ABSTRACT Porcine deltacoronavirus (PDCoV) is a newly emerged enteric pathogen that causes diarrhea, dehydration, vomiting, and in suckling piglets, a high rate of death. Here, we used endocytosis inhibitors, dominant-negative mutants, and small interfering RNA (siRNA) interference to investigate PDCoV entry into swine testicle (ST) cells. Results showed that chlorpromazine, dynasore, and methyl-β-cyclodextrin (MβCD) inhibited PDCoV internalization; immunofluorescence assay (IFA) of dominant-negative (DN) mutants and siRNA interference further demonstrated these results, indicating that PDCoV utilizes clathrin-mediated endocytosis and requires dynamin and cholesterol to enter ST cells. The role of endosomes in PDCoV infection was also investigated and we found that PDCoV does not require Rab5, Rab7, or Rab11 for productive infection, indicating that the transport of PDCoV in ST cells does not require the participation of the endosomal system. Our findings will provide additional understanding of the entry mechanisms of PDCoV and possible antiviral targets. IMPORTANCE Porcine deltacoronavirus (PDCoV) is a newly emerged enteric virus threatening pig industries worldwide. Our previous work showed that PDCoV enters porcine kidney (PK-15) cells through a caveolae-dependent pathway, but the entry mechanism for PDCoV into swine testicle (ST) cells remains unclear. Mechanisms of virus entry can be different with different virus isolates and cell types. Here, we determined that PDCoV enters ST cells via clathrin-mediated endocytosis. Additionally, we found that PDCoV entry does not require Rab5, Rab7, or Rab11. These findings provide additional understanding of the entry mechanisms of PDCoV and possible antiviral targets.

Enterovirus G belongs to the family Picornaviridae and are associated with a variety of animal diseases. We isolated and characterized a novel EV-G2 strain, CHN-SCMY2021, the first genotype 2 strain isolated in China. CHN-SCMY2021 is about 25 nm diameter with morphology typical of picornaviruses and its genome is 7341 nucleotides. Sequence alignment and phylogenetic analysis based on VP1 indicated that this isolate is a genotype 2 strain. The whole genome similarity between CHN-SCMY2021 and other EV-G genotype 2 strains is 78.3–86.4%, the greatest similarity is to EVG/Porcine/JPN/Iba26–506/2014/G2 (LC316792.1). Recombination analysis indicated that CHN-SCMY2021 resulted from recombination between 714,171/CaoLanh_VN (KT265894.2) and LP 54 (AF363455.1). Except for ST cells, CHN-SCMY2021 has a broad spectrum of cellular adaptations, which are susceptible to BHK-21, PK-15, IPEC-J2, LLC-PK and Vero cells. In piglets, CHN-SCMY2021 causes mild diarrhea and thinning of the intestinal wall. The virus was mainly distributed to intestinal tissue but was also found in heart, liver, spleen, lung, kidney, brain, and spinal cord. CHN-SCMY2021 is the first systematically characterized EV-G genotype 2 strain from China, our results enrich the information on the epidemiology, molecular evolution and pathogenicity associated with EV-G.

The craft beer was continuously sterilized by the non-thermal plasma reactor, and its sterilization and quality were evaluated. The total count of aerobic plate, yeast and Escherichia coli were used as microbial indexes, the effects of plasma sterilization and pasteurization on the bactericidal effect of craft beer were studied. Meanwhile, the beer color, foam hold and pH were evaluated to obtain the optimum sterilization condition with the response surface. The results showed that, compared with pasteurization, the sterilization rate after plasma treatment was almost the same, but the chroma, brightness and bubble retention were better, which was closer to the original beer. The total number of colonies and Escherichia coli were almost completely killed, and the number of yeast was reduced by 97.5% with the 40 kV plasma treatment voltage, 10 s and 60 mL/min gas flow rate. According to the analysis of Design Expert software, the sterilization effect and brightness were the best when the voltage, plasma treatment times and gas flow were 40 kV, 10 s and 60 mL/min, respectively. The predicted values were consistent with the measured values. The influence of each factors were voltage > time > gas flow rate.

