We present the discovery of the radio afterglow of the short gamma-ray burst (GRB) 210726A, localized to a galaxy at a photometric redshift of z ∼ 2.4. While radio observations commenced ≲1 day after the burst, no radio emission was detected until ∼11 days. The radio afterglow subsequently brightened by a factor of ∼3 in the span of a week, followed by a rapid decay (a “radio flare”). We find that a forward shock afterglow model cannot self-consistently describe the multiwavelength X-ray and radio data, and underpredicts the flux of the radio flare by a factor of ≈5. We find that the addition of substantial energy injection, which increases the isotropic kinetic energy of the burst by a factor of ≈4, or a reverse shock from a shell collision are viable solutions to match the broadband behavior. At z ∼ 2.4, GRB 210726A is among the highest-redshift short GRBs discovered to date, as well as the most luminous in radio and X-rays. Combining and comparing all previous radio afterglow observations of short GRBs, we find that the majority of published radio searches conclude by ≲10 days after the burst, potentially missing these late-rising, luminous radio afterglows.

Pipe re-sizing of water distribution networks (WDNs) aims at improving the service performance to the required level, while minimizing the cost of replacing pipes in the network. The main challenge comes from the identification of the most effective pipes to re-size from a large number of interacting components. Performing a global search over all pipes in large WDNs does not guarantee a feasible and efficient solution due to the enormous search space, even by employing advanced algorithms, e.g., evolutionary algorithms. This paper proposes a novel method to reduce the search space for optimal re-sizing based on topological metrics from Complex Network Theory and hydraulic metrics, while providing suboptimal solutions comparable to the full search solutions, i.e., considering all pipes as candidates. The topological metrics are based on the edge-betweenness tailored for WDN analysis. Hydraulic metrics are unit head loss and flow rates of pipes computed based on simulation of the WDN in the current configuration. The optimal re-sizing plans obtained, particularly those using edge betweenness, were tested on a real WDN. The results are comparable with the full search solutions but they are much more efficient to obtain and require replacing mostly contiguous pipes, i.e., easier for practical fieldwork. Background: Grape agri-food wastes, such as skin, seeds, and other discarded by-products, contain phytochemical compounds that offer potential health benefits. Methods: This study aimed to investigate the polyphenol composition and bioactivities of different extracts obtained from grape marc and seeds, with the goal of exploring their potential for application as natural food additives. Results: Regardless of the extraction method used (dynamic maceration, ultrasound-assisted extraction (UAE), and microwave-assisted extraction (MAE)), all extracts exhibited relatively high concentrations of phenolic compounds. The chemical characterization of the extracts revealed the presence of specific compounds and chemical groups associated with each extraction methodology. Moreover, the extracts displayed satisfactory antioxidant activities, especially in inhibiting lipoperoxidation as assessed by the TBARS assay. Additionally, the extracts demonstrated effective inhibition against different strains of bacteria and fungi known as food contaminants. Taken together, these findings indicate that those extracts have the potential to be tested as natural antioxidants and preservatives with sustainable origins in food and beverage systems. Among the extraction methods evaluated, traditional maceration and UAE provided extracts with the highest antioxidant and antimicrobial activities. Conclusions: Our results suggest the opportunity to explore grape marc and seeds discarded by the winery industry in Portugal as natural sources of bioactive compounds, which could be employed as functional food ingredients or technological additives. The valorization of grape biowastes offers a promising strategy to reduce waste and harness their potential health benefits.

ABSTRACTThe Asian lineage of Zika virus (ZIKV) is responsible for the recent epidemics in the Americas and severe disease, whereas the African lineage of ZIKV has not been reported to cause epidemics or severe disease. We constructed a cDNA infectious clone (IC) of an African ZIKV strain, which, together with our previously developed Asian ZIKV strain IC, allowed us to engineer chimeric viruses by swapping the structural and non-structural genes between the two lineages. Recombinant parental and chimeric viruses were analyzed in A129 and newborn CD1 mouse models. In the A129 mice, the African strain developed higher viremia, organ viral loading, and mortality rate. In CD1 mice, the African strain exhibited a higher neurovirulence than the Asian strain. A chimeric virus containing the structural genes from the African strain is more virulent than the Asian strain, whereas a chimeric virus containing the non-structural genes from the African strain exhibited a virulence comparable to the Asian strain. These results suggest that (i) African strain is more virulent than Asian strain and (ii) viral structural genes primarily determine the virulence difference between the two lineages in mouse models. Other factors may contribute to the discrepancy between the mouse and epidemic results.

