1. Cech and de Rham Cohomology of Integral Forms

Abstract

We present a study on the integral forms and their Cech/de Rham cohomology. We analyze the problem from a general perspective of sheaf theory and we explore examples in superprojective manifolds. Integral forms are fundamental in the theory of integration in supermanifolds. One can define the integral forms introducing a new sheaf containing, among other objects, the new basic forms delta(dtheta) where the symbol delta has the usual formal properties of Dirac's delta distribution and acts on functions and forms as a Dirac measure. They satisfy in addition some new relations on the sheaf. It turns out that the enlarged sheaf of integral and "ordinary" superforms contains also forms of "negative degree" and, moreover, due to the additional relations introduced, its cohomology is, in a non trivial way, different from the usual superform cohomology., Comment: 20 pages, LaTeX, we expanded the introduction, we add a complete analysis of the cohomology and we derive a new duality between cohomology groups

Abstract

We first review the definition of superprojective spaces from the functor-of-points perspective. We derive the relation between superprojective spaces and supercosets in the framework of the theory of sheaves. As an application of the geometry of superprojective spaces, we extend Donaldson's definition of balanced manifolds to supermanifolds and we derive the new conditions of a balanced supermanifold. We apply the construction to superpoints viewed as submanifolds of superprojective spaces. Abstract

We study de Rham cohomology for various differential calculi on finite groups G up to order 8. These include the permutation group S_3, the dihedral group D_4 and the quaternion group Q. Poincare' duality holds in every case, and under some assumptions (essentially the existence of a top form) we find that it must hold in general. A short review of the bicovariant (noncommutative) differential calculus on finite G is given for selfconsistency. Exterior derivative, exterior product, metric, Hodge dual, connections, torsion, curvature, and biinvariant integration can be defined algebraically. A projector decomposition of the braiding operator is found, and used in constructing the projector on the space of 2-forms. Abstract

OBJECTIVES: Thrombocytopenia is a hallmark for patients with cirrhosis and it is perceived as a risk factor for bleeding events. However, the relationship between platelet count and bleeding is still unclear. METHODS: We investigated the relationship between platelet count and major or clinical relevant nonmajor bleedings during a follow-up of ∼4 years. RESULTS: A total of 280 cirrhotic patients with different degrees of liver disease (67% males; age 64±37 years; 47% Child–Pugh B and C) were followed up for a median of 1,129 (interquartile range: 800–1,498) days yielding 953.12 patient-year of observation. The annual rate of any significant bleeding was 5.45%/year (3.57%/year and 1.89%/year for major and minor bleeding, respectively). Fifty-two (18.6%) patients experienced a major (n=34) or minor (n=18) bleeding event, predominantly from gastrointestinal origin. Platelet counts progressively decreased with the worsening of liver disease and were similar in patients with or without major or minor bleeding: a platelet count ≤50×103/μl was detected in 3 (6%) patients with and in 20 (9%) patients without any bleeding event. Conversely, prothrombin time-international normalized ratio was slightly higher in patients with overall or major bleeding. On Cox proportional hazard analysis, only a previous gastrointestinal bleeding (hazard ratio (HR): 1.96; 95% confidence interval: 1.11–3.47; P=0.020) and encephalopathy (HR: 2.05; 95% confidence interval: 1.16–3.62; P=0.013) independently predicted overall bleeding events. CONCLUSIONS: Platelet count does not predict unprovoked major or minor bleeding in cirrhotic patients.

