Abstract

The kinesin light chain 3 protein (KLC3) is the only member of the kinesin light chain protein family that was identified in post-meiotic mouse male germ cells. It plays a role in the formation of the sperm midpiece through its association with both spermatid mitochondria and outer dense fibers (ODF). Previous studies showed a significant correlation between its expression level and sperm motility and quantitative semen parameters in humans, while the overexpression of a KLC3-mutant protein unable to bind ODF also affected the same traits in mice. To further assess the role of KLC3 in fertility, we used CRISPR/Cas9 genome editing in mice and investigated the phenotypes induced by the invalidation of the gene or of a functional domain of the protein. Both approaches gave similar results, i.e. no detectable change in male or female fertility. Testis histology, litter size and sperm count were not altered. Apart from the line-dependent alterations of Klc3mRNA levels, testicular transcriptome analysis did not reveal any other changes in the genes tested. Western analysis supported the absence of KLC3 in the gonads of males homozygous for the inactivating mutation and a strong decrease in expression in males homozygous for the allele lacking one out of the five tetratricopeptide repeats. Overall, these observations raise questions about the supposedly critical role of this kinesin in reproduction, at least in mice where its gene mutation or inactivation did not translate into fertility impairment.Although the kinesin light chain 3 protein has been proposed to play a critical role in midpiece formation and sperm parameters in humans and mice, mutation or inactivation of its encoding gene has no adverse effect on mouse fertility.Graphical Abstract

Abstract

AROMATASE is encoded by the CYP19A1 gene and is the cytochrome enzyme responsible for estrogen synthesis in vertebrates. In most mammals, a peak of CYP19A1 gene expression occurs in the fetal XX gonad when sexual differentiation is initiated. To elucidate the role of this peak, we produced 3 lines of TALEN genetically edited CYP19A1 knockout (KO) rabbits that were devoid of any estradiol production. All the KO XX rabbits developed as females with aberrantly small ovaries in adulthood, an almost empty reserve of primordial follicles, and very few large antrum follicles. Ovulation never occurred. Our histological, immunohistological, and transcriptomic analyses showed that the estradiol surge in the XX fetal rabbit gonad is not essential to its determination as an ovary, or for meiosis. However, it is mandatory for the high proliferation and differentiation of both somatic and germ cells, and consequently for establishment of the ovarian reserve. [ABSTRACT FROM AUTHOR]

Abstract

It has been shown that Forkhead boxL2 gene (FOXL2) is a master gene for female gonadal development. Firstly, it is involved in ovarian determination[1] and/or maintenance[2], and secondly it is necessary for ovarian follicle growth[3]. Interestingly, long-range deletions affecting FOXL2 expression have been reported in different species. In goats, a natural mutation called PIS for “Polled Intersex Syndrome” is responsible for female-to-male sex-reversal in homozygous XX PIS-/- mutant animals. This mutation corresponds to a 11.7kb homozygous deletion[4] lying at around 300 kb upstream of FOXL2 gene[5,6]. The PIS mutation is responsible for FOXL2 expression silencing and the sex-reversal[1]. In humans, various heterozygous long-range deletions have been shown to be responsible for decreasing FOXL2 expression, resulting in Blepharophimosis Ptosis Epicanthus inversus Syndrome[7]. To date, the Smallest Region of deletion Overlap (SRO) described in human patients, is a 7.4 kb region, encompassing the long non-coding RNA PISRT1 that was shown, in the goat species, to be closely regulated with FOXL2. All this data suggest that this distant region located at around 300 kb of FOXL2 may contain crucial regulatory elements for its expression. Analyzes of the epigenetic state of this region in human ovary (ChIP-Seq, ENCODE data), or of its counterpart in mouse ovary (ChIP-qPCR, personal data) revealed H3K27ac enrichment, which is an epigenetic mark preferentially observed on active gene enhancer or promoter. In order to prove that this region contains crucial enhancer(s), but also to demonstrate that long-range epigenetic modifications are able to control FOXL2 expression, we developed a program of epigenome editing. The CRISPR/dCas9 technology has been previously deflected to induce epigenetic modifications at specific loci[8]. Thus we designed several guide RNAs, coupled to a CRISPR/dCas9-p300 which is capable to acetylate H3K27. In a cellular model, different regions will be targeted, focusing on the most conserved elements of the SRO counterpart.

