1. The Influence of Patient Preference on Surgeons' Treatment Recommendations in the Management of Intraductal Papillary Mucinous Neoplasms.

Abstract

Objective: We aimed to determine whether surgeon variation in management of intraductal papillary mucinous neoplasms (IPMN) is driven by differences in risk perception and quantify surgeons' risk threshold for changing their recommendations. Background: Surgeons vary widely in management of IPMN. Methods: We conducted a survey of members of the Americas HepatoPancreatoBiliary Association, presented participants with 2 detailed clinical vignettes and asked them to choose between surgical resection and surveillance. We also asked them to judge the likelihood that the IPMN harbors cancer and that the patient would have a serious complication if surgery was performed. Finally, we asked surgeons to rate the level of cancer risk at which they would change their treatment recommendation. We examined the association between surgeons' treatment recommendations and their risk perception and risk threshold. Results: One hundred and fifty surgeons participated in the study. Surgeons varied in their recommendations for surgery [19% for vignette 1 (V1) and 12% for V2] and in their perception of the cancer risk (interquartile range: 2%--10% for V1 and V2) and risk of surgical complications (V1 interquartile range: 10%--20%, V2 20%--30%). After adjusting for surgeon characteristics, surgeons who were above the median in cancer risk perception were 22 percentage points (27% vs. 5%) more likely to recommend resection than those who were below the median (95% CI: 11.34%; P<0.001). The median risk threshold at which surgeons would change their recommendation was 15% (V1 and V2). Surgeons who recommended surgery had a lower risk threshold for changing their recommendation than those who recommended surveillance (V1: 10.0 vs. 15.0, P=0.06; V2: 7.0 vs. 15.0, P= 0.05). Conclusions: The treatment that patients receive for IPMNs depends greatly on how their surgeons perceive the risk of cancer in the lesion. 4. Characterization of Capsicum chlorosis virus infecting chilli (Capsicum annuum. L) in southern India

Abstract

During a field survey in Tamil Nadu (2014–16), chilli plants showing chlorotic and necrotic ring spots and necrotic streaks on leaves and stems were observed. Disease incidence ranged between 5 and 50% among five surveyed districts. RT-PCR assay using species specific primers detected 25% of samples infected with Capsicum chlorosis virus (CaCV) and 50% of samples infected with groundnut bud necrosis virus (GBNV). For simultaneous detection of CaCV and GBNV, duplex RT-PCR (dRT-PCR) has been validated. Complete nucleotide sequences of L-RNA, M-RNA, NSs and N genes of CaCV isolate (CaCV-TN-CBE) were determined.The L-RNA constitutes 8913 nt, M-RNA of 4848 nt, NSs gene of 1320 nt and the N gene of 828 nt. Sequence analysis showed that CaCV-TN-CBE isolate is closely related to China isolate based on L- and M- RNA whereas with Taiwan isolate based on S RNA. Also, based on nucleotide identity and phylogenetic analysis CaCV-TN-CBE isolate is different from the previously reported isolate from India. Further nucleotide sequences of L- and S- RNA have shown recombination events. This study suggested the isolate infecting chilli in Tamil Nadu might have evolved by undergoing recombination and reassortment evolutionary mechanisms.

Abstract

Chayote (Sechium edule (Jacq. Sw.) is a single seeded cucurbitaceous vegetable crop mainly grown for its fruit. During 2015–2016, mosaic and leaf distortion type of symptoms were observed in chayote plants in hilly regions of Tamil Nadu. The disease incidence was 50. 3–100% and yield loss was about 69.9% in Dindigul district. The infected chayote plants showed yellow spots, yellow mosaic, leaf curling, puckering, and enations. The fruits of infected plants were malformed and were not marketable. The begomovirus causing the disease was identified as a variant of tomato leaf curl New Delhi virus. The chayote isolates of ToLCNDV share only 91 to 92% identity with other ToLCNDV isolates, deserving to be designated as distinct strain. The phylogenetic analysis on the basis of DNA A component nucleotides clearly indicated common origin of chayote, ridge gourd, ash gourd isolates of India along with Spanish isolates of ToLCNDV. This was contrasting to diverse origin of ToLCNDV isolates from other countries. The virus was sap transmissible to selected cucurbitaceous hosts. The whitefly population (Asia-I) reared in the glass house transmitted the virus to bottle gourd with 4 h of acquisition access period (AAP) and 24 h of inoculation feeding period (IFP). Heavy infestation of greenhouse whiteflies, Trialeurodes vaporariorum on infected chayote plant in Kodaikanal paved way to investigations on its role in vector transmission of ToLCNDV. The field population of T. vaporariorum was found to be viruliferous in PCR using virus specific primers. The greenhouse whitefly efficiently transmitted the virus with 4 h of AAP and 24 h IFP. Seed-borne nature of ToLCNDV was confirmed in PCR by using Roja’s and ToLCNDV specific primers in different parts of the fruit viz., pericarp, mesocarp, seed coat, endosperm and embryo. This is the first report of seed transmissible nature of ToLCNDV, its implication in transboundary movement of the virus across several countries is discussed.