Il presente lavoro intende tracciare un’analisi storico-critica dell’epistemologia evoluzionistica, prendendo in esame le opere di Konrad Lorenz, Karl Raimund Popper e Donald T. Campbell, dopo averle inserite all’interno di una linea storica che risale al pensiero ed all’opera di Charles Darwin. L’epistemologia evoluzionistica, secondo la quale la conoscenza deve essere intesa primariamente come un prodotto dell’evoluzione biologica e considera quindi l’evoluzione biologica come la precondizione del comportamento cognitivo, culturale e sociale che un organismo, un gruppo o una specie può esprimere, ha le sue origini appunto già nei lavori di Charles Darwin. Il quadro esplicativo proposto da Darwin portava a considerare la vita, in modo totalmente “laico”, non più come il culmine di un processo attuato da una mente sovrannaturale, ma come l’esito conclusivo, ma contingente, di uno sviluppo evolutivo segnato da eventi inaspettati. Darwin poneva le premesse per una naturalizzazione della mente, ricollocandola nel contesto di una natura in evoluzione, senza alcuna finalità. Darwin rigettò ogni tentativo di rinunciare al naturalismo metodologico e di escludere a-priori l’applicazione del principio di selezione naturale alla mente umana e non arrivò mai a sostenere una posizione discontinuista. Dopo Darwin fu un filosofo, Herbert Spencer, che abbracciò entusiasticamente la teoria dell’evoluzione arrivando a considerare tutto come una manifestazione di una spinta evolutiva tesa verso il progresso. Fu però soprattutto negli Stati Uniti che la rivoluzione darwiniana impattò sulla filosofia. E’ noto che fin dalla sua apparizione L’origine delle specie venne accolta con grande interesse. E’ importante sottolineare che l’Epistemologia Evoluzionistica è strettamente connessa da una parte alla sintesi moderna, nel campo degli studi di biologia evoluzionistica e dall’altra all’ epistemologia naturalizzata di Willard van Orman Quine. Abstract

Chimeras and Consciousness is a collection of essays coming mainly from an international conference held in 2004 in Bellagio, Italy, hosted by the Rockefeller Foundation. In the section ‘What’s in Chimeras and Conscious- ness’ we summarize the content of the 23 essays appearing in the book organized in five parts: ‘Selves’, ‘Groups’, ‘Earth’, ‘Chimeras’, and ‘Consciousness’. We also make a few comments on such a structure, prob- ing both the boundaries and internal coherence of these topics. The main goal of the book seems a kind of innovative unification of entities and processes like consciousness, micro-organisms, evolution, and the chemico-physical processes involving the Earth as a whole. The editors and authors want to break up with a tradition that, in their view, keeps all these topics separated and marginalize some of them, thus hindering understanding. Then we assess this unifiying endeavor, analyzing it in terms of an all-encompassing glance, of the discovery of sameness, and/or of an explanatory history or theory. The conclusion summarizes and makes an over- all evaluation of the book as more an inspirational than a scientific work, related to some ancient philosophical themes.

Abstract

We study de Rham cohomology for various differential calculi on finite groups G up to order 8. These include the permutation group S3, the dihedral group D4 and the quaternion group Q. Poincaré duality holds in every case, and under some assumptions (essentially the existence of a top form) we find that it must hold in general. A short review of the bicovariant (noncommutative) differential calculus on finite G is given for selfconsistency. Exterior derivative, exterior product, metric, Hodge dual, connections, torsion, curvature, and biinvariant integration can be defined algebraically. A projector decomposition of the braiding operator is found, and used in constructing the projector on the space of two-forms. 30. Ca2+ inhibition of beta-adrenergic receptor- and forskolin-stimulated cAMP accumulation in C6-2B rat glioma cells is independent of protein kinase C.