Abstract

National audience; In mammals, gonad differentiation initiates early in foetal life. The molecular mechanisms that underlie gonad differentiation through foetal and neonatal life until puberty have already been studied intensively in mouse species. A cascade of genes has thus been described as the main actors, which regulate specific steps. It is now accepted that early events, which occur during foetal life profoundly determine the sequence of further gonad development. However, foetal life is extremely short in mice when compared with other mammalian species (especially livestock species such as cows, goats, sheep and pigs) and humans. Even though the same events punctuate overall differentiation, the different milestones are not reached at the same time and probably do not have the same effect in each species. It is therefore difficult to rely on data obtained in the mouse to further explain the origin of aberrations in gonad development that may be encountered in other species, especially in humans. The aim of the present study was to report a first insight into gonad differentiation in the rabbit that is an attractive species because of its 31-day long gestation, the timing of female meiosis around birth and the 15-day delay between gonadal switch and the onset of meiosis in the female. The study was achieved by measuring the expression of genes already characterized as being important actors in sex determination and gonad differentiation in mouse species such as the SRY, Aromatase, STRA8, DMC1 and NANOS2 genes. At the same time, gonads were analysed histologically throughout the developmental period, alongside immunohistological localizations of proteins when this was possible. Interestingly, our data highlighted some rabbit-specific findings with respect to the gonad differentiation process.

Abstract

Rabbit is a non-rodent species, which genome sequence is closer to the human than the mouse genome. It is now generally admitted that the rabbit is a more pertinent species than the mouse for the production of physiological model relevant to the human species. In the field of sex determination and gonadal development, the mouse offers a powerful and well- established model thanks to the multiple possibilities to perform functional genetics through genome modifications. However, the foetal development in this species is extremely short, leading to the difficulty to study overlapping events and to compare with other species. The rabbit is especially valuable because it fills an important niche between the laboratory mouse and larger farm mammals. This species has been extensively studied in the past to lay the foundations of sex determination. To date, it is possible to produce genetically modified rabbits by additive transgenesis using the classical DNA microinjection procedure, or through targeted deletion induced by specific nucleases (zinc finger nucleases (ZFN) or transcription activator-like effector nucleases (TALEN)). Thus, our present study was undertaken to investigate in the rabbit the molecular pathways already characterized in the mouse and involved in sex determination and gonad development. A series of developmental stages were selected from the early genital crest stage 13 days post coitum (dpc) until birth (30 dpc) and few months after birth until puberty (3-5 months). Gonad differentiation was analysed by histological methods. The expression of a series of more than 35 candidate-genes was determined by RT-qPCR, and whenever possible, the proteins were localised in the tissues by immunohistochemistry. Among the various expression patterns, some have retained our attention. As in other mammals that have been studied to date, and by opposition to what has been described in the mouse, there is a short peak of aromatase gene expression in the ovary at 20 dpc. The role of this peak is under investigation through invalidation of the aromatase gene by a specific ZFN. In both sexes, rabbit germ cells undergo a series of mitosis during foetal life before entering meiosis that occurs in perinatal life in the ovary (29-30 dpc), and 7-9 weeks after birth in the testis. Interestingly, we observed in males at 29-30 dpc also a small but significant peak of expression of a set of genes characteristic of the onset of meiosis (Stra8, Spo11) without any subsequent meiosis but followed by a peak of expression of the Nanos2 gene considered as a meiosis suppressor. The significance of these gene expression profiles is now under investigation with the aim to revisit the molecular mechanisms that regulate germ cell differentiation and meiosis.
