$\gamma p \rightarrow K^+ \Lambda$ differential cross sections and recoil polarisation data from threshold for extremely forward angles are presented. The measurements were performed at the BGOOD experiment at ELSA, utilising the high angular and momentum resolution forward spectrometer for charged particle identification. The BGO-OD experiment at the ELSA accelerator facility uses an energy tagged bremsstrahlung photon beam to investigate the excitation structure of the nucleon via meson photoproduction. The setup with a BGO calorimeter surrounding the target and an open dipole spectrometer covering the for ward region is ideally suited for investigating low momentum transfer processes, in particular in strangeness photoproduction. The associated photoproduction of K0S and hyperons is essential to understand the role of K* exchange mech anisms. A cusp-like structure observed in the yp → K0SΣ+ reaction at the K* threshold is described by models including dynamically generated resonances from vector meson-baryon interactions. Such interactions are pre dicted to give a peak like structure in K0SΣ0 photoproduction off the neutron. A very preliminary cross section is determined and compared to the prediction, the results appear to support the model

First key results at low $t$ indicate a cusp-like structure in $K^+\Sigma^0$ photoproduction at $W = 1900$\,MeV, line shapes and differential cross sections for $K^+\Lambda$(1405)$\rightarrow K^+\Sigma^0\pi^0$, and a peak structure in $K^0_S\Sigma^0$ photoproduction. The peak in the $K^0_S\Sigma^0$ channel appears consistent with meson-baryon generated states, where equivalent models have been used to describe the $P_C$ pentaquark candidates in the heavy charmed quark sector., Comment: Proceedings of The 12th International Workshop on the Physics of Excited Nucleons (NSTAR 2019)

The unexpected nodal structure of the beam asymmetry recently reported by the GRAAL collaboration in η′ photoproduction very close to threshold could be explained by a previously unobserved narrow resonance. The BGO-OD experiment is ideally suited to verify this measurement via the detection of forward going charged particles which in the threshold region of interest allows the identification of the reaction γp → η′ p solely based on the proton going in the forward direction. This yields unprecedented statistics if, in the missing mass analysis of the η′ meson, the background can be sufficiently well controlled. Preliminary results using a linearly polarised photon beam are shown. The reaction γp → η′ p was identified in the BGO forward spectrometer, with simulated data used to seperate signal and background.

The γ p → K + Σ 0 differential cross section at extremely forward angles was measured at the BGOOD experiment. A three-quarter drop in strength over a narrow range in energy and a strong dependence on the polar angle of the K + in the centre-of-mass of the reaction is observed at a centre-of-mass energy of 1900 MeV. Residing close to multiple open and hidden strangeness thresholds, the structure appears consistent with meson-baryon threshold effects which may contribute to the reaction mechanism.

Since the discovery of the Λ(1405), it remains poorly described by conventional constituent quark models, and it is a candidate for having an “exotic” meson-baryon or “penta-quark” structure, similar to states recently reported in the hidden charm sector.The Λ(1405) can be produced in the reactionγpK+Λ(1405). The pure I=0 decay mode into Σ0π0is prohibited for the mass-overlapping Σ(1385). Combining a large aperture forward magnetic spectrometer and a central BGO crystal calorimeter, the BGO-OD experiment is ideally suited to measure this decay with theK+in the forward direction. The BGO-OD experiment at the ELSA accelerator facility uses an energy tagged bremstrahlung photon beam to investigate the excitation spectrum of the nucleon. The setup consists of a highly segmented BGO calorimeter surrounding the target, with a particle tracking magnetic spectrometer at forward angles. BGO-OD is ideal for investigating the photoproduction of hadrons of non-zero strangeness. The high momentum resolution at forward angles covers a kinematic region where t-channel exchange mechanisms play a dominant role. Access to this low momentum transfer region also allows the investigation of degrees of freedom not derived from constituent quark models, for example, the role of (vector) meson - baryon interactions and dynamically generated states in photoproduction reactions. Data taking for the first part of an extensive physics programme is complete and preliminary results and ongoing analysis are presented. (Less)

Photoproduction of $\eta$ mesons from neutrons} \abstract{Results from measurements of the photoproduction of $\eta$ mesons from quasifree protons and neutrons are summarized. The experiments were performed with the CBELSA/TAPS detector at the electron accelerator ELSA in Bonn using the $\eta\to3\pi^{0}\to6\gamma$ decay. A liquid deuterium target was used for the measurement of total cross sections and angular distributions. The results confirm earlier measurements from Bonn and the MAMI facility in Mainz about the existence of a narrow structure in the excitation function of $\gamma n\rightarrow n\eta$. The current angular distributions show a forward-backward asymmetry, which was previously not seen, but was predicted by model calculations including an additional narrow $P_{11}$ state. Furthermore, data obtained with a longitudinally polarized, deuterated butanol target and a circularly polarized photon beam were analyzed to determine the double polarization observable $E$. Both data sets together were also used to extract the helicity dependent cross sections $\sigma_{1/2}$ and $\sigma_{3/2}$. The narrow structure in the excitation function of $\gamma n\rightarrow n\eta$ appears associated with the helicity-1/2 component of the reaction.

