1. Type IIA String Theory and tmf with Level Structure

Abstract: We look at a new string$^h$ tangential structure first introduced by Devalapurkar and relate it to the $W_7=0$ condition of Diaconescu-Moore-Witten for type IIA string theory and M-theory. We show that a string$^h$ structure on the target space automatically satisfies the $W_7=0$ condition and we also explain when the $W_7=0$ condition gives rise to a string$^h$ structure. Devalapurkar initially constructed $MString^h$ in such a way that it orients $tmf_1(3)$, we extend Devalapurkar's orientation and show that $MString^h$ orients $tmf_1(n)$. We compute the homotopy groups of $MString^h$ in the dimensions relevant for physical applications, and apply them to anomaly cancellation applications for compactifications of type IIA string theory., Comment: 46 pages

Abstract: We investigate the stability of weak symmetry-protected topological phases (SPTs) in the presence of short-range interactions, focusing on the tenfold way classification. Using Atiyah's Real $\mathit{KR}$-theory and Anderson-dualized bordism, we classify free and interacting weak phases across all Altland-Zirnbauer symmetry classes in low dimensions. Extending the free-to-interacting map of Freed-Hopkins, we mathematically compute how the behavior of free weak SPTs changes when interactions are introduced as well as predict intrinsically-interacting weak phases in certain classes. Our mathematical techniques involve T-duality and the James splitting of the torus. Our results provide a mathematical framework for understanding the persistence of weak SPTs under interactions, with potential implications for experimental and theoretical studies of these phases., Comment: 48 pages. 3. Impeding LLM-assisted Cheating in Introductory Programming Assignments via Adversarial Perturbation

Abstract: While Large language model (LLM)-based programming assistants such as CoPilot and ChatGPT can help improve the productivity of professional software developers, they can also facilitate cheating in introductory computer programming courses. Assuming instructors have limited control over the industrial-strength models, this paper investigates the baseline performance of 5 widely used LLMs on a collection of introductory programming problems, examines adversarial perturbations to degrade their performance, and describes the results of a user study aimed at understanding the efficacy of such perturbations in hindering actual code generation for introductory programming assignments. The user study suggests that i) perturbations combinedly reduced the average correctness score by 77%, ii) the drop in correctness caused by these perturbations was affected based on their detectability.

4. Simulability of non-classical continuous-variable quantum circuits

Abstract: In continuous-variable quantum computation, identifying key elements that enable a quantum computational advantage is a long-standing issue. Starting from the standard results on the necessity of Wigner negativity, we develop a comprehensive and versatile framework that not only enables the identification of a potential quantum computational advantage, but also allows to pinpoint the contribution of each quantum gate in achieving this objective. As such, it can be straightforwardly applied to current continuous-variables quantum circuits, while also constraining the tolerable amount of losses above which any potential quantum advantage can be ruled out. We use $(s)$-ordered quasiprobability distributions on phase-space to capture the non-classical features in the protocol, and focus our model entirely on the ordering parameter $s$. This allows us to highlight the resourcefulness and robustness to loss of a universal set of unitary gates comprising three distinct Gaussian gates, and a fourth one, the cubic gate, providing important insight on the role of non-Gaussianity., Comment: 16 pages, 5 figures

Abstract: In this article, we answer two questions of Buchanan-McKean (arXiv:2312.08209) about bordism for manifolds with spin$^h$ structures: we establish a Smith isomorphism between the reduced spin$^h$ bordism of $\mathbb{RP}^\infty$ and pin$^{h-}$ bordism, and we provide a geometric explanation for the isomorphism $\Omega_{4k}^{\mathrm{Spin}^c} \otimes\mathbb Z[1/2] \cong \Omega_{4k}^{\mathrm{Spin}^h} \otimes\mathbb Z[1/2]$. 6. Alfv\'en waves at low Magnetic Reynolds number

