The objective of this study was to evaluate the potential of selection for feed utilization on associated blood plasma metabolite and hormone traits. Dry matter intake (DMI) was recorded in 970 Holsteins from 11 commercial farms in Pennsylvania and used to derive dry matter efficiency (DME; fat-corrected milk yield/DMI), crude protein efficiency (CPE; protein yield/crude protein intake), and residual feed intake (RFI, defined as actual feed intake minus expected feed intake for maintenance and milk production, based on calculation of DMI adjusted for yield, body weight, and body condition score). Estimated breeding values for the 4 feed utilization traits (DMI, DME, CPE, and RFI), yield traits, body traits, and days open were standardized according to their respective genetic standard deviations. Up to 631 blood samples from 393 cows from 0 to 60 d in milk (DIM) were evaluated for blood plasma concentrations of glucose, nonesterified fatty acids (NEFA), β-hydroxybutyrate (BHB), creatinine, urea, growth hormone (GH), 3,5,3′-triiodothyronine (T3), and other parameters. Blood plasma traits were regressed on DIM, lactation number, herd, and standardized genetic merit. Cows with higher genetic merit for yield had significantly higher concentrations of GH, NEFA (milk and protein yield), and BHB (fat yield) from 31 to 60 DIM, but lower concentrations of glucose from 0 to 30 DIM, and T3 (milk yield, 0–60 DIM). The high GH–low glucose–low T3 concentration pattern was further accentuated for cows with genetic merit for enhanced feed efficiency (higher DME and lower RFI). Cows with a genetic tendency to be thin (low body condition score) also had elevated GH concentrations, but lower blood glucose, creatinine, and T3 concentrations. Those characteristics associated with enhanced feed efficiency (higher GH and lower glucose and T3 concentrations) were unfavorably associated with fertility, as indicated by elevated days open. Elevated NEFA and BHB concentrations were also associated with extended days open. Consideration of metabolic profiles when evaluating feed efficiency might be a method of maintaining high levels of health and reproductive fitness when selecting for feed efficiency. Colostrum formation is thought to occur slowly over an extended period (4 wk) prepartum. Furthermore, colostrum formation is highly variable among cows in total volume, IgG1 concentration, and mass obtained at first postpartum milking. Recent work has suggested that a rapid transfer of IgG1 to secretions may occur if animals are milked prepartum. Our objective was to establish the concentration, mass, and mass transfer rates of IgG1 in multiparous Holstein cows (n = 11, parity = 3.6 ± 1.1) milked prepartum (-74 to -1 h) and again around 4 h postpartum. Blood concentrations of IgG1 were very low (<1 mg/mL) in 7 cows at prepartum milking and did not decline following prepartum milking. Cows showed variability in the capacity to recover total volume, IgG1 concentration, and IgG1 mass. Three groupings of cows were considered based on the time between the 2 milkings (prepartum + 4 h postpartum): long-time (-74 to -54 h, n = 3), medium-time (-25 to -17 h, n = 4), and short-time (< -13 h, n = 4) groups. The average rates of transfer of these groups were 1.4 ± 0.8, 3.0 ± 1.3, and 25.1 ± 15.8 g/h, respectively. The data indicate that a longer time between prepartum and postpartum milking is not a main factor in IgG1 secretion transfer. Furthermore, because blood concentrations did not change after prepartum milking and the mass of blood plasma IgG1 was not sufficient to account for the mass occurring in postpartum colostrum, a source of IgG1 other than blood circulation appears to be present during colostrogenesis. The objectives of this study were to quantify the relationships of various definitions of feed utilization with both fertility and productive life. Intake and body measurement data were collected monthly on 970 cows in 11 tie-stall herds for 6 consecutive months. Measures of feed utilization for this study were dry matter intake (DMI), dry matter intake efficiency (DME, defined as 305-d fat-corrected milk/305-d DMI), DME with intake adjusted for maintenance requirements (DMEM), crude protein efficiency (defined as 305-d protein yield/305- d crude protein intake), and 2 definitions of residual feed intake (RFI). The first, RFIreg, was calculated by regressing daily DMI on daily milk, fat, and protein yields, body weight (BW), daily body condition score (BCS) gain or loss, the interaction between BW and BCS gain or loss, and days in milk. The second, RFINRC, was estimated by subtracting 305-d DMI predicted according to their fat-corrected milk and BW from actual 305-d DMI. Data were analyzed with 8-trait animal models and included one measure of feed utilization and milk, fat, and protein yields, BW, BCS, days open (DO), and productive life (PL). The genetic correlation between DME and DO was 0.53 (±0.19) and that between DME and PL was 0.66 (±0.10). These results show that cows who had higher feed efficiency had greater DO (undesirable) and greater PL (desirable). Results were similar for the genetic correlation between DO and crude protein efficiency (0.42). Productive life had genetic correlations of -0.22 with BW and -0.48 with BCS, suggesting that larger, fatter cows in this study had shorter PL. Correlations between estimated breeding values for feed utilization and official sire genetic evaluations for fertility were in agreement with the results from the multiple-trait models. Telomere length variation may provide a quantitative measure of the effects of dairy management and selection practices on animal stress and welfare. The objective of this study was to evaluate the association between telomere length in Holstein cattle with age, herd, and survival. A multiplex quantitative PCR (qPCR) procedure was utilized to estimate telomere length for 201 Holstein cows from 10 herds following DNA extraction from blood. Primers were designed to amplify a 79-bp telomere product and a 144-bp product of a standard reference gene (β-globin). Both primer sets were included in the same reaction well to enable the analysis of relative quantity (qT) of telomere product compared with β-globin product. Triplicate samples were run for each cow, and mixed models were used to analyze the qPCR results. Younger cows were significantly associated with higher qT, and significant variation was observed among herds for qT. Cows with short telomeres were more likely to be culled in the subsequent year than cows with above-average telomere lengths. Multiplex qPCR provides a cost-effective method of assessing telomere length. Variation in telomere length might provide insights into how management practices and genetic selection influence cow stress and physiological responses to stress. [ABSTRACT FROM AUTHOR]

Data from 879 Holstein cows from 11 tie-stall herds in Pennsylvania were analyzed to determine the effects of nutritional management practices on the level of genetic expression for milk, fat, and protein yields. Environments were defined according to the amount of dry matter refusals at the end of 24 h for the average cow (DMR), diet crude protein percentage (CP), and diet NEL concentration. Sire predicted transmitting ability (PTA) was available for all cows, whereas 775 cows were genotyped and received a molecular breeding value (MBV) for milk, fat, and protein yields. Milk, fat, and protein yields were regressed on sire PTA and cow MBV independently in addition to combined breeding values (CBV) of sire PTA and cow MBV. Four-trait animal models with fat-corrected milk yield in high and low environments plus either body weight or body condition score in high and low environments treated as separate traits were also evaluated. Regressions on sire PTA (0.31 for fat yield to 0.54 for milk yield) were significantly lower in the 5 herds that had the lowest average DMR than in the 6 herds with highest average DMR (0.82 for fat yield to 1.11 for protein yield). The regressions of milk and protein yield on CBV were also significantly lower in the 5 herds with low NEL concentration in the ration than in herds that had high NEL concentration. Genetic correlations from animal models showed that large cows were more affected by low DMR, CP, and NEL concentration than smaller cows. Efforts to minimize feed wastage must ensure that cows receive adequate nutrient intake to avoid suppression of genetic potential for yield, particularly for larger cows. [ABSTRACT FROM AUTHOR]