Abstract

Rice is affected by many diseases, among which sheath blight incited by Rhizoctonia solani causes substantial loss to the crop worldwide. The present investigation aims to assess the efficacy of Bacillus subtilis strain EBPBS4 against rice sheath blight and to delineate the mechanism of resistance through a proteomic approach. Eighty endophytic Bacillus isolates were screened for the antagonistic activity against rice sheath blight disease. Among them, Bacillus subtilis isolate EBPBS4, displayed highest antagonistic activity, possessed 13 antimicrobial peptide genes viz., iturin A, iturin D, iturin C, surfactin, bacilysin, fengycin, ericin, mycosubtilin, subtilosin, mersacidin, and plant growth promoting genes. Sequential application of the endospore-based liquid formulation of Bacillus subtilis, EBPBS4 as seed treatment + seedling dip + soil application + foliar spray reduced sheath blight severity significantly when compared to control under in vivo. Unraveling the molecular mechanism of resistance through 2D-PAGE-MALDI-TOF Mass spectrometry (MS) analysis revealed a total of 15 defense/resistant proteins viz., β-1,3-glucanase, ascorbate peroxidases, PAL, chitinase, putative disease resistance protein RGA1, NBS-LRR L-type lectin-domain containing receptor kinase, serine threonine protein kinase, and HSP20-like chaperone domain containing protein which are found to be differentially regulated when there is a tripartite interaction of host, pathogen and biocontrol agent. Thus endospore-based bioformulation of B. subtilis EBPBS4 was found to induce the expression of defense/resistance genes in plants and thus can be commercially exploited for the management of sheath blight of rice.

Abstract

Bud necrosis and chlorotic spots causing virus affecting chilli crop in Tamil Nadu (India) was identified as Capsicum chlorosis virus (CaCV). Specific primers were used for amplification and sequencing of the nucleocapsid protein (NP) gene. Polyclonal antibody against the bacterially expressed NP from the CaCV-TN-CBE isolate was produced using recombinant DNA technology. NP gene was subcloned into the pET-28a (+) vector and expressed by transformation in BL21 (DE3) pLysS. The expressed protein was about ∼34 kDa and was confirmed through western blot analysis using Groundnut bud necrosis virus (GBNV) polyclonal antiserum from ICRISAT, India. The purified recombinant protein was used to immunize rabbits to generate CaCV-specific polyclonal antiserum. The sensitivity levels of polyclonal antiserum thus raised was assayed through indirect ELISA or direct antigen coating (DAC)-ELISA using the recombinant protein as antigen. The recombinant antiserum produced in this study successfully detected the natural infection of CaCV on chilli plants collected from the field as well as on cowpea plants artificially inoculated with CaCV by using DAC-ELISA, DIBA and western blotting.

12. Efficacy of fungicides, botanicals and bioagents against Rhizoctonia solani inciting leaf blight on turmeric (Curcuma longa L.)

