This study explored the influence of genetic and environmental factors on adolescent anxiety. Ninety-eight monozygotic and dizygotic twins from Chongqing, China (aged 15–18 years) were assessed for anxiety with the Self-Rating Anxiety Scale (SAS). The Parenting Styles and Dimensions Questionnaire (PSDQ) and Strengths and Difficulties Questionnaire (SDQ) were applied to assess environmental factors. Venous blood was drawn from the twins for zygosity determination. Structural equation modeling was performed to evaluate the effects of additive genetic factors (A), common environmental factors (C), and individual-specific environmental factors (E) on adolescent anxiety. The estimates of A and E on adolescent anxiety were 0.34 (95% CI = 0.12–0.53) and 0.66 (95% CI:0.47–0.89), respectively. The environment played an important role in adolescent anxiety. Adolescent anxiety was significantly positively correlated with peer relations (r = 0.606, p < 0.05) and negatively correlated with prosocial behavior (r = 0.207, p < 0. 05). No sex differences were observed. Adolescent anxiety was influenced by both genetic and environmental factors. The individual-specific environmental factors played an important role. The sleep homeostatic process in adults is moderately stable over time and unique to an individual. Work in transgenic mice has suggested a role of genes in sleep homeostasis. The current study quantified the genetic contribution to sleep homeostasis in adolescence. We use slow wave energy (SWE) as a metric for sleep pressure dissipation during sleep. This measure reflects both sleep intensity and duration. High‐density (58 derivations) sleep electroencephalogram (EEG) was recorded in 14 monozygotic and 12 dizygotic adolescent twin pairs (mean age = 13.2 years; standard deviation [SD] = 1.1; 20 females). SWE at the end of sleep was quantified as the cumulative delta power (1–4.6 Hz) over the night. We also examined the time constant of the decay and the level of slow wave activity (SWA) at the beginning of the sleep episode. Structural equation modelling was used to quantify the amount of variance in SWE and the dissipation of sleep pressure due to genes. We found that most (mean = 76% across EEG derivations) of the variance in SWE was due to genes. In contrast, genes had a small (mean = 33%) influence on the rate of dissipation of sleep pressure, and this measure was largely (mean = 67%) driven by environmental factors unique to each twin. Our results show that the amount of dissipated sleep pressure is largely under genetic control; however, the rate of sleep pressure dissipation is largely due to unique environmental factors. Objective: Hoarding disorder is a complex condition that significantly impacts individuals' lives, characterized by excessive acquiring, difficulty discarding, clutter, distress, and impairment. This study aimed to examine the extent to which genetics and environment influence difficulty discarding, excessive acquisition, and clutter through the implementation of a classical twin study. Method: This classical twin study, conducted between April and September 2021, enrolled 194 twins (97 pairs) from Isfahan, recruited through the Isfahan Twins Registry (ITR). A total of 194 twins, consisting of 100 monozygotic (MZ) and 94 dizygotic (DZ) twins, participated in this study. Participants aged 16-50 were invited electronically and completed an online consent form and questionnaire. Hoarding symptoms were assessed using the saving inventory-revised. Zygosity was determined using a self-report method based on Song et al.'s questionnaire. To estimate the heritability of hoarding symptoms, the classical univariate twin model was employed. Results: Based on the univariate analysis, the heritability estimates for difficulty discarding and excessive acquisition were found to be 0.43 and 0.52, respectively. However, the results did not provide support for the role of genetics in clutter. Instead, it was indicated that the common environment accounted for 0.54 of the variance in clutter, while the specific environment contributed 0.46 to this symptom. Conclusion: The difficulty discarding and excessive acquisition were found to be moderately heritable. On the other hand, considering the contribution of genetics and environment to clutter, the results raise doubts about the association of clutter with hoarding. Humans spend a significant portion of their waking hours day-dreaming, so gaining a deeper comprehension of the phenomenon is of substantial importance. To better characterize the day-dreaming phenomenon, in the present study we asked: (1) What is the relationship between day-dreaming and the age of young adults? (2) What is the relationship between day-dreaming and the education level of young adults? (3) Is there a difference in day-dreaming propensity between females and males? and (4) Is day-dreaming propensity genetically determined? To address these questions, we examined individual differences in day-dreaming propensity by using the large Human Connectome Project Young dataset of 1,200 young adult participants. We found that: (1) Day-dreaming propensity was negatively correlated with age of participants; (2) Day-dreaming propensity was positively correlated with education level of participants; (3) There was no difference in day-dreaming propensity between genders, but positive correlation between day-dreaming propensity and education level was much higher for females than males; and (4) Monozygotic compared to dizygotic twins were more similar to each other with regard to propensity to day-dream, suggesting that propensity to day-dream is at least to some extent genetically determined. One of the major roles of parenting twins is to ensure their parallel development, as well as their psychological differentiation and self‐individuation.The study included 91 Israeli adult participants (41 same‐sex fraternal twins; 50 same‐sex nontwin siblings), sampled through social media, who answered a series of questionnaires.Results indicated that twins reported less self‐differentiation, a poorer attachment history with their parents, and less secure attachment than nontwin siblings. Moderation analysis established that the positive link between parental differentiation and self‐differentiation was significantly stronger for twins than nontwin siblings. In contrast, sibling status had no moderating effect on the link between attachment history and style.These findings emphasize the significance of parental differentiation in twins' personal development and the importance of recognizing the unique bond between them.Implications for practitioners focusing on nurturing healthy development, strong sibling relationships, and individual well‐being are discussed. Objective: This scoping review aimed to summarize current knowledge from twin studies on migraine. Migraine heritability, genetic correlations with migraine comorbid disorders, and the use of discordant twin pairs in migraine research are described. Further, the review considers the unused potential of twin studies in migraine research and reflects on future directions. Background: Twin studies can be used to understand how