Abstract

Leaf anatomy, photosynthetic performance, and the role of poly(ADP-ribosyl)ation in maintaining genomic stability in winter and summer leaves of Cistus incanus L. were investigated to evaluate the strategies allowing this species to grow in a Mediterranean climate. Measurements on summer leaves (SL) were conducted at midday and on the following morning to assess the reversibility of changes induced by high temperatures and irradiances reached at midday. Winter leaves (WL) were thicker with a lower leaf density and higher intercellular spaces compared with SL, which showed reduced net photosynthesis ([P.sub.n]) and a decrease in Rubisco activity. Compared with WL, SL showed a higher thermal dissipation ([Φ.sub.NPQ]) in compensating for the reduced [P.sub.n]. This allowed minimization of ROS production (low [Φ.sub.NO]) and hence prevented photo-oxidative damage. In SL, poly(ADP-ribose) polymerase (PARP) activity was down-regulated, probably allowing leaves to become tolerant to high temperatures and irradiances. SL are also characterised by chloroplasts occupying larger cell volume and well exposed to intercellular spaces. This may well favour better liquid-phase C[O.sub.2] diffusion towards carboxylation sites, contributing to reducing the negative influence of high leaf density and the low percentage of intercellular spaces on photosynthesis. Les auteurs ont examine l'anatomie foliaire, la performance photosynthetique et le role de la poly(ADP-ribosyl)ation dans le maintient de la stabilite genomique des feuilles estivales et hivernales du Cistus incanus L., afin d'evaluer les strategies permettant a cette espece de croitre en climat mediterraneen. Ils ont effectue des mesures sur les feuilles estivales (FE) au milieu du jour et le matin suivant pour evaluer la reversibilite des changements induits par des temperatures et les irradiances elevees atteintes en mi-journee. Les feuilles hivernales (FH), plus epaisses ont une densite plus faible et de plus grands espaces intercellulaires, comparativement aux FE et montrent une photosynthese reduite ([P.sub.n]) et une activite Rubisco amoindrie. Comparativement aux FH, les FE montrent une dissipation thermique ([Φ.sub.NPQ]) en compensation pour la [P.sub.n] reduite. Ceci permet de minimiser la production de ROS (faible [Φ.sub.NO]) et consequemment de prevenir le dommage par photooxydation. Chez les FE, l'activite de la poly(ADP-ribose) polymerase (PARP) est sous regulee, probablement permettant ainsi aux feuilles de devenir tolerantes aux temperatures et irradiances elevees. Les FE se caracterisent egalement par des chloroplastes occupant une volume cellulaire plus important et bien exposes aux espaces intercellulaires. Ceci pourrait bien favoriser la diffusion en phase liquide du C[O.sub.2] vers les sites de carboxylation, contribuant a la reduction de l'influence negative de la forte densite foliaire et du faible pourcentage des espaces intercellulaires, sur la photosynthese. Abstract

Long-storage tomato landraces are niche crops traditionally cultivated in southern Italy for drought tolerance and high nutritional and organoleptic quality of fruits. Nowadays, there is a growing interest in these crops as for exploiting biodiversity and the "Made in Italy" food production. We studied the physiological and biochemical performances of two landraces in response to soil water deficiency and re-watering. The potted-plant experiments were carried out in greenhouse during 2012 and 2013. During the first year, an intense water stress determined a decrease of stomatal conductance (gs) and net CO2 assimilation (A) along with an increase in both proline and ABA content, without any effect on photochemistry. In the second year, a more severe water stress leaded to a greater decline of both gs and A, and a reduction in photochemistry. During the re-watering, the full recover of gas exchanges and photochemistry, indicated the occurrence of efficient stomatal control and photochemical regulation, rather than impairment of photosynthetic apparatus. The activity of poly ADP-ribose polymerase (PARP), a key regulator of the energy homeostasis during stress conditions, showed during the first cycle of stress an evident increase, followed by a decrease. Re-watering, instead, produced a new non-expected increase of activity, which returned to control values during the second cycle of water stress. The two tomato landraces succeeded to cope with water stress by a useful synergy of biochemical and physiological regulatory mechanisms. Abstract

Ionising radiation may have different effects on plant metabolism, growth and reproduction, depending on radiation dose, plant species, developmental stage and physiological traits. In this study, exposure of dwarf bean plants to different doses of X-rays (0.3, 10, 50, 100 Gy) was investigated with a multidisciplinary approach consisting of morphological, ecophysiological and biochemical analysis. Both mature and young leaves still growing during the X-rays exposure were compared with non-irradiated control leaves. In particular, leaf expansion, leaf anatomy and functional traits, as well as photosynthetic pigment content and Rubisco expression were analysed. Moreover, the activity of poly(ADP-ribose) polymerase (PARP) was also measured as an indicator of radiation-induced DNA damage. Our data showed that leaf growth is affected by high levels of radiation and demonstrate that mature leaves are more radio-resistant than young leaves, which experience severe dose-dependent changes in leaf functional traits. In particular, young leaves exhibited a reduction of area and an increase in specific mass and dry matter content, as well as a decline in Rubisco activity. Moreover, they showed elevated PARP activity and an increase in phenolic compounds in wall cells if compared with mature leaves. Both of these strategies have been interpreted as a way to help developing leaves withstand irradiation.