Abstract: This paper seeks whether Alfv\'en waves (AW) can be produced in laboratory-scale liquid metal experiments, \emph{i.e.} at low-magnetic Reynolds Number ($R\!m$). AW are incompressible waves propagating along magnetic fields typically found geo and astrophysical systems. Until now, only faint linear waves have been experimentally produced in liquid metals because of the large magnetic dissipation they undergo when $R\!m\ll1$. Yet, controlling laboratory AW could emulate such far remote processes as anomalous heating in the solar corona, oscillations of the Earth inner core or turbulence in the solar wind. To answer this question, we force AW with an AC electric current in a liquid metal channel in a transverse magnetic field. We derive a wave-bearing extension of the usual low$-R\!m$ MHD approximation to identify two linear regimes: The purely diffusive regime exists when $N_\omega$, the ratio of the oscillation period to the timescale of diffusive two-dimensionalisation by the Lorentz force, is small. The propagative regime is governed by the ratio of the forcing period to the AW propagation timescale which, we call the Jameson number $J\!a$ after Jameson (1964), JFM. In this regime, AW are dissipative and dispersive as they propagate more slowly where velocity gradients are higher. Both regimes are recovered in the FLOWCUBE experiment, in excellent agreement with the model up to $J\!a \lesssim 0.85$ but near the $J\!a=1$ resonance, high amplitude waves become clearly nonlinear. Hence, in electrically driving AW, we were able to produce some of the propagative, diffusive and nonlinear processes of astro and geophysical AW., Comment: 36 pages, 18 figures

7. The Smith Fiber Sequence of Invertible Field Theories

Abstract: Smith homomorphisms are maps between bordism groups that change both the dimension and the tangential structure. We give a completely general account of Smith homomorphisms, unifying the many examples in the literature. We provide three definitions of Smith homomorphisms, including as maps of Thom spectra, and show they are equivalent. Using this, we identify the cofiber of the spectrum-level Smith map and extend the Smith homomorphism to a long exact sequence of bordism groups, which is a powerful computation tool. We discuss several examples of this long exact sequence, relating them to known constructions such as Wood's and Wall's sequences. Furthermore, taking Anderson duals yields a long exact sequence of invertible field theories, which has a rich physical interpretation. 8. Aluminum nuclear demagnetization refrigerator for powerful continuous cooling

Abstract: Many laboratories routinely cool samples to 10 mK, but relatively few can cool condensed matter below 1 mK. Easy access to the microkelvin range would propel fields such as quantum sensors and quantum materials. Such temperatures are achieved with adiabatic nuclear demagnetization. Existing nuclear demagnetization refrigerators (NDR) are ``single-shot'', and the recycling time is incompatible with some sub-mK experiments. Furthermore, a high cooling power is required to overcome the excess heat load of order nW on NDR pre-cooled by cryogen-free dilution refrigerators. We report the performance of an aluminum NDR designed for powerful cooling when part of a dual stage continuous NDR (CNDR). Its thermal resistance is minimized to maximize the cycling rate of the CNDR and consequently its cooling power. At the same time, its susceptibility to eddy current heating is minimized. A CNDR based on two of the aluminum NDR presented here would achieve a cooling power of approximately 40 nW at 560 $\mu$K less than six days after cooling from room temperature, with a small offset in electronic temperature that decreases as the time-dependent heat load decays.

9. Exploring possible magnetic monopoles-induced magneto-electricity in spin ices

Abstract: The possibilities of combining several degrees of freedom inside a unique material have recently been highlighted in their dynamics and proposed as information carriers in quantum devices where their cross-manipulation by external parameters such as electric and magnetic fields could enhance their functionalities. An emblematic example is that of electromagnons, spin-waves dressed with electric dipoles, that are fingerprints of multiferroics. Point-like objects have also been identified, which may take the form of excited quasiparticles. This is the case for magnetic monopoles, the exotic excitations of spin ices, that have been recently proposed to carry an electric dipole although experimental evidences remain elusive. Presently, we investigate the electrical signature of a classical spin ice and a related compound that supports quantum fluctuations. Our in-depth study clearly attributes magnetoelectricity to the correlated spin ice phase distinguishing it from extrinsic and single-ion effects. Our calculations show that the proposed model conferring magnetoelectricity to monopoles is not sufficient, calling for higher order contributions.

