The SARS-CoV-2 (COVID-19) pandemic has attracted attention to the challenge of neuroinflammation as an unavoidable component of viral infections. Acute neuroinflammatory responses include activation of resident tissue macrophages in the CNS followed by release of a variety of cytokines and chemokines associated with activation of oxidative stress and delayed neuron damage. This makes the search for treatments with indirect anti-inflammatory properties relevant. From this point of view, attention is focused on further study of the treatment of patients with COVID-19 with dipyridamole (Curantil) which, having antiviral and anti-inflammatory effects, can inhibit acute inflammatory activity and progression of fibrosis, is a drug with potential, especially among patients with early increases in the D-dimer concentration and severe signs of microangiopathy.

We consider the Cauchy problem for a Schrödinger equation whose Hamiltonian is the difference of the operator of multiplication by the potential and the operator of taking the second derivative. Here the potential is a real differentiable function of a real variable such that this function and its derivative are bounded. This equation has been studied since the advent of quantum mechanics and is still a good model case for various methods of solving partial differential equations. We find solutions of the Cauchy problem in the form of quasi-Feynman formulae by using Remizov’s theorem. Quasi-Feynman formulae are relatives of Feynman formulae containing multiple integrals of infinite multiplicity. Their proof is easier than that of Feynman formulae but they give longer expressions for the solutions. We provide detailed proofs of all theorems and deliberately restrict the spectrum of our results to the domain of classical mathematical analysis and elements of real analysis trying to avoid general methods of functional analysis. As a result, the paper is long but accessible to readers who are not experts in the field of functional analysis.

The formation of a resultant voltage vector of a symmetrical five-phase ABCDE winding is considered when it is connected to a symmetrical five-phase voltage with an ACEBD time phase sequence. It is found that, with the ACEBD phase sequence, the space–time resultant voltage vector is a third harmonic vector, and the modulus of the space–time resultant voltage vector in the first harmonic is equal to zero. The rotation speed of the resultant voltage vector is three times greater than the frequency of the converter. Switching the AEDCB phase sequence to ACEBD leads to a discrete increase in the rotation speed of the resultant voltage vector by three times at a constant switching frequency of the converter keys. Using control algorithms of the converter, to which a symmetrical five-phase winding is connected, it is possible to amplify the third time harmonic of the phase voltage and to weaken the first time harmonic. The implementation of the algorithm amplifying the third phase voltage harmonic requires an increase in the number of commutations per period, while the switching frequency of the keys decreases.

COVID‑19 is acute respiratory infection caused by the new coronavirus SARS-CoV‑2 (2019-nCoV). Until now, the pathogenesis of the virus has not been fully studied, but the global medical practice is dominated by the theory of systemic inflammation as the main damaging factor of viral infection. A special role is given to the study of the state of the endothelium. Understanding the molecular components underlying functional disorders and vascular damage, together with the ability of new laboratory technologies to determine the effect of endothelial damage on the entire body, provides a key to modern treatment strategies. Antihypertensive, antiplatelet drugs and statins are the drugs whose effect on the functional state of the endothelium has been studied most fully.

Stearyl methacrylate–glycidyl methacrylate copolymers were synthesized by controlled atom transfer radical polymerization. These copolymers can be used as multifunctional additives improving the low-temperature properties and thermal oxidation resistance of diesel fuel. The additives synthesized well compete in depressant properties with commercial imported additives and surpass some of them in the effect on the thermal oxidation resistance of hydrotreated diesel fuel of Euro-5 class, appreciably decelerating the fuel oxidation.

Multiphase valve electromechanical systems with an odd number of phases (three, five, seven, nine, etc.) have been investigated. It has been established that, with an increase in the number of phases, the copper consumption and losses in copper do not increase as compared to a three-phase machine. The increase in the number of phases of the multiphase motor winding, while maintaining unchanged power, makes it necessary to reduce the supply voltage of the converter; i.e., the phase voltages of the motor are reduced (the motor becomes “low-voltage motor”). However, this increases the input current of the converter. The increase in the number of phases of the motor at a constant power leads either to a decrease in the phase inductance, or to a decrease in the phase current compared to a three-phase winding. This can help improve switching the keys of the converter. As the number of phases increases, the loading current of the converter keys decreases. The results of experiments confirming the theoretical studies are presented.