In order to maintain the stability of the transmitter to ensure the quality of the output signal, we propose a novel technique of automatic bias control based on the pilot signal that can be used in high-order modulation formats. The proposed scheme requires loading a pair of orthogonal pilot signals to the modulator and tapping the frequency-shifted local oscillation light with the signal light output from the modulator to achieve coherent detection. The control of the bias voltage is achieved by means of correlation operation and monitoring the value of the correlation coefficient. This scheme combines coherent detection and correlation detection techniques, which can effectively improve the sensitivity and control accuracy of the system. The feasibility of this scheme in high-order modulation systems and the improvement of sensitivity are also fully verified by simulation.

Residues of fipronil and its metabolites in paddy fish soil were detected using high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS). Samples were extracted with oscillation and acetonitrile in constant temperature, and subsequently purified by solid phase extraction using ENVI 18 column and 10 mg GCB. The target compounds were separated on an Agilent ZORBAX Extend C18 column (2.1 mm× 100 mm×1.8 μm), and determined using electrospray ionization (ESI) source at negative mode with multiple reation monitoring (MRM). The analytes were quantified with external standard using the matrix matched standard calibration curve method. The results indicated that the average spiked recoveries of fipronil, fipronil desulfinyl, fipronilsulfone and fipronilsulfide in paddyfish soil were 83.5%~110.3% with the relative standard deviations (RSDs) of 1.9%~11.5%. Good linearities were obtained in the range of 0.5~100 μg/L, and the correlation coefficients were greater than 0.998, the limits of detection (LODs) were 1.0 μg/kg, and the limits of quantification (LOQs) were 3.0 μg/kg. This method is simple, accurate and reliable, which can meet the detection requirements of fipronil and its metabolites in paddy fish soil.

The microstructure of as-cast Mg-6Zn-xCe alloy was investigated by optical microscope (OM), X-ray diffraction (XRD), scanning election microscopy (SEM), energy dispersive spectrometer (EDS), transmission electron microscopy (TEM)and the changing rule of thermal conductivity with the content of Ce was investigated using the flash method. The results show that Mg-6Zn alloy is mainly composed of α-Mg and Mg7Zn3 phases. The ternary phase Ce5 (Mg,Zn)41 which is mainly distributed between grain boundaries and dendrites appears in the alloy after the addition of the rare earth element Ce, and the appearance of ternary phase has an inhibitory effect on Mg7Zn3 phase. The content of eutectic structure is increased by the addition of Ce, and the distribution continuity is improved with the increase of Ce content. The thermal conductivity of the alloy is decreased with the increase of Ce content, which may be caused by the increase of volume fraction, more continuous distribution of eutectic structure in the alloy, the enhanced electron scattering, the extended electron conduction paths and the increased thermal resistance, ultimately the thermal conductivity of the alloy is reduced.

With precursor modification by regulating dianhydride hydrolysis degree, the viscosity of polyimide precursor solution, derived from 3,3',4,4'-biphenyl tetracarboxylic dianhydride (BPDA) and para-phenylenediamine (PDA), was facilely manipulated. The chemical structure of the precursor was characterized by infrared (IR) spectrometry and proton nuclear magnetic resonance (1H-NMR) spectrometry. The effects of dianhydride hydrolysis on the viscosity, imidization process and properties were investigated. The results show that the precursor can be formed from hydrolytic BPDA and PDA, with amide acid and carboxylic ammonium salt groups in the molecular structure. The viscosity of the precursor can be reduced by the introduction of carboxylic ammonium salt group, achieving the effective regulation of viscosity in the 10-105cP range. Importantly, the existence of carboxylic ammonium salt shows no influence on the complete imidization of the precursor, which guarantees the maintenance of mechanical property. Interestingly, the coefficient of thermal expansion (CTE) of this BPDA-PDA polyimide films can be reduced by this viscosity manipulation method.

