Agrivoltaics is presented as a possible solution to the need for new sources of renewable energies, also responding to the increasing demand for feed/food and energy in a strongly efficient and sustainable manner. To this aim, agrivoltaics proposes to combine agricultural and renewable energy production on the same land using photovoltaic technology. The performance of this new production model strongly depends on the interaction between the two systems, agricultural and photovoltaic. In that sense, one of the most important aspects to consider are the effects of the shadows of the photovoltaic panels on the crop land. Overall, the experiment clearly indicated that a fourth cycle of escarole is possible under the PVs of agrivoltaics. Both fresh weight and size of the salad bowls were significantly increased by the shade provided by the PVs. Escarole appeared to be very tolerant to the shade and commercial yields were boosted, compared to full sun treatments, even under extended shade conditions. Such an effect can be likely explained by an overall amelioration of the water status in shaded plots. Therefore, a further study of the behavior of escarole under agrivoltaic conditions will be desirable.

With a view to understanding the dynamics of ancient trade and agrobiodiversity, archaeobotanical remains provide a means of tracing the trajectories of certain agricultural commodities. A prime example is cotton in Arabia, a plant that is non-native but has been found in raw seed and processed textile form at Hegra and Dadan, in the region of al-ʿUlā, north-western Saudi Arabia—sites of critical importance given their role in the trans-Arabian trading routes during Antiquity. Here, we demonstrate that the measurement of strontium isotopes from pre-cleaned archaeological cotton is methodologically sound and is an informative addition to the study of ancient plant/textile provenance, in this case, putting forward evidence for local production of cotton in oasis agrosystems and possible external supply. The presence of locally-grown cotton at these sites from the late 1st c. BCE–mid 6th c. CE is significant as it demonstrates that cotton cultivation in Arabia was a Pre-Islamic socio-technical feat, while imported cotton highlights the dynamism of trade at that time.

Hay is one of the primary constituents of ruminant feed, and rapid classification systems of nutritional value are essential. A reliable approach to evaluating hay quality is a combination of visual combined inspection by NIRS analysis. The analysis was carried out on 1,639 samples of hay collected from 2016 to 2021 in northern Italy. Discriminant analysis (DAPC) on five hay types (FOM, forage mixtures; APG, first alfalfa cutting with prevalence of graminaceous >50%; PRA, prevailing alfalfa >50%; PUA, purity alfalfa >95%; and PEM, permanent meadows) was performed by ex-ante visual inspection categorization and NIRS analysis. This study aimed to provide a complementary method to differentiate hay types and classify unknown samples. Two scenarios were used: i) all data were used for model training, and the discriminant functions were extracted based on all samples; ii) the assignment of each group was assessed without samples belonging to the training set group. DAPC model resulted in an overall assignment success rate of 66%; precisely, the success was 84, 79, 69, 37, and 27% for PUA, FOM, PRA, APG, and PEM, respectively. In the second scenario, three groups showed percentages of posterior assignment probability higher than 70% to only one group: PUA with PRA (~ 99%), PRA with PUA (~71%), and PEM with FOM (~75%). Discriminant analysis can be successfully used to differentiate hay types and could also be used to assess factors related to hay quality in addition to NIRS analysis.

