Long noncoding RNA (lncRNA) is emerging as a vital regulator in various tumors. However, the biological function of ZFPM2-antisense RNA 1 (ZFPM2-AS1) in hepatocellular carcinoma (HCC) remains unclear. The present study aims to explore the function and mechanism of ZFPM2-AS1 in hepatocellular carcinoma progression.The ZFPM2-AS1 expression in HCC cells and tissues was measured by quantitative real-time polymerase chain reaction (qRT-PCR). Effects of ZFPM2-AS1 on tumor cell proliferation and invasion were detected by CCK8 assay or EdU assay or matrigel migration assay and Western blot. The Luciferase reporter assay, RNA pulldown assay, qRT-PCR, and Western blot were performed to explore and confirm the interaction between ZFPM2-AS1 and miR-1226-3p and integrin β1 (ITGB1).ZFPM2-AS1 was overexpressed in HCC tissues and cell lines. High levels of ZFPM2-AS1 were correlated with advanced TNM stage, distant metastasis and a poorer overall survival rate. ZFPM2-AS1 knockdown inhibited cell proliferation and invasion. ZFPM2-AS1 could directly bind to and negatively regulate miR-1226-3p expression. Moreover, ITGB1 was identified as a target gene of miR-1226-3p. ITGB1 was found to be directly negatively regulated by miR-1226-3p and indirectly upregulated by ZFPM2-AS1. Rescue assays demonstrated that ZFPM2-AS1 promotes HCC cell proliferation and invasion through modulating miR-1226/ITGB1 axis.ZFPM2-AS1 promotes cell proliferation and migration by regulating miR-1226-3p/ITGB1 axis in HCC.

Modified paraffin wax instead of natural bee wax has been used in the modified research of the automobile polish wax formula. The influence of the quality fraction of self-prepared emulsifier and thickener, as well as the emulsified craft conditions such as emulsified temperature, emulsified time, and agitate speed, on the emulsified wax performance were investigated separately. The results indicated that with the paraffin wax and the modified paraffin wax as the raw material, the quality fraction of the compound emulsifier was 2.1 ~ 2.5 wt% and the quality fraction of the thickener was 2.9 ~ 3.4 wt% at the technical condition of the emulsified temperature 85°C, the emulsified time of 40 min, and the agitate speed of 1000 r/min. Hepatitis C virus (HCV) is one of the major causes of liver inflammation. The aim of this study was to investigate the associations of T-cell immunoglobulin and mucin domain-3 (Tim-3) polymorphisms and the alternate reading frame protein (F protein) with the outcomes of HCV infection. Three single-nucleotide polymorphisms (SNPs; rs10053538, rs12186731, and rs13170556) of Tim-3 were genotyped in this study, which included 203 healthy controls, 558 hepatitis C anti-F-positive patients, and 163 hepatitis C anti-F-negative patients. The conventional methods of slices are commonly used for the analysis of slope stability. When anchor loads are involved, they are often treated as point loads, which may lead to abrupt changes in the normal stress distribution on the potential slip surface. As such abrupt changes are not reasonable and do not reflect reality in the field, an alternative approach based on the limit equilibrium principle is proposed for the evaluation of the stability of anchor-reinforced slopes. With this approach, the normal stress distribution over the slip surface before the application of the anchor (i.e., σ0) is computed by the conventional, rigorous methods of slices, and the normal stress on the slip surface purely induced by the anchor load (i.e., λpσp, where λp is the load factor) is taken as the analytical elastic stress distribution in an infinite wedge approximating the slope geometry, with the anchor load acting on the apex. Then the normal stress on the slip surface for the anchor-reinforced slope is assumed to be the linear combination of these two normal stresses involving two auxiliary unknowns, η1 and η2; that is, σ = η1σ0 + η2λpσp. Simultaneously solving the horizontal force, the vertical force, and the moment equilibrium equations for the sliding body leads to the explicit expression for the factor of safety (Fs)—or the load factor (λp), if the required factor of safety is prescribed. The reasonableness and advantages of the present method in comparison with the conventional procedures are demonstrated with two illustrative examples. The proposed procedure can be readily applied to designs of excavated slopes or remediation of landslides with steel anchors or prestressed cables, as well as with soil nails or geotextile reinforcements.Key words: slopes, factor of safety, anchors, limit equilibrium method.

