to describe the curvature of a softrobot; Motion capture is a prevailing technique in research but require thatthe robot is confined to spaces where motion capture can be used. Since thisis not possible in many promising applications of soft robotics, including butnot limited to minimally invasive surgery and disaster site navigation, manyare looking to other ways to make robots capable of proprioception withoutusing external sensors. One option is incorporating fiber optic sensing intosoft robots. Fiber is flexible and a staggering amount of information abouta system can be determined by reading changes in wavelength, phase shift,etc, which makes it an excellent solution for soft systems. The problemwith incorporating fiber optics into soft robotics is that equipment andtechnologies such as optical spectrum analyzers, interrogators, fiber Bragggratings are prohibitively expensive. Some work has been done showcasingthat the optical intensity transmitted through a fiber can be correlated withthe deformation that the fiber is experiencing (also known as macrobendloss or fiber optic intensity modulation), which can be read with muchmore accessible equipment. While promising, existing research only usesthis technique to measure axial strain. We propose extending the methodto determine arbitrary shapes of a continuum arm. We believe the powerof data-driven methods will allow us to glean enough information frombending loss to achieve shape reconstruction currently only possible withhigh-cost optical spectrum analyzers. Specifically, this work aims to use adata-driven method to link the pose of a flexible arm with the intensityoutputs of a number of fiber optic sensors attached to its body, using motioncapture as ground truth. This work serves as the groundwork necessary forestablishing a new low-cost optical sensing technique for soft robotics thatcould potentially allow important advancements in sensing and thus control.

11. Mental Health and Behavioral Problems of Youth in the Child Welfare System: Residential Treatment Centers Compared to Therapeutic Foster Care in the Odyssey Project Population

Abstract

This is the first multisite, prospective study of behavioral and mental health disorders of youth in residential treatment centers (RTC) and therapeutic foster care (TFC), and the first study to compare the two. This study addressed two questions in a sample of 22 agencies in 13 states: (1) how prevalent were emotional and behavioral disorders in the youth admitted to RTCs and TFC?, and (2) were the youth in RTCs significantly more likely to be disturbed than youth served in TFCs? Data were drawn from the Time 1 phase of the longitudinal national "Odyssey Project" developed by the Child Welfare League of America (1995). Measures included an extensive child and family characteristics form (CFC) and the Child Behavior Checklist (CBCL). The results revealed extremely high levels of behavioral and mental health disorders in the sample as a whole, well above the norms for a non-child welfare population. The prevalence of disorder in the RTC population was substantially greater than in the TFC population. (Contains 3 tables and 4 footnotes.)

12. Youth Characteristics Associated with Behavioral and Mental Health Problems during the Transition to Residential Treatment Centers: The Odyssey Project Population

Abstract

This study aimed to determine what youth characteristics were associated with emotional and behavioral problems exhibited within the first three months of placement in residential treatment centers (RTCs) in a sample of youth from 20 agencies in 13 states. Two primary research questions were addressed: 1) What characteristics were associated with behavior during the transition to care? 2) Were the characteristics associated with behavior during the transition the same for boys and girls? Data were drawn from the Time 1 phase of the longitudinal national Odyssey Project dataset developed by the Child Welfare League of America. Measures included an extensive child and family characteristics (CFC) form and the Achenbach Child Behavior Checklist (CBCL). The results revealed significant gender-specific patterns of associations between youth characteristics and behavior exhibited during the transition to RTC placement. Notably, a sexual abuse history was associated with Externalizing for girls and Internalizing for boys and entering on psychotropic medication was associated with Internalizing for girls and boys and Externalizing for boys only. 13. Sexual Abuse Histories of Youth in Child Welfare Residential Treatment Centers: Analysis of the Odyssey Project Population

Abstract

This multi-site examination of sexual abuse histories of youth in residential treatment centers asked, for the sample as a whole and by youth's gender: (a) How many perpetrators did each youth have? (b) What was the gender of the perpetrator? (c) What proportion of youth was abused by family members? (d) What proportion of youth was abused in a child welfare setting? And, (e) what proportion of sexually abused youth were also victims of physical abuse and/or neglect? Results revealed that males were more likely to have one perpetrator, a female perpetrator, and to have been abused in a child welfare setting and less likely be abused by a family member. Implications for refining programs and practices are discussed. (Contains 7 tables.)