BACKGROUND: Populations of the plant pathogenic fungus Verticillium dahliae display a complex and rich genetic diversity, yet the existence of sexual reproduction in the fungus remains contested. As pivotal genes, MAT genes play a crucial role in regulating cell differentiation, morphological development, and mating of compatible cells. However, the functions of the two mating type genes in V. dahliae, VdMAT1-1-1, and VdMAT1-2-1, remain poorly understood. RESULTS: In this study, we confirmed that the MAT loci in V. dahliae are highly conserved, including both VdMAT1-1-1 and VdMAT1-2-1 which share high collinearity. The conserved core transcription factor encoded by the two MAT loci may facilitate the regulation of pheromone precursor and pheromone receptor genes by directly binding to their promoter regions. Additionally, peptide activity assays demonstrated that the signal peptide of the pheromone VdPpg1 possessed secretory activity, while VdPpg2, lacked a predicted signal peptide. Chemotactic growth assays revealed that V. dahliae senses and grows towards the pheromones FO-a and FO-α of Fusarium oxysporum, as well as towards VdPpg2 of V. dahliae, but not in response to VdPpg1. The findings herein also revealed that VdMAT1-1-1 and VdMAT1-2-1 regulate vegetative growth, carbon source utilization, and resistance to stressors in V. dahliae, while negatively regulating virulence. CONCLUSIONS: These findings underscore the potential roles of VdMAT1-1-1 and VdMAT1-2-1 in sexual reproduction and confirm their involvement in various asexual processes of V. dahliae, offering novel insights into the functions of mating type genes in this species.

PDCoV is an emerging enteropathogenic coronavirus that mainly causes acute diarrhea in piglets, seriously affecting pig breeding industries worldwide. To date, the molecular mechanisms of PDCoV-induced immune and inflammatory responses or host responses in LLC-PK cells in vitro are not well understood. HSP90 plays important roles in various viral infections. In this study, HSP90AB1 knockout cells (HSP90AB1KO) were constructed and a comparative transcriptomic analysis between PDCoV-infected HSP90AB1WT and HSP90AB1KO cells was conducted using RNA sequencing to explore the effect of HSP90AB1 on PDCoV infection. A total of 1295 and 3746 differentially expressed genes (DEGs) were identified in PDCoV-infected HSP90AB1WT and HSP90AB1KO cells, respectively. Moreover, most of the significantly enriched pathways were related to immune and inflammatory response-associated pathways upon PDCoV infection. The DEGs enriched in NF-κB pathways were specifically detected in HSP90AB1WT cells, and NF-κB inhibitors JSH-23, SC75741 and QNZ treatment reduced PDCoV infection. Further research revealed most cytokines associated with immune and inflammatory responses were upregulated during PDCoV infection. Knockout of HSP90AB1 altered the upregulated levels of some cytokines. Taken together, our findings provide new insights into the host response to PDCoV infection from the transcriptome perspective, which will contribute to illustrating the molecular basis of the interaction between PDCoV and HSP90AB1.

(1) Background: Porcine deltacoronavirus (PDCoV) is a newly emerged enteric virus affecting pig breeding industries worldwide, and its pathogenic mechanism remains unclear. (2) Methods: In this study, we preliminarily identified the endocytic pathway of PDCoV in PK-15 cells, using six chemical inhibitors (targeting clathrin-mediated endocytosis, caveolae-mediated endocytosis, macropinocytosis pathway and endosomal acidification), overexpression of dominant-negative (DN) mutants to treat PK-15 cells and proteins knockdown. (3) Results: The results revealed that PDCoV entry was not affected after treatment with chlorpromazine (CPZ), 5-(N-ethyl-N-isopropyl) amiloride (EIPA)or ammonium chloride (NH4Cl), indicating that the entry of PDCoV into PK-15 cells were clathrin-, micropinocytosis-, PH-independent endocytosis. Conversely, PDCoV infection was sensitive to nystatin, dynasore and methyl-β-cyclodextrin (MβCD) with reduced PDCoV internalization, indicating that entry of PDCoV into PK-15 cells was caveolae-mediated endocytosis that required dynamin and cholesterol; indirect immunofluorescence and shRNA interference further validated these results. (4) Conclusions: In conclusion, PDCoV entry into PK-15 cells depends on caveolae-mediated endocytosis, which requires cholesterol and dynamin. Our finding is the first initial identification of the endocytic pathway of PDCoV in PK-15 cells, providing a theoretical basis for an in-depth understanding of the pathogenic mechanism of PDCoV and the design of new antiviral targets.