The most specific method of the recording of the rate offree radical reactions is the method of electron paramagnetic resonance (EPR) spectroscopy, but it is rarely used in applied biology due to expensive equipment and complexity of the execution of measurements. However chemists have found a number of colored organic radicals which lose the coloring under transition into diamagnetic form. In the given paper there are presented results of our studies on the development of methods for the assessment of oxidant equilibrium in biological media with a use of stable radical 2,2-diphenyl-1-picrylhydrazyl (DPPH) and cation-radicals of N,N-diethyl-p-phenylenediamine (DEPPD). We have developed the new modification of DPPH test, replacing methanol-based incubation medium by non-ionic detergent solution, compatible with native blood serum. Modified DPPH test conserved typical biphasic kinetics of the origin variant, had the similar sensitivity to model antioxidants (IC values 49, 38 and 13 mkMfor ascorbate, a-tocopherol and quercetine, correspondingly) and was applied in experiments on laboratory animals treated with nano- and ionic silver, carbon nanotubes, microfine coal and electrolytic dust. We have tried also the assay of serum lipid hydroperoxides based on Fe-initiated DEPPD oxidation (Alberti et al., 2000). The comparison of kinetics of DEPPD oxidation in model (HO/Fe) and biologic (rat serum/Fe) systems, before and after Fe addition, seems to be an evidence that ceruloplasmin (CP) was involved in the resulting process, but failed to determine its polynomial kinetics, at least for the rat serum and DEPPD excess. The use of CP monoclonal antibodies seems to be the best way for the clarification of the mechanism of this reaction.

In the process of optimization of heterologous expression of thermostable chemotaxis proteins CheW and CheY as industrially useful polypeptides, their direct influence on the cell growth kinetics and morphology of Escherichia coli was observed. CheW and CheY of bacteria of the genus Thermotoga, being expressed in recombinant form in E. coli cells, are involved in the corresponding signal pathways of the mesophilic microorganisms. The effects of such involvement in the metabolism of "host" cells are extremely diverse: from rapid aging of the culture to elongation of the stationary growth phase. We also discuss the mechanisms of the influence of the heterologous chemotaxis proteins on cells, their positive and negative effects, as well as potential applications in industry and biomedicine.

Normal human CD4+ T-lymphocytes can undergo malignant transformation during prolong cultivation in conditions of high endonuclease G (EndoG) expression or after DNA damage. The aim of this work was to study biochemical and cytogenetic features of transformed ex vivo human malignant CD4+ T- lymphocytes, as well as biochemical and morphological characteristics of tumors that develop in athymic mice after transplantation of these cells. The telomerase activity was higher and telomere length was shorter in tumor cells than in control cells. Transformed malignant cells exhibited a high level of chromosomal aberrations. Expression of genes regulating the cell cycle changed in transformed malignant CD4+ T-lymphocytes and tumor cells. The tumors that developed were classified as multicomponent Т-cell lymphomas and panniculitis-like T-cell lymphomas. Thus, transformed CD4+ T-lymphocytes can generate malignant tumors of various histogenesis.

The method of the evaluation of the antioxidant activity (AOA) with a help of stable radical 2,2-diphenyl-1-picrylhydrazyl test (DPPH test) widely applied in pharmacological and food chemistry, was used also in some clinic investigations for the assessment of the total activity of serum low-molecular-weight antioxidants, but never was applied earlier for toxicological and environmental studies. We have developed the new modification of DPPH test, by replacing methanol-based incubation medium to non-ionic water detergent solution, compatible with native blood serum. Modified DPPH test was approved in 5 toxicological experiments on laboratory animals (Gig. Sanit., 2016, 9: 884-90) and in the hygienic study of the influence of ambient air pollution on the health state in a cohort of 142 capable Moscow residents (aged of 43.5 ± 11.8 years). As a results of the cohort observation, AOA of human serum in DPPH test had close to the normal distribution without gender differences, the slowly decrease with age by nearly 0.4% per year and was positively associated with serum concentrations of catabolic antioxidant - uric acid (R=0.257; p=0.002). The influence of outdoor air pollution in the points of the persons permanent homes was studied by 2 methods: 1) dividing Moscow territory into 2 zones accordingly the long-term monitoring of Hydrometereology and Environmental Monitoring Service; 2) by cartographic adjustment of corresponding home buildings to the nearest of 46 route stations of Moscow Agency for Health and Consumer Rights. No significant differences was found between residents of 2 preferred city zones in 12 used serum biochemical markers, the results of DPPH test including. Significant correlation exposure-response was found between the variables “serum AOA in DPPH test” and “benzene annual outdoor concentration” (R=0.342; p=0.00003) by individual exposure evaluation according to the data of route stations. Two main independent predictors of serum AOA values in the observed cohort of Moscow residents (outdoor air benzene and serum uric acid concentrations) detected 14 and 8.5 % of AOA total variance (p = 0.000002 and p =0.00018 correspondingly).

