Abstract Multiple myeloma (MM) remains incurable despite the availability of novel agents. This multi-center retrospective cohort study used the Canadian Myeloma Research Group Database to describe real-world outcomes of patients withanti-CD38 monoclonal antibody (mAb) refractory MM subsequently treated with standard of care (SoC) regimens. Patients with triple class refractory (TCR) disease (refractory to a proteasome inhibitor, immunomodulatory drug, and anti-CD38 mAb) were examined as a distinct cohort. Overall, 663 patients had disease progression on anti-CD38 mAb therapy, 466 received further treatment (346 with SoC regimens were included, 120 with investigational agents on clinical trial and were excluded). The median age at initiation of subsequent SoC therapy of 67.9 (range 39.6–89.6) years with a median of 3 prior lines (range 1–9). The median PFS and OS from the start of subsequent therapy was 4.6 (95% CI 4.1–5.6) months and 13.3 (95% CI 10.6–16.6) months, respectively. The median PFS and OS of patients with TCR disease (n = 199) was 4.4 (95% CI 3.6–5.3) months and 10.5 (95% CI 8.5–13.8) months. Our results reinforce that real-world patients with relapsed MM, particularly those with TCR disease, have dismal outcomes. There remains an urgent unmet need for the development of and access to effective therapeutics for these patients.

ABSTRACT: This study evaluated the effects of 25-hydroxycholecalciferol (25-OHD) on performance, gut health, and bone quality of broilers fed with reduced calcium (Ca) and phosphorus (P) diet during Eimeria spp. challenge. A total of 576 fourteen-day-old Cobb 500 male chicks were randomly distributed in a 2 × 2 × 2 factorial arrangement, with 6 replicates of 12 birds each. The main factors were 25-OHD level (0 or 3,000 IU/kg of feed), mineral level (0.84% of Ca/0.42% of P, the levels recommended for the grower phase (NOR) or 0.64% of Ca/0.22% of P (RED), and mid-high mixed Eimeria challenge or nonchallenge. 25-OHD improved phosphorus retention (P = 0.019), bone ash weight (P = 0.04), cortical bone trabecular connectivity (P = 0.043) during coccidiosis. For birds fed with reduced mineral levels, 25-OHD supplementation increased bone ash weight (P = 0.04). However, 25-OHD did not improve bone ash weight when birds were challenged and fed with reduced mineral levels. The dietary 3,000 IU of 25-OHD supplementation did not improve performance or gut morphology but support bone health during coccidiosis. Future investigations are needed for better understand 25-OHD role on bone microarchitecture and oxidative metabolism during coccidiosis.

Abstract Presented here is a magnetic hydrogel particle enabled workflow for capturing and concentrating SARS-CoV-2 from diagnostic remnant swab samples that significantly improves sequencing results using the Oxford Nanopore Technologies MinION sequencing platform. Our approach utilizes a novel affinity-based magnetic hydrogel particle, circumventing low input sample volumes and allowing for both rapid manual and automated high throughput workflows that are compatible with Nanopore sequencing. This approach enhances standard RNA extraction protocols, providing up to 40 × improvements in viral mapped reads, and improves sequencing coverage by 20–80% from lower titer diagnostic remnant samples. Furthermore, we demonstrate that this approach works for contrived influenza virus and respiratory syncytial virus samples, suggesting that it can be used to identify and improve sequencing results of multiple viruses in VTM samples. These methods can be performed manually or on a KingFisher automation platform.

Fibre-optic sensing technology has recently become popular for oil and gas extraction, mining, geotechnical engineering, and hydrogeology applications. With a successful track record in many applications, distributed acoustic sensing using straight fibre-optic cables has become a method of choice for seismic studies. However, distributed acoustic sensing using straight fibre-optic cables cannot detect off-axial strain at high incident angles (the angle between the ray and normal vector of the surface); hence, a helically wound cable design was introduced to overcome this limitation. The helically wound cable field data at the New Afton deposit in British Columbia, Canada, showed that the quality of the data is highly dependent on the incident angle and surrounding media. A 3D finite element model developed using COMSOL Multiphysics quickly and efficiently assessed the effects of various materials surrounding a helically wound cable for simple geometry for scenarios corresponding to a real deployment of such cable underground at the New Afton mine. The proposed numerical modelling workflow could be applied to more complicated scenarios (e.g., non-linear material constitutive behaviour and the effects of pore fluids). The results of this paper can be used as a guideline for analyzing the impact of surrounding media and incident angle on the response of helically wound cable, optimizing the installation of helically wound cable in various conditions, and validating boundary conditions of 3D numerical models built for analyzing complex scenarios.