10. Small Advances and Swift Retreat: Race-Conscious Educational Policy in the Obama and Trump Administrations

Abstract: The three terms comprising the Obama and Trump presidencies provide an opportunity to understand the evolution of race-conscious education policy in an increasingly multiracial, unequal, and divided society. Through document review and interviews with civil rights lawyers, government officials, congressional staffers, and intermediary organization personnel, we sought to understand how Obama officials envisioned and changed the role of the federal government in fostering K-12 race-conscious educational policies and what mechanisms they used to advance priorities. We also explored changes Trump administration officials made to federal civil rights policies and through which institutional means. Our findings reveal through-lines between past and present political agendas and the methods for enactment. Obama's interagency efforts to reinvigorate civil rights oversight and enforcement in education harkened back to the mid-1960s era of bipartisan cooperation around school desegregation. Yet the decades-long legal and policy retrenchment against civil rights advances made in the 1960s constrained further progress. Trump's administration advocated for the privatization of public education through increased choice and opposed race-consciousness in education law and policy. The reshaping of the federal judiciary under Trump presents challenges for race-consciousness in the law for years to come. Recognizing these consistent through-lines and constraints will be essential for advocates and policymakers going forward.

11. Re-Centering Civil Rights in the Reauthorization of ESEA: An Equitable, Ecological, Evidence-Based Framework

Abstract: This article considers the reauthorization of the Elementary and Secondary Education Act (ESEA) in light of current educational inequities and the impact of the pandemic. The reauthorization presents an opportunity to center equity and justice and revitalize the civil rights aspects of the law. The authors review recent studies about the myriad ways the COVID-19 pandemic affected students, teachers and school systems and situates the reauthorization within the broader political context, including federal aid to states and districts during the pandemic. The authors present overarching policy recommendations: (1) a restructuring of the law's titles to focus on students, teachers, and systems (i.e. schools and districts); (2) a focus on the principles of racial equity, an ecosystem approach to serve students' needs across policy silos (i.e., housing and health), and a focus on research evidence; (3) policy targets for students; (4) policy targets for educators, including professional renewal and retention; (5) policy targets for the education system as a whole, including a focus on fiscal equity and facilitating regional approaches to cross-sector collaborations. The article presents a coherent conceptual framework for a redesign of ESEA with an emphasis on equity, evidence use, and ecosystems that is argued would directly address the needs of students, teachers, and systems.

15. Bosonization and Anomaly Indicators of (2+1)-D Fermionic Topological Orders

Abstract: We provide a mathematical proposal for the anomaly indicators of symmetries of (2+1)-d fermionic topological orders, and work out the consequences of our proposal in several nontrivial examples. Our proposal is an invariant of a super modular tensor category with a fermionic group action, which gives a (3+1)-d topological field theory (TFT) that we conjecture to be invertible; the anomaly indicators are partition functions of this TFT on $4$-manifolds generating the corresponding twisted spin bordism group. Our construction relies on a bosonization construction due to Gaiotto-Kapustin and Tata-Kobayashi-Bulmash-Barkeshli, together with a ``bosonization conjecture'' which we explain in detail. In the second half of the paper, we discuss several examples of our invariants relevant to condensed-matter physics. The most important example we consider is $\mathbb{Z}/4^T\times \mathbb{Z}/2^f$ time-reversal symmetry with symmetry algebra $\mathcal T^2 = (-1)^FC$, which many fermionic topological orders enjoy, including the $\mathrm{U}(1)_5$ spin Chern-Simons theory. Using newly developed tools involving the Smith long exact sequence, we calculate the cobordism group that classifies its anomaly, present the generating manifold, and calculate the partition function on the generating manifold which serves as our anomaly indicator. Our approach allows us to reproduce anomaly indicators known in the literature with simpler proofs, including $\mathbb{Z}/4^{Tf}$ time-reversal symmetry with symmetry algebra $\mathcal T^2 = (-1)^F$, and other symmetry groups in the 10-fold way involving Lie group symmetries., Comment: 82 pages, comments welcome