Abstract

Leaf blight caused by Rhizoctonia solani is one of the most serious diseases that cause damage to turmeric crop. Fifty percent disease incidence was observed in turmeric growing areas of Andhiyur (Erode district) and Mettur (Salem district) of Tamil Nadu, India. The antifungal efficacy of botanicals, bioagents and fungicides were tested against R. solani in vitro. It was observed that seed extract and oil cake of Madhuca longifolia suppressed the mycelia growth (34.81; 49.63%), sclerotial formation (100%) and production of biomass (2.18; 2.41 mg) of R. solani followed by leaf extracts of Azadirachta indica and Littorina littorea. Among the bioagents native isolate of BSoya1 of Bacillus spp., Cg1 of Chaetomium spp., Pf1 of Pseudomonas spp. and Tv1 of Trichoderma spp. shown more than 50% antagonistic effect on mycelia growth and 100% on sclerotial formation. The complete fungal growth inhibition was observed in Nativo and Bavistin fungicides amended medium. Then, the basal soil application of mahua cake and T. viride in the ratio of 1:2 (1 g of mahua cake mixed with 2 g T. viride per kg of pot soil) followed by foliar spray of Nativo @ 0.5% significantly mitigated the leaf blight (8.6%) over the inoculated control (50.51%) in pot culture experiments. Abstract

Nomenclatural type definitions are one of the most important concepts in biological nomenclature. Being physical objects that can be re-studied by other researchers, types permanently link taxonomy (an artificial agreement to classify biological diversity) with nomenclature (an artificial agreement to name biological diversity). Two proposals to amend the International Code of Nomenclature for algae, fungi, and plants (ICN), allowing DNA sequences alone (of any region and extent) to serve as types of taxon names for voucherless fungi (mainly putative taxa from environmental DNA sequences), have been submitted to be voted on at the 11th International Mycological Congress (Puerto Rico, July 2018). We consider various genetic processes affecting the distribution of alleles among taxa and find that alleles may not consistently and uniquely represent the species within which they are contained. Should the proposals be accepted, the meaning of nomenclatural types would change in a fundamental way from physical objects as sources of data to the data themselves. Such changes are conducive to irreproducible science, the potential typification on artefactual data, and massive creation of names with low information content, ultimately causing nomenclatural instability and unnecessary work for future researchers that would stall future explorations of fungal diversity. We conclude that the acceptance of DNA sequences alone as types of names of taxa, under the terms used in the current proposals, is unnecessary and would not solve the problem of naming putative taxa known only from DNA sequences in a scientifically defensible way. Abstract

Yellow mosaic disease is the major limitation in the production of grain legumes in India. This disease is caused by bipartite begomovirus, Mungbean yellow mosaic virus. In addition to the bipartite genomic components, the yellow mosaic disease affected urdbean plants which contain satellite like DNA-1 component called as alphasatellites. The present study has been attempted to characterise the alphasatellites associated with Mungbean yellow mosaic virus. Nucleotide sequence analysis of alphasatellites showed 98% identity with Vernonia yellow vein Fijian alphasatellite, VYVFA (JF733780). Since the sequence identity is more than 98%, the threshold value for demarcation of alphasatellites species, the alphasatellites of the present study are named as Vernonia yellow vein Fijian alphasatellite. Comparison with other, alphasatellites shared 51–55% identity with alphasatellites associated with monopartite begomovirus and it shared only 41–42% identity with an unusual alphasatellites, DNA-2. Abstract

Field experiments were conducted to ascertain the efficacy of leaf extracts from botanical species viz., Vitex negundo (10%), Psoralea corylifola (10 %), Winter Green oil 30 EC (2%) against Sterility Mosaic Disease (SMD) in pigeonpea under field conditions. Resistant inducing chemicals viz., 200 ppm Salicylic acid and acaricides viz ; 0 .1% Fenazaquin and 0. 2% Dicofol were also evaluated against SMD. Comprehensive analysis of data demonstrated that the two sprays of 0.1% Fenazaquin first at 30 days after sowing and second at 15 days after first spray recorded the lowest (8.8 per cent) SMD incidence as compared to 46.1 per cent in untreated control which accounted for the disease reduction of 80.9 per cent. Among the plant products tested, winter green oil 30 EC (2 %) was effective by recording 19.0 per cent SMD incidence with 58.7 per cent disease reduction. Fenazaquin (0 .1%) spray reduced the mite (Aceria cajani) population by 81.9 per cent followed by winter green oil 30 EC (2 %) with the population reduction of 72.3 per cent. Fenazaquin (0 .1%) spray also influenced to record the highest yield of 815 kg per ha as against 492 kg per ha in untreated plot and therefore, 0.1% Fenazaquin could be the best alternative for the management of SMD in pigeonpea under field conditions.