Objectives: The optimal healing of skin wounds, deep burns, and chronic ulcers is an important clinical problem. Attempts to solve it have been driving the search for skin equivalents based on synthetic or natural polymers. Methods: Consistent with this endeavor, we used regenerated silk fibroin (SF) from Bombyx mori to produce a novel compound scaffold by welding a 3D carded/hydroentangled SF-microfiber-based nonwoven layer (C/H-3D-SFnw; to support dermis engineering) to an electrospun 2D SF nanofiber layer (ESFN; a basal lamina surrogate). Next, we assessed—via scanning electron microscopy, attenuated total reflectance Fourier transform infrared spectroscopy, differential scanning calorimetry, mono- and co-cultures of HaCaT keratinocytes and adult human dermal fibroblasts (HDFs), dsDNA assays, exosome isolation, double-antibody arrays, and angiogenesis assays—whether the C/H-3D-SFnws/ESFNs would allow the reconstitution of a functional human skin analog in vitro. Results: Physical analyses proved that the C/H-3D-SFnws/ESFNs met the requirements for human soft-tissue-like implants. dsDNA assays revealed that co-cultures of HaCaTs (on the 2D ESFN surface) and HDFs (inside the 3D C/H-3D-SFnws) grew more intensely than did the respective monocultures. Double-antibody arrays showed that the CD9+/CD81+ exosomes isolated from the 14-day pooled growth media of HDF and/or HaCaT mono- or co-cultures conveyed 35 distinct angiogenic/growth factors (AGFs). However, versus monocultures’ exosomes, HaCaT/HDF co-cultures’ exosomes (i) transported larger amounts of 15 AGFs, i.e., PIGF, ANGPT-1, bFGF, Tie-2, Angiogenin, VEGF-A, VEGF-D, TIMP-1/-2, GRO-α/-β/-γ, IL-1β, IL-6, IL-8, MMP-9, and MCP-1, and (ii) significantly more strongly stimulated human dermal microvascular endothelial cells to migrate and assemble tubes/nodes in vitro. Conclusions: Our results showed that both cell–cell and cell–SF interactions boosted the exosomal release of AGFs from HaCaTs/HDFs co-cultured on C/H-3D-SFnws/ESFNs. Hence, such exosomes are an asset for prospective clinical applications as they advance cell growth and neoangiogenesis and consequently graft take and skin healing. Moreover, this new integument analog could be instrumental in preclinical and translational studies on human skin pathophysiology and regeneration.

Mastitis is a most common disease of dairy cows and causes tremendous economic loss to the dairy industry worldwide. Somatic cell counts (SCC) reflect the inflammatory response to infections and is a metric used as key indicator in mastitis screening programs, typically within the framework of national milk recording schemes. Besides the determination of total SCC, the differentiation of cell types has been described to be beneficial for a more definite description of the actual udder health status of dairy cows. Differential somatic cell count (DSCC) represents the combined proportion of polymorphonuclear leukocytes (PMN) and lymphocytes expressed as a percentage of the total. The aim of this study was to investigate the relationship between SCC and differential somatic cell count (DSCC) in individual quarter milk samples collected at different time points: at dry-off, after calving and at the lactation peak. We used individual quarter data from farms representing the specialized production system of Parmigiano Reggiano cheese in Northern Italy. Average DSCC values ranged between 44.9% and 56.3%, with higher values (60.4%-72.1%) in milk samples with ≥ 1 million SCC/ml (where the proportion of samples with DSCC > 70% can be as high as 0.73). Moderate overall correlations between DSCC and log(SCC) were estimated (Pearson = 0.42, Spearman = 0.38), with a clear increasing trend with parity and around the lactation peak (e.g. Pearson = 0.59 at 60 DIM in parity 4). Taking SCC values as indicators of subclinical mastitis, DSCC would diagnose mastitis with 0.75 accuracy. Data editing criteria do have an impact on results, with stricter filtering leading to lower correlations between log(SCC) and DSCC. In conclusion DSCC and SCC provide different descriptions of the udder health status of dairy cows which, at least to some extent, are independent. DSCC alone doesn't provide more accurate information than SCC at quarter level but, used in combination with SCC, can be of potential interest within the framework of milk recording programs, especially in the context of selective dry-cow therapy (SDCT). However, this needs further investigation and updated threshold values need to be selected and validated.