heritable and environmental factors influence human traits and disorders. The classical twin design compares the resemblance of a trait in monozygotic twins to that in dizygotic twins. The classical twin design can be extended to estimate the genetic correlation between disorders, model causality, and describe differences within discordant twin pairs. Methods: Studies focusing on migraine and using a twin study design were included. The search was performed on the PubMed‐MEDLINE database using the search terms "migraine" AND "twin" OR "twins." It was done in May 2023, rerun in November 2023, and managed with the Covidence software. Results: The search identified 52 twin studies on migraine. In 24 papers, the heritability of migraine was estimated with a classical twin design. Heritability estimates ranged from 0.36 to 0.48 for studies with adults, both men and women, and unspecified migraine. Migraine heritability was predominantly estimated with twin cohorts of North European ancestry, and only two studies examined migraine subtypes. A multilevel classical twin design was used in 11 studies to examine the co‐occurrence between migraine and comorbid disorders. The differences within migraine discordant twin pairs were examined in nine studies. Conclusion: The heritability of migraine was estimated with a classical twin design in twin cohorts from seven different countries, with remarkably similar results across studies. Future studies should include migraine subtypes and twin cohorts of non‐North European ancestry to better reflect the global population. Beyond heritability estimations, the twin method is a valuable tool for understanding causality and describing differences within discordant twin pairs. Despite more than 80 years of twin studies in migraine research, the twin design has a large unused potential to advance our understanding of migraine. Purpose: We aimed to identify the genetic causes of hypospadias in children using targeted gene panel sequencing for disorders of sex development (DSD). Materials and Methods: This study included 18 twin boys with hypospadias: seven and two pairs were monozygotic and dizygotic twins, respectively, and six were discordant and three were concordant twins. Targeted gene panel sequencing for 67 known DSD genes was performed. Sequence variants were classified into five different categories, pathogenic, likely pathogenic, variants of uncertain significance, likely benign, and benign, following the American College of Medical Genetics and Genomics Standards and Guidelines. Results: The mean gestational age and birth weight were 35.3±2.0 weeks and 1.96±0.61 kg, respectively, with seven patients being small for gestational age. Hypospadias was present in 12 patients, with posterior type in 33.3% and anterior type in 66.7%. In three families with twins, both siblings had hypospadias. In addition, cryptorchidism was observed in one subject. Surgical correction of hypospadias was performed at a mean age of 22.1 months. Molecular analysis identified 12 different genetic variants, including two pathogenic mutations in the AMH (p.E389*) and SRD5A2 (p.R246Q) genes, found in subjects with hypospadias, respectively. However, only heterozygous mutations were detected. Conclusions: This study did not identify a definitive genetic component contributing to the development of hypospadias; however, the findings suggest that intrauterine growth retardation may play a significant role. Trichotillomania is a clinical psychiatric manifestation involving significant hair loss in association with recurrent hair-pulling behavior, the etiology of which is still unknown. The aim of this study was to investigate the genetic and environmental and behavioral contribution to trichotillomania (hair-pulling disorder) and to relate the findings to contemporary theories about the etiology. This was a descriptive correlational and twins study design. 672 twins (MZ=474; DZ=202) were selected from Iranian twins associations in 2018. The Massachusetts General Hospital Hair-pulling Scale(1995) and the Self-Report of Zygosity were used as research instrument. Twin modeling methods were employed to decompose the variance in the liability trichotillomania (hair-pulling disorder) into additive genetic and shared and non-shared environmental factors. The data was analyzed by SPSS, STATA and M-plus. Univariate model-fitting analyses showed that genetic factors accounted for approximately 64% of the variance in TTM. The best extracted model for TTM was the DE model. The correlation coefficient results indicated that the correlation rate was 0/61 and 0/31 in MZ and DZ group respectively. It can be concluded that TTM phenotype is the results of a polygenetic. There is no intermediate despite of the dominance effect and it results from a gene-gene interaction or a dominant gene.

The objective of this study was to assess the relative contributions of genetic and environmental factors to variation in palatal parameters in twins with completed maxillary growth. The subjects of this study comprised digital dental casts of 50 monozygotic and 35 dizygotic twin pairs. The subjects' average age was 17.95 ± 2.83 years. Zygosity determination was carried out using 15 specific DNA markers and an amel fragment of the amelogenin gene. The interdental distances were measured between selected dental landmarks at the occlusal and gingival planes. The palatal height, surface area and volume were measured between the gingival plane and the midpalate suture. High heritability estimates were observed for all transverse intra-arch measurements. The palate height (a2 = 0.8), dental arch width in the molar area (a2 = 0.86), palatal surface area (a2 = 0.61) and palate volume (a2 = 0.69) were under strong additive genetic control. Moderate genetic dominance was observed for dental arch widths at the gingival line in the canine (d2 = 0.5) and premolar regions (d2 = 0.78–0.81). Sexual dimorphism was shown, with males exhibiting a greater arch width, palate surface area and volume than females (p < 0.01). Variation in political preferences is increasingly understood to stem from both environmental influences and genetics. Research in political psychology has argued that a pathway for genetic effects on ideology is via cognitive performance, showing a sizable genetic overlap between the traits. Yet the link between actual trait cognitive performance and economic conservatism is itself highly ambiguous, with both positive, null, and negative estimates prevalent in the literature. In this study, I argue that this puzzle can only be understood from a gene–environment interaction (GxE) perspective. Drawing on traditional theories of political preference formation, I argue that genetics associated with cognitive performance should cause more left‐wing economic preferences if you grow up in relative poverty, but more right‐wing economic preferences if you grow up affluent. Utilizing variation in a polygenic index (PGI) of cognitive performance within dizygotic twin pairs, coupled with unique register data on economic conditions for the twins, their parents, and their childhood neighborhood, I show that the causal effect