Abstract Predator–prey interactions are important for regulating populations and structuring communities but are affected by many dynamic, complex factors across large scales, making them difficult to study. Integrated population models (IPMs) offer a potential solution to understanding predator–prey relationships by providing a framework for leveraging many different datasets and testing hypotheses about interactive factors. Here, we evaluate the coyote–deer (Canis latrans–Odocoileus virginianus) predator–prey relationship across the state of North Carolina (NC). Because both species have similar habitat requirements and may respond to human disturbance, we considered net primary productivity (NPP) and urbanization as key mediating factors. We estimated deer survival and fecundity by integrating camera trap, harvest, and biological and hunter observation datasets into a two‐stage, two‐sex Lefkovich population projection matrix. We allowed survival and fecundity to vary as functions of urbanization, NPP, and coyote density, and projected abundance forward to test eight hypothetical scenarios. We estimated initial average deer and coyote densities to be 11.83 (95% CI: 5.64, 20.80) and 0.46 (95% CI: 0.02, 1.45) individuals/km2, respectively. We found a negative, though highly uncertain, relationship between the current levels of coyote density and deer fecundity in most areas that became more negative under hypothetical conditions of lower NPP or higher urbanization, leading to lower projected deer abundances. Though not conclusive, our results indicate the possibility that coyotes could have stronger effects on deer populations in NC if their densities rise, but primarily in less productive and/or more suburban habitats. Our case study provides an example of how IPMs can be used to better understand the complex relationships between predator and prey under changing environmental conditions.

Malaria remains a major public health issue, infecting nearly 220 million people every year. The spread of drug-resistant strains of Plasmodium falciparum around the world threatens the progress made against this disease. Therefore, identifying druggable and essential pathways in P. falciparum parasites remains a major area of research. One poorly understood area of parasite biology is the formation of disulfide bonds, which is an essential requirement for the folding of numerous proteins. Specialized chaperones with thioredoxin (Trx) domains catalyze the redox functions necessary for breaking incorrect and forming correct disulfide bonds in proteins. Defining the substrates of these redox chaperones is difficult and immunoprecipitation based assays cannot distinguish between substrates and interacting partners. Further, the substrate or client interactions with the redox chaperones are usually transient in nature. Activity based crosslinkers that rely on the nucleophilic cysteines on Trx domains and the disulfide bond forming cysteines on clients provide an easily scalable method to trap and identify the substrates of Trx-domain containing chaperones. The cell permeable crosslinker divinyl sulfone (DVSF) is active only in the presence of nucleophilic cysteines in proteins and, therefore, traps Trx domains with their substrates, as they form mixed disulfide bonds during the course of their catalytic activity. It’s so good to be in Cambridge, it feels almost like home. Let me start by stating that in the past year ITMB, a successful map publisher in British Columbia, Canada published more paper map titles than at any time in their history. Similarly, the U.S. Geological Survey (USGS) recently announced that they have ceased producing paper from their aerial photography archive and will only produce digital copies. I believe that both of these facts speak to the future of maps and digital data. It means there will be paper maps well into the future and there will be an increasing array of digital data - some of it reformatted, as in the USGS case, and most of it will be born digital. When asked to speak about GIS and its role in libraries I often find myself in a conundrum - am I here to slay the dragon, or to pet the dragon. The role of technology in libraries is not one that has been embraced by everyone, and often the technology itself seems to have been force-fed upon us. The library profession is not one that has historically been a proponent of change and the very nature of GIS is change. In one sense, we have been given the choice of becoming paper museums or, at the very least, making GIS technology available in our collections. Today, I would like to review the many ways that GIS is, or will, affect our collections. I will divide the presentation into a general overview of GIS in libraries, how it affects our acquisitions or collection development policies, its effect on cataloging, on reference services, staffing, and our web services. Then I will shift the focus a little and discuss the current situation at the Harvard Map Collection, the future role of legacy collections, and a look to the future.

Curative-intent radiotherapy plays an integral role in the treatment of lung cancer and therefore improving its therapeutic index is vital. MR guided radiotherapy (MRgRT) systems are the latest technological advance which may help with achieving this aim. The majority of MRgRT treatments delivered to date have been stereotactic body radiation therapy (SBRT) based and include the treatment of (ultra-) central tumors. However, there is a move to also implement MRgRT as curative-intent treatment for patients with inoperable locally advanced NSCLC. This paper presents the initial clinical experience of using the two commercially available systems to date: the ViewRay MRIdian and Elekta Unity. The challenges and potential solutions associated with MRgRT in lung cancer will also be highlighted.

Background and purposeIn prostate cancer treatment with external beam radiation therapy (EBRT), prostate motion and internal changes in tissue distribution can lead to a decrease in plan quality. In most currently used planning methods, the uncertainties due to prostate motion are compensated by irradiating a larger treatment volume. However, this could cause underdosage of the treatment volume and overdosage of the organs at risk (OARs). To reduce this problem, in this proof of principle study we developed and evaluated a novel adaptive planning method. The strategy proposed corrects the dose delivered by each beam according to the actual position of the target in order to produce a final dose distribution dosimetrically as similar as possible to the prescribed one.Material and methodsOur adaptive planning method was tested on a phantom case and on a clinical case. For the first, a pilot study was performed on an in-silico pelvic phantom. A "library" of intensity modulated RT (IMRT) plans corresponding to possible positions of the prostate during a treatment fraction was generated at planning stage. Then a 3D random walk model was used to simulate possible displacements of the prostate during the treatment fraction. At treatment stage, at the end of each beam, based on the current position of the target, the beam from the library of plans, which could reproduce the best approximation of the prescribed dose distribution, was selected and delivered. In the clinical case, the same approach was used on two prostate cancer patients: for the first a tissue deformation was simulated in-silico and for the second a cone beam CT (CBCT) taken during the treatment was used to simulate an intra-fraction change. Then, dosimetric comparisons with the standard treatment plan and, for the second patient, also with an isocenter shift correction, were performed.ResultsFor the phantom case, the plan generated using the adaptive planning method was able to meet all the dosimetric requirements and to correct for a misdosage of 13% of the dose prescription on the prostate. For the first clinical case, the standard planning method caused underdosage of the seminal vesicles, respectively by 5% and 4% of the prescribed dose, when the position changes for the target were correctly taken into account. The proposed adaptive planning method corrected any possible missed target coverage, reducing at the same time the dose on the OARs. For the second clinical case, both with the standard planning strategy and with the isocenter shift correction target coverage was significantly worsened (in particular uniformity) and some organs exceeded some toxicity objectives. While with our approach, the most uniform coverage for the target was produced and systematically the lowest toxicity values for the organs at risk were achieved.ConclusionsIn our proof of principle study, the adaptive planning method performed better than the standard planning and the isocenter shift methods for prostate EBRT. It improved the coverage of the treatment volumes and lowered the dose to the OARs. This planning method is particularly promising for hypofractionated IMRT treatments in which a higher precision and control on dose deposition are needed. Further studies will be performed to test more extensively the proposed adaptive planning method and to evaluate it at a full clinical level.