A concise algorithm is proposed in this paper for the calculation of the factor of safety of a slope using the Morgenstern–Price method. Based on force and moment equilibrium considerations, two expressions are derived for the factor of safety Fs and the scaling factor λ, respectively, both in relatively simple forms. With this algorithm and assumed initial values of Fs and λ, the solutions for Fs and λ are found to converge within a few iterations. Compared to other procedures, the present algorithm possesses the advantages of simplicity and high efficiency in application. It is rather straightforward to implement this algorithm into a computer program.Key words: slope, stability, factor of safety, limit equilibrium method.

A generalised framework is proposed in this paper incorporating almost all of the existing limit equilibrium methods of slices for slope stability analysis with general slip surfaces. The force and moment equilibrium equations are derived in terms of the factor of safety and the initially assumed normal stress distribution over the slip surface, multiplied by a modification function involving two auxiliary unknowns. These equations are then analytically solved to yield explicit expressions for the factor of safety. Various assumptions regarding the interslice forces can be transformed into a unified form of expression for the normal stress distribution along the slip surface. An iterative procedure is developed to expedite the convergence of the solution for the factor of safety. Experience to date indicates that the process generally converges within a few iterations. Computation schemes are suggested to avoid numerical difficulty, especially in computing the factor of safety associated with the rigorous Janbu method. The present framework can be readily implemented in a computer program, giving solutions of slope stability associated with a number of conventional methods of slices.

The Terzaghi superposition assumption has been widely used to determine the bearing capacity of shallow footings. Although this assumption always errs on the safe side, a rigorous procedure to calculate the bearing capacity is still of engineering value. This paper presents such a procedure that is free from errors as a result of the superposition assumption. It demonstrates that the ultimate bearing capacity can be precisely expressed by the Terzaghi equation, except that the bearing capacity factor Nγis dependent upon the surcharge ratio. A recently developed numerical method, i.e., the critical slip field method, is used to calculate the modification coefficient for modifying Nγ. It is found that this modification coefficient increases with the surcharge ratio at small values of surcharge ratio and then remains constant for large values of surcharge ratio. However, the errors invoked by the superposition assumption do not exceed 10%. On the basis of numerical calculations, a simple closed-form expression of the modification coefficient is proposed that yields the theoretically rigorous ultimate bearing capacity. In the later part of the paper, errors in bearing capacity calculations owing to the use of conventional procedures are analyzed. It is concluded that the continued use of conventional procedures is justified, but the inherent errors should not be neglected in assessing the performance of shallow foundations.Key words: shallow foundation, strip footing, ultimate bearing capacity, critical slip field.

Conventional methods of slices used for slope stability analysis satisfying all equilibrium conditions involves generally solving two highly non-linear equations with respect to two unknowns, i.e. the factor of safety and the associated scaling parameter. To solve these two equations, complicated numerical iterations are required with non-convergence occasionally occurring. This paper presents an alternative procedure to derive the three equilibrium equations (horizontal and vertical forces equations and moment equation) based on an assumption regarding the normal stress distribution along the slip surface. Combination of these equations results in a single cubic equation in terms of the factor of safety, which is explicitly solved. Theoretical testing demonstrates that the proposed method yields a factor of safety in reasonable agreement with a closed-form solution based on the theory of plasticity. Example studies show that the difference in values of factor of safety between the proposed method, the Spencer method and the Morgenstern–Price method is within 5%. Application of the proposed method to practical slope engineering problems is rather straightforward, but its solution is of the same precision as those given by the conventional rigorous methods of slices since it is still within the rigorous context. Values of the bearing capacity factor Nγare numerically computed using the method of triangular slices. Three assumptions of the value of ψ, the base angle of the active wedge, are analyzed, corresponding to the following three cases: (1) ψ = ϕ, the internal friction angle; (2) ψ = 45° + ϕ/2; and (3) ψ has a value such that Nγis a minimum. The location of the critical failure surface is presented and the numerical solutions to Nγfor the three cases are approximated by simple equations. The influence of the base angle on the value of Nγis investigated. By employing the same assumption regarding interslice forces as that used in the Morgenstern-Price method, two concise recurrence relations between interslice forces and interslice moments are derived which satisfy both force and moment equilibrium conditions. The Newton-Raphson method is used for determining the factor of safety and the associated scaling parameter of the interslice force function. Algebraic derivatives required in the solution process are evolved in a recursive manner which can be easily implemented in a computer program. The choices of initial values of safety factor and scaling parameter are suggested. The procedure proposed in this paper proves to be efficient and solutions converge rapidly.Key words: slope, stability, factor of safety, limit equilibrium method.

