The development of rootstocks that are less sensitive to abiotic and biotic stresses can help mitigate the negative effects of climate change on crop productivity, soil health, and water use. Though, the phenomenon of graft incompatibility is a significant limitation to the spread of new rootstock genotypes. Numerous studies have focused on this issue, highlighting the role of certain phenolic molecules as predictive markers of incompatibility. Given the lack of specific research on table grape, , this study proposes a protocol for the extraction of polyphenols from its woody tissues, which is a fundamental prerequisite for further analysis on secondary metabolites involved in graft incompatibility. The proposed eco-friendly method coupled a traditional maceration using solvent with a green technique utilizing an ultrasound-assisted extractor. The following parameters were compared: (1) sample weight (0.1 g, 0.5 g, and 1 g), (2) time of ultrasound-assisted extraction (10 min, 20 min, and 30 min), and (3) solvent volume for maceration (10 mL, 15 mL, and 20 mL). Four phenol classes were considered based on previous works on Vitis spp.: cinnamic acids, flavonols, benzoic acids, and catechins. The characterization of polyphenolic biomarkers was carried out via HPLC. 1 g of plant material, 30 minutes of sonification, and 20 mL of organic solvent was the combination of factors that resulted in the most efficient fingerprint, both quantitatively (267.68 ± 3.91 mg/100 g fresh weight [FW]) and qualitatively, with the four classes analyzed significantly represented. This is the first work to come up with an extraction protocol for phenol compounds in table grape woody tissue based on both ecological and routine techniques.

Morella spathulata (Myricaceae family) is a common plant from Madagascar and is present on the IUCN Red List of threatened species classified at the ’least concern’ level, used by the local population to treat numerous illnesses and pain. Despite its frequent use, comprehensive phytochemical and pharmacological research on the species is limited. This study evaluated the antioxidant, analgesic, and anti-inflammatory properties, as well as the toxicity of methanol extracts from the leaves (MS_L) and bark (MS_B) of M. spathulata. The research involved the analysis of nutritional traits such as sugars, organic acids, vitamin C, polyphenolic content (TPC) and the main phytochemicals by HPLC analysis. Antioxidant capacity was assessed through DPPH and FRAP assays. Analgesic and anti-inflammatory activities were evaluated using acetic acid-induced writhing and carrageenan-induced paw oedema tests in mice. The results showed a high content of phenolic and bioactive components in the leaf and bark extracts, associated with antioxidant, analgesic and anti-inflammatory properties. The interaction of key compounds such as ferulic acid and ellagic acid with proteins involved in pH regulation and immune modulation provides clues to the mechanisms underlying the therapeutic effects. However, conservation efforts are crucial due to habitat loss and illegal logging, and further studies are needed to fully explore the plant’s therapeutic potential.

Background: The aerial parts of Micromeria madagascariensis Baker and M. flagellaris Baker are used by the population of the Vakinankaratra and Itasy regions (Madagascar) to treat breathing difficulty, fever and/or headache, wounds, and sores. Purpose: This work aimed to characterise plant materials from M. madagascariensis and M. flagellaris to report i) chemical composition, ii) antimicrobial properties, and iii) antioxidant capacity of the essential oils extracted from the aerial parts of these species. Materials and methods: The essential oils from M. madagascariensis (MMO) and M. flagellaris (MFO) were obtained by hydrodistillation. Their chemical composition was quantified using gas chromatography coupled with mass spectrometry (GC-MS). MMO and MFO were also tested against 7 microbial strains using the disk diffusion method and their antioxidant capacity was assessed using the DPPH scavenging assay. Results: Hydrodistillation yielded 0.26% MMO and 0.29% MFO (w/w) in relation to the fresh weight. Twenty-seven compounds were identified by GC-MS in MMO extract against 36 in MFO one. The main compounds in MMO were pulegone (24.67%), trans-menthone (24.67%), eucalyptol (8.12%), β-caryophyllene (4.98%), α-guanene (4.47), iso-menthone (3.85%), iso-pulegone (3.34%), azulene (3.28%) and 2-isopropyl-5-methylcyclohexenone (2.82%). The main compounds in the MFO were eudesma-4,11-dien-2-ol (13.88%), δ-guanene (6.62%), pulegone (6.40%), cyperone (5.56%), 4-epi-dehydrobietinol acetate (5.39%), eucalyptol (5.12%), trans-menthone (4.67%), limonene (3.77%) and sabinene (2.29%). Regarding the chemotaxonomy, M. flagellaris was very different from M. madagascariensis and both species also differed from the other Micromeria species, as confirmed by multivariate statistical analysis. Both MMO and MFO exerted activities against a large microbial spectrum; the antimicrobial activity of MMO was higher than MFO one against S. pneumoniae and C. albicans due to the presence of pulegone as the main component. MFO showed an excellent scavenging capacity with an SC50 value of 2.17 ± 0.03 μg/mL. Conclusion: The biological properties of the essential oils extracted from the selected species may explain their therapeutic value showing that Malagasy Micromeria species may be very important as new natural sources of bioactive compounds. This study may promote the effectiveness and quality of Malagasy Micromeria species, contributing to sustainable development and commercial valorisation of traditional preparations based on natural local resources.

Abstract Within the world of natural food supplements, organic extracts deriving from young plant meristematic tissue (bud-derivatives) are becoming attractive, thanks to their richness in bioactive molecules. This natural source is scarce, but every year, tons of plant material, including buds, come from city pruning. If this sustainable source is rather promising from a circular economy point of view, the safety of the obtained supplements must be assessed. In fact, anthropic microcontaminants, such as polycyclic aromatic hydrocarbons (PAHs), could adsorb onto the urban buds, leading to a possible contamination of the bud-derivatives. In this study, we developed a magnetic dispersive solid phase extraction (m-dSPE) based on molecularly imprinted microparticles, combined with GC-MS, to quantify the 16 priority PAHs in such extracts. The D-optimal experimental design was implemented to maximize analytes’ recovery with the smallest set of experiments. The optimized method was characterized by great selectivity thanks to the molecular imprinted polymer and ease of use provided by m-dSPE. Moreover, it complies with green principles, thanks to the minimum consumption of organic solvent (1.5 mL of acetone per sample). The recoveries ranged from 76 to 100% and procedural precision was below 10% for most PAHs. Despite the matrix complexity, low quantification limits (0.7–12.6 μg kg−1) were reached. This guaranteed the PAHs’ quantitation at levels below those indicated as safe by a European Community regulation on food supplements. None of the analyzed samples, coming from different anthropically impacted areas, showed concerning PAHs levels.

