Deuterated and tritiated compounds are widely used in numerous applications in chemistry, biology and material science. In the drug discovery and development process, ADME studies require quick access to labelled molecules, otherwise the drug development costs and timeline are significantly impacted. The rapid development of metabolomics has also increased the need for isotopically labelled compounds. In particular, deuterated molecules are used as internal standards for quantitative LC-MS/MS analysis of metabolites in biological fluids and tissues. In this context, a general method allowing the deuterium and tritium labelling of bioactive thioethers using a HIE reaction is described in the first chapter. From a fundamental point of view, this transformation is the first example of (Csp³)-H activation directed by a sulfur atom. In terms of application, this new reaction has been proved to be useful for the preparation of deuterated LC-MS/MS reference materials and tritiated pharmaceuticals owning high specific activity.In the second chapter of this manuscript, the development of a method allowing the cross-dehydrogenative homocoupling of 2-arylpyridines catalyzed by Ru/C is developed. Various substrates with different substituents were efficiently coupled to give the desired dimers in good yield. In terms of application, a series of pyridine-boron complexes derived from the phenyl pyridine dimers were also synthesized and their photophysical properties were studied.In the third chapter, a regioselective palladium catalyzed intramolecular arylation reaction allowing the synthesis of pyridine containing polycyclic compounds is described.; Les molécules marquées par des isotopes de l’hydrogène possèdent de nombreuses applications dans divers domaines tels que la chimie, la biologie ou en science des matériaux. Dans le domaine de la recherche de nouveaux médicaments, les études liées à la pharmacocinétique nécessitent un accès rapide à des molécules marquées afin de ne pas impacter les coûts et les délais de développement. Le développement de la métabolomique a aussi entrainé une augmentation du besoin en molécules marquées isotopiquement. En effet, les molécules deuterées peuvent être utilisées en tant qu’étalons internes pour la quantification rapide des métabolites présents dans des tissus ou des fluides biologiques. La première partie de cette thèse concerne le développement d’une méthode générale de marquage de motifs de type thioéther dans des molécules complexes à l’aide d’une nouvelle réaction d’échange isotopique (catalysée par des nanoparticules de Ruthénium). D’un point de vue fondamental cette transformation représente le premier exemple de (Csp³)-H activation dirigée par un atome de soufre. En termes d’application, cette nouvelle réaction permet la synthèse rapide d’étalons internes pour la quantification LC-MS/MS et le marquage tritium de molécules complexes. La seconde partie de cette thèse relate le développement d’une nouvelle méthode d’homocouplage de phénylpyridines catalysée par Ru/C. Différents substrats comportant des substituants riches et pauvres en électron ont été couplés avec de bons rendements. Ces dimères ont ensuite été utilisés pour synthétiser de nouveaux complexes de bore dont les propriétés photophysiques ont été étudiées. Dans une troisième partie, la mise au point d’une réaction palladocatalysée permettant d’obtenir des molécules polycycliques contenant un motif de type pyridine est développée.

The synthesis and characterisation of a series of dinuclear ruthenium and osmium polypyridyl metal complexes based on the bridging ligands [5-(5′-bipyridin-2′,2′-yl)-3-(pyridin-2-yl)]-1,2,4-triazole (Hpytr-bipy), 2,2′-bis(pyridin-2′yl)-5,5′-bis(pyridin-3′-yl) (bipy-bipy) and 5,5′-bis(pyridin-2′-yl)-3,3′-bis(1,2,4-triazole) (Hpytr-Hpytr) are reported. The dinuclear complexes have been synthesised via a Ni(0) catalysed cross-coupling reaction from brominated precursors. With this approach a mixture of three products is obtained, which are separated by chromatographic methods. The compounds obtained are characterised by elemental analysis, 1H NMR, absorption and emission spectroscopy. The synthetic approach developed offers a new route to asymmetric multinuclear supramolecular structures that is complimentary to the complexes as ligands/complexes as metal approaches.

The application of deuteriation of both ligands and solvents on the photophysical properties of transition metal complexes in solution and glassy matrices is reviewed. The reduction in amplitude and frequency of vibrational modes due to deuterium’s increased mass, relative to hydrogen, has a significant effect on non-radiative deactivation processes, which can occur through both intra- and inter-molecular vibrational coupling. The effect of deuteriation on excited state lifetimes has allowed for its application in probing the nature of excited state decay processes. The effects of isotopic exchange on vibrational spectroscopies such as resonance Raman and low temperature high-resolution emission spectroscopies are also addressed briefly.

