The China Spallation Neutron Source is an accelerator-based user facility for neutron, proton, muon applications and so on. The designed proton beam power is 100 kW for the first phase. Overall, the scope of accelerator control system covers four parts, the global systems, the device layer local controls and remote controls, the high-level applications and services and the integration of conventional facilities. The principle guideline is to adopt as many as possible matured technologies and commercial products in the design and implementation. The control system is based on Experimental Physics and Industrial Control System. VME64x bus is chosen for timing system and fast protection system. PLCs and embedded industrial computers are used in the device layer slow controls. Control System Studio is adopted as the high-level applications. Blade server-based virtualization platform is deployed for the computer system. The accelerator control system was completed in September 2017 and then transited to routine operations. The design, implementation and operation of the accelerator control system are described in this paper.

In this study, we analyzed the role of mammalian STE20-like protein kinase 2 (Mst2), a serine-threonine protein kinase, in Lipopolysaccharides (LPS)-mediated inflammation and apoptosis in the H9C2 cardiomyocytes. Mst2 mRNA and protein levels were significantly upregulated in the LPS-treated H9C2 cardiomyocytes. LPS treatment induced expression of IL-2, IL-8, and MMP9 mRNA and proteins in the H9C2 cardiomyocytes, and this was accompanied by increased caspase-3/9 mediating H9C2 cardiomyocyte apoptosis. LPS treatment also increased mitochondrial reactive oxygen species (ROS) and the levels of antioxidant enzymes, such as GSH, SOD, and GPX, in the H9C2 cardiomyocytes. The LPS-treated H9C2 cardiomyocytes showed lower cellular ATP levels and mitochondrial state-3/4 respiration but increased mitochondrial fragmentation, including upregulation of the mitochondrial fission genes Drp1, Mff, and Fis1. LPS-induced inflammation, mitochondrial ROS, mitochondrial fission, and apoptosis were all significantly suppressed by pre-treating the H9C2 cardiomyocytes with the Mst2 inhibitor, XMU-MP1. However, the beneficial effects of Mst2 inhibition by XMU-MP1 were abolished by carbonyl cyanide-4-(trifluoromethoxy) phenylhydrazone (FCCP), a potent activator of mitochondrial fission. These findings demonstrate that Mst2 mediates LPS-induced cardiomyocyte inflammation and apoptosis by increasing mitochondrial fission.

The China Spallation Neutron Source (CSNS) accelerator consists of an 80-MeV H-linac, a 1.6-GeV rapid cycling synchrotron (RCS) and two beam transport lines. The designed proton beam power is 100 kW in Phase-I. The CSNS accelerator can operate in different beam modes and beam destinations, and for each beam destination, the beam power must be strictly restricted; meanwhile, the linac and the RCS may operate independently during the commissioning stage. To satisfy these operation requirements, the run management system (RMS) was designed and deployed for CSNS. The run management system, including a hardware layer and a software layer, operates on the top level of the global control system. RMS coordinates the machine protection system, personnel protection system, timing system and other systems, to set and publish the machine states and machine modes, to verify beam parameters and to permit the beam extraction from ion source. The RMS has been operating for more than one and half years and has played an important role during the commissioning and operation of CSNS facility. This paper will introduce the design and implementation of the RMS for CSNS.

Purpose Emerging evidence has demonstrated that gut microbiome plays essential roles in the pathogenesis of human diseases in distal organs. Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by the progressive loss of motor neurons. Treatment with the only drug approved by the US Food and Drug Administration for use in ALS, riluzole, extends a patient׳s life span by only a few months. Thus, there is an urgent need to develop novel interventions that for alleviate disease progression and improve quality of life in patients with ALS. Here we present evidence that intestinal dysfunction and dysbiosis may actively contribute to ALS pathophysiology. Methods We used G93A transgenic mice as a model of human ALS. The G93A mice show abnormal intestinal microbiome and damaged tight junctions before ALS disease onset. The mice were given 2% butyrate, a natural bacterial product, in the drinking water. Results In mice fed with butyrate, intestinal microbial homeostasis was restored, gut integrity was improved, and life span was prolonged compared with those in control mice. At the cellular level, abnormal Paneth cells—specialized intestinal epithelial cells that regulate the host–bacterial interactions—were significantly decreased in the ALS mice treated with butyrate. In both ALS mice and intestinal epithelial cells cultured from humans, butyrate treatment was associated with decreased aggregation of the G93A superoxide dismutase 1 mutated protein. Implications The findings from this study highlight the complex role of the gut microbiome and intestinal epithelium in the progression of ALS and present butyrate as a potential therapeutic reagent for restoring ALS-related dysbiosis.