Apoptotic endonucleases act cooperatively to fragment DNA and ensure the irreversibility of apoptosis. However, very little is known regarding the potential regulatory links between endonucleases. Deoxyribonuclease 1 (DNase I) inactivation is caused by alternative splicing (AS) of DNase I pre-mRNA skipping exon 4, which occurs in response to EndoG overexpression in cells. The current study aimed to determine the role of EndoG in the regulation of DNase I mRNA AS and the modulation of its enzymatic activity. A strong correlation was identified between the EndoG expression levels and DNase I splice variants in human lymphocytes. EndoG overexpression in CD4+ T cells down-regulated the mRNA levels of the active full-length DNase I variant and up-regulated the levels of the non-active spliced variant, which acts in a dominant-negative fashion. DNase I AS was induced by the translocation of EndoG from mitochondria into nuclei during the development of apoptosis. The DNase I spliced variant was induced by recombinant EndoG or by incubation with EndoG-digested cellular RNA in an in vitro system with isolated cell nuclei. Using antisense DNA oligonucleotides, we identified a 72-base segment that spans the adjacent segments of exon 4 and intron 4 and appears to be responsible for the AS. DNase I-positive CD4+ T cells overexpressing EndoG demonstrated decreased progression towards bleomycin-induced apoptosis. Therefore, EndoG is an endonuclease with the unique ability to inactivate another endonuclease, DNase I, and to modulate the development of apoptosis.

The pandemic caused by the SARS-CoV-2 virus (COVID-19) made it necessary to evaluate in more detail the processes of neuroinflammation as an integral component of the pathogenesis of viral infection. The acute neuroinflammatory response includes the activation of resident tissue macrophages in the CNS and the subsequent release of various cytokines and chemokines, which probably activates oxidative stress, causing long-term neuronal damage. This makes urgent the search for drugs with indirect anti-inflammatory effects with proven effectiveness. From this point of view, it is worth further studying the treatment of patients with COVID-19 with dipyridamole, which, with its antiviral activity and anti-inflammatory effect, inhibiting acute inflammation and progressive fibrosis, is the drug of choice, especially for patients with early signs of elevated D-dimer concentrations and pronounced clinical symptoms of microangiopathy.Пандемия, вызванная вирусом SARS-CoV-2 (COVID-19), привлекла внимание к проблеме нейровоспаления как неотъемлемому компоненту вирусной инфекции. С этой точки зрения заслуживает внимания дальнейшее изучение терапии пациентов с COVID-19 препаратом дипиридамол (Курантил), который, обладая противовирусным и противовоспалительным эффектами, способностью ингибировать активность острого воспаления и прогрессирования фиброза, является перспективным препаратом, особенно у пациентов с ранним повышением концентрации D-димера и выраженными проявлениями микроангиопатии.

Market segments demonstrated high growth rates, following in line with global trends. State institutions had adopted a number of programs aimed at supporting and developing the sector of practical implementation of post-genomic technologies. the policy of the Government of the Russian Federation, aimed at importing foreign drugs and medical products, created a full cycle of domestic production of targeted drugs for personalized therapy of malignant tumors.