of the PGI on economic conservatism is zero on average, but indeed sizable and sign‐discordant by class background. Objective: This study examined whether phenotypic correlations between psychopathological dimensions and personality traits of different hierarchical levels originate from common genetic and environmental sources of variance. Method: Participants were 386 monozygotic and 204 dizygotic twins. The Psychiatric Diagnostic Screening Questionnaire (PDSQ) was applied along with the Revised NEO Personality Inventory (NEO‐PI‐R). The results of the CFA confirmed the hypothesis of the internalizing and externalizing dimensions underlying PDSQ scales. Results: The results indicated a significantly greater role of genetic compared to environmental factors in the relationship between internalizing psychopathology and personality traits. Facets of neuroticism showed positive genetic links with internalizing disorders, while negative genetic links were shown for all facets of extraversion except excitement‐seeking, competence, self‐discipline, achievement striving, actions, and trust. Lower‐order personality traits were shown to be associated with internalizing disorders more intensively than the broader domains to which they belong, both at the phenotypic and genetic levels. Conclusions: High neuroticism, together with several facets from the domain of extraversion and conscientiousness seems to represent an increased genetic susceptibility to the disorders from the internalizing spectrum. The human subcortex plays a pivotal role in cognition and is widely implicated in the pathophysiology of many psychiatric disorders. However, the heritability of functional gradients based on subcortico-cortical functional connectivity remains elusive. Here, leveraging twin functional MRI (fMRI) data from both the Human Connectome Project (n = 1023) and the Adolescent Brain Cognitive Development study (n = 936) datasets, we construct large-scale subcortical functional gradients and delineate an increased principal functional gradient pattern from unimodal sensory/motor networks to transmodal association networks. We observed that this principal functional gradient is heritable, and the strength of heritability exhibits a heterogeneous pattern along a hierarchical unimodal-transmodal axis in subcortex for both young adults and children. Furthermore, employing a machine learning framework, we show that this heterogeneous pattern of the principal functional gradient in subcortex can accurately discern the relationship between monozygotic twin pairs and dizygotic twin pairs with an accuracy of 76.2% (P < 0.001). The heritability of functional gradients is associated with the anatomical myelin proxied by MRI-derived T1-weighted/T2-weighted (T1w/T2w) ratio mapping in subcortex. This study provides new insights into the biological basis of subcortical functional hierarchy by revealing the structural and genetic properties of the subcortical functional gradients. ABSTRACT Objective Method Results Discussion Pediatric loss‐of‐control (LOC) eating is associated with high BMI and predicts binge‐eating disorder and obesity onset with age. Research on the etiology of this common comorbidity has not explored the potential for shared genetic risk. This study examined genetic and environmental influences on LOC eating and its shared influence with BMI.Participants were 499 monozygotic and 398 same‐sex dizygotic twins (age = 17.38 years ± 0.67, BMIz = 0.03 ± 1.03, 54% female) from the Colorado Center for Antisocial Drug Dependence Study. LOC eating was assessed dichotomously. Self‐reported height and weight were converted to BMIz. Univariate and bivariate twin models estimated genetic and environmental influences on LOC eating and BMIz.More girls (21%) than boys (9%, p < 0.001) reported LOC eating. The phenotypic correlation with BMIz was 0.03 in girls and 0.18 in boys. Due to the nonsignificant phenotypic correlation in girls, bivariate twin models were fit in boys only. Across all models, the best‐fitting model included genetic and unique environmental effects. Genetic factors accounted for 0.51 (95% CI: 0.23, 0.73) of the variance of LOC eating in girls and 0.54 (0.18, 0.90) in boys. The genetic correlation between LOC eating and BMIz in boys was 0.45 (0.15, 0.75).Findings indicate moderate heritability of LOC eating in adolescence, while emphasizing the role of unique environmental factors. Previous studies have shown that mothers of naturally conceived dizygotic (DZ) twins tend to be taller, older, and smoke more than mothers of naturally conceived monozygotic (MZ) twin and mothers of singletons. Here, we investigate whether mothers of naturally conceived DZ twins differ from mothers who conceived their DZ twins after medically assisted reproduction (MAR) in eight maternal traits related to fertility based on observational survey data. We include data from 33,648 mothers from the Netherlands Twin Register (NTR) and 1660 mothers of twins from the Norwegian Mother, Father and Child Cohort Study (MoBA). We contrast mothers of naturally conceived DZ twins with mothers of MAR DZ twins. Next, we further segment the MAR group into mothers who underwent hormonal induction of ovulation but not in vitro fertilization (IVF) and those who IVF twins, comparing them both to each other and against the mothers of naturally conceived DZ twins. Mothers of naturally conceived DZ twins smoke more often, differ in body composition, have a higher maternal age and have more offspring before the twins than mothers of MZ twins. Compared to MAR DZ twin mothers, mothers of naturally conceived DZ twins have fewer miscarriages, lower maternal age and increased height, more offspring and are more often smokers. BMI before the twin pregnancy is similar in both natural and MAR DZ twin mothers. Mothers who received hormonal induction of ovulation (OI) have a lower maternal age, fewer miscarriages, and a higher number of offspring before their twin pregnancy than twin mothers who received IVF and/or intracytoplasmic sperm injection (ICSI) treatments. Our study shows that twin mothers are a heterogenous group and the differences between twin mothers should be taken into account in epidemiological and genetic research that includes twins. Purpose: To investigate ocular and systemic factors associated with the retinal arterial wall‐to‐lumen ratio (WLR) and to determine the relative contribution of genetic and environmental variation to WLR in healthy adults. Methods: This cross‐sectional twin study included 78 monozygotic and 67 dizygotic same‐sex twin pairs aged 58.4 ± 9.8 (mean ± SD) years. Lumen diameter (LD) and outer diameter (OD) of a superotemporal retinal artery were measured using adaptive optics fundus photography, and the WLR was calculated. Linear mixed model regression analysis of associations with WLR comprised the descriptive variables ocular axial length (AL), intraocular pressure (IOP), height, weight, body mass index (BMI), smoking, blood pressure, high density (HDL), low density (LDL) and very low density (VLDL) lipoproteins, total cholesterol and triglycerides. The relative influence of genes and environment on WLR was