Yerba mate (Ilex paraguariensis) is a plant originating from South America. From this plant, mate a drink is produced. Brazil is the first mate producer in the world. The diffusion of mate is mainly limited to the native regions of South America, as it is consumed with cuia and bombilla. Trying to expand mate culture and export it towards Europe makes it necessary to offer this drink in a new guise that sets it apart from traditional preparation patterns. In this instance, the introduction of green tea mate may represent a solution. This innovative product comes from I. paraguariensis sprouts, which are further processed and consumed as green tea from Camellia sinensis. The present study aimed to investigate the physico-chemical and nutraceutical characteristics and consumer perceptions towards the visual aspect of the innovative green tea mate (L1, L2, and L3) compared to commercial traditional Brazilian mate (CI and CB) and roasted mate (L). Moreover, a preliminary sensory evaluation with untrained panellists was carried out. Significant results were detected, highlighting the higher content of bioactive compounds in the innovative product. Total Phenolic Content (TPC) almost doubled in green tea mate (L1 117.14 mg GAE/g d.p., L2 128.10 mg GAE/g d.p., L3 126.21 mg GAE/g d.p.) compared to the other samples (CI 71.91 mg GAE/g d.p., CB 54.23 mg GAE/g d.p., L 34.16 mg GAE/g d.p.) (p < 0.05). Considering caffeine, batches L1 and L3 had a content of 3.68 mg/g d.p. and 3.58 mg/g d.p., respectively, significantly higher when compared to the amount retrieved in the CB sample, 1.57 mg/g d.p. (p < 0.05). The consumer survey demonstrated the interest of consumers towards the new product, as it was perceived similar to more commonly consumed teas, while from a sensory point of view, a vegetal and hay flavour were more defining for the product.

Chestnut processing has increasingly grown in recent years. All the processes involved in the chestnut supply chain are characterized by the production of high levels of by-products that cause several environmental and disposal issues. The Castanea spp. fruit production is related to a high number of chestnut episperm. This underutilized agricultural by-product may be evaluated as a good resource for the extraction of health-promoting natural molecules, such as phenolics. This preliminary study aimed to develop and optimize, using a multivariate statistical approach, a sustainable protocol for the ultrasound-assisted extraction (UAE) of the main phenolics from chestnut episperm (cv Marsol, C. sativa × C. crenata). A design of experiment (DoE) approach was employed. This approach focused on the two quantitative UAE process factors: the extraction time (X1), within a timeframe ranging from 10 to 30 min, and the sample-to-solvent (w/v) ratio (X2), ranging from 1/30 to 1/10. These variables were investigated to determine their impact on phenol extraction yield. Exploratory analysis, in particular principal component analysis (PCA) and multiple linear regression (MLR), were carried out on the studied responses. The phenolic characterization of ten different extracts was also performed using high-performance liquid chromatography (HPLC), both to define the levels of specific phenolics selected for their health-promoting properties and to evaluate some important features, such as the total antioxidant capacity. The values of total polyphenolic content (TPC) obtained in the different experiments ranged between 97 (extract 4) and 142 (extract 6) mg GAE/g of dried weight (DW). Moreover, results from the ferric reducing antioxidant power (FRAP) test confirmed the high TPC values, highlighting that all the ultrasound extracts contained excellent levels of molecules with good antioxidant properties. In particular, extracts 2 and 3 showed the highest AOC values (about 490–505 mmol Fe2+/Kg of dried weight). The proposed optimized protocol allowed for obtaining formulations characterized by high levels of tannins, phenolic acids, and catechins. Indeed, episperm extracts contained high levels of chlorogenic acid (15–25 mg/100 g DW), ferulic acid (80–120 mg/100 g DW), castalagin (20–80 mg/100 g DW), and vescalagin (40–75 mg/100 g). Finally, in this research study, the potential of chestnut episperm as a source of polyphenolic molecules to be extracted by green technologies and used in several food and/or pharmaceutical applications was evaluated to valorize a sustainable reuse strategy of agri-food processing by-products, also reducing the environmental impact of this waste derived from chestnut processing.

The phytochemical characterization of Castanea spp. fruits is very important for the development of effective strategies for the biodiversity conservation and traceability of chestnuts, as the chestnut is one of the most important Italian and European nut and forest species. In this study, several cultivars of C. sativa (sweet chestnuts and “marrone-type”), C. crenata, and hybrids of C. sativa × C. crenata were characterized by spectrophotometric (Folin–Ciocalteu assay for the total polyphenolic content and ferric reducing antioxidant power test for the antioxidant capacity) and chromatographic (high-performance liquid chromatography coupled to a diode array UV-Vis detector) protocols to define their phytochemical composition and nutraceutical properties. The phytochemical results were then used to build a multivariate statistical model (by principal component analysis) and obtain an effective and rapid tool to discriminate unknown cultivars (i.e., no information about their origin) belonging to different species. The multivariate approach showed that the genotype was a significantly discriminating variable (p < 0.05) for the phytochemical composition. Polyphenols (in particular, phenolic acids and tannins) have been identified as the main bioactive classes with the highest discriminating power among the different genotypes. The total polyphenol content (TPC) and antioxidant capacity (AOC) showed a rich presence of bioactive compounds (74.09 ± 15.10 mgGAE 100 g−1 DW and 11.05 ± 1.35 mmol Fe2+ kg−1 DW, respectively), underlining the potential health benefits and functional traits of chestnuts. The principal component analysis applied to phytochemical variables has proved to be an excellent and effective tool for genotype differentiation to be used as a preliminary method for identifying the species of Castanea spp. fruits with an unknown origin. The present study showed that a multivariate approach, based on phytochemical data and preliminary to genetic analysis, may represent a rapid, effective, and low-cost tool for the traceability and quality evaluation of chestnuts from different species and hybrids with no information on their origin.

Bud derivatives (BDs) represent a category of botanicals obtained by macerating the meristematic tissues (buds or young sprouts) of plants; widely used since ancient times in complementary medicine, they remain poorly investigated to date. In this study, a contribution is made toward the identification of the correct “a posteriori” attribution of the botanical species in commercial BDs, which is very important for ensuring the quality and safety of these natural products. Excitation–emission matrix (EEM) fluorescence spectroscopy proved to be a rapid, non-destructive and low-cost analytical instrument for providing a preliminary qualitative characterization of the BDs, allowing for the identification of specific spectral regions related to flavonol compounds and cinnamic and benzoic acids, in agreement with the targeted chromatographic analysis (HPLC-DAD), which completely defined a phytochemical profile for each BD considered. This information will be implemented in the first web application for the recognition of vegetable buds, named GEMMAPP, which was designed by the Authors and is proposed as a tool and future scientific data repository for providing information about the main, typical BDs of the France–Italy Alcotra territory.