Background Axin1 is a scaffold protein in the β-catenin destruction complex, which, if disrupted, contributes to pathogenesis of various human diseases, including colorectal carcinogenesis and inflammatory bowel diseases (IBD). We have previously demonstrated that Salmonella infection promotes the degradation and plasma sequestration of Axin1, leading to bacterial invasiveness and inflammatory responses. Vitamin D and the vitamin D receptor (VDR) appear to be important regulators of IBD and colon cancer. Although VDR and Axin1 are all involved in intestinal inflammation, it remains unclear whether these processes are related or function independently. In the current study, we hypothesize that VDR is an important regulator for the maintenance of physiological level of Axin1. Methods Using the intestinal epithelial conditional VDR knockout mouse model (VDR ΔIEC ) and cultured cell lines, influences of VDR status on the expression of Axin1 was evaluated by Western blots and real-time PCR. Loss- and gain-of-function assays were used to investigate the regulation of VDR on Axin1 at the transcriptional and translational levels. Cells were treated with cycloheximide or actinomycin for molecular mechanistic studies. Candidate genomic VDR binding sites for Axin1 were tested by chromatin immunoprecipitation (ChIP) assay. Physical interactions among VDR, Axin1, and β-catenin were tested by immunoprecipitation. Cellular localization of Axin1 with different VDR status was determined by fractionation and immunohistochemistry. Results We found that VDR deletion led to lower protein and mRNA levels of Axin1 , whereas knockdown of Axin1 did not change the expression level of VDR protein. Immunoprecipitation data did not support physical interaction between VDR and Axin1. The VDR regulation of Axin1 was through a VDR genomic binding site for Axin1 gene on the regulatory region. Fractionation data showed that cytosolic Axin1 was significantly reduced due to VDR deletion, leaving the nuclear fraction unchanged. In ileum, Axin1 was distributed in the cytosol of apical epithelium and crypts. Conclusion VDR is important for the maintenance of physiological level of Axin1. The discovery of Axin1 as a VDR target gene provides novel and fundamental insights into the interactions between the VDR and β-catenin signaling pathways.

Based on a sample of 1.31 billion J/psi events collected with the BESIII detector, we report the study of the doubly radiative decay eta' -> gamma gamma pi(0) for the first time, where the eta' meson is produced via the J/psi -> gamma eta' decay. The branching fraction of eta' -> gamma gamma pi(0) inclusive decay is measured to be B(eta' -> gamma gamma pi(0))(Incl) = (3.20 +/- 0.07(stat) +/- 0.23(sys)) x 10(-3), while the branching fractions of the dominant process eta' -> gamma omega and the non-resonant component are determined to be B(eta' -> gamma omega) x B(omega -> gamma pi(0)) = (23.7 +/- 1.4(stat) +/- 1.8(sys)) x 10(-4) and B(eta' -> gamma gamma pi(0))(NR) = (6.16 +/- 0.64(stat) +/- 0.67(sys)) x 10(-4), respectively. In addition, the M-gamma gamma(2)-dependent partial widths of the inclusive decay are also presented.

Adipose-derived stem cells/adipose-derived progenitor cells (ADPCs) are multipotent stem cells that can differentiate in vitro into many cell types. However, the vast majority of experimental materials were obtained from human, mouse, rabbit, and other mammals but rarely from poultry. In this study, ADPCs were isolated from 1-day-old chicks. Primary ADPCs were subcultured to passage 15. The surface markers of ADPCs, CD29, CD44, CD71, and CD73, were detected by immunofluorescence and RT-polymerase chain reaction assays. The growth curves of different passages were all typically sigmoidal. In addition, ADPCs of different passages were successfully induced to differentiate into osteoblasts, adipocytes, and myocardial cells. The results suggest that the ADPCs isolated from chicken possess similar biological characteristics with those derived from other species, and their multilineage differentiation provides many potential applications.

