11. Toxicity assessment and bioimaging potential of carbon dots synthesized from banana peel in zebrafish model

Abstract

Zebrafish serve as a pivotal model for bioimaging and toxicity assessments; however, the toxicity of banana peel-derived carbon dots in zebrafish has not been previously reported. The aim of this study was to assess the toxicity of carbon dots derived from banana peel in zebrafish, focusing on two types prepared through hydrothermal and pyrolysis methods. Banana peels were synthesized using hydrothermal and pyrolysis techniques and then compared for characteristics, bioimaging ability, and toxicity in zebrafish as an animal model. Pyrolysis-derived banana peel and hydrothermal-derived banana peel showed blue emission under ultraviolet light, indicating excitation-dependent behavior. To test their potential application for bioimaging, a soaking method was used using zebrafish that showed fluorescence intensity in the eyes, abdomen, and tail of zebrafish. Toxicity comparison showed that pyrolysis-derived banana peel had lower toxicity with 50% lethal concentrations (LC50) of 1707.3 ppm than hydrothermal-derived banana peel (LC50 993 ppm) in zebrafish. Both types of carbon dots showed significant differences (p<0.05) in hatching rates at 96 and 120 hours of exposure. Of the two methods for carbon dot synthesis from banana peel, the pyrolysis method had a higher toxicity threshold than the hydrothermal method, as indicated by the LC50 value and the number of zebrafish embryos that died, hatched delayed, and experienced malformation during their development.

Abstract

Four derivatives of 2'‐hydoxychalcone were synthesized through Claisen‐Schmidt condensation by combining 2'‐hydroxyacetophenone and four different benzaldehyde derivatives (o‐vanillin, salicylaldehyde, p‐anisaldehyde, and benzaldehyde) in the presence of a base. The reactions took place in an ionic liquid, 1‐dodecyl‐3‐methylimidazolium bromide ([DDMIm]Br), and were facilitated by microwave irradiation. The reactions were completed within 10 to 15 minutes, yielding products with chemical yields ranging from 52.64 % to 83.42 %. The structures of the synthesized products were confirmed using 1H NMR and 13C NMR spectroscopy. Additionally, antimicrobial activity tests demonstrated the effectiveness of specific derivatives against Staphylococcus aureus and Escherichia coli, underscoring the importance of hydroxy and methoxy groups on the aromatic ring in inhibiting microbial growth. These findings provide valuable knowledge for developing chalcone derivatives with improved properties and potential applications in drug discovery. Furthermore, the synthesized products′ reactivity was analyzed using Frontier Molecular Orbital (FMO) and their reactivity site was mapped using Molecular Electrostatic Potential (MEP). Lastly, molecular docking analysis was attempted to explain antibacterial activities from all products. Abstract

Eleutherine americana (E. americana) is a medicinal plant commonly found on the island of Borneo, Indonesia. This plant is known to have several biological activities. However, anthraquinone residues are generally present as contaminants. This study was aimed to develop a method of determining the levels of 9,10-anthraquinone in plant extracts and fractions using High Performance Liquid Chromatography (HPLC). The research aims to optimize the mobile phase, the system suitability test, and the system validation. The optimal mobile phase was acetonitrile:distilled water 1:1 v/v with a flow rate of 1.25 mL/min. The validation result shows that the linearity was obtained with a correlation coefficient (r) of 0.9995 and an r2 coefficient of 0.9991. The estimated limits for detection and quantification values were 0.178 and 0.594 µg/mL, respectively. In the intraday and inter-day accuracy test, the coefficient of variance for reference was 0.627 and 0.774, while the results for the sample were 2.966 and 2.658. The percentage recovery rate for reference was between 98.976–101.452%, and for the sample, the result was 89.191–94.667%. The average 9,10-anthraquinone content in the acetate fraction of E. americana plant was 9.799 µg/g ± 5.243.

Abstract

Invasive fungal infections cause serious illness and death worldwide. Long-term therapeutic and preventative use of antifungal drugs in high-risk patients has caused resistance. Triazole antifungals are widely used to prevent and treat fungal infections, and therapeutic drug monitoring has been suggested to improve outcomes, reduce toxicity, and prevent drug resistance. Common methods used for monitoring triazole antifungal drugs in biological matrices such as blood, serum, and plasma include bioassay and instrumentation methods, especially liquid chromatography. Sample preparation is needed to remove interference from liquid chromatography for reliable results. This paper evaluates the use of liquid chromatography to analyze triazole antifungal agents. We provided various chromatographic techniques combined with different detector types to analyze triazole antifungal drugs in biological matrices. We also compared chromatography systems with different sample preparation methods in order to select the most suitable analytical method for bioanalysis. [ABSTRACT FROM AUTHOR]

Abstract

Red bulbs of Eleutherine americana (Aubl.) Merr. ex K. Heyne has been known for its high content of naphthoquinones that have antifungal and antiparasitic activities. In this research, in silico interaction study was performed between 31 compounds reported to be found in E. americana with the selected target proteins for antifungal and antitoxoplasmosis activity using the molecular docking method. An ORPs (OSBP-related proteins), Osh4 (PDB ID: 1ZHX), and N-myristoyltransferase (Nmt, PDB ID: 1IYL) were used as the antifungal target proteins. Toxoplasma gondii purine nucleoside phosphorylase (TgPNP, PDB ID: 3MB8) and calcium-dependent protein kinase-1 (TgCDPK1, PDB ID: 4M84) were used as antitoxoplasmosis target proteins. Three-dimensional structures of the test compounds were made and optimized using GaussView 6.0 and Gaussian 09W. The target proteins were prepared using the Discovery Studio 2016 Program. Aquatic toxicity prediction as the preliminary assessment of the safety of the compounds was performed using ECOSAR v2.0. The results suggest that the compound having both the smallest free binding energy compared with positive control and other test compounds and low predicted toxicity is β-sitosterol with a free binding energy of ‒11.55 and ‒11.18 kcal/mol towards Osh4 and Nmt and ‒8.06 and ‒10.29 kcal/mol towards TgPNP and TgCDPK1, respectively.

Abstract

Hepatitis is a liver illness caused by virus such as hepatitis A virus, hepatitis B virus and hepatitis C virus. Hepatitis B and C are considerably more usual and induce more cirrhosis and dead worldwide than hepatitis A. Although drugs that are currently often used in the medication of hepatitis B and C, the finding of recent drug from various resources including herbal has been intensively developed. Therefore, the purpose of this review is to consider the possibility of plant’s compounds as anti-HBV and anti-HCV. From the results of a review of several articles, several plant’s compound have shown effectiveness againts HBV and HCV by in silico, in vitro and in vivo studies. In conclusion, several plant’s active compounds are possibility to be developed as anti-hepatitis B and C.