calculated through polygenetic modelling. Results: Increasing age and arterial blood pressure were associated with a higher WLR, while increasing retinal artery OD and ocular AL were associated with a lower WLR. Sex, smoking status, BMI, IOP, cholesterol levels or triglycerides had no detectable impact on the WLR. Broad‐sense heritability of WLR was 21% (95% CI: 1–41%), while environmental factors accounted for the remaining 79% of the interindividual variance (95% CI: 59–99%). The prevalence of autism has been increasing at an alarming rate. Even accounting for the expansion of autism spectrum disorder diagnostic (ASD) criteria throughout the 1990's, there has been an over 300% increase in ASD prevalence since the year 2000. The often debilitating personal, familial, and societal sequelae of autism are generally believed to be lifelong. However, there have been several encouraging case reports demonstrating the reversal of autism diagnoses, with a therapeutic focus on addressing the environmental and modifiable lifestyle factors believed to be largely underlying the condition. This case report describes the reversal of autism symptoms among dizygotic, female twin toddlers and provides a review of related literature describing associations between modifiable lifestyle factors, environmental exposures, and various clinical approaches to treating autism. The twins were diagnosed with Level 3 severity ASD "requiring very substantial support" at approximately 20 months of age following concerns of limited verbal and non-verbal communication, repetitive behaviors, rigidity around transitions, and extensive gastrointestinal symptoms, among other common symptoms. A parent-driven, multidisciplinary, therapeutic intervention involving a variety of licensed clinicians focusing primarily on addressing environmental and modifiable lifestyle factors was personalized to each of the twin's symptoms, labs, and other outcome measures. Dramatic improvements were noted within several months in most domains of the twins' symptoms, which manifested in reductions of Autism Treatment Evaluation Checklist (ATEC) scores from 76 to 32 in one of the twins and from 43 to 4 in the other twin. The improvement in symptoms and ATEC scores has remained relatively stable for six months at last assessment. While prospective studies are required, this case offers further encouraging evidence of ASD reversal through a personalized, multidisciplinary approach focusing predominantly on addressing modifiable environmental and lifestyle risk factors. The association between circadian rhythms and diseases has been well established, while the association with mental health is less explored. Given the heritable nature of circadian rhythms, this study aimed to investigate the relationship between genes underlying circadian rhythms and mental health outcomes, as well as a possible gene-environment correlation for circadian rhythms. Polygenic scores (PGSs) represent the genetic predisposition to develop a certain trait or disease. In a sample from the Netherlands Twin Register (N = 14,021), PGSs were calculated for two circadian rhythm measures: morningness and relative amplitude (RA). The PGSs were used to predict mental health outcomes such as subjective happiness, quality of life, and depressive symptoms. In addition, we performed the same prediction analysis in a within-family design in a subset of dizygotic twins. The PGS for morningness significantly predicted morningness (R 2 = 1.55%) and depressive symptoms (R 2 = 0.22%). The PGS for RA significantly predicted general health (R 2 = 0.12%) and depressive symptoms (R 2 = 0.20%). Item analysis of the depressive symptoms showed that 4 out of 14 items were significantly associated with the PGSs. Overall, the results showed that people with a genetic predisposition of being a morning person or with a high RA are likely to have fewer depressive symptoms. The four associated depressive symptoms described symptoms related to decision-making, energy, and feeling worthless or inferior, rather than sleep. Based on our findings future research should include a substantial role for circadian rhythms in depression research and should further explore the gene-environment correlation in circadian rhythms. Aim: Genetic influences on cerebral activity have been described previously, but data are scarce in preterms. We aimed to investigate whether a genetic influence causes amplitude-integrated electroencephalography (aEEG) signals to differ between singletons and twin preterm newborns. Methods: This was a retrospective single-centre study conducted at Innsbruck Medical University Hospital, Austria. Preterm infants born before 32 weeks of gestation between 6 November 2010 and 6 December 2022 were eligible for the study. The aEEG was analysed for the total maturation score, its component scores and the number of sleep--wake cycles per hour. Results: We enrolled 240 preterm twin infants (57.5% male) with a mean gestational age of 30 (range: 24-32) weeks and a mean birth weight of 1324 (range: 600--2116) grams. We compared 240 singleton matched preterms. No differences were found between preterm singletons and twin preterm infants regarding the total maturation and component scores, or the number of sleep--wake cycles. aEEG showed no difference between monozygotic and dizygotic twins. Conclusion: Compared to singletons, twin infants born preterm showed no differences in aEEG signals in the first 4 weeks of life. This cohort study investigates the association of adverse childhood experiences with adult psychiatric disorders after adjusting for familial confounding. Key Points: Question: Are adverse childhood experiences (ACEs) associated with poor mental health in adulthood after adjustment for familial confounding due to shared genetic and environmental factors? Findings: In this cohort study using twin data, there were associations between ACEs and adult mental health outcomes in dizygotic and monozygotic twin pairs, while odds ratios were attenuated compared with the full cohort. Objective: To investigate whether associations between ACEs and adult mental health outcomes remain after adjusting for familial (genetic and environmental) confounding. Design, Setting, and Participants: This Swedish twin cohort study used a discordant twin pair design based on monozygotic (MZ) and dizygotic (DZ) twins. A total of 25 252 adult twins (aged 18-47 years) from the Swedish Twin Registry born between 1959 and 1998 were followed up from age 19 years until 2016, with a maximum follow-up time of 39 years. Data were analyzed from April 2022 to November 2023. Exposures: A total of 7 ACEs, including family violence, emotional abuse or neglect, physical neglect, physical abuse, sexual abuse, rape, and hate crime, were assessed with items from the Life Stressor Checklist-Revised in a web-based survey. Main Outcomes and Measures: Adult (ages >18 years) clinical diagnosis of psychiatric disorders (ie, depressive, anxiety, alcohol or drug misuse, or stress-related disorders) were obtained from the Swedish National Patient Register. Results: Of 25 252 twins included in the study (15 038 female [59.6%]; mean [SD] age at