Abstract

In C6-2B rat glioma cells, agonist-stimulated cAMP accumulation is potently inhibited after the stimulation of endogenous bradykinin receptors or stably transfected substance K receptors, coupled to phosphatidylinositol hydrolysis. In the present report, pharmacological tools were used to selectively stimulate either protein kinase C or Ca2+, the two final effectors activated upon phosphatidylinositol hydrolysis, and their role in the inhibition of the C6-2B cell cAMP signaling pathway was investigated. Activation of protein kinase C by an acute treatment with phorbol 12-myristate 13-acetate or L-alpha-1-oleoyl-2-acetyl-sn-3-glycerol did not reduce, but rather enhanced, the cAMP accumulation elicited by forskolin, a direct activator of adenylyl cyclase [ATP pyrophosphate-lyase (cyclizing), EC 4.6.1.1]. This effect was antagonized by the protein kinase inhibitor H-7 and mimicked by the protein phosphatase inhibitor okadaic acid. Thapsigargin, a selective microsomal Ca(2+)-ATPase inhibitor, evoked a sustained increase in the intracellular free Ca2+ concentration, with an EC50 of 24.8 +/- 4.3 nM, and inhibited the cAMP accumulation induced by the beta-adrenergic receptor agonist isoproterenol with comparable potency (IC50 = 19.3 +/- 0.2 nM), strongly suggesting a causal relationship between the two phenomena. The inhibition by thapsigargin of isoproterenol- or forskolin-stimulated cAMP accumulation was not affected by pertussis toxin or down-regulation or inhibition of protein kinase C. Dantrolene, a blocker of Ca2+ release from intracellular stores, antagonized 1) the Ca2+ transient in response to thapsigargin and substance K and 2) the inhibitory effect of these compounds on isoproterenol- or forskolin-induced cAMP accumulation. Moreover, sequestration of intracellular Ca2+ with the cell-permeable Ca2+ chelator ethylene glycol bis(beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid acetoxymethyl ester abolished the cAMP inhibition mediated by thapsigargin. Finally, isoproterenol- or forskolin-stimulated adenylyl cyclase activity in digitonin-permeabilized cells was not affected by either thapsigargin or substance K. These data provide compelling evidence that increases in intracellular free Ca2+ concentration without activation of protein kinase C suffice and are responsible for the inhibition of cAMP accumulation in C6-2B cells.

31. Chromosomal localization of the human diazepam binding inhibitor gene.

Abstract

We have used in situ chromosome hybridization and human-mouse somatic cell hybrids to map the gene(s) for human diazepam binding inhibitor (DBI), an endogenous putative modulator of the gamma-aminobutyric acid receptor acting at the allosteric regulatory center of this receptor that includes the benzodiazepine recognition site. In 784 chromosome spreads hybridized with human DBI cDNA, the distribution of 1476 labeled sites revealed a significant clustering of autoradiographic grains (11.3% of total label) on the long arm of chromosome 2 (2q). Furthermore, 63.5% of the grains found on 2q were located on 2q12-21, suggesting regional mapping of DBI gene(s) to this segment. Secondary hybridization signals were frequently observed on other chromosomes and they were statistically significant mainly for chromosomes 5, 6, 11, and 14. In addition, DNA from 32 human-mouse cell hybrids was digested with BamHI and probed with human DBI cDNA. A 3.5-kilobase band, which probably represents the human DBI gene, was assigned to chromosome 2. Four higher molecular weight bands, also detected in BamHI digests, could not be unequivocally assigned. A chromosome 2 location was excluded for the 27-, 13-, and 10-kilobase bands. These results assign a human DBI gene to chromosome 2 (2q12-21) and indicate that three of the four homologous sequences detected by the human DBI probe are located on three other chromosomes.

32. Diphtheria toxin prevents catecholamine desensitization of A431 human epidermoid carcinoma cells.

Abstract

We proposed that a rapidly turning over protein, induced in response to catecholamine stimulation of C6-2B rat astrocytoma cells, inhibits subsequent hormonal activation of adenylate cyclase. Studies upon which our hypothesis is based and confirmatory work in a variety of other cell lines and in vivo have utilized actinomycin D and cycloheximide to inhibit RNA and protein synthesis, respectively. These inhibitors, however, are not specific and have been reported also to interfere with other cellular processes. Diphtheria toxin is a specific protein synthesis inhibitor that acts only by ADP-ribosylating elongation factor 2, thus preventing peptide chain elongation. We thus tested whether diphtheria toxin could prevent catecholamine-induced desensitization in A431 human epidermoid carcinoma cells. The toxin inhibited protein synthesis and altered the time course of isoproterenol-stimulated cAMP accumulation as did the less-specific protein synthesis inhibitor cycloheximide. Cellular cAMP content after a 30-min exposure to isoproterenol was similar in control and in toxin-treated cells. However, after 4 hr of treatment with isoproterenol, toxin-treated cells accumulated up to six times more cAMP than controls. When cells or cell-free adenylate cyclase preparations were rechallenged with agonists, toxin-mediated inhibition of protein synthesis prevented desensitization. These results show that diphtheria toxin, a specific inhibitor of protein synthesis, can interfere with the normal physiological regulation of cAMP metabolism in eukaryotic cells and provide compelling evidence that catecholamine stimulation of adenylate cyclase promotes the synthesis of a protein(s) that, in some way, inhibits hormone-stimulated adenylate cyclase.

