Abstract

Nerve damage is a prevalent and debilitating condition with limited treatment options. Recent years have seen an increased incidence of neural damage due to factors such as aging populations and traumatic brain injuries. Addressing the urgent need for effective therapies, this study explores the controlled delivery of mesenchymal stem cells (MSCs) secretome, a complex mixture of bioactive factors, which is currently being investigated for its potential in nerve regeneration. The secretome offers significant advantages over stem cells themselves, as it can be more easily characterized and controlled, enabling precise regulation of therapeutic interventions. However, the challenge lies in delivering the secretome specifically to the target anatomical region. To overcome this limitation, we propose a novel approach utilizing near-infrared (NIR) radiation-responsive bioprinted alginate-graphene oxide (AGO) microbeads. Graphene oxide (GO) is a highly biocompatible material with unique properties, including NIR responsiveness, enabling controlled release of therapeutic agents upon NIR exposure. We hypothesized that AGO microbeads could encapsulate MSCs secretome and release it in a controlled manner using NIR radiation. To investigate our hypothesis, controlled damage was induced to hippocampal neurons, and MSCs secretome was encapsulated within AGO microbeads. Subsequently, NIR radiation was applied to trigger the release of the secretome. We compared the efficacy of MSCs secretome with that of astrocytes, which also possess nerve growth and proliferation-promoting capabilities. Our findings demonstrated that the controlled release of MSCs secretome from AGO microbeads through non-invasive NIR radiation significantly promoted the proliferation and regeneration of neurons following nerve injury. Abstract

Glioblastoma is the most aggressive and lethal brain cancer. Current treatments involve surgical resection, radiotherapy and chemotherapy. However, the life expectancy of patients with this disease remains short and chemotherapy leads to severe adverse effects. Furthermore, the presence of the blood-brain barrier (BBB) makes it difficult for drugs to effectively reach the brain. A promising strategy lies in the use of graphene quantum dots (GQDs), which are light-responsive graphene nanoparticles that have shown the capability of crossing the BBB. Here we investigate the effect of GQDs on U87 human glioblastoma cells and primary cortical neurons. Non-functionalized GQDs (NF-GQDs) demonstrated high biocompatibility, while dimethylformamide-functionalized GQDs (DMF-GQDs) showed a toxic effect on both cell lines. The combination of GQDs and the chemotherapeutic agent doxorubicin (Dox) was tested. GQDs exerted a synergistic increase in the efficacy of chemotherapy treatment, specifically on U87 cells. The mechanism underlying this synergy was investigated, and it was found that GQDs can alter membrane permeability in a manner dependent on the surface chemistry, facilitating the uptake of Dox inside U87 cells, but not on cortical neurons. Therefore, experimental evidence indicates that GQDs could be used in a combined therapy against brain cancer, strongly increasing the efficacy of chemotherapy and, at the same time, reducing its dose requirement along with its side effects, thereby improving the life quality of patients.

Abstract

Recent evidence has shown that graphene quantum dots (GQDs) are capable of crossing the blood–brain barrier, the barrier that reduces cancer therapy efficacy. Here, we tested three alternative GQDs’ surface chemistries on two neural lineages (glioblastoma cells and mouse cortical neurons). We showed that surface chemistry modulates GQDs’ biocompatibility. When used in combination with the chemotherapeutic drug doxorubicin, GDQs exerted a synergistic effect on tumor cells, but not on neurons. This appears to be mediated by the modification of membrane permeability induced by the surface of GQDs. Our findings highlight that GQDs can be adopted as a suitable delivery and therapeutic strategy for the treatment of glioblastoma, by both directly destabilizing the cell membrane and indirectly increasing the efficacy of chemotherapeutic drugs.

Abstract

High-fat diet (HFD) and metabolic diseases cause detrimental effects on hippocampal synaptic plasticity, learning, and memory through molecular mechanisms still poorly understood. Here, we demonstrate that HFD increases palmitic acid deposition in the hippocampus and induces hippocampal insulin resistance leading to FoxO3a-mediated overexpression of the palmitoyltransferase zDHHC3. The excess of palmitic acid along with higher zDHHC3 levels causes hyper-palmitoylation of AMPA glutamate receptor subunit GluA1, hindering its activity-dependent trafficking to the plasma membrane. Accordingly, AMPAR current amplitudes and, more importantly, their potentiation underlying synaptic plasticity were inhibited, as well as hippocampal-dependent memory. Hippocampus-specific silencing of Zdhhc3 and, interestingly enough, intranasal injection of the palmitoyltransferase inhibitor, 2-bromopalmitate, counteract GluA1 hyper-palmitoylation and restore synaptic plasticity and memory in HFD mice. Our data reveal a key role of FoxO3a/Zdhhc3/GluA1 axis in the HFD-dependent impairment of cognitive function and identify a novel mechanism underlying the cross talk between metabolic and cognitive disorders.