ACE assessment, 29.9 [8.7] years), 9751 individuals (38.6%) reported exposure to at least 1 ACE. A greater number of ACEs was associated with increased odds of any psychiatric disorder in the full cohort (odds ratio [OR] per additional ACE, 1.52; 95% CI, 1.48-1.57). The association remained but ORs per additional ACE were attenuated in DZ (1.29; 95% CI, 1.14-1.47) and MZ (1.20; 95% CI, 1.02-1.40) twin pairs. Individuals who were exposed to sexual abuse compared with those who were not exposed had increased odds of any clinically confirmed psychiatric disorder in all comparisons: full cohort (OR, 3.09; 95% CI, 2.68-3.56), DZ twin pairs (OR, 2.10; 95% CI, 1.33-3.32), and MZ twin pairs (1.80; 95% CI, 1.04-3.11). Conclusions and relevance: This study found that associations between ACEs and adult mental health outcomes remained after controlling for shared genetic and environmental factors, which was particularly evident after multiple ACEs or sexual abuse. Background: Sex differences exist in the prevalence and clinical manifestation of several mental disorders, suggesting that sex-specific brain phenotypes may play key roles. Previous research used machine learning models to classify sex from imaging data of the whole brain and studied the association of class probabilities with mental health, potentially overlooking regional specific characteristics. Methods: We here investigated if a regionally constrained model of brain volumetric imaging data may provide estimates that are more sensitive to mental health than whole brain-based estimates. Given its known role in emotional processing and mood disorders, we focused on the limbic system. Using two different cohorts of healthy subjects, the Human Connectome Project and the Queensland Twin IMaging, we investigated sex differences and heritability of brain volumes of limbic structures compared to non-limbic structures, and subsequently applied regionally constrained machine learning models trained solely on limbic or non-limbic features. To investigate the biological underpinnings of such models, we assessed the heritability of the obtained sex class probability estimates, and we investigated the association with major depression diagnosis in an independent clinical sample. All analyses were performed both with and without controlling for estimated total intracranial volume (eTIV). Results: Limbic structures show greater sex differences and are more heritable compared to non-limbic structures in both analyses, with and without eTIV control. Consequently, machine learning models performed well at classifying sex based solely on limbic structures and achieved performance as high as those on non-limbic or whole brain data, despite the much smaller number of features in the limbic system. The resulting class probabilities were heritable, suggesting potentially meaningful underlying biological information. Applied to an independent population with major depressive disorder, we found that depression is associated with male–female class probabilities, with largest effects obtained using the limbic model. This association was significant for models not controlling for eTIV whereas in those controlling for eTIV the associations did not pass significance correction. Conclusions: Overall, our results highlight the potential utility of regionally constrained models of brain sex to better understand the link between sex differences in the brain and mental disorders. To investigate the genetic determinants of the models, we assessed the heritability of the estimates between identical twins and fraternal twins. The estimates of all our models were heritable, suggesting a genetic component contributing to brain sex. Finally, to investigate the association with mental health, we compared brain sex estimates in healthy subjects and in a depressed population. We found an association between depression and brain sex in females for the limbic model, but not for the non-limbic model. No effect was found in males. Overall, our results highlight the potential utility of machine learning models of brain sex based on relevant structures to better understand the link between sex differences in the brain and mental disorders. This paper proposes the utilisation of twin studies as a novel and powerful methodological approach to investigate critical research questions pertaining to cancer prevention, screening, diagnosis, treatment and survivorship within primary care contexts. The inherent genetic similarity between monozygotic (MZ) (identical) twins provides a unique opportunity to disentangle genetic and environmental influences on cancer-related outcomes. MZ twins share virtually identical genetic makeup, offering a unique opportunity to discern the relative contributions of genetic and environmental factors to cancer-related outcomes. In contrast, dizygotic (DZ) twins, also known as fraternal twins, develop from two separate eggs fertilised by two different sperm and share on average 50% of their genetic material, the same level of genetic similarity found in non-twin siblings. Comparisons between MZ and DZ twins enable researchers to disentangle hereditary factors from shared environmental influences. This methodology has the potential to advance our understanding of the multifaceted interplay between genetic predisposition, lifestyle factors and healthcare interventions in the context of cancer care. We compared dyad-focused maternal speech (protoconversations and maternal speech describing the dyad) in interactions of mothers with their twin and singleton infants. Nine twins and nine singletons, coming from urban areas of Crete (Greece), were video-recorded at home in spontaneous face-to-face interactions with their mothers, from the 2nd to the 6th month at 30-day intervals. Continuous micro-analysis revealed that the following: (a) variations in proto-conversations favoured singleton compared to twin infants; (b) twin and singleton mothers addressed different, but possibly of equivalent communicative significance, verbal comments to their infants within protoconversations; (c) both twin and singleton mothers showed age-variant efforts for conversation with their young infants. Though we showed quantitative variations of maternal dyad-focused speech between mother-singleton infant and mother-twin infant dyads, both singleton and twin mothers used devices of equivalent communicative significance and age-variant attempts to respond sensitively to their infants' expressive behaviours according to the latter's age-related motives for communication. Enteroviruses (EVs) are the most common causes of viral myocarditis in neonates. Neonatal enterovirus myocarditis manifestations range from nonspecific febrile illness to congestive heart failure and cardiogenic shock with high risk of in-hospital mortality and long-term cardiac sequelae. Early recognition is essential to undertake appropriate therapy and predict outcomes. Echocardiography and echo-derived left ventricular strain measures seem promising for these purposes. The widespread introduction of molecular genetic research methods into health care practice has made it possible to diagnose rare microdeletion