Abstract Background Cyclic β-1,2-glucans (CβG) are bacterial cyclic homopolysaccharides with interesting biotechnological applications. These ring-shaped molecules have a hydrophilic surface that confers high solubility and a hydrophobic cavity able to include poorly soluble molecules. Several studies demonstrate that CβG and many derivatives can be applied in drug solubilization and stabilization, enantiomer separation, catalysis, synthesis of nanomaterials and even as immunomodulators, suggesting these molecules have great potential for their industrial and commercial exploitation. Nowadays, there is no method to produce CβG by chemical synthesis and bacteria that synthesize them are slow-growing or even pathogenic, which makes the scaling up of the process difficult and expensive. Therefore, scalable production and purification methods are needed to afford the demand and expand the repertoire of applications of CβG. Results We present the production of CβG in specially designed E. coli strains by means of the deletion of intrinsic polysaccharide biosynthetic genes and the heterologous expression of enzymes involved in CβG synthesis, transport and succinilation. These strains produce different types of CβG: unsubstituted CβG, anionic CβG and CβG of high size. Unsubstituted CβG with a degree of polymerization of 17 to 24 glucoses were produced and secreted to the culture medium by one of the strains. Through high cell density culture (HCDC) of that strain we were able to produce 4,5 g of pure unsubstituted CβG /L in culture medium within 48 h culture. Conclusions We have developed a new recombinant bacterial system for the synthesis of cyclic β-1,2-glucans, expanding the use of bacteria as a platform for the production of new polysaccharides with biotechnological applications. This new approach allowed us to produce CβG in E. coli with high yields and the highest volumetric productivity reported to date. We expect this new highly scalable system facilitates CβG availability for further research and the widespread use of these promising molecules across many application fields.

Modern mass spectrometry, including electrospray and MALDI, is applied for analysis and structure elucidation of carbohydrates. Cyclic oligosaccharides isolated from different sources (bacteria and plants) have been known for decades and some of them (cyclodextrins and their derivatives) are widely used in drug design, as food additives, in the construction of nanomaterials, etc. The peculiarities of the first- and second-order mass spectra of cyclic oligosaccharides (natural, synthetic and their derivatives and modifications: cyclodextrins, cycloglucans, cyclofructans, cyclooligoglucosamines, etc.) are discussed in this minireview.

Diverse flavonoids are abundant in dietary food constituents and possess useful biological activities. However, some flavonoids have limited bioavailability due to their low solubility in water. As an important approach to enhance aqueous solubility, inclusion of hydrophobic guest molecules in hydrophilic hosts such as cyclic glucans has been used. This review summarizes applications of β-cyclodextrin, synthetic β-cyclodextrin derivatives, and newly synthesized derivatives of cyclosophoraoses as complexing agents to enhance the bioavailability of flavonoids such as baicalein, kaempferol, and naphthoflavones. The article focuses on a study which demonstrated the preparation of cyclosophoraoses (Cys)-immobilized chiral stationary phase (CSP) in high-performance liquid chromatography (HPLC). Discussed is a comparison between cyclodextrins (CD) and Cys. Information are also offered about flavanones, a class of flavonoids. Also offered is an illustration of a Cys-CSP column scheme.

Abstract: Methanolysis of four ethyl esters, N-acetyl-l-phenylalanine ethyl ester, N-acetyl-l-tyrosine ethyl ester, N-acetyl-l-tryptophan ethyl ester, and ethyl phenylacetate was catalyzed by a mixture of microbial cyclooligosaccharides termed cyclosophoraoses isolated from Rhizobium meliloti. Cyclosophoraoses [cyclic-(1→2)-β-d-glucans, collectively ‘Cys’] are a mixture of large-ring molecules consisting of various numbers of glucose residues (17–27) linked by β-(1→2)-glycosidic bonds. Cys as a catalytic carbohydrate enhanced the methanolysis about 233-fold for N-acetyl-l-tyrosine ethyl ester in comparison with a control. The effect of dry organic solvents on the methanolysis of N-acetyl-l-tyrosine ethyl ester was investigated by high-performance liquid chromatography (HPLC), and it was found that the rate enhancement correlated closely with the hydrophobicity of the solvent. A novel catalytic methanolysis can be induced by a natural cyclooligosaccharide, a cyclosophoraose (cyclic-(1 → 2)-β-d-glucan, Cys), which is a member of a family of unbranched cyclooligosaccharides produced as intra- or extraoligosaccharides by soil microorganisms of the genus, Rhizobium. Cys catalyzed the methanolysis for 5(4H)-oxazolones and various phospholipids. Cys enhanced the methanolysis reaction about 9200-fold for a benzylidene oxazolone or 250-fold for dipalmitoylphosphatidylcholine comparing with control. In this study, we describe that natural cyclosophoraoses isolated from the Rhizobium species function as catalytic carbohydrates for the methanolysis. [Copyright &y& Elsevier]

Cyclosophoraoses, cyclic β-(1→2)-d-glucans produced by Rhizobium meliloti 2011, were used as a novel chiral additive for the separation of terbutaline, amethopterin, thyroxine and N-acetylphenylalanine enantiomers in aqueous capillary electrophoresis (CE). Enantioseparation took place in the normal- or reversed-polarity mode when a high concentration of neutral (60 mM) or anionic (40 mM) cyclosophoraoses was added to the background electrolyte (BGE). Cyclosophoraoses (cyclic-(1→2)-β-d-glucans) produced by Rhizobium meliloti were used as a novel chiral NMR solvating agent. 13C NMR spectroscopic analysis as an enantiodiscriminating tool was carried out where NMR signal splittings were observed on the interactions of cyclosophoraoses with the enantiomers of N-acetylphenylalanine, catechin and propranolol. The 13C chemical shifts of cyclosophoraoses induced by the enantiomeric interactions predominantly occurred at the C-1 and C-2 carbons associated with the -glycosidic linkage.