Regulatory T cells (Tregs) play a fundamental role in the maintenance of immunological tolerance by suppressing effector target T, B and NK lymphocytes. Contact-dependent suppression mechanisms have been well-studied, though contact-independent Treg activity is not fully understood. In the present study, we showed that human native Tregs, as well as induced ex vivo Tregs, can cause in vitro telomere-dependent senescence in target T, B and NK cells in a contact-independent manner. The co-cultivation of target cells with Tregs separated through porous membranes induced alternative splicing of the telomerase catalytic subunit hTERT (human Telomerase Reverse Transcriptase), which suppressed telomerase activity. Induction of the hTERT splicing variant was associated with increased expression of the apoptotic endonuclease EndoG, a splicing regulator. Inhibited telomerase in target cells co-cultivated with Tregs for a long period of time led to a decrease in their telomere lengths, cell cycle arrest, conversion of the target cells to replicative senescence and apoptotic death. Induced Tregs showed the ability to up-regulate EndoG expression, TERT alternative splicing and telomerase inhibition in mouse T, B and NK cells after in vivo administration. The results of the present study describe a novel mechanism of contact-independent Treg cell suppression that induces telomerase inhibition through the EndoG-provoked alternative splicing of hTERT and converts cells to senescence and apoptosis phenotypes.

Regulatory T cells (Tregs) suppress the activity of effector T, B and NK lymphocytes and sustain immunological tolerance, but the proliferative activity of suppressed cells remains unexplored. In the present study, we report that mouse Tregs can induce replicative senescence and the death of responder mouse CD4+CD25- T cells, CD8+ T cells, B cells and NK cells in vitro and in vivo. Contact-independent in vitro co-cultivation with Tregs up-regulated endonuclease G (EndoG) expression and its translocation to the nucleus in responder cells. EndoG localization in the nucleus induced alternative mRNA splicing of the telomerase catalytic subunit Tert and telomerase inhibition. The lack of telomerase activity in proliferating cells led to telomere loss followed by the development of senescence and cell death. Injection of Tregs into mice resulted in EndoG-associated alternative splicing of Tert, telomerase inhibition, telomere loss, senescence development and increased cell death in vivo. The present study describes a novel contact-independent mechanism by which Tregs specify effector cell fate and provides new insights into cellular crosstalk related to immune suppression.

Mutant homologues of small chemotactic and DNA-binding proteins from thermophilic bacteria Thermotoga petrophila RKU-1 and Thermotoga naphthophila were obtained. These proteins can be expressed in the recombinant form in E. coli cells. A wide range of properties and parameters that are important for isolation of these proteins were revealed: stability in a wide temperature and pH range, high level of expression, solubility, and the possibility of using simple purification schemes with low number of successive steps. The positive effect of proteins on in vitro fibroblasts growth was demonstrated. The described properties of the target proteins indicate the possibility of their use in different biotechnology industries as an inexpensive source of L-amino acids.

The work is aimed at selecting a promising grade of a tool material, whose physical-mechanical characteristics would allow using it for processing the surfaces of discontinuous parts in the presence of shock loads. An analysis of the physical-mechanical characteristics of most common tool materials is performed and the data on a possible provision of the metal-working processes with promising composite grades are presented.

It is known that apoptotic endonuclease G (EndoG) induces alternative splicing (AS) of telomerase catalytic subunit TERT (telomerase reverse transcriptase) mRNA and inhibits telomerase activity in tumor cells and activated human T cells. The aim of this study was to investigate the possibility of TERT mRNA AS induction and inhibition of telomerase activity by EndoG in activated mouse and rat lymphocytes. To induce EndoG expression, mouse and rat CD4+, CD8+ T cells, B cells, and NK cells were transfected with the pEndoG-GFP plasmid or incubated with the DNA-damaging agent cisplatin in vitro. The increase in the EndoG expression resulted in decreased expression of full-length active TERT variant, enhanced synthesis of the truncated splicing variant, and decreased telomerase activity. An increase in the EndoG expression, a change in the mRNA pool of TERT splicing variants, and inhibition of telomerase activity were observed in mouse and rat lymphocytes after cisplatin administration in vivo. Thus, EndoG is capable of inducing TERT mRNA AS and regulating telomerase activity in mouse and rat lymphocytes.