Abstract

Impairment of adult hippocampal neurogenesis is an early event in Alzheimer’s disease (AD), playing a crucial role in cognitive dysfunction associated with this pathology. However, the mechanisms underlying defective neurogenesis in AD are still unclear. Recently, the nucleoporin Nup153 has been described as a new epigenetic determinant of adult neural stem cell (NSC) maintenance and fate. Here we investigated whether Nup153 dysfunction could affect the plasticity of NSCs in AD. Nup153 expression was strongly reduced in AD-NSCs, as well as its interaction with the transcription factor Sox2, a master regulator of NSC stemness and their neuronal differentiation. Similar Nup153 reduction was also observed in WT-NSCs treated with amyloid-beta (Abeta) or stimulated with a nitric oxide donor. Accordingly, AD-NSCs treated with either a gamma-secretase inhibitor or antioxidant compounds showed higher Nup153 levels suggesting that both nitrosative stress and Abetaaccumulation affect Nup153 expression. Of note, restoration of Nup153 levels in AD-NSCs promoted their proliferation, as assessed by BrdU incorporation, neurosphere assay and stemness gene expression analysis. Nup153 overexpression also recovered AD-NSC response to differentiation, increasing the expression of pro-neuronal genes, the percentage of cells positive for neuronal markers and the acquisition of a more mature neuronal phenotype. Electrophysiological recordings revealed that neurons differentiated from Nup153-transfected AD-NSCs displayed higher Na+ current density, comparable to those deriving from WT-NSCs. Our data uncover a novel role for Nup153 in NSCs from animal model of AD and point to Nup153 as potential target to restore physiological NSC behavior and fate in neurodegenerative diseases.

Abstract

Resilience and vulnerability to neuropsychiatric disorders are linked to molecular changes underlying excitability that are still poorly understood. Here, we identify glycogen-synthase kinase 3b (GSK3b) and voltage-gated Na+ channel Nav1.6 as regulators of neuroplasticity induced by environmentally enriched (EC) or isolated (IC) conditions—models for resilience and vulnerability. Transcriptomic studies in the nucleus accumbens from EC and IC rats predicted low levels of GSK3b and SCN8A mRNA as a protective phenotype associated with reduced excitability in medium spiny neurons (MSNs). In vivo genetic manipulations demonstrate that GSK3b and Nav1.6 are molecular determinants of MSN excitability and that silencing of GSK3b prevents maladaptive plasticity of IC MSNs. In vitro studies reveal direct interaction of GSK3b with Nav1.6 and phosphorylation at Nav1.6T1936 by GSK3b. A GSK3b-Nav1.6T1936 competing peptide reduces MSNs excitability in IC, but not EC rats. These results identify GSK3b regulation of Nav1.6 as a biosignature ofMSNs maladaptive plasticity.

Abstract

Amyloid beta-peptide (A beta) plays a fundamental role in the pathogenesis of Alzheimer's disease. We recently reported that the redox state of the methionine residue in position 35 of amyloid beta-peptide (A beta) 1-42 (Met35) strongly affects the peptide's ability to trigger apoptosis and is thus a major determinant of its neurotoxicity. Dysregulation of intracellular Ca(2+) homeostasis resulting in the activation of pro-apoptotic pathways has been proposed as a mechanism underlying A beta toxicity. Therefore, we investigated correlations between the Met35 redox state, A beta toxicity, and altered intracellular Ca(2+) signaling in human neuroblastoma IMR32 cells. Cells incubated for 6-24 h with 10 mu M A beta 1-42 exhibited significantly increased KCl-induced Ca(2+) transient amplitudes and resting free Ca(2+) concentrations. Nifedipine-sensitive Ca(2+) current densities and Ca(v)1 channel expression were markedly enhanced by A beta 1-42. None of these effects were observed when cells were exposed to A beta containing oxidized Met35 (A beta 1-42(Met35-Ox)). Cell pre-treatment with the intracellular Ca(2+) chelator 1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid acetoxymethyl ester (1 mu M) or the Ca(v)1 channel blocker nifedipine (5 mu M) significantly attenuated A beta 1-42-induced apoptosis but had no effect on A beta 1-42(Met35-Ox) toxicity. Collectively, these data suggest that reduced Met35 plays a critical role in A beta 1-42 toxicity by rendering the peptide capable of disrupting intracellular Ca(2+) homeostasis and thereby provoking apoptotic cell death.