Abstract

Fungicidal activity of azoxystrobin on Pyricularia grisea (Cooke) Sacc, causing blast disease in rice was investigated under in vitro and in vivo conditions. The results of the experiments revealed that under in vitro conditions, azoxystrobin at 1, 2 and 4 ppm concentrations completely (100%) restricted mycelial growth of P. grisea by inhibiting the mitochondrial respiration of the organism and causing cell death. This was also found to be true in case of field experiments. Considering the systemic, translaminar and rainfastness properties of azoxystrobin, three foliar sprays (one at initial appearance of disease and subsequently two sprays at 15 days intervals) were administered under in vivo conditions. The results showed that there was hardly any phytotoxicity effect at different concentrations of azoxystrobin. Abstract

Biological and nutrient management of soil borne disease is increasingly gaining stature as a possible practical and safe approach. Inhibitory effects of fungal and bacterial antagonists were tested under in vitro conditions against the wilt pathogen of alfalfa Fusarium oxysporum f. sp. medicaginis. Trichoderma harzianum and Pseudomonas fluorescens (PI 5) were found to be effective against the alfalfa wilt pathogen. Manganese sulphate at 500 and 750 ppm inhibited the mycelial growth of F. oxysporumf. sp. medicaginis under in vitro conditions. In pot culture studies, manganese sulphate at 12.5 mg/kg reduced the wilt incidence (23.33%). Combined application of manganese sulphate 12.5 mg/kg + T. harzianum 1.25 mg/kg of soil significantly reduced the wilt incidence accompanied by improved plant growth and yield in pot culture. Abstract

Three antagonists: Pseudomonas fluorescens (Pf1), Bacillus subtilis and Trichoderma viride, were tested alone and in combination for suppression of onion leaf blight (Alternaria palandui) disease under glasshouse and field conditions. The average mean of disease reduction was 24.81% for single strains and 42.44% for mixtures. In addition to disease suppression, treatment with a mixture of antagonists promoted plant growth in terms of increased plant height and ultimately bulb yield. Though seed treatment of either single strain or strain mixtures alone could reduce the disease, subsequent application to root, leaves or soil further reduced the disease and enhanced the plant growth. The mixture consisting of Pseudomonas fluorescens Pf1 plus Bacillus subtilis plus Trichoderma viride was the most effective in reducing the disease and in promoting plant growth and bulb yield in greenhouse and field tests.

Abstract

Azoxystrobin residue was determined in mango fruits using high performance liquid chromatography (HPLC). Residues of azoxystrobin were extracted with acetonitrile and purified by both liquid liquid extraction (LLE) and solid phase extraction (SPE) clean up. The HPLC analysis was carried out on ODS2 column with acetonitrile: water (80:20 v/v) as the mobile phase with UV detection at 255 nm. The rate of average recoveries and limits of determinations were 85.57 % and 0.004 µg per g of sample, respectively. The results revealed that the half life of azoxystrobin in mango fruit was one day for recommended dose, hence no concern regarding contamination of the food chain and environment by azoxystrobin. O resíduo de azoxistrobina em mangas foi determinado por cromatografia líquida de alta performance (HPLC). O resíduo foi extraído com acetonitrila e purificado tanto por extração líquido-líquido (LLE) quanto por extração de fase sólida (SPE). A análise cromatográfica foi realizada em uma coluna ODS2 com fase móvel acetonitrila: água 80:20 v/v e detecção por UV em 255 nm. As recuperações médias e limites de determinação foram iguais a 85.57 % e 0.004 µg g-1 de amostra, respectivamente. Os resultados reveleram que a meia vida da azoxistrobina foi igual a um dia, para a dose recomendada. Portanto, não há problemas de contaminação da cadeia alimentar e do meio ambiente pela adição de azoxistrobina em mangas.