17. Global anomalies & bordism of non-supersymmetric strings

Abstract: The three tachyon-free non-supersymmetric string theories in ten dimensions provide a handle on quantum gravity away from the supersymmetric lamppost. However, they have not been shown to be fully consistent; although local anomalies cancel due to versions of the Green-Schwarz mechanism, there could be global anomalies, not cancelled by the Green-Schwarz mechanism, that could become fatal pathologies. We compute the twisted string bordism groups that control these anomalies via the Adams spectral sequence, showing that they vanish completely in two out of three cases (Sugimoto and $SO(16)^2$) and showing a partial vanishing also in the third (Sagnotti 0'B model). We also compute lower-dimensional bordism groups of the non-supersymmetric string theories, which are of interest to the classification of branes in these theories via the Cobordism Conjecture. We propose a worldvolume content based on anomaly inflow for the $SO(16)^2$ NS5-brane, and discuss subtleties related to the torsion part of the Bianchi identity. Abstract: Background: Cardiac rehabilitation programs provide a valuable opportunity to promote the adoption of healthy lifestyle behaviors in patients with atherosclerotic cardiovascular diseases (ASCVDs) and metabolic comorbidities, including metabolic syndrome and prediabetes. However, strategies to reverse these conditions remain to be explored. The DIABEPIC-1 study aimed to assess the feasibility of an enhanced 6-month cardiac rehabilitation program for patients with ASCVD while investigating prediabetes and metabolic syndrome remission. Methods: The study combined exercise training with a comprehensive nutritional intervention, emphasizing reduction in intake of ultraprocessed foods, adoption of a Mediterranean diet, and implementation of time-restricted eating. Baseline, 3-month, and 6-month assessments included segmental body-composition measurements, blood analysis, maximal exercise testing, nutritional diaries recorded with the Keenoa AI app, and lifestyle questionnaires. Remission criteria included a return to an HbA1c level of < 5.7%, and < 3 National Cholesterol Education Program Adult Treatment Panel III (NCEP/ATP-III) criteria for prediabetes and metabolic syndrome, respectively. Results: A total of 36 participants were recruited. The study demonstrated completion rates of 94.4% at 3 months, and 88.9% at 6 months, and a mean compliance rate of 92.5% for planned clinical appointments. Significant reductions in waist circumference (–9.2 cm, P < 0.001) and weight (–8.0 kg, P < 0.001) were observed. Improvement in glycemic and lipid profiles, insulin-resistance marker levels, and liver health were noted. Participants enhanced their cardiorespiratory fitness, reduced their consumption of ultraprocessed food, and increased their adherence to the Mediterranean diet and time-restricted eating. Notably, 50% achieved prediabetes remission, and 70% with metabolic syndrome at baseline achieved remission. Conclusions: The study demonstrates the possibility of enhancing cardiac rehabilitation with an intensive nutritional intervention, yielding clinically significant outcomes, including remission of key risk factors in a substantial number of ASCVD patients. Les évaluations de base, à 3 mois et à 6 mois, comprenaient des mesures de la composition corporelle par segments, des analyses sanguines, des tests d'exercice maximal, la tenue de journaux nutritionnels enregistrés avec l'application Keenoa AI et des questionnaires sur le mode de vie. Les critères de rémission comprenaient un retour à un niveau d'hémoglobine glyquée HbA1c < 5,7 %, et < 3 critères de la table du « National Cholesterol Education Program Adult Treatment Panel III » (NCEP/ATP-III) pour le prédiabète et le syndrome métabolique, respectivement. Résultats: Au total, 36 participants ont été recrutés. L'étude a montré des taux d'achèvement de 94,4 % à 3 mois et de 88,9 % à 6 mois, ainsi qu'un taux de conformité moyen de 92,5 % pour les rendez-vous cliniques planifiés. Des réductions significatives du tour de taille (-9,2 cm, p < 0,001) et du poids (-8,0 kg, p < 0,001) ont été observées. Une amélioration des profils glycémiques et lipidiques, des niveaux de marqueurs de résistance à l'insuline et de la santé hépatique a été constatée. Les participants ont amélioré leur condition cardiorespiratoire, réduit leur consommation d'aliments ultra-transformés et augmenté leur adhésion au régime méditerranéen et à l'alimentation restreinte dans le temps. Notamment, 50 % des participants ont obtenu une rémission du prédiabète et 70 % des participants atteints du syndrome métabolique au début de l'étude ont atteint la rémission. Conclusions: L'étude démontre la possibilité d'améliorer la réadaptation cardiaque par une intervention nutritionnelle intensive, produisant des résultats cliniquement significatifs, y compris la rémission des principaux facteurs de risque chez un nombre substantiel de patients atteints de MCVA. 23. A Civil Rights Framework for the Reauthorization of ESEA