How to scientifically reflect the soil environmental quality of a large area through limited soil point monitoring is a major direction of current research. The Huaihe River Basin in Anhui Province was selected as the study area and soil environmental quality monitoring for five major soil pollution types during five consecutive years was conducted. The monitoring project includes Cd, Hg, As, Pb, Cr, Cu, Zn and Ni in the soil. The single pollution index and Nemerow pollution index methods were used to evaluate 426 monitoring sites. The Kriging interpolation method was used to plot the distribution of heavy metal elements in regional soil. In general, the soil environmental quality of the Anhui section of the Huaihe River Basin is relatively safe according to the evaluation results. 96.48% of the surveyed sites belong to clean and relatively clean levels, but some sites were severe. According to the distribution of heavy metal elements in the evaluation area, the overall content is low and the local area is slightly higher. The method of special type monitoring was applied and the characteristics of regional distribution through soil point monitoring results are discussed. The proposed method has great application potential. This method is suitable for preliminary investigation and research of large-scale environmental quality of soil.

Isobaric ions interfere with two isotopes of Ag during determination by Inductively Coupled Plasma-Mass Spectrometry (ICP-MS), i.e., 90Zr16O1H and 91Zr16O on 107Ag, and 92Zr16O1H and 93Nb16O on 109Ag. Although an interference correction coefficient method was used for the calibration of the analytical results, the analytical errors remain serious. In the proposed method, samples were closed decomposed by nitric acid, hydrofluoric acid, and perchloric acid. Ammonia was used to precipitate Zr and the purified Ag was determined by ICP-MS using 103Rh as the internal standard. The interference caused by the residual Zr was calibrated by the interference coefficient method. The method has a detection limit (3σ) of 4.1 ng/g and relative standard deviation (n=12) ranging from 2.5% to 7.8%. Determination of Ag in 53 primary certified materials of geochemical exploration indicates no loss of Ag. Using ammonium as the reagent, separation of Zr and Ag was successfully achieved due to the formation of the stable Ag-ammonium complex and Zr-hydroxide precipitate. The separation of Ag from Zr has reduced the interferences of Zr on Ag. Compared with the high-pressure sealed decomposition method and the P507 extraction resin separation method, the closed sample decomposition method is much simpler with less acid used and a lower blank.