Abstract

“Il ruolo dell’insegnante tra metodo e qualità dell’esperienza scolastica” intitola un percorso di ricerca-formazione realizzato tra aprile e ottobre 2011 con gli insegnanti delle scuole dell’infanzia, primarie e secondarie di primo grado del Comune di Cinisello Balsamo (MI). L’obiettivo della ricerca ha riguardato la possibilità di ricostruire, a partire da situazioni scolastiche concrete, in cosa consistesse il “modo di fare scuola” agito quotidianamente da ogni insegnante; come esso incidesse sulla qualità dell’esperienza che allievi e insegnanti vissuta a scuola e quindi come, a partire dalla pratica, si potesse ridefinire ciò che qualifica la professionalità dell’insegnante oggi, rispetto a situazioni cosiddette “di disagio”. Dal punto di vista pedagogico, la ricerca presuppone la specificità dell’esperienza educativa, sottolineandone da un lato la complessità intrinseca, dall’altro la problematicità delle connessioni con l’educazione informale. Inserita nell’ambito delle ricerche pedagogiche empiriche di tipo qualitativo, idiografico e intensivo, la ricerca ha sviluppato un percorso strutturato in gruppo. Riferendosi all’approccio di Clinica della Formazione, è stata animata dalla preoccupazione di consentire ai partecipanti di pervenire a una propria consapevolezza didattico-metodologica e di individuare i propri riferimenti deontologici a partire da una riflessione sull’esperienza agita. Tra i risultati, è stata evidenziata la presenza di una pratica didattico-educativa esperta e in divenire, che, se riesce a fronteggiare situazioni difficili, tuttavia non viene legittimata come “modalità di fare scuola” nel confronto con modelli di insegnamento tradizionali, a cui, pur riconoscendo l’anacronismo rispetto alla situazione attuale, insegnanti e dispositivi scolastici sembrano comunque riferirsi. Dopo aver esplicitato i presupposti epistemologici e metodologici, le domande e le finalità della ricerca, il contributo ne mostra i risultati. Abstract

We report the development and optimization of reagents for in-solution, hybridization-based capture of the mouse exome. By validating this approach in a multiple inbred strains and in novel mutant strains, we show that whole exome sequencing is a robust approach for discovery of putative mutations, irrespective of strain background. We found strong candidate mutations for the majority of mutant exomes sequenced, including new models of orofacial clefting, urogenital dysmorphology, kyphosis and autoimmune hepatitis.

Abstract

A new procedure for the encapsulation of non-diffusible drugs into human erythrocytes was developed. With as little as 50 ml of blood and by using a new apparatus, it was possible to encapsulate a variety of biologically active compounds into erythrocytes in 2 h at room temperature and under blood-banking conditions, The process, which is based on two sequential hypotonic dilutions of washed red cells followed by concentration with a haemofilter and resealing of red cells, allows a 35-50% cell recovery and approx, 30% encapsulation of added drugs. The resulting processed erythrocytes have a normal survival in vivo and can be modified further, with the same apparatus, to; Increase their recognition by tissue macrophages to perform as a drug-targeting system. The new equipment designed and built for this procedure was named 'Red Cell Loader'.

43. Biochemical properties of two protein kinases involved in disease resistance signaling in tomato.

Abstract

In tomato plants, resistance to bacterial speck disease is mediated by a phosphorylation cascade, which is triggered by the specific recognition between the plant serine/threonine protein kinase Pto and the bacterial AvrPto protein. In the present study, we investigated in vitro biochemical properties of Pto, which appears to function as an intracellular receptor for the AvrPto signal molecule. Pto and its downstream effector Pti1, which is also a serine/threonine protein kinase, were expressed in Escherichia coli as maltose-binding protein and glutathione S-transferase fusion proteins, respectively. The two kinases each autophosphorylated at multiple sites as determined by phosphopeptide mapping. In addition, Pto and Pti1 autophosphorylation occurred via an intramolecular mechanism, as their specific activity was not affected by their molar concentration in the assay. Moreover, an active glutathione S-transferase-Pto fusion failed to phosphorylate an inactive maltose-binding protein-Pto(K69Q) fusion excluding an intermolecular mechanism of phosphorylation for Pto. Pti1 phosphorylation by Pto was also characterized and found to occur with a Km of 4.1 microM at sites similar to those autophosphorylated by Pti1. Pto and the product of the recessive allele pto phosphorylated Pti1 at similar sites, as observed by phosphopeptide mapping. This suggests that the inability of the kinase pto to confer resistance to bacterial speck disease in tomato is not caused by altered recognition specificity for Pti1 phosphorylation sites.