15. A First Look at the Need for Enhanced Support Services for Kinship Caregivers.

Abstract

This study used focus group discussions with formal kinship caregivers in Maryland to examine the need for support services. Findings revealed that kinship caregivers were committed to providing safety and stability for the children placed with them and suggested that caregiving can be a significant adjustment for many kinship caregivers. Findings suggest that expanded support services are needed to enhance kinship caregivers' relationship with the child welfare agency. (Author/KB)

20. Child Welfare League of America 1997 Salary Study.

Abstract

This report details a study of trends in the salaries of Chief Executive Officers (CEOs) and other personnel at member agencies of the Child Welfare League of America. Responding to the study's mail survey were 377 out of 780 member agencies (48 percent return) and 327 out of 640 direct service CWLA agencies (51 percent return), including state and local public agencies, voluntary agencies, and supporting advocates. The bulk of the report is comprised of tables and reports of findings. Most of the tables delineate salaries by agency type, with CEO salaries also delineated by CWLA membership region, selected states, agency income, educational level, gender, and race. Findings indicated that CEOs of voluntary child welfare agencies had a median salary of $78,750, an increase of 5 percent from 1995. Almost half the full-time position categories in voluntary agencies had salary increases less than the inflation rate for 1995-96. Salaries for Supervisor and Residential Supervisor, unlike most other positions, have not kept pace with inflation over the past 16 years. All day care positions experienced increases less than inflation or decreases in median salary. CEOs of child welfare agencies did not earn as much as CEOs from other nonprofit organizations. Salaries were higher in the public than voluntary sector, with turnover highest among voluntary service providers. Salaries were lowest in the southern region of the country. Positions lost due to agency budget cuts were low and were offset by staffing increases. The fringe benefit cost in public agencies was higher than in voluntary agencies. (KB)

This report provides a comprehensive portrait of America's most vulnerable children and families, gathered as a quick reference to national and state data on the U.S. child welfare system. Data were derived from the Child Welfare League of America's 1996 State Survey of Child Welfare and from U.S. Government agencies and other national sources. The report documents the differences and disparities among state data collection, providing a useful baseline for measuring the effects of child welfare service experiments and cutbacks. The figures and tables presented in the report provide child welfare data based on rates for children in each state, comparisons over time to show child well-being and related trend data, and state child welfare budgets and related financial information. Designed to provide a basis for sound child welfare policy decisions, this book presents current state and national data, as well as data from many cities and counties, on child maltreatment. The six chapters in the book present statistics on: (1) child abuse and neglect, including child abuse fatalities; (2) out-of-home care; (3) adoption; (4) finance and administration of child welfare organizations; (5) risk factors; and (6) possibilities for prevention. Chapter one draws attention to the use of different definitions of neglect and abuse, and investigation procedures by states. Chapter two describes various forms of out-of-home care. Chapter three discusses different types of adoption, all of which provide permanence for children. Chapter four illustrates the expenditures incurred by different states to safeguard and maintain child well-being. The fifth chapter focuses on the condition of poverty as a factor in child maltreatment. The last chapter considers current strategies that are used to prevent child abuse and neglect, as well as the issue of corporal punishment. Most of the data are presented in graphic formats and tables, which are meant to illustrate the wide variations in how states are addressing child abuse and neglect, not to judge the effectiveness of one state compared to another. 29. Conceptualization of the Holobiont Paradigm as It Pertains to Corals

Abstract

Corals’ obligate association with unicellular dinoflagellates, family Symbiodiniaceae form the foundation of coral reefs. For nearly a century, researchers have delved into understanding the coral-algal mutualism from multiple levels of resolution and perspectives, and the questions and scope have evolved with each iteration of new techniques. Advances in genetic technologies not only aided in distinguishing between the multitude of Symbiodiniaceae but also illuminated the existence and diversity of other organisms constituting the coral microbiome. The coral therefore is a meta-organism, often referred to as the coral holobiont. In this review, we address the importance of including a holistic perspective to understanding the coral holobiont. We also discuss the ramifications of how different genotypic combinations of the coral consortium affect the holobiont entity. We highlight the paucity of data on most of the coral microbiome. Using Symbiodiniaceae data, we present evidence that the holobiont properties are not necessarily the sum of its parts. We then discuss the consequences of the holobiont attributes to the fitness of the holobiont and the myriad of organisms that contribute to it. Considering the complexity of host-symbiont genotypic combinations will aid in our understanding of coral resilience, robustness, acclimation, and/or adaptation in the face of environmental change and increasing perturbations.