High-redshift gamma-ray bursts (GRBs) are useful to probe the early Universe, but only a few candidates have been detected so far. Here, we report the optical and near-infrared observations of the afterglow of a relatively high-redshift event GRB 220101A, which was triggered on New Year’s Day of 2022, and therefore referred to as the “New Year’s burst.” With the optical spectra obtained by XL2.16/BFOSC and NOT/ALFOSC, we determine the redshift of the burst to be z = 4.615. We find that the optical afterglow of GRB 220101A is one of the most luminous ever detected. Based on our optical and near-infrared data, and combined with the X-ray observations, we perform a multiband fit with the Python package afterglowpy . The jet opening angle is constrained to ∼3.°4, which is consistent with the jet-break time at ∼0.7 day. We also determine the circumburst density of n _0 = 0.15 cm ^−3 and kinetic energy E _K,iso = 3.5 × 10 ^54 erg. In the prompt phase of the burst, we find a “mirror” feature in the lightcurve from 80 s to 120 s. The physical origin of such a mirror feature is unclear.

The viral genus Henipavirus includes two highly virulent zoonotic viruses of serious public health concern. Hendra henipavirus and Nipah henipavirus outbreaks are restricted to Australia and Southeast Asia, respectively. The Henipavirus genus comprises mostly bat-borne viruses, but exceptions have already been described as novel viruses with rodents and shrews as reservoir animals. In the Americas, scarce evidence supports the circulation of these viruses. In this communication, we report a novel henipa-like virus from opossums (Marmosa demerarae) from a forest fragment area in the Peixe-Boi municipality, Brazil, after which the virus was named the Peixe-Boi virus (PBV). The application of next-generation sequencing and metagenomic approach led us to discover the original evidence of a henipa-like virus genome in Brazil and South America and the original description of a henipa-like virus in marsupial species. These findings emphasize the importance of further studies to characterize PBV and clarify its ecology, impact on public health, and its relationship with didelphid marsupials and henipaviruses.

The COVID-19 pandemic is the biggest public health threat facing the world today. Multiple vaccines have been approved; however, the emergence of viral variants such as the recent Omicron raises the possibility of booster doses to achieve adequate protection. In Brazil, the CoronaVac (Sinovac, Beijing, China) vaccine was used; however, it is important to assess the immune response to this vaccine over time. This study aimed to monitor the anti-SARS-CoV-2 antibody responses in those immunized with CoronaVac and SARS-CoV-2 infected individuals. Samples were collected between August 2020 and August 2021. Within the vaccinated cohort, some individuals had a history of infection by SARS-CoV-2 prior to immunization, while others did not. We analyzed RBD-specific and neutralizing-antibodies. Anti-RBD antibodies were detected in both cohorts, with a peak between 45–90 days post infection or vaccination, followed by a steady decline over time. In those with a previous history of COVID-19, a higher, longer, more persistent response was observed. This trend was mirrored in the neutralization assays, where infection, followed by immunization, resulted in higher, longer lasting responses which were conditioned on the presence of levels of RBD antibodies right before the vaccination. This supports the necessity of booster doses of CoronaVac in due course to prevent serious disease.

Abstract Quinoa, classified as a pseudocereal, presents greater nutritional value compared to traditional cereals. Considering the potential for cultivation presented by the species and the benefits of studying plant morphology and morphobiometry, this paper describes seed and seedling morphobiometric characteristics of quinoa (Chenopodium quinoa Willd.) cultivar BRS Piabiru during germination and emergence. To evaluate seed morphobiometry the 1000-seeds weight, moisture content, seed morphological characterization and the tetrazolium test were performed. The morphological characterization of germination and seedling emergence were performed by periodic observations during the development, allowing the description using pictures and drawings. Quinoa seeds cv. BRS Piabiru present an average diameter of 2.05 mm and 1.07 width. 1000-seeds weight of 2.68 g and moisture content of 11%. Externally, quinoa seeds present the pericarp as testa, the hilum and the raphe and, internally, the embryonic axis (cotyledons, radicle and hypocotyl-radicle), perisperm and endosperm. The germination is characterized as epigeal, phanerocotylar type, with radicle protrusion at 3 hours after sowing and complete formed seedlings at 24 hours after sowing. Emergence occurs at 9 days after sowing and plants are completely formed at 12 days after sowing.

