The precise measurement of a target depth has applications in biophysics and nanophysics, and non-linear optical methods are sensitive to intensity changes on very small length scales. By exploiting the high sensitivity of an autocorrelator's dependency on path length, we propose a technique that achieves $\approx$30 nm depth resolution for each pixel in 30 seconds. Our method images up-converted pulses from a non-linear crystal using a sCMOS camera and converts the intensity recorded by each pixel to a delay. By utilising statistical estimation theory and using the data from a set of 32$\times$32 pixels, the standard error of the detected delay falls below 1 nm after 30 seconds of measurement. Numerical simulations show that this result is extremely close to what can be achieved with a shot-noise-limited source and is consistent with the precision that can be achieved with a sCMOS camera.

Multifocal plane microscopy (MUM) allows three dimensional objects to be imaged in a single camera frame. Our approach uses dual orthogonal diffraction phase gratings with a quadratic distortion of the lines to apply defocus to the first diffraction orders which, when paired with a relay lens, allows for 9 focal planes to be imaged on a single camera chip. This approach requires a strong signal level to ensure sufficient intensity in the diffracted light, but has the advantage of being compact and straightforward to implement. As the microscope begins to focus deeper into the sample, aberrations caused by refractive index mismatch and inhomogeneity in the sample's media have an adverse effect on the signal's quality. In this paper, we investigate the image quality improvement brought by applying adaptive optics (AO) to multifocal plane microscopy. A single correction device (an 8x8 deformable mirror (DM)) is combined with an image-based AO control strategy to perform the correction of optical aberrations. We compare full end-to-end modelling results using an established numerical modelling system adapted for microscopy to laboratory results both on a test sample and on a number of biological samples. Finally, we will demonstrate that combining AO and MUM, we are able to improve the image quality of biological samples and provide a good correction throughout the volume of the biological sample., Comment: 4 pages, 5 figures. We present a dual color laser scanning endomicroscope capable of fluorescence lifetime endomicroscopy at one frame per second (FPS). The scanning system uses a coherent imaging fiber with 30,000 cores. High-speed lifetime imaging is achieved by distributing the signal over an array of 1024 parallel single-photon avalanche photo--diode detectors (SPADs), minimizing detection dead time maximizing the number of photons detected per excitation pulse without photon pile--up to achieve the high frame rate. Dual color fluorescence imaging is achieved by temporally shifting the dual excitation lasers, with respect to each other, to separate the two spectrally distinct fluorescent decays. Combining the temporal encoding, to provide spectral separation, with lifetime measurements we show a one FPS, multi-channel endomicroscopy platform for clinical applications and diagnosis. We demonstrate the potential of the system by imaging smartprobe-labeled bacteria in ex vivo samples of human lung using lifetime to differentiate bacterial fluorescence from the strong background lung auto--fluorescence which was used to provide structural information.

We demonstrate that photonic lanterns based on tapered multicore fibres provide an efficient way to couple multimode states of light to a two-dimensional array of Single-Photon Avalanche Detectors (SPADs), each of which has its own Time-to-Digital Converter (TDC) for Time-Correlated Single-Photon-Counting (TCSPC). Exploiting this capability, we demonstrate multiplexed single-mode wavelength-to-time conversion of multimode states of light using a multicore-fibre photonic lantern with 121 single-mode cores, coupled in a one-to-one fashion to 121 SPADS on a 32 $\times$ 32 pixel CMOS SPAD array. The application of photonic lanterns for coupling multimode light to SPAD arrays in this manner may find wide-ranging applications in areas such as Raman spectroscopy, coherent LIDAR and quantum optics., Comment: 15 pages, 4 figures

SPAD cameras offer single photon detection sensitivity, high frame rates and zero readout noise. They are a core technology for widefield FLIM, but have further potential in ultra-fast imaging applications. However, in practice sensitivity falls behind that of EMCCD and sCMOS devices due to the lower photon detection efficiency of SPAD arrays. This paper considers the application of a binary SPAD camera to the capture of blinking molecules as part of a superresolution (dSTORM/PALM) experiment. Simulation and experimental studies demonstrate that the effective sensitivity of the camera can be improved significantly by aggregation of signal only binary frames. The simulations also indicate that with future advances in SPAD camera technology, SPAD devices could in time outperform existing scientific cameras when capturing fast temporal dynamics.

The response of a single InGaAs quantum dot, embedded in a miniaturized charge-tunable device, to an applied GHz bandwidth electrical pulse is investigated via its optical response. Quantum dot response times of 1.0 \pm 0.1 ns are characterized via several different measurement techniques, demonstrating GHz bandwidth electrical control. Furthermore a novel optical detection technique based on resonant electron-hole pair generation in the hybridization region is used to map fully the voltage pulse experienced by the quantum dot, showing in this case a simple exponential rise., Comment: 7 pages, 4 figures