In a search for $\omega$ mesic states, the production of $\omega$-mesons in coincidence with forward going protons has been studied in photon induced reactions on $^{12}$C for incident photon energies of 1250 - 3100 MeV. The $\pi^0 \gamma$ pairs from decays of bound or quasi-free $\omega$-mesons have been measured with the CBELSA/TAPS detector system in coincidence with protons registered in the MiniTAPS forward array. Structures in the total energy distribution of the $\pi^0 \gamma$ pairs, which would indicate the population and decay of bound $\omega~^{11}$B states, are not observed. The $\pi^0 \gamma$ cross section of 0.3 nb/MeV/sr observed in the bound state energy regime between -100 and 0 MeV may be accounted for by yield leaking into the bound state regime because of the large in-medium width of the $\omega$-meson. A comparison of the measured total energy distribution with calculations suggests the real part $V_0$ of the $\omega~^{11}$B potential to be small and only weakly attractive with $V_0(\rho=\rho_0) = -15\pm$ 35(stat) $\pm$20(syst) MeV in contrast to some theoretical predictions of attractive potentials with a depth of 100 - 150 MeV., Comment: 13 pages, 8 figures

The excitation function and momentum distribution of $\eta^\prime$ mesons have been measured in photon induced reactions on $^{12}{}$C in the energy range of 1250-2600 MeV. The experiment was performed with tagged photon beams from the ELSA electron accelerator using the Crystal Barrel and TAPS detectors. The data are compared to model calculations to extract information on the sign and magnitude of the real part of the $\eta^\prime$-nucleus potential. Within the model, the comparison indicates an attractive potential of -($37 \pm 10(stat)\pm10(syst)$) MeV depth at normal nuclear matter density. Since the modulus of this depth is larger than the modulus of the imaginary part of the $\eta^\prime$-nucleus potential of -($10\pm2.5$) MeV, determined by transparency ratio measurements, a search for resolved $\eta^\prime$-bound states appears promising., Comment: 13 pages, 6 figures

The new BGO-OD experiment at the electron accelerator ELSA, of the University of Bonn, is designed to study the reaction dynamics of nucleon excitations in meson photoproduction. It consists of a central BGO calorimeter with a magnetic spectrometer in forward direction. The physics programme includes the measurement of polarisation observables using linearly and circularly polarised photon beams. Linear polarisation is obtained by coherent bremsstrahlung off a diamond crystal, and circular polarisation is obtained via bremsstrahlung from longitudinally polarised electrons. The degree of linear polarisation is determined from the bremsstrahlung spectrum itself. To determine the polarisation of the circularly polarised photon beam, the polarisation of the electron beam is measured by a Moller polarimeter. As a preliminary consistency check, the (linear) polarisation observable, Σ, was compared to world data for π^0 and η photoproduction. To determine the degree of circular polarisation, a Moller polarimeter was setup and first measurements of the electron beam polarisation performed.

BGO-OD is a new meson photoproduction experiment at the ELSA facility of Bonn University. It aims at the investigation of non strange and strange baryon excitations, and is especially designed to be able to detect weekly bound meson-baryon type structures. The setup for the BGO-OD experiment is presented, the characteristics of the photon beam and the detector performances are shown and the initial experimental program is discussed.

This paper reports on a measurement of the double-polarization observable G in $\pi^0$ photoproduction off the proton using the CBELSA/TAPS experiment at the ELSA accelerator in Bonn. The observable G is determined from reactions of linearly-polarized photons with longitudinally-polarized protons. The polarized photons are produced by bremsstrahlung off a properly oriented diamond radiator. A frozen spin butanol target provides the polarized protons. The data cover the photon energy range from 617 to 1325 MeV and a wide angular range. The experimental results for G are compared to predictions by the Bonn-Gatchina (BnGa), J\"ulich-Bonn (J\"uBo), MAID and SAID partial wave analyses. Implications of the new data for the pion photoproduction multipoles are discussed.