As crossbreeding is extensively used in some livestock species, we aimed to evaluate the performance of single-step GBLUP (ssGBLUP) and weighted ssGBLUP (WssGBLUP) methods to predict Genomic Estimated Breeding Values (GEBVs) of crossbred animals. Different training population scenarios were evaluated: (SC1) ssGBLUP based on a single-trait model considering purebred and crossbred animals in a joint training population; (SC2) ssGBLUP based on a multiple-trait model to enable considering phenotypes recorded in purebred and crossbred training animals as different traits; (SC3) WssGBLUP based on a single-trait model considering purebred and crossbred animals jointly in the training population (both populations were used for SNP weights' estimation); (SC4) WssGBLUP based on a single-trait model considering only purebred animals in the training population (crossbred population only used for SNP weights' estimation); (SC5) WssGBLUP based on a single-trait model and the training population characterized by purebred animals (purebred population used for SNP weights' estimation). A complex trait was simulated assuming alternative genetic architectures. Different scaling factors to blend the inverse of the genomic (G−1) and pedigree (A22-1) relationship matrices were also tested. The predictive performance of each scenario was evaluated based on the validation accuracy and regression coefficient. The genetic correlations across simulated populations in the different scenarios ranged from moderate to high (0.71–0.99). The scenario mimicking a completely polygenic trait (hQTL2= 0) yielded the lowest validation accuracy (0.12; for SC3 and SC4). The simulated scenarios assuming 4,500 QTLs affecting the trait and hQTL2=h2 resulted in the greatest GEBV accuracies (0.47; for SC1 and SC2). The regression coefficients ranged from 0.28 (for SC3 assuming polygenic effect) to 1.27 (for SC2 considering 4,500 QTLs). In general, SC3 and SC5 resulted in inflated GEBVs, whereas other scenarios yielded deflated GEBVs. The scaling factors used to combine G−1 and A22-1 had a small influence on the validation accuracies, but a greater effect on the regression coefficients. Due to the complexity of multiple-trait models and WssGBLUP analyses, and a similar predictive performance across the methods evaluated, SC1 is recommended for genomic evaluation in crossbred populations with similar genetic structures [moderate-to-high (0.71–0.99) genetic correlations between purebred and crossbred populations].

Coral reefs are threatened by climate change on a global scale with thermal stress events and mass coral bleaching being widely reported. The reefs off the east coast of Brazil (and other turbid areas) have, however, historically escaped such thermal stress events, with relatively low levels of background coral mortality (5–10%). This has recently changed. Here we show that, in 2019, degree heating weeks (DHW) of 19.65 coincided with catastrophic declines in coral cover, especially in the major reef building hydrocoral Millepora alcicornis. The decline was due to bleaching associated with exposure to high temperature stress culminating in DHW values exceeding 15 for a period of 50 days. At two independent sites, surveys showed upwards of 83.5 ± 9.0 and 89.1 ± 3.9% mortality, and a third site showed relatively lower (albeit still high) mortality rates of 43.3 ± 12.0%. The mass die-off in 2019 is unprecedented in the South Atlantic reefs and coincides with increased heating events.

Abstract Background In recent years, there has been increased interest in the study of the molecular processes that affect semen traits. In this study, our aim was to identify quantitative trait loci (QTL) regions associated with four semen traits (motility, progressive motility, number of sperm cells per ejaculate and total morphological defects) in two commercial pig lines (L1: Large White type and L2: Landrace type). Since the number of animals with both phenotypes and genotypes was relatively small in our dataset, we conducted a weighted single-step genome-wide association study, which also allows unequal variances for single nucleotide polymorphisms. In addition, our aim was also to identify candidate genes within QTL regions that explained the highest proportions of genetic variance. Subsequently, we performed gene network analyses to investigate the biological processes shared by genes that were identified for the same semen traits across lines. Results We identified QTL regions that explained up to 10.8% of the genetic variance of the semen traits on 12 chromosomes in L1 and 11 chromosomes in L2. Sixteen QTL regions in L1 and six QTL regions in L2 were associated with two or more traits within the population. Candidate genes SCN8A, PTGS2, PLA2G4A, DNAI2, IQCG and LOC102167830 were identified in L1 and NME5, AZIN2, SPATA7, METTL3 and HPGDS in L2. No regions overlapped between these two lines. However, the gene network analysis for progressive motility revealed two genes in L1 (PLA2G4A and PTGS2) and one gene in L2 (HPGDS) that were involved in two biological processes i.e. eicosanoid biosynthesis and arachidonic acid metabolism. PTGS2 and HPGDS were also involved in the cyclooxygenase pathway. Conclusions We identified several QTL regions associated with semen traits in two pig lines, which confirms the assumption of a complex genetic determinism for these traits. A large part of the genetic variance of the semen traits under study was explained by different genes in the two evaluated lines. Nevertheless, the gene network analysis revealed candidate genes that are involved in shared biological pathways that occur in mammalian testes, in both lines.

The Asian lineage of Zika virus (ZIKV) has acquired a mutation in NS1 that enhances mosquito infection. Here, Xia et al. show that the same mutation interferes with interferon production through interaction with TBK1 and affects ZIKV replication in mice.

Reporter virus neutralization test (RVNT) has been used as an alternative to the more laborious and time-demanding conventional PRNT assay for both DENV and ZIKV. However, few studies have investigated how these techniques would perform in epidemic areas with the circulation of multiple flavivirus. Here, we evaluate the performance of ZIKV and DENV Rluc RVNT and ZIKV mCh RVNT assays in comparison to the conventional PRNT assay against patient sera collected before and during ZIKV outbreak in Brazil. These samples were categorized into groups based on (1) acute and convalescent samples according to the time of disease, and (2) laboratorial diagnostic results (DENV and ZIKV RT-PCR and IgM-capture ELISA). Our results showed that DENV Rluc assay presented 100% and 78.3% sensitivity and specificity, respectively, with 93.3% accuracy, a similar performance to the traditional PRNT. ZIKV RVNT90, on the other hand, showed much better ZIKV antibody detection performance (around nine-fold higher) when compared to PRNT, with 88% clinical sensitivity. Specificity values were on average 76.8%. Even with these results, however, ZIKV RVNT90 alone was not able to reach a final diagnostic conclusion for secondary infection in human samples due to flavivirus cross reaction. As such, in regions where the flavivirus differential diagnosis represents a challenge, we suggest the establishment of a RVNT panel including other flaviviruses circulating in the region, associated with the other serological techniques such as IgM ELISA and the investigation of seroconversion, in order to help define an accurate diagnostic conclusion using serology.