Verticillium dahliae is a soilborne fungal pathogen that causes disease on many economically important crops. Based on the resistance or susceptibility of differential cultivars in tomato, isolates of V. dahliae are divided into three races. Avirulence (avr) genes within the genomes of the three races have also been identified. However, the functional role of the avr gene in race 3 isolates of V. dahliae has not been characterized. In this study, bioinformatics analysis showed that VdR3e, a cysteine-rich secreted protein encoded by the gene characterizing race 3 in V. dahliae, was likely obtained by horizontal gene transfer from the fungal genus Bipolaris. We demonstrate that VdR3e causes cell death by triggering multiple defense responses. In addition, VdR3e localized at the periphery of the plant cell and triggered immunity depending on its subcellular localization and the cell membrane receptor BAK1. Furthermore, VdR3e is a virulence factor and shows differential pathogenicity in race 3-resistant and -susceptible hosts. These results suggest that VdR3e is a virulence factor that can also interact with BAK1 as a pathogen-associated molecular pattern (PAMP) to trigger immune responses. IMPORTANCE Based on the gene-for-gene model, research on the function of avirulence genes and resistance genes has had an unparalleled impact on breeding for resistance in most crops against individual pathogens. The soilborne fungal pathogen, Verticillium dahliae, is a major pathogen on many economically important crops. Currently, avr genes of the three races in V. dahliae have been identified, but the function of avr gene representing race 3 has not been described. We investigated the characteristics of VdR3e-mediated immunity and demonstrated that VdR3e acts as a PAMP to activate a variety of plant defense responses and induce plant cell death. We also demonstrated that the role of VdR3e in pathogenicity was host dependent. This is the first study to describe the immune and virulence functions of the avr gene from race 3 in V. dahliae, and we provide support for the identification of genes mediating resistance against race 3.

BACKGROUND: The extracellular space between the cell wall and plasma membrane is a battlefield in plant-pathogen interactions. Within this space, the pathogen employs its secretome to attack the host in a variety of ways, including immunity manipulation. However, the role of the plant secretome is rarely studied for its role in disease resistance. RESULTS: Here, we examined the secretome of Verticillium wilt-resistant Gossypium hirsutum cultivar Zhongzhimian No.2 (ZZM2, encoding 95,327 predicted coding sequences) to determine its role in disease resistance against the wilt causal agent, Verticillium dahliae. Bioinformatics-driven analyses showed that the ZZM2 genome encodes 2085 secreted proteins and that these display disequilibrium in their distribution among the chromosomes. The cotton secretome displayed differences in the abundance of certain amino acid residues as compared to the remaining encoded proteins due to the localization of these putative proteins in the extracellular space. The secretome analysis revealed conservation for an allotetraploid genome, which nevertheless exhibited variation among orthologs and comparable unique genes between the two sub-genomes. Secretome annotation strongly suggested its involvement in extracellular stress responses (hydrolase activity, oxidoreductase activity, and extracellular region, etc.), thus contributing to resistance against the V. dahliae infection. Furthermore, the defense response genes (immunity marker NbHIN1, salicylic acid marker NbPR1, and jasmonic acid marker NbLOX4) were activated to varying degrees when Nicotina benthamiana leaves were agro-infiltrated with 28 randomly selected members, suggesting that the secretome plays an important role in the immunity response. Finally, gene silencing assays of 11 members from 13 selected candidates in ZZM2 displayed higher susceptibility to V. dahliae, suggesting that the secretome members confer the Verticillium wilt resistance in cotton. CONCLUSIONS: Our data demonstrate that the cotton secretome plays an important role in Verticillium wilt resistance, facilitating the development of the resistance gene markers and increasing the understanding of the mechanisms regulating disease resistance.

To study the beneficial effects of ivabradine in dilated cardiomyopathy mice, which evolved from coxsackievirus B3-induced chronic viral myocarditis. Four-to-five-week-old male balb/c mice were inoculated intraperitoneally with coxsackievirus B3 (Strain Nancy) on day 1, day 14 and day 28. The day of the first virus inoculation was defined as day 1. Thirty-five days later, the surviving chronic viral myocarditis mice were divided randomly into two groups, a treatment group and an untreated group. Ivabradine was administered by gavage for 30 consecutive days in the treatment group, and the untreated group was administered normal saline. Masson’s trichrome stain was used to evaluate the fibrosis degree in myocardial tissue. The expression levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), interleukin-6 (IL-6), Collagen I, Collagen III and p38-MAPK signaling pathway proteins were detected by western blot. Electrocardiogram was used to investigate the heart rate and rhythm. The thickness of the ventricular septum and left ventricular posterior wall, left ventricular end diastolic dimension, left ventricular end systolic dimension, left ventricular ejection fractions and fractional shortening were studied by echocardiography. Compared with the untreated chronic viral myocarditis mice, ivabradine significantly increased the survival rate, attenuated the myocardial lesions and fibrosis, improved the impairment of the left ventricular function, diminished the heart dimension, decreased the production of collagen I and collagen III, reduced the expression of the proinflammatory cytokines TNF-α, IL-1β, and IL-6, and lowered the production of phospho-p38MAPK. The findings indicate the therapeutic effect of ivabradine in preventing the progression from viral myocarditis to dilated cardiomyopathy in mice with chronic viral myocarditis induced by coxsackievirus B3, is associated with inhibition of the p38MAPK pathway, downregulated inflammatory responses and decreased collagen expression. Ivabradine appears a promising approach for the treatment of patients with viral myocarditis.