C35 Bacteriohopanoids represent the majority of hopanoids produced by bacteria. They bear an additional C5 side chain linked by a carbon/carbon bond to the isopropyl group of the hopane skeleton. The C5 side chains present an impressive structural diversity and carry taxonomic and physiological information. The most common C35 bacteriohopanoids are bacteriohopanetetrol (BHT) and aminobacteriohopanetriol. Moreover, these two compounds are proposed as the parents of most complex bacteriohopanoids. Therefore, elucidation of the biosynthesis of hopanoid side chains is in great interest for a better understanding of the physiological distribution and biological importance of bacteriohopanoids.In this work, ribosylhopane was isolated for the first time from a bacterium. This discovery is a solid proof for the role of ribosylhopane as an intermediate in the biosynthesis of hopanoid side chain. In addition, two genes involved in the hopanoid production in Streptomyces coelicolor have been characterized. Adnosylhopane may be converted into ribosylhopane by a phosphorylase; and an aminotransferase is required for the formation of aminobacteriohopantriol from ribosylhopane. Moreover, we have developed a concise strategy for the hemisynthesis of adenosylhopane and a deuteriated isotopmer. The subsequent incorporation of the deuteriated adenosylhopane into BHT by a cell-free system in Methylobacterium organophilum proved that adenosylhopane is indeed a precursor of C35 bacteriohopanoids., Les bactériohopanepolyols (BHPs) en C35 sont les principaux hopanoïdes trouvés chez les bactéries. Ces composés présentent une chaîne latérale polyhydroxylée en C5 liée par une liaison carbone/carbone au groupement isopropyle du squelette hopane. Ils présentent de nombreuses variations structurales au niveau de la chaîne latérale qui apportent des informations taxonomiques et physiologiques. Le bactériohopanetétrol (BHT) et l’aminobactériohopanetriol sont les composés majoritaires. En outre, ces deux BHPs seront les parents de la plupart des BHPs complexes. L'élucidation de la biosynthèse de la chaîne latérale en C5 des hopanoïdes est donc très intéressante pour une meilleure compréhension de la distribution phylogénétique et la signification biologique des BHPs.Au cours de ce travail, le ribosylhopane a été isolé pour la premier fois chez une bactérie. Cette découverte est une preuve solide confirmant le rôle du ribosylhopane comme intermédiaire dans la biosynthèse des hopanoïdes. En outre, deux gènes, impliqués dans la formation de la chaîne latérale des BHPs chez Streptomyces coelicolor, ont été caractérisés. L’adénosylhopane serait converti par une phosphorylase en ribosylhopane et une aminotransférase est nécessaire pour la formation de l’aminobactériohopanetriol du ribosylhopane. De plus, nous avons développé des synthèses concises de l’adénosylhopane et d’un isotopomère bisdeutérié. L’analogue marqué a par la suite été incorporé dans le BHT par un système acellulaire de Methylobacterium organophilum et le suivi du marquage nous a permis de démontrer l’implication de l’adénosylhopane dans la biosynthèse des hopanoïdes en C35.

In aqueous solutions, [RuCl2(TPPMS)(2)](2) (TPPMS = 3-sulfonatophenyldiphenylphosphine) catalyzes the exchange between H-2 and D2O or between D-4 and H2O with high efficiency (up to 1 252 turnovers per hour) under mild conditions (298 K, 2 MPa H-2). The exchange rate strongly depends on the solution pH. Similar H/D exchange is observed with the water soluble tertiary phosphine complexes [RhCl(TPPMS)(3)], [RhCl(TPPTS)(3)] and [RuCl2(PTA)(4)] as catalysts (TPPTS = tris(3-sulfonatophenyl)phosphine. PTA = 1,3,5-triaza-7-phosphadamantane) which, however, show lower activity. The results described in the literature and our previous research confirms the efficiency of the reduction reactions of α, β-unsaturated compounds and carbonyl compounds mediated by the Ni-Al alloy in alkaline (NaOH) aqueous solution. Some particularly experiments for reduction of crotonic and cinnamic acid, was designed to bring information on the mechanism of reduction reaction with Ni-Al alloys in alkaline aqueous media. Also it was developed several deuteriation reaction of this α, β-unsaturated acids. The analysis of the isotopic distribution shows a significant and reproducible difference between Cα and Cβ that help us to draw some conclusions. In addition, recent researches give us additional arguments concerning the existence of anionic aluminum clusters with water. All this information contributes to propose a mechanism of reduction reaction and could provide new possibilities for development of this reaction: improving yield and selectivity, promoting a new synthesis etc.