The objectives of this study were to estimate genetic parameters for body weight (BW) and BW change (BWC) and genetic correlations of BW and BWC with diseases and genomic predicted transmitting abilities (PTA) of productive and conformation traits of Holsteins during the first 120 DIM. Daily BW data were from the Afiweigh cow body weighing system (SAE Afikim, Kibbutz Afikim, Israel), which records BW as a cow exits the milking parlor. Disease categories included metabolic diseases, ketosis, infectious diseases, mastitis, reproductive diseases, and other diseases. Edited data included 68,914 and 11,615 daily BW observations from 441 Pennsylvania State University and 72 Virginia Tech Holstein cows, respectively. Two-trait random regression models were used to estimate relationships between BW, BWC, and diseases at 25, 38, and 58 mo of age at calving. Fixed effects for BW were age at calving nested within lactation group, week of lactation, and herd date; random effects for BW included animal, permanent environment, and error. Fixed effects for disease were herd-year-season of calving and age at calving nested within lactation group; random effects for disease were animal, permanent environment (for mastitis only), and error. Correlations of PTA for BW and BWC with genomic PTA for productive and type traits were also estimated with data from 117 cows. Heritability estimates for daily BW ranged from 0.34 to 0.63. Greater BW and less BWC were favorably correlated with ketosis, metabolic diseases, infectious diseases, and other diseases. The genetic correlation estimate between BW and ketosis was strongest at 60 DIM (-0.51), and genetic correlation estimates at 60 DIM with metabolic diseases (-0.52), infectious diseases (-0.81), and other diseases (-0.48) followed the same trend as ketosis. The genetic correlation estimate between BWC and ketosis was strongest for the change from 5 to 20 DIM (0.70) and was similar for metabolic diseases (0.37), infectious diseases (0.74), and other diseases (0.49). Correlations of BW and BWC with reproductive diseases tended to be in the reverse direction of those reported for ketosis. A larger PTA for BW was significantly correlated with smaller genomic PTA for milk yield, dairy form, rear udder height, and udder cleft. Predicted transmitting ability for BWC was negatively correlated with genomic PTA for protein percentage, strength, and hip width (ranging from -0.26 to -0.13 across lactation) and was positively correlated with dairy form, rear udder height, and udder cleft (ranging from 0.20 to 0.37 across lactation). The objectives of this study were to compare alternative mastitis definitions and to estimate genetic correlations of producer-recorded mastitis with somatic cell score (SCS) and yield. Cow health events and lactation records from June 2002 through October 2007 were provided by Dairy Records Management Systems (Raleigh, NC). First- through fifth-lactation records from cows calving between 20 and 120 mo of age and that calved in a herd-year with at least 1% of cows with a clinical mastitis event were retained. The edited data contained 118,516 lactation records and 1,072,741 testday records of 64,893 cows. Mastitis occurrence (1 = at least one mastitis event during lactation or test-day interval, 0 = no mastitis events), number of mastitis events during lactation, SCS, and yield were analyzed with animal models (single trait) or sire-maternal grandsire models (multiple trait) in ASREML. Comparisons were made among models assuming a normal distribution, a binary distribution, or Poisson distribution (for total episodes). The overall incidence of clinical mastitis was 15.4%; and heritability estimates ranged from 0.73% (test-day interval mastitis with a linear model) to 11.07% (number of mastitis episodes with a Poisson model). Increased mastitis incidence was genetically correlated with higher SCS (range 0.66 to 0.88) and was generally correlated with higher yield (range -0.03 to 0.40), particularly during first lactation (0.04 to 0.40). Significant genetic variation exists for clinical mastitis; and health events recorded by producers could be used to generate genetic evaluations for cow health. Sires ranked similarly for daughter mastitis susceptibility regardless of how mastitis was defined; however, test-day interval mastitis and a total count of mastitis episodes per lactation allow a higher proportion of mastitis treatments to be included in the genetic analysis. The objectives were to describe culling patterns and reasons for culling across lactation, estimate mortality and the proportion of cows leaving from 21 d before an expected calving date through 60 d in milk (DIM; CULL60) for Pennsylvania (PA) dairy herds, and to describe production measures for herds with high and low mortality and CULL60. Weekly culling frequencies and reasons for culling from 3 wk before a reported expected calving date through ≥100 wk of lactation were calculated for all PA cows with at least 1 Dairy Herd Improvement test in 2005. It was estimated that at least 5.0% of PA dairy cows died in 2005, and that at least 7.6% were culled by 60 DIM. The majority of cows exiting the herd by 60 DIM either died (35.1%) or had a disposal code of injury/other (29.9%). A total of 137,951 test-day records from 20,864 cows in herds with high mortality (>8.0%) and CULL60 (>12.0%) and 136,906 test-day records from 12,993 cows in herds with low mortality (<1.4%) and CULL60 (<2.9%) were retained to describe differences among herds with high and low survival. Least squares means for weekly milk yield, fat and protein percentages, and somatic cell score (SCS) were estimated with a model that included fixed effects for herd environment (high or low survival) and week nested within herd environment and lactation; random effects were cow, herd-test-day, and error. Cows from herds with high mortality and CULL60 produced more milk in lactations 1 (+1.9 ± 0.15 kg/d) and 2 (+0.9 ± 0.16 kg/d), but less in lactations 4 (-0.7 ± 0.22 kg/d), 5 (-1.4 ± 0.29 kg/d), and ≥6 (-0.7 ± 0.32 kg/d) and had higher SCS (+0.24 ± 0.02), more change in early-lactation fat percentage (-1.77% vs. -1.59%), and a greater frequency of fat-protein inversions (3.6 ± 0.3%). There is an opportunity to manipulate management practices to reduce mortality and early-lactation culling rates, which will improve cow welfare and the efficiency of dairy production by capturing a greater proportion of potential lactation milk yield, increasing cow salvage values, and reducing replacement costs. The objectives of this study were to estimate heritability for daily body weight (BW) and genetic correlations of daily BW with daily milk yield (MY), body condition score (BCS), dry matter intake, fat yield, and protein yield. The Afiweigh cow body weighing system records BW of every cow exiting the milking parlor. The Afiweigh system was installed at the Pennsylvania State University dairy herd in August 2001 and in July 2004 at the Virginia Tech dairy herd. The edited data included 202,143 daily BW and 290,930 daily MY observations from 575 Pennsylvania State University and 120 Virginia Tech Holstein cows. Data were initially analyzed with a series of 4-trait animal models, followed by random regression models. The models included fixed effects for age within lactation group, week of lactation, and herd-date. Random effects included animal, permanent environment, and error. The order of the polynomials for random animal and permanent environmental effects with the random regression model for daily BW was 4 and 6, respectively. Heritability estimates for daily BW ranged from 0.48 to 0.57 and were largest between 200 and 230 and smallest at 305 d of lactation. Genetic correlations were large between BW and BCS (0.60). The genetic correlation between daily BW and MY was -0.14 but was positive (0.24) after adjusting for BCS. Electronically recorded daily BW is highly heritable, and genetic evaluations of daily BW and BW change across the lactation could be used to select for less early lactation BW loss. The objectives of this study were to compare (co)variance parameter estimates among subsets of data that were pooled from herds with high, medium, or low individual herd heritability estimates and to compare individual herd heritability estimates to REML heritability estimates for pooled data sets. A regression model was applied to milk yield, fat yield, protein yield, and somatic cell score (SCS) records from 20,902 herds to generate individual-herd heritability estimates. Herds representing the 5th percentile or less (P5), 47th through the 53rd percentile (P50), and the 95th percentile or higher (P95) for herd heritability were randomly selected. Yield or SCS from the selected herds were pooled for each percentile group and treated as separate traits. Records from P5, P50, and P95 were then analyzed with a 3-trait animal model. Heritability estimates were 23, 31, 26, and 8% higher in P95 than in P5 for milk yield, fat yield, protein yield, and SCS, respectively. The regression techniques successfully stratified individual herds by heritability, and additive genetic variance increased progressively, whereas permanent environmental variance decreased progressively as herd heritability increased. The objectives of this study were to estimate heritabilities within herds participating in Dairy Herd Improvement and determine the relationship of the individual herd heritability with sire misidentification rate. Individual herd heritabilities for milk, fat, and protein yield and somatic cell score (SCS) were calculated with daughter-dam regression and daughter-sire predicted transmitting ability (PTA) regression using 4,712,166 records from 16,336 herds available for August 2000 evaluations and 7,084,953 records from 20,920 herds available for August 2006 evaluations. Herd heritabilities were estimated using regression models that included fixed breed, age within parity, herd-year-season of calving, dam records nested within state, sire PTA within state, and an interaction between sire PTA and herd variance; random regression coefficients were dam records within