Abstract

This study was conducted to analyse the induction of lignification-related enzymes and phenolic content in rice to blast disease caused by Pyricularia grisea using azoxystrobin. The severity of rice blast was reduced (70% over control) through treatment by azoxystrobin. This reduction in disease severity was mainly associated with induction of host defense mechanisms by azoxystrobin. Increased production of secondary metabolite – phenolic and lignification – related enzymes, namely, peroxidase (POD), polyphenol oxidase (PPO) and phenylalanine ammonia-lyase (PAL) were observed in rice plants treated with azoxystrobin.

Abstract

In this Letter, comprehensive results on π±, K±, KoS, p(p) and A(A) production at mid-rapidity (0 < yCMS < 0.5) in p-Pb collisions at √SNN = 5.02 TeV, measured by the ALICE detector at the LHC, are reported. The transverse momentum distributions exhibit a hardening as a function of event multiplicity, which is stronger for heavier particles. This behavior is similar to what has been observed in pp and Pb-Pb collisions at the LHC. The measured pT distributions are compared to d-Au, Au-Au and Pb-Pb results at lower energy and with predictions based on QCD-inspired and hydrodynamic models. © 2013 The Authors. Abstract

Yellow mosaic disease caused by mungbean yellow mosaic virus (MYMV) belonging to the genus Begomovirus (the family Geminiviridae) is a major constraint in cultivation of grain legumes in India. The urdbean (Vigna mungo (L.) Hepper) and mungbean (Vigna radiata (L.) R. Wilczek) samples affected with yellow mosaic disease exhibits yellow mosaic symptoms along with leaf puckering and leaf distortion in Tamil Nadu. Hence the study was performed to find out if there was any association and influence of betasatellite DNA on the symptom expression of MYMV. Full length viral clones of DNA A and DNA B were obtained through rolling circle amplification from YMD infected samples and identified as mungbean yellow mosaic virus. Interestingly, betasatellite was found to associate with MYMV, and its nucleotide sequence analysis showed its 95% identity with papaya leaf curl betasatellite (DQ118862) from cowpea. The present study represents the first report about the association of papaya leaf curl betasatellite with MYMV and represents a new member of the emerging group of bipartite begomovirus associated with betasatellite DNA.

Abstract

Abatract: The present study explored pathogenic and genetic variability among the eleven isolates of Rhizoctonia bataticola (Taub.) Butler from different pulse crops. Based on morphological characters, 11 isolates were categorized into three groups viz., linear, fluffy, and linear at the end with fluffy growth at the center. Isolates also showed variability in sclerotial characters (intensity and shape) and intensity of pigment synthesis. All isolates were more aggressive on the original host from which it was isolated, which was shown by the variability in pathogenic characters. RAPD-PCR analysis has shown that genetic clustering agreed with the above findings in dendrogram analysis (2 clusters A and B). The black gram root isolates showed a maximum genetic similarity of 73 % with soya bean shoot isolate. Red gram shoot isolate showed 61% genetic similarity with green gram isolates. The findings from this study confirm the variability in R. bataticola isolates from pulses, according to their pathological as well as genetic characters. In the future, variability in pathogens will determine effective management practices.

Abstract

Superstition's impingement on conventional cognition and its transformation to generations are evidently explored in this study. Superstitions as beliefs, rituals, community laws, folklore and urban legend suppress potential knowledge system of Tamil Nadu. This research was reinforced with contagion theory, which proves instinctive influence of group behaviour on an individual's behaviour. In addition, it identifies the gap between the reasons behind the greatly influenced uncontrollable group behavioural superstition. This research negates the knowledge gap theory that verifies increased knowledge gap between two groups of socioeconomic status. This study intricate cross-sectional face-to-face survey of 300 individuals, who are exposed towards the communication interventions of social organizations. In-depth semi-structured interviews from organizational members were conducted to discover the extent of destruction of traditional knowledge systems and its impact in Tamil Nadu. Largely, reveals the reality behind the traditional-religious superstition and steps taken by the social organization towards superstition eradication and locates remarkable and astounding facts on superstitious impact on conventional cognition of Tamil Nadu.