More information on the effects of traditional cooking methods (roasting or boiling) on the chestnut composition may be important if health-promoting aspects are considered. The main aims of this study were to investigate and describe the phenolic profile and antioxidant capacity of raw, boiled, and roasted chestnuts from several Castanea spp. genotypes, evaluating the influence of the application of different traditional cooking methods on the nut phytochemical composition by chromatographic and spectroscopic strategies. The amounts of phenolics were used as selected variables together with total polyphenol content and antioxidant capacity to perform a Principal Component Analysis (PCA). Catechins and tannins were the main molecules in the phenolic phytocomplex, reaching 30–40% of the total, followed by phenolic acids (5–20%) and flavonols (about 5%). Gallic and ellagic acids were the most important phenolic acids in raw and processed chestnuts (about 20–70 mg·100 g−1 dried weight-DW and 10–50 mg·100 g−1 DW, respectively). Both of the cooking processes significantly influenced the polyphenolic content and the relative antioxidant capacity. This research may support and confirm the potential use of chestnuts for human health, increasing the information on the phenolic pattern of differently processed Castanea spp. fruits from different genotypes to (i) assess the potential health-positive effects, (ii) help processing companies to select specific varieties to commercialise in the market, and (iii) increase the use of these fruits with the relative increase in income for the producers.

Antioxidants are important supplements for the human body for their roles in human life for the maintenance of homeostasis. Tapia fruits (Uapaca bojeri) are used by the riverain population of the Tapia forests in Madagascar as complementary foods. This study aims to quantify the main antioxidants in the U. bojeri fruits to verify their contribution to the enhancement of their anti-inflammatory and antihyperglycemic effects. Standard phytochemical screening was used for qualitative analysis, while spectrophotometric (TPC, TAC, and TFC) and chromatographic analyses (HPLC) were used to quantify several phytochemicals in U. bojeri fruits. The antioxidant activity was evaluated using DPPH and FRAP assays. The writhing test was used for the analgesic effects, the carrageenan-induced paw edema was used for the anti-inflammatory activity, and OGTT was used to test the anti-hyperglycemia property of the MEUB in mice. Several phytocompounds were detected and quantified in the fruits, including succinic acid (67.73%) as the main quantified compound. Fruits exerted a good antioxidant capacity and showed analgesic, anti-inflammatory, and antihyperglycemic activities in mice. Isolation of the bioactive compounds should be carried out to confirm these pharmacological properties and develop health-promoting food products or medicinal applications derived from this species.

Traditional medicine plays an important role in the Malagasy health system. In Madagascar, medicinal plants are the main remedies for several diseases, especially chronic diseases. Scientific studies are performed to valorize the use of Imperata cylindrica (Ic), Uapaca bojeri (Ub), Vaccinium secondiflorum (Vs), and Ravenala madagascariensis (Rm). Ic and Ub are used to treat some inflammatory-related diseases, Vs has an anti-diabetic value, and Rm is known as an antihypertensive. Phytochemical and pharmacological studies were carried out using standard scientific models to justify their properties. Firstly, the antioxidant, analgesic, anti-inflammatory, and/or vasorelaxant activities of their crude methanol extracts (ME) were evaluated according to ethnomedicinal information. The ME of Ic, Ub, and Vs showed potent antioxidant activities on DPPH and FRAP methods. These species are rich in phenolics, flavonoids, and organic acids, known for their antioxidant activities. They also possess significant analgesic and anti-inflammatory activities, respectively, assessed on the pain model caused by acetic acid (1%) and on inflammatory edema induced by carrageenan in mice. Sitostenone was isolated as an analgesic and anti-inflammatory compound from Ic. Ub and Vs ME significantly reduced the glycemia level after 30 min of glucose loading in mice compared to glibenclamide. Androsta-1,4-dien-3,16-dione was isolated as the vasorelaxant molecule from Rm, responsible for its antihypertensive activity. These results showed that there are some scientific reasons that can justify the therapeutic properties of these plants. It is very important to investigate the local use of biodiversity to identify new bioactive compounds, to support biodiversity conservation and sustainable development projects in Madagascar.

In European countries, the utilisation of chestnut flours (CF) has been increasing in the bakery industry because the CF ingredients provide not only nutritional and health benefits but also improve organoleptic and health-promoting properties. This work aimed to describe the phytochemical properties and antioxidant capacity of chestnut flours from different Castanea spp. genotypes and origins, studying the effects of the addition of CF to traditional wheat-based cookies on their quality and sensory traits. Commercial chestnut flours were also considered. CF used in addition to wheat-based flours may increase the quality and health-promoting value of bakery products for its many benefits: (i) nutritional and phytochemical value; (ii) zero-Km availability in many producing areas (Europe, Asia, Australia, and North and South America); (iii) for these areas, food security is connected to a vulnerable wheat-based food system. Chromatographic and spectroscopic methods were utilised to assess the composition and antioxidant properties of the considered chestnut flours. The sensory value of the prepared chestnut/wheat-based cookies was also assessed by a panel of common consumers (hedonistic test by a 9-points hedonistic scale). Monoterpenes were the main substances in the flour phytocomplex, reaching 80–90% of the total, followed by phenolics (8–12%) and vitamin C in trace (1–3%). Antioxidant capacity ranged from 9.64 ± 0.96 mmol Fe+2 kg−1 DW (BOUC flour from cv Bouche de Bétizac) to 17.33 ± 1.35 mmol Fe+2 kg−1 DW (CANA flour from cv Canalutta). In this research study, the cookies derived from CANA and BOUC flours were considered the most appreciated products by consumers, with values of 7.09 ± 0.46 and 6.88 ± 0.18, respectively. These results confirmed that integrating phytochemical data with sensory results is very important for food industries to obtain a complete description of the analysed flours and consequently of the derived products to produce new bakery products highly appreciated by consumers with high health value in comparison to the traditional products.