33. P2U-purinergic receptors on C6-2B rat glioma cells: modulation of cytosolic Ca2+ and cAMP levels by protein kinase C.

Abstract

The activation of P2-purinergic receptors on C6-2B rat glioma cells caused a transient increase in cytosolic-free Ca2+ concentration ([Ca2+]i) as detected by Fura 2 fluorescence ratio imaging of single cells. These purinergic receptors are of the P2U subtype because UTP and ATP were equipotent and substantially more potent than the P2X- and P2Y-selective agonists alpha,beta-methylene ATP and 2-methylthio ATP, respectively. There was homologous desensitization of the Ca2+ responses between UTP and ATP but no heterologous desensitization between these nucleotides and another Ca(2+)-mobilizing receptor agonist, alpha-thrombin. The UTP-induced peak [Ca2+]i rise was insensitive to chelation of extracellular Ca2+ with EGTA. However, the response was abolished after either depletion of intracellular Ca2+ stores with the microsomal Ca(2+)-ATPase inhibitor thapsigargin or blockade of Ca2+ release from intracellular stores with the muscle relaxant dantrolene. The activation of P2U-purinergic receptors and thrombin receptors increased the formation of total inositol phosphates (IPs) and inhibited cAMP accumulation elicited with either the beta-adrenergic receptor agonist (-)-isoproterenol, or forskolin, a direct activator of adenylyl cyclase. UTP- and alpha-thrombin-induced changes in the levels of IPs, cytosolic Ca2+, and agonist-elicited cAMP accumulation were dramatically inhibited (> 80%) by acute treatment of the cells with the protein kinase C activator 4 beta-phorbol 12-myristate 13-acetate but not with the inactive ester 4 alpha-phorbol 12,13-didecanoate. We conclude that in C6-2B cells, the increase in [Ca2+]i after activation of P2U-purinergic receptors is primarily a result of IPs-mediated release of Ca2+ from intracellular stores with secondary influx of Ca2+ by capacitative mechanisms. Also, the inhibition by UTP and alpha-thrombin of agonist-elicited cAMP accumulation is mediated through an increase in [Ca2+]i.

Konrad Lorenz ritiene che il progressivo processo di un sempre miglior adattamento degli organismi all’ambiente sia leggibile come una crescita di informazione sull’ambiente, che viene definito, in modo analogico, come “sapere”. ambpell non fu né un filosofo né uno storico della filosofia quanto a status accademico, ma uno psicologo. Questo fatto non è casuale: l’epistemologia evoluzionistica occupa uno spazio “extraterritoriale” all’interno della geografia accademica attuale ed è anche per questo motivo che non ha mai assunto un’identità ben definita, trovandosi ad occupare un territorio di confine fra la filosofia, la biologia evoluzionistica, la psicologia e la sociologia. Si tratta di un aspetto essenziale che va rimarcato in modo nettissimo, perché ha rappresentato una notevole difficoltà nella ricostruzione che è stata fatta del dibattito, rendendo quanto mai arduo definire i contorni e i limiti di questo ambito di ricerca. D.T. Campell adottò una prospettiva totalmente selezionista, applicandola a partire dal mondo del vivente su su fino alla scienza.

Lynn Margulis is the first of the three editors. She hardly needs an introduction: in the late 1960s, she pioneered and forcefully pushed the symbiogenetic theory of the eukaryotic cell (Sagan 1967; Margulis 1970), revolutionary at that time, but today confirmed by an overwhelming mass of researches and data, renewed and deepened at the molecular level in the genomic era (Margulis & Sagan 2003). Eukaryotes have originated by repeated symbiogenesis, the most famous of which is the incorporation of former free-living bacteria (Lane & Martin 2010). Details of the sequence and mechanisms of the Serial Endosymbiosis Theory (SET) are still today an exciting field of research (for example, Sassera et al. 2011). Margulis is unanimously seen as the scientist who opened this new field of research. She is also known for her tendency to radicalize and generalize her views against the established ones, being a forerunner not only for some specific theories but also for a way of looking to the living world based on symbiosis and association (cf. Abstract