Bombesin/gastrin-releasing peptide (BN/GRP) antagonists RC-3940-II and RC-3940-Et, and growth hormone-releasing hormone (GHRH) antagonists MZ-J-7-118 and RC-J-29-18 inhibit the growth of human androgen-independent PC-3 and DU-145 prostate cancers in nude mice. Additive inhibitory effects were observed after treatment with both classes of analogs. In the present study, we investigated the effects of these antagonists on intracellular signalling pathways of protein kinase C (PKC), mitogen activated protein kinases (MAPK) and c-fos and c-jun oncogenes that are involved in tumour cell proliferation. In PC-3 tumours, antagonists of BN/GRP and GHRH decreased significantly the expression of PKC isoforms alpha (alpha), eta (eta) and zeta (zeta) and increased that of delta (delta) PKC protein. MAPK was not detectable. In DU-145 tumours, which constitutively express MAPK, all treatments strongly decreased the levels of p42/44 MAPK. Treatment with the antagonists tended to reduce m-RNA for c-jun in both tumour models. In proliferation assays in vitro, inhibitors of PKC and MAPK diminished growth of DU-145 and PC-3 cells. These findings suggest that antagonists of BN/GRP and GHRH inhibit the growth of androgen-independent prostate cancer by affecting intracellular signalling mechanisms of PKC, MAPK and c-jun.

BACKGROUND Antagonists of growth hormone releasing hormone (GHRH) as well as antagonists of bombesin/gastrin releasing peptide (BN/GRP) inhibit the growth of various malignancies (cancers) including prostate cancer. METHODS We investigated the effects of GHRH antagonists MZ-J-7-118 and RC-J-29-18, BN/GRP antagonists RC-3940-II and RC-3940-Et and the combination of MZ-J-7-118 and RC-3940-II on the growth of PC-3 and DU-145 human androgen independent prostate cancers xenografted s.c. into nude mice. To elucidate the mechanisms of action of these analogs, growth factors like IGF-II (insulin-like growth factor-II), vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF), and epidermal growth factor receptor/human epidermal growth factor receptor (EGF-R/HER) family were measured in tumors as well as IGF-I in serum. RESULTS Antagonists of GHRH and BN/GRP alone or in combination significantly inhibited growth of PC-3 and DU-145 tumors, the greatest inhibition of tumor volume being achieved by combination of MZ-J-7-118 (5 μg/day) and RC-3940-II (10 μg/day). BN/GRP and GHRH antagonists and their combination also decreased the expression of VEGF significantly in PC-3 and non-significantly in DU-145, as measured by radioimmunoassay for VEGF protein and RT-PCR for mRNA levels of VEGF. GHRH and BN/GRP antagonists reduced bFGF concentrations and the maximal binding capacity of EGF receptors, and their mRNA levels in PC-3 and DU-145 tumors. mRNA levels for HER-2 and -3 were also diminished in PC-3 tumors by GHRH and BN/GRP antagonists. No changes in HER-4 were found after treatment. Serum IGF-I and tumoral IGF-II levels were not affected by the analogs. CONCLUSIONS BN/GRP and GHRH antagonists inhibit growth of PC-3 and DU-145 prostate cancers by suppressing the expression of tumoral growth factors such as VEGF and bFGF as well as the receptors for EGF and related HER-2 and -3. Additive effects on tumor inhibition (TI) in vivo, but not on VEGF, bFGF, or members of the EGF/HER receptor family, can be achieved by the joint administration of both classes of analogs. © 2005 Wiley-Liss, Inc.

We investigated the effects of antagonists of growth hormone-releasing hormone (GHRH) alone and in combination with bombesin/gastrin-releasing peptide (BN/GRP) antagonist RC-3940-II on the growth of H-69 human small cell lung carcinoma (SCLC) xenografted into nude mice. Since the activation of the signaling pathways involving protein kinase C (PKC) and the subsequent steps involving mitogen-activated protein kinase (MAPK) and c-fos and c-jun oncogenes are known to be important mechanisms implicated in cellular growth, we investigated how the blockade of tumoral GHRH receptor splice variants and BN/GRP receptors by these antagonists could interfere with these intracellular signaling pathways. The results obtained at the BGO-OD experiment with the BGO calorimeter, equipped with the new electronic readout based on sampling ADCs, during the tests performed with the beam time of March and June 2012 are presented. The proper functioning of the apparatus has allowed the reconstruction of the pseudo-scalar mesons π0 and η invariant masses. The simulation of the η' photoproduction reaction prepared for a proposal to the joint ELSA-MAMI Physics Advisory Committee is also presented.