Abstract Background There are significant challenges in ensuring sufficient clinician participation in quality improvement training. Clinician capability has been identified as a barrier to the delivery of evidence-based care. Clinician training is an effective strategy to address this barrier, however, there are significant challenges in ensuring adequate clinician participation in training. This study aimed to assess the extent of participation by antenatal clinicians in evidence-based training to address alcohol consumption during pregnancy, and to assess differences in participation by profession. Methods A 7-month training initiative based on six evidence-based principles was implemented in a maternity service in New South Wales, Australia. Descriptive statistics described participation in training (% attending: any training; six evidence-based principles of training; all principles). Regression analyses examined differences by profession. Results Almost all antenatal clinicians participated in some training (182/186; 98%); 69% participated in ≥1 h of training (μ = 88.2mins, SD:56.56). The proportion of clinicians participating in training that satisfied each of the six principles ranged from 35% (training from peers and experts) to 82% (training was educational and instructional). Only 7% participated in training that satisfied all principles. A significantly higher proportion of midwifery compared to medical clinicians participated in training satisfying five of the six training principles. Conclusions A training initiative based on evidence-based principles resulted in almost all clinicians receiving some training and 69% participating in at least 1 h of training. Variability between professions suggests training needs to be tailored to such groups. Further research is required to determine possible associations with care delivery outcomes. ABSTRACT: Vitamin D is essential for the metabolism of calcium (Ca) and phosphorus (P) in birds. The objective of the study was to evaluate the effect of different isoforms of dietary vitamin D on Ca and P utilization, egg quality, and bone mineralization of laying hens. A total of 42 Lohmann white laying hens at 57 wk of age were randomly assigned to 7 dietary treatments for 6 wk. Dietary treatments were: 3,000 IU/kg Vit D3 as control, and control with additional 3,000 IU/kg 25-hydroxyvitamin D3 (T1), 9,000 IU/kg 25-hydroxyvitamin D3 (T2), 3,000 IU/kg vitamin D3 (T3), 9,000 IU/kg vitamin D3 (T4), 3,000 IU/kg of vitamin D2 (T5), or 9,000 IU/kg of vitamin D2 (T6). Egg production and egg quality were measured weekly. Fecal samples were collected at weeks 2 and 6 to measure Ca and P utilization. After 6 wk, the left tibia and femurs were collected to measure bone mineral density (BMD) and bone mineral content (BMC). A 1-way ANOVA with Tukey HSD means separation test was used for statistical analysis. There were no significant differences in egg production, egg quality, BMD, or BMC of tibia and femurs among the treatments (P > 0.05). T6 significantly reduced feed intake (P < 0.05). The apparent total tract digestibility (ATTD) of Ca was higher (P < 0.012) in treatments supplemented with additional vitamin D, irrespective of forms. The ATTD of P was higher (P < 0.0001) in T5 compared to the other treatments at both time points. The utilization of Ca and P by laying hens can be improved through the addition of different isoforms of vitamin D in diets. However, additional vitamin D supplementation to laying hens, regardless of forms, had no effect on either bone mineralization or measures of egg quality.

Thirty-six borehole temperature–depth profiles were analysed to reconstruct the ground surface temperature history (GSTH) of eastern Tasmania for the past 5 centuries. We used the singular value decomposition method to invert borehole temperatures to produce temperature histories. The quality of borehole data was classified as high or low based on model misfit. The quality of the borehole data was not dependent on topography or land use. Analysis reveals that three to five high-quality borehole temperature–depth profiles were adequate to reconstruct robust paleotemperature records from any area. Average GSTH reconstructed from Tasmanian boreholes shows temperature increases about 1.2 ± 0.2 °C during the past 5 centuries. Reconstructed temperatures were consistent with meteorological records and other proxy records from Tasmania during their period of overlap. Temperature changes were greatest around the north-east coast and decreased towards the centre of Tasmania. The extension of the East Australian Current (EAC) further south and its strengthening around the north-east coast of Tasmania over the past century was considered a prime driver of warmer temperatures observed in north-east Tasmania.

In contrast to physical processes, biogeochemical processes are inherently patchy in the ocean, which affects both the observational sampling strategy and the representativeness of sparse measurements in data assimilating models. In situ observations from multiple glider deployments are analysed to characterize spatial scales of variability in both physical and biogeochemical properties, using an empirical statistical model. We find that decorrelation ranges are strongly dependent on the balance between local dynamics and mesoscale forcing. The shortest horizontal (5–10 km) and vertical (45 m) decorrelation ranges are for chlorophyll a fluorescence, whereas those variables that are a function of regional ocean and atmosphere dynamics (temperature and dissolved oxygen) result in anisotropic patterns with longer ranges along (28–37 km) than across the shelf (8–19 km). Variables affected by coastal processes (salinity and coloured dissolved organic matter) have an isotropic range similar to the baroclinic Rossby radius (10–15 km).

The application of spatiotemporal (ST) analytics to integrated data from major sources such as the World Bank, United Nations, and dozens of others holds tremendous potential for shedding new light on the evolution of cultural, health, economic, and geopolitical landscapes on a global level. Realizing this potential first requires an ST data model that addresses challenges in properly merging data from multiple authors, with evolving ontological perspectives, semantical differences, and changing attributes, as well as content that is textual, numeric, categorical, and hierarchical. Equally challenging is the development of analytical and visualization approaches that provide a serious exploration of this integrated data while remaining accessible to practitioners with varied backgrounds. The WSTAMP project at Oak Ridge National Laboratory has yielded two major results in addressing these challenges: 1) development of the WSTAMP database, a significant advance in ST data modeling that integrates 10,000+ attributes covering over 200 nation states spanning over 50 years from over 30 major sources and 2) a novel online ST exploratory and analysis tool providing an array of modern statistical and visualization techniques for analyzing these data temporally, spatially, and spatiotemporally under a standard analytic workflow. We discuss the status of this work and report on major findings.