Abstract

Detection of Capsicum chlorosis virus (CaCV), An Emerging Virus Infecting Chilli in Tamil Nadu, India Chilli plants in the farmers’ fields in Tamil Nadu showing symptoms of concentric chlorotic and necrotic rings on the leaves were found infected with a tospo virus through RT- PCR by using tospo virus degenerate primer (gL3637 /F and gL4435C). From the BLAST result the virus associated with the disease is identified as Capsicum chlorosis virus (CaCV). For further confirmation, infected samples collected from Coimbatore were amplified using newly designed primer pair (GKCaCVCPF1/R1) corresponding to the nucleocapsid region of the CaCV for further confirmation. In the sequence analysis, complete nucleocapsid protein region shared a nucleotide and amino acid identity of 99.1% and 100% with the PB4 and PB1 isolates of CaCV from India respectively. In the phylogenetic analysis, this isolate grouped with the CaCV isolates from Aurangabad and Tamil Nadu, India than with isolates from other countries. About fifteen hosts from different families were inoculated through sap, of which nine hosts produce local lesion and four showed systemic symptoms. The positive reaction of extract from these plants in DAS- ELISA (double antibody sandwich ELISA) against the Nucleocapsid protein of CaCV (DSMZ, Germany), confirmed the presence of CaCV in infected chilli plants in Tamil Nadu..

Abstract

The Pseudomonas fluorescens isolate Pfl was found to inhibit the growth of pathogen Alternaria palandui, in vitro. In the present study, foliar application of a talc-based formulation of Pfl significantly reduced the incidence of leaf blight of onion, caused by A. palandui. Induction of defense-related proteins viz., chitinase, beta-1,3 glucanase, peroxidase (PO) and polyphenol oxidase (PPO) by application of Pfl, was studied against A. palandui infection in resistant (IHR 56) and susceptible (MDUI) onion cultivars. Chitinase in both cultivars, with or without challenge-inoculation of A. palandui revealed changes in the isoform pattern. The Native-PAGE of PO showed induction of PO2 isoform in both the cultivars, in response to inoculation of pathogen. Isoform analysis of PPO also exhibited induction in the Pfl-treated plants challenged with pathogen. Similarly, the activity of beta-1,3-glucanase was greatly induced in Pfl-treated plants, challenged with pathogen as compared to controls. Thus, the P. fluorescens-treated plants showed significant increase in the levels of the defense enzymes, in comparison to the plants challenged with the pathogen.

Abstract

Four antibiotics were tested against Erwinia causing soft rot of onion (Allium cepa var. aggregatum) of which streptomycin sulphate 90% and tetracycline hydrochloride 10% (streptocycline) recorded the maximum inhibition zone of 27.66 mm. In the enzyme studies the maximum inhibition of pectinlyase (PL), polygalacturonase (PG) and protopectinase production was recorded by the same antibiotic. The antibiotics have a significant influence on the production and activity of cell wall degrading enzymes produced by the plant pathogenic microorganisms. Garlic clove extract was equally effective in inhibiting the growth and enzyme production.

Abstract

Nutritional knowledge, attitudes, and food patterns of women athletes at the University of Nebraska at Lincoln were studied. Mean test scores for nutritional knowledge were higher for questions relating to "nutrition for the athlete" than to "general nutrition." Attitudes toward nutrition were generally favorable, with a positive correlation between nutritional knowledge and attitudes. Athletes who had received some form of nutrition education had higher nutritional knowledge and attitude scores than those who had not. Respondents were especially concerned with diet as it relates to weight control; those with higher knowledge scores consistently followed food patterns for weight-control diets similar to those used in training and pre-event diets.

Abstract

A data collection tool was developed to teach nursing students basic principles of nutritional assessment. The tool was used to help nurses coordinate and interpret data they routinely collected so they would be more aware of real and potential nutritional problems. Emphasis was on those aspects of nutritional care relevant to the nurse's advocates in communicating these concerns to the dietitian. Observation and early referral of potential diet-related problems enables the dietitian to more effectively provide for nutritional care.

Abstract

To promote nutrition and dietitians, the Nebraska Dietetic Association approached various other associations in the state about featuring articles on nutrition in their newsletters. This public relations strategy permitted greater dissemination of nutritional information to specific target groups through existing channels of communication. Dietitians were presented to other professional groups as nutrition experts. In addition, this activity opened the door to further professional exchanges between dietitians and the various associations.