New therapeutic strategies are necessary to improve the treatment of lung cancer. We investigated the effects of bombesin/gastrin-releasing peptide (GRP) antagonist, RC-3940-II, and growth hormone-releasing hormone (GHRH) antagonists, MZ-J-7-114 and MZ-J-7-118, on the expression of epidermal growth factor receptor (EGFR)/HER (-2, -3, and -4) family, angiogenic factors, VEGF-A and VEGF receptors (VEGF-R1 and VEGF-R2), and the apoptotic molecules Bax and Bcl-2, in H-460 and A-549 non-small cell lung carcinomas (NSCLC). Nude mice bearing xenografts of H-460 and A-549 NSCLC were treated daily with these peptide analogues for 4 weeks. The treatment resulted in growth inhibition of H-460 by 22-77% and A-549 NSCLCs by 64-84%. The inhibition of tumor growth was associated with a down-regulation of members of EGFR/HER family. A significant reduction of the levels of expression of EGFR/HER family on both tumors varied from 29-96%: the greatest inhibition being induced by RC-3940-II. Similarly, a significant decrease in the levels of VEGF-A in tumors by 19-60% and VEGF receptors (VEGF-R1, 24-74% and VEGF-R2, 25-50%) was detected after therapy. An up-regulation of Bax by 21-63% and a down-regulation of Bcl-2 by 23-39% was observed only for H-460 NSCLC. Our study demonstrates that human H-460 and A-549 NSCLC, express receptors for GHRH and bombesin/GRP, and respond to the respective antagonists. The antagonists of bombesin/GRP and GHRH could provide a new strategy for treatment of NSCLC through down-regulation of EGFR/HER family and an interference with the angiogenic and apoptotic pathways.

Purpose: To determine whether the cytotoxic analogue of bombesin/gastrin-releasing peptide (GRP) AN-215 can inhibit the in vivo growth of four human ovarian cancer cell lines. AN-215 consists of 2-pyrrolinodoxorubicin (AN-201), a superactive derivative of doxorubicin linked to a bombesin antagonist carrier des-D-Tpi-RC-3095. This conjugate binds strongly to receptors for bombesin/GRP and can be targeted to tumors that express these receptors. Bombesin/GRP receptors are found in 77% of human ovarian cancer specimens. Experimental Design: Nude mice bearing xenografts of ES-2, SKOV-3, OV-1063, and UCI-107 human ovarian carcinomas were treated with AN-215. The antitumor effects and the toxicity were determined. The expression of bombesin receptor subtypes was measured by reverse-transcriptase PCR analysis, and the presence of bombesin/GRP receptors was determined by radioligand binding assays. Results: AN-215 significantly (P < 0.05) inhibited growth of ES-2, OV-1063, and UCI-107 tumors, prevented the metastatic spread of ES-2 cancers, and prolonged the survival of nude mice bearing i.p. ES-2 xenografts. Cytotoxic radical AN-201, the unconjugated mixture of bombesin antagonist RC-3095 and AN-201 or RC-3095 alone had no significant effects. Blockade of bombesin/GRP receptors abolished the effect of AN-215. The expression of bombesin/GRP receptors was not changed after repeated treatment with AN-215. Conclusions: Our findings indicate that targeted chemotherapy with cytotoxic bombesin/GRP analogue AN-215 can inhibit ovarian tumors, which express bombesin/GRP receptors. AN-215 might provide a new treatment modality for women with advanced ovarian carcinoma.

The cytotoxic analogue of somatostatin, AN-238, consisting of 2-pyrrolinodoxorubicin (AN-201), a superactive derivative of doxorubicin (DOX), linked to somatostatin analogue carrier RC-121 binds with high affinity to receptors for somatostatin and can be targeted to tumors that express these receptors. Because somatostatin receptors are found in a high percentage of human non-Hodgkin's lymphomas (NHLs), we evaluated the antitumor effect of AN-238 in 2 human NHL cell lines in vivo. Nude mice bearing xenografts of RL and HT human NHL were treated with AN-238 or its components at equimolar doses, and antitumor effects were determined. Expression of mRNA for somatostatin receptor subtypes was measured by RT-PCR, and the presence of somatostatin receptors was determined by radioligand binding. Toxicity was evaluated by following white blood cell count (WBC) and body weight. AN-238 significantly (p < 0.05) inhibited growth of RL and HT xenografts and prolonged the tumor doubling time. Cytotoxic radical AN-201, the unconjugated mixture of somatostatin analogue RC-121 and AN-201 or RC-121 alone had no significant effects. Blockade of somatostatin receptors by excess RC-121 abolished the effect of AN-238, demonstrating targeting. Expression of somatostatin receptors was not changed after repeated treatment with AN-238. AN-201, but not AN-238, significantly lowered the WBC and caused a greater decrease in body weight than AN-238. Our findings demonstrate that targeted chemotherapy with AN-238 can strongly inhibit the growth of NHL cells, which express somatostatin receptors. AN-238 could be considered for the treatment for patients with NHL.