As global demand for livestock products (such as meat, milk and eggs) is expected to double by 2050, necessary increases to future production must be reconciled with negative environmental impacts that livestock cause. This paper describes the LivestockPlus concept and demonstrates how the sowing of improved forages can lead to the sustainable intensification of mixed crop-forage-livestock-tree systems in the tropics by producing multiple social, economic and environmental benefits. Sustainable intensification not only improves the productivity of tropical forage-based systems but also reduces the ecological footprint of livestock production and generates a diversity of ecosystem services (ES) such as improved soil quality and reduced erosion, sedimentation and greenhouse gas (GHG) emissions. Integrating improved grass and legume forages into mixed production systems (crop-livestock, tree-livestock, crop-tree-livestock) can restore degraded lands and enhance system resilience to drought and waterlogging associated with climate change. When properly managed tropical forages accumulate large amounts of carbon in soil, fix atmospheric nitrogen (legumes), inhibit nitrification in soil and reduce nitrous oxide emissions (grasses), and reduce GHG emissions per unit livestock product. The LivestockPlus concept is defined as the sustainable intensification of forage-based systems, which is based on 3 interrelated intensification processes: genetic intensification - the development and use of superior grass and legume cultivars for increased livestock productivity; ecological intensification - the development and application of improved farm and natural resource management practices; and socio-economic intensification - the improvement of local and national institutions and policies, which enable refinements of technologies and support their enduring use. Increases in livestock productivity will require coordinated efforts to develop supportive government, non-government organization and private sector policies that foster investments and fair market compensation for both the products and ES provided. Effective research-for-development efforts that promote agricultural and environmental benefits of forage-based systems can contribute towards implemention of LivestockPlus across a variety of geographic, political and socio-economic contexts.Keywords: Eco-efficiency, environmental benefits, livestock and environment, mixed farming, pastures, smallholders.DOI: 10.17138/TGFT(3)59-82

The research objectives of the Aquistore CO2 storage project are to design, adapt, and test non-seismic monitoring methods for measurement, and verification of CO2 storage, and to integrate data to determine subsurface fluid distributions, pressure changes and associated surface deformation. Aquistore site is located near Estevan in Southern Saskatchewan on the South flank of the Souris River and west of the Boundary Dam Power Station and the historical part of Estevan coal mine in southeastern Saskatchewan, Canada. Several monitoring techniques were employed in the study area including advanced satellite Differential Interferometric Synthetic Aperture Radar (DInSAR) technique, GPS, tiltmeters and piezometers. The targeted CO2 injection zones are within the Winnipeg and Deadwood formations located at > 3000 m depth. An array of monitoring techniques was employed in the study area including advanced satellite Differential Interferometric Synthetic Aperture Radar (DInSAR) with established corner reflectors, GPS, tiltmeters and piezometers stations. We used airborne LIDAR data for topographic phase estimation, and DInSAR product geocoding. Ground deformation maps have been calculated using Multidimensional Small Baseline Subset (MSBAS) methodology from 134 RADARSAT-2 images, from five different beams, acquired during 20120612–20140706. We computed and interpreted nine time series for selected places. MSBAS results indicate slow ground deformation up to 1 cm/year not related to CO2 injection but caused by various natural and anthropogenic causes.

The Great Artesian Basin mound springs (Australia) are unique wetland ecosystems of great significance. However, these unique ecosystems are endangered by anthropogenic water extraction. Relationships have been established between the vegetated wetland area and the discharge associated with individual springs, providing a potential means of monitoring groundwater flow using measurements of wetland area. Previous studies using this relationship to monitor Great Artesian Basin springs have used aerial photography or high resolution satellite images, giving sporadic temporal information. These "snapshot " studies need to be placed within a longer and more regular context to better assess changes in response to aquifer draw-downs. In this study, the potential of medium resolution MODIS Normalized Difference Vegetation Index data for studying the long-term and high frequency temporal dynamics of wetland vegetation at the Dalhousie Spring Complex of the GAB is tested. Photosynthetic activity within Dalhousie wetlands could be differentiated from surrounding land responses. The study showed good correlation between wetland vegetated area and groundwater flow, but also the important influence of natural species phenologies, rainfall, and human activity on the observed seasonal and inter-annual vegetation dynamic. Declining trends in the extent of wetland areas were observed over the 2000– 2009 period followed by a return of wetland vegetation since 2010. This study underlined the need to continue long-term medium resolution satellite studies of the Great Artesian Basin as these data provide a good understanding of variability within the wetlands, give temporal context for less frequent studies and a strong baseline for assessment of future changes.