In view of problem that conventional PID controller is difficult to obtain satisfactory control effect due to sampling frequency and other factors, a control strategy of fractional order calculus for shunt active power filter using improved Oustaloup algorithm for solving fractional calculus was studied. The Matlab simulation results show that the control strategy can better stabilize DC side voltage to set point, and can get smoother grid current which is closer to sine wave than conventional PID control strategy. Impact current at the compensation instant is small, and tracking and compensation current can track command current effectively.

Dysfunctional pancreatic islet beta cells are a hallmark of type 2 diabetes (T2D), but a comprehensive understanding of the underlying mechanisms, including gene dysregulation, is lacking. Here we integrate information from measurements of chromatin accessibility, gene expression and function in single beta cells with genetic association data to nominate disease-causal gene regulatory changes in T2D. Using machine learning on chromatin accessibility data from 34 nondiabetic, pre-T2D and T2D donors, we identify two transcriptionally and functionally distinct beta cell subtypes that undergo an abundance shift during T2D progression. Subtype-defining accessible chromatin is enriched for T2D risk variants, suggesting a causal contribution of subtype identity to T2D. Both beta cell subtypes exhibit activation of a stress-response transcriptional program and functional impairment in T2D, which is probably induced by the T2D-associated metabolic environment. Our findings demonstrate the power of multimodal single-cell measurements combined with machine learning for characterizing mechanisms of complex diseases.

This study investigated the compensating mediating mechanism of resilience in the relationship between loneliness and life satisfaction in Thai and Chinese college students during a risk period of coronavirus disease 2019 transmission. A total of 262 Thai and 247 Chinese college students were enrolled in the study between December 2020 and January 2021. A measurement model was constructed with the items of loneliness, resilience, and life satisfaction by using structural equation models. Subsequently, structural mediating models were constructed according to the valid items and a multigroup comparison was performed. The relationship between loneliness and life satisfaction differed significantly between Thai and Chinese college students as a result of the mediating effect of resilience. Resilience in Thai college students partly compensated for the negative influence of loneliness on life satisfaction, and resilience in Chinese college students compensated completely for the negative influence of loneliness on life satisfaction.

Understanding how plant pathogenic fungi adapt to their hosts is of critical importance to securing optimal crop productivity. In response to pathogenic attack, plants produce reactive oxygen species (ROS) as part of a multipronged defense response. Pathogens, in turn, have evolved ROS scavenging mechanisms to undermine host defense. Thioredoxins (Trx) are highly conserved oxidoreductase enzymes with a dithiol-disulfide active site, and function as antioxidants to protect cells against free radicals, such as ROS. However, the roles of thioredoxins in Verticillium dahliae, an important vascular pathogen, are not clear. Through proteomics analyses, we identified a putative thioredoxin (VdTrx1) lacking a signal peptide. VdTrx1 was present in the exoproteome of V. dahliae cultured in the presence of host tissues, a finding that suggested that it plays a role in host-pathogen interactions. We constructed a VdTrx1 deletion mutant ΔVdTrx1 that exhibited significantly higher sensitivity to ROS stress, H2O2, and tert-butyl hydroperoxide (t-BOOH). In vivo assays by live-cell imaging and in vitro assays by western blotting revealed that while VdTrx1 lacking the signal peptide can be localized within V. dahliae cells, VdTrx1 can also be secreted unconventionally depending on VdVps36, a member of the ESCRT-II protein complex. The ΔVdTrx1 strain was unable to scavenge host-generated extracellular ROS fully during host invasion. Deletion of VdTrx1 resulted in higher intracellular ROS levels of V. dahliae mycelium, displayed impaired conidial production, and showed significantly reduced virulence on Gossypium hirsutum, and model plants, Arabidopsis thaliana and Nicotiana benthamiana. Thus, we conclude that VdTrx1 acts as a virulence factor in V. dahliae.