The dissolution of Bombyx mori silk fibroin (SF) films in formic acid (FA) for the preparation of electrospinning dopes is widely exploited to produce electrospun SF scaffolds. The SILKBridge® nerve conduit is an example of medical device having in its wall structure an electrospun component produced from an FA spinning dope. Though highly volatile, residual FA remains trapped into the bulk of the SF nanofibers. The purpose of this work is to investigate the type and strength of the interaction between FA and SF in electrospun mats, to quantify its amount and to evaluate its possible toxicological impact on human health. The presence of residual FA in SF mats was detected by FTIR and Raman spectroscopy (new carbonyl peak at about 1,725 cm−1) and by solid state NMR, which revealed a new carbonyl signal at about 164.3 ppm, attributed to FA by isotopic 13C substitution. Changes occurred also in the spectral ranges of hydroxylated amino acids (Ser and Thr), demonstrating that FA interacted with SF by forming formyl esters. The total amount of FA was determined by HS-GC/MS analysis and accounted for 247 ± 20 μmol/g. The greatest part was present as formyl ester, a small part (about 3%) as free FA. Approximately 17% of the 1,500 μmol/g of hydroxy amino acids (Ser and Thr) theoretically available were involved in the formation of formyl esters. Treatment with alkali (Na2CO3) succeeded to remove the greatest part of FA, but not all. Alkali-treated electrospun SF mats underwent morphological, physical, and mechanical changes. The average diameter of the fibers increased from about 440 nm to about 480 nm, the mat shrunk, became stiffer (the modulus increased from about 5.5 MPa to about 7 MPa), and lost elasticity (the strain decreased from about 1 mm/mm to about 0.8 mm/mm). Biocompatibility studies with human adult dermal fibroblasts did not show significant difference in cell proliferation (313 ± 18 and 309 ± 23 cells/mm2 for untreated and alkali-treated SF mat, respectively) and metabolic activity. An in-depth evaluation of the possible toxicological impact of residual FA was made using the SILKBridge® nerve conduit as case study, following the provisions of the ISO 10993-1 standard. The Potential Patient Daily Intake, calculated from the total amount of FA determined by HS-GC/MS, was 2.4 mg/day and the Tolerable Exposure level was set to 35.4 mg/day. This allowed to obtain a value of the Margin of Safety of 15, indicating that the amount of FA left on SF mats after electrospinning does not raise concerns for human health.

Increasingly prevalent acute and chronic human brain diseases are scourges for the elderly. Besides the lack of therapies, these ailments share a neuroinflammation that is triggered/sustained by different innate immunity-related protein oligomers called inflammasomes. Relevant neuroinflammation players such as microglia/monocytes typically exhibit a strong NLRP3 inflammasome activation. Hence the idea that NLRP3 suppression might solve neurodegenerative ailments. Here we review the recent Literature about this topic. First, we update conditions and mechanisms, including RNAs, extracellular vesicles/exosomes, endogenous compounds, and ethnic/pharmacological agents/extracts regulating NLRP3 function. Second, we pinpoint NLRP3-activating mechanisms and known NLRP3 inhibition effects in acute (ischemia, stroke, hemorrhage), chronic (Alzheimer’s disease, Parkinson’s disease, Huntington’s disease, MS, ALS), and virus-induced (Zika, SARS-CoV-2, and others) human brain diseases. The available data show that (i) disease-specific divergent mechanisms activate the (mainly animal) brains NLRP3; (ii) no evidence proves that NLRP3 inhibition modifies human brain diseases (yet ad hoc trials are ongoing); and (iii) no findings exclude that concurrently activated other-than-NLRP3 inflammasomes might functionally replace the inhibited NLRP3. Finally, we highlight that among the causes of the persistent lack of therapies are the species difference problem in disease models and a preference for symptomatic over etiologic therapeutic approaches. Therefore, we posit that human neural cell-based disease models could drive etiological, pathogenetic, and therapeutic advances, including NLRP3’s and other inflammasomes’ regulation, while minimizing failure risks in candidate drug trials.