Abstract: The last several years have been trying for students, communities, and schools. As states and local educational agencies work to meet the significant health and educational needs of students, and address the vast racial and socioeconomic inequities that have been heightened by the pandemic, the federal role in education is critical. Because the Elementary and Secondary Education Act (ESEA) remains the government's most consequential mechanism to promote equity in public education, the upcoming reauthorization of the law's latest version, the Every Student Succeeds Act (ESSA), presents an opportunity to re-envision this federal role in promoting access to quality educational opportunities. In this brief, the authors describe an equitable, evidence-based, and ecological civil rights framework to guide the redesign of ESSA. It places students, staff, school systems, and cross-sector collaboration at the center of ESEA and considers the complexity of racial, socioeconomic, and other inequities along with the strengths nested within communities.

Abstract: We study defects in symmetry breaking phases, such as domain walls, vortices, and hedgehogs. In particular, we focus on the localized gapless excitations which sometimes occur at the cores of these objects. These are topologically protected by an 't Hooft anomaly. We classify different symmetry breaking phases in terms of the anomalies of these defects, and relate them to the anomaly of the broken symmetry by an anomaly-matching formula. We also derive the obstruction to the existence of a symmetry breaking phase with a local defect. We obtain these results using a long exact sequence of groups of invertible field theories, which we call the "symmetry breaking long exact sequence" (SBLES). The mathematical backbone of the SBLES is studied in a companion paper. Our work further develops the theory of higher Berry phase and its bulk-boundary correspondence, and serves as a new computational tool for classifying symmetry protected topological phases., Comment: 31 pages, 4 figures. 25. Directed Test Program Generation for JIT Compiler Bug Localization

Abstract: Bug localization techniques for Just-in-Time (JIT) compilers are based on analyzing the execution behaviors of the target JIT compiler on a set of test programs generated for this purpose; characteristics of these test inputs can significantly impact the accuracy of bug localization. However, current approaches for automatic test program generation do not work well for bug localization in JIT compilers. This paper proposes a novel technique for automatic test program generation for JIT compiler bug localization that is based on two key insights: (1) the generated test programs should contain both passing inputs (which do not trigger the bug) and failing inputs (which trigger the bug); and (2) the passing inputs should be as similar as possible to the initial seed input, while the failing programs should be as different as possible from it. We use a structural analysis of the seed program to determine which parts of the code should be mutated for each of the passing and failing cases. Experiments using a prototype implementation indicate that test inputs generated using our approach result in significantly improved bug localization results than existing approaches.

Abstract: When $R$ is one of the spectra $\mathit{ku}$, $\mathit{ko}$, $\mathit{tmf}$, $\mathit{MTSpin}^c$, $\mathit{MTSpin}$, or $\mathit{MTString}$, there is a standard approach to computing twisted $R$-homology groups of a space $X$ with the Adams spectral sequence, by using a change-of-rings isomorphism to simplify the $E_2$-page. This approach requires the assumption that the twist comes from a vector bundle, i.e. the twist map $X\to B\mathrm{GL}_1(R)$ factors through $B\mathrm{O}$. We show this assumption is unnecessary by working with Baker-Lazarev's Adams spectral sequence of $R$-modules and computing its $E_2$-page for a large class of twists of these spectra. Abstract: Abstract The possibilities of combining several degrees of freedom inside a unique material have recently been highlighted in their dynamics and proposed as information carriers in quantum devices where their cross-manipulation by external parameters such as electric and magnetic fields could enhance their functionalities. An emblematic example is that of electromagnons, spin-waves dressed with electric dipoles, that are fingerprints of multiferroics. Point-like objects have also been identified, which may take the form of excited quasiparticles. This is the case for magnetic monopoles, the exotic excitations of spin ices, that have been recently proposed to carry an electric dipole, although experimental evidences remain elusive. Presently, we investigate the electrical signature of a classical spin ice and a related compound that supports quantum fluctuations. Our in-depth study clearly attributes magneto-electricity to the correlated spin ice phase distinguishing it from extrinsic and single-ion effects. Our calculations show that the proposed model conferring magneto-electricity to monopoles is not sufficient, calling for higher-order contributions.