BackgroundVerticillium dahliae is a fungal pathogen that causes a vascular wilt on many economically important crops. Common fungal extracellular membrane (CFEM) domain proteins including secreted types have been implicated in virulence, but their roles in this pathogen are still unknown.ResultsNine secreted small cysteine-rich proteins (VdSCPs) with CFEM domains were identified by bioinformatic analyses and their differential suppression of host immune responses were evaluated. Two of these proteins, VdSCP76 and VdSCP77, localized to the plant plasma membrane owing to their signal peptides and mediated broad-spectrum suppression of all immune responses induced by typical effectors. Deletion of either VdSCP76 or VdSCP77 significantly reduced the virulence of V. dahliae on cotton. Furthermore, VdSCP76 and VdSCP77 suppressed host immunity through the potential iron binding site conserved in CFEM family members, characterized by an aspartic acid residue in seven VdSCPs (Asp-type) in contrast with an asparagine residue (Asn-type) in VdSCP76 and VdSCP77. V. dahliae isolates carrying the Asn-type CFEM members were more virulent on cotton than those carrying the Asp-type.ConclusionsIn the iron-insufficient xylem, V. dahliae is likely to employ the Asp-type CFEM members to chelate iron, and Asn-type CFEM members to suppress immunity, for successful colonization and propagation in host plants.

BackgroundDuring the disease cycle, plant pathogenic fungi exhibit a morphological transition between hyphal growth (the phase of active infection) and the production of long-term survival structures that remain dormant during "overwintering." Verticillium dahliae is a major plant pathogen that produces heavily melanized microsclerotia (MS) that survive in the soil for 14 or more years. These MS are multicellular structures produced during the necrotrophic phase of the disease cycle. Polyketide synthases (PKSs) are responsible for catalyzing production of many secondary metabolites including melanin. While MS contribute to long-term survival, hyphal growth is key for infection and virulence, but the signaling mechanisms by which the pathogen maintains hyphal growth are unclear.ResultsWe analyzed the VdPKSs that contain at least one conserved domain potentially involved in secondary metabolism (SM), and screened the effect of VdPKS deletions in the virulent strain AT13. Among the five VdPKSs whose deletion affected virulence on cotton, we found that VdPKS9 acted epistatically to the VdPKS1-associated melanin pathway to promote hyphal growth. The decreased hyphal growth in VdPKS9 mutants was accompanied by the up-regulation of melanin biosynthesis and MS formation. Overexpression of VdPKS9 transformed melanized hyphal-type (MH-type) into the albinistic hyaline hyphal-type (AH-type), and VdPKS9 was upregulated in the AH-type population, which also exhibited higher virulence than the MH-type.ConclusionsWe show that VdPKS9 is a powerful negative regulator of both melanin biosynthesis and MS formation in V. dahliae. These findings provide insight into the mechanism of how plant pathogens promote their virulence by the maintenance of vegetative hyphal growth during infection and colonization of plant hosts, and may provide novel targets for the control of melanin-producing filamentous fungi.

