Film-mulched rock outcrops are a proven way to effectively prevent preferential flow at the rock–soil interface in rocky karst areas, but the impact on rainwater redistribution and maize growth in farmland areas has never been studied. This paper used the dye tracer method at a sprinkling intensity of 1 mm min−1 to study the rainwater redistribution in soils for three different treatments: away from rock (AR), close to rock (CR), and close to film-mulched rock (CFMR). The growth situation of maize (Zea mays L.) according to the different treatments was also studied. It was shown in the study that the rainwater gathered by rock outcrops was mainly confined only within a narrow flow path at the rock–soil interface in the CR treatment, with a preferential flow fraction of 35.78~55.54% and dyeing depth of 15.37~20.00 cm across the three dye application amounts (850 mL, 1700 mL, and 3400 mL) in contrast to the uniform distribution of the rainwater in the soil of the AR treatment, with a preferential flow fraction of 12.31~37.15% and dyeing depth of 6.93~14.00 cm. Interestingly, in the CFMR treatment, there was no preferential flow at the rock–soil interface benefiting from the film's blocking action, and the redistribution of rainwater was mainly based on the matrix flow, with a preferential flow fraction of 24.93~39.62% and dyeing depth of 10.27~18.00 cm, indicating that the film-mulched rock outcrops improved the rock's output capability for gathering rainwater into the surrounding soil. In addition, film-mulched rock outcrops can promote the growth of maize, as indicated by the results for plant height, stem diameter, maximum leaf area, SPAD value, and floral development of maize, which increased in order from AR, CR, to CFMR. Our study suggests that film-mulched rock outcrops have important implications for the efficient use of rainwater and the growth of crops in rocky karst areas, as well as in regions with similar geological characteristics. The present work examined morphological and physiological aspects of osmoregulation in some differently adapted populations of Typha domingensis. Six populations of T. domingensis were grown in waterlogged conditions under four levels of nickel (0, 50, 100 and 1500 mg/kg of Ni). The Ni tolerance was explored on the basis of different characteristics of shoots. Along with the increasing level of Ni stress, all populations showed an increment of proline and glycinebetaine content, low photosynthetic and transpiration rates and decreased chlorophyll content. Analyzed morphological parameters revealed possible adaptations to deal with Ni stress. The less tolerant Sahianwala population produced more aerenchyma, less accumulation of osmolytes and reduced shoot fresh weight under Ni stress than the tolerant ones. Our results suggested that T. domingensis has the capacity to survive under Ni stress and could be used for the absorption of toxic metal ions from industrially polluted water. The complete chloroplast (cp) genome ofCurcuma flaviflora, a medicinal plant in Southeast Asia, was sequenced. The genome size was 160 478 bp in length, with 36.3% GC content. A pair of inverted repeats (IRs) of 26 946 bp were separated by a large single copy (LSC) of 88 008 bp and a small single copy (SSC) of 18 578 bp, respectively. The cp genome contained 132 annotated genes, including 79 protein coding genes, 30 tRNA genes, and four rRNA genes. And 19 of these genes were duplicated in inverted repeat regions. Effect of altitude on leaf responses in Phleum himalaicum populations was evaluated at three different elevation levels, viz. (Low 1200 m.a.s.l.), middle (1600 m a.s.l.) and high (1900 m a.s.l.) in western part of Himalaya. We hypothesized that physico-chemical properties of soil varied along elevation and Phleum populations located at high elevation would adapt more distinct morphological and physiological traits than those originating from middle and low elevation sites. Our study revealed that soil pH, Ec Mg, Ca, and P decreased at high elevation however, significant increase was recorded in soil K, organic matter, and total nitrogen along the elevation gradient. A significant correlation between leaf characteristics and elevation sites was recorded along the gradient. The outcomes of this study showed that highland population had better adjustments under low temperature and exhibited adaptive traits. These were, decreased number of leaves and leaf area, increased leaf blade thickness, intensive sclerification, and greater stomatal and trichome density. Apart from these, high elevation population had more physiological adjustment in terms of low stomatal conductance, low transpiration rate, high water use efficiency, and synthesis of more osmolytes in leaf. We argued that certain level of sugar and protein must be attained by high population to dodge the aggressive climatic forces in order to grow successfully at the highest elevation. Furthermore, altitude between 1600 and 1900 m was more likely an optimum zone for vigorous growth of P. himalaicum at the highest level of elevation. [ABSTRACT FROM AUTHOR]

Skin cutaneous melanoma (SKCM) is a highly fatal form of skin cancer that develops from the malignant transformation of epidermal melanocytes. There is substantial evidence linking autophagy to cancer etiology and immunotherapy efficacy. This study aimed to conduct a comprehensive analysis of autophagy-related genes (ARGs) using TCGA datasets and further explore the potential function of critical ARGs in SKCM progression. We performed comprehensive bioinformatics analysis uses the TCGA dataset. RT-PCR was applied to examine the expression of CAPNS1 in SKCM cells. Lost-of-function experiments were performed to detect the expression of the related proteins. In this search, we screed 70 differentially expressed autophagy-related genes (DE-ARGs), including 33 up-DE-ARGs and 37 down-DE-ARGs. Enrichment assays revealed that these 70 DE-ARGs may exert influence on critical cellular processes such as autophagy, protein kinase activity, and signaling pathways, impacting cell growth, differentiation, survival, and tumor development. Then, we further explore the prognostic value of 70 DE-ARGs and confirmed 18 survival-related DE-ARGs in SKCM patients. Nearly all the 18 DE-ARGs' methylation was negatively correlated with their corresponding expression in SKCM. The 12 survival-related DE-ARGs were used to develop a unique predictive model that effectively classified SKCM patients into high- and low-risk groups with regard to overall survival. Furthermore, tumor environment analysis indicated that the risk score was associated with several immune cells. Among the 12 survival-related DE-ARGs, our attention focused on CAPNS1 which was highly expressed in SKCM patients and predicted a poor prognosis. In addition, we confirmed that knockdown of CAPNS1 distinctly suppressed the proliferation, metastasis and EMT of SKCM cells, and promoted autophagy via regulating Notch signaling pathway.