Bexarotene is a specific retinoid X receptor agonist that has been used for the treatment of cutaneous T-cell lymphoma (CTCL). Because bexarotene causes hypothyroidism, it requires the administration of levothyroxine. However, levothyroxine, in addition to its ubiquitous nuclear receptors, can activate the αVβ3 integrin that is overexpressed in CTCL, potentially interfering the antineoplastic effect of bexarotene. We thus investigated the biological effect of levothyroxine in relation to bexarotene treatment. Although in isolated CTCL cells levothyroxine decreased, in an αVβ3-dependent manner, the antineoplastic effect of bexarotene, levothyroxine supplementation in preclinical models was necessary to avoid suppression of lymphoma immunity. Accordingly, selective genetic and pharmacologic inhibition of integrin αVβ3 improved the antineoplastic effect of bexarotene plus levothyroxine replacement while maintaining lymphoma immunity. Abstract

Background: Pregnancy and breastfeeding are associated with bone density loss. Fracture occurrence during pregnancy and post-partum, and its determinants, remain poorly known in Osteogenesis Imperfecta (OI). The aim of this study was to characterize fractures that occurred during pregnancy and post-partum in OI patients., Results: We conducted a retrospective multicentric study including a total of 50 previously pregnant OI women from 10 Bone Centers in France. Among these patients, 12 (24%) patients experienced fractures during pregnancy or in the 6 months following delivery, and 38 (76%) did not experience any fracture. The most frequent localizations were: proximal femur (25%), spine (25%), distal femur (12.5%), and pelvis (12.5%). Fractures during pregnancy occurred during the third trimester and post-partum fractures occurred with a mean delay of 2 months following delivery. No fractures occurred during childbirth. We next compared the 12 patients with pregnancy or post-partum fractures with the 38 patients without fractures. Mean age at pregnancy was 32.7 ± 3.1 years-old in the fractured group, vs 29.3 ± 5.0 years-old in the non-fractured group (p = 0.002). Breastfeeding was reported in 85.7% of patients in the fractured group, vs 47.1% in the non-fractured group (p = 0.03). All patients with post-partum fractures were breastfeeding. Bone mineral density was significantly lower in patients with pregnancy-related fractures compared with other patients: spine Z-score - 2.9 ± 1.6DS vs - 1.5 ± 1.7DS (p = 0.03), and total hip Z-score - 2.0 ± 0.7DS vs - 0.5 ± 1.4DS (p = 0.04). At least one osteoporosis-inducing risk factor or disease other than OI was identified in 81.8% vs 58.6% of fractured vs non-fractured patients (not significant). Fracture during pregnancy or post-partum was not associated with the severity of OI. Bisphosphonates before pregnancy were reported in 16.7% and 21.1% of patients with pregnancy-related fractures and non-fractured patients, respectively (not significant)., Conclusions: OI management during pregnancy and post-partum should aim for optimal control of modifiable osteoporosis risk factors, particularly in patients with low BMD. Abstract

This work provides information on the features of low molecular weight hyaluronic acid (HA)-decorated liposomes to target resveratrol (RSV) in the skin. Deformable liposomes were made of soy-phosphatidylcholine with Tween 80 as the fluidizing agent. For HA conjugation, three different phosphoethanolamines were tested: 1,2-dipalmitoyl- sn -glycero-3-phosphoethanolamine (DPPE), 1,2-dimyristoyl- sn -glycero-3-phosphoethanolamine (DMPE), and 1,2-dioleoyl- sn -glycero-3-phosphoethanolamine (DOPE). The different phosphoethanolamine-HA conjugates were inserted into the liposome bilayer by hydration (HA on both faces of the bilayer) or by the postinsertion method (HA only on the external face of the bilayer). The effect of these variables on deformability was experimentally assessed by an in-house method ( K value, the lower the value, the higher the deformability) and molecular dynamics (MD) simulations. The results showed that the K values of HA-liposomes obtained by hydration were higher than the K values of HA-liposomes prepared by postinsertion, and both were at least 10-fold higher than the K values of the corresponding plain liposomes. The nature of the lipid anchor played a key role in deformability (DMPE > DOPE > DPPE) with high variability in the case of DOPE formulations. These data were justified by the trends found in silico for the bilayer bending modulus and the HA end-to-end distance. In addition to liposome flexibility, the HA extent seems to be the key factor governing the skin penetration of RSV. When the extent is higher, the amount of the drug retained in the skin is larger. Regarding skin permeation, a parabolic trend was recorded, and the optimal amount to favor skin permeation was an approximately 30 HA/phospholipid (μg/mmol) ratio. This study reports the first piece of evidence that it is possible to control drug delivery in the skin by tuning the amount of HA on the vesicle surface.