From 2016 to 2018, Brazil faced the biggest yellow fever (YF) outbreak in the last 80 years, representing a risk of YF reurbanization, especially in megacities. Along with this challenge, the mass administration of the fractionated YF vaccine dose in a naïve population brought another concern: the possibility to increase YF adverse events associated with viscerotropic (YEL-AVD) or neurological disease (YEL-AND). For this reason, we developed a quantitative real time RT-PCR (RT-qPCR) assay based on a duplex TaqMan protocol to distinguish broad-spectrum infections caused by wild-type yellow fever virus (YFV) strain from adverse events following immunization (AEFI) by 17DD strain during the vaccination campaign used to contain this outbreak. A rapid and more accurate RT-qPCR assay to diagnose YFV was established, being able to detect even different YFV genotypes and geographic strains that circulate in Central and South America. Moreover, after testing around 1400 samples from human cases, non-human primates and mosquitoes, we detected just two YEL-AVD cases, confirmed by sequencing, during the massive vaccination in Brazilian Southeast region, showing lower incidence than AEFI as expected.

Since pre- and postnatal development are programmed during early prenatal life, studies addressing the complete transcriptional landscape during organogenesis are needed. Therefore, we aimed to disentangle differentially expressed (DE) genes between fetuses (at 35 days old) and embryos (at 25 days old) through RNA-sequencing analysis using the pig as model. In total, 1705 genes were DE, including the top DE IBSP, COL6A6, HBE1, HBZ, HBB, and NEUROD6 genes, which are associated with developmental transition from embryos to fetuses, such as ossification, skeletal muscle development, extracellular matrix organization, cardiovascular system, erythrocyte differentiation, and neuronal system. In pathway analysis, embryonic development highlighted those mainly related to morphogenic signaling and cell interactions, which are crucial for transcriptional control during the establishment of the main organs in early prenatal development, while pathways related to myogenesis, neuronal development, and cardiac and striated muscle contraction were enriched for fetal development, according to the greater complexity of organs and body structures at this developmental stage. Our findings provide an exploratory and informative transcriptional landscape of pig organogenesis, which might contribute to further studies addressing specific developmental events in pigs and in other mammals.

Zika virus infection can result in congenital disorders and cause disease in adults, and there is currently no approved vaccine. Here Shan et al. show that a single dose of a live-attenuated Zika vaccine prevents infection, testis damage and transmission to the fetus during pregnancy in different animal models.

Background and rationale. Epidemics of hepatitis B and C are a public health burden, and their prevalence in Brazil varies among regions. We determined the prevalence of hepatitis markers in an urban university population in order to support the development of a comprehensive program for HBV immunization and HBV/HCV diagnosis. Students, employees, and visitors (n = 2,936, 31 years IQR 24.5-50, female = 69.0% and 81.1% with at least 12 years of education) were enrolled from May to November 2013. Antibodies against hepatitis B surface antigen (anti-HBs), against hepatitis B core antigen (anti-HBc), and hepatitis B surface antigen (HBsAg) were detected with enzyme immunoassays and anti-hepatitis C virus (anti-HCV) antibodies with a chemiluminescence immunoassay. The results were confirmed with polymerase chain reaction for HCV and nucleic acid amplification test for hepatitis B virus (HBV).Results. The overall prevalence of markers among the participants was 0.136% (95% confidence interval [CI]: 0.003-0.270) for HBsAg, 6.44% (95% CI: 5.55-7.33%) for anti-HBc, 50.8% (95% CI: 48.9-52.7%) for anti-HBs > 10 mIU/mL, and 0.44% (95% CI: 0.20-0.68) for anti-HCV. Almost 30.4% had anti-HBs titers > 100 mIU/mL. Participants with a detectable HCV viral load (n = 9) were infected with genotype 1a.Conclusions. In an urban university population, in which 80% of participants had > 11 years of education, prevalence increased with age, and self-declared ethnicity for anti-HBc and with age, marital status and professional activity for anti-HCV antibodies. A periodical offer of HCV rapid testing should be implemented, and HBsAg rapid testing should be offered to individuals above 20 years of age.

