International audience; Date palm (Phoenix dactylifera) is an important crop providing a valuable nutrition source for people in many countries including the Middle East and North Africa. In recent years, the amount of rain in North Africa and especially in the Tunisian palm grove areas has dropped significantly. We investigated the growth and cell wall remodelling of fruits harvested at three key development stages from trees grown with or without water supply. During development, cell wall solubilization and remodelling was characterized by a decrease of the degree of methylesterification of pectin, an important loss of galactose content and a reduction of the branching of xylan by arabinose in irrigated condition. Water deficit had a profound effect on fruit size, pulp content, cell wall composition and remodelling. Loss of galactose content was not as important, arabinose content was significantly higher in the pectin‐enriched extracts from non‐irrigated condition, and the levels of methylesterification of pectin and O‐acetylation of xyloglucan were lower than in irrigated condition. The lower levels of hydrophobic groups (methylester and O‐acetyl) and the less intensive degradation of the hydrophilic galactan, arabinan and arabinogalactan in the cell wall may be implicated in maintaining the hydration status of the cells under water deficit.

Plant cells are surrounded by a carbohydrate-rich compartment called the cell wall. This compartment plays a central role in growth, development, sexual reproduction and defence (Vicre et al., 2005; Mollet et al., 2007; Hematy et al., 2009; Seifert & Blaukopf, 2010). It is a carbohydrate-based structure composed mostly of polysaccharides (~90%) and glycoproteins (~10%). Polysaccharides include cellulose, pectin domains such as homogalacturonans (HG) with different levels of methylesterification, xylogalacturonan, rhamnogalacturonan-I (RG-I) and rhamnogalacturonanII (RG-II) and hemicelluloses (depending on the species, xyloglucan [XyG] or arabinoxylan) (Liepman et al., 2010), whereas cell wall proteins are either enzymatic such as glucanases (Gilbert, 2010) or nonenzymatic such as arabinogalactan proteins (AGP) (Ellis et al., 2010, Driouich & Baskin, 2008).

Au cours de la reproduction sexuée des plantes, la germination du pollen et la croissance des tubes polliniques sont soumises à un contrôle spatio-temporel dans les tissus femelles afin d'acheminer les gamètes mâles jusqu'aux ovules. Le tube pollinique est une cellule à croissance rapide entourée d'une matrice extracellulaire riche en polysaccharides. La croissance polarisée des tubes polliniques et leurs capacités à percevoir des signaux externes permettent de les guider à travers le tissu de transmission de l'organe femelle. Parmi ces signaux, un mécanisme de guidage par adhésion a été postulé chez le lys. Peu de travaux concernent les tubes polliniques de la plante modèle Arabidopsis thaliana. Dans le but d'identifier les interactions impliquées dans l'adhésion des tubes polliniques, nous avons entrepris 1- de caractériser les composés pariétaux des tubes et des cellules du tissu de transmission qui sont la première interface de contact entre ces cellules ; 2 - d'utiliser une approche de génétique chimique pour identifier des molécules impliquées dans l'adhésion cellulaire. Nos résultats indiquent que la paroi des tubes polliniques d'Arabidopsis se compose d'une double paroi riche en callose, pour la paroi interne et en pectines et xyloglucanes pour la paroi externe. Nous avons alors caractérisé la structure des xyloglucanes et mis en évidence que ce polymère est fortement fucosylé et O-acétylé. Cette caractéristique structurale semble être conservée parmi les espèces. L'abondance de l'arabinose provenant des chaînes latérales du rhamnogalacturonane de type 1 a été également mise en évidence. Pour étudier l'adhésion, nous avons reconstitué une matrice à partir de polysaccharides pariétaux extraits de fleurs mais aussi d'organes végétatifs de la plante. Les tubes polliniques adhèrent sur des pectines extraites à l'imidazole de fleurs et de feuilles d'Arabidopsis. Cette matrice nous a permis de constituer un test d'adhésion assez efficace et significatif pour le soumettre à une analyse de génétique chimique. Cette méthode consiste à étudier des processus biologiques en les perturbant via le criblage de petites molécules chimiques. Parmi les 258 molécules en notre possession, cinq ont montré des résultats prometteurs en réduisant significativement l'adhésion des tubes polliniques. De plus, avec certaines molécules, un effet dose-réponse a pu être mis en évidence.; During sexual plant reproduction, pollen germination and pollen tube growth require a tight spatial and temporal control while traveling through the female tissues in order to deliver properly the sperm cells to the ovules. Pollen tubes are unique, fast growing cells surrounded by a cell wall enriched in polysaccharides. They are tip-polarized cells able to perceive external signal cues to guide them through the transmitting tract of the female organs. Among these signals, guidance by adhesion has been postulated in lily. Few data are available concerning the cell wall of the pollen tube in the model plant Arabidopsis Thaliana. In order to identify the interactions implicated in pollen tube adhesion, we first characterized the cell wall components from the pollen tube and the transmitting tract tissue. Secondly, we used a chemical genetic approach to disturb cell adhesion. Our data indicate that Arabidopsis pollen tube cell wall is composed of a double cell wall, enriched in callose in the inner layer and pectins and xyloglucans in the outer layer. Characterization of the xyloglucan structure showed that it was highly fucosylated and O-acetylated. These structural features seemed to be preserved among species. In addition, the cell wall is composed of a large amount of arabinosyl residues coming from the rhamnogalacturonan-1 side chains. In order to identify molecules involved on pollen tube adhesion, we have used a chemical genetic approach. This new method uses small molecules to alter a specific biological process and identify protein(s) involved. To this end, we elaborated an artificial matrix where pollen tubes were able to adhere. The matrix constitutes of cell wall pectins extracted with imidazole from Arabidopsis flowers and vegetative organs. Then, the assay was used to screen 258 small molecules from our library. Our data showed that five out of the 258 tested molecules, were able to disturb pollen tube adhesion in a dose-dependent manner.

Au cours de la reproduction sexuée des plantes, la germination du pollen et la croissance des tubes polliniques sont soumises à un contrôle spatio-temporel dans les tissus femelles afin d'acheminer les gamètes mâles jusqu'aux ovules. Le tube pollinique est une cellule à croissance rapide entourée d'une matrice extracellulaire riche en polysaccharides. La croissance polarisée des tubes polliniques et leurs capacités à percevoir des signaux externes permettent de les guider à travers le tissu de transmission de l'organe femelle. Parmi ces signaux, un mécanisme de guidage par adhésion a été postulé chez le lys. Peu de travaux concernent les tubes polliniques de la plante modèle Arabidopsis thaliana. Dans le but d'identifier les interactions impliquées dans l'adhésion des tubes polliniques, nous avons entrepris 1- de caractériser les composés pariétaux des tubes et des cellules du tissu de transmission qui sont la première interface de contact entre ces cellules ; 2 - d'utiliser une approche de génétique chimique pour identifier des molécules impliquées dans l'adhésion cellulaire. Nos résultats indiquent que la paroi des tubes polliniques d'Arabidopsis se compose d'une double paroi riche en callose, pour la paroi interne et en pectines et xyloglucanes pour la paroi externe. Nous avons alors caractérisé la structure des xyloglucanes et mis en évidence que ce polymère est fortement fucosylé et O-acétylé. Cette caractéristique structurale semble être conservée parmi les espèces. L'abondance de l'arabinose provenant des chaînes latérales du rhamnogalacturonane de type 1 a été également mise en évidence. Pour étudier l'adhésion, nous avons reconstitué une matrice à partir de polysaccharides pariétaux extraits de fleurs mais aussi d'organes végétatifs de la plante. Les tubes polliniques adhèrent sur des pectines extraites à l'imidazole de fleurs et de feuilles d'Arabidopsis. Cette matrice nous a permis de constituer un test d'adhésion assez efficace et significatif pour le soumettre à une analyse de génétique chimique. Cette méthode consiste à étudier des processus biologiques en les perturbant via le criblage de petites molécules chimiques. Parmi les 258 molécules en notre possession, cinq ont montré des résultats prometteurs en réduisant significativement l'adhésion des tubes polliniques. De plus, avec certaines molécules, un effet dose-réponse a pu être mis en évidence., During sexual plant reproduction, pollen germination and pollen tube growth require a tight spatial and temporal control while traveling through the female tissues in order to deliver properly the sperm cells to the ovules. Pollen tubes are unique, fast growing cells surrounded by a cell wall enriched in polysaccharides. They are tip-polarized cells able to perceive external signal cues to guide them through the transmitting tract of the female organs. Among these signals, guidance by adhesion has been postulated in lily. Few data are available concerning the cell wall of the pollen tube in the model plant Arabidopsis Thaliana. In order to identify the interactions implicated in pollen tube adhesion, we first characterized the cell wall components from the pollen tube and the transmitting tract tissue. Secondly, we used a chemical genetic approach to disturb cell adhesion. Our data indicate that Arabidopsis pollen tube cell wall is composed of a double cell wall, enriched in callose in the inner layer and pectins and xyloglucans in the outer layer. Characterization of the xyloglucan structure showed that it was highly fucosylated and O-acetylated. These structural features seemed to be preserved among species. In addition, the cell wall is composed of a large amount of arabinosyl residues coming from the rhamnogalacturonan-1 side chains. In order to identify molecules involved on pollen tube adhesion, we have used a chemical genetic approach. This new method uses small molecules to alter a specific biological process and identify protein(s) involved. To this end, we elaborated an artificial matrix where pollen tubes were able to adhere. The matrix constitutes of cell wall pectins extracted with imidazole from Arabidopsis flowers and vegetative organs. Then, the assay was used to screen 258 small molecules from our library. Our data showed that five out of the 258 tested molecules, were able to disturb pollen tube adhesion in a dose-dependent manner.

- Abstract
Chlamydomonas reinhardtiiis a green unicellular eukaryotic model organism for studying relevant biological and biotechnological questions. The availability of genomic resources and the growing interest in C. reinhardtiias an emerging cell factory for the industrial production of biopharmaceuticals require an in-depth analysis of protein N-glycosylation in this organism. Accordingly, we used a comprehensive approach including genomic, glycomic, and glycoproteomic techniques to unravel the N-glycosylation pathway of C. reinhardtii.Using mass-spectrometry-based approaches, we found that both endogenous soluble and membrane-bound proteins carry predominantly oligomannosides ranging from Man-2 to Man-5. In addition, minor complex N-linked glycans were identified as being composed of partially 6-O-methylated Man-3 to Man-5 carrying one or two xylose residues. These findings were supported by results from a glycoproteomic approach that led to the identification of 86 glycoproteins. Here, a combination of in-source collision-induced dissodiation (CID) for glycan fragmentation followed by mass tag-triggered CID for peptide sequencing and PNGase F treatment of glycopeptides in the presence of 18O-labeled water in conjunction with CID mass spectrometric analyses were employed. In conclusion, our data support the notion that the biosynthesis and maturation of N-linked glycans in the endoplasmic reticulum and Golgi apparatus occur via a GnT I-independent pathway yielding novel complex N-linked glycans that maturate differently from their counterparts in land plants.

- Abstract
Plant sexual reproduction involves the growth of tip-polarized pollen tubes through the female tissues in order to deliver the sperm nuclei to the egg cells. Despite the importance of this crucial step, little is known about the molecular mechanisms involved in this spatial and temporal control of the tube growth. In order to study this process and to characterize the structural composition of the extracellular matrix of the male gametophyte, immunocytochemical and biochemical analyses of Arabidopsis pollen tube wall have been carried out. Results showed a well defined localization of cell wall epitopes with highly esterified homogalacturonan and arabinogalactan-protein mainly in the tip region, weakly methylesterified homogalacturonan back from the tip and xyloglucan and (1→5)-α-L-arabinan all along the tube. Here, we present complementary data regarding 1) the ultrastructure of the pollen tube cell wall and 2) the immunolocalization of homogalacturonan and arabinan epitopes in 16h-old pollen tubes and in the stigma and the transmitting tract of the female organ. Discussion regarding the pattern of the distribution of the cell wall epitopes and the possible mechanisms of cell adhesion between the pollen tubes and the female tissues is provided.

48. In silicoprediction of proteins related to xyloglucan fucosyltransferases in Solanaceae genomes

- Abstract
Two independent studies have shown that the cell wall of pollen tubes from tobacco and tomato species contained fucosylated xyloglucan (XyG). These findings are intriguing as many reports have shown that XyG of somatic cells of these species is not fucosylated but instead is arabinosylated. In order to produce fucosylated XyG, plants must express a functional galactoside α-2-fucosyltransferase. Here, using a bioinformatics approach, we show that several candidate genes coding for XyG fucosyltransferases are present in the genome of coffee and several Solanaceae species including tomato, tobacco, potato, eggplant and pepper. BLAST and protein alignments with the 2 well-characterized XyG fucosyltransferases from Arabidopsis thalianaand Pisum sativumrevealed that at least 6 proteins from different Solanaceae species and from coffee displayed the 3 conserved motifs required for XyG fucosyltransferase activity.

- Abstract
Date palm (Phoenix dactylifera) is an important crop providing a valuable nutrition source for people in many countries including the Middle East and North Africa. In recent years, the amount of rain in North Africa and especially in the Tunisian palm grove areas has dropped significantly. We investigated the growth and cell wall remodelling of fruits harvested at three key development stages from trees grown with or without water supply. During development, cell wall solubilization and remodelling was characterized by a decrease of the degree of methylesterification of pectin, an important loss of galactose content and a reduction of the branching of xylan by arabinose in irrigated condition. Water deficit had a profound effect on fruit size, pulp content, cell wall composition and remodelling. Loss of galactose content was not as important, arabinose content was significantly higher in the pectin-enriched extracts from non-irrigated condition, and the levels of methylesterification of pectin and O-acetylation of xyloglucan were lower than in irrigated condition. The lower levels of hydrophobic groups (methylester and O-acetyl) and the less intensive degradation of the hydrophilic galactan, arabinan and arabinogalactan in the cell wall may be implicated in maintaining the hydration status of the cells under water deficit., (© 2012 Blackwell Publishing Ltd.)