A new method of solving earth pressure problems is proposed in this paper within the framework of the limit equilibrium approach. The concept of the critical slip field (CSF) is postulated: the active critical slip field (ACSF) in the active case, and the passive critical slip field (PCSF) in the passive case. Based on the principle of extremum thrust force (which is theoretically consistent with the principle of optimality) in conjunction with the method of slices, a numerical procedure is presented for the determination of such fields and consequently the distribution of earth pressures on retaining walls. For simplicity at this stage, the backfill material is assumed to be a homogeneous cohesionless soil with sloping ground surface carrying uniform and vertical surcharge, while the retaining wall is allowed to be battered, and the strength of the soil may be either fully or partially mobilised. Compared with the rigorous method (i.e. the method of characteristics), the proposed method is simple in principle and easily implemented in a computer program. However, it is more accurate than other approximate techniques and agrees well with available closed-form solutions. A number of examples of ACSF and PCSF are given in this paper, and the validity and efficiency of the proposed method are demonstrated. In addition, it is easy to extend this numerical procedure to obtain more general ACSF and PCSF accommodating non-homogeneous c, ϕ soils subject to complicated loading conditions.

Danshen is the dried root and rhizome of the Chinese medicinal plant Salvia miltiorrhiza Bunge (Labiatae), which has been used to treat hypertension and myocardial infarction. One of its water-soluble active components is magnesium tanshinoate B (MTB). The present study examined and compared the cardiovascular effects of the water-soluble extract of danshen (SME) and its MTB-enriched form (containing 70% of MTB (MTB70)). Anaesthetized rats were infused intravenously with saline or phenylephrine to achieve a normal or elevated blood pressure, respectively. Different doses of SME, MTB70 or vehicle were then injected intravenously and their effect on blood pressure was monitored. The results indicate that SME and MTB70 reduce blood pressure dose-dependently. Independently of the initial blood pressure, SME caused a smaller reduction in blood pressure than MTB70. In rats infused with phenylephrine, MTB70 caused greater decreases in blood pressure than in rats infused with saline, while the responses to SME did not differ between the two groups. From these findings, it appears that MTB is one of the major components responsible for the cardiovascular effects of danshen, and that the beneficial cardiovascular effect of the extract is more prominent under conditions of elevated blood pressure.

Liver X receptors (LXRs) are important transcriptional regulators of lipid homeostasis and proliferation in several cell types. However, the roles of LXRs in pancreatic beta cells have not been fully established. The aim of this study was to investigate the effects of LXRs on pancreatic beta cell proliferation.Gene expression was analysed using real-time RT-PCR. Transient transfection and reporter gene assays were used to determine the transcriptional activity of LXRs in pancreatic beta cells. Cell viability and proliferation were analysed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), DNA fluorometric, BrdU labelling and [(3)H]thymidine incorporation assays. Cell cycle distribution was investigated by flow cytometry analysis. Adenovirus-based RNA interference was used to knockdown LXRalpha, LXRbeta and p27 in MIN6 cells and mouse islets.We found that both Lxralpha (also known as Nr1h3) and Lxrbeta (also known as Nr1h2) were expressed and transactivated the LXR response element in HIT-T15 and MIN6 cells. Activation of LXRs dose-dependently inhibited pancreatic beta cell viability and proliferation. This was accompanied by beta cell cycle arrest at the G1 phase. Furthermore, LXR activation increased levels of the p27 protein by inhibiting its degradation. Knockdown of p27 reversed these effects of LXR activation on growth inhibition and cell cycle arrest.Our observations indicate that LXR activation inhibits pancreatic beta cell proliferation through cell cycle arrest. A well-known regulator of pancreatic beta cell cycle progression, p27, is upregulated and mediates the effects of LXRs on growth inhibition in beta cells. These observations suggest the involvement of aberrant activation of LXR in beta cell mass inadequacy, which is an important step in the development of type 2 diabetes.

