5. Rapid self-healing and anion selectivity in metallosupramolecular gels assisted by fluorine–fluorine interactions

Abstract

Simple ML2 [M = Fe(ii), Co(ii), Ni(ii)] complexes obtained from a perfluoroalkylamide derivative of 4-aminophenyl-2,2',6,2'-terpyridine spontaneously, yet anion selectively, self-assemble into gels, which manifest an unprecedented rapid gel strength recovery, viz. self-healing, and thermal rearrangement in aqueous dimethyl sulfoxide. The key factor for gelation and rheological properties emerges from the fluorine-fluorine interactions between the perfluorinated chains, as the corresponding hydrocarbon derivative did not form metallogels. The perfluoro-terpyridine ligand alone formed single crystals, while its Fe(ii), Co(ii) or Ni(ii) complexes underwent rapid gelation leading to highly entangled fibrillar networks visualized by electron microscopy. The thermodynamic parameters of gelation based on variable temperature NMR 1H and 19F resonances showed that gelation was enthalpically favourable and entropically disfavourable. The step strain rheological experiments revealed that the gels undergo rapid self-healing and the morphological features, thermal stability and mechanical properties were found to depend on the nature of the metal ion.

6. Structural Insights into Low-Temperature Copolymerization of Thermodegradable Amino Acids Mediated by Pyroglutamic Acid

Abstract

The demand for biocompatible multifunctional systems for bioimaging is driving the interest in new-generation fluorescent peptide based on nonaromatic amino acids such as l-glutamic acid and l-alanine. In general, due to the high melting points characterizing the zwitterionic structures of amino acids, their thermal polymerization is performed at temperatures between 200 and 300 °C. However, in this range of temperatures, most of the amino acids tend to decompose rather than undergo polymerization. The present work shows how to obtain nonaromatic fluorescent peptides at temperatures as low as 160 °C by copolymerizing l-glutamic acid with other high-melting amino acids, such as l-alanine, l-valine, and l-leucine. The low-temperature conversion of l-glutamic acid into pyroglutamic acid and its copolymerization with another amino acid were fully characterized by infrared and NMR spectroscopies, MS spectrometry, and thermal analysis. The reaction is mediated by the in situtransformation of l-glutamic acid into pyroglutamic acid, which acts simultaneously as an inomer, initiator + monomer, as well as a dispersing agent, allowing copolymerization with another amino acid. The resulting peptides exhibit fluorescent emission in the visible range typical of PGA derivatives, but they also possess a different polar nature that is inherited by the side chain of the second amino acid.

Esta depositada la versión del artículo enviada a la revista, A series of blue and blue-green emitters based on neutral bis- and tris-cyclometalated Ir(III) complexes with 1-benzyl-4-(2,6-difluorophenyl)-1H-1,2,3-triazole (dfptrBn) as cyclometalating ligand is reported. The bis-cyclometalated complexes of the type [Ir(dfptrBn)(2)((LX)-X-boolean AND)] with different ancillary ligands, (LX)-X-boolean AND = picolinate (pic) (2) or 2-(5-(perfluorophenyl)-2H-1,2,4-triazol-3-yl)pyridine (pytrF(5)) (3), are described and their photophysical properties compared with the analogous complexes containing the archetypal 2-(2,4-difluorophenyl)-pyridinato (dfppy) as cyclometaled ligand ((CN)-N-boolean AND). Complex 2 exhibits a marked solvatochromic behavior, from 475 nm in toluene to 534 nm in formamide, due to the strong MLCT character of its emissive excited state. Complex 3 displays a true-blue emission, narrower in the visible part than FIrpic. In addition, the homoleptic complex [Ir(dfprBn)(3)] (4) and the heteroleptic compounds with mixed arylpyridine/aryltriazole ligands, [Ir(dfptrBn)(2)((CN)-N-boolean AND)] ((CN)-N-boolean AND = 2-phenylpyridinato (ppy) (5) or dfppy (6)), have been synthesized and fully characterized. The facial (fac) complex fac-4 is emissive at 77 K showing a deep-blue emission, but it is not luminescent in solution at room temperature similarly to their phenylpyrazole counterparts. However, the fac isomers, fac-5 and fac-6, are highly emissive in solution and thin films, reaching emission quantum yields of 76%, with emission colors in the blue to blue-green region. The photophysical properties for all complexes have been rationalized by means of quantum-chemical calculations. Abstract

Platinum-based chemotherapy is the first-line treatment for different cancer types, and in particular, for malignant pleural mesothelioma patients (a tumor histotype with urgent medical needs). Herein, a strategy is presented to stabilize, transport, and intracellularly release a platinum(IV) (Pt-IV) prodrug using a breakable nanocarrier. Its reduction, and therefore activation as an anticancer drug, is promoted by the presence of glutathione in neoplastic cells that also causes the destruction of the carrier. The nanocage presents a single internal cavity in which the hydrophobic complex (Pt(dach)Cl-2(OH)(2)), (dach = R,R-diaminocyclohexane) is encapsulated. The in vitro uptake and the internalization kinetics in cancer model cells are evaluated and, using flow cytometry analysis, the successful release and activation of the Pt-based drug inside cancer cells are demonstrated. The in vitro findings are confirmed by the in vivo experiments on a mice model obtained by xenografting MPM487, a patient-derived malignant pleural mesothelioma. MPM487 confirms the well-known resistance of malignant pleural mesothelioma to cisplatin treatment while an interesting 50% reduction of tumor growth is observed when mice are treated with the Pt-IV, entrapped in the nanocages, at an equivalent dose of the platinum complex.

The self‐assembly of platinum complexes is a well‐documented process that leads to interesting changes of the photophysical and electrochemical behavior as well as to a change in reactivity of the complexes. However, it is still not clear how many metal units must interact in order to achieve the desired properties of a large assembly. This work aimed to clarify the role of the number of interacting PtII units leading to an enhancement of the spectroscopic properties and how to address inter‐ versus intramolecular processes. Therefore, a series of neutral multinuclear PtII complexes were synthesized and characterized, and their photophysical properties at different concentration were studied. Going from the monomer to dimers, the growth of a new emission band and the enhancement of the emission properties were observed. Upon increasing the platinum units up to three, the monomeric blue emission could not be detected anymore and a concentration independent bright‐yellow/orange emission, due to the establishment of intramolecular metallophilic interactions, was observed.

The synthesis and characterization of neutral heteroleptic molecular iridium(III) complexes with (2,4-difluoro)phenylpyridine and different pyridine-1,2,4-triazole ligands are described. The studies have shown high external quantum efficiencies and color points better than the standard for blue iridium(III) bis[(4,6-difluorophenyl)pyridinato-N,[C.sup.2]]picolinate.