herd and sire PTA within herd. Average daughter-dam herd heritability estimates ranged from 0.21 (SCS in 2000) to 0.73 (protein percentage in 2006), whereas daughter-sire herd heritability ranged from 0.10 (SCS in 2000) to 0.42 (protein percentage in 2006). Verification of sire identification with DNA marker analysis was provided by Accelerated Genetics and Alta Genetics Inc. Daughter-sire herd heritability was more strongly correlated with sire misidentification rate than daughter-dam herd heritability. The correlation between the first principal component for all measures of herd heritability and sire misidentification rate was -0.38 (176 herds) and -0.50 (230 herds) in 2000 and 2006, respectively. Herd heritability can be estimated with simple regression techniques for several thousand herds simultaneously. The herd heritability estimates were correlated negatively with sire misidentification rates and could be used to identify herds that provide inaccurate data for progeny testing. The objectives of this study were to compare Holstein (HO), Brown Swiss (BS), and their crosses for milk, fat, and protein yields, somatic cell score (SCS), days open (DO), and age at first calving (AFC), and to estimate the effects of heterosis and recombination. First through fifth lactation records were obtained from 19 herds milking crosses among BS and HO. The edited data set included 6,534 lactation records from 3,473 cows of the following breed combinations: 2,125 pure HO, 926 pure BS, 256 BS sire x HO dam (SH), 105 backcrosses to BS (SX), 18 HO sire x BS dam, and 43 backcrosses to HO. Least squares means for daily milk, fat, and protein yields, mature-equivalent milk, fat, and protein yields, SCS, DO, and AFC were calculated for breed combinations with a model that included fixed effects of age within parity (except for AFC), days in milk for daily yield and SCS, herd-year-season of calving, and breed combination. Cow and error were random effects. Breed combination was replaced with regressions on coefficients for heterosis and recombination in a second analysis. Last, data were analyzed with a 5-trait animal model that included a single pedigree file for both breeds and coefficients for heterosis and recombination. The least squares means for fat production were 1.21, 1.15, 1.27, and 1.16 kg for HO, BS, SH, and SX, respectively, which corresponds to a heterosis estimate of 7.30% and a recombination estimate of -3.76%. Heterosis and recombination estimates for protein production were 5.63% and -3.31%, respectively. Heterosis estimates increased for fat yield (10.38%) and protein yield (7.07%) when maternal grandsire identification from a known artificial insemination sire was required. Regression coefficients indicated an 11.44-d reduction in DO due to heterosis. Heterosis estimates for SCS were inconsistent. Regression on heterosis for SCS was significant and favorable (-0.22) when the breed of sire was BS, but nonsignificant and unfavorable when sire breed was HO (0.43). Heterosis estimates were favorable for all traits, whereas recombination effects tended to be unfavorable for yield traits. Reduced performance of future generations did not appear to be the result of inseminating crossbred cows with inferior sires. Results indicated that first-generation crosses among BS and HO compared favorably with HO. Yield in subsequent generations was somewhat below expectations, perhaps due to recombination loss in HO. Estimates of heritability within herd (h²WH) that were generated with daughter-dam regression, daughter-sire regression, and REML were compared, and effects of adjusting lactation records for within-herd heritability on genetic evaluations were evaluated. Holstein records for milk, fat, and protein yields and somatic cell score (SCS) from the USDA national database represented herds in the US Northeast, Southeast, Midwest, and West. Four data subsets (457 to 499 herds) were randomly selected, and a large-herd subset included the 15 largest herds from the West and 10 largest herds from other regions. Subset heritabilities for yield and SCS were estimated assuming a regression model that included fixed covariates for effects of dam yield or SCS, sire predicted transmitting ability (PTA) for yield or SCS, herd-year-season of calving, and age within parity. Dam records and sire PTA were nested within herd as random covariates to generate within-herd heritability estimates that were regressed toward mean h²WH for the random subset. Heritabilities were estimated with REML using sire models (REMLSIRE), sire-maternal grandsire models (REMLMGS), and animal models (REMLANIM) for each herd individually in the large-herd subset. Phenotypic variance for each herd was estimated from herd residual variance after adjusting for effects of year-season and age within parity. Deviations from herd-year-season mean were standardized to constant genetic variance across herds, and records were weighted according to estimated error variance to accommodate h²WH when estimating breeding values. Mean h²WH tended to be higher with daughter-dam regression (0.35 for milk yield) than with daughter-sire regression (0.24 for milk yield). Heritability estimates varied widely across herds (0.04 to 0.67 for milk yield estimated with daughter-dam regression), and h²WH deviated from subset means more for large herds than for small herds. Correlation with REMLANIM h²WH was 0.68 for daughter-dam and was 0.45 for daughter-sire h²WH for milk yield. The correlation between daughter-sire h²WH and REMLMGS was greater than the correlation between daughter-dam h²WH and REMLMGS. Data adjustments had a minimal impact on breeding value bias. Within-herd heritability can be estimated rapidly using regression techniques with moderate accuracy, but adjusting lactation records for h²WH resulted in only a small improvement in the accuracy of genetic evaluations. The objectives of this study were to estimate genetic correlations among body condition score (BCS), dairy form, milk yield, and days open in US Holsteins and investigate the potential of using BCS or dairy form evaluations as early indicators of days open. Dairy form and BCS obtained from the Holstein Association USA, Inc., were merged with mature equivalents (ME) for milk yields and days open data from AIPL-USDA. Cows were required to be classified between 24 and 60 mo of age, before 335 d in milk (DIM) and have ME milk of at least 4537 kg. A minimum of 20 daughters per sire and 10 cows per herd-classification visit (HV) or herd-year-season of calving (HYS) were required. The final data set included 159,700 records. Heritabilities and correlations among dairy form, BCS, milk yield, and days open were estimated with multiple trait sire models. Fixed effects included age at classification for dairy form and BCS, age at calving for milk yield, HV for dairy form and BCS, HYS for milk yield and days open, DIM within lactation group for dairy form and BCS and lactation group for milk yield and days open. Correlations among dairy form, BCS, and days open were generated with and without a ME milk covariable. Correlations between ME milk and days open were generated with and without covariables for dairy form or BCS. Random effects included sire and error. The genetic correlation estimates of days open with dairy form, BCS, and ME milk were 0.48, -0.30, and 0.38, respectively. The genetic correlation estimate between days open and dairy form was 0.38 after adjustment for ME milk, whereas the genetic correlation between days open and BCS was -0.24 after adjustment for ME milk. Combining dairy form evaluations with days open evaluations for 19 recently proven bulls resulted in an average The addition of BCS evaluations did not increase reliability when dairy form observations were available. The objectives of this study were to estimate genetic correlations among body condition scores (BCS) from various sources, dairy form, and measures of cow health. Body condition score and dairy form evaluated during routine type appraisal was obtained from the Holstein Association USA, Inc. A second set of BCS was obtained from Dairy Records Managements Systems (DRMS) and was recorded by producers that use PCDART dairy management software. Disease observations were obtained from recorded veterinarian treatments in several dairy herds in the United States. Estimated breeding values for diseases in Denmark were also obtained. Genetic correlations among BCS, dairy form, and cow health traits in the United States were generated with sire models. Models included fixed effects for age, DIM, and contemporary group. Random effects included sire, permanent environment, herd-year season for health traits, and error. Predicted transmitting abilities (PTA) for BCS and dairy form were correlated with estimated breeding values for disease in Denmark. The genetic correlation estimate between BCS from DRMS and BCS from the Holstein Association USA, Inc., was 0.85. The genetic correlation estimate between BCS and a composite of all diseases in the United States was -0.79, and PTA for BCS was favorably correlated with an index of resistance to disease other than mastitis in Denmark (0.27). Dairy form was positively correlated with a composite of all diseases in the United States (0.85) and was unfavorably correlated with an index for resistance to