Abstract Background GRAS, an important family of transcription factors, have played pivotal roles in regulating numerous intriguing biological processes in plant development and abiotic stress responses. Since the sequencing of the sorghum genome, a plethora of genetic studies were mainly focused on the genomic information. The indepth identification or genome-wide analysis of GRAS family genes, especially in Sorghum bicolor, have rarely been studied. Results A total of 81 SbGRAS genes were identified based on the S. bicolor genome. They were named SbGRAS01 to SbGRAS81 and grouped into 13 subfamilies (LISCL, DLT, OS19, SCL4/7, PAT1, SHR, SCL3, HAM-1, SCR, DELLA, HAM-2, LAS and OS4). SbGRAS genes are not evenly distributed on the chromosomes. According to the results of the gene and motif composition, SbGRAS members located in the same group contained analogous intron/exon and motif organizations. We found that the contribution of tandem repeats to the increase in sorghum GRAS members was slightly greater than that of fragment repeats. By quantitative (q) RT-PCR, the expression of 13 SbGRAS members in different plant tissues and in plants exposed to six abiotic stresses at the seedling stage were quantified. We further investigated the relationship between DELLA genes, GAs and grain development in S. bicolor. The paclobutrazol treatment significantly increased grain weight, and affected the expression levels of all DELLA subfamily genes. SbGRAS03 is the most sensitive to paclobutrazol treatment, but also has a high response to abiotic stresses. Conclusions Collectively, SbGRAs play an important role in plant development and response to abiotic stress. This systematic analysis lays the foundation for further study of the functional characteristics of GRAS genes of S. bicolor.

We investigated the change in seed shape and weight as well as the accumulation and distribution of anthocyanins, melanin, and flavonoids during tartary buckwheat seed development. Pericarp dry weight increased slowly while the increase in flour dry weight was much greater. Increases in the dry weight of the embryo and endosperm were associated with change in seed shape. The highest anthocyanin content was observed in stage two while the highest melanin content occurred in stage five. There was a considerable decrease in total flavonoids, rutin, quercetin and kaempferol in mature seeds relative to seeds that were still developing. Background: Tartary buckwheat (Fagopyrum tataricum), an excellent edible and medicinal crop, has been widely used as a daily diet and traditional medicine for a long time. The major functional components of Fagopyrum tataricum have been demonstrated to be flavonoids (i.e. rutin and quercetin), which had notable andioxidant, antidiabetic, hypocholesterolemic and antitumor activities. Hairy root culture is a convenient and efficient plant tissue culture system for large scale production of bioactive metabolites. Objective: To enhance the functional flavonoids production in hairy root culture of F. tataricum. Materials and Methods: The elicitation treatment in combination with medium renewal strategy was applied for efficient promoting flavonoids production in F. tataricum hairy root cultures. Results: The exogenous yeast polysaccharide (YPS) elicitor notably stimulated the functional metabolites production in F. tataricum hairy root cultures, and the stimulation effect was concentration-dependent. Combination with the YPS elicitation (200 mg/L) and medium renewal process, the maximal flavonoids yield was enhanced to 47.13 mg/L, about 3.2-fold in comparison with the control culture of 14.88 mg/L. Moreover, this research also revealed the accumulation of these bioactive metabolites resulted from the stimulation of the phenylpropanoid pathway by YPS treatment. Background: Buckwheat (Fagopyrum spp., Polygonaceae) is a widely planted food crop. Flavonoids, including quercetin, rutin, and kaempferol, are the main bioactive components in tartary buckwheat (Fagopyrum tataricum (L.) Gaertn). From the nutriological and pharmacological perspectives, flavonoids have great value in controlling blood glucose and blood pressure levels, and they also have antioxidant properties. Objective: To optimize the conditions for extraction of quercetin, rutin, and kaempferol from F. tataricum. Materials and Methods: A combination of ultrasound-assisted extraction (UAE) and response surface methodology (RSM) was used for flavonoid extraction and yield assessment. The RSM was based on a three-level, three-variable Box-Behnken design. Results: Flavonoids were optimally extracted from F. tataricum by using 72% methanol, at 60°C, for 21 minutes. Under these conditions, the obtained extraction yield of the total flavonoids was 3.94%. Increase of planting density has been widely used to increase grain yield in crops. However, it may lead to higher risk of lodging hence causing significant yield loss of the crop. To investigate the effects of planting density on lodging-related morphology, lodging rate (LR), and yield of tartary buckwheat, an experiment was carried out with a split-plot randomized block design at the experimental farm of Chengdu University (Sichuan, China) in the 2012 and 2013 growing seasons. Results showed that plant density significantly affected characteristics of stem and root. In each season, with the increasing of planting density, light transmittance, main root length, number of first lateral root, root volume, internode number, and first internode diameter decreased, the plant height, first internode length, abortion rate and LR increased. Increasing density caused decreased grains number per plant, the dry matter weight and yield displayed an acceleration first and then deceleration. The correlation analysis indicated that the internode number, first internode diameter, number of first lateral roots, and root volume were significantly negatively correlated with LR, but positively correlated with stem breaking strength and lodging resistance index. The LR was significantly positively correlated with plant height and first internode length. In both years, the D2 (9 × 105 plant ha−1) and D3 (12 × 105 plant ha−1) yielded more grains than in other treatments, and the effects of density on two cultivars showed the same trend. The results suggested that planting density could alter lodging-related traits, lodging resistance, and yield of tartary buckwheat.