Permanent grasslands represent the main terrestrial ecosystem and serve as an important global reservoir of biodiversity, providing a wide range of benefits to humans and ecosystems. The effects of environment on permanent meadows (in our survey, they were centuries-old meadows that had not been plowed, mowed, or fertilized with manure) production have been adequately investigated in literature. However, plant species composition impact on potential feed value of first cut has still to be understood, in particular regarding different agronomic management. Our field trial was carried out in five farms, in a territory involved in the value chain of the Parmigiano Reggiano PDO (Val d’Enza, Northern Italy), over a two-year period (2017–2018). Differences in botanical composition, biomass, and Pastoral Value index (PV), which synthesizes grassland yield and nutritional parameters, were investigated in depth. The herbage dry matter (DM) yield was affected by year, farm, and their interaction factors. Its highest value across the two years was recorded in farm 5 (11.7 tons of DM ha−1), which applied the highest rate of nitrogen fertilization. The botanical composition of the first cut has favored the presence of both Poaceae and ‘other species’ (each one around 40 plants per transect) compared to Fabaceae (seven plants per transect). However, higher numbers of Fabaceae plants (13 and 10) plausibly determined increases in PV in farms 3 and 5 (56.4 and 58.7, respectively). Although differences were observed among the most important nutritional parameters of grassland (crude protein, digestible and undigested neutral detergent fiber contents), suitable net energy for lactation (NEL) values for feeding lactating cows were always recorded during the two years of survey. The present study provides a contribution of knowledge on how the botanical composition of permanent meadows may affect their potential nutritive value as fresh herbage for feeding dairy cows. Considering these results, the agronomic management should seek a level of plant biodiversity that at the same time might guarantee satisfactory yield and feed value, also in a context of climate change.

The objective of this study was to quantify the climate change (CC) impact of the honey supply chain in different beekeeping systems and farms, over two consecutive years. The CC impact category is quantified as kg CO2 equivalent and it evaluates the GHG emissions, mainly CO2, N2O, and CH4. The results ranged from 0.44 to 3.18 (p = 0.039) kg CO2e/kg honey with higher values in 2021 than 2020. The main contributors to climate change of the honey supply chain are represented by transport and supplemental feeding inputs. The beekeeping system (migratory or stationary) influenced CC: the contribution to CC for stationary farms was estimated at 0.58 kg CO2e/kg honey and 2.48 for migratory ones (p < 0.001). Given the close connection between honey yield and LCA results due to the unit of measurement of impact, i.e., kg of honey produced, an index was developed (wildflower honey climate index) as a simple benchmark tool for prediction of honey yield in the survey context. Using the data from the present study, we found that the index is positively related to honey yield (r = 0.504; p < 0.05) but negatively related to supplemental feeding (r = −0.918; p < 0.01) and overall carbon footprint (r = −0.657; p < 0.05). Further studies are needed to better explain the effects of weather on honey production, as well as environmental impact.

Fresh samples of Longissimus thoracis of Charolais (n = 12), Romagnola (n = 15), Limousine (n = 77), and crossbreed (n = 62) animals were evaluated with different storage periods (0, 4, 9, and 14 days). Proximate analysis (i.e., pH, humidity, color, free water content, and physical parameters) was performed for each sample. The data obtained were evaluated with a mixed model, setting 5 fixed effects (breed, storage time, animals age, EUROP conformation, number of animal transports) and the animal as random. The results demonstrated that meat quality was affected mainly by the wet ageing period and that the visual and tactile parameters were also found to be susceptible to the storage time. The conservation entailed a decrease in meat humidity and an increase in L* and b* traits; it also led to a decrease in the hardness of the sample, in turn affecting the other texture profile analysis parameters considered. Fixed effects affected in different ways the traits analyzed i.e., pH and humidity changed with breed, as well as with EUROP classification, animals’ age for some TPA parameters, and the number of animal transports for both visual and tactile parameters. Wet ageing influenced the meat quality, often improving it, confirming how important further research would be to identify precise storage times in relation to the parameters studied.