disease other than mastitis in Denmark (-0.29). Adjustment for protein yield PTA had a minimal affect on correlations between PTA for BCS or dairy form and disease in Denmark. Selection for higher body condition or lower dairy form with continued selection for yield may slow deterioration in cow health as a correlated response to selection for increased yield. The objectives of the current study were to investigate the relationship between body condition score (BCS) and dairy form and changes in genetic parameters for BCS and dairy form within and across lactations and age. Body condition score and dairy form were obtained from the Holstein Association USA, Inc. Records were edited to include those cows classified between 24 and 60 mo of age and between 0 and 335 d in milk (DIM). A minimum of 20 daughters per sire and 15 cows per herd-classification visit were required. The dataset consisted of 135,178 records from 119,215 cows. Repeatability, multiple trait, and random regression models were used to analyze the data. All models included fixed effects for herd-classification visit, age within lactations 1, 2, and 3 or higher, and 5th-order polynomials for DIM. Random effects included sire and permanent environment for all models. Random regression models included age at classification nested within sire or DIM and lactation number nested within sire. Genetic variance for both BCS and dairy form was lowest in early lactation and highest in midlactation. Genetic correlations within and across lactations were high. The genetic correlation between DIM 0 in lactation 1 and DIM 305 in lactation 3 was estimated to be 0.77 for BCS and 0.60 for dairy form. The genetic correlation estimate between 30 mo of age at classification and 50 mo of age at classification was 0.94 for both dairy form and BCS. A total of 1,257,333 insemination records from January of 2000 through December of 2004 were provided by Genex Cooperative, Inc. Insemination date, herd number, and cow identification number were used to determine if cows were re-inseminated within 90 days (NR90) of insemination. First and second services by Holstein sires in herd-years that had at least 300 recorded services and herd-months with at least 8 services were retained. A minimum of 10 services per sire, 50 services per technician, and a herd non-return rate between 10% and 80% were required for inclusion in the dataset. The final dataset included 60,718 records from 152 herds. Daily maximum temperature-humidity index (THI) from weather stations representing six regions in Pennsylvania was merged with insemination data. Maximum THI on the day of insemination, 1 day after insemination, and average maximum THI for the week of insemination were analyzed. Statistical analyses were performed in ASReml with NR90 treated as a binary response variable. Fixed effects included herd-year, service number, technician and THI. Random effects were cow, bull and error. Month of insemination was included as a fixed effect for one analysis. Average NR90 was 50%, which represents the percentage of cows that were not re-inseminated within 90 days. The odds of not returning to service were highest for services in March and lowest in August, and the odds ratio for March to August non-return was 1.36 to 1. Temperature-humidity index effects were strongest when the average maximum THI for the week of insemination was considered. The odds ratio of NR90 when THI was in the 75th percentile (23°C) to NR90 at median THI (15°C) was 0.92:1.00. The odds ratios of NR90 at the 95th percentile THI (28°C) to the median THI was 0.81:1.00. Results indicated that heat stress can have severe impacts on conception rates in Pennsylvania. The objectives of this study were to investigate the relationship between body weight change between calving and 30 days in milk and occurrence of dairy cattle disease. Daily body weight was available from 515 cows, of which 346 Holstein cows were from the Penn State herd, and 120 Holstein and 49 Jersey cows were from the Virginia Tech herd. Daily body weights were recorded twice daily by AfiFarm upon a cow exiting the milking parlor. The average of at least five weights during the first week of lactation and at least five weights between 27 and 33 days in milk were calculated for each cow. Body weight change was defined as average BW during the first week of lactation minus average body weight during the fourth week of lactation. Diseases were recorded by herd managers at the respective universities; disease categories (1 = diseased, 0 = not diseased) included displaced abomassum, ketosis, all metabolic disease, uterine disease, mastitis, and all diseases. The total disease frequency was 48% in the Penn State herd and 31% in the Virginia Tech herd. The LOGISTIC procedure of SAS was used to analyze disease from the Penn State herd with fixed lactation number, age, year-season, and body weight change effects. The average body weight change during the first 30 days of lactation was -44 kg. The effect of body weight change was significant for displaced abomasum, ketosis, metabolic disease, uterine disease, and all diseases. Odds ratios were calculated for weight change at the 75th percentile (-69 kg) to weight change at the 25th percentile (-20 kg). Cows losing more weight had higher odds of developing a displaced abomasum (1.41:1), ketosis (2.24:1), metabolic disease (1.62:1), and all diseases (1.43:1). Cows with uterine diseases (0.80:1) lost less weight. The objectives of this study were to determine if within-herd heritability (WHH) estimated with regression techniques accurately reflects heritability (h2) differences among herds and to compare sire and animal model genetic parameter estimates among herds varying in WHH. Milk, fat, and protein yield and somatic cell score (SCS) were obtained from states representing the Northeast, Southeast, Midwest and Western regions of the United States. Four random subsets of data were selected across regions. Within-herd heritabilities were estimated with daughter-dam regression (h2 DD) and daughter-sire predicted transmitting ability (PTA) regression (h2 DS) in ASReml with a model that included fixed effects of herd-year-season of calving, age, dam yield, and sire PTA. Dam yield and sire PTA were also treated as random covariables nested within herd to create h2 DD and h2 DS that were regressed toward the subset average. Herds were ranked from lowest to highest WHH for each trait and average WHH rank was calculated. Fifty herds with the highest and lowest average WHH were retained for further analysis in each subset. Milk, fat, protein and SCS were each considered two separate traits (one in the high, another in the low h2 herds) and analyzed with two trait sire and animal models in ASReml. Fixed effects included herd-year-season of calving, age and lactation number. Random effects included animal, permanent environment and error. Sire and animal model h2 estimates were greater in herds with high h2 for all traits. Average h2 DD across subsets for milk yield was 0.41 for high h2 herds and 0.29 for low h2 herds, while average milk yield h2 DS were 0.35 and 0.13 from high and low h2 herds, respectively. Sire model h2 estimates were more highly correlated with h2 DS (0.93) than with h2 DD (0.85). Animal model h2 estimates were more highly correlated with h2 DD (0.91) than with h2 DS (0.72). There are significant differences among herds for h2 and it appears WHH estimated with daughter-dam and daughter-sire PTA regression reflect those differences with some accuracy. The objectives of this study were to estimate the heritability of daily body weight and to estimate the genetic correlation between body weight and daily milk production. The Afiweigh cow body weighing system records the weight of every cow exiting the milking parlor. The Afiweigh system was installed at the Penn State dairy herd in August of 2001 and in July of 2004 at the Virginia Tech dairy herd. Daily body weight and milk yield were available for 542 Penn State Holstein cows and 120 Holstein cows from the Virginia Tech dairy herd. Body weight and milk yield recorded after 365 days in milk were eliminated. Outliers were detected by generating a body weight and milk yield curve for each cow and daily records more than four standard deviations from the predicted curves were removed. A total of 159,388 daily body weights and 235,144 daily milk records from the Penn State herd were analyzed with a two-trait animal model in ASReml. The model included lactation, age, date and days in milk as fixed effects and animal, permanent environment, and error as random effects. Average daily milk yield was 36.5 kg and average body weight was 654 kg. Heritability estimates for body weight and milk yield were 0.62 and 0.21, respectively, and repeatability estimates were 0.86 and 0.66, respectively. The genetic correlation estimate between body weight and milk yield was -0.61 with a standard error of 0.13. The phenotypic correlation estimate was -0.33 with a standard error of 0.04. Body weight can be measured accurately as cows exit the milking parlor and could be used to generate genetic evaluations for body weight. The genetic and phenotypic relationship between body weight and milk yield was negative, indicating that most body weight differences at the Penn State dairy herd may be due to