Prostaglandin E(2) (PGE(2)) is a well-recognised inhibitor of glucose-stimulated insulin secretion (GSIS). The aim of this study was to investigate the signalling pathway of PGE(2) in beta cell function regulation in HIT-T15 cells and isolated rat islets.mRNA levels of the prostaglandin E receptor 3 (Ptger3) were measured by real-time PCR. Western blot analysis was used to detect changes in the levels of PTGER3, phosphorylated and total Akt, phosphorylated and total forkhead box 'Other' (Foxo). Transient transfection and reporter assays were used to measure Foxo transcriptional activity. The biological significance of PGE(2) in beta cell function was analysed using MTT, flow cytometry and GSIS assays.We found that treating HIT-T15 cells with exogenous PGE(2) stimulated Ptger3 gene expression specifically, and diminished cAMP generation. These were accompanied by the downregulation of Akt and Foxo phosphorylation in HIT-T15 cells and isolated rat islets. Moreover, PGE(2) upregulated basal and partially reversed constitutively active Akt-inactivated Foxo transcriptional activity. Furthermore, GSIS was impaired in PGE(2)-treated HIT-T15 cells and isolated islets. However, the dosage used in the above experiments did not affect beta cell viability and apoptosis. In addition, insulin-like growth factor 1 (IGF-1) pretreatment reversed the effects of PGE(2), and wortmannin treatment abolished the preventive effects of IGF-1.Our observations strongly suggest that PGE(2) can induce pancreatic beta cell dysfunction through the induction of Ptger3 gene expression and inhibition of Akt/Foxo phosphorylation without impacting beta cell viability. These results shed light on the mechanisms of PGE(2) actions in pancreatic beta cell dysfunction.

To investigate the antioxidative activity of the constituents of the roots of Scrophularia ningpoensis (Chinese name: Xuanshen).The main compounds from the roots of Scrophularia ningpoensis were isolated and identified by chromatography and FABMS, NMR etc. Using the techniques of pulse radiolysis, the electron transfers from iridoid glycosides (IG) or phenylpropanoid glycosides (PG) to oxidized OH radical adducts of 2'-deoxyadenosine-5'-monophosphate acid (dAMP) or 2'-deoxyguanosine-5'-monophosphate acid (dGMP) were observed.Two IG: harpagoside and harpagide, two PG: angoroside C and acteoside were obtained as the main hydrophilic constituents of the plant. At 0.1 mmol/L concentration, angoroside C and acteoside were able to repair the oxidized OH adducts dAMP and dGMP significantly. However, harpagoside and harpagide had no such effect. The electron transfer rate constants of angoroside C with dAMP and dGMP were 4.2 x 10(8) and 10.3 x 10(8) L.mol-1.s-1; the electron transfer rate constants of acteoside with dAMP and dGMP were 5.3 x 10(8) and 20.2 x 10(8) L.mol-1.s-1.PG from Scrophularia ningpoensis have a potent antioxidative activity for reducing of the oxidized OH adducts of dAMP and dGMP.

Four new 2, 2-dimethylchromenes named leptol B (I), ethylleptol B (II), methylleptol B (III), leptene B (IV), along with a known chromene-methylevodionol (V) have been isolated from the aerial parts of Evodia lepta. Their structures were elucidated by spectroscopic analysis and chemical techniques.

The activation of stress-activated protein (SAP) kinase may lead to an induction of apoptosis that is responsible for part of the cardiomyocyte death in reperfusion injury. The objective of the present study was to investigate the mechanism by which magnesium tanshinoate B (MTB), a bioactive compound isolated from Danshen, prevents apoptosis in cardiomyocytes in the ischemic/reperfused heart. Isolated adult rat hearts were perfused by the Langendorff mode with medium containing MTB prior to the induction of normothermic global ischemia. At the end of the 30-min ischemic period, the heart was reperfused with the same medium with or without MTB for an additional 20 min. In the MTB-treated ischemic/reperfused heart, the number of apoptotic nuclei was reduced by 2.5-fold in comparison to that in untreated ischemic/reperfused controls [23 +/- 4 vs 57 +/- 7 (mean +/- SD) TUNEL-positive cells, respectively, N = 3-4, P0.001]. SAP kinase activity was elevated 1.7-fold in ischemic/reperfused rat hearts [35.6 +/- 3.8 vs 21.2 +/- 3.3 (control) (mean +/- SEM) relative densitometric units, N = 4-6, P0.05]. Treatment with MTB abolished this elevation in SAP kinase activity (25.0 +/- 5.2 relative densitometric units), which was also decreased by 40% in the nucleus. When the heart was subjected to ischemia alone, there was no significant change in SAP kinase activity in the presence or absence of MTB. MTB did not appear to affect the p38 mitogen-activated protein kinase activity in this model system. In conclusion, MTB was shown to have cardioprotective activity against apoptosis, probably through the inhibition of SAP kinase activity.

