Abstract

Bacterial contamination is a hazard in many industries, including food, pharmaceuticals, and healthcare. The availability of a rapid and simple method for detecting this type of contamination in sterile areas enables immediate intervention to avoid or reduce detrimental effects. Among these methods, colorimetric indicators are becoming increasingly popular due to their affordability, ease of use, and quick visual interpretation of the signal. In this article, a bacterial contamination indicator system was designed by incorporating MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) into an electrospun PADAS matrix, which is a biodegradable poly(ester amide) synthesized from L-alanine, 1,12-dodecanediol, and sebacic acid. Uniaxial stress testing, thermogravimetric analysis and scanning electron microscopy were used to examine the mechanical properties, thermal stability, and morphology of the mats, respectively. The capacity for bacterial detection was not only analyzed with agar and broth assays but also by replicating important environmental conditions. Among the MTT concentrations tested in this study (0.2%, 2%, and 5%), it was found that only with a 2% MTT content the designed system produced a color response visible to the naked eye with optimal intensity, a sensitivity limit of 104 CFU/mL, and 86% cell viability, which showed the great potential for its use to detect bacterial contamination. In summary, by means of the process described in this work, it was possible to obtain a simple, low-cost and fast-response bacterial contamination indicator that can be used in mask filters, air filters, or protective clothing. 7. Screening and Assessment of Antimicrobial Susceptibility of Periodontopathic Bacteria in Peruvian Patients with Periodontitis: A Pilot Study

Abstract

Background. Severe periodontal disease is highly prevalent worldwide, affecting 20% of the population between the ages of 35 and 44 years. The etiological epidemiology in Peru is scarce, even though some studies describe a prevalence of 48.5% of periodontal disease in the general population. Periodontitis is one of the most prevalent oral diseases associated with site-specific changes in the oral microbiota and it has been associated with a socioeconomic state. This study aimed to determine the etiology and resistance profile of bacteria identified in a group of Peruvian patients with periodontal disease. Methods. Six subgingival plaque samples were collected from eight patients with severe periodontitis. Bacterial identification was carried out by an initial culture, PCR amplification, and subsequently DNA sequencing. We evaluated the antibiotic susceptibility by the disk diffusion method. Results. Variable diversity in oral microbiota was identified in each one of the eight patients. The bacterial genus most frequently found was Streptococcus spp. (15/48, 31.3%) followed by Rothia spp. (11/48, 22.9%), Actinomyces spp. (9/48, 18.8%), and Eikenella spp. (4/48, 8.3%). The most common species found was Rothia dentocariosa (8/48, 16.7%). The antimicrobial susceptibility assay varied according to the species tested; however, among all the isolates evaluated, Actinomyces naeslundii was resistant to penicillin and tetracycline; Eikenella corrodens was resistant to dicloxacillin; and Rothia dentocariosa was resistant to amoxicillin + clavulanic acid and metronidazole but also susceptible to trimethoprim-sulfamethoxazole. Conclusions. The most prevalent periodontal bacterium found in this study was Rothia dentocariosa. Specific antimicrobial therapy is required to improve the treatment outcomes of patients with periodontal disease and avoid antibiotic resistance.

Abstract

Introduction: Nowadays there is a high consumption of probiotics in food products due to its ability to fight pathogens. However, less is known of the effects of probiotics in oral microbial ecology. Aim: To determinate the antimicrobial effect of the probiotics Lactobacillus rhamnosus GG and Bifidobacterium bifidum against the cariogenic bacteria Streptococcus mutans and its antagonist Streptoccoccus sanguinis. Material and methods: For the present experimental study in vitro, strains of S. mutans (ATCC 27175), S. sanguinis (ATCC10556), LGG (ATCC 53103) and Bifidobacterium bifidum (ATCC 11863) were used. The two first bacteria were grown in Brain Heart Infusion broth (BHI) and the rest in Man Rogosa Sharpe (MRS). The samples were incubated under anaerobic conditions at 37 °C for 72 hours. For inhibition tests, four groups were prepared: S. mutans inhibition by LGG and B. bifidum (1); S. sanguinis inhibition by LGG and B. bifidum (2); LGG inhibition by S. mutans and S. sanguinis (3) and B. bifidum inhibition by S. mutans and S. sanguinis (4). The inhibitory effect was evaluated with the cup-plate diffusion method in which each well contained 100 µL of the bacteria selected as inhibitory agent. The wells were made on a BHI agar containing the microorganism to be inhibited. Chlorhexidine was the positive control of agar bacterial growth and periopathogenic bacteria the indicators of the antibacterial effect of probiotics., Introducción: actualmente, existe un alto consumo de probióticos debido a su capacidad de combatir microorganismos patógenos. Pese a ello, existe poca evidencia científica del efecto de éstos sobre la microbiota oral. Objetivo: evaluar la capacidad antimicrobiana del Lactobacillus rhamnosus GG (LGG) y del Bifidobacterium bifidum frente a la bacteria cariogénica Streptococcus mutans y su antagonista Streptoccoccus sanguinis. Materiales y Métodos: para este estudio experimental in vitro, se utilizaron cepas de S. mutans (ATCC 27175), S. sanguinis (ATCC10556), LGG (ATCC 53103) y Bifidobacterium bifidum (ATCC 11863). Las dos primeras cultivadas en caldo Brain Heart Infusion (BHI) y las dos últimas en Man Rogosa Sharpe (MRS), incubadas en condiciones de anaerobiosis, a 37ºC, durante 72 horas. Se prepararon 4 grupos de estudio: prueba de inhibición de S. mutans por LGG y B. bifidum (grupo 1); de S. sanguinis por LGG y B. bifidum (grupo 2); de LGG por S. mutans y S. sanguinis (grupo 3) y de B. bifidum por S. mutans y S. sanguinis (grupo 4). El efecto inhibitorio se evaluó con la prueba de difusión con pocillos, los cuales contenían el inóculo de la bacteria seleccionada como agente antibacteriano. El agar BHI contenía el cultivo del microorganismo que se buscaba inhibir. La clorhexidina fue el control positivo del crecimiento bacteriano en agar y las bacterias periopatógenas los cultivos indicadores del efecto antibacteriano de los probióticos. Resultados: el LGG y el B. bifidum demostraron no tener un efecto antibacteriano frente al S. mutans y S. sanguinis. Conclusiones: los probióticos LGG y Bifidobacterium bifidum no poseen un efecto antibacteriano contra Streptococcus mutans y Streptococcus sanguinis, siendo de relevancia clínica para la adecuada elección de microorganismos para las nuevas estrategias preventivas de caries en base a terapias de reemplazo., Tesis