Effects of in vivo treatment with antagonists of growth hormone-releasing hormone (GHRH), JV-1-65 and MZ-J-7-110, and bombesin/gastrin-releasing peptide antagonist RC-3940-II, on the EGF receptor (EGFR) family, were investigated in H-69 SCLC. Tumors were analyzed by RT-PCR, immunoblotting and binding assays. Treatment with these analogs reduced the binding capacity of EGFR by 18-64%, and inhibited the mRNA expression for EGFR, HER-2 and -3 by 27-75.4, 17-26.3, and 13.8-46.6%, respectively. The antagonists also decreased the protein levels for EGFR by 21-34%, HER-2 by 36-68% and HER-3 by 43-49%. This is the first demonstration that antiproliferative effects of GHRH antagonists are associated with a downregulation of EGF/HER receptors.

Meson photoproduction is a key tool for the experimental investigation of the nucleon excitation spectrum. To disentangle the specific couplings of resonances, in addition to the rather well measured pion and eta photoproduction channels it is mandatory to obtain information on channels involving strange and vector mesons and higher mass pseudoscalar mesons, and the associated multi-particle final states with both charged and neutral particles. In this respect, the new BGO-OD experiment at the ELSA accelerator of the University of Bonn's Physikalisches Institut provides unique instrumentation. We describe the experiment, present its status and the initial program of measurements.

The effects of short episodes (1 min) of vagally-mediated bradycardia were studied in 9 anesthetized dogs utilizing vagal stimulation and slow atrial pacing (120 and 80 beats/min) before and after graded coronary constriction of the left anterior descending (LAD) and the left circumflex (CCA). In the presence of 90% LAD stenosis, bradycardia tended to restore both the elevated total LAD coronary vascular resistance (CVR-LAD) and the reduced, total CVR-CCA towards control levels obtained at corresponding slow rates in the absence of coronary stenosis; as a result, LAD coronary flow (F-LAD) was relatively less reduced and the accessory rise of F-CCA disappeared. In the presence of combination of 90% LAD plus 70% CCA stenosis, the effects of bradycardia on total CVR-LAD and F-LAD were similar to those obtained with single 90% LAD stenosis, but the accessory flow through the CCA was abolished resulting in no significant difference of the rate-dependent alterations of total CVR-CCA and F-CCA as compared with those observed in the absence of coronary stenosis. In the presence of single or combined coronary stenosis, bradycardia restored the depressed aortic pressure and cardiac output towards control values obtained at comparable slow rates before coronary stenosis. The results support the concept that in the presence of 90% LAD stenosis vagally-mediated bradycardia 1) decreases the tension-time index (myocardial nutritional demand) shifting cardiac performance to less expensive "flow work" and 2) facilitates antegrade flow through the highly stenotic LAD thereby inhibiting accessory flow through the nonstenotic CCA.

The effect of short bouts (1 min) of electrically induced ventricular tachycardias (VT) of increasing rates (160-240/min) was studied in 8 anesthetized dogs before and after graded constrictions of the left anterior descending (LAD) and the circumflex (CCA) coronary arteries. In the absence of coronary stenosis, paroxysmal VT caused a significant decrease in tension-time index (TTI), coronary blood flow (CBF) and coronary vascular resistance (CVR). Single and combined coronary stenosis caused relatively small alterations of the VT-induced depression of the systemic hemodynamics but reversed the effect of paroxysmal VT on the CVR. In the presence of single 90% LAD stenosis, VT resulted in an increase in CVR-LAD and a decrease in F-LAD associated with a fall in CVR-CCA and a rise in F-CCA. Combination of 90% LAD plus 70% CCA stenosis abolished the compensatory fall of CVR-CCA resulting in a pronounced reduction of F-CCA during VT. The results support the concept that in the presence of severe coronary stenosis brief paroxysmal ventricular tachycardias do increase myocardial nutritional demand but rather decrease nutritional supply.

The BGOOD experiment at the ELSA facility in Bonn has been commissioned within the framework of an international collaboration. The experiment pursues a systematic investigation of non-strange and strange meson photoproduction, in particular $t$-channel processes at low momentum transfer.