Nitric oxide (NO) is a potent vasodilator which plays an important role in regulating vascular tones. Danshen, a Chinese herbal medicine has been widely used for the treatment of cardiovascular diseases. The objective of this study was to investigate the effect of magnesium tanshinoate B (MTB), a compound purified from Danshen, on the production of NO in human endothelial cell line (ECV304). After cells were incubated with MTB (1-10 microM) for 1 or 4 h, amounts of NO metabolites released by cells were quantified and cellular NOS activities were determined following the conversion of [3H]arginine to [3H]citrulline. The NOS protein expression was determined by Western immunoblotting analysis. MTB (1-10 microM) stimulated the release of NO and its metabolites from endothelial cells. Following MTB treatment, the cellular NOS activities were significantly enhanced with a concomitant increase in the levels of constitutive NOS (cNOS) protein mass (110-178%). Selective activation of cNOS by MTB may be employed therapeutically in modulating NO production in endothelial cells.

To study the actions of salvianolic acid A (SA-A) on liver injury and liver fibrosis.The liver fibrotic rat was made by i.p. injection of CCl4. The model rats were divided into 3 groups treated with SA-A, colchicine (Col), and Salvia miltiorrhiza Bunge (SMB), respectively. Six wk later the rat liver pathology was examined, type I and III collagen in the liver were examined by immunohistochemistical method. Also hydroxyproline (Hyd) and malondialdehyde (MDA) in the liver, alanine aminotransferase (AlaAT), aspartate aminotransferase (AspAT), and albumin (Alb) levels in the serum were measured.SA-A inhibited serum AlaAT and AspAT activities, decreased MDA and Hyd contents, alleviated liver fibrogenesis, protected deposition of type I and III collagen in liver matrix. The actions of SA-A on liver fibrosis were similar to those of Col and SMB, action of SA-A decreasing MDA was better than that of Col.SA-A has marked effects against liver injury and fibrosis, associated with its anti-lipid peroxidation actions.

For the purpose of searching for new drug with high potency and simple chemical structure, the dominant conformation and structural parameters of Guan-Fu base (GFA) molecule were modelled and calculated with a SGI-4D 25G computer. The propanediolamine chain in GFA might be considered to be a pharmacophore responsible for the bioactivity and the configuration of the chain seemed important. Thus, thirteen compounds of (erythro)-p-x-PhCHOHCHOHCH2NHR(x = H, I1-7; X = NO2, II1-6) were prepared. Among them, 10 compounds showed antiarrhythmic effect on aconitine-induced arrhythmia in rats. The ED50(to stop VT) of I2 and ED50(to stop VP) of I3 were shown to be comparable with those of GFA. In the synthesis, no stereoselectivity was found in the Prevost reaction with allylamine analogues (a1-7). After a1-7 were acetylated, the erythro type products(I1-7) were obtained.

To study the effects of isovanihuperzine A (IVHA) on cholinesterase and scopolamine-induced memory deficit.AChE and BuChE activities were determined by the colorimetric method of Ellman. The Ki value was determined by the plotting method of Lineweaver and Burk. In a behavioral test, rats were trained to perform a radial arm maze task using a partially baited procedure.The anti-AChE activity of IVHA was comparable to huperzine A (Hup-A), and was more potent than those of physostigmine and galanthamine with an IC50 value of 0.11 mumol.L-1. IVHA was a mixed competitive type with a Ki value of 32 nmol.L-1. It bound to AChE in a reversible manner. IVHA at a dose of 0.2 mg.kg-1 ip significantly reversed scopolamine-induced working memory and reference memory impairments in radial arm maze.IVHA is a new potential reversible AChE inhibitor and merits further study as a cognitive enhancer.

An extensive investigation of the petroleum ether extract of TRIPTERYGIUM WILFORDII plants have revealed the presence of four new compounds, triptonoterpene, neotriptonoterpene, triptonodiol, and neotriptonolide, in addition to the diterpenes isolated and characterized earlier. The structures of these novel natural products have been elucidated.