differences in body condition. The objectives of this study were to compare milk, fat, and protein yield, somatic cell score, days open, and age at first calving between Holstein (HO) and Brown Swiss (BS), and among BS and HO crosses and backcrosses. A minimum of five first lactation cows entering a herd in the same year was required and only cows with a first lactation record and a registered HO or BS sire were retained. Data from 16 herds included 5,148 total records from 1,604 HO, 736 BS, 238 BS x Ho (SH), 19 HO x BS, and 60 BS x SH backcrosses (SSH). Data was retrieved through PCDART or Dairy Comp 305 for lactations one through five. Average daily milk, fat, and protein yield, SCS, days open, and age at first calving were analyzed using the MIXED procedure of SAS. The fixed effects were days in milk, age, parity, and breed. Random effects included cow and residual error. Breed effects were significant for all yield traits and age at first calving, approached significance for days open (P = 0.12), but were not significant for SCS (P = 0.48). Least square means (LSM) for daily milk yield were 32.5 kg for HO, 28.0 kg for BS, 31.4 kg for SH, and 28.8 kg for SSH. Least square means for daily fat yield were 1.17 kg for HO, 1.12 kg for BS, 1.22 kg for SH, and 1.14 kg for SSH. Daily protein yield LSM were 0.98 kg, 0.93 kg, 1.00 and 0.93 kg for HO, BS, SH, and SSH, respectively. Days open LSM were 159.1, 156.2, 148.3, and 149.2 for HO, BS, SH, and SSH respectively. Heterosis in SH crosses for yield traits ranged from 3.83% for daily milk yield to 6.56% for daily fat yield. Heterosis estimates were -5.93% for days open and -2.29% for age at first calving in SH crosses. Milk yield of SH were less than HO, but fat and protein yields were increased slightly. The objective of this study was to investigate the phenotypic relationship between common health disorders in dairy cows and lactation persistency, uncorrelated with 305-d yield. The relationships with peak yield and days in milk (DIM) at peak were also studied. Daily milk weights and treatment incidence records of 991 Holstein lactations from experimental dairy herds at Virginia Tech and Pennsylvania State University were used. Persistency was calculated as a function of daily yield deviations from standard lactation curves, developed separately for first (FL) and later lactations (LL), and deviations of DIM around reference dates: 128 for FL and 125 for LL. Days in milk at peak and peak yield were computed for each lactation by using Wood's function. The disease traits studied were mastitis (MAST) only during the first 100 d (MAST1), only after 100 DIM (MAST2), both before and after 100 DIM (MAST12), and at any stage of lactation (MAST1/2), as well as metritis, displaced abomasums, lameness, and metabolic diseases. Each disease was defined as a binary trait, distinguishing between lactations with at least one incidence (1) and lactations with no incidences (0). The relationships of diseases to persistency, DIM at peak, and peak yield were investigated separately for FL and LL for all disease traits except MAST12, which was investigated across parities. The relationships of persistency to probability of the diseases in the same lactation and in the next lactation were examined using odds ratios from a logistic regression model. Metritis and displaced abomasums in FL and LL were associated with significantly higher persistencies. Metabolic diseases and MAST1 in LL were significantly related to higher persistencies. The relationships of MAST2 in both FL and LL, and MAST12 across parities were significant but negative. Cows affected by MAST tended to have less persistent lactations. Most of the diseases had a significant impact on DIM at peak in LL. In LL, metritis, metabolic diseases, and displaced abomasums tended to significantly delay DIM at peak. Mastitis only after 100 DIM was associated with significantly earlier DIM at peak in LL. Increasing persistency was associated with low MAST2 and MAST1/2 in primiparous cows. The objectives of this study were to estimate the heritability of milk urea nitrogen (MUN) concentration and describe the genetic relationship between MUN and reproductive performance and between MUN and diseases in Holsteins. Dairy Records Management Systems (Raleigh, NC) provided lactation data. The Danish Agricultural Advisory Center provided breeding value estimates for diseases. Infrared (IR) and wet chemistry (WC) data were analyzed separately. Heritabilities and genetic correlations for 2 different measures of MUN and reproductive performance were estimated with an animal model using ASREML. Heritabilities for MUN were estimated using all lactations combined (lactations 1 through 5) and separately for first lactation and second lactation. Genetic correlations with reproduction and health were estimated separately for parities 1 and 2. Herd-test-day or herd-year-season along with age at calving and days in milk were included as fixed effects in all models. Heritability estimates for all lactations combined were 0.15 for WC MUN and 0.22 for IR MUN. Genetic correlations between WC MUN and 2 measures of reproductive performance, days to first service, and first service conception were not different from zero. In contrast, the genetic correlation between WC MUN and days open of 0.21 in first lactation and 0.41 in second lactation indicated that higher WC MUN values were associated with increased days open. Correlations among estimated breeding values for MUN and estimated breeding values for Danish diseases identified no significant relationships. Although the results of this study indicate that heritable variation for MUN exists, the inability to identify significant genetic relationships with several measures of disease or reproductive performance appears to limit the value of MUN in selection for disease resistance and improved reproduction. The objective of this study was to understand the associations of calf growth traits with subsequent milk yield and body weight (BW). Data were collected for 281 Holstein heifer calves from 6 different calf trials of varying lengths (4 to 8 wk) conducted at Pennsylvania State University between 2003 and 2010. Calves were classified as high, medium, or low for hip height, starter feed intake, BW, and growth rate. Milk yield and cow BW were recorded during subsequent lactations. In total, there were 169,734 daily milk records and 136,153 cow BW records available. Data were evaluated using mixed model equations. Separate models were used for each calf growth trait initially, followed by models that considered multiple growth measures. Each model included age at calving, treatment within trial, parity, days in milk, lactation, and one of the calf growth traits as well as the interaction between lactation and days in milk as fixed effects. Cow and calendar week by year were fitted as random effects. Heifers from the low hip height classification as calves produced less milk across lactations after accounting for BW differences. Cows from the medium BW classification as calves produced more milk in early lactation than cows from the high BW classification as calves after accounting for differences in height. Calves that grew more quickly, ate more, and weighed more were heavier as first-lactation heifers and as mature cows. Our results suggest that the type of preweaning growth is an important consideration for future milk yield. Changes in milk composition during a milking are well characterized, but variation in milk fatty acid (FA) profile is not well described and may affect the accuracy of in-line milk composition analyzers and could potentially be used for selective segregation of milk. Within-milking samples were collected from 8 multiparous high-producing Holstein cows (54.86 ± 6.8 kg of milk/d; mean ± standard deviation). A milk-sampling device was designed to allow collection of multiple samples during a milking without loss of vacuum or interruption of milk subsampling. Milk was collected during consecutive morning and afternoon milkings (12-h intervals) and was replicated 1 wk later. Each sample represented approximately 20% of the milking and was analyzed for fat, true protein, and lactose concentration and FA profile. Milk fat concentration markedly increased over the course of milk let down (4.4 and 4.2 percentage units at the a.m. and p.m. milking, respectively), whereas milk fat globule size did not change. Milk protein and lactose concentration decreased slightly during milking. Modest changes in milk FA profile were also observed, as milk de novo and 16-C FA concentrations increased approximately 10 and 8%, respectively, whereas the concentration of preformed FA decreased about 7% during the milking. In agreement, mean milk FA chain length and unsaturation modestly decreased during milking (0.59 and 0.014 U, respectively). The observed changes in milk fat concentration during a milking are consistent with previous reports and reflect the dynamic nature of milk fat secretion from the mammary gland. Changes in milk FA profile are not expected to practically affect the accuracy of spectroscopy methods for determination of milk fat concentration. Bovine IgG1 is thought to be specifically transported by a process of transcytosis across the mammary epithelial cells during colostrogenesis. Mammary