Bacterial cellulose produced from Gluconacetobacter xilinus was used to produce cellulose nanocrystals by sulfuric acid hydrolysis. Hydrolysis was performed with 64% sulfuric acid at 50 ºC with the hydrolysis time ranging between 5 and 90 min. The production of nanocrystals was observed to have size distributions that were dependent on hydrolysis times up to 10 min, after which time the suspensions showed distributions closer in size. Results from thermal analysis and X-ray diffraction showed that the amorphous cellulose was removed, leaving only the crystalline portion. Self-supported films were formed from the suspension of nanocrystals and had iridescence characteristics. The films were characterized by microscopy measures and specular reflectance.

This work presents a new method for assessing global ionospheric maps (GIM) using ionosonde data. The method is based on the critical frequency at the F2 layer directly measured by ionosondes to validate VTEC (vertical total electron content) values from GIMs. The analysis considered four different approaches to using foF2. The study was performed over one of the most challenging scenarios, the Brazilian region, considering four ionosondes (combined in six pairs) and thirteen GIM products available at CDDIS (Crustal Dynamics Data Information System). Analysis was conducted using daily, weekly, one year (2015), and four years (2014–2017) of data. Additional information from the ionosphere was estimated to complement the daily analysis, such as slab thickness and shape function peak. Results indicated that slab thickness and shape function peak could be used as alternative indicators of periods and regions where this method could be applied. The weekly analysis indicated the squared frequency ratio with local time correction as the best approach of using foF2, between the ones evaluated. The analysis of one-year data (2015) was performed considering thirteen GIMs, where CODG and UQRG were the two GIMs that presented the best performance. The four-year time series (2014–2017) were analyzed considering these two products. Regional and temporal ionospheric influences could be noticed in the results, with expected larger errors during the solar cycle peak in 2014 and at locations with pairs of ionosondes with the larger distance apart. Therefore, we have confirmed the viability of the developed approach as an assessment method to analyze GIMs quality based on ionosonde data.

The present work employs the so-called Evolutionary Polynomial Regression (EPR) algorithm to build up a formula for the assessment of mean wave overtopping discharge for smooth sea dikes and vertical walls. EPR is a data-mining tool that combines and integrates numerical regression and genetic programming. This technique is here employed to dig into the relationship between the mean discharge and main hydraulic and structural parameters that characterize the problem under study. The parameters are chosen based on the existing and most used semi-empirical formulas for wave overtopping assessment. Besides the structural freeboard or local wave height, the unified models highlight the importance of local water depth and wave period in combination with foreshore slope and dike slope on the overtopping phenomena, which are combined in a unique parameter that is defined either as equivalent or imaginary slope. The obtained models aim to represent a trade-off between accuracy and parsimony. The final formula is simple but can be employed for a preliminary assessment of overtopping rates, covering the full range of dike slopes, from mild to vertical walls, and of water depths from the shoreline to deep water, including structures with emergent toes.

RESUMO Este estudo teve por objetivo caracterizar a diversidade carcinológica de dois manguezais (igarapés Buenos Aires e Tronco) da Baía de São Marcos, na costa amazônica maranhense, Brasil. Foram realizadas quatro coletas trimestrais entre setembro de 2011 a junho de 2012. Em cada coleta foram analisados três pontos por área (zona 1, zona 2 e zona 3), totalizando 24 amostras. O material biológico foi coletado por meio de arrastos com rede do tipo puçá, catação manual e técnica de braceamento. Paralelamente à coleta do material biológico, foram verificados a salinidade, temperatura e oxigênio dissolvido de cada área analisada. Para avaliar a similaridade de agrupamento entre as zonas dos manguezais foi aplicada a análise de cluster e consecutiva elaboração de dendrogramas. Foi coletado um total de 873 indivíduos, representando nove famílias e 21 espécies, das quais Ocypodidae e Penaeidae foram as mais abundantes. Em relação à distribuição espacial, percebe-se que Clibanarius vittatus (Bosc, 1802), Clibanarius tricolor (Gibbes, 1850), Clibanarius foresti Holthuis, 1959 e Uca maracoani (Latreille, 1802) se restringiram à primeira zona dos dois manguezais, enquanto outras dez espécies foram observadas por todo o manguezal, o que pode estar intimamente relacionado ao seu hábito de vida. De um modo geral, o igarapé Buenos Aires apresentou maior número de espécies em relação ao igarapé Tronco, no entanto, existe grande similaridade faunística de crustáceos decápodes entre as duas áreas amostradas.