Diabetic foot ulcers (DFUs) present significant challenges due to their associated amputation rates, mortality, treatment complexity and excessive costs. Our earlier work introduced a wound surgical integrated treatment (WSIT) for DFUs, yielding promising outcomes. This study focuses on a specific WSIT protocol employing antibiotic‐loaded bone cement (ALBC) in the first Stage, and free vastus lateralis muscle‐sparing (VLMS) flaps and split‐thickness skin grafts (STSGs) in the second stage to repair non‐weight‐bearing DFUs. From July 2021 to July 2023, seven DFU patients (aged 47–71 years) underwent this treatment. Demographic data, hospital stay and repair surgery times were collected. Histological and immunohistochemical analyses assessed angiogenesis, collagen deposition and inflammation. SF‐36 questionnaire measured pre‐ and postoperative quality of life. Preoperative ultrasound Doppler showed that the peak blood flow velocity of the recipient area artery was significantly >30 cm/s (38.6 ± 6.8 cm/s) in all patients. Muscle flap sizes varied from 8 × 3.5 × 1 to 18 × 6 × 2 cm. The operation time of the repair surgery was 156.9 ± 15.08 minutes, and the hospital stay was 18.9 ± 3.3 days. Histological analysis proved that covering DFUs with ALBC induced membrane formation and increased collagen, neovascularization and M2 macrophages fraction while reducing M1 macrophages one. All grafts survived without amputation during a 7‐ to 24‐month follow‐up, during which SF‐36 scores significantly improved. A combination of ALBC with free VLMS flaps and STSGs proved to be safe and effective for reconstructing non‐weight‐bearing DFUs. It rapidly controlled infection, enhanced life quality and foot function, and reduced hospitalization time. We advocate integrating this strategy into DFU treatment plans. [ABSTRACT FROM AUTHOR]

A byproduct from orange juice processing known as pastazzo represents a significant organic waste stream. Rich in essential oils and known for its inhibitory effect on plant germination, pastazzo could serve as a valuable input for agricultural purposes. This study assesses the effects of a 40% w/v orange pastazzo water extract (OPWE) produced by hydrodynamic cavitation on the germination of two species, one of economic interest (Lactuca sativa L.) and one common weed (Chenopodium album L.). Three dilutions of OPWE in water (25%; 50%; 75%) were compared to a control treatment in four experiments, using (i) seeds in Petri dishes; (ii) seeds in commercial substrate; (iii) C. album seeds and transplanted L. sativa in commercial substrate; and (iv) other weeds in an open-field plantation of L. sativa. Highly rich in limonene, OPWE applied at higher concentrations in Petri dishes caused the effective inhibition of germination in C. album and a germination delay in L. sativa. Similar results were observed in the germination of the two species in commercial substrate, with none of the dilutions affecting L. sativa biomass. In the field experiment, despite a relatively low number of weeds in the control treatment, higher OPWE concentrations reduced the number of grasses and forbs, largely confirming the inhibitory effects. We conclude that OPWE produced with hydrodynamic cavitation, an efficient and affordable method of extraction, represents an effective crop treatment due to the species-specific effects of its constituent limonene on plant germination. Abstact Cutaneous regeneration at the wound site involves several intricate and dynamic processes which require a series of coordinated interactions implicating various cell types, growth factors, extracellular matrix (ECM), nerves, and blood vessels. Mesenchymal stromal cells (MSCs) take part in all the skin wound healing stages playing active and beneficial roles in animal models and humans. Exosomes, which are among the key products MSCs release, mimic the effects of parental MSCs. They can shuttle various effector proteins, messenger RNA (mRNA) and microRNAs (miRNAs) to modulate the activity of recipient cells, playing important roles in wound healing. Moreover, using exosomes avoids many risks associated with cell transplantation. Therefore, as a novel type of cell-free therapy, MSC-exosome -mediated administration may be safer and more efficient than whole cell. In this review, we provide a comprehensive understanding of the latest studies and observations on the role of MSC-exosome therapy in wound healing and cutaneous regeneration. In addition, we address the hypothesis of MSCs microenvironment extracellular vesicles (MSCs-MEVs) or MSCs microenvironment exosomes (MSCs-MExos) that need to take stock of and solved urgently in the related research about MSC-exosomes therapeutic applications. This review can inspire investigators to explore new research directions of MSC-exosome therapy in cutaneous repair and regeneration.