The aim of this work was to seek a different approach to conserving important domestic animals in imminent danger. A Qingyuan partridge chicken embryonic fibroblast line, containing 336 cryovials with 8x10(6) cells each, was successfully established from 60 Qingyuan partridge chicken embryos using explant culture and cryopreservation techniques. The cells were morphologically consistent with fibroblasts. The growth curve was a typical "S" shape having a detention phase, a logarithmic phase, and a plateau phase. The population doubling time was approximately 72 h. Tests for bacteria, fungi, viruses, and Mycoplasma were all negative. Isoenzyme polymorphism indicated that the genetic characteristics of the cell line were stable in vitro. Karyotyping analysis suggested that the chromosome number of a normal cell was 2n=78 and 93.4% of the entire population was diploid. The transfection efficiencies of 6 fluorescent proteins (pEGFP-C1, pEGFP-N3, pDsRed-N1, pEYFP-N1, pECFP-N1, and pECFP-mito) optimal at 48 h were from 15.1 to 39.8%. The cell line met all of the criteria from the American Type Culture Collection. Not only has the germline of this important chicken breed been preserved at the cell level, but also valuable material has been provided for genome, postgenome, and somacloning research. Moreover, the establishment of this technical platform may provide both technical and theoretical support for storing the genetic resources of other animals and poultry at the cell level.

Y chromosome acts as a single nonrecombining unit that is male specific and in effect haploid, thus ensuring the preservation of mutational events as a single haplotype via male lines. In this study, 6 Y chromosome-specifc micro-satellites (SSR) were tested for the patrilineal genetic variations of 573 male samples from Chinese domestic horse (30 breeds), Przewalski's horse, and donkey. All the 6 loci appeared as a haplotype block in Przewalski's horse and the domestic donkey. There were notable differences, however, at Y chromosome markers between horse and donkey. There were 2 haplotypes of Eca. YA16 in the domestic horse breeds, Haplotype A (Allele A: 156 bp) and Haplotype B (Allele B: 152 bp). Allele A was the common allele among 30 horse breeds, and Allele B was found in I I horse breeds. This is the first description of a Y chromosome variant for horses. The 2 haplotypes of Y chromosome discovered in the domestic horse breeds in China could be helpful in unveiling their intricate genetic genealogy.

Up to 32 Landrace ear marginal tissue samples were used for establishing a fibroblast cell bank by the means of primary explantation and cryopreservation. Biological analysis suggested that the Population Doubling Time (PDT) of the cell line was approximately 24h. The diploid accounted for 97.2% of the whole population; isozyme analysis of Lactic Dehydrogenase (LDH) and Malic Dehydrogenase(MDH) disproved cross-contamination from other cell lines. The results of bacterium, fungus, virus and mycoplasma tests were all negative. The transfection rates of three fluorescent proteins were high, indicating that the exogenous genes could be effectively expressed in the cells. It had not only preserved the precious germplasm resource of the Landrace on the cell level but also provided valuable material for the researches of genomics, postgenomics, somatic cloning and so on.

Abstract For preserving the genetic resources of the Chinese Game chicken and exploring a new method to preserve endangered animals, a Chinese Game chicken embryo fibroblast cell bank was established by the attachment culture method and the cryopreservation biotechniques. This bank includes 43 embryo samples and has stocks of 178 cryovials, with each containing 4.0×106 cells respectively. Quality assays and biological characteristics of the bank established showed that the cells cultured in vitro were all typical fibroblast and the cell population doubling time (PDT) was about 2 days. The cell viability before freezing was 97.37% ± 2.25% and 91.43% ± 3.22% after thawing. Tests for microbial contamination by bacteria, fungus, viruses and mycoplasmas were all negative. The frequency of cells having the diploid number of chromosomes (78) was 94.65% ± 3.29%. Isoenzyme analysis confirmed that there was no cross-contamitation in the culture. The fluorescence could be observed well-distributed in cytoplasm and nucleus except for some cryptomere vesicles at 24 h after transfection, and the expression efficiency of pEGFP-C1, pEGFP-N3, pEYFP-N1, pECFP-N1, pECFP-mito and pDsRed1-N1 were all between 9.5%∼34.6% at 24 h, 48 h and 72 h after transfection. This research provided both technical and theoretical support for preserving the genetic resources of other animals and poultry at the cell level.

The Electromagnetic Calorimeter (EMC) sub-trigger system is one of the important parts of the trigger system of the BESIII spectrometer. Together with the conditions from the other sub-trigger systems, cluster-related and energy-related trigger conditions are provided to the Global Trigger system for effective selection of events interested and rejection of backgrounds. Total two types of PCB boards are designed to implement the whole functions, which are Trigger Cell and energy Block Adding (TCBA) and Energy Adding and Cluster Counting (EACC). Simulations and hardware arrangement of the EMC sub-trigger system are introduced in this paper.

Rare decay modes J / ψ → D s − π + + c.c. , J / ψ → D − π + + c.c. , and J / ψ → D ¯ 0 K ¯ 0 + c.c. are searched for using 5.77 × 10 7 J / ψ events collected with the BESII detector at the BEPC. No signal above background is observed. We present upper limits on the branching fractions of B ( J / ψ → D s − π + ) 1.3 × 10 −4 , B ( J / ψ → D − π + ) 7.5 × 10 −5 , and B ( J / ψ → D ¯ 0 K ¯ 0 ) 1.7 × 10 −4 at the 90% confidence level.

R values measured with the BESII detector at center-of-mass energies between 3.7 and 5.0 GeV are fitted to determine resonance parameters (mass, total width, electron width) of the high mass charmonium states, ψ ( 3770 ) , ψ ( 4040 ) , ψ ( 4160 ) and ψ ( 4415 ) . Various effects, including the interferences and relative phases between the resonances, the energy-dependence of the full widths, and the initial state radiative correction, are examined. The results are compared to previous studies.

The decays of J / ψ and ψ ( 2 S ) to n K S 0 Λ ¯ + c.c. are observed and measured for the first time, and the perturbative QCD “12%” rule is tested, based on 5.8 × 10 7 J / ψ and 1.4 × 10 7 ψ ( 2 S ) events collected with BESII detector at the Beijing Electron–Positron Collider. No obvious enhancement near n Λ ¯ threshold in J / ψ → n K S 0 Λ ¯ + c.c. is observed, and the upper limit on the branching ratio of J / ψ → K S 0 X , X → n Λ ¯ is determined.

With singly-tagged D ¯ samples selected from the data collected at and around 3.773 GeV with the BESII detector at the BEPC collider, we have measured the branching fractions for the inclusive K ± decays of D + and D 0 mesons, which are BF ( D + → K − X ) = ( 24.7 ± 1.3 ± 1.2 ) % , BF ( D + → K + X ) = ( 6.1 ± 0.9 ± 0.4 ) % , BF ( D 0 → K − X ) = ( 57.8 ± 1.6 ± 3.2 ) % and BF ( D 0 → K + X ) = ( 3.5 ± 0.7 ± 0.3 ) % , respectively. We have also measured the branching fractions for the inclusive semileptonic decays of D + and D 0 mesons to be BF ( D + → e + X ) = ( 15.2 ± 0.9 ± 0.8 ) % and BF ( D 0 → e + X ) = ( 6.3 ± 0.7 ± 0.4 ) % . These yield the ratio of their partial widths to be Γ ( D + → e + X ) / Γ ( D 0 → e + X ) = 0.95 ± 0.12 ± 0.07 .

By analyzing the data sets of 17.3 pb−1 taken at s = 3.773 GeV and 6.5 pb−1 taken at s = 3.650 GeV with the BESII detector at the BEPC collider, we have measured the observed cross sections for 12 exclusive light hadron final states produced in e + e − annihilation at the two energy points. We have also set the upper limits on the observed cross sections and the branching fractions for ψ ( 3770 ) decay to these final states at 90% C.L.

With a sample of 14 × 10 6 ψ ( 2 S ) events collected by the BESII detector at the Beijing Electron Positron Collider (BEPC), the decay channels ψ ( 2 S ) → B 8 B ¯ 8 ( p p ¯ , Λ Λ ¯ , Σ 0 Σ ¯ 0 , Ξ − Ξ ¯ + ) are measured, and their branching ratios are determined to be ( 3.36 ± 0.09 ± 0.25 ) × 10 −4 , ( 3.39 ± 0.20 ± 0.32 ) × 10 −4 , ( 2.35 ± 0.36 ± 0.32 ) × 10 −4 , ( 3.03 ± 0.40 ± 0.32 ) × 10 −4 , respectively. In the decay ψ ( 2 S ) → p p ¯ , the angular distribution parameter α is determined to be 0.85 ± 0.24 ± 0.04 .

The microbiome modulates numerous aspects of human physiology and is a crucial factor in the development of various human diseases. Vitamin D deficiency and downregulation of the vitamin D receptor (VDR) are also associated with the pathogenesis of diseases such as inflammatory bowel disease, cancers, obesity, diabetes, and asthma. VDR is a nuclear receptor that regulates the expression of antimicrobial peptides and autophagy regulator ATG16L1. Vitamin D may promote a balanced intestinal microbiome and improve glucose homeostasis in diabetes. However, how VDR regulates microbiome is not well known. In the current study, we hypothesize that VDR status regulates the composition and functions of the intestinal bacterial community.Fecal and cecal stool samples were harvested from Vdr knockout (Vdr(-/-)) and wild-type mice for bacterial DNA and then sequenced with 454 pyrosequencing. The sequences were denoised and clustered into operational taxonomic units, then queried against the National Center for Biotechnology Information database. Metagenomics were analyzed, and the abundances of genes involved in metabolic pathways were compared by reference to the Kyoto Encyclopedia of Genes and Genomes and Clusters of Orthologous Groups databases.In the Vdr(-/-) mice, Lactobacillus was depleted in the fecal stool, whereas Clostridium and Bacteroides were enriched. Bacterial taxa along the Sphingobacteria-to-Sphingobacteriaceae lineage were enriched, but no genera reached statistical significance. In the cecal stool, Alistipes and Odoribacter were depleted, and Eggerthella was enriched. Notably, all of the taxa upstream of Eggerthella remained unchanged. A comparison of Vdr(-/-) and wild-type samples revealed 40 (26 enriched, 14 depleted) and 72 (41 enriched, 31 depleted) functional modules that were significantly altered in the cecal and fecal microbiomes, respectively (both, P0.05), due to the loss of Vdr. In addition to phylogenetic differences in gut microbiome with different intestinal origins, we identify several important pathways, such as nucleotide-binding oligomerization domain-like receptor, affected by Vdr status, including amino acid, carbohydrate, and fatty acid synthesis and metabolism, detoxification, infections, signal transduction, and cancer and other diseases.Our study fills knowledge gaps by having investigated the microbial profile affected by VDR. Insights from our findings can be exploited to develop novel strategies to treat or prevent various diseases by restoring VDR function and healthy microbe-host interactions.

Inflammatory bowel diseases (IBD), including Crohn’s disease and ulcerative colitis, are featured by chronic intestinal inflammation, which is becoming increasingly prevalent in Western societies, and is spreading to the rest of the world. Although the etiology of IBD is poorly understood, it is widely accepted that several factors may be crucial, which includes genetics, diet and lifestyle, immunity, environment, and microbiota. In the past decade, huge advances have been made toward a better understanding of IBD, epitomized by far-reaching progress in the field of microbiome. In this review, we summarize the current knowledge of how changes in microbiota may affect the pathogenesis of IBD. Commensal bacteria, benign normally, are essentially opportunists that may readily take over, and potentially contributes to dysbiosis, which, in turn, promotes pathogenesis. Several pathogens, mainly Mycobacterium avium paratuberculosis, adherent invasive Escherichia coli, Clostridium difficile, Campylobacter, and Salmonella, have been shown to be associated with IBD, but the causality remains unproven. The microbiome comprises of not only bacteria, but also viruses, bacteriophages, and fungi. However, little is known as per the role of the latter. We highlight recent research on viruses, bacteriophages, and fungi in IBD. We also discuss the progress on manipulating the microflora to serve therapeutic purposes. The methodology for manipulating the microbiota covers fecal transplantation, pre-, pro-, syn- and post-biotics, helminth therapy, bacteriocins, bacteriophage, etc., which greatly enriches the arsenal against IBD. Manipulation of intestinal microbiome represents a promising type of therapy for IBD.

Animal genetic resources top the list of the most fundamental properties for animal husbandry and medical sciences, play an irreplaceable role in human survival and social prosperity, constitute an invaluable substrate for scientific researches, and have an enormous bearing on sustainable development of economy. Biodiversity takes an underlying part in ecological balance. The havoc people wreaked on nature had been aggravating ever since agricultural civilization, accelerating the extinction of animal species and breed incomparable than gradual natural loss, which became more and more apparent upon the advent of industrial age. The statistics of Food and Agriculture Organization (FAO) in 1995 revealed that approximately 15% of the total 738 registered livestock and poultry breeds in Sub-Saharan Africa were on the brink of extinction. The situation has been aggravating ever since. To date, the proportion of livestock breed in danger has increased from 8% to 19%, while that of poultry has risen from 20% to 34%. Among the 1251 registered breeds in Asia, 10% are severely endangered. From 1995 to 1999, livestock breeds about to be extinct grew from 11% to 14%, and the proportion of poultry was 32% to 37%. Owing to economic pressure, some low yield breeds are being subjected to marketing elimination and shrinkage in population, for instance, the production of poultry and swine depends heavily on only a few breeds. The trend is extremely obvious in Eastern Europe, which is further worsened by political unstability. Similarly, sustaining intensification of animal husbandry makes the food production rely more and more on a small number of high yield breeds, thereby exacerbating the animal diversity crisis. In Latin America, the number of endangered breeds accounts for 20% of the whole. As was reported by the FAO in 2000, livestock and poultry throughout the earth are disappearing at the rate of 2 breeds per week. Worse still, 1350 breeds are next to imminent extinction. For all that matter, animal genetic resources are confronted with a progressive narrowing in diversity. Accordingly, it’s absolutely exigent to protect and preserve them with effective measures.

A millimeter wave ultra-wide band (UWB) tapered slot antenna is presented based on a multilayer liquid crystal polymer (LCP) circuit process. The multilayer circuit board consists of eight copper layers and seven LCP layers. In order to enhancing the bandwidth, two overlapped tapered slots with different flare angle are applied in the 3rd and the 5th copper layers, respectively. A low backward radiation characteristic is achieved due to using the ground-backed structure. A simple design guideline is detected and the dimension is optimized. The designed results indicate that the millimeter wave UWB slot antenna can work in the frequency band of 33GHz-60GHz (a relative bandwidth of 58%). The radiation patterns are calculated at three typical frequencies and the analysis demonstrates that an elliptical polarization performance is obtained within the total operation frequency band. The study also demonstrates that the LCP process is suitable for developing lightweight and high performance millimeter wave antennas.

The subject of this paper is a dual frequency microstrip antenna operating at 28.5–30.4 GHz and 58.8–60.9 GHz. Local multipoint distribution system (LMDS) and The WirelessHD (WiHD) special interest group utilizes the band above 26GHz and 60 GHz, respectively. Antenna design was based on the ultra wideband tapered slot antenna. Dual frequency operation is realized on a multilayer substrate with multi-arm stubs and corrugation structure. Corrugation structure is used to confine the energy formed by the fields approximately from 30.4 GHz to 58.8 GHz.

Recently, wearable wireless communication system has attracted much attention. It is used widely in the areas of military operations, medical field and personal communication applications. With the rapid development of the wearable systems, wearable antennas are facing with a considerable design challenge, especially in the bandwidth of the antenna. This paper proposed a wearable antenna for ultra-wideband (UWB) communication systems. The antenna is composed of a quasi-trapeziform ground and a rectangular patch with two symmetrical bevel slots at each side of the lower edge of the patch. The two bevel slots on the patch can enhance the bandwidth of the antenna to satisfy the requirements of UWB communication systems. And the size of the quasi-trapeziform ground can influence the match of the antenna. The antenna shows UWB bandwidth with high performance. Then, we study the performance of the antenna worn on body. It also shows good performance. Simulation was accomplished using Ansoft HFSS and CST Microwave Studio, both simulated results are presented and they are in good agreement.

An ATCA and FPGA-based Data Processing Unit for PANDA Experiment has been developed. The Unit features 5 Xilinx Virtex-4 FX60 FPGA chips and up to 10 GBytes DDR2 memory. High bandwidth connectivity is provided by 8 additional optical links using RocketIO ports and 5 Gbit Ethernet links to receive data from front-end electronics and transmit the data passed high level trigger to mass storage. A single ATCA shelf can host up to 14 boards which are interconnected via a full mesh backplane. A prototype system has been set up and some tests have been made.