syndromes in patients with multiple congenital malformations. Aim of the study is to present the results of a literature search and demonstrate a clinical observation of Williams-Beuren syndrome in 10-month-old monochorionic twins with congenital malformations of the cardiovascular system in combination with kidney pathology and an additional spleen. Results. Williams-Beuren syndrome (WBS) is a rare congenital disorder characterized by specific craniofacial dysmorphisms (elphic face) and a hoarse voice in combination with cardiovascular damage, mental retardation, musculoskeletal disorders, and hypercalcemia. WBS occurs in the population with a frequency of 1:7,500-1000 infants. The presence of a specific phenotype is associated with a hemizygous microdeletion of the long arm of chromosome 7 at region 7q11.23. The size of the deletion varies from 1.5 to 1.8 Mb and results in the loss of several neighboring genes. The diagnosis is made syndromologically and confirmed by modern molecular cytogenetic methods. Pathologically significant WBS mutations include loss of the ELN gene and loss of neighboring genes such as LIMK1, RFC2, BAZ1B, GTF2I, STX1A, CLIP2, GTF2IRD, NCF. Haploinsufficiency of ELN gene is the main marker of WBS and causes insufficient synthesis of elastin protein, which leads to development of pathology of heart and blood vessels (elastin arteriopathy), disorders of connective apparatus of joints, abnormalities of vocal cords and skin. LIMK1 hemizygosity is associated with impaired visual-spatial constructive cognition. Deletion of the RFC2 gene can cause growth retardation and developmental delay. Reduced intelligence can be caused by a mutation of the GTF2I gene and hypercalcemia by a mutation of the BAZ1B gene. The phenotypic manifestations of WBS are also thought to be influenced by the reduced expression of flanking intact genes. The diagnosis, treatment, and adjustment of patients with WBS require an interdisciplinary team of specialists. The presented clinical case demonstrates multisystem pathology in 10-month-old monochorionic dizygotic twins in whom Williams-Beuren syndrome was clinically diagnosed and confirmed by FISH: ish del (7)(q11.23q11.23)(ELN-). Conclusion. To confirm the genetic component in congenital multisystem pathology, it is necessary to use modern molecular genetic diagnostic methods. Determination of genetic mutation, its size and origin is important for medical genetic counseling. This study examines the hypothesis that temperamental (dis)similarity is associated with twin relationship quality. In a longitudinal study that followed 322 monozygotic twins (who share close to 100% of their genes) and 1199 dizygotic twins (who on average, share 50% of their segregating genes) throughout childhood, mothers (N = 1547) and fathers (N = 536) reported on their twins' relationships on at least one of four measurement points when the twins were between 3 and 8–9 years of age. Mothers also reported on the twins' temperament. Negative associations were found between reports by both parents on the twins' closeness and their temperament difference throughout childhood, while positive associations were found between twins' conflict and their temperament difference in late childhood. Latent growth modeling indicated that the association between temperament differences and the twins' mother-reported closeness was evident beyond the effect of zygosity. A different pattern was found for twin conflict: the more the twins differed in their temperament (specifically negative emotionality) with age, the more the conflict between them increased. Our findings support the hypothesis that personality similarities can contribute to positive relationships from early childhood, and vice versa, beyond the effect of genetic similarity. Key Clinical Message: To raise awareness about the increasing incidence of superfetation and heterotopic pregnancy in patients with ovarian induction, their insidious symptoms of abdominal pain, anemia, and hemodynamic instability in early pregnancy, and the usefulness of transvaginal ultrasound (TVUS) and quantitative beta human chorionic gonadotrophin (b‐hCG) for diagnosis. Superfetation, occurrence of ovulation, fertilization, and implantation during an ongoing pregnancy and heterotopic pregnancy (HP) simultaneous presence of intrauterine and extrauterine pregnancies are infrequent phenomena. We report a case where both coexisted, challenges in diagnosis and management and association with the widespread use of assisted reproductive technologies (ARTs). A 32‐year‐old woman, who previously underwent ovulation induction therapy, presented with abdominal pain at 8 weeks pregnancy according to her last menstrual period. The patient had high quantitative serum beta‐human chorionic gonadotropin (b‐hCG) (30,883 mIU/mL). She was vitally stable and not anemic. Transvaginal ultrasound (TVUS) revealed two pregnancies at different gestational ages: an intrauterine pregnancy at 5 weeks and 3 days, and a right intact tubal ectopic pregnancy at 10 weeks and 5 days. Superfetation resulting in HP was then diagnosed. Subsequently, the patient underwent right laparoscopic salpingectomy. The intrauterine pregnancy progressed normally, resulting in delivery of a healthy full‐term neonate via Cesarean section at 38 weeks. Superfetation is typically rare from suppression of follicular development and ovulation during pregnancy. Various theories have been proposed to explain its etiology, including polyovulation, delayed blastocyst implantation, and abnormal estrogen and b‐hCG surges. In superfetation, an embryo resulting from a previous conception coexists with another embryo, either intrauterine, resulting in diamniotic dizygotic twins with significantly different gestational ages, or extrauterine resulting in HP. Despite being particularly challenging to diagnose because its presenting symptoms can overlap with those of other more common clinical conditions in early pregnancy, HP is increasingly seen with ARTs. In addition, the treatment of HP is versatile, ranging from expectant management to laparoscopic surgery. High level of suspicion for HP and superfetation is crucial in patients who, after ART, present with abdominal pain, hemodynamic instability, or anemia. Additionally, patients planning to undergo subsequent ART cycles should be thoroughly screened with b‐hCG and TVUS to exclude an ongoing intrauterine or extrauterine pregnancy. The functional connectome supports information transmission through the brain at various spatial scales, from exchange between broad cortical regions to finer-scale, vertex-wise connections that underlie specific information processing mechanisms. In adults, while both the coarse- and fine-scale functional connectomes predict cognition, the fine scale can predict up to twice the variance as the coarse-scale functional connectome. Yet, past brain-wide association studies, particularly using large developmental samples, focus on the coarse connectome to understand the neural underpinnings of individual differences in cognition. Using a large cohort of children (age 9-10 years; n = 1,115 individuals; both sexes; 50% female, including 