IgG1 appearance in cow colostrum has typically been reported as a concentration and shows IgG1 concentration to be extremely variable because of animal variation, colostrum milking time, and water dilution effects. To identify animal IgG1 transfer capacity and separate it from the other effects, our objective was to determine first colostrum IgG1 total mass. We collected 214 samples of totally milked first colostrum with recorded colostrum weights from 11 Pennsylvania dairy farms that participated in Pennsylvania Dairy Herd Improvement Association, analyzed colostrum for IgG1 by ELISA, and calculated total IgG1 mass. Median and mean concentrations of IgG1 were 29.4 mg/mL and 37.5±30.2 mg/mL, respectively, with a range of 9 to 166 mg/mL. However, total mass of IgG1 had a median of 209.1g, mean of 291.6±315.8g, and a range of 14 to 2,223g. Colostrum IgG1 concentration showed no relationship with colostrum volume, but IgG1 mass had a positive relationship with volume. Colostrum IgG1 mass was related to IgG1 concentration (R2=0.58). Using DHIA records for 196 animals, we established milk production for these animals to a 15-d equivalent. An established milk secretion relationship to mammary parenchyma tissue (secretory tissue) was calculated and showed no relationship of IgG1 mass with mammary parenchyma tissue. In addition, we show that approximately 10% of the sampled animals had IgG1 mass greater than 1 standard deviation above the mean (high mass transfer) and represented all parities tested (1-7). Whereas first-lactation animals showed less overall calculated parenchyma tissue when compared with other parities, approximately 10% of the first-lactation group animals were capable of high mass transfer, with one transporting 2,029g into first colostrum. Concentration variance of IgG1 can be attributed to water inclusion, whereas mass transfer provides a clear indication of animal IgG1 transfer capacity. The specific mechanism of bovine mammary IgG1 transfer is not clear, but secretory tissue mass does not explain the variation observed. A recessive haplotype resulting in elevated calf mortality but with apparent incomplete penetrance was previously linked to the end of chromosome 16 (78.7-80.7 Mbp). Genotype analysis of 5.6 million Holsteins indicated that the haplotype was common and traced back to 1952, with a key ancestor born in 1984 (HOUSA1964484, Southwind) identified from chip genotypes as homozygous for the suspect haplotype. Sequence data from Southwind (an affected calf) and the sire of the affected calf were scanned for candidate mutations. A missense mutation with a deleterious projected impact at 79,613,592 bp was homozygous in the affected calf and heterozygous in the calf's sire and Southwind. Sequence data available from the Cooperative Dairy DNA Repository for 299 other Holsteins indicated a 97% concordance with the haplotype and an 89% call rate. The exon amino acid sequence appears to be broadly conserved in the CACNA1S gene, and mutations in humans and mice can cause phenotypes of temporary or permanent paralysis analogous to those in calves with the haplotype causing muscle weakness (HMW). Improved methods for using pedigree to track new mutations within existing haplotypes were developed and applied to the haplotypes for both muscle weakness and Holstein cholesterol deficiency (HCD). For HCD, concordance of the gene test with its haplotype status was greatly improved. For both defects, haplotype status was matched to heifer livability records for 558,000 calves. For HMW, only 46 heifers with livability records were homozygous and traced only to Southwind on both sides. Of those, 52% died before 18 mo at an average age of 1.7 ± 1.6 mo, but that death rate may be underestimated if only healthier calves were genotyped. The death rate was 2.4% for noncarriers. Different reporting methods or dominance effects may be needed to include HMW and other partially lethal effects in selection and mating. Direct tests are needed for new mutations within existing common haplotypes because tracking can be difficult even with accurate pedigrees when the original haplotype has a high frequency., (The Authors. In the United States, it is becoming common for dairy herds to mate a portion of cows to beef semen to create a value-added calf. The objectives of this study were to determine if dystocia risk, stillbirth (SB) risk, gestation length (GL), probability of early-lactation clinical disease events, early-lactation culling risk, or subsequent milk production differ between cows that carried calves sired by different beef breeds and those that carried Holstein-sired calves. Records from 10 herds contained 75,256 lactations from 39,249 cows that had calves with known Holstein or beef breed sires from the years 2010 to 2023. Calf sire breeds with ≥150 records included in analyses were Holstein, Angus, Simmental, Limousin, crossbred beef, and Charolais. Additional beef sire breeds that existed in lower frequency (n < 150 records) were condensed together and classified as "other." Because GL is a continuous variable, sire breed inclusion criteria were reduced to n ≥ 100 records; thus, Wagyu sires were included as their own breed group. Some records did not contain all variables of interest, thus models included fewer lactations depending on variable. Binomial generalized mixed models evaluated dystocia risk (defined as calving ease score ≥4 or calving ease score ≥3), SB risk, clinical health event risk (defined as lameness, mastitis, metabolic, reproductive, other, or any health events occurring within 60 d in milk [DIM]), and early culling risk (defined as death or culling within 60 DIM). Gestation length and test-date milk, fat, and protein yields were evaluated with mixed models. Calves sired by crossbred beef bulls had a greater probability of being stillborn (5%; 95% confidence interval lower = 2.9% upper = 9.0%) than those sired by Holstein bulls (2%; 95% confidence interval lower = 1.5%, upper = 2.7%). All beef-sired calves increased GL from that of Holstein-sired calves (277 ± 0.15 d) with Limousin (282 ± 0.81 d) and Wagyu-sired calves (285 d ± 0.79) resulting in the longest GL. The risk of dystocia, clinical health events, and early-lactation culling did not differ by calf sire breed nor did subsequent milk and component yield. Study aims were to investigate associations of hyperketolactia (HYKL) status of Holstein dairy cows between 6 and 60 d in milk (DIM), defined by milk acetone (mACE) and β-hydroxybutyrate (mBHB) content, with daily milk yield and composition. Milk samples (∼5.0 million) were collected over a 5-yr period (2014-2019) within the milk recording system in Poland. Concentrations of mACE and mBHB determined by Fourier-transform infrared spectroscopy were used to categorize samples into 4 ketolactia groups. Based on threshold values of ≥0.15 mmol/L mACE and ≥0.10 mmol/L mBHB, ketolactia groups were normoketolactia (NKL; mACE <0.15 mmol/L and mBHB <0.10 mmol/L), BHB hyperketolactia (HYKL BHB ; mACE <0.15 mmol/L and mBHB ≥0.10 mmol/L), ACE hyperketolactia (HYKL ACE ; mACE ≥0.15 mmol/L and mBHB <0.10 mmol/L), and ACE and BHB hyperketolactia (HYKL ACEBHB ; mACE ≥0.15 mmol/L and mBHB ≥0.10 mmol/L). To investigate ketolactia association with production outcomes, a linear model was developed, including ketolactia group, DIM, parity, their interactions, year-season as fixed effects, and random effects of herd and cow. Among all milk samples, 31.2% were classified as HYKL, and of these, 52.6%, 39.6%, and 7.8% were HYKL ACEBHB , HYKL BHB , and HYKL ACE , respectively. Ketolactia groups differed for all traits studied in all parities and DIM. Among HYKL groups, lowest milk yield was found in HYKL ACEBHB cows, except for 6 to 30 DIM in first- and second-lactation cows. Milk yield of HYKL BHB cows was higher than that of NKL cows until 20 to 30 DIM, and then it was lower than NKL cows. Milk yield of HYKL ACE cows was mostly lower than NKL cows. Energy-corrected milk (ECM) yield of HYKL ACEBHB cows was higher than that of NKL cows until 30 to 35 DIM for second lactation and third lactation or greater, and in the whole study period for first lactation. The yield of ECM for HYKL BHB cows was mostly higher than that of NKL cows, whereas HYKL ACE cows had higher ECM than NKL cows until 15 to 25 DIM and then was lower for the HYKL ACE group. Milk composition differed among HYKL groups. Highest milk fat (MF) and lowest milk lactose (ML) contents were observed in HYKL ACEBHB cows. Cows in HYKL ACEBHB and HYKL BHB groups had higher MF and lower milk protein (MP; except in 6-8 DIM in first lactation) and ML content than NKL cows. Milk fat content was higher in HYKL ACE than NKL cows in first lactation and during the first 30 to 40 DIM in older cows. Lactose content was lower in HYKL ACE than in NKL cows within 30 to 40 DIM; afterward it was higher in NKL cows. Lower MP content was found in HYKL ACE than in NKL cows, except during 6 to 9 DIM for cows in first lactation and third lactation or greater. In conclusion, HYKL is associated with altered milk production in all parities, but a range of these negative relations depends on ketone status addressing both ACE and BHB contents. Acetate supplementation increases milk fat production, but interactions with animal-related factors have not been investigated. The objective