Abstract

We hypothesized that small molecule transcriptional perturbation could be harnessed to target a cellular dependency involving protein arginine methyltransferase 5 (PRMT5) in the context of methylthioadenosine phosphorylase (MTAP) deletion, seen frequently in malignant pleural mesothelioma (MPM). Here we show, that MTAP deletion is negatively prognostic in MPM. In vitro, the off-patent antibiotic Quinacrine efficiently suppressed PRMT5 transcription, causing chromatin remodelling with reduced global histone H4 symmetrical demethylation. Quinacrine phenocopied PRMT5 RNA interference and small molecule PRMT5 inhibition, reducing clonogenicity in an MTAP-dependent manner. This activity required a functional PRMT5 methyltransferase as MTAP negative cells were rescued by exogenous wild type PRMT5, but not a PRMT5E444Q methyltransferase-dead mutant. We identified c-jun as an essential PRMT5 transcription factor and a probable target for Quinacrine. Our results therefore suggest that small molecule-based transcriptional perturbation of PRMT5 can leverage a mutation-selective vulnerability, that is therapeutically tractable, and has relevance to 9p21 deleted cancers including MPM.

11. The 8 billion milestone: Risk perceptions of global population growth among UK and US residents.

Abstract

In November 2022, the global human population reached 8 billion and is projected to reach 10 billion by 2060. Theories, models, and evidence indicate that global population growth (GPG) increases the likelihood of many adverse outcomes, such as biodiversity loss, climate change, mass migrations, wars, and resource shortages. A small body of research indicates that many individuals are concerned about the effects of GPG, and these concerns are strongly related to the willingness to engage in mitigative and preventative actions. However, scientific understanding of the factors that influence GPG risk perceptions remains limited. To help address this research gap, we conducted a study of the perceived risk of GPG among UK and US residents (N = 1029) shortly after the "8 billion milestone." Our results confirmed that GPG is perceived as a moderate‐to‐high risk and these perceptions have a strong positive relationship with the willingness to engage in and support risk management actions. Our participants believed that the worst effects of GPG were yet to come but would largely be geographically and socially remote. Despite their willingness to engage in risk management actions, our participants reported low self‐efficacy and that governments (cf. individuals and communities) have the greatest capacity to influence GPG. Risk perceptions were strongly predicted by worldviews and were higher among our UK (cf. US) participants. We also found that the perceived benefits of GPG were low and found no evidence to suggest that risk perceptions were affected by exposure to media coverage of the 8 billion milestone. Abstract

Insular evolution on archipelagos generates a significant proportion of global biodiversity, yet islands are among the ecosystems most sensitive to accelerating anthropogenic disturbance, introductions of non‐native species, and emerging pathogens, among other conservation challenges. The Alexander and Haida Gwaii archipelagos along North America's North Pacific Coast support a disproportionate number of endemic taxa compared to other high‐latitude terrestrial ecosystems. In this region, endemics in Canada are explicitly protected, but in the United States, endemics have been operationally ignored. We reviewed regional research on terrestrial mammals and endemics from 2000–2022 to guide wildlife management. Elevated regional endemism is due to a combination of deep and shallow temporal processes (i.e., long‐term refugial isolation vs. recent colonization). With adequate sampling, genomic analyses are well‐suited to identifying nuanced patterns of divergence and endemism, thereby facilitating a deeper understanding of regional diversity. We identified 18 mammalian endemics in Southeast Alaska, USA, at varying taxonomic scales, but research effort has significant taxonomic biases and sampling infrastructure remains inadequate. Of the 66 terrestrial and aquatic mammal species in Southeast Alaska, only 55% are represented by ≥10 archived samples over the last 2 decades. Across taxa, major spatial and temporal sampling gaps limit interpretations of wildlife responses to changing environmental conditions. The Tongass National Forest is spread across an island archipelago, and climate change is projected to have disproportionate impacts on island endemics worldwide. In this case, the United States Forest Service is not closely monitoring endemic taxa, as was required by the Tongass Land Management Plan in 1997. Our review underscores a need for increased consideration of how endemism can be incorporated into land and wildlife management across the Alexander Archipelago. Moving forward, we encourage state and federal agencies, Indigenous communities, and international collaborators to continue to partner with natural history biorepositories to ensure strategic wildlife sampling infrastructure is built and made accessible to the broader scientific community as part of the land management process. Abstract

The Muller F element (4.2 Mb, ~80 protein-coding genes) is an unusual autosome of Drosophila melanogaster; it is mostly heterochromatic with a low recombination rate. To investigate how these properties impact the evolution of repeats and genes, we manually improved the sequence and annotated the genes on the D. erecta, D. mojavensis, and D. grimshawi F elements and euchromatic domains from the Muller D element. We find that F elements have greater transposon density (25-50%) than euchromatic reference regions (3-11%). Among the F elements, D. grimshawi has the lowest transposon density (particularly DINE-1: 2% vs. 11-27%). F element genes have larger coding spans, more coding exons, larger introns, and lower codon bias. Comparison of the Effective Number of Codons with the Codon Adaptation Index shows that, in contrast to the other species, codon bias in D. grimshawi F element genes can be attributed primarily to selection instead of mutational biases, suggesting that density and types of transposons affect the degree of local heterochromatin formation. F element genes have lower estimated DNA melting temperatures than D element genes, potentially facilitating transcription through heterochromatin. Most F element genes (~90%) have remained on that element, but the F element has smaller syntenic blocks than genome averages (3.4-3.6 vs. 8.4-8.8 genes per block), indicating greater rates of inversion despite lower rates of recombination. Overall, the F element has maintained characteristics that are distinct from other autosomes in the Drosophila lineage, illuminating the constraints imposed by a heterochromatic milieu.

