3. In Vitro Propagation of Anthurium andreanum ′Atlanta′ Developed for Pot Culture

In order to establish micropropagation system Anthurium andreanum `Atlanta`, dwarf type, shoots of A. andreanum were cultured on medium supplemented with cytokinin. Callus was formed from the base of shoots. high frequency callus induction was obtained on medium with 10.0mg/L BA or 10.0mg/L TDZ(thidiazuron) at more than 71.8%. The shoots were cultured on media with various combinations and concentrations of TDZ, BA and 2.4-D to enhance callus induction. Callus was induced at more than 72.6% and grew vigorously on media containing 10.0mg/L BA and 0.0∼0.5mg/L 2.4-D, or 1.0mg/L TDZ. Stimulation effects of cytokinin by 2.4-D did not occur in combined treatments of cytokinin and 2.4-D. Callus was cut into sections(710mm), and then cultured on media with BA alone or BA and 2.4-D to regenerate shoots and to stimulate the callus growth. Shoot regeneration and callus growth were effective on media with 10.0mg/L BA alone, or 10.0mg/L BA and 0.1mg/L 2.4-D. In combined treatments of BA and 2.4-D, stmulation effects of cytocinin by 2.4-D also did not occur. Callus growth was decreased, accordiong to increasing the concentration of 2.4-D. In cimbined treatments of TDZ and 2.4-D in shoot regeneration and callus proliferation, stimulated effects of cytokinin by 2.4-D did not occur entirely. Media with 0.5∼1.0mg/L TDZ ingibited the regeneration and rooting of shoots, and callus growth from callus sections. Addition of 2.4-D on medium with TDZ ingibited the regeneration and rooting of shoots, and callus growth. Rooted plantdts were acclimatized in greenhouse. The plantlets were survived more than 98% in soil of vermiculite alone or mixed perlite 1 and vermiculite 1. more...

4. An experimental study of the effect of low-noise wheels in reducing noise and vibration

Wheel/rail interaction is a major source of railway noise. A low-noise wheel structure is developed and its effect on noise reduction is investigated. This low-noise wheel employs a rubber material inserted into the steel rim or mounted on the wheel surface. The low-noise wheel has low stiffness and a high-damping ratio compared to a solid wheel. Measurement shows that it reduces rolling and squealing. It turns out that a subway line with the proposed wheel could reduce its interior noise level by 4–5 dB(A) and vehicle vibration level by 7–8 dB. While the proposed structure seems promising in noise reduction for railway vehicles, the endurance and cost effectiveness of the low-noise wheel are yet to be verified. more...

Lecithinized superoxide dismutase reduces cold ischemia-induced chronic allograft dysfunction.BackgroundChronic renal allograft failure (CAF) is influenced by both allo-dependent and independent factors and is a major cause of graft loss in clinical renal transplantation. We evaluated a novel membrane-bound free radical scavenger, lecithinized superoxide dismutase (lec-SOD), to determine its potential in limiting the harmful effects of ischemia/reperfusion injury on CAF.MethodsFisher rat kidneys were stored for either 1 hour or 18 hours in cold Marshall's preservation solution either with or without lec-SOD and transplanted into Lewis recipients.ResultsWithin 3 days of transplantation, an early inflammatory response involving granulocytes and macrophages was detected in renal allografts exposed to 18 hours cold ischemia that was significantly reduced by preservation with lec-SOD. By 24 weeks post-transplantation, elevated proteinuria and detection of apoptotic cells was observed in kidneys exposed to 18 hours of cold ischemia, that was attenuated by preservation with lec-SOD (P < 0.05). However, up-regulated expression of intracellular adhesion molecule-1 (ICAM-1) and major histocompatibility complex (MHC) Class II together with a T lymphocyte infiltration were observed at 24 weeks that was not prevented by preservation with lec-SOD.ConclusionsThese results demonstrate that ischemia/reperfusion injury, apoptotic cell death and allo-immune responses may be exacerbated by cold ischemia and accelerate the development of CAF. Preservation with lec-SOD may protect against the early damage induced by cold ischemia and reperfusion injury. more...

Background TH-302 is a hypoxia targeted prodrug with a hypoxia-triggered 2-nitroimidazole component designed to release the DNA alkylator, bromo-isophosphoramide mustard (Br-IPM), when reduced in severe hypoxia. A randomized Phase 2B study (NCT01144455) was conducted to assess the benefit of G + T to standard dose G as first-line therapy of PAC. Materials and methods An open-label multi-center study of two dose levels of TH-302 (240 mg/m2 or 340 mg/m2) in combination with G versus G alone (randomized 1:1:1). G (1000 mg/m2) and T were administered IV over 30-60 minutes on Days 1, 8 and 15 of a 28-day cycle. Patients on the G could crossover after progression and be randomized to a G + T arm. The primary efficacy endpoint was a comparison of progression-free survival (PFS) between the combination arms and G alone (80% power to detect 50% improvement in PFS with one-sided alpha of 10%). Summary PFS outcome has previously been reported; more detailed PFS as well as the initial overall survival (OS) data are presented. Results 214 pts were treated; 164 (77%) Stage IV and 50 (23%) Stage IIIB. Median age 65 (range 29-86); 126 M/88 F; 40% ECOG 0/60% ECOG 1. Receiving 6 or more cycles: 32% G; 45% G + T240; 55% G + T340. Median PFS was 3.6 mo in G vs 5.5 mo in G + T240 (p = 0.031) and 6.0 mo in G + T340 (p = 0.008). Poorer prognostic factors (older age, poorer performance status, reduced albumin) were associated with larger treatment effect. Median OS was 7.0 mo in G vs 9.0 in G + T240 and 9.5 mo in G + T340. RECIST best response was 12% in G vs 17% in G + T240 and 27% in G + T340. CA19-9 decreases were significantly greater G + T340. A >50% CA19-9 decrease was 52% with G vs 50% with G + T240 and 70% with G + T340. AEs leading to discontinuation were: 16% G, 15% G + T240 and 11% G + T340. Rash (45% in G + T340) and stomatitis (36% in G + T340) were greater in combination, 4 pts Grade 3 rash. Grd 3/4 thrombocytopenia were 11% G, 39% G + T240 and 59% G + T340 and Grd 3/4 neutropenia were 28% G, 56% G + T240 and 59% G + T340. Conclusions The combination of G plus TH-302 improved the efficacy of G. A TH-302 dose of 340 mg2 was identified for future studies. Skin and mucosal toxicity and myelosuppression were the most common TH-302 related AEs with no increase in treatment discontinuation.