In the work a recombinant chemotaxis protein CheW from Thermotoga petrophila RKU-1 (TpeCheW) and its mutant homolog (TpeCheW-mut) were created. It was shown that, despite the low homology with CheW prototypes from intestinal bacteria, these proteins didn't cause metabolic overload and were well expressed by cells of E. coli laboratory strains. We have discovered a broad spectrum of industrial valuable properties of the TpeCheW-mut protein such as stability in a wide range of temperatures and pH, high expression level, solubility and possibility of the application of a simple low-stage purification methodology with the use of preliminary heat treatment. The complexity of the technology of machining by cutting complex surfaces of parts heterogeneous in terms of physical and mechanical properties and different machinability, and, in particular, of metals (aluminum) and plastics (polyurethane and polyethylene), which form an assembly component, a section of the container-type countermine system is defined in the paper. The description of technological process operations of machining of a basic detail of an item, the body, is given. Features, the presence of which creates not only difficulties in the implementation of the technological process of manufacturing a base part, but is not permissible in the operation of a defense-purpose item are revealed. The article proposes design and technological solutions for the use of a new cutting tool design, capable to eliminate errors of creep feed drilling, avoid sticking of the material of the protective shell and casing on the active cutting part, exclude damage to the contact body and thereby ensure the specified performance of the item , Field tests of a container-type countermine system with basic parts manufactured using the new tool and technology have shown high efficiency of the decisions taken.

Aim. To study the predictive ability of the NEWS2, 4C Mortality Score, COVID-GRAM and qSOFA scales in predicting clinical outcomes in patients with severe coronavirus disease 2019 (COVID-19) hospitalized in a multidisciplinary hospital.Material and methods. The pilot retrospective cohort study used data from 90 patients (52 — intensive care unit subgroup, 38 — general unit subgroup) with a confirmed diagnosis of COVID-19 hospitalized in the O.M. Filatov City Clinical Hospital № 15 (Moscow) from January to March 2021.Results. The probability of a positive outcome of the disease significantly negatively correlates with the patient’s age (R=-0,514; p=0,0002). The best correlation with the COVID-19 outcome had a 4C Mortality Score (R=0,836; p=0,0001). Logistic regression revealed a significant dependence of the “outcome” and “age” parameters with the greatest accuracy in the form of age subgroups according to the World Health Organization classification with odds ratio (OR) of 4,29 (p=0,0001). As a result of ROC analysis, the best predictive ability of disease outcomes was shown for the 4C Mortality Score (area under curve (AUC)=0,878; 95% confidence interval (CI): 0,782- 0,975 (p=0,00001)) and COVID-GRAM (AUC=0,807; 95% CI: 0,720- 0,895 (p=0,00001)); taking into account the division of patients into age subgroups, optimal predictive tools were obtained: in subgroups 18-44 years old and 45-59 years old — the 4С Mortality Score (AUC=0,892, 95% CI: 0,762-0,980 (p=0,002) and AUC=0,853, 95% CI: 0,784-0,961 (p=0,0014), respectively); in the subgroup 60-74 years old — the COVID-GRAM (AUC=0,833, 95% CI: 0,682-0,990 (p=0,038)); in subgroups 75-90 years and >90 years — NEWS2 (AUC=0,958, 95% CI: 0,807-1,0 (p=0,002) and AUC=0,818, 95% CI: 0,713-0,996 (p=0,006), respectively). ROC analysis showed that the age of 70 years is the threshold value, above which the probability of an unfavorable COVID-19 outcome increases significantly (OR=11,63; 95% CI: 9,72- 12,06 (p=0,0052)).Conclusion. The pilot study showed the significance of predicting the hospitalization outcome of patients with severe COVID-19. The 4C Mortality Score and COVID-GRAM scales had the best predictive accuracy. The specificity and sensitivity of the scores depended on the age of a patient. The age of 70 years was the threshold value at which the risk of an adverse outcome increased significantly. Based on the data obtained, it is planned to study the problem of predicting the disease course, taking into account the severity of COVID-19.