In recent years, research has focused on phenolic compounds and their putative role as markers of graft incompatibility. Thus far, no studies have been conducted on the role of phenolic compounds in chestnut (Castanea spp.). The present study investigated the content of phenolic compounds in different combinations of Castanea spp. cultivars and rootstocks. Analyses were performed on the inner and outer tissues of chestnut grafts at two phenological sampling stages. The separation, identification and quantification of the phenolic markers via HPLC were preceded by an ultrasonic green extraction. Two chromatographic methods were tested for a total of 15 phenol compounds. Flavonol compounds were not detected, while cinnamic acids were found in low concentrations. The amount of gallic acid turned out to be higher at the graft union of the incompatible combination (20.11 ± 1.47 mg/100 gFW vs. 8.94 ± 1.08 mg/100 gFW). The same pattern was observed for catechin (15.79 ± 1.83 mg/100 gFW vs. 9.63 ± 1.98 mg/100 gFW). Differences in tannin concentrations seemed to be species-specific, and were apparently not related to graft incompatibility. The present work underlines the potential application of certain phenol compounds for the early prediction of graft incompatibility in Castanea spp.

The production and consumption of nuts are increasing in the world due to strong economic returns and the nutritional value of their products. With the increasing role and importance given to nuts (i.e., walnuts, hazelnut, pistachio, pecan, almond) in a balanced and healthy diet and their benefits to human health, breeding of the nuts species has also been stepped up. Most recent fruit breeding programs have focused on scion genetic improvement. However, the use of locally adapted grafted rootstocks also enhanced the productivity and quality of tree fruit crops. Grafting is an ancient horticultural practice used in nut crops to manipulate scion phenotype and productivity and overcome biotic and abiotic stresses. There are complex rootstock breeding objectives and physiological and molecular aspects of rootstock–scion interactions in nut crops. In this review, we provide an overview of these, considering the mechanisms involved in nutrient and water uptake, regulation of phytohormones, and rootstock influences on the scion molecular processes, including long-distance gene silencing and trans-grafting. Understanding the mechanisms resulting from rootstock × scion × environmental interactions will contribute to developing new rootstocks with resilience in the face of climate change, but also of the multitude of diseases and pests.

Herbal products are now considered among the most important sources of phenolic compounds: the FINNOVER project aimed at the creation and development of sustainable supply chains to extract and use natural biologically active agents. Vitis vinifera is one of the most utilised herbal products derived from buds and sprouts as polyphenolic food supplements for its homeostatic and astringent properties. This research was aimed to describe the antioxidant capacity and the phytochemical composition of V. vinifera herbal products by the application of spectroscopic and chromatographic fingerprints considering phenolics as potential markers to significantly differentiate traditional preparations (macerates) from innovative extracts obtained by an ultrasound extraction from V. vinifera buds. Two different commercial products were also considered. Flavonols were the most abundant class in ultrasound extracts (45%), while phenolic acids were the most important class in traditional macerates (49%) and commercial bud-preparations (about 50%). This study may support the potential use of V. vinifera bud-products (starting from pruning byproducts) as food supplements to integrate human diet with good amounts of phenolics. Finally, the use of different extraction methods on the same plant material could be an important development to produce innovative herbal products with a phytochemical composition similar to traditional preparations.

The present study aimed to evaluate the health-promoting potential of breadfruit (Artocarpus altilis (Parkinson) Fosberg, Moraceae family), a traditional Comorian food, considering the sample variability according to geographic localisation. Moreover, the main aims of this research were also to promote its consumption in the Comoros Islands as potential health-promoting food and evaluate it as a source of bioactive molecules for the food industry thanks to its antioxidant and antibacterial properties. Investigations on biologically active substances were carried out on the extracts obtained from breadfruit flours from five regions of Grande Comore (Ngazidja), the main island in Comoros. Phytochemical screening revealed the presence of tannins and polyphenols, flavonoids, leucoanthocyanins, steroids, and triterpenes. The considered secondary metabolites were phenolic compounds, vitamin C, monoterpenes, and organic acids. The contents of total phenolic compounds (mgGAE/100 g of dry weight—DW) in the extracts ranged from 29.69 ± 1.40 (breadfruit from Mbadjini—ExMBA) to 96.14 ± 2.07 (breadfruit from Itsandra—ExITS). These compounds included flavanols, flavonols, cinnamic acid and benzoic acid derivatives, and tannins which were detected at different levels in the different extracts. Chlorogenic acid presented the highest levels between 26.57 ± 0.31 mg/100 g DW (ExMIT) and 43.80 ± 5.43 mg/100 g DW (ExMBA). Quercetin was by far the most quantitatively important flavonol with levels ranging from 14.68 ± 0.19 mg/100 g DW (ExMIT) to 29.60 ± 0.28 mg/100 g DW (ExITS). The extracts were also rich in organic acids and monoterpenes. Quinic acid with contents ranging from 77.25 ± 6.04 mg/100 g DW (ExMBA) to 658.56 ± 0.25 mg/100 g DW of ExHAM was the most important organic acid in all the breadfruit extracts, while limonene was quantitatively the main monoterpene with contents between 85.86 ± 0.23 mg/100 g DW (ExMIT) and 565.45 ± 0.24 mg/100 g DW (ExITS). The antibacterial activity of the extracts was evaluated on twelve pathogens including six Gram (+) bacteria and six Gram (−) bacteria. By the solid medium disc method, except for Escherichia coli and Pseudomonas aeruginosa, all the bacteria were sensitive to one or more extracts. Inhibitory Halo Diameters (IHDs) ranged from 8 mm to 16 mm. Salmonella enterica, Clostridium perfringens, and Vibrio fischeri were the most sensitive with IHD > 14 mm for ExITS. By the liquid microdilution method, MICs ranged from 3.12 mg/mL to 50 mg/mL and varied depending on the extract. Bacillus megaterium was the most sensitive with MICs ≤ 12.5 mg/mL. Pseudomonas aeruginosa, Shigella flexneri, and Vibrio fischeri were the least sensitive with all MICs ≥ 12.5 mg/mL. ExHAM was most effective with a MIC of 3.12 mg/mL on Staphylococcus aureus and 6.25 mg/mL on Salmonella enterica. The antioxidant power of the extracts was evaluated by the FRAP method. The activity ranged from 5.44 ± 0.35 (ExMBA) to 14.83 ± 0.11 mmol Fe2+/kg DW (ExHAM). Breadfruit from different regions of Comoros contained different classes of secondary metabolites well known for their important pharmacological properties. The results of this study on phenolics, monoterpenes, and organic acids have provided new data on these fruits. The obtained results showed that breadfruit from the biggest island of the Union of Comoros also presented antimicrobial and antioxidant properties, even if some differences in effectiveness existed between fruits from different regions.

Foods confer many health-promoting benefits to humans for the treatment/prevention of different diseases [...]