Mycobacterium avium subsp. paratuberculosis (MAP) is the causative agent of paratuberculosis [Johne's disease (JD)], a chronic disease that causes substantial economic losses in the dairy cattle industry. The long incubation period means clinical signs are visible in animals only after years, and some cases remain undetected because of the subclinical manifestation of the disease. Considering the complexity of JD pathogenesis, animals can be classified as infected, infectious, or affected. The major limitation of currently available diagnostic tests is their failure in detecting infected non-infectious animals. The present study aimed to identify metabolic markers associated with infected and infectious stages of JD. Direct analysis in real time coupled with high resolution mass spectrometry (DART-HRMS) was, hence, applied in a prospective study where cohorts of heifers and cows were followed up annually for 2–4 years. The animals' infectious status was assigned based on a positive result of both serum ELISA and fecal PCR, or culture. The same animals were retrospectively assigned to the status of infected at the previous sampling for which all JD tests were negative. Stored sera from 10 infected animals and 17 infectious animals were compared with sera from 20 negative animals from the same herds. Two extraction protocols and two (-/+) ionization modes were tested. The three most informative datasets out of the four were merged by a mid-level data fusion approach and submitted to partial least squares discriminant analysis (PLS-DA). Compared to the MAP negative subjects, metabolomic analysis revealed the m/z signals of isobutyrate, dimethylethanolamine, palmitic acid, and rhamnitol were more intense in infected animals. Both infected and infectious animals showed higher relative intensities of tryptamine and creatine/creatinine as well as lower relative abundances of urea, glutamic acid and/or pyroglutamic acid. These metabolic differences could indicate altered fat metabolism and reduced energy intake in both infected and infectious cattle. In conclusion, DART-HRMS coupled to a mid-level data fusion approach allowed the molecular features that identified preclinical stages of JD to be teased out.

The study investigated the relationship between the temperature humidity index (THI) and the behaviour of 24 young fattening Limousin bulls reared in two farms in Tuscany, Italy. In each farm, six animals were undergone to ceiling fans (switched on at THI values up to 72), and six animals represented the control group. The trial lasted three days for two consecutive weeks in August 2020. Behavioural observations were conducted using scan sampling technique and eating, ruminating, drinking, resting and other social activities were registered every 5 min, from 9.30 am to 4.00 pm. Two different microclimatic conditions were evaluated to assess the effect of the ventilation system: normal (THI < 78) and alert (THI ≥ 78) conditions. Results showed that the ventilation system had significant effects increasing inactivity and lying down compared to control groups and decreasing eating and drinking activities. THI alert condition caused a significant decrease in eating and an increase in lying down behaviours. Ventilation system did not influence the animals’ cleanliness. The ceiling fans’ efficiency in changing the behaviour of young fattening bulls was demonstrated but further studies are needed to assess the ventilation system effects, especially during longer heat stress periods.

In this work we present a framework for blood cholesterol levels prediction from genotype data. The predictor is based on an algorithm for cholesterol metabolism simulation available in literature, implemented and optimized by our group in the R language. The main weakness of the former simulation algorithm was the need of experimental data to simulate mutations in genes altering the cholesterol metabolism. This caveat strongly limited the application of the model in the clinical practice. In this work we present how this limitation could be bypassed thanks to an optimization of model parameters based on patient cholesterol levels retrieved from literature. Prediction performance has been assessed taking into consideration several scoring indices currently used for performance evaluation of machine learning methods. Our assessment shows how the optimization phase improved model performance, compared to the original version available in literature.