of this study was to characterize the interaction of acetate supplementation with parity and genetic potential for milk fat synthesis including the DGAT1 K232A polymorphism (AA and KA genotypes). In total, 47 primiparous and 49 multiparous lactating cows were used in 2 blocks in a crossover design. The basal diet was formulated to have a low risk of biohydrogenation-induced milk fat depression and had 32.8% and 32.0% neutral detergent fiber and 21.7% and 23.6% starch [all on a dry matter (DM) basis] in block 1 and 2, respectively. The control treatment received the basal diet, and the acetate supplementation treatment included anhydrous sodium acetate supplemented to the basal diet at 3.2% and 3.1% of DM of the diet for block 1 and 2, respectively (targeting 10 mol/d of acetate). The DGAT1 genotype frequency of the experimental cows was 45% AA and 51% KA, with 4% cows with either a KK or unimputable genotype. Acetate supplementation increased DM intake (DMI) in KA multiparous cows, but acetate did not change DMI in AA multiparous or primiparous cows of either genotype. Acetate supplementation increased the frequency of meals by 8% and decreased the length of each meal by ∼5 min compared with control. There was no effect of acetate on milk yield. Acetate supplementation increased milk fat yield and concentration by 117 g/d and 0.31 percentage units, respectively, regardless of DGAT1 polymorphism or parity. The increase in milk fat yield was mostly due to an increase in yield of 16C mixed-sourced fatty acids, suggesting that acetate supplementation drives mammary de novo synthesis toward completion. Response to acetate supplementation was not related to genomic predicted transmitting ability of milk fat concentration and yield or to pretrial milk fat percent and yield, suggesting that acetate increases milk fat production regardless of genetic potential for milk fat yield and level of milk fat synthesis. Interestingly, analyzing the temporal effect on the interaction between treatment and DGAT1 polymorphism on milk fat yield suggested that DGAT1 polymorphism may affect the short-term response to acetate supplementation during the first ≤7 d on treatment. Acetate supplementation also increased plasma β-hydroxybutyrate concentration and decreased plasma glucose concentration. This experiment was designed to investigate the relation of high and low methane-yield phenotypes with body weight (BW), dry matter intake (DMI), lactation performance, enteric CH 4 emissions, and rumen fermentation parameters in lactating dairy cows. A total of 130 multi- and primiparous Holstein cows were screened for enteric CH 4 emissions using the GreenFeed system (C-Lock Inc.). Out of these 130 cows, 5 were identified as phenotypically high (HM) and 5 as phenotypically low (LM) CH 4 emitters. Cows in the LM group had lower daily enteric CH 4 emissions than cows in the HM group (on average 346 vs. 439 g/d, respectively), lower CH 4 yield (15.5 vs. 20.4 g of CH 4 /kg of DMI), and CH 4 intensity (13.2 vs. 17.0 g of CH 4 / kg of energy-corrected milk yield). Enteric emissions of CO 2 and H 2 did not differ between HM and LM cows. These 10 cows were blocked by parity, days in milk, and milk production, and were used in a 5-wk randomized complete block design experiment. Milk composition, production, and BW were also not different between LM and HM cows. The concentration of total volatile fatty acids in ruminal contents did not differ between CH 4 phenotypes, but LM cows had a lower molar proportion of acetate (57 vs. 62.1%), a higher proportion of propionate (27.5 vs. 21.6%, respectively), and therefore a lower acetate-to-propionate ratio than HM cows. Consistently, the 16S cDNA analysis revealed the abundance of Succinivibrionaceae and unclassified Veillonellaceae to be higher in LM cows compared with HM cows, bacteria that were positively correlated with ruminal propionate concentration. Notably, Succinivibrionaceae trigger the formation of propionate via oxaloacetate pathway from phosphoenolpyruvate via Enzyme Commission: 4.1.1.49, which showed a trend to be higher in LM cows compared with HM cows. Additionally, LM cows possessed fewer transcripts of a gene encoding for methyl-CoM reductase enzyme compared with HM. In this study, low and high CH 4 -yield cows have similar production performance and milk composition, but total-tract apparent digestibility of organic matter and fiber fractions was lower in the former group of animals., (The Authors. Minimizing the incidence of disease on organic dairy farms is important for both economic and animal welfare purposes. The objective of this study was to estimate genetic parameters for total disease treatment costs using producer-recorded treatments in organic Holstein dairy calves and cows. Individual cow and calf health data were collected from 16 USDA certified organic farms from across the United States. Eleven of these farms provided treatment costs for some or all of the following cow health issues (mean cost): mastitis ($46.10), milk fever ($39.05), ketosis ($29.81), metritis ($28.66), retained placenta ($45.59), displaced abomasum ($439.71), lameness ($66.36), indigestion ($22.94), respiratory ($48.35), and died ($64.98). These farms also provided the following health costs for calves (mean cost): respiratory ($56.37) and scours ($25.21). Costs included consultant fees, therapeutics, and producer labor. The total lactational health cost (HCOST) was analyzed using animal models adjusted for the fixed effects of lactation and herd and the random effect of herd-year-season of calving with animal relationships based on the blending of pedigree and genomic relationships established from 2,347 genotyped cows. Along with HCOST, the binary traits stayability and presence of disease were included in a trivariate model such that lactations absent of disease were considered to be missing HCOST. To estimate the genetic relationship between nulliparous and primiparous health costs, a 2-trait linear model was fitted for total nulliparous health costs (NHCOST) and first lactation HCOST. The most expensive cow-lactation was $643.86 and 26.5% of lactations encountered disease. The heritability for HCOST was 0.03 ± 0.01, and the repeatability was 0.21 ± 0.01. The heritability of NHCOST was 0.06 ± 0.01, and the genetic correlation between NHCOST and HCOST was 0.98 ± 0.51. Traits representing the repeated nature of disease have a genetic component that should foster improved disease resistance among organic Holstein dairy cows. However, total cost of disease did not lead to gains in genetic variation over consideration of disease traits considered as binary variables and is a more laborious phenotype to obtain, diminishing its appeal for use in routine genetic evaluations., (© 2023.)

Thirty-four Holstein calves from multiple farms were found recumbent during the neonatal period with no detectable neurologic, infectious, or metabolic abnormalities. Most calves did not survive beyond 6 wk of age. The objective of this study was to conduct a genome-wide association and pedigree analysis to determine if a genetic origin was plausible. There were 101,917 DNA markers for 18 affected calves and 26 unaffected family controls available for analysis. Genome-wide association, homozygosity screening, and a parental based transmission disequilibrium test were conducted in PLINK. A genomic region on the end of chromosome 16 that contained 78 markers based on a recessive inheritance model and that spanned 5.1 million bp was considered the most probable region for a genetic defect; the region was narrowed to 2.1 million bp following homozygosity screening and the transmission disequilibrium test with all affected calves homozygous in the candidate region and 1 homozygous control. A genotyped sire and 2 dams with imputed genotypes were heterozygous in the candidate region. A common sire born in 2008 was identified that was present for both paternal and maternal lineages of all affected calves; nearly all lineages traced through a prolific son born in 2010 who was genotyped and was heterozygous for the candidate region. Therefore, a possible genetic defect with incomplete penetrance on chromosome 16 that results in recumbency has been identified. Among other regulations, organic cows in the United States cannot receive antibiotics and preserve their organic status, emphasizing the importance of prevention of illness and benefit of high genetic merit for disease resistance. At the same time, data underlying national genetic evaluations primarily come from conventional cows, drawing concern to the possibility of a genotype by environment interaction whereby the value of a genotype varies depending on the environment, and potentially limits the relevance of these evaluations to organic cows. The objectives of this study were to characterize the genetics of and determine the presence of genotype by environment interaction for health traits in US organic dairy cows. Individual cow health data were obtained from 16 US Department of Agriculture certified organic dairy farms from across the United States that used artificial insemination and maintained detailed records. Data were obtained for the following traits: died, lameness, mastitis, metabolic diseases (displaced abomasum, ketosis, and milk fever), reproductive diseases (abortion, metritis, and retained placenta), transition health events (any health event occurring 21 d before or after parturition), and all health events. Binary phenotypes (1 = diseased, 0 = otherwise) for 38,949 lactations on 19,139 Holstein cows were used. Genotypes from 2,347 cows with 87.5% or greater Holstein breed-based representation were incorporated into single-step multitrait threshold animal models that included stayability (1 = completed lactation, 0 = otherwise). Gibbs sampling was used. Genomic predicted transmitting abilities (gPTA) from national genetic evaluations were obtained for sires for production, fitness, health, and conformation traits. We approximated genetic correlations for sires using these gPTA and our estimated breeding values. We also regressed health phenotypes on cow estimated breeding values and sire gPTA. Heritabilities (± standard error) ranged from 0.03 ± 0.01 (reproductive diseases) to 0.11 ± 0.03 (metabolic diseases). Most genetic correlations among health traits were positive, though the genetic correlation between metabolic disease and mastitis was -0.42 ± 0.17. Approximate genetic correlations between disease resistance for our health trait categories and disease resistance for the nationally-evaluated health traits generally carried the expected sign with the strongest correlation for mastitis (0.72 ± 0.084). Regression coefficients carried the expected sign and were mostly different from zero, indicating that evaluations from primarily conventional herd data predicted health on organic farms. Horn flies (Haematobia irritans [L.]) contribute to major economic losses of pastured cattle operations, particularly in organic herds because of limitations on control methods that can be used. The objectives of this research were to determine if resistance to horn flies is a heritable trait in organic Holstein cattle, determine associations with yield traits, and to detect genomic regions associated with fly infestation. Observations of fly load were recorded from 1,667 pastured Holstein cows, of which 640 were genotyped, on 13 organic dairies across the United States. Fly load score was determined using a 0 to 4 scale based on fly coverage from chine to loin on one side of the body, with 0 indicating few to no flies and 4 indicating high infestation. The scoring system was validated by counting flies from photographs taken at the time of scoring from 252 cows. To mitigate the effect of our data structure on potential selection bias effects on genetic parameter estimates, survival to subsequent lactations of scored animals and herd-mates that had been culled before the trial was accounted for as the trait stayability. Genetic parameters were estimated using single-step genomic analysis with 3-trait mixed models that included fly score, stayability, and a third phenotype. Model effects differed by variable, but fixed effects generally included a contemporary group, scorer, parity, and stage of lactation; random effects included animal, permanent environment, and residual error. A genome-wide association study was performed by decomposing estimated breeding values into marker effects to detect significant genomic regions associated with fly score. The rank correlation between the subjective fly score and the objective count was 0.79. The average heritability of fly score (± standard error) estimated across multiple models was 0.25 ± 0.04 when a known Holstein maternal grandsire was required and 0.19 ± 0.03 when only a known Holstein sire was required. Genetic correlation estimates with yield traits were moderately positive, but a greater fly load was associated with reduced yield after accounting for genetic merit. Lower fly loads were associated with white coat coloration; a significant genomic region on Bos taurus autosome 6 was identified that contains the gene KIT, which was the most plausible candidate gene for fly resistance because of its role in coat pattern and coloration. Fourier transform infrared spectroscopy (FTIR) allows for the determination of milk acetone (mACE) and β-hydroxybutyrate (mBHB) concentrations, providing a potential herd monitoring tool for hyperketolactia, defined as elevated milk ketone bodies. The study aim was to characterize mACE and mBHB concentration dynamics during early lactation in Polish Holstein-Friesian cows. Milk samples (n = 3,867,390) were collected within 6 to 60 days in milk (DIM) over a 4-yr period (April 1, 2013 to March 31, 2017) from approximately 21,300 dairy herds (average 38.7 cows/herd). Fixed effects of parity, DIM, and their interaction on mACE and mBHB concentrations were determined using a mixed model with a herd-year-season fixed effect and random cow effect. Published hyperketolactic mACE (≥0.15 mmol/L) and mBHB (≥0.10 mmol/L) threshold concentrations were used to classify study milk samples into ketolactia groups of normal (mACE <0.15 mmol/L and mBHB <0.10 mmol/L) and hyperketolactic (HYKL; either mACE ≥0.15 mmol/L or mBHB ≥0.10 mmol/L). Additionally, HYKL samples were categorized into subpopulations as having elevated mBHB and mACE (HYKL ACEBHB , mACE ≥0.15 mmol/L and mBHB ≥0.10 mmol/L), only elevated mBHB (HYKL BHB ; mACE <0.15 mmol/L and mBHB ≥0.10 mmol/L), or only elevated mACE (HYKL ACE ; mACE ≥0.15 mmol/L and mBHB <0.10 mmol/L). Effects of parity, DIM, ketolactia group or subpopulation, and their interactions on mACE and mBHB concentrations were also determined using the mixed model that included ketolactia group or subpopulation as an independent variable. Across all data, mACE and mBHB concentrations were influenced by effects of parity, DIM, and their interaction as well as parity, DIM, ketolactia group or subpopulation, and their interactions. For all samples, mACE and mBHB concentrations decreased with increasing DIM, with mACE concentration declining more rapidly compared with mBHB. In the data set, 68% and 32% of all samples were defined as normal or HYKL, respectively. Among HYKL samples, mACE was elevated soon after calving and declined over time. In contrast, mBHB started lower after calving and increased reaching peak concentrations around 30 DIM, and then decreased. Within HYKL samples, 50.8, 41.3, and 7.9% were categorized as HYKL ACEBHB , HYKL BHB , and HYKL ACE respectively. Between 6 and 21 DIM, 11.3% of HYKL were classified as HYKL ACE . Primiparous cows had greater (14.8%) HYKL ACE samples in this time period. In conclusion, this study has characterized mACE and mBHB concentrations during early lactation and determined effects of parity, DIM, and their interaction. Using published criteria interpreting mACE and mBHB concentrations, it was intriguing to identify a unique population of samples having elevated mACE without mBHB in early lactation, especially in primiparous cows. Milk and dairy products provide highly sustainable concentrations of essential amino acids and other required nutrients for humans; however, amount of milk currently produced per dairy cow globally is inadequate to meet future needs. Higher performing dairy cows and herds produce more milk with less environmental impact per kg than lower performing cows and herds. In 2018, 15.4% of the world's dairy cows produced 45.4% of the world's dairy cow milk, reflecting the global contribution of high-performing cows and herds. In high-performing herds, genomic evaluations are utilized for multiple trait selection, welfare is monitored by remote sensing, rations are formulated at micronutrient levels, health care is focused on prevention and reproduction is managed with precision. Higher performing herds require more inputs and generate more waste products per cow, thus innovations in environmental management on such farms are essential for lowering environmental impacts. The objectives of this study were to estimate genetic parameters of calf health in organic US Holstein calves. Calves were born on farms across the United States from 2006 to 2019. Three calf health traits were evaluated in the study: calf respiratory disease until 365 d of age, calf scours until 60 d of age, and heifer stayability until 365 d of age. For respiratory disease and scours, animals were assigned a phenotype of 0 if they were healthy and a phenotype of 1 if they were diseased. For stayability, animals were assigned a phenotype of 0 if they were removed from the herd by 365 d of age and 1 if they remained in the herd at 365 d of age. Genetic parameters were estimated from threshold models that included the fixed effects of mean, year-season of birth, and dam age (respiratory disease and scours only) as well as the random effects of herd-year of birth and additive genetics. Heritability estimates were 0.100, 0.075, and 0.085 for respiratory disease, scours, and stayability, respectively. Solutions for estimated breeding values for respiratory disease and scours were transformed from disease risk to disease resistance by reversing the signs before calculating genetic correlations such that higher values of scours, respiratory disease, and stayability were favored. There was a moderate favorable genetic correlation estimate between respiratory disease resistance and stayability of 0.675. However, genetic correlation estimates between respiratory disease resistance and scours resistance (0.148) and between scours resistance and stayability (0.165) were low. