Cationic liposome - CpG DNA complexes (lipoplexes) are known as stimulators of innate immunity via Toll-like receptor 9 (TLR9)-triggered activation of the nuclear factor kappa B (NF-κB) pathway. More recent reports suggest that DNA lipoplexes also engage DNA sensors in the cytosol leading to the stimulation of the interferon response factor (IRF) pathway. In this study a range of lipoplexes were formulated by using an invariable helper lipid, three different cationic lipids (DOTAP, DOTMA and DDA) and three different CpG-containing plasmids of different sizes. These lipoplexes exhibited similar hydrodynamic diameters, zeta-potentials and plasmid loading rates, despite the different lipid blends and CpG-containing plasmids. Binding and uptake of liposomal lipids by J774.A1 macrophages and JAWSII dendritic cells increased significantly (up to 4-fold) upon lipoplex formation. Cellular plasmid DNA uptake via lipoplexes compared to naked DNA was increased up to 18-fold. Analysis of signal transduction pathway activation in J774-DUAL™ reporter cells by liposomes or naked CpG plasmid DNA compared to their derived lipoplexes showed only minor activation of the NF-κB pathway, while the IRF pathway displayed massive activation factors of up to 46-fold. DOTAP- and DOTMA lipoplexes also led to massive interferon-alpha and -beta secretion of J774A.1 macrophages and JAWSII dendritic cells, which is a hallmark of IRF pathway activation. Cellular distribution studies on DOTAP lipoplexes suggest delivery of plasmid DNA via vesicular compartments into the cytosol. The cationic dimethyldioctadecylammonium/trehalose 6,6,9-dibehenate (DDA/TDB) liposome is as a strong adjuvant system for vaccines, with remarkable immunostimulatory activity. The mucosal administration of vaccines is a potential strategy for inducing earlier and stronger mucosal immune responses to infectious diseases. In this study, we assessed whether the intranasal administration of cationic DDA/TDB liposomes combined with influenza antigen A (H3N2) can be used as a highly efficacious vaccine to induce mucosal and systemic antibody responses. Confocal laser scanning microscopy and a flow-cytometric analysis showed that the uptake of the cationic DDA/TDB liposome carrier was significantly higher than that of neutral 1,2-distearoyl-sn-glycero-3-phosphocholine/cholesterol (DSPC/Chol) or cationic 1,2-dioleoyl-3-trimethylammonium-propane/3β-(N-[N',N'-dimethylaminoethane]-carbamoyl (DOTAP/DC-Chol) liposomes. Our results indicate that the cationic DDA/TDB liposome is more effective in facilitating its uptake by dendritic cells (DCs) in vitro than the DSPC/Chol or DOTAP/DC-Chol liposome. DCs treated with DDA/TDB liposomes strongly expressed CD80, CD86, and MHC II molecules, whereas those treated with DSPC/Chol or DOTAP/DC-Chol liposomes did not. C57BL/6 mice intranasally immunized with H3N2-encapsulating cationic DDA/TDB liposomes had significantly higher H3N2-specific s-IgA levels in their nasal wash fluid than those treated with other formulations. The DDA/TDB liposomes also simultaneously enhanced the serum IgG IgG2a, IgG1, and IgG2b antibody responses. In summary, DDA/TDB liposomes effectively facilitated their uptake by DCs and DCs maturation in vitro, and induced significantly higher mucosal IgA, systemic IgG, IgG1, and IgG2b antibody titres than other formulations after their intranasal administration in vivo. These results indicate that DDA/TDB liposomes are a promising antigen delivery carrier for clinical antiviral applications.

Loss-of-function of barley mildew locus o ( Mlo ) confers durable broad-spectrum penetration resistance to the barley powdery mildew pathogen, Blumeria graminis f. sp. hordei ( Bgh ). Given the importance of mlo mutants in agriculture, surprisingly few molecular components have been identified to be required for this type of resistance in barley. With the aim to identify novel cellular factors contributing to mlo -based resistance, we devised a pharmacological inhibitor screen. Of the 41 rationally chosen compounds tested, five caused a partial suppression of mlo resistance in barley, indicated by increased levels of Bgh host cell entry. These chemicals comprise brefeldin A (BFA), 2',3'-dideoxyadenosine (DDA), 2-deoxy-d-glucose, spermidine, and 1-aminobenzotriazole. Further inhibitor analysis corroborated a key role for both anterograde and retrograde endomembrane trafficking in mlo resistance. In addition, all four ribonucleosides, some ribonucleoside derivatives, two of the five nucleobases (guanine and uracil), some guanine derivatives as well as various polyamines partially suppress mlo resistance in barley via yet unknown mechanisms. Most of the chemicals identified to be effective in partially relieving mlo resistance in barley also to some extent compromised powdery mildew resistance in an Arabidopsis mlo2 mlo6 double mutant. The Palos Verdes Shelf (PVS) and the continental slope off the Palos Verdes Peninsula are highly contaminated by degradation products of the pesticide DDT (1-chloro-4-[2,2,2-trichloro-1-(4-chlorophenyl)ethyl]benzene). Sediment samples from two box cores were analyzed to obtain further information about the fate of DDT and its degradation products within the environment. After solvent extraction, an alkaline hydrolysis procedure was applied. A comprehensive screening for 26 DDT compounds revealed that DDT and its degradates contaminate not only the extractable fraction but also the fraction released by alkaline hydrolysis. A comparison of the quantitative distribution of DDT degradation products in the extractable fraction and released by alkaline hydrolysis showed a distinct difference. DDE (1-chloro-4-[2,2-dichloro-1-(4-chlorophenyl)ethenyl]benzene), DDD (1-chloro-4-[2,2-dichloro-1-(4-chlorophenyl)ethyl]benzene), DDMS (1-chloro-4-[2-chloro-1-(4-chlorophenyl)ethyl]benzene), and DDMU (1-chloro-4-[2-chloro-1-(4-chlorophenyl)ethenyl]benzene) were predominant in the sediment extracts but minor components of the hydrolyzable fraction. The most abundant compounds released by the alkaline hydrolysis were DBP (bis(4-chlorophenyl)methanone), DDNU (1-chloro-4-[1-(4-chlorophenyl)ethenyl]benzene), DDM (1-chloro-4-[(4-chlorophenyl)methyl]benzene) and the water-soluble DDA (2,2-bis(4-chlorophenyl)acetic acid). The release of DDA may point to the presence of an important degradation pathway in marine environments. Concentration levels of DDT-related compounds showed corresponding vertical profiles in both fractions, but were significantly lower in the fraction released by alkaline hydrolysis. In contrast to fluvial sediments contaminated by DDT and its degradates the alkaline hydrolysis products represented a minor portion of the total sedimentary burden in the analyzed marine sediments. These findings show the necessity of a comprehensive screening for all DDT isomers and breakdown products in the extractable and non-extractable fraction to assess the total contamination abundance and potential environmental risks., (Copyright © 2017 Elsevier Ltd. Abiotic and biotic processes can be used to remediate DDX (DDT, DDD, DDE, and DDNS) contaminated soils; these processes can be fostered using specific carbon-amendments to stimulate particular soil indigenous microbial communities to improve rates or extent of degradation. In this study, toluene and glycerol were evaluated as cosubstrates under aerobic and anoxic conditions to determine the degradation efficiencies of DDX and to elucidate possible degradation mechanisms. Slurry microcosms experiments were performed during 60 days using pretreated soil with zero-valent iron (ZVI). Toluene addition enhanced the percentage of degradation of DDX. DDNS was the main compound degraded (around 86%) under aerobic conditions, suggesting cometabolic degradation of DDX by toluene-degrading soil bacteria. Glycerol addition under anoxic conditions favored the abiotic degradation of DDX mediated by sulfate-reducing bacteria activity, where DDT was the main compound degraded (around 90%). The 16S rDNA metagenomic analyses revealed Rhodococcus ruber and Desulfosporosinus auripigmenti as the predominant bacterial species after 40 days of treatment with toluene and glycerol additions, respectively. The temporal-spatial distributions of DDT-related contaminants (DDXs), including DDT (dichlorodiphenyltrichloroethane), DDE (dichlorodiphenyldichloroethylene) and DDD (dichlorodiphenyldichloroethane), in the sediments of Lake Chaohu and their influencing factors were studied. p,p-DDE and p,p-DDD were found to be the two dominant components of DDXs in both surface and core sediments. The parent DDT compounds were still detectable in sediment cores after the late 1930s. Historical usage of technical DDT was identified as the primary source of DDXs in sediments, as indicated by DDT/(DDD + DDE) ratios of less than one. The residual levels of DDXs were higher in the surface and core sediments in the western lake area than in other lake areas, which might be due to the combined inflow effects of municipal sewage, industrial wastewater and agricultural runoff. The DDX residues in the sediment cores reached peak values in the late 1970s or early 1980s. There were significant positive relationships between DDX residues in sediment cores with annual DDT production and with fine particulate sizes (<4.5 μm). The relationship between the DDXs and TOC in sediment was complex, as indicated by the significant differences among the surface and core sediments. The algae-derived organic matter significantly influenced the amount of residue, composition and distribution of DDXs in the sediments. The DDD/DDE ratios responded well to the anaerobic conditions in the sediments that were caused by algal blooms after the late 1970s in the western lake area. This suggests that the algae-derived organic matter was an important factor and served as a biomarker of eutrophication and also affected the DDX residues and lifecycle in the lake ecosystem., (Copyright © 2016 Elsevier Ltd. The fate of hydrophobic organic compounds in aquatic environment are largely determined by their exchange at sediment-water interface, which is highly dynamic and subject to rapidly evolving environmental conditions. In turn, environmental conditions may be governed by both physicochemical parameters and anthropogenic events. To examine the importance of various impact factors, passive sampling devices were deployed at the seafloor of Hailing Bay, an urbanized estuarine bay in Guangdong Province of South China to measure the sediment-water diffusion fluxes of several metabolites of dichlorodiphenyltrichloroethane (DDT), p,p'-DDE, p,p'-DDD and o,p'-DDD. The physicochemical properties of water (temperature, pH, salinity and dissolved oxygen) and surface sediment (sediment organic matter, physical composition, pH, water content, colony forming unit and catalase activity) were also measured. The results showed that the diffusion fluxes of o,p'-DDD, p,p'-DDD and p,p'-DDE at sites A1 and A2 near a fishing boat maintenance facility ranged from 0.42 to 4.73 ng m -2 d -1 (from sediment to overlying water), whereas those at offshore sites varied between -0.03 and -3.02 ng m -2 d -1 (from overlying water to sediment), implicating A1 and A2 as the sources of the target compounds. The distribution patterns of the diffusion fluxes of the target compounds were different from those of water and sediment parameters (water temperature, salinity, sediment texture, pH, colony forming unit and catalase activity) at six sampling sites. This finding suggested that none of these parameters were critical in dictating the sediment-water diffusion fluxes. Besides, decreases in the contents of kerogen and black carbon by 6.7% and 11% would enhance the diffusion fluxes of the target compounds by 11-14% and 12-23%, respectively, at site A1, indicating that kerogen and black carbon were the key factors in mediating the sediment-water diffusion fluxes of DDT-related compounds in field environments., (Copyright © 2016 Elsevier Ltd. The incidence of autoimmune Type 1 diabetes (T1D) has been steadily rising in developed countries. Although the exact cause of T1D remains elusive, it is known that both genetics and environmental factors play a role in its immunopathogenesis. Whereas a positive association between p,p'-DDE, a DDT metabolite, and Type 2 diabetes (T2D) has been well established, its role in T1D development in an experimental animal model has never been elucidated. This study seeks to investigate the effects of DDE exposure on the development of T1D in a NOD mouse model. As T1D is a T-cell-mediated disease, the underlying mechanism of DDE action on T-cells was studied in vitro and, in the context of acute and chronic DDE exposure, in vivo by investigating lymphocytes' viability, proliferation, their subsets and cytokine profiles. Chronic high-dose DDE treatment, initiated in pre-diabetic 8-week-old NOD females administered twice weekly intraperitoneally with 50 mg/kg DDE, significantly increased diabetes incidence and augmented disease severity in treated animals. Whereas T-cell proliferation and cell viability in the spleens of treated mice were not affected, diabetogenic action of chronic DDE exposure was associated with a decrease in regulatory T-cells and a suppression of secretion of protective cytokines, such as IL-4 and IL-10. Interestingly, an acute high-dose in vivo treatment of 8-week-old NOD males with 100 mg DDE/kg, administered intraperitoneally every other day over a period of 10 days, increased T-cell proliferation and potentiated pro-inflammatory and TH1-type cytokine secretion, without affecting the splenocytes viability and the T-cell sub-populations. These results confirm that high-dose DDE treatments affect the immune system, in particularly T-cell function. In conclusion, this study shows for the first time that high-dose chronic DDE exposure exhibits a diabetogenic potential, with an underlying immunomodulatory mechanism of action, in the development of T1D in an experimental mouse NOD model.

Human exposure to persistent organic pollutants (POPs) is a certainty, even to long banned pesticides like o,p'-dichlorodiphenyltrichloroethane (o,p'-DDT), and its metabolites p,p'-dichlorodiphenyldichloroethylene (p,p'-DDE), and p,p'-dichlorodiphenyldichloroethane (p,p'-DDD). POPs are known to be particularly toxic and have been associated with endocrine-disrupting effects in several mammals, including humans even at very low doses. As environmental estrogens, they could play a critical role in carcinogenesis, such as in breast cancer. With the purpose of evaluating their effect on breast cancer biology, o,p'-DDT, p,p'-DDE, and p,p'-DDD (50-1000 nM) were tested on two human breast adenocarcinoma cell lines: MCF-7 expressing estrogen receptor (ER) α and MDA-MB-231 negative for ERα, regarding cell proliferation and viability in addition to their invasive potential. Cell proliferation and viability were not equally affected by these compounds. In MCF-7 cells, the compounds were able to decrease cell proliferation and viability. On the other hand, no evident response was observed in treated MDA-MB-231 cells. Concerning the invasive potential, the less invasive cell line, MCF-7, had its invasion potential significantly induced, while the more invasive cell line MDA-MB-231, had its invasion potential dramatically reduced in the presence of the tested compounds. Altogether, the results showed that these compounds were able to modulate several cancer-related processes, namely in breast cancer cell lines, and underline the relevance of POP exposure to the risk of cancer development and progression, unraveling distinct pathways of action of these compounds on tumor cell biology., (© 2013 Wiley Periodicals, Inc.)

The purpose of this study was to develop a rapid and cost efficient hair extraction method, using the headspace solid phase microextraction (HSSPME) technique for the simultaneous determination and biomonitoring of 1,1,1-trichloro-2,2-bis(4-chlorophenyl)ethane) (DDT) and its isomers/metabolites and polychlorinated biphenyls (PCBs) in hair samples. A total of 72 head hair samples were collected from children living in urban and rural regions of the island of Crete. Two hundred milligrams of hair were digested under alkaline conditions and thermostated for 30 min at 90°C while a 65 µm PDMS/DVB fibre was exposed into the headspace of the vial. Analytical parameters of the method (time of incubation, agitation speed, recovery, precision, accuracy, carry over, matrix effect, linearity, and selectivity) were examined. Recoveries of the DDTs in the spiked hair samples were calculated from 42.3% for opDDD to 87.1% for opDDE, while recoveries for PCB congeners were from 52.6% for PCB138 to 96.6 % for PCB28. The method was applied for the analysis of authentic hair samples. Significant differences (p=0.001) of the burden to total DDTs (sumDDTs) as well as of the frequencies of detection of positive samples (p=0.020) were observed between the examined regions. Moreover, significant differences in the detected concentrations of PCB congeners were observed for PCB52 (p<0.001) and PCB28 (p=0.017) as well for their prevalence between urban and rural regions. Application of HSSPME for the biomonitoring of DDTs and PCBs biomarkers in hair was tested and successfully applied to the analysis of spiked and authentic hair samples. Recent understanding of the pathogenesis of tuberculosis allows the possible application of immunotherapy for the treatment of tuberculosis. Therapies that would upregulate the host antimycobacterial immune response and/or attenuate T-cell suppressive and macrophage-deactivating cytokines may prove to be useful in the treatment of tuberculosis. ESAT6, 6-kDa early secreted antigenic target, is a potent protective antigen and is considered as major target for long-lived memory cells. In the present study the immunotherapeutic potential of ESAT-6 has been evaluated in mouse model of experimental tuberculosis. In the present study the ESAT-6 protein was cloned in Escherichia coli using pET23a(+) plasmid and purified by Ni(2+)-NTA chromatography. Further, the immunotherapeutic potential of the recombinant ESAT-6 (in terms of CFU enumeration in the target organs and histopathological analysis of lungs) was evaluated against experimental tuberculosis. The recombinant ESAT-6 with C-terminal histidine-tag and free N-terminus mimics the natural form of ESAT-6 has been successfully cloned and purified. The recombinant ESAT-6 protein adjuvanted with dimethyl dioctadecylammonium bromide (DDA) moderately reduced the bacterial load in the target organs of infected mice. Further, the formulation (ESAT-6-DDA) was able to act synergistically when given in combination with antituberculosis drugs. DDT has enjoyed the reputation of a successful pesticide in disease control programme and agricultural practices along with the serious opposition and ban later on due to its biomagnification and toxic action against non-target organisms. The present work was carried out to develop QSAR models for analysing DDT analogues for their pesticidal activity and in silico validation of these models. A 2D-QSAR model was generated using stepwise with multiple regression, and the model with a value of r(2) = 0.7324; q(2) = 0.6215; pred r(2) = 0.7038, containing five descriptors, was selected for further study. The 3D QSAR with CoMFA analysis showed that steric contribution of 21% and electrostatic contribution of about 79% were required for larvicidal activity of DDT analogues. A set of 3430 molecules was generated using the basic DDT skeleton as template, and these were evaluated for their mosquito larvicidal activity using the generated QSAR models and DDT as standard. Eleven molecules were selected for in silico validation of these models. For this, a docking study of the selected molecules against the homology-modelled voltage-gated sodium channel of Anopheles funestus was conducted. The study showed that the activities of these analogues as predicted by 2D-QSAR, 3D-QSAR with CoMFA and dock score were observed to be well correlated.

The presence of endocrine disruptors in source water is of great concern because of their suspected adverse effects on humans, even when present at very low levels. As the main source of potable water supply, rivers in Malaysia are highly susceptible to contamination by various endocrine disruptors originating from anthropogenic activities. In this study, the contamination levels of 1,1,1-trichloro-2,2-bis (4-chlorophenyl) ethane (DDT) and its metabolites and di-(2-ethylhexyl) phthalate (DEHP) in rivers of Selangor were examined using gas chromatography-mass spectrometry. Samples were collected from sites representing source water for 18 drinking water treatment plants in Selangor between July 2008 and July 2009. DDT and its metabolites were detected in only 14% of the 192 samples analysed at levels ranging from 0.6 to 14.6 ng/L. Meanwhile DEHP was detected in 96.8% of the samples at levels ranging from below quantitation level (18 ng/L) to 970 ng/L. The detected levels of DDTs and DEHP were lower than the WHO and Malaysian Guidelines for Drinking Water Quality. Data obtained from this study should also serve as a reference point for future surveillance on these endocrine disruptors.

Organochlorine pesticides (OCPs) are persistent endocrine disruptors. OCPs cross the placenta; this prenatal exposure has been associated with adverse pregnancy outcomes. We investigated associations between prenatal exposure to OCPs and gestational age and birth weight in 600 infants born between 1960 and 1963. The primary OCP was 1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane (p,p'-DDT), its primary metabolite, 1,1'-dichloro-2,2'-bis(p-chlorophenyl)ethylene (p,p'-DDE) and the contaminant, 1,1,1-trichloro-2-(p-chlorophenyl)-2-(o-chlorophenyl)-ethane (o,p'-DDT). Regression analysis indicated that for each natural log unit increase in p,p'-DDT, birth weight increased by 274 g (95% CI: 122, 425) when controlling for p,p'-DDE and o,p'-DDT. At a given level of p,p'-DDT exposure, o,p'-DDT and p,p'-DDE were associated with decreased birth weight. p,p'-DDE was negatively associated with length of gestation, controlling for p,p'-DDT and o,p'-DDT. These findings suggest opposing associations between exposure to p,p'-DDT and p,p'-DDE and birth weight. The purpose of this study was to investigate the effect of 1,1,1-trichloro-2,2-bis-(chlorophenyl)ethane (DDT), 1,1-bis-(chlorophenyl)-2,2-dichloroethene (DDE), and 1,1-dichloro-2,2-bis(chlorophenyl)ethane (DDD) isomers on COX-2 expression in a human trophoblast-derived cell line. Cultured HTR-8/SVneo trophoblast cells were exposed to DDT isomers and its metabolites for 24 h, and COX-2 mRNA and protein expression were assessed by RT-PCR, Western blotting, and ELISA. Prostaglandin E₂ production was also measured by ELISA. Both COX-2 mRNA and protein were detected under control (unexposed) conditions in the HTR-8/SVneo cell line. COX-2 protein expression and prostaglandin E₂ production but not COX-2 mRNA levels increased only after DDE and DDD isomers exposure. It is concluded that DDE and DDD exposure induce the expression of COX-2 protein, leading to increased prostaglandin E2 production. Dichlorodiphenyltrichloroethane (DDT) is ubiquitous in the environment, and the exposure to DDT and its related pesticides has long been linked to endocrine disruption. The mechanism of endocrine disruption toward targeted receptors, however, remains unclear. Probing the molecular recognition of DDT analogs by targeted receptors at the atomic level is critical for deciphering this mechanism. Molecular dynamics (MD) simulations were applied to probe the molecular recognition process of DDT and its five analogs, including dichlordiphenyldichloroethylene (DDE), dichlorodiphenyldichloroethane (DDD), methoxychlor (MXC), p,p'-hydroxy-DDT (HPTE), and dicofol by human estrogen receptor (ER) α and human ER-related receptor (ERR) γ. Van der Waals interactions mainly drive the interactions of DDT analogs with ERα ligand-binding domain (LBD) and ERRγ LBD. Minor structural changes of DDT analogs in the number and position of chlorine and phenolic hydroxyl moiety cause differences in binding modes through aromatic stacking and hydrogen bonding and thus affect differently conformational changes of ERα LBD and ERRγ LBD. The binding of DDT analogs affects the helix 12 orientation of ERα LBD but causes no rearrangement of helix 12 of ERRγ LBD. DDT, DDE and some additional lipophilic derivatives are recognized contaminants in sediments of Southern California Bight (SCB). Only about 10% of total DDTs discharged into the SCB are accounted for using available monitoring data (sediment, water, and biota). DDA represented up to 0.03% DDTs (DDT/DDE/DDD) in SCB surface sediments (top 2 cm) in amounts up to 76 μg DDA/kg dry weight. Highest DDA levels were found where DDT and DDD levels were maximal at the primary wastewater outfall indicating a natural precursor-product relationship for DDT and DDA. Still culture of SCB sediment revealed limited DDA formation following DDT fortification. DDA residues have also been found in contaminated Long Island, NY sediments provided by USGS. The organochlorine insecticide DDT (1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane) is still used for malaria vector control in certain areas of South Africa. The strict Stockholm Convention on Persistent Organic Pollutants (POPs) allows spraying on the inside of traditional dwellings with DDT. In rural villages contaminated dust presents an additional pathway for exposure to DDT. We present a new method for the determination of DDT in indoor air where separate vapour and particulate samples are collected in a single step with a denuder configuration of a multi-channel open tubular silicone rubber (polydimethylsiloxane (PDMS)) trap combined with a micro quartz fibre filter. The multi-channel PDMS trap section of the denuder concentrates vapour phase insecticide whereas particle associated insecticide is transferred downstream where it is collected on a micro-fibre filter followed by a second multi-channel PDMS trap to capture the blow-off from the filter. The multi-channel PDMS trap and filter combination are designed to fit a commercial thermal desorber for direct introduction of samples into a GC-MS. The technique is solvent-free. Analyte extraction and sample clean-up is not required. Two fractions, vapour phase and particulate phase p,p'-DDT, o,p'-DDT; p,p'-DDD, o,p'-DDD; p,p'-DDE and o,p'-DDE in 4 L contaminated indoor air, were each quantitatively analysed by GC-MS using isotopically labelled ring substituted (13)C(12) -p,p'-DDT as an internal standard. Limits of detection were 0.07-0.35 ng m(-3) for p,p'-DDT, o,p'-DDT, p,p'-DDD, o,p'-DDD, p,p'-DDE and o,p'-DDE. Ratios of airborne p,p'-DDD/p,p'-DDT and of o,p'-DDT/p,p'-DDT are unusual and do not match the ideal certified ingredient composition required of commercial DDT. Results suggest that the DDT products used for indoor residual spraying (IRS) prior to, and during 2007, may have been compromised with regards to insecticidal efficacy, demonstrating the power of this new environmental forensics tool., (Copyright © 2012 Elsevier B.V. Introduction: In Egypt, the picture of threats to humans and the environment from the exposure to organic pollutants is still incomplete. Thus the objectives of this study were to assess the occurrence and distribution of polychlorinated biphenyls (PCBs), organochlorine pesticides, and chlorpyrifos in sediments and mussels of Abu Qir Bay and their risks for environment and human health., Materials and Methods: Twenty-three different compounds organochlorines were determined in 20 surfacial sediment and 10 mussel samples by gas chromatography-electron capture detector. A Screening Level Ecological Risk Assessment (SLERA) and a Human Health Risk Assessment (HHRA) were performed with the data., Results and Discussion: ΣDDT (DDT, DDE, DDD) (average concentration 27 µg/kg dw) dominated the detected organic pollutants in the sediments, followed by CHLs (chlordane, heptachlor, heptachloro epoxide), hexachlorocyclohexane, chlorpyrifos, endosulfane, dieldrine, Σ6 PCBs, aldrine, hexachlorobenzene, pentachlorobenzene, methoxychlor, and mirex. In general, concentrations of Σ6 PCBs in mussels were higher than their corresponding sediment concentrations reflecting their relatively high bioavailability and bioaccumulative potential. However, concentrations of the organochlorine pesticides in mussels were lower than their corresponding sediment samples. Nevertheless, the SLERA on the bay sediments revealed that adverse ecological effects to benthic species are expected to occur whereas the HHRA showed that adverse health effects are not expected to occur from the consumption of the mussels., Conclusions: With the help of a SLERA, it was possible to indicate which class of chlorinated organic compounds is of highest concern to assess and to improve the environmental quality of the bay. Monitoring of organochlorines and chlorpyrifos would be needed to control the future trend of pollution.

Background, Aim, and Scope: 2,2-bis(chlorophenyl)-1,1,1-trichloroethane (DDT) metabolites, other than those routinely measured [i.e., 2,2-bis(chlorophenyl)-1,1-dichloroethylene (DDE) and 2,2-bis(chlorophenyl)-1,1-dichloroethane (DDD)], have recently been detected in elevated concentrations not only in the surface water of Teltow Canal, Berlin, but also in sediment samples from Elbe tributaries (e.g., Mulde and Havel/Spree). This was paralleled by recent reports that multiple other metabolites could emerge from the degradation of parent DDT by naturally occurring organisms or by interaction with some heavy metals. Nevertheless, only very few data on the biological activities of these metabolites are available to date. The objective of this communication is to evaluate, for the first time, the cytotoxicity, dioxin-like activity, and estrogenicity of the least-studied DDT metabolites., Methods: Four DDT metabolites, p,p'-2,2-bis(chlorophenyl)-1-chloroethylene (DDMU), p,p'-2,2-bis(chlorophenyl)-1-chloroethane (DDMS), p,p'-2,2-bis(4-ch1oropheny1)acetonitrile (DDCN), and p,p'-2,2-bis(chlorophenyl)acetic acid (DDA), were selected based on their presence in environmental samples in Germany such as in sediments from the Mulde River and Teltow Canal. O,p'-DDT was used as reference in all assays. Cytotoxicity was measured by neutral red retention with the permanent cell line RTG-2 of rainbow trout (Oncorhynchus mykiss). Dioxin-like activity was determined using the 7-ethoxyresorufin-O-deetylase assay. The estrogenic potential was tested in a dot blot/RNAse protection-assay with primary hepatocytes from male rainbow trout (O. mykiss) and in a yeast estrogen screen (YES) assay., Results: All DDT metabolites tested revealed a clear dose-response relationship for cytotoxicity in RTG-2 cells, but no dioxin-like activities with RTL-W1 cells. The dot blot/RNAse protection-assay demonstrated that the highest non-toxic concentrations of these DDT metabolites (50 μM) had vitellogenin-induction potentials comparable to the positive control (1 nM 17β-estradiol). The estrogenic activities could be ranked as o,p'-DDT > p,p'-DDMS > p,p'-DDMU > p,p'-DDCN. In contrast, p,p'-DDA showed a moderate anti-estrogenic effect. In the YES assay, besides the reference o,p'-DDT, p,p'-DDMS and p,p'-DDMU displayed dose-dependent estrogenic potentials, whereas p,p'-DDCN and p,p'-DDA did not show any estrogenic potential., Discussion: The reference toxicant o,p'-DDT displayed a similar spectrum of estrogenic activities similar to 17β-estradiol, however, with a lower potency. Both p,p'-DDMS and p,p'-DDMU were also shown to have dose-dependent estrogenic potentials, which were much lower than the reference o,p'-DDT, in both the vitellogenin and YES bioassays. Interestingly, p,p'-DDA did not show estrogenic activity but rather displayed a tendency towards anti-estrogenic activity by inhibiting the estrogenic effect of 17β-estradiol. The results also showed that the p,p'-metabolites DDMU, DDMS, DDCN, and DDA do not show any dioxin-like activities in RTL-W1 cells, thus resembling the major DDT metabolites DDD and DDE., Conclusions: All the DDT metabolites tested did not exhibit dioxin-like activities in RTL-W1 cells, but show cytotoxic and estrogenic activities. Based on the results of the in vitro assays used in our study and on the reported concentrations of DDT metabolites in contaminated sediments, such substances could, in the future, pose interference with the normal reproductive and endocrine functions in various organisms exposed to these chemicals. Consequently, there is an urgent need to examine more comprehensively the risk of environmental concentrations of the investigated DDT metabolites using in vivo studies. However, this should be paralleled also by periodic evaluation and monitoring of the current levels of the DDT metabolites in environmental matrices., Recommendations and Perspectives: Our results clearly point out the need to integrate the potential ecotoxicological risks associated with the "neglected" p,p'-DDT metabolites. For instance, these DDT metabolites should be integrated into sediment risk assessment initiatives in contaminated areas. One major challenge would be the identification of baseline data for such risk assessment. Further studies are also warranted to determine possible additive, synergistic, or antagonistic effects that may interfere with the fundamental cytotoxicity and endocrine activities of these metabolites. For a more conclusive assessment of the spectrum of DDT metabolites, additional bioassays are needed to identify potential anti-estrogenic, androgenic, and/or anti-androgenic effects.

Previous studies of in utero exposure to dichlorodiphenyltrichloroethane (DDT) have shown mixed results for the harmful effects on fetal growth and development. This cross-sectional study was designed to: (1) examine the extent of DDT exposure in 1578 women, aged 28.5±6.0 who delivered between June 2005 and 2006 in the area of Al-Kharj, Saudi Arabia; and (2) assess its influence on neonatal anthropometric measurement of newly born babies. DDT and its metabolites, namely 1,1-dichloro-2,2-bis (p-chlorophenyl) ethylene (p,p'-DDE), 1,1-dichloro-2,2-bis (p-chlorophenyl) ethane (p,p'-DDD) and 1,1,1-trichloro-2,2' bis (p-chlorophenyl) ethane (p,p'-DDT) were measured in cord and maternal serum as well as placenta by Gas Chromatography coupled with an Electron Capture Detector (GC/ECD). p,p'-DDE was detected in 28.3% of cord and 54.4% of maternal serum, reflecting past exposure, whereas the p,p'-DDT was only found in 0.46% cord and 1.2% maternal samples. As expected the p,p'-DDE cord levels (0.197±0.961μg/L) were 2.8 times lower than the maternal levels (0.551±1.778μg/L), and both were significantly correlated (r=0.517) suggesting its transplacental transfer. The picture was different in placental tissues. p,p'-DDE and p,p'-DDT were detected in 84% and 99% of placental tissues, with the highest p,p'-DDT in placental tissues (29.62±158.282µg/kg dry wt.) compare to p,p'-DDE (10.167±18.851μg/kg dry wt.). In general, the presence of DDT metabolites in our participants indicates that these chemicals are still relevant despite the fact that they have been banned or restricted in the study area for many years. Our results support the view for an association between low in utero exposure to DDT and the anthropometric development of the fetus leading to a reduction in its head circumference, crown-heel length, birth weight and birth height. Since the reduction in these measures was independent of gestational age and/or preterm births, our findings suggest a detrimental effect of the DDT exposure on fetal growth. Neonatal anthropometric measures are considered as an important tool to detect newborns at higher risk of morbidity and impairment of growth. This study evaluated the interactions of flight, fasting, and 1,1,1-trichloro-bis(4-chlorophenyl)ethane (p,p'-DDT) loading on residue metabolism and distribution in recently exposed white-crowned sparrows (Zonotrichia leucophrys). Female sparrows were dosed with 5 mg p,p'-DDT per kg body weight over 3 d. Following 1 d of recovery, sparrows were flown in a wind tunnel for up to 140 min, in 15-min blocks. Food was withheld from the start of the flight period until birds were euthanized. DDT, 1,1-dichloro-2,2-bis(4 chlorophenyl)ethane (DDD), and 1,1-dichloro-2,2-bis(4-chlorophenyl)ethylene (DDE) were present in all tissues examined. 1-Chloro-2,2-bis(4-chlorophenyl)ethene (DDµ), 1,1-bis(4-chlorophenyl)ethane (p,p'-DDη), and 2,2-bis(4-chlorophenyl)ethanol (p,p'-DDOH) were not found. Fasting did not significantly affect the rate of residue increase over time in any of the tissues examined. When sparrows flew and fasted simultaneously, fasting seldom contributed to an increase in tissue residues. However, the length of time flown was significantly correlated with increasing toxicant concentrations in the brain, kidney, and liver, effectively demonstrating the potential for brief flights to enhance mobilization of DDT and its metabolites. Dose, flight, and fasting also increased residues in brain tissue. The novel molecularly imprinted microsphere (MIM) that could be applied as special sorbent was synthesized by aqueous suspension polymerization using 1,1-bis(4-chlorophenyl)-1,2,2,2-tetrachloroethane (α-chloro-DDT) as the dummy template. The obtained MIM exhibited good recognition and selectivity to dicofol and it was successfully applied as selective sorbent of solid-phase extraction for the determination of dicofol from aquatic products. At the optimum conditions of the molecularly imprinted solid-phase extraction (MISPE) coupled with GC-ECD, good linearity for dicofol was achieved in a range of 0.4-100 ng g(-1) (r(2)=0.9995) and the recoveries at three spiked levels were ranged from 85.8% to 101.2% for aquatic products with the relative standard deviation (RSD) less than 5.6%. The presented MISPE-GC-ECD method exhibited the advantages of simplicity, selectivity and sensitivity, and could be potentially applied to the determination of dicofol in complicated aquatic products., (Copyright © 2011 Elsevier B.V. Although the production and use of dichlorodiphenyltrichloroethane (DDT), a legacy component of persistent organic pollutants, have been highly restricted worldwide, the environmental fate of DDT has remained a great concern as it is not only ubiquitous and bioaccumulative but can also be degraded to a series of metabolites that may be more hazardous ecologically. The present study, taking advantage of the abundant levels of DDT and its metabolites in a subtropical coastal region of China, investigated into the degradation pathways of DDT in natural coastal sediment. Sediment profiles indicated that degradation of 1,1,1-trichloro-2,2-bis-(p-chlorophenyl)ethane (p,p'-DDT) to 1,1-dichloro-2,2-bis-(p-chlorophenyl)ethane (p,p'-DDD) mainly occurred in sediment of the top 20 cm layer. 1,1-dichloro-2,2-bis-(p-chlorophenyl)ethylene (p,p'-DDE), aerobically transformed from p,p'-DDT prior to sedimentation, was likely to degrade to 1-chloro-2,2-bis-(p-chlorophenyl)ethylene (p,p'-DDMU) which was further converted to 2,2-bis(p-chlorophenyl)ethylene (p,p'-DDNU). In addition, p,p'-DDNU could be transformed to 2,2-bis(p-chlorophenyl)ethane (p,p'-DDNS) and other high-order metabolites. On the other hand, the conversions of p,p'-DDD to p,p'-DDMU and 1-chloro-2,2-bis-(p-chlorophenyl)ethane (p,p'-DDMS) to p,p'-DDNU were deemed slow in anaerobic sediment. Therefore, the present study confirmed all the degradation pathways involving reductive dechlorination and p,p'-DDE being a more important precursor for p,p'-DDMU than p,p'-DDD in anaerobic sediment, as proposed previously. On the other hand, the present study suggested that p,p'-DDMU instead of p,p'-DDMS was more likely the precursor for formation of high-order metabolites. Based on the current assessments, use of (DDD+DDE)/DDTs to indicate whether there is fresh DDT input may lead to large uncertainties if the concentrations of high-order metabolites are not negligible. Similarly, ecological risk assessment associated with DDT should be conducted with consideration of high-order DDT metabolites.

Proteolysis in skeletal muscle is mainly carried out by the activity of the ubiquitin-dependent proteolytic system. For the study of protein degradation through the ubiquitin-proteasome pathway, we used a model of hyperthermia in murine myotubes. In C2C12 cells, hyperthermia (41°C) induced a significant increase in both the rate of protein synthesis (18%) and degradation (51%). Interestingly, the addition of the β(2) -adrenoceptor agonist formoterol resulted in a significant decrease in protein degradation (21%) without affecting protein synthesis. The decrease in proteolytic rate was associated with decreases in gene expression of the different components of the ubiquitin-dependent proteolytic system. The effects of the β(2) -agonist on protein degradation were dependent exclusively on cAMP formation, because inhibition of adenylyl cyclase completely abolished the effects of formoterol on protein degradation. It can be concluded that hyperthermia is a suitable model for studying the anti-proteolytic potential of drugs used in the treatment of muscle wasting., (Copyright © 2011 Wiley Periodicals, Inc.)

Background Aims: Obesity is correlated with chronic low-grade inflammation. Thus the induction of inflammation could be used to stimulate adipose tissue formation in tissue-engineering approaches. As nitric oxide (NO) is a key regulator of inflammation, we investigated the effect of NO and its downstream signaling molecule guanosine 3',5'-cyclic monophosphate (cGMP) as well as adenosine 3',5'-cyclic monophosphate (cAMP) on preadipocytes in vitro., Methods: Preadipocytes were isolated from human subcutaneous adipose tissue, cultured until confluence, and differentiated. The NO donor diethylenetriamine (DETA)/NO (30-150 microm) was added during proliferation and differentiation. Additionally, cGMP/cAMP analogs 8-bromoguanosine 3',5'-cyclic monophosphate (8-Br-cGMP), 8-(4-chlorophenylthio)-guanosine 3',5'-cyclic monophosphate (8-pCPT-cGMP) and 8-bromoadenosine 3',5'-cyclic monophosphate (8-Br-cAMP), and the adenylyl cyclase activator forskolin, specific guanylyl cyclase inhibitor 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one (ODQ) and adenylyl cyclase inhibitor 2'-5'-dideoxyadenosine (ddA), were applied. Proliferation and differentiation were evaluated., Results: DETA/NO in combination with the standard differentiation procedure significantly enhanced maturation of precursor cells to adipocytes. Proliferation, in contrast, was inhibited in the presence of NO. The application of cGMP and cAMP, respectively, increased pre-adipocyte differentiation to an even higher extent than NO. Inhibitors of the underlying pathways caused a significant decrease in adipogenic conversion., Conclusions: Our results support the application of NO donors during transplantation of preadipocytes in a 3-dimensional setting to accelerate and optimize differentiation. The results suggest that, instead of the rather instable and reactive molecule NO, the application of cGMP and cAMP would be even more effective because these substances have a stronger adipogenic effect on preadipocytes and a longer half-life than NO. Also, by applying inhibitors of the underlying pathways, the induced inflammatory condition could be regulated to the desired level.

Spatial distribution of selected contaminants in the surface sediments of Santa Monica Bay (SMB), California was investigated. Sediments were analyzed for DDTs (DDT and metabolites), polychlorinated biphenyls (PCBs), polycyclic aromatic hydrocarbons (PAHs), linear alkylbenzenes (LABs) and coprostanol. Effluent samples from the Hyperion Treatment Plant (HTP), which discharges treated municipal wastewater effluents into SMB, were also analyzed. The inter-correlation in the distribution trends of contaminants was examined. The concentrations of contaminants were interpolated in a geographic information system to visualize their spatial distribution in the Bay. Inventories of the contaminants were also estimated. The concentrations of coprostanol, LABs and PCBs are very high only in the vicinity of the sewage outfall whereas PAHs and DDTs occur widespread in the Bay. The poor correlation of DDTs with LABs, PAHs or coprostanol content confirms the historic origin of DDTs and their absence in the contemporary wastewaters. Moderate correlation of DDTs with PCBs implies historic deposits as a major origin of PCBs. There are hot spots of DDTs at water depths of 60 and 100m and the inventory of DDTs in Bay sediments is insignificant compared to that estimated in the Palos Verdes Shelf which extends from the southern edge of Redondo Canyon around Palos Verdes Peninsula. The concentration of toxic contaminants was examined according to published sediment quality guidelines. About 20 stations contain p, p'-DDE and/or total DDTs above ERM and, PCBs between ERL and ERM indicating potential for adverse biological effects.

In a previous work we found that the insecticide 1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane (DDT), inhibits the accumulation of cAMP as induced by the bovine thyroid stimulating hormone (bTSH) in cells transfected with the TSH receptor. In this work, we demonstrate that the DDT molecular analogues, diethylstilbestrol and quercetine, are more potent inhibitors of the TSH receptor activity than DDT itself. The notion that all these compounds interfere with nuclear estrogen receptors, as either agonists (DDT and diethylstilbestrol) or antagonists (quercetin), prompted us to test the ability of the steroid hormone 17-beta-estradiol to inhibit the TSH receptor activity. We found that estrogen exposure causes a modest but significant inhibition of the bTSH induced cAMP accumulation both in transfected CHO-TSH receptor and Fischer Rat Thyroid Low Serum 5% (FRTL-5) cells. When applied to CHO cells transfected with the luteinizing hormone receptor, 17-beta-estradiol proved capable of inhibiting the hCG induced cAMP accumulation at a concentration as low as 10nM, though the effect was not greater than 35%. The effect of 17-beta-estradiol was not estrogen receptors mediated, as co-transfection of the estrogen receptor alpha and beta subunits with LH receptor caused cAMP to increase above the level attained by the sole hCG stimulation, and not to decrease it as expected. These data suggest the presence of a steroidal-like allosteric binding site on glycoprotein hormone receptors.

DDT metabolism in humans yields DDA as the principal urinary metabolite and potential exposure biomarker. A method for DDA analysis in human urine was developed using pentafluorobenzyl bromide and diisopropylethyl amine. Dried hexane extracts were reacted for 1 hour at room temperature. The stable DDA-pentafluorobenzyl-ester derivative was analyzed by gas chromatography-electron capture detector (GC-ECD) and confirmed by gas chromatography-mass spectrometry (GC-MS) in selective ion monitoring mode. The limit of detection for DDA was 0.1 microg/L urine by GC-ECD and 2 microg/L urine by GC-MS, with a relative standard deviation of 12%. Urine specimens from DDT applicators in Swaziland and South Africa were analyzed to evaluate the method. The mean DDA levels during the spray season and post season were 59 and 11 microg/L, respectively. These results must be interpreted cautiously because different groups of workers provided urine specimens in each case. The DDA urinalysis may be a feasible monitoring strategy for low-level occupational and residential DDT exposure assessment in antimalaria campaigns.

This paper presents results based on determination of residue levels of 1,1,1-trichloro-2,2-bis[p-chlorophenyl]ethane (DDT) and its metabolites in five fish species from Rwenshama landing site on Lake Edward, Uganda. The residue levels were analysed by using a GC-ECD and confirmed by GC-MS. The DDT residues detected in fish samples from Lake Edward were p,p'-DDT, o,p'-DDT, p,p'-DDE, o,p'-DDE, p,p'-DDD and o,p'-DDD. All the analysed samples of fish presented mean DDT residues ranging from non-detectable levels to 68 microg kg(-1) fresh weight. High levels of p,p'-DDT were detected in comparison to the metabolites (p,p'-DDE and p,p'-DDD). The detection of higher levels of p,p'-DDT than p,p'-DDE and p,p'-DDD, in most fish samples, suggests recent exposure of fish to DDT. Generally, most of the fish samples had residue levels below the maximum residue limits (MRL) recommended by FAO/WHO Codex Alimentarius Commission.

In this work, a large-volume on-column injection method for the analysis of 21 organochlorine pesticides, including HCH isomers, DDT derivatives and cyclodiene derivatives, was optimized. The solvent selected to carry out the study was ethyl acetate and the injection volume was 100 microl. Some factors were introduced in a Plackett-Burman design to determine their influence in the vaporization efficiency. The significant factors were then studied by a univariate procedure and sorted according to their importance on the response. The effect of the injection conditions on the peak resolution was also noted. The conditions selected according to sensitivity and resolution were: initial oven temperature 75 degrees C, injection speed 20 microl s(-1), solvent vapor exit (SVE) valve closure time 60 s, initial pressure 100 kPa and isothermal oven time 1 min. Analytical characteristics expressed in terms of precision, linear range, and limit of detection have been determined and compared with those obtained by splitless injection. The degradation of endrin and p,p'-DDT thermolabile pesticides was evaluated for both injection techniques. Finally the developed method was successfully applied to the ultratrace analysis of pesticides in natural waters. With this purpose a micro liquid-liquid extraction method using 2 ml of ethyl acetate to extract 10 ml of water sample was proposed. Recoveries between 69 and 107% were obtained with a very good precision (0.2-1.3%) for the studied pesticides, except for p,p'-DDD. Detection limits between 0.3 and 25 ng l(-1), which fulfill the limits established by the new water directive 2008/105/EC, were achieved. The MLLE method was compared with the SPE method by the analysis of some water samples using both procedures, and good concordance was obtained.

The coastal lagoon system of Laguna de Terminos, Campeche, Mexico, a natural reserve since 1994, was investigated for contamination by agricultural and industrial chemical residues. Water, sediment and biota samples were analyzed for a wide variety of organochlorine and organophosphorus compounds. Chlorpyrifos was detected in water in concentrations up to 72 pgL(-1) and, amongst organochlorine compounds, summation operator PCB were measured averaging 1177 pgL(-1) and summation operator DDT 279 pgL(-1). Residues of chlorinated compounds were present in sediments and in biota with summation operator DDT averaging 190 pg g(-1) and 5876 pg g(-1) in sediment and oysters, respectively. Results show that the more widespread contaminants in the Laguna were residues of chlorinated hydrocarbons, such as DDTs, PCBs, endosulfan, and lindane. Concentrations of residues were not at an alarming level and were even lower than reported for other costal lagoons of the region. Still there is a need to implement control measures on persistent and bioaccumulative compounds that may reach the aquatic system of Laguna de Terminos.

Bioaccumulation factors (BAFs) and biota-sediment accumulation factors (BSAFs) are frequently used to predict contaminant bioaccumulation in risk assessments. Development of these parameters is often hindered by uncertainty regarding the spatial scale of contaminant transfer from sediments to biota. We present a simple statistical method for optimizing bioaccumulation parameters (BAF and BSAF) in aquatic species, such as fish, whose exposure history may occur over broad spatial scales. For 6 finfish species sampled in San Francisco Bay, San Diego Bay, or the Southern California Bight, California, USA, the spatial scale of correlation was optimized using regression analysis. The ranges identified for pairing biota and sediment observations generally corresponded to the known life histories of the species and with laboratory tests comparing relationships observed for 28-d Macoma spp. This procedure may be useful for identifying appropriate species and spatial scales to predict bioaccumulation and for developing data sets of corresponding sediment and tissue residues.

This paper presents the results of an investigation on association between dietary intakes and human milk concentrations of dichlorodiphenyltrichloroethane and metabolites (DDTs) of two populations from Beijing and Shenyang, China. We analyzed human milk samples from 76 women and 287 composite food samples covering major food categories for concentrations of DDTs. We also collected information on food consumptions and calculated dietary intakes of DDTs of the participants on individual basis. The median values of the measured DDTs in human milk were 125 ng/g lipid and 132 ng/g lipid for the samples from Beijing and Shenyang, respectively. The mean (+/-standard deviation) daily dietary intakes of DDTs by the two groups were 32.0 +/- 14.2 ng/kg day and 27.9 +/- 11.3 ng/kg day, respectively. The temporal trends of decreasing in DDTs and increasing in DDE/DDT ratio suggested that the residuals were primarily from historical application. We found a significant correlation between human milk concentration and daily dietary intake of DDTs, while the dietary intake could explain 22% of the variation in the DDTs in human milk. In addition to dietary exposure, we also found that maternal body mass index (body weight divided by the squared height), body weight, body height, and mother's age contributed significantly to the variation of DDTs in human milk after intake normalization. The result of a probabilistic risk assessment indicated that the exposure of infants to DDTs through breast feeding would be a public health concern for years to come, although breast feeding is still recommended.

A useful tool based on a single-step extraction and clean-up procedure for the determination of 15 organohalogenated pollutants (including brominated flame retardants) in aquaculture samples, using aluminium oxide basic and acidic silica gel in the supercritical extraction cell followed by gas chromatography with electron capture detection or mass spectrometry has been developed. This effective clean-up step ensures a minimum of chromatographic difficulties related to complex matrix components such as aquaculture feed. The extraction procedure has been screened by a fractional factorial design for the preliminary statistically significant parameters. The factors selected were extraction temperature, pressure, static extraction time, dynamic extraction time and carbon dioxide flow rate. The Doehlert design, followed by a multicriteria decision-making strategy, was then performed in order to determine the optimum conditions for the two most significant factors: pressure (165 bar) and dynamic extraction time (27 min). Under optimal conditions, the procedure developed with GC-MS/MS provides an excellent linearity, detection (0.01-0.2 ng g(-1)) and quantification limits (0.05-0.8 ng g(-1)) for most of the analytes investigated. The feasibility of the proposed supercritical fluid extraction method was validated by analysing two reference materials and fish feed and shellfish samples with satisfactory results.

In this study, organochlorine insecticides (OCPs), DDT metabolites and HCH isomers were quantified in mustard, groundnut and sunflower oils collected from different regions of India. The maximum level of 222.0 ng g(-1) and minimum level of 0.2 ng g(-1) of SigmaDDT were detected in mustard oil from Deoria and Varanasi while those of SigmaHCH i.e., 1500 ng g(-1) and 8.0 ng g(-1) in mustard oil from Deoria and Kanpur, respectively. However, the maximum total concentration of alpha-, beta- and gamma-HCH was in mustard oil, whereas maximum delta-HCH was in groundnut oil. However, the maximum percentage of p,p'-DDT was in mustard oil while maximum p,p'-DDE was in groundnut oil. The study calls for periodical monitoring to ensure safe supply of edible oils to consumers.

The bioconcentration and distribution pattern of p,p'-DDT 1,1,1-1trichloro-2,2-bis(2-chlorophenyl-4-chlorophenyl)-ethane] and its main metabolites (p,p'-DDD [1,1-dichloro-2,2-bis (4-chlorophenyl) ethane] and p,p'-DDE [1,1-dichloro-2,2-bis (4-chlorophenyl) in adipose tissue, liver, brain, kidney, thymus, and testis were examined in young rats after 10 days of intraperitoneal injection of 50 and 100 mg of p,p'-DDT/kg of body weight. Analyses were performed by high-resolution gas chromatography. p,p'-DDT was found to be accumulated in a dose-dependent manner with the highest concentration in adipose tissue. However, in brain, the accumulation of pesticide was low and remained unchanged at the higher dose. This difference may relate to the protective role of the blood-brain barrier, which limits the access of the xenobiotic in the cerebral compartment, and to the differential tissue lipid composition. Although tissues concentration of p,p'-DDE and p,p'-DDD correlated positively to total p,p'-DDT levels, the active role in detoxification of pollutants may explain why p,p'-DDD is more abundant in liver than in the rest of organs. On the contrary, in brain, the concentration of p,p'-DDE is higher than that of p,p'-DDD, suggesting that the metabolism of the parent insecticide proceeds via more than one pathway.

Objective: We investigated the role of the inward rectifier potassium (KIR) channel and the cyclic AMP-dependent pathway in mediating vasorelaxation induced by the prostacyclin analogue cicaprost., Methods: Small vessel myography was used to assess responses to cicaprost in segments of rat tail artery contracted with phenylephrine. Microelectrode recordings were made from helical strips to assess effects on membrane potential., Results: Cicaprost caused relaxation and hyperpolarisation that were significantly inhibited by Ba2+ (30-100 microM), a known blocker of KIR channels. Raising extracellular K+ from 5 to 15 mM elicited membrane hyperpolarisation and an endothelium-independent relaxation that was blocked by Ba2+ (30-100 microM), suggesting the existence of functional KIR channels on the smooth muscle. In contrast, neither glibenclamide (10 microM), a blocker of ATP-sensitive K+ channels, nor fluoxetine hydrochloride (100 microM), a blocker of G-protein-gated inward rectifier K+ channels, nor pertussis toxin (PTX; 1 microg/ml), which irreversibly inhibits Gi/Go, reduced relaxation to cicaprost. Indeed, PTX significantly potentiated responses. Relaxation to cicaprost was not mediated by NO but was partially endothelium-dependent, consistent with a similar inhibition by a combination of charybdotoxin (0.1 microM) and apamin (0.5 microM), blockers of endothelium-derived hyperpolarising factor (EDHF). However, relaxation was unaffected by adenylyl cyclase (SQ22536, dideoxyadenosine) or protein kinase A (Rp-2-O-monobutyryl-cAMP) inhibitors, consistent also with Ba2+ only weakly inhibiting relaxation to the adenylyl cyclase activator forskolin., Conclusion: We conclude that cicaprost relaxes rat tail artery by activating KIR channels with some involvement from EDHF. The mechanism appears to be largely independent of cyclic AMP and Gi/Go, although the latter appears to counteract relaxation through an unknown pathway and/or receptor.

The aim of this study was to evaluate sediments of the Dniester River, in the former Soviet republic of Moldova, for the occurrence of agricultural pesticides, polychlorinated biphenyls (PCBs), and heavy metals. In October 2001, sediment samples were collected at three locations of the Dniester River: upstream, downstream, and at the tributary of the River Byk. DDT and its metabolites were observed most frequently. Total DDT sediment concentrations ranged from 8.2 to 34.7 ng/g dry weight with the highest average concentration detected at the tributary location. Heptachlor epoxide (< 0.2-3.0 ng/g dry weight), chlordane (< 0.1-6.1 ng/g dry weight), endrin (< 0.2-2.5 ng/g dry weight), phosalone (< 0.2-1.1 ng/g dry weight), and methylparathion (< 0.2-16.8 ng/g dry weight) were also detected. Total PCB concentrations observed in sediments ranged from 68 to 763 ng/g dry wt. Total average PCB concentrations were significantly (p < or = 0.004) higher for sediments from the downstream sampling sites compared to the upstream samples. Tetra- and trichlorobiphenyls accounted for 84 and 88% of the total residues measured in the downstream and tributary locations, respectively. In contrast, heptachlorobiphenyls were the prevailing homologues in sediments from the upstream sampling site, contributing 51% of the total PCB concentration. Predominant PCB homologues were: trichlorobiphenyl congener 28 and 37, tetrachlorobiphenyl congeners 44, 49, 52, 74, 77, and 81, and heptachlorobiphenyl congener 170. Sediment concentrations of SigmaDDE (19.7 ng/g dry weight) in the tributary, heptachlor epoxide (3.0 ng/g dry weight) in the downstream, and nickel (128-170 microg/g dry weight) in all locations exceeded Probable Effect Levels (PELs) established for sediment quality in fresh water, indicating probable environmental stress and the potential for adverse effects to benthic organisms in the Dniester River.