29. Well-being amid (im)mobility struggles: Youth’s experiences in Casamance, Senegal

Abstract: Background Large numbers of young people worldwide, especially in the Global South, wish to migrate but lack the capacity to do so, with potentially detrimental consequences for their well-being and mental health. Termed ‘involuntary immobility’, this phenomenon is numerically larger than migration, but remains for now a largely underexplored area of research. Focusing on young Senegalese living in Casamance, this paper contributes to the limited literature on the implications of immobility for subjective well-being. It does so by (i) considering different degrees and types of involuntary immobility and their intersections, (ii) laying out the implications of (interacting) social and spatial immobilities for well-being with particular attention to youth’s agency in navigating a lack of capabilities, and (iii) by accounting for heterogenous experiences taking a gendered approach. Methods For this study, 35 semi-structured interviews were conducted with 18- to 39-year-olds in Ziguinchor region, recruited through purposive and snowball sampling. Thematic analysis was employed on verbatim transcriptions of audio-recorded interviews. Results Our study reveals a nuanced reality where aspirations to (temporarily) move abroad coexist with aspirations to stay in Casamance. Participants’ life aspirations and overarching projects clash, however, both with a lack of capabilities to move abroad and to enact these locally. This pervasive immobility decreases life satisfaction and generates negative emotions, such as stress, anxiety, discouragement, and distress. Despite these obstacles, our findings also underscore the agency and resourcefulness displayed by the youth as they navigate their limited control over life choices and paths. Conclusions Involuntary spatial immobility exacerbates the dominant experience of social immobility, magnifying its effect on youth’s well-being, revealing a previously unacknowledged phenomenon. Our findings further emphasize the pressing need for a more cohesive alignment between migration policies and information campaigns on one hand and the real experiences and challenges encountered by their intended audience on the other.

Abstract: In this work we investigate the Swampland Cobordism Conjecture in the context of type IIB string theory geometries with non-trivial duality bundle. Quite remarkably, we find that many non-trivial bordism classes with duality bundles in Mp$(2,\mathbb{Z})$, a double cover of SL$(2,\mathbb{Z})$ related to fermions, correspond to asymptotic boundaries of well-known supersymmetric F-theory backgrounds. These include $[p,q]$-7-branes, non-Higgsable clusters, S-folds, as well as various lower-dimensional generalizations. These string theoretic objects break the global symmetries associated to the non-trivial bordism groups, providing a strong test of the Cobordism Conjecture. Further including worldsheet orientation reversal promotes the duality group to the Pin$^+$ cover of GL$(2,\mathbb{Z})$. The corresponding bordism groups require a new non-supersymmetric "reflection 7-brane" and its compactifications to ensure the absence of global symmetries, thus providing an interesting prediction of the Cobordism Conjecture for non-supersymmetric type IIB backgrounds. A major component of the present work is the explicit derivation of the involved bordism groups as well as their generators, which correspond to asymptotic boundaries of explicit string theory backgrounds. The main tool is the Adams spectral sequence, to which we provide a detailed introduction. Abstract: Missing data is a common problem in medical research, and is commonly addressed using multiple imputation. Although traditional imputation methods allow for valid statistical inference when data are missing at random (MAR), their implementation is problematic when the presence of missingness depends on unobserved variables, i.e. the data are missing not at random (MNAR). Unfortunately, this MNAR situation is rather common, in observational studies, registries and other sources of real-world data. While several imputation methods have been proposed for addressing individual studies when data are MNAR, their application and validity in large datasets with multilevel structure remains unclear. We therefore explored the consequence of MNAR data in hierarchical data in-depth, and proposed a novel multilevel imputation method for common missing patterns in clustered datasets. This method is based on the principles of Heckman selection models and adopts a two-stage meta-analysis approach to impute binary and continuous variables that may be outcomes or predictors and that are systematically or sporadically missing. 35. Long-term functional course of Sjögren's disease-associated interstitial lung disease