170 monozygotic and 219 dizygotic twin pairs and 337 unrelated individuals), we examine the reliability, heritability, and behavioral relevance of resting-state functional connectivity computed at different spatial scales. We use connectivity hyperalignment to improve access to reliable fine-scale (vertex-wise) connectivity information and compare the fine-scale connectome with the traditional parcel-wise (coarse scale) functional connectomes. Though individual differences in the fine-scale connectome are more reliable than those in the coarse-scale, they are less heritable. Further, the alignment and scale of connectomes influence their ability to predict behavior, whereby some cognitive traits are equally well predicted by both connectome scales, but other, less heritable cognitive traits are better predicted by the fine-scale connectome. Together, our findings suggest there are dissociable individual differences in information processing represented at different scales of the functional connectome which, in turn, have distinct implications for heritability and cognition. MZT were also associated with higher odds of preterm birth (prior to 37 weeks) (adjusted OR: 2.31; 95% CI, 1.48–3.67), low birth weight (adjusted OR: 1.92; 95% CI, 1.27–2.93), and small for gestational age (adjusted OR: 2.18; 95% CI, 1.21–3.69) in the fully adjusted analyses. The effect of MZT on neonatal death was partially mediated by preterm birth and low birth weight (P < 0.05). Conclusions: This study indicates that MZT conceived by FET are related to an increased risk of neonatal death, emphasizing a potential need for comprehensive antenatal surveillance in these at-risk pregnancies. Familial twinning and fertility traits were investigated in Nigerian mothers of dizygotic (DZ) twins (MoDZT; N = 972) and controls (N = 525) who responded to our person-to-person interview, which included questions on pregnancy history and family history of DZ twinning. Controls were defined as women who are not twins themselves and do not have twins in their first-degree relatives. Over 95% of the participants were Yoruba. We found that Nigerian MoDZT had an average of 4.0 (±2.6) pairs of twins among their relatives, and of these, the prevalence of DZ twins was significantly higher than that of monozygotic (MZ) twins (45.9% vs. 25.8%). Controls had an average of 0.5 (±0.4) pairs, and over 95% of the controls had no twins in their relatives. These results suggest genetic influences on DZ twinning in Nigerians. MoDZT were significantly younger in their mean age at first child, and had higher parity than controls, suggesting increased fertility in MoDZT. As compared to mothers with a single set of twins, mothers (N = 130) with multiple sets had significantly more twins among their relatives (5.4 pairs vs. 3.7 pairs) and had their first twins at a younger age (28.4 vs. 30.7 years), indicating that mothers with multiple sets of twins might have higher genetic propensity for twinning associated with earlier age at twin pregnancy. Our findings argue for genomewide association studies for DZ twinning in Nigerians, and may help to develop intervention strategies to overcome infertility/subfertility problems. Although the focus of the work is still within the critical framework of modern and contemporary literature, as the subconscious mind of text generation, drama theory seems to require more pre-emptive attention and outlook. Snow is a vital component of the earth system, yet no snow-focused satellite remote sensing platform currently exists. In this study, we investigate how synthetic observations of snow water equivalent (SWE) representative of a synthetic aperture radar remote sensing platform could improve spatiotemporal estimates of snowpack. We use a fraternal twin observing system simulation experiment, specifically investigating how much snow simulated using widely used models and forcing data could be improved by assimilating synthetic observations of SWE. We focus this study across a 24∘×37∘ domain in the western USA and Canada, simulating snow at 250 m resolution and hourly time steps in water year 2019. We perform two data assimilation experiments, including (1) a simulation excluding synthetic observations in forests where canopies obstruct remote sensing retrievals and (2) a simulation inferring snow distribution in forested grid cells using synthetic observations from nearby canopy-free grid cells. Results found that, relative to a nature run, or assumed true simulation of snow evolution, assimilating synthetic SWE observations improved average SWE biases at maximum snowpack timing in shrub, grass, crop, bare-ground, and wetland land cover types from 14 %, to within 1 %. However, forested grid cells contained a disproportionate amount of SWE volume. In forests, SWE mean absolute errors at the time of maximum snow volume were 111 mm and average SWE biases were on the order of 150 %. Here the data assimilation approach that estimated forest SWE using observations from the nearest canopy-free grid cells substantially improved these SWE biases (18 %) and the SWE mean absolute error (27 mm). Simulations employing data assimilation also improved estimates of the temporal evolution of both SWE and runoff, even in spring snowmelt periods when melting snow and high snow liquid water content prevented synthetic SWE retrievals. In fact, in the Upper Colorado River region, melt-season SWE biases were improved from 63 % to within 1 %, and the Nash–Sutcliffe efficiency of runoff improved from -2.59 to 0.22. These results demonstrate the value of data assimilation and a snow-focused globally relevant remote sensing platform for improving the characterization of SWE and associated water availability. Background: Twin babies provide a good study model since they have the same GA and are exposed to the same prenatal risk factors. Thus, it helps us to analyze the role of systemic complications in development and progression of ROP in two premature babies exposed to same risk factors. hence, the study was undertaken. The objectives of the study were to determine, profile of ROP in twin babies. Materials and Methods: A prospective interventional clinical study was done from, November 2016 to May 2018 in 100 preterm babies, less than or equal to 32 weeks of gestational age (GA) & less than or equal to 1500 gram of birth weight (BW) at Vani Vilas Hospital attached to Bangalore Medical College and Research Institute, Bangalore. ROP screening was performed by dilated fundoscopy using indirect ophthalmoscope with 20D lens and all babies were followed up till complete vascularisation of the retina. The babies who developed any stage of ROP were taken as cases and the babies who did not have ROP were taken as controls. Results: 100babies/50 pairs (76.92%) satisfied the inclusion criteria and were enrolled in the study. Out of 50 pairs of twins, 10 pairs developed ROP. In our study we found that most commonly identical twins (p 0.001, significant) affected than non-identical twins, smaller baby affected more than larger baby (p 0.007, significant), Male babies affected more than female babies (p 0.07), ROP development between 2nd born and born