Apoptotic endonuclease EndoG plays a key role in the alternative splicing of mRNA of human TERT telomerase catalytic subunit. The aim of this work was to test the ability of EndoG to induce alternative splicing of mRNA of other genes and in other organisms. To determine new mRNA splice-variants, EndoG overexpression was induced in human, mouse and rat CD4+-T-lymphocytes followed by sequencing of total RNA of these cells. Sequencing results showed that besides TERT, EndoG induced alternative splicing of deoxyribonuclease I (DNase I), caspase-2 (Casp-2) and BCL-x. The expression level of EndoG strongly correlated with mRNA splicing-variants of TERT, DNase I, Casp-2, and BCL-x in intact CD4+-T cells of healthy donors as well as different lines of mice and rats. EndoG overexpression induced down-regulation of fulllength mRNAs of TERT, DNase I, Casp-2, and BCL-x and up-regulation of their short-length mRNAs. Alternative splicing of studied mRNAs resulted in down-regulation of enzymatic activity of proteins in vitro and in vivo. The results of this work confirm the ability of endonuclease EndoG to induce alternative splicing of several mRNAs in human, mice and rats.

The study of the poultry needs in basic nutrients allowed the development of a scheme for obtaining feed polypeptides (“polypeptide cassettes”) enriched with L-amino acids, which are necessary for the metabolism of birds. The amino acid and nucleotide profiles of about 500 bioinformation sequences of thermostable plant proteins and archaea were studied, on the basis of which candidate sequences were selected. In silico, the amino acid and domain composition of the thermostable polypeptides has been optimized. A library of genetically engineered constructs encoding optimized polypeptides with the necessary composition of L-amino acids irreplaceable for poultry has been created. Primary E. coli producer strains were obtained, and the expression and thermostability of the target polypeptides were studied.

Rhodospirillum rubrum L‐asparaginase mutant E149R, V150P, F151T (RrA) down‐regulates telomerase activity due to its ability to inhibit the expression of telomerase catalytic subunit hTERT. The aim of this study was to define the effect of short‐term and long‐term RrA exposure on proliferation of cancer Jurkat cell line and normal human CD4+ T lymphocytes. RrA could inhibit telomerase activity in dose‐ and time‐dependent manner in both Jurkat and normal CD4+ T cells. Continuous RrA exposure of these cells resulted in shortening of telomeres followed by cell cycle inhibition, replicative senescence, and development of apoptosis. Complete death of Jurkat cells was observed at the day 25 of RrA exposure while normal CD4+ T cells died at the day 50 due to the initial longer length of telomeres. Removal of RrA from senescent cells led to a reactivation of hTERT expression, restoration telomerase activity, re‐elongation of telomeres after 48 h of cultivation, and survival of cells. These findings demonstrate that proliferation of cancer and normal telomerase‐positive cells can be limited by continuous telomerase inhibition with RrA. Longer telomeres of normal CD4+ T lymphocytes make such cells more sustainable to RrA exposure that could give them an advantage during anti‐telomerase therapy. These results should facilitate further investigations of RrA as a potent anti‐telomerase therapeutic protein.

Published data on the influence of sulfur and its compounds on the ecological properties and performance characteristics of diesel fuels have been analyzed. The main characteristics of depressant, antiwear, and antioxidant additives to modern hydrotreated diesel fuels with low and ultralow sulfur content have been considered. A special emphasis is made on the additives based on synthetic and natural high-molecularweight compounds, including complex multifunctional broad-spectrum additives for clean diesel fuels, conforming to the European requirements and quality standards.

The effect of a number of quinoid compounds on methyl methacrylate polymerization initiated by azo-bis(isobutyronitrile) has been studied. It has been revealed that N-aryl-o-iminobenzoquinones, in contrast to o-benzoquinones, can provide radical polymerization of methyl methacrylate in controllable mode. The efficiency of the compounds as chain growth regulators has been found to depend on their composition and reaction conditions. It has been established that 4,6-di-tert-butyl-N-(2,6-diethylphenyl)-o-iminobenzoquinone and 4,6-di-tert-butyl-N-(2,6-diisopropylphenyl)-o-iminobenzoquinone under radical initiation conditions provide the synthesis of poly(methyl methacrylate) with wide-range molecular weight, retaining polydispersity indices about ~1.4–1.8 up to deep conversions.

An algorithm for a gyrovertical based on MEMS sensors for detecting pitch and roll angles of aircraft is considered. It is suggested to integrate inertial sensors with an air data system. A flight simulator is used to optimize the algorithm coefficients.