Abstract: Background Interstitial lung disease (ILD) is common in primary Sjögren's disease (pSD); its functional course is poorly known. Our aim was to characterise the long-term functional course and prognosis in patients with pSD-ILD. We determined the role of baseline demographic and clinical variables in the evolution of lung function and identified risk factors for death or transplantation. Methods In a retrospective observational cohort study, patients with pSD and ILD were retrospectively identified from two French ILD centres. Forced vital capacity (FVC) and diffusing capacity of the lungs for carbon monoxide (DLCO) slopes were obtained from joint models. Latent class mixed models identified clusters of FVC and DLCO trajectories. Results We included 73 patients (63% women, mean age 63 years), with a median follow-up of 9.3 years. At baseline, mean FVC was 73±21% and DLCO 51±16%. On average, FVC was stable, while there was an annual decline in DLCO of 1% of the predicted value. Male sex, a pattern of usual interstitial pneumonia (UIP) or indeterminate for UIP on high-resolution computed tomography (HRCT), and features of fibrosis on HRCT, were associated with an accelerated decline in FVC and DLCO. Conclusion We identified clusters of lung function evolution. 1) Two FVC trajectories: patients with stable FVC (n=56, 78%); patients with FVC decline (n=16, 22%) of 2.4% per year, characterised by a low baseline DLCO (39%) and a higher risk of death or transplantation (HR 52, 95% CI 10–273). 2) Three DLCO trajectories: patients with stable DLCO (n=44, 66%); patients with a slow decline in DLCO (n=12, 18%) of 2.8% per year; patients with a rapid decline in DLCO (n=11, 16%) of 4.8% per year, characterised by a low baseline DLCO (41%) and a higher risk of death or transplantation (HR 156, 95% CI 18–1352).

Abstract: Abstract Observational data provide invaluable real-world information in medicine, but certain methodological considerations are required to derive causal estimates. In this systematic review, we evaluated the methodology and reporting quality of individual-level patient data meta-analyses (IPD-MAs) conducted with non-randomized exposures, published in 2009, 2014, and 2019 that sought to estimate a causal relationship in medicine. We screened over 16,000 titles and abstracts, reviewed 45 full-text articles out of the 167 deemed potentially eligible, and included 29 into the analysis. Unfortunately, we found that causal methodologies were rarely implemented, and reporting was generally poor across studies. Specifically, only three of the 29 articles used quasi-experimental methods, and no study used G-methods to adjust for time-varying confounding. To address these issues, we propose stronger collaborations between physicians and methodologists to ensure that causal methodologies are properly implemented in IPD-MAs. In addition, we put forward a suggested checklist of reporting guidelines for IPD-MAs that utilize causal methods. This checklist could improve reporting thereby potentially enhancing the quality and trustworthiness of IPD-MAs, which can be considered one of the most valuable sources of evidence for health policy.

44. Evaluating individualized treatment effect predictions: a model-based perspective on discrimination and calibration assessment

Abstract: In recent years, there has been a growing interest in the prediction of individualized treatment effects. While there is a rapidly growing literature on the development of such models, there is little literature on the evaluation of their performance. In this paper, we aim to facilitate the validation of prediction models for individualized treatment effects. The estimands of interest are defined as based on the potential outcomes framework, which facilitates a comparison of existing and novel measures. In particular, we examine existing measures of measures of discrimination for benefit (variations of the c-for-benefit), and propose model-based extensions to the treatment effect setting for discrimination and calibration metrics that have a strong basis in outcome risk prediction. The main focus is on randomized trial data with binary endpoints and on models that provide individualized treatment effect predictions and potential outcome predictions. We use simulated data to provide insight into the characteristics of the examined discrimination and calibration statistics under consideration, and further illustrate all methods in a trial of acute ischemic stroke treatment. The results show that the proposed model-based statistics had the best characteristics in terms of bias and accuracy. While resampling methods adjusted for the optimism of performance estimates in the development data, they had a high variance across replications that limited their accuracy. Therefore, individualized treatment effect models are best validated in independent data. To aid implementation, a software implementation of the proposed methods was made available in R.