Oltre alle questioni storiografiche il saggio motiva le ragioni dell'intero volume sul piano del recupero della memoria scolastica collettiva, nel quadro della valorizzazione del patrimonio educativo e scolastico del passato, a cui le biografie magistrali possono contribuire.

© 2019 Elsevier Ltd The dopamine D 3 receptor (D 3 R), in the nucleus accumbens (NAc), plays an important role in alcohol reward mechanisms. The major neuronal type within the NAc is the GABAergic medium spiny neuron (MSN), whose activity is regulated by dopaminergic inputs. We previously reported that genetic deletion or pharmacological blockade of D 3 R increases GABA A α6 subunit in the ventral striatum. Here we tested the hypothesis that D 3 R-dependent changes in GABA A α6 subunit in the NAc affect voluntary alcohol intake, by influencing the inhibitory transmission of MSNs. We performed in vivo and ex vivo experiments in D 3 R knockout (D 3 R −/− ) mice and wild type littermates (D 3 R +/+ ). Ro 15-4513, a high affinity α6-GABA A ligand was used to study α6 activity. At baseline, NAc α6 expression was negligible in D 3 R +/+ , whereas it was robust in D 3 R −/− ; other relevant GABA A subunits were not changed. In situ hybridization and qPCR confirmed α6 subunit mRNA expression especially in the NAc. In the drinking-in-the-dark paradigm, systemic administration of Ro 15-4513 inhibited alcohol intake in D 3 R +/+ , but increased it in D 3 R −/− ; this was confirmed by intra-NAc administration of Ro 15-4513 and furosemide, a selective α6-GABA A antagonist. Whole-cell patch-clamp showed peak amplitudes of miniature inhibitory postsynaptic currents in NAc medium spiny neurons higher in D 3 R −/− compared to D 3 R +/+ ; Ro 15-4513 reduced the peak amplitude in the NAc of D 3 R −/− , but not in D 3 R +/+ . We conclude that D 3 R-dependent enhanced expression of α6 GABA A subunit inhibits voluntary alcohol intake by increasing GABA inhibition in the NAc.

Abstract

Gli anni recenti hanno visto un’evoluzione nella pratica degli sport outdoor. Infatti sia in ambito ricreativo che competitivo sono emerse nuove discipline e tipologie di manifestazioni. In questo capitolo, partendo col definire cosa si intende con sport outdoor, verranno analizzati gli sport tradizionali, propriamente detti (istituzionalizzati) e genericamente intesi (come pratica saltuaria). Successivamente verranno prese in esame le pratiche motorie e sportive outdoor sviluppatesi più di recente, come i playground, i lifestyle sport e gli adventure sport. Accomunati dall’intento di distaccarsi dagli sport tradizionali, questi ultimi vengono presentati attraverso l’analisi dei numerosi elementi comuni che li caratterizzano, ed attraverso i quali hanno raggiunto una sempre maggiore popolarità. Verrà inoltre considerato come, partendo dalla semplice camminata di alcune ore fino ad arrivare alle gare epiche, l’interesse per queste attività è oggetto di studio e promozione nei più svariati settori, a cominciare dagli studi sociologici e psicologici per passare a quelli di industrie che realizzano materiali tecnici, fino alle campagne pubblicitarie finalizzate alla promozione di aree turistiche.

Abstract

This paper focuses on the figure of Franco Salvo, a teacher of history and philosophy at the classical high school Umberto I in Palermo from 1945 to 1981. He was a master of freedom rights who taught democratically and critically the civic education in a historical perspective. Salvo was a representative figure of the Sicilian school after the Second World War, a master of life. As his students recall, in the 1968 he supported the student movement, forming their consciences for a critical reflection, a responsible freedom, a social and political commitment.

Abstract

The fundamental role of d -serine as coagonist at the n -methyl- d -aspartate receptor (NMDAR), a major glutamate receptor subtype involved in synaptic plasticity, is well documented and experimental evidence indicates now that this d -amino acid is an influential player in the context of psychiatric diseases such as schizophrenia and depression. More recently, a direct link between cocaine addiction, another neuropsychiatric disorder, and d -serine signaling has been proposed by findings that d -serine levels are decreased in the nucleus accumbens of cocaine-treated rats. Such a deficit in d -serine content leads to impairment of NMDAR-dependent synaptic plasticity and locomotor sensitization to cocaine, a behavioral hallmark of cocaine addiction. The d -serine hypothesis for cocaine addiction, here proposed, provides considerable insight into the understanding of the cocaine-induced neuroadaptations in reward-related neuronal circuits and opens new attractive perspectives for therapeutic approaches to treat this addictive state.