We tested the effectiveness of combined DNA vaccines encoding antigens Ag85B, MPT64 and MPT83 from Mycobacterium tuberculosis on cattle. Our results showed that calves treated with combined DNA vaccines in the presence of dimethyldioctyldecyl ammonium bromide (DDA) or saline elicited a strong gamma interferon (IFN-gamma) response 1 or 2 months after the third vaccination. All three antigens induced substantial levels of IFN-gamma production 1 month after the bacterial challenge, when the BCG-driven IFN-gamma levels dropped to less than one third of their peak values. Animals receiving combined DNA vaccines produced highest amounts of IgG antibody titer 2 months after the third vaccination. Steady state low IgG levels were found 2 months after bacterial challenge. A few small lung and lymph node lesions were detected in 1/5 animals treated with combined DNA vaccines, whereas 3/5 of BCG-treated and 5/5 of vector-control calves showed larger and significantly more lesions. About 70- to 100-fold fewer bacteria were found in the lungs and lymph nodes of combined DNA vaccine-treated animals compared to those of the control group. Histopathological analyses showed that vaccinated calves possessed substantially improved post-infection lung and lymph node pathology relative to the controls. Our data indicate that combined DNA vaccines may be used in cattle to combat bovine tuberculosis.

The content of 21 organochlorine pesticides were studied in vegetation samples of a highly contaminated area by isomers of hexachlorocyclohexane (HCH) located close to a former industrial area in Galicia (NW Spain). Five species of plants were collected at different points of the contaminated area and the different parts of the plants were separated in order to study differences in accumulation capabilities. Samples were extracted employing microwave energy followed by a clean-up step using solid phase extraction and finally determined by GC-ECD. The results obtained show that the most abundant pesticides are HCHs isomers, being the main isomers beta-HCH and alpha-HCH in all samples whereas delta-HCH and gamma-HCH were at lower levels. Some other pesticides such as p,p'-DDT, p,p'-DDD and p,p'-DDE were also present in much lower amount in some of the samples. Several degradation products of HCH were also identified in some samples by GC-MS.

Organochlorine pesticides and polychlorinated biphenyls are widely used in agriculture and industry, respectively. The present study assessed the burden of environmental pollutants in the follicular fluid of farm animals (cattle, sheep, goats, and pigs). An analytical method combining a solid-phase extraction with (C(18)) for clean-up and GC-electron capture detection using a capillary column was implemented for isolation and determination of organochlorine pesticides and polychlorinated biphenyls (PCBs). Of the organochlorine pesticides, hexachlorobenzene (HCB), hexachlorocyclohexane (HCH, alpha-, beta-, and gamma-isomers), dieldrin, heptachlor epoxide, and the DDT-related chemicals (o,p'-DDE, p,p'-DDE, p,p'-DDD, p,p'-DDT) were detected and of the PCBs, the congeners PCB-52, -101, -138, -153, and -180 were detected. In all species of farm animals, the most frequently detected pollutant was gamma-HCH (90-100% of samples) followed by HCB (80-100%), and p,p'-DDE (75-90.91%). Species differences in the concentrations of HCB, beta-HCH, heptachlor epoxide, and DDT-related chemicals in follicular fluid were noted as well as differences in the concentrations of some pollutants within the same species.

DDT homologues and PCBs were determined in eggs of two stationary species, great crested grebe (Podiceps cristatus) and mallard (Anas platyrhynchos), in three breeding areas of Lake Maggiore in 1998. As expected, much higher contamination levels of both DDTs and PCBs were found in the fish-eating great crested grebe than in the mallard. In many eggs these values exceeded the safety thresholds proposed for aquatic bird protection. However, no significant correlation could be found between the two classes of pollutants and shell thickness in great crested grebe eggs. While a DDT manufacturer was detected as the main source of DDT pollution in the Lake Maggiore environment, the reason for the surprisingly high PCB level remains unknown.

Molecularly imprinting sol-gel materials for DDT using both a noncovalent and a covalent approach was examined. A nonpolar porous sol-gel network was created through the use of the bridged polysilsesquioxane, bis-(trimethoxysilylethyl)benzene (BTEB), as the principal sol-gel component. Noncovalent molecular imprinting was deemed unsuccessful, presumably because of the lack of strong intermolecular interactions that can be established between the DDT and the sol-gel precursor. A covalent imprinting strategy was employed by generating a sacrificial spacer through the reaction of two 3-isocyanatopropyltriethoxysilanes with one of two different template molecules: 4,4'-ethylenedianiline (EDA) or 4,4'-ethylidenebisphenol (EBP). After formation of the sol-gel, the bonds linking the spacer template to the matrix were cleaved in a manner that generated a pocket of the appropriate size bordered by amine groups that could aid in the binding of DDT through weak hydrogen bonding interactions. Experiments indicated that DDT could be bound selectively by such an approch. To generate a sensor, an environmentally sensitive fluorescent probe, 7-nitrobenz-2-oxa-1,3-diazole, (NBD) located adjacent to the DDT binding site was used to transduce the binding of analyte. EDA-imprinted sol-gels, deposited as films on glass microscope slides, were shown to quantitatively detect DDT in water to a limit-of-detection of 50 ppt with a response time of <60 s. Repeat measurements could be made with the same sensing films after rinsing with acetone between each measurement. The EDA sensing material was selective for DDT and other structurally similar molecules. However, the sensing film design was limited by the relatively minor changes in fluorescence intensity upon binding DDT. This situation may be remedied by an alternative methodology that can facilitate attachment of the NBD fluorophore in an optimal position proximal to the binding pocket.

Polychlorinated biphenyls (PCBs), DDT-related substances and hexachlorocyclohexane (HCH) isomers were analysed in fish from Estonian coastal waters. The content of persistent organic pollutants (POPs) in fish (perch (Perca fluviatilis) and flounder (Platichtys flesus)) from the coastal waters of Estonia is not a danger to human health. Even the maximum concentrations that we have determined are lower than the standards set by the World Health Organization (WHO). The results do not exclude the need to monitor toxicants in fish also in the future, because the use of hazardous chemicals in the Baltic Sea region will probably continue.

The potential for anaerobic biodegradation of 1,1,1-trichloro-2,2-bischlorophenylethane (DDT), 1,1-dichloro-2,2,-bischlorophenylethane (DDD), and dichlorodiphenylchloroethylene (DDE) in anoxic sediment slurries collected from the Keelung River was investigated in this study. o,p'- and p,p'-DDT were dechlorinated to o,p'- and p,p'-DDD, respectively, and then transformed to other compound(s). 1-Chloro-2,2-bis (p-chlorophenyl) ethylene (DDMU) and trace amount of dichlorobenzophenone (DBP) were detected in sediment slurries amended with p,p'-DDT or p,p'-DDD. DDMU was also detected in sediment slurries amended with p,p'-DDE. The relative transformation rates for both o,p'- and p,p'-isomers of DDT, DDD, and DDE were DDT>DDD>DDE. Re-addition of DDT, DDD, or DDE to the sediment slurries after initial removal enhanced the respective dechlorination rates. The transformation rates of the p,p'-isomers of both DDT and DDD were faster than those of the respective o,p'-isomers. p,p'-DDT dechlorination in the p,p'-DDT-adapted sediment slurries were inhibited by the addition of molybdate, or molybdate plus sulfate, but not inhibited by the addition of sulfate. Addition of bromoethane-sulfonic acid (BESA) slightly inhibited p,p'-DDT dechlorination. Non-adapted sediment slurries lost the ability to dechlorinate pentachlorophenol during adaptation to p,p'-DDT. p,p'-DDD was the major transformation product of p, p'-DDT in 3,4,4',5-tetrachlorobiphenyl-adapted sediment slurries, which suggested that the microbial community in the 3,4,4',5-CB-adapted sediment was unable to remove chlorine from the aromatic rings of p,p'-DDT.

Organochlorine compounds such as o,p'DDT can mimic estrogen effects. We compared the effects of o,p'DDT and six other DDT congeners to the effects of estradiol by comparing in vivo color changes in the reed frog (Hyperolius argus). Premature female color pattern induction in H. argus is specific to estrogens and the current study suggests that this assay has potential for use in discriminating between xenobiotic estrogens and non-estrogens. Animals were treated at forelimb emergence and maintained in treated solution until final evaluation. Estradiol, o,p.DDT (0.1 microg/ml), o,p'DDE (1 microg/ml) and o,p'DDD (1 microg/ml) prematurely induced adult female coloration patterns in juvenile animals, whereas p,p'DDT, p, p'DDE and p,p'DDD did not.