25. A Descriptive Analysis of the Perspectives of Neville High School's Teachers Regarding the School's Renovation.

Abstract

It has been suggested since the early 80's that America's public schools are not meeting the needs of America's citizenry and that the schools are in need of repair. This paper describes a study conducted at Neville High School (Louisiana) in 1996-1997 and designed to examine the effects of facility renovation on faculty morale. Data collection included comments from 10 teachers along with direct observations, participant observation, and document review. Questions considered participant views of the renovation process; their levels of frustration; their concerns over changes in certain physical properties that may effect school tradition and heritage; and feelings of pride, motivation, and new attitudes. Findings show a wide spread of competing emotions, e.g., participants expressed negative comments about change while indicating how excited they were about the newness of it all. Despite numerous negative comments, overall morale appeared to be higher as a result of the renovations. Teachers were more active, showed greater collegiality, and were interested in showing visitors the newly renovated areas. Appendices describe the participants and present the history of the school. 26. Understanding and improving people's judgments of synergistic risks

Abstract

Certain hazard combinations present a risk that is greater than the sum of the risk attributable to each constituent hazard. These ‘synergistic risks’ occur in several domains, can vary in magnitude, and often have life-threatening consequences. However, research concerning the extent to which people understand synergistic risks is in its infancy, and extant studies investigating this topic have encountered problems in identifying valid measures of subjective risk judgments for combined hazards. Consequently, few firm conclusions can be made about the extent to which people understand synergistic risks. This thesis presents four original research papers that aim to provide greater insight into peoples’ judgments of synergistic risks, and investigates how such judgments may be assessed and improved. Each of the studies presented in the four papers employs data obtained via questionnaires specifically designed to address each research question. In the first paper, two studies are presented that examine whether people believe that combined hazards can present synergistic risks. In the second paper, qualitative data is analysed to explore the cognitive reasoning that individuals employ when assessing the risk for combined hazards. The third paper presents a study that assess a new metric for the assessment of risk judgments for combined hazards, and a second study in which domain-experts’ and non-experts’ judgments are compared. The final paper features a study that investigates which message content (i.e., antecedent vs. probabilistic data) most effectively informs people about synergistic risks. The results show that many people can make veridical judgments of synergistic risks. The findings indicate such judgments may depend on factors that include hazard-specific knowledge, judgmental experience and a rudimentary awareness of an xiii underlying causal mechanism for the increased risk. However, many people also make non/less veridical judgments; often underestimating the magnitude of the synergistic risk or employing an additive risk model which corresponds to the notion of ‘adding’ one hazard to another. Furthermore, the findings suggest risk judgments for combined hazards do not vary according to hazard domain but, rather, according to the hazard characteristics. Importantly, the research also identifies both (a) a valid method of assessing peoples’ risk judgments for combined hazards, and (b) risk communications contents that can lead to significant improvement in individuals’ understanding of synergistic risks

Abstract

Study designPosterolateral L4-L5 transverse process fusions were done on 14 adult beagles. Six were implanted with recombinant human bone morphogenetic protein-2 carried by open-cell polylactic acid polymer delivery vehicle. Six received autogenous iliac bone graft. Two received carrier alone. Eleven were killed 3 months after implantation. One in each group was maintained for 8 months.ObjectivesTo compare recombinant human bone morphogenetic protein-2 and open-cell polylactic acid polymer with autogenous iliac bone for inducing transverse process fusion in the canine by 3 months and to determine whether transverse process decortication and implantation of carrier alone causes spontaneous transverse process fusion in the canine.Summary of background dataRecombinant human bone morphogenetic proteins have healed segmental long bone defects in several models. They have induced interlaminar and facet fusions in canines. Interlaminar and facet fusions have occurred after sham decortications in canines. Recombinant human bone morphogenetic protein-2 has not been evaluated for transverse process fusion in canines. Transverse process fusion is a preferred clinical method for achieving posterior lumbar fusion.MethodsFusion sites were evaluated by serial computed tomography scans. After the dogs were killed, explanted spines were subjected to manual testing, mechanical testing, high resolution radiography, and histologic analysis.ResultsOne hundred percent of recombinant human bone morphogenetic protein-2-implanted sites had solid transverse process fusion by 3 months according to all measures. No autografted sites were fused at this interval. Osseous bridging of posterolateral gutters occurred in the recombinant human bone morphogenetic protein-2-implanted sites after 2 months, the earliest radiographic measure. None of the carrier-only sites showed bone formation.ConclusionsRecombinant bone morphogenetic protein-2 carried by open-cell polyactic acid polymer is superior to autogenous iliac bone for producing radiographically and mechanically solid transverse process fusions in canines by 3 months. Spontaneous transverse process fusion does not occur in canines after decortication and open-cell polylactic acid polymer implantation.

This initiative examined systematically the extent to which a large set of archival research findings generalizes across contexts. We repeated the key analyses for 29 original strategic management effects in the same context (direct reproduction) as well as in 52 novel time periods and geographies; 45% of the reproductions returned results matching the original reports together with 55% of tests in different spans of years and 40% of tests in novel geographies. Some original findings were associated with multiple new tests. Reproducibility was the best predictor of generalizability—for the findings that proved directly reproducible, 84% emerged in other available time periods and 57% emerged in other geographies. Overall, only limited empirical evidence emerged for context sensitivity. In a forecasting survey, independent scientists were able to anticipate which effects would find support in tests in new samples. Publisher PDF