Abstract

We report first results on elliptic flow of identified particles at midrapidity in Au+Au collisions at sqrt[sNN] = 130 GeV using the STAR TPC at RHIC. The elliptic flow as a function of transverse momentum and centrality differs significantly for particles of different masses. This dependence can be accounted for in hydrodynamic models, indicating that the system created shows a behavior consistent with collective hydrodynamical flow. The fit to the data with a simple model gives information on the temperature and flow velocities at freeze-out.

Abstract

Microcinematography was used to examine fruiting body development of Myxococcus xanthus. Wild-type cells progress through three distinct phases: a quiescent phase with some motility but little aggregation (0 to 8 h), a period of vigorous motility leading to raised fruiting bodies (8 to 16 h), and a period of maturation during which sporulation is initiated (16 to 48 h). Fruiting bodies are extended vertically in a series of tiers, each involving the addition of a cell monolayer on top of the uppermost layer. A pilA (MXAN_5783) mutant produced less extracellular matrix material and thus allowed closer examination of tiered aggregate formation. A csgA (MXAN_1294) mutant exhibited no quiescent phase, aberrant aggregation in phase 2, and disintegration of the fruiting bodies in the third phase.

Abstract

Fruiting body formation of Myxococcus xanthus, like biofilm formation of many other organisms, involves the production of an extracellular matrix (ECM). While the polysaccharide component has been studied, the protein component has been largely unexplored. Proteins associated with the ECM were solubilized from purified ECM by boiling with sodium dodecyl sulfate and were identified by liquid chromatography-tandem mass spectrometry of tryptic fragments. The ECM is enriched in proteins of novel function; putative functions were assigned for only 5 of the 21 proteins. Thirteen putative ECM proteins had lipoprotein secretion signals. The genes for many ECM proteins were disrupted in the wild-type (WT), fibA, and pilA backgrounds. Disruption of the MXAN4860 gene had no effect in the WT or fibA baekground but in the pilA background resulted in a 24-h delay in aggregation and sporulation compared to its parent. The results of this study show that the M. xanthus ECM proteome is diverse and novel.

Abstract

The DivJ-DivK-PleC signaling system ofCaulobacter crescentusis a signaling network that regulates polar development and the cell cycle. This system is conserved in related bacteria, including the sister genusBrevundimonas. Previous studies had shown unexpected phenotypic differences between theC. crescentus divKmutant and the analogous mutant ofBrevundimonas subvibrioides, but further characterization was not performed. Here, phenotypic assays analyzing motility, adhesion, and pilus production (the latter characterized by a newly discovered bacteriophage) revealed thatdivJandpleCmutants have mostly similar phenotypes as theirC. crescentushomologs, butdivKmutants maintain largely opposite phenotypes than expected. Suppressor mutations of theB. subvibrioides divKmotility defect were involved in cyclic-di-GMP (c-di-GMP) signaling, including the diguanylate cyclasedgcB, andcleDwhich is hypothesized to affect flagellar function in a c-di-GMP dependent fashion. However, the screen did not identify the diguanylate cyclasepleD.Disruption ofpleDinB. subvibrioidescaused hypermotility in wild-type, but not in thedivKbackground. Analysis of c-di-GMP levels in these strains revealed incongruities between c-di-GMP levels and displayed phenotypes with a notable result that suppressor mutations altered phenotypes but had little impact on c-di-GMP levels in thedivKbackground. Conversely, when c-di-GMP levels were artificially manipulated, alterations of c-di-GMP levels in thedivKstrain had minimal impact on phenotypes. These results suggest that DivK performs a critical function in the integration of c-di-GMP signaling into theB. subvibrioidescell cycle.ImportanceCyclic-di-GMP signaling is one of the most broadly conserved signaling systems in bacteria, but there is little understanding of how this system directly affects the physiology of the organism. InC. crescentus, c-di-GMP has been integrated into the developmental cell cycle, but there is increasing evidence that environmental factors can impact this system as well. The research presented here suggests that developmental signaling could impact physiological Processes in c-di-GMP dependent and independent ways. This hints that the integration of these signaling networks could be more complex than previously hypothesized, which could have a bearing on the larger field of c-di-GMP signaling. In addition, this work further examines how much models developed in one organism can be extrapolated to related organisms.