The stable slurry was prepared by ball-milling with a certain proportion of silica fume as silicon source,glucose as carbon source and metal niobium (Nb) as additives mixed with alcohol water. After the slurry was dried and pulverized, put the powder into a tubular furnace heated to 650℃ for 2h under nitrogen atmosphere to prepare precursor, the heating rate was 5℃/min. Treated the precursor under vacuum by carbothermal reduction method to prepare silicon carbide (SiC) powder. The effect of temperature and additive content on the morphology of synthesised silicon carbide and the impurity removal order on product purity were explored. After firing at 1300℃,1400℃,1500℃ for 2 h, the 3C-SiC powders are detected, and as the temperature increased, the crystallinity of the product become better. When the content of the additive is 1% of the quality of the silica fume, the particle size of the silicon carbide synthesized at the temperature of 1500℃ is even and the dispersion is better. As for the impurity removal order, removed SiO2 first, then removed C can effectively remove the impurities in the product.

Verticillium dahliae is a notorious soil-borne pathogen that enters hosts through the roots and proliferates in the plant water-conducting elements to cause Verticillium wilt. Historically, Verticillium wilt symptoms have been explained by vascular occlusion, due to the accumulation of mycelia and plant biomacromolecule aggregation, and also by phytotoxicity caused by pathogen-secreted toxins. Beyond the direct cytotoxicity of some members of the secretome, this review systematically discusses the roles of the V. dahliae secretome in vascular occlusion, including the deposition of polysaccharides as an outcome of plant cell wall destruction, the accumulation of fungal mycelia, and modulation of plant defence responses. By modulating plant defences and hormone levels, the secretome manipulates the vascular environment to induce Verticillium wilt. Thus, the secretome of V. dahliae colludes with plant defence responses to modulate Verticillium wilt symptoms, and thereby bridges the historical concepts of both toxin production by the pathogen and vascular occlusion as the cause of wilting symptoms.

The arms race between fungal pathogens and plant hosts involves recognition of fungal effectors to induce host immunity. Although various fungal effectors have been identified, the effector functions of ribonucleases are largely unknown. Herein, we identified a ribonuclease secreted by Verticillium dahliae (VdRTX1) that translocates into the plant nucleus to modulate immunity. The activity of VdRTX1 causes hypersensitive response (HR)-related cell death in Nicotiana benthamiana and cotton. VdRTX1 possesses a signal peptide but is unlikely to be an apoplastic effector because its nuclear localization in the plant is necessary for cell death induction. Knockout of VdRTX1 significantly enhanced V. dahliae virulence on tobacco while V. dahliae employs the known suppressor VdCBM1 to escape the immunity induced by VdRTX1. VdRTX1 homologs are widely distributed in fungi but transient expression of 24 homologs from other fungi did not yield cell death induction, suggesting that this function is specific to the VdRTX1 in V. dahliae. Expression of site-directed mutants of VdRTX1 in N. benthamiana leaves revealed conserved ligand-binding sites that are important for VdRTX1 function in inducing cell death. Thus, VdRTX1 functions as a unique HR-inducing effector in V. dahliae that contributes to the activation of plant immunity.

Parnassia wightiana Wall. ex Wight & Arn., a perennial herb, is an alpine plant distributed across three biodiversity hotspots in China. This species offers an excellent study system to analyze the distribution pattern and genetic structure of high mountainous plant populations. Three chloroplast DNA (cpDNA) regions (rpl32-trnL, trnL-F, and trnS-G) from 442 individuals of 39 populations and internal transcribed spacers (ITS) from 418 individuals of 38 populations were sequenced and analyzed. We identified a total of 26 haplotypes based on the concatenated sequences of cpDNA and a total of 36 based on ITS sequences. A hierarchical analysis of molecular variance indicated significant phylogeographic structure among populations. Mismatch analysis of multipeak and Tajima's D neutral test demonstrated Parnassia wightiana populations within China had not experienced abrupt expansion recently. The allopatric fragmentation resulted in the geographic isolation due to environmental heterogeneity, producing a high proportion of private haplotypes in the distribution of Parnassia wightiana within China.