Background: There is little evidence in the literature to suggest that safety training is effective in reducing injuries at the workplace., Objective: This study aimed at assessing the impact of a safety training programme on injury rates during construction work on the Turin-Novara high-speed railway line (2002-2006)., Methods: We adopted a before-after study design. Since workers were enrolled and trained at different times, pre- and post-training periods were calculated individually for each worker, Results: At the end of the training programme, the incidence of occupational injuries had fallen by 16% in the case of basic training and 25% for specific training. In the construction workers group (63.5% of trained workers) the reductions were 21% for basic training and 27% for specific training. All variations were statistically significant., Conclusion: Implementation of the training programme described led to a reduction in injury rates.

Abstract

Background: The optimal prophylactic regimen to prevent cytomegalovirus (CMV) infection and disease in orthotopic liver-transplant patients remains to be established. We tested whether a combination of intravenous ganciclovir (GCV) followed by high dosages of oral acyclovir (ACV) for 4 months provided a higher degree of protection from CMV than oral ACV alone., Methods: One hundred sixty-seven liver-transplant recipients were randomized to receive 120 days of antiviral treatment starting at the time of transplantation consisting of either ACV 800 mg orally four times daily (n=84) or 14 days of GCV 5 mg/kg intravenously every 12 hr followed by oral ACV 800 mg four times daily (n=83). Prospective laboratory and clinical surveillance was performed to determine primary endpoints (onset of CMV infection and CMV disease) and secondary endpoints (rates of fungal and bacterial infection, allograft rejection, and survival after transplantation). One-year event rates are presented as cumulative percentages., Results: During the first year after transplantation, CMV infection developed in 57% of patients treated with ACV and in 37% of patients treated with GCV + ACV (P=0.001). CMV disease developed in 23% of patients treated with ACV and in 11% of patients treated with GCV + ACV (P=0.03). In seronegative recipients of allografts from CMV-seropositive donors (D+/R-), CMV disease developed in 58% of patients treated with ACV and in 25% of patients treated with GCV + ACV (P=0.04). In the D+/R- group, 54% of patients treated with ACV and 17% of patients treated with GCV + ACV developed infection with Candida albicans (P=0.05)., Conclusions: Prophylaxis of CMV infection in liver-transplant patients with 14 days of intravenous GCV followed by high-dosage oral ACV is more effective than high-dosage oral ACV alone at reducing CMV infection and disease, even for patients in the D+/R- CMV serological group.

Abstract

We previously proposed that intracellular cyclic AMP accumulation induces a putative, rapidly turning over protein inhibitory to further hormone activation of adenylate cyclase. In the present study, 2-aminopurine, which has been reported to selectively block c-fos gene expression, was used to test the hypothesis that c-fos protein might be involved in the desensitization to catecholamines was observed in 2-aminopurine-treated C6-2B rat glioma cells. However, we found 2-aminopurine to inhibit, in a concentration-dependent manner, total cellular RNA and protein synthesis in C6-2B, HeLa, Swiss 3T3 and BALB/c cells. mRNA synthesis was also markedly reduced in 2-aminopurine-treated cells. These unexpected findings, while supporting our hypothesis of a protein synthesis-sensitive step in the development of refractoriness, raise concern about the specificity of action of 2-aminopurine to inhibit c-fos induction and thus any cellular process, including desensitization, which might be regulated by c-fos gene expression.