In state of Acre, most anuran research has been carried out in areas of terra firme, whereas alluvial forests remain relatively unexplored. We document the richness, breeding environments and calling season of the anurans of the Lower Moa River in the Alto Juruá (state of Acre), a region considered to harbor high biodiversity. Sampling was conducted in two main areas with different forest typologies (Terra firme forest and Alluvial forest), totaling 256 ha. The diversity of anurans was analyzed using the Shannon-Wiener index, evenness and Berger - Parker dominance. The anurans were sampled from October 2008 to September 2009, using the following sampling methods: sampling at breeding sites, pitfall traps, time constrained visual search during the day (TCVSD); at night (NVTCS) and active search (AS). Fifty anuran species, belonging to eight families, were recorded: Aromobatidae (2), Bufonidae (5), Hemiphractidae (1), Hylidae (25), Leptodactylidae (9), Microhylidae (4), Ranidae (1) and Craugastoridae (3). Pitfall traps totaling 1440 day-buckets in the year. DVTCS had a total sampling effort 72 hours/person/year; NVTCS - 72 hours/person/year and AS - 24 hours/person/year. Greater richness (35 species) was recorded in the alluvial forest, low diversity index and high dominance (H' = 1.69, J' = 0.51, d = 0.62). Most likely, these values were influenced by the high relative abundance of Rhinella margaritifera in this area. The richness of the alluvial forest may be associated with environmental heterogeneity according with the theory of intermediate disturbances. The breeding activity of most species occurred in lentic environments (lakes and temporary ponds) and during the rainy season. When sampling anurans in the Amazon it is important to use two or more sampling method and to consider the different forest typologies of this biome for a better characterization of its richness.

ABSTRACT Compared with other flaviviruses, Zika virus (ZIKV) is uniquely associated with congenital diseases in pregnant women. One recent study reported that (i) ZIKV has higher thermostability than dengue virus (DENV [a flavivirus closely related to ZIKV]), which might contribute to the disease outcome; (ii) the higher thermostability of ZIKV could arise from an extended loop structure in domain III of the viral envelope (E) protein and an extra hydrogen-bond interaction between E molecules (V. A. Kostyuchenko, E. X. Y. Lim, S. Zhang, G. Fibriansah, T.-S. Ng, J. S. G. Ooi, J. Shi, and S.-M. Lok, Nature 533:425–428, 2016, https://doi.org/10.1038/nature17994 ). Here we report the functional analysis of the structural information in the context of complete ZIKV and DENV-2 virions. Swapping the prM-E genes between ZIKV and DENV-2 switched the thermostability of the chimeric viruses, identifying the prM-E proteins as the major determinants for virion thermostability. Shortening the extended loop of the E protein by 1 amino acid was lethal for ZIKV assembly/release. Mutations (Q350I and T351V) that abolished the extra hydrogen-bond interaction between the E proteins did not reduce ZIKV thermostability, indicating that the extra interaction does not increase the thermostability. Interestingly, mutant T351V was attenuated in A129 mice defective in type I interferon receptors, even though the virus retained the wild-type thermostability. Furthermore, we found that a chimeric ZIKV with the DENV-2 prM-E and a chimeric DENV-2 with the ZIKV prM-E were highly attenuated in A129 mice; these chimeric viruses were highly immunogenic and protective against DENV-2 and ZIKV challenge, respectively. These results indicate the potential of these chimeric viruses for vaccine development. IMPORTANCE Analysis of a recently observed high-resolution structure of ZIKV led to a hypothesis that its unusual stability may contribute to the associated, unique disease outcomes. Here we performed a functional analysis to demonstrate that viral prM-E genes are the main determinants for the high stability of ZIKV. The extra hydrogen-bond interaction (observed in the high-resolution structure) between ZIKV E proteins did not enhance virion stability, whereas the extended loop of E protein (CD loop in domain III) was essential for ZIKV assembly. More importantly, we found that a chimeric ZIKV with DENV-2 prM-E genes and a chimeric DENV-2 with ZIKV prM-E genes were highly attenuated in A129 mice. Mice immunized with these chimeric viruses generated robust neutralizing antibody responses and were fully protected from DENV-2 and ZIKV challenge, respectively, indicating that these chimeric viruses could be further developed as vaccine candidates.

To study the dynamics of wild rodent populations and identify potential hosts for hantavirus, we conducted an eco-epidemiologic study in Campo Novo do Parecis, Mato Grosso State, Brazil. We detected and genetically characterized Castelo dos Sonhos virus found in a species of pygmy rice rat (Oligoryzomys utiaritensis).

To confirm circulation of Anajatuba virus in Maranhão, Brazil, we conducted a serologic survey (immunoglobulin G ELISA) and phylogenetic studies (nucleocapsid gene sequences) of hantaviruses from wild rodents and persons with hantavirus pulmonary syndrome. This virus is transmitted by Oligoryzomys fornesi rodents and is responsible for hantavirus pulmonary syndrome in this region.