is 6:4(p 0.37). With regarding to laterality both eyes affected in all 10 babies and stage and zone of affection remains same in both the eyes. In our study, 6 babies (60%) developed zone-2 stage-1 ROP, 3 babies (30%) developed Zone-2 Stage-2 ROP with plus disease & one baby (10%) developed Zone-2 APROP with plus disease. Conclusion: Thorough fundus examination with indirect ophthalmoscopy should be done in all preterm babies as per the guidelines and screening should be intensified in the presence of factors like RDS, oxygen administration. Due to recent advances in the development of land data assimilation systems (LDAS) and the availability of high-quality, satellite-based surface soil moisture (SSM) retrieval products, we now have unambiguous evidence that the assimilation of SSM retrievals, or their proxy, can improve the precision (i.e., correlation versus truth) of surface state estimates provided by a land surface model (LSM). However, this clarity does not yet extend to the estimation of LSM surface water fluxes that are key to hydrologic and numerical weather forecasting applications. Here, we hypothesize that a key obstacle to extrapolating realized improvements in water state precision into comparable improvements in water flux accuracy (i.e., mean absolute error) is the presence of water state–water flux coupling strength biases existing in LSMs. To test this hypothesis, we conduct a series of synthetic fraternal twin data assimilation experiments where realistic levels of state–flux coupling strength bias—involving both evapotranspiration and runoff—are systematically introduced into an assimilation LSM. Results show that the accuracy of the resulting water flux analysis is sharply reduced by the presence of such bias, even in cases where the precision of soil moisture state estimates (e.g., SSM) is improved. The rescaling of SSM observations prior to their assimilation (i.e., the most common approach for addressing systematic differences between LSMs and assimilated observations) is not always a robust strategy for addressing these errors and can, in certain circumstances, degrade water flux accuracy. Overall, results underscore the critical need to assess, and correct for, LSM water state–water flux coupling strength biases during the operation of an LDAS. Significance Statement: Land data assimilation is the process by which land surface model estimates of water states (e.g., soil moisture) and water fluxes (e.g., runoff and evapotranspiration) are improved via the incorporation of observations. Over the past decade, substantial improvements have been made in the precision of land surface model states via the assimilation of satellite-based soil moisture information. However, to date, these improvements have not yet been extended into water flux estimates like runoff and evapotranspiration. This is a critical shortcoming since advances in important weather and hydrologic forecasting applications are dependent on the improved estimation of such fluxes. We demonstrate that this shortcoming is linked to the inability of existing land surface models to accurately describe the impact of variations in water states on water fluxes and propose strategies for overcoming this issue in future land data assimilation systems. Nutrition and diet are key modifiable risk factors for the rising burden of non-communicable diseases like cardio-vascular diseases and diabetes in low- and middle- income countries (LMICs). The nutritional transition in dietary behaviours in LMICs has most likely contributed to this problem. Although traditionally assumed to be environmental, dietary choices are also genetically influenced. Twin study designs can be used to investigate the relative influence of genes and environment on nutrition intake, eating behaviours and associated psychological health. The overall aim of this project is to: provide proof-of-concept for the feasibility of using dietary (biomarker) data within the Children-of-Twin design in nutrition studies, develop laboratory skills and statistical genetic skills and establish a Sri Lankan-specific food composition database. Currently, a pilot study is being conducted with 304 individuals (38 Monozygotic twin pairs, 38 Dizygotic twin pairs and their male or female adult offspring). Questionnaire data on nutritional intake, eating behaviours, psychological well-being, physical health, and bio-specimens are being collected. A Sri Lankan-specific food composition database was developed, training sessions on macro and micro element analysis in biological samples and statistical genetics skills development were conducted and Community Engagement and Involvement programs were carried out in two districts of Sri Lanka. Objectives The educational gradient in late-life health is well established. Despite this, there are still ambiguities concerning the role of underlying confounding by genetic influences and gene-environment (GE) interplay. Here, we investigate the role of educational factors (attained and genetic propensities) on health and mortality in late life using genetic propensity for educational attainment (as measured by a genome-wide polygenic score, PGSEdu) and attained education. Methods By utilizing genetically informative twin data from the Swedish Twin Registry (n = 14,570), we investigated influences of the educational measures, familial confounding as well as the possible presence of passive GE correlation on both objective and subjective indicators of late-life health, that is, the Frailty Index, Multimorbidity, Self-rated health, cardiovascular disease, and all-cause mortality. Results Using between-within models to adjust for shared familial factors, we found that the relationship between educational level and health and mortality later in life persisted despite controlling for familial confounding. PGSEdu and attained education both uniquely predicted late-life health and mortality, even when mutually adjusted. Between-within models of PGSEdu on the health outcomes in dizygotic twins showed weak evidence for passive GE correlation (prGE) in the education-health relationship. Discussion Both genetic propensity to education and attained education are (partly) independently associated with health in late life. Vocabulary acquisition is the fundamental aspect of intelligence and is the process of learning new words. The classical twin study provided robust information regarding the contribution of genetics and environmental propensities on the vocabulary acquisition of twins. The twin study has been planned with the objective to assess the impact of genetics and environment on vocabulary acquisition ability. This investigation was performed in the Bhiwani and Hisar districts of Haryana. A sample of 150 pairs of twins in the age groups of 3-6 years was attained from 55 villages of two districts. Results of heritability estimates showed that the contribution of genetics for vocabulary acquisition ability ranged from 51-60% for 3-6 years. The conclusion of heritability estimates indicated that the impact of genetic factors on vocabulary acquisition ability of twins was more than the environmental factors. [ABSTRACT FROM AUTHOR]