Abstract: Alterations in dopaminergic transmission are associated with neurological disorders, such as depression, autism, and Parkinson's disease. Exposure of rats to ambient fine (FP) or ultrafine (UFP) particles induces oxidative and inflammatory responses in the striatum, a neuronal nucleus with dense dopaminergic innervation and critically involved in the control of motor activity.Objectives: We used an ex vivo system to evaluate the effect of in vivo inhalation exposure to FP and UFP on motor activity and dopaminergic transmission.Materials and Methods: Male adult Wistar rats were exposed to FP, UFP, or filtered air for 8 weeks (subchronic exposure; 5 h/day, 5 days/week) in a particle concentrator. Motor activity was evaluated using the open-field test. Uptake and release of [3H]-dopamine were assessed in striatal synaptosomes, and dopamine D2 receptor (D2R) affinity for dopamine was evaluated by the displacement of [3H]-spiperone binding to striatal membranes.Results: Exposure to FP or UFP significantly reduced spontaneous motor activity (ambulatory distance: FP -25%, UFP -32%; ambulatory time: FP -24%, UFP -22%; ambulatory episodes: FP -22%, UFP -30%), decreased [3H]-dopamine uptake (FP -18%, UFP -24%), and increased, although not significantly, [3H]-dopamine release (113.3 ± 16.3 and 138.6 ± 17.3%). Neither FP nor UFP exposure affected D2R density or affinity for dopamine.Conclusions: These results indicate that exposure to ambient particulate matter reduces locomotion in rats, which could be related to altered striatal dopaminergic transmission: UFP was more potent than FP. Our results contribute to the evidence linking environmental factors to changes in brain function that could turn into neurological and psychiatric disorders.HIGHLIGHTSYoung adult rats were exposed to fine (FP) or ultrafine (UFP) particles for 40 days.Exposure to FP or UFP reduced motor activity.Exposure to FP or UFP reduced dopamine uptake by striatal synaptosomes.Neither D2R density or affinity for dopamine was affected by FP or UFP.UFP was more potent than FP to exert the effects reported.

Abstract: The above-ground (phyllosphere) plant microbiome is increasingly recognized as an important component of plant health. We hypothesized that phyllosphere bacterial recruitment may be disrupted in a greenhouse setting, and that adding a bacterial amendment would therefore benefit the health and growth of host plants. Using a newly developed synthetic phyllosphere bacterial microbiome for tomato (Solanum lycopersicum), we tested this hypothesis across multiple trials by manipulating microbial inoculation of leaves and measuring subsequent plant growth and reproductive success, comparing results from plants grown in both greenhouse and field settings. We confirmed that greenhouse-grown plants have a relatively depauperate phyllosphere bacterial microbiome, which both makes them an ideal system for testing the impact of phyllosphere communities on plant health and important targets for microbial amendments as we move towards increased agricultural sustainability. We find that the addition of the synthetic microbial community early in greenhouse growth leads to an increase in fruit production in this setting, implicating the phyllosphere microbiome as a key component of plant fitness and emphasizing the role that these bacterial microbiomes likely play in the ecology and evolution of plant communities.

Abstract: Summary: Human genetic studies of critical COVID-19 pneumonia have revealed the essential role of type I interferon-dependent innate immunity to SARS-CoV-2 infection. Conversely, an association between the HLA-B∗15:01 allele and asymptomatic SARS-CoV-2 infection in unvaccinated individuals was recently reported, suggesting a contribution of pre-existing T cell-dependent adaptive immunity. We report a lack of association of classical HLA alleles, including HLA-B∗15:01, with pre-omicron asymptomatic SARS-CoV-2 infection in unvaccinated participants in a prospective population-based study in the United States (191 asymptomatic vs. 945 symptomatic COVID-19 cases). Moreover, we found no such association in the international COVID Human Genetic Effort cohort (206 asymptomatic vs. 574 mild or moderate COVID-19 cases and 1,625 severe or critical COVID-19 cases). Finally, in the Human Challenge Characterisation study, the three HLA-B∗15:01 individuals infected with SARS-CoV-2 developed symptoms. As with other acute primary infections studied, no classical HLA alleles favoring an asymptomatic course of SARS-CoV-2 infection were identified.

50. The Politics of Fair and Affordable Housing in Metropolitan Atlanta: Challenges for Educational Opportunity

Abstract: This qualitative case study considers the politics of the coalitions that emerged to address fair and affordable housing in the Atlanta metropolitan area between 2017 and 2020 and the connections or barriers they identified to educational opportunity. It applies regional, civic capacity, and social construction of policy frameworks to explore in which arenas the two issue areas of housing and education have been linked by policymakers, and by selected civic entities, non-profit organizations, and philanthropies. The case draws on demographic data, interviews, and documents in the analysis of the barriers to, and possibilities for, coordination of housing and education policy instruments to promote educational opportunity in the region. The author found that the majority of efforts to bridge the two policy areas were developed and led by non-profit actors, and mostly took the form of place-based interventions rather than mobility programs. Implications of these findings for policy design are discussed.