New technology development and globalisation have led to extreme changes in the agri-food sector in recent years that need an important food supply chain characterisation from plant materials to commercial productions. Many analytical strategies are commonly utilised in the agri-food industry, often using complementary technologies with different purposes. Chromatography on-line coupled to mass spectrometry (MS) is one of the most selective and sensitive analytical methodologies. The purpose of this overview is to present the most recent MS-based techniques applied to food analysis. An entire section is dedicated to the recent applications of high-resolution MS. Covered topics include liquid (LC)– and gas chromatography (GC)–MS analysis of natural bioactive substances, including carbohydrates, flavonoids and related compounds, lipids, phenolic compounds, vitamins, and other different molecules in foodstuffs from the perspectives of food composition, food authenticity and food adulteration. The results represent an important contribution to the utilisation of GC–MS and LC–MS in the field of natural bioactive compound identification and quantification.

Anxiety disorders are common and complex psychiatric syndromes affecting a broad spectrum of patients. On top of that, we know that aging produces an increase in anxiety vulnerability and sedative consumption. Moreover, stress disorders frequently show a clear gender susceptibility. Currently, the approved pharmacological strategies have severe side effects such as hallucinations, addiction, suicide, insomnia, and loss of motor coordination. Dietary integration with supplements represents an intriguing strategy for improving the efficacy and the safety of synthetic anxiolytics. Accordingly, a recent article demonstrated that glyceric bud extracts from Tilia tomentosa Moench (TTBEs) exert effects that are consistent with anxiolytic activity. However, the effects of these compounds in vivo are unknown. To examine this question, we conducted behavioral analysis in mice. A total of 21 days of oral supplements (vehicle and TTBEs) were assessed by Light Dark and Hole Board tests in male and female mice (young, 3 months; old, 24 months). Interestingly, the principal component analysis revealed gender and age-specific behavioral modulations. Moreover, the diet integration with the botanicals did not modify the body weight gain and the daily intake of water. Our results support the use of TTBEs as dietary supplements for anxiolytic purposes and unveil age and gender-dependent responses.

Changes in lifestyle and demographics, rising consumer incomes, and shifting preferences due to advanced knowledge about the relationships between food and health contribute to generate new needs in the food supply. Today, the role of food is not only intended as hunger satisfaction and nutrient supply but also as an opportunity to prevent nutrition-related diseases and improve physical and mental well-being. For this reason, there is a growing interest in the novel or less well-known plant foods that offer an opportunity for health maintenance. Recently, interest in plant foods and underutilized fruits is continuously growing, and agrobiodiversity exploitation offers effective and extraordinary potentialities. Plant foods could be an important source of health-promoting compounds and functional food ingredients with beneficial properties: the description of the quality and physicochemical traits, the identification and quantification of bioactive compounds, and the evaluation of their biological activities are important to assess plant food efficacy as functional foods or source of food supplement ingredients.

Saffron is a well-known spice, produced for a long time, mainly in the Mediterranean area. In the last few years, saffron has been seen as an alternative crop for the diversification of agricultural production and a new source of income, due to its high price, including in Alpine areas. Apart from the spice, constituted by the stigmas, saffron production provides a high amount of fresh tepals, which have so far been considered agricultural waste. Recently, studies on the composition of saffron tepals have been revealing their potential as a rich source of bioactive compounds. In this study, we evaluated the productive traits and the quality of the spice of saffron cultivated in open fields in the north western Italian Alps, for 2 years. In addition, in order to valorize the crop residues, we evaluated the bioactive compound content in tepals by using maceration and ultrasound assisted extraction with different rates of water and methanol as solvents. Higher yields in spice were obtained in the second year of cultivation. However, in both years, the spice had a high quality (ISO 3632), and a very high total phenolic content and antioxidant activity. Thirteen bioactive compounds were identified, including flavonols, cinnamic acids, benzoic acids, catechins, and vitamin C, with few differences between sites and year of cultivation. In tepals, the extraction method and the solvent used influenced the evaluated parameters, i.e., total phenolics and total anthocyanins content, antioxidant activity, and the amount of the four phenolic compounds found (hyperoside, rutin, ellagic acid, and epicatechin). Overall, this study revealed that both saffron spice and its by-product, the fresh tepals, produced in the north western Italian Alps can be considered a source of bioactive compounds with nutraceutical properties, having an antioxidant capacity that is often similar or higher than those of some vegetables and fruits.

The use of herbal food supplements, as a concentrate form of vegetable extracts, increased so much over the past years to count them among the relevant sources of dietetic polyphenols. Bud-derivatives are a category of botanicals perceived as a “new entry” in this sector since they are still poorly studied. Due to the lack of a manufacturing process specification, very different products can be found on the market in terms of their polyphenolic profile depending on the experimental conditions of manufacturing. In this research two different manufacturing processes, using two different protocols, and eight species (Carpinus betulus L., Cornus mas L., Ficus carica L., Fraxinus excelsior L., Larix decidua Mill., Pinus montana Mill., Quercus petraea (Matt.) Liebl., Tilia tomentosa Moench), commonly used to produce bud-derivatives, have been considered as a case study. An untargeted spectroscopic fingerprint of the extracts, coupled to chemometrics, provide to be a useful tool to identify these botanicals. The targeted phytochemical fingerprint by HPLC provided a screening of the main bud-derivatives polyphenolic classes highlighting a high variability depending on both method and protocol used. Nevertheless, ultrasonic extraction proved to be less sensitive to the different extraction protocols than conventional maceration regarding the extract polyphenolic profile.

A large species diversity characterises the wide distribution of chestnuts in Asia, North America, and Europe, hence reflecting not only the adaptation of the genus Castanea to diverse environmental conditions, but also to different management strategies encompassing orchards. The characterisation and description of chestnut populations and cultivars are crucial to develop effective conservation strategies of one of the most important Italian and European fruit and wood species. Chestnut cultivars grown in the same pedoclimatic conditions and on the same clonal rootstock were characterised with sensory, spectrophotometric, and chromatographic analysis to determine the phytochemical composition and nutraceutical properties. A multivariate approach, including principal component analysis and conditional inference tree models, was also performed. The ease of peeling, seed colour, and intensity of sweetness were the sensory descriptors that allowed us to differentiate C. sativa cultivars. Antioxidant capacity ranged from 9.30 ± 0.39 mmol Fe+2 kg−1 DW (‘Bouche de Bètizac’) to 19.96 ± 1.89 mmol Fe+2 kg−1 DW (‘Garrone Rosso’). Monoterpenes represented the main component, reaching 88% for hybrids, followed by polyphenols (10–25% for hybrids and chestnuts, respectively). A multivariate approach showed that phenolic acids and tannins were the bioactive classes with the highest discriminating power among different genotypes, and that genotype is a significant variable (p < 0.05). In addition, most of the analysed chestnut cultivars showed a content of bioactive compounds similar to or higher than the main hazelnut, walnut, and almond varieties. Chestnut agrobiodiversity could be intended as strictly associated to the genotype effect and underlines the large variability within the genus Castanea, and therefore, the importance of in farm and ex situ conservation of local germplasm is part of a global strategy aimed at increasing the levels of agrobiodiversity.

Autumn olive (Elaeagnus umbellata Thunb.) is a deciduous shrub tree widely distributed in Asia and Southern Europe and grown as ornamental species. It is locally used for human consumption, as relevant medical value is attributed to the berries. Information about its composition, especially concerning the characterization of bioactive and health-promoting compounds, is limited. The aim of the present study is to evaluate the main bioactive compounds and nutraceutical proprieties of autumn olive fruits, via high-performance liquid chromatography (HPLC) fingerprint and spectrophotometric analysis, in order to strengthen the knowledge about this underrated species and promote potential applications as a food supplement. Concerning nutraceutical traits, total polyphenolic content (325.366 ± 13.019 mg of gallic acid equivalents (mg GAE)/100 of fresh weight (g FW)) and total anthocyanin content (194.992 ± 0.817 mg of cyanidin-3-O-glucoside (mg C3G)/100 g FW) recorded considerable values. The phytochemical fingerprint revealed the presence 23 bioactive compounds. Polyphenols (65.56%) were the largest class, followed by monoterpenes (27.40%) and vitamin C (7.04%). Anthocyanins were the most represented compounds among polyphenols (71.9%). The antioxidant capacity (20.031 ± 1.214 mmol Fe2+/kg) was similar to that recorded for other small fruits with proven health-promoting properties. The present work underlined the potential of E. umbellata as a source of health-promoting bioactive compounds. Further studies should deepen the knowledge of nutraceutical aspects, which turned out to be interesting.

Uapaca bojeri is an endemic Malagasy plant used by the local population. This work aimed to evaluate antioxidant, anti-inflammatory, and antidiabetic activities of the methanol extracts of U. bojeri leaves and stems and to report their total phenolic content and the bioactive compound content by HPLC methods. Antioxidant capacity was determined by DPPH and ferric reducing antioxidant power (FRAP) assays. An in vivo carrageenan-induced paw oedema and acetic acid-induced writhing test in mice were used for anti-inflammatory activity evaluation. An oral glucose tolerance test was performed in mice to evaluate antidiabetic activity. The total bioactive compound content of leaves was higher than that of stems. Stem methanol extract inhibited the free radical DPPH more than the leaf methanol extract. Leaf methanol extract inhibited, in a dose-dependent manner, the carrageenan-induced paw oedema more than the stem extract, but their inhibition of the pain symptoms caused an acetic acid-induced decrease similar to the number of writhes in the dose-dependent case. The leaf and stem methanol extracts significantly reduced blood glucose levels after 30 min of glucose loading in mice compared to the control group blood glucose reduction. The presence of several bioactive compounds in U. bojeri contributed to the different biological activities, but isolation and identification of these bioactive molecules are necessary to confirm these pharmacological properties.

Comoros Union presents a considerable biodiversity of food resources that are neglected or still not valorised, as breadfruit. This study aimed to evaluating nutritional and nutraceutical traits of Artocarpus altilis (Parkinson) Fosberg by characterizing its main bioactive compounds, nutritional traits, and antioxidant properties in order to contribute to the development of traditional and innovative uses of this species as functional food (e.g., infant flour). Bioactive compound composition, antioxidant properties, protein and sugar content, lipids, fibre, and macro- and microelements were observed in these fruits after a specific drying process. Breadfruit showed positive nutritional traits. The main identified phenolic groups were cinnamic acids (with a maximum of 51.88 ± 2.63 mg/100 gDW for chlorogenic acid) and tannins. The highest value of antioxidant activity was 6.40 ± 1.02 mmol·Fe2+/kgDW. This preliminary phytochemical investigation may provide a contribution to the identification and quantification of lead compounds responsible for traditional nutritional and therapeutic claims.

Madagascar is characterized by one of the highest rates of endemism and biodiversity in the world. Brachylaena preparations are extensively used in Malagasy folk medicine for treating gastrointestinal diseases and blenorrhagia. The aim of this study was a preliminary phytochemical fingerprint of Brachylaena ramiflora leaves infusions and bark decoctions, in order to characterize this species as source of biologically active compounds and their relative antioxidant activity by high-performance liquid chromatography-diode array detector. Sixteen and twenty-three biomarkers (molecules with health properties selected for their demonstrated positive role on human organism) were identified in B. ramiflora leaf infusions and bark decoctions, respectively: the main compounds identified in the infusions were quinic acid (334.55±0.99 mg/100 gFW), chlorogenic acid (208.27±7.74 mg/100 gFW), and g-terpinene (144.19±1.00 mg/100 gFW), while the major components in the decoctions were castalagin (2002.64±13.96 mg/100 gFW), citric acid (1171.81±1.05 mg/100 gFW), and chlorogenic acid (646.44±2.31 mg/100 gFW). B. ramiflora could be considered as a promising source of natural antioxidants that may provide health-benefits. The development of pharmaceuticals based on a sustainable exploitation of wild medicinal plants or their cultivation by local villagers could offer a number of benefits to a wide range of people as an alternative source of income and a natural and accessible health remedy. In this research, for the first time, an alternative method to produce Ribes nigrum bud derivatives is presented. Pulsed ultrasound-assisted extraction (PUAE), using a food-grade solvent according to green chemistry principles, has been employed and compared to the conventional extraction method. Traditionally, bud derivatives, a category of botanicals marketed as plant food supplements in the European Community, are produced by macerating meristematic tissues of trees and plants mainly spontaneously collected. Buds are a challenging raw material for the UAE, since meristematic tissues are much softer and fragile than their corresponding adult phenological stage. It is therefore important to assess whether the polyphenolic fraction, very susceptible to degradation, is conserved after UAE. Untargeted polyphenolic fingerprints (UV-Visible and fluorescence) coupled with chemometrics are employed to quickly screen the best extraction conditions, evaluated by the design of experiment (DoE) method. The polyphenolic fraction of the optimized PUAE extract was quantified by targeted HPLC fingerprint and its antiradical activity was determined. PUAE on a lab pilot reactor was proven to be the most practical approach for a rapid (20 min vs. 21 days maceration) and efficient extraction of bioactive polyphenolics from Ribes nigrum buds, encouraging the scale up to an industrial plan.

Dried fruits are important, healthy and popular snacks, despite the limited information on their nutritional profiles and phytochemical composition. The present work was aimed to study the chemical composition of freeze-dried fruits from four fruit species: two common commercial snacks (apple and goji) and two innovative products (kaki and kiwi). Sugar and organic acid levels, total phenolics (TPC), and main health-promoting phytochemicals were studied by HPLC fingerprinting analysis. Furthermore, in vitro antioxidant capacity (AOC) was preliminarily observed in these products. A Principal Component Analysis (PCA) was carried out as a multivariate approach as well. The TPC ranged from 210.9 mg GAE/100g DW (kiwi) to 872.6 mg GAE/100g DW (kaki), while dried fruit antioxidant capacity ranged from 23.09 mmol Fe2+/kg DW (goji) to 137.5 mmol Fe2+/kg DW (kaki). The most important phytochemical class in apple (two cultivars), kiwi, and kaki dried fruits was phenolics (from 74.6% to 93.3%), while monoterpenes were the first class in goji (67.5%). No anthocyanins have been identified in dried fruits because these compounds are most likely converted to phenolic acids during the drying process. This research intended to stimulate large-scale exploitation of commercial dried fruits as functional foods as well.

Saffron (Crocus sativus L.) is cultivated worldwide. Its stigmas represent the highest-priced spice and contain bioactive compounds beneficial for human health. Saffron cultivation commonly occurs in open field, and spice yield can vary greatly, from 0.15 to 1.5 g m−2, based on several agronomic and climatic factors. In this study, we evaluated saffron cultivation in soilless systems, where plants can benefit from a wealth of nutrients without competition with pathogens or stresses related to nutrient-soil interaction. In addition, as plant nutrient and water uptake can be enhanced by the symbiosis with arbuscular mycorrhizal fungi (AMF), we also tested two inocula: a single species (Rhizophagus intraradices) or a mixture of R. intraradices and Funneliformis mosseae. After one cultivation cycle, we evaluated the spice yield, quality (ISO category), antioxidant activity, and bioactive compound contents of saffron produced in soilless systems and the effect of the applied AMF inocula. Spice yield in soilless systems (0.55 g m−2) was on average with that produced in open field, while presented a superior content of several health-promoting compounds, such as polyphenols, anthocyanins, vitamin C, and elevated antioxidant activity. The AMF symbiosis with saffron roots was verified by light and transmission electron microscopy. Inoculated corms showed larger replacement corms (+50% ca.). Corms inoculated with R. intraradices performed better than those inoculated with the mix in terms of spice quality (+90% ca.) and antioxidant activity (+88% ca.). Conversely, the mixture of R. intraradices and F. mosseae increased the polyphenol content (+343% ca.). Thus, soilless systems appeared as an effective alternative cultivation strategy for the production of high quality saffron. Further benefits can be obtained by the application of targeted AMF-based biostimulants.

Arbuscular mycorrhizal fungi (AMF) establish mutualistic symbiotic associations with plant roots and act as biofertilizers by enhancing plant nutrient and water uptake. Moreover, AMF colonization may influence the biosynthesis of plant bioactive compounds in medicinal and aromatic plants. There is limited information on AMF associations with Crocus sativus L. (saffron) roots and their effect on crop performances and spice quality. In the present work we verified the efficiency of root mycorrhization in potted conditions, then we evaluated the yield and quality of the saffron produced in two Alpine sites during two cultivation cycles with the application of AMF. Two inocula were applied, either a single-species (Rhizophagus intraradices) or a multi-species mixture (R. intraradices and Funneliformis mosseae). The trial conducted in potted conditions confirmed that both AMF commercial inocula established symbiotic relationships with saffron roots. The multi-species inoculation yielded the highest content of arbuscules in colonized portions of the root (100%), while the single-species was slightly less (82.9%) and no AMF were recorded in untreated control corms. In open-field conditions, AMF colonization of the root systems, flower production, and saffron yields were monitored, and bioactive compounds contents and antioxidant activity in the dried spice were analysed using spectrophotometry and high performance liquid chromatography. Overall, the saffron produced was high quality (ISO category) and had high contents of bioactive compounds, with very high total polyphenol content and elevated antioxidant activity. The use of arbuscular mycorrhizal symbionts as biostimulants positively affected saffron cultivation, improving the crop performances and the content of important nutraceutical compounds. In particular, the inoculum composed by R. intraradices and F. mosseae increased flower production and the saffron yield. R. intraradices alone enhanced the spice antioxidant activity and the content of bioactive compounds such as picrocrocin, crocin II, and quercitrin. Since saffron is the world’s highest priced spice, the increases in yield and quality obtained using AMF suggests that farms in marginal areas such as alpine sites can increase profitability by inoculating saffron fields with arbuscular mycorrhiza.

The influence of roasting temperature on the final quality of Piedmont hazelnut (cultivar Tonda Gentile delle Langhe) was evaluated. Sensory and colorimetric analyses were performed to define a quality profile of the fruit relating to the roasting technique parameters. Sensory analysis was conducted on roasted kernels through a trained panel. The effects of the different temperatures on colour were evaluated with two different techniques, colorimeter and scan-marker (scanner), in order to define the best method to individuate correlations between roasting temperature and colorimetric results. The quality traits of nuts roasted at different temperatures were compared: the samples roasted at lower temperature showed lighter colour; the hue values were higher in the roasted chopped nuts, while the chroma was higher in the paste samples. For both paste and chopped kernels a larger range of values was found in comparison with the results given by the colorimeter. As for the sensory analysis, we found that "hazelnut odour" varied significantly among the samples: this descriptor is able to significantly differentiate between different samples. Our study showed that the modulation temperature is a very important parameter with a strong influence on the characteristic roasted flavour and colour of the product.

In the Comoros Islands, as in other developing countries, malnutrition and food insecurity affect a very large percentage of the population. Developing fruit-based products in order to make profit, reduce poverty and improve indigenous people diet could be very important for local population of countries as Comoros Islands. The aim of the present work was to study the chemical composition of jams and jellies produced from seven fruit species harvested in Grand Comore Island. The following parameters were studied sugars and organic acids, total phenolics, total anthocyanins and high-performance liquid chromatography (HPLC) fingerprint of the main phytochemicals. Antioxidant activity was also measured. A multivariate approach (Principal Component Analysis) was performed in order to better characterize the products and to set a potential analytical tool for jam characterisation. Results showed that the analysed products are a good source of polyphenolic constituents, as caffeic and gallic acids, catechin and quercetin and volatile compounds, as limonene and γ-terpinene: these molecules may be considered as suitable markers for these fruit-derived products as characterizing the chromatographic patterns. The characterisation of these products and their nutritional and nutraceutical traits is important as valorisation of local food production for poverty reduction and rural development. Further benefits of this approach include the maintenance of local agro-biodiversity as raw material for fruit-based products and the strengthening of food security practices.

European bird cherry (Prunus padus L.) has been known since the Middle Ages for its medical/food use and high health-promoting value. This study aimed to assess the potential of these fruits as a source of bioactive compounds through the characterization of its physicochemical traits, nutraceutical properties, phytochemical composition via HPLC fingerprint, and antioxidant capacity. Fully ripened fruits of Prunus padus L. (Colorata cv) were collected in mid-July 2017 in Chieri, north-western Italy. The TPC (194.22 ± 32.83 mgGAE/100 gFW) and TAC (147.42 ± 0.58 mgC3G/100 gFW) values were obtained from the analyzed extracts. The most important phytochemical class was organic acids (48.62 ± 2.31%), followed by polyphenols (35.34 ± 1.80%), monoterpenes (9.36 ± 0.64%), and vitamin C (6.68 ± 0.22%). In this research the most important flavonols selected as marker were quercitrin (16.37 ± 3.51 mg/100 gFW) and quercetin (11.86 ± 2.36 mg/100 gFW). Data were reported based on fresh weight. Moreover, fresh fruits showed a mean antioxidant activity value of 17.78 ± 0.84 mmol Fe2+·kg−1. Even though the seeds and leaves contain cyanogenic glycosides, this study showed that these fruits could be a natural source of bioactive compounds with high antioxidant properties, due to the contents of organic and phenolic acids, catechins, and a synergetic effect of vitamin C and flavonoids.

It is well known that plants are important sources for the preparation of natural remedies as they contain many biologically active compounds: in particular, phenolic compounds are one of the most widely occurring groups of phytochemicals. Some endemic species may be used for the production of herbal preparations containing phytochemicals with significant antioxidant capacities and health benefits: blackberry sprouts and blackcurrant buds are known to contain appreciable levels of phenolic compounds, including flavonols, phenolic acids and catechins. The aim of this research was to perform an analytical study of blackcurrant and blackberry bud-preparations, in order to identify the main bioactive polyphenolic compounds, to study the total polyphenolic content and to obtain a specific profile of the main polyphenols contained in these products using a High Performance Liquid Chromatograph - Diode Array Detector; the same analyses were performed both on the University lab preparations and on commercial preparations. Differentchromatographic methodswere usedto determine concentrations of phytochemical compounds in the preparations, allowing to quantifystatistically significant differencesin their polyphenolic content both in the case of Ribes nigrum and Rubus ulmifolius. The assessment of chemical composition and bioactivities of the plant-derived products could help in find out new sources of natural antioxidants and other health-promoting compounds: only with the deep knowledge of the bioactive composition of plant preparations it will be possible to develop a new generation of standardized, effect-optimized, mono- and multi-extract preparations.

The purpose of this study was to evaluate the effects of the application of an agro-industrial waste extract (AWE) on the quality of kiwifruit. The AWE was obtained by extraction from apple seeds, rapeseed and rice husks and then Boron (0,6%) and Zinc (1,4%) were added. The effect of AWE as a fertilizer/biostimulant on several parameters of kiwifruit quality (weight, total soluble solids, firmness, dry matter percentage, pH, titratable acidity, antioxidant capacity, ascorbic acid) was then evaluated. The application was carried out on two cultivars of Actinidia deliciosa, Hayward and Green Light, and also in two different cultivation environments. AWE increased the dry matter percentage in Green Light and in Hayward fruits grown in Piedmont but no increase was observed for Hayward fruits grown in Latium. The most relevant effect of AWE was the significant increase of the Ascorbic Acid (AA) content in the fruits of all the tested orchards. Fertilization with biostimulants shows a promising future in functional plant nutrition linked to an increase in food quality parameters.

Berry fruits show high antioxidant capacity among plant foods. Medical research has uncovered medicinal properties of pigmented polyphenols, such as flavonoids, anthocyanins, tannins and other phytochemicals, mainly localized in berry skin and seeds. The aim of this work was to contribute to the study of nutraceutical features of some berry fruits (currants, gooseberries and strawberries). The content in different antioxidant compounds in fresh fruits of different cultivars and selections of Ribes spp. and Fragaria x ananassa Duch was analyzed . Fruits of 29 cultivars of 3 different species of Ribes spp. and 5 cultivars of strawberry were analysed by High Performance Liquid Chromatograph coupled to a UV/Vis detector and a mass detector (MS) to identify and quantify the main antioxidant compounds. Regarding the cultivars of Ribes spp., it was confirmed the presence of a high content in phenolic compounds, representing, therefore, an important source of antioxidant compounds. Moreover, results showed that the considered cultivars and selections of strawberries are a good source of bioactive compounds, especially phenolics. The results of this study contributed to give some new insights into the nutraceutical aspects of the considered berry fruit species.

It is well known that plants are important sources for the preparation of natural remedies as they contain many biologically active compounds. In particular, polyphenols, terpenic compounds, organic acids, and vitamins are the most widely occurring groups of phytochemicals. Some endemic species may be used for the production of herbal preparations containing phytochemicals with significant bioactivity, as antioxidant activity and anti-inflammatory capacities, and health benefits. Blackberry sprouts and blackcurrant buds are known to contain appreciable levels of bioactive compounds, including flavonols, phenolic acids, monoterpenes, vitamin C, and catechins, with several clinical effects. The aim of this research was to perform an analytical study of blackcurrant and blackberry bud-preparations, in order to identify and quantify the main biomarkers, obtaining a specific phytochemical fingerprint to evaluate the single botanical class contribution to total phytocomplex and relative bioactivity, using a High Performance Liquid Chromatograph−Diode Array Detector; the same analyses were performed both on the University laboratory and commercial preparations. Different chromatographic methods were used to determine concentrations of biomolecules in the preparations, allowing for quantification of statistically significant differences in their bioactive compound content both in the case of Ribes nigrum and Rubus cultivated varieties at different harvest stages. In blackcurrant bud-extracts the most important class was organic acids (50.98%) followed by monoterpenes (14.05%), while in blackberry preparations the main bioactive classes were catechins (50.06%) and organic acids (27.34%). Chemical, pharmaceutical and agronomic-environmental knowledge could be important for obtaining label certifications for the valorization of specific genotypes, with high clinical and pharmaceutical value: this study allowed to develop an effective tool for the natural preparation quality control and bioactivity evaluation through the chemical fingerprinting of bud preparations.