OBJETIVO: Detecção de oito vírus respiratórios mais comuns: vírus respiratório sincicial humano (VRSH), vírus influenza tipo A e B (IA e IB), vírus da parainfluenza 1, 2 e 3 (VPIH1, 2 e 3), adenovírus (Ad) e metapneumovírus humano (MPVH), a fim de estabelecer a etiologia das infecções respiratórias agudas (IRA) e a epidemiologia desses vírus em crianças pequenas atendidas no Hospital Universitário da Universidade de São Paulo, em São Paulo, Brasil, durante o ano de 2003. MÉTODOS: A vigilância epidemiológica foi realizada em todas as crianças menores de 5 anos hospitalizadas por causa de doenças do trato respiratório inferior (DTRI) entre 1º de janeiro de 2003 e 20 de dezembro de 2003, no hospital universitário. Amostras coletadas de nasofaringe foram analisadas quanto à presença de vírus respiratórios através da reação em cadeia da polimerase e detectadas pelo programa GeneScan. RESULTADOS: Das 336 amostras coletadas, 187 (55,6%) foram positivas para pelo menos um dos vírus respiratórios estudados. De todas as crianças, o VRSH foi identificado em 24,1%, o MPVH em 17,8%, o VPIH3 em 8,3%, o Ad em 6,8%, o IA em 5%, o VPIH1 em 0,6%, sendo que nenhum vírus foi detectado em 44,1%. Infecções virais duplas foram detectadas em 7,1% de todas as amostras (12,8% das amostras positivas). O VPIH2 e o IB não foram detectados no presente estudo. CONCLUSÕES: Este estudo confirma que as crianças menores de 5 anos, e especialmente aquelas menores de 1 ano, apresentam uma alta taxa de hospitalização devido aos seguintes vírus: VRSH, MPVH, VPIH, influenza e adenovírus. Foi possível determinar a etiologia e epidemiologia da maioria das IRAs e traçar o perfil de sazonalidade dos vírus respiratórios mais comuns entre as crianças pequenas.OBJECTIVE: Detection of the eight most common respiratory viruses: Human respiratory syncytial virus (HRSV), influenza virus A and B (IA and IB), parainfluenza viruses 1, 2 and 3 (HPIV1, 2 and 3), adenovirus (Ad) and human metapneumovirus (HMPV), in order to establish the etiology of acute respiratory infections (ARIs) and the epidemiology of these viruses in young children seen at Hospital Universitário, Universidade de São Paulo, in São Paulo, Brazil, during 2003. METHODS: The epidemiological surveillance was conducted in all children younger than 5 years hospitalized at the Hospital for lower respiratory tract infections (LRTI) from January 1, 2003 to December 30, 2003. Nasal and throat samples were scanned for respiratory viruses by polymerase chain reaction and detected by the GeneScan assay. RESULTS: Of 336 samples collected from 336 patients, 187 (55.6%) were positive for at least one of the respiratory viruses studied. Of all the children, HRSV was identified in 24.1%, HMPV in 17.8%, HPIV3 in 8.3%, Ad in 6.8%, IA in 5%, HPIV1 in 0.6%, but no virus could be detected in 44.1%. Dual virus infections were detected in 7.1% of all samples (12.8% of positive samples). HPIV2 and IB were not detected in the present study. CONCLUSIONS: This study confirms that children younger than 5 years and particularly younger than 1 year have a high hospitalization rate due to HRSV, HMPV, HPIV, influenza and adenovirus. We were able to determine the etiology and epidemiology of most ARIs and trace the seasonal profile of the commonest respiratory viruses among young children.

Contexto: O processo de envelhecimento está associado às doenças que podem comprometer o bom funcionamento do sistema nervoso central. Objetivo: Avaliar característica de declínio cognitivo e prevalência de sintomas depressivos e sua associação com dados sociodemográficos de idosos institucionalizados em três instituições de longa permanência do sul de Santa Catarina. Métodos: Estudo transversal, avaliando todos idosos institucionalizados em Tubarão e Laguna/SC. Participaram da presente pesquisa 66 idosos, com idade maior ou igual a sessenta anos, de ambos os gêneros. Para coleta de dados utilizou-se um questionário padronizado abordando dados sociodemográficos, a Escala de Depressão Geriátrica reduzida (EDG-15) e o Mini-Exame do Estado Mental. Resultados: A população estudada era predominantemente feminina, viúva e média de idade 77,3±9,9anos. Sintomas depressivos estiveram presentes em 62,1% dos idosos. Possível déficit cognitivo esteve presente em 37,87% da população. Conclusão: Alta prevalência de sintomas depressivos e déficit cognitivo estiveram presentes nos idosos residentes nas instituições de longa permanência pesquisadas. Diante de tais fatos, o serviço de saúde do Brasil deve estar preparado para avaliar e tratar tais pacientes, a fim de melhorar a qualidade de vida dos idosos institucionalizados.

Background. Dietary pattern plays a causative role in the rising of noncommunicable diseases. The SESI (Serviço Social da Indústria) study was designed to evaluate risk factors for noncommunicable diseases. We aimed to describe food items consumed by Brazilian workers and to assess their association with socioeconomic status. Methods. Cross-sectional study was carried out among Brazilian industrial workers, selected by multistage sampling, from 157 companies. Interviews were conducted at the work place using standardized forms. Results. 4818 workers were interviewed, aged 35.4±10.7 years, 76.5% were men. The workers had an average of 8.7±4.1 years of schooling and 25.4±4.1 kg/m2 of BMI. Men and individuals with less than high school education were less likely to consume dairy products, fruits, and vegetables daily, even after control for confounding factors. Men consumed rice and beans daily more often than women. In comparison to workers aged 50–76 years, those under 30 years old consumed less fruits and green leafy vegetables daily. Conclusion. The food items consumed by Brazilian workers show that there are insufficient consumption according to the guidelines of healthy foods, particularly of dairy products, vegetables, and fruits.

