It is well known that nikethamide (N,N-diethylnicotinamide, CoramineR) is metabolized very rapidly to nicotinamide. Hence, there is difficulty in proving that nikethamide has been used as a doping substance because nicotinamide is a normal physiological metabolite in the organism as well as a vitamin preparation. However, an intermediate metabolite (N-ethylnicotinamide) was found by us in the urine of horses treated with CoramineR. This was characterized by gas chromatography/mass spectrometry, and synthesized and identified as being N-ethylnicotinamide. The excretion and metabolism of nikethamide after intramuscular injection in the horse was followed using quantitative gas chromatography of urine extracts over a period of several hours and the results of these experiments are reported. Changes in urinary pH had no significant effect upon either the metabolism or rate of excretion of the drug. [ABSTRACT FROM PUBLISHER]

A severe case is described of fluoride poisoning in cattle on different farms, caused by the emission of an enamel factory in their vicinity. The paper presents the results of the fluoride analysis during a seven months investigation period in urine and bones of several animals grazing in the surrounding polluted area and also of the forage consumed. In a seven years old cow, which died with severe symptoms of lameness, the fluoride content was measured of nearly all the bones. From each type of bone several samples of fat-free dry bone dust were collected and analysed for fluoride in order to obtain more information about the most respresentative sample area for the diagnosis of fluorosis. Taking into account a five percent deviation of the mean vague for all the bones analysed and using our sampling technique, the fluoride levels of ribs, thoracic vertebrae, lumbar vertebrae, sacral bones and pelvis seem to be the most representative. However, due to the unequal distribution of fluoride, between the compact and cancellous fraction, especially in long bones, the method of sampling seems to be extremely important. A case of fluoride pollution involving injury to cattle, caused by a brickworks in the Flemish countryside of Belgium, is described. The results of a one-year investigation, including a fluoride analysis of urine of all the animals grazing in the vicinity, are presented. Fluoride concentrations in the forage consumed and the different materials used and produced at the brickworks are also measured.