This systematic review aimed to identify the level of impact of educational strategies for teaching tooth carving on the carving ability of undergraduate dental students. The PubMed-NCBI, Cochrane-CENTRAL, LILACS, Ibecs, BBO, Web of Science, and Scopus databases were searched in May 2019, for randomized controlled trials (RCTs) and two-arm non-randomized studies of interventions (NRSI) addressing educational interventions toward the dental carving of undergraduate students. Studies from the year 2000 until the search date, written in English, Portuguese, and Spanish were included. Study screening and data extraction were performed in duplicate and blinded. The data were presented narratively, considering the dental carving ability of students the primary outcome. The risk of bias was assessed using the RoB tool 2.0 and ROBINS-I, and the level of evidence was determined with GRADE. Of 3,574 studies, 6 were included, with 3 RCTs and 3 NRSIs. Very low level of evidence was provided from the NRSIs that flipped classroom (1 study; n = 140) and a student-driven revised module (1 study; n = 264) improved the carving ability of students. Additionally, there was moderate evidence of online complementary material (1 RCT; n = 30) and reinforcement class improving the carving ability of students (1 RCT; n = 29). The replacement of traditional classes by an instructional DVD (1 RCT; n = 73) and assessment of carving projects through digital systems (1 NRSI; n = 79) did not enhance the carving ability of students. Study design, risk of bias, and imprecision downgraded the level of evidence. There was a very low to moderate evidence on the effectiveness of student-driven educational approaches and complementary classes of dental anatomy in improving the dental carving ability of students.

This study aimed to investigate the effect of extraction methods and solvents (Soxhlet and supercritical fluid extraction, SFE) on the chemical profile (gas chromatography [GC]/mass spectrometry and GC/flame ionization detection) and antimicrobial, antioxidant, antitumor, and anti‐inflammatory activities of quinoa Piabiru (QP) husk crude extracts. Soxhlet was applied using 100% water (QPSH2O), and ethanolic solutions of 50% ethanol (ethanol:water 50:50 v/v, QPSetOH50), 70% ethanol (QPSetOH70), and 99% ethanol (QPSetOH99) as solvents. SFE was applied using CO2 and n‐propane as solvents (QPSF). QPSH2O extract showed a higher concentration of phytosterols (stigmasterol, β‐sitosterol, 7,8‐epoxylanostan‐11‐ol,3‐acetoxy) and carotenoids (rhodopin) and oleic acid, displaying intermediate thiobarbituric acid reactive substance (TBARS) antioxidant activity. Intermediate concentrations of ethanol (QPSetOH50 and QPSetOH70 extracts) increased the extraction yields and the antibacterial activity of the extracts (Pseudomonas aeruginosa, Salmonella enterica, Bacillus cereus, and Staphylococcus aureus). Higher concentrations of ethanol (QPSetOH99 extract) contributed to increased antioxidant activity as assessed by TBARS and higher recoveries of 4‐(allyloxy)‐2‐methyl‐2‐pentanol, nonadecane, and lauric, myristic, palmitic, linoleic, stearic, arachidic, behenic, and lignoceric acids. Finally, the QPSF extract presented higher antioxidant activity by DPPH, ABTS, and ferric‐reducing antioxidant power, higher content of 5‐methoxy‐2‐pentanone, 5‐methoxy‐2‐methyl‐2‐pentanol, 1‐(1,3‐dimethylbutoxy)‐2‐propanol, oxalic, undecanoic, myristoleic, tricosanoic, pentadecanoic, elaidic, 11‐eicosenoic, and erucic acids, and better antifungal activity against Aspergillus brasiliensis than the other extracts. Crude extracts were not cytotoxic against non‐tumor cells (Vero) and did not show antitumor or anti‐inflammatory activities. Thus, antagonistic or synergistic effects of the phytochemical profile of quinoa husk crude extracts may present potential food and pharmaceutical applications. Practical Applications: The exceptional nutritional properties of quinoa seeds have boosted their cultivation in more than 123 countries. However, quinoa husks are generally considered waste. This study, which is of utmost importance, demonstrates the potential of extracting bioactive compounds from quinoa husks via pressurized fluids, turning them into a health‐promoting co‐product. This approach could minimize the current shortage of new antibiotics, antifungals, antitumor agents, and anti‐inflammatory substances in the pharmaceutical and food sectors. By converting quinoa husks into valuable bioactive extracts, we contribute significantly to developing effective natural compounds, underlining the significance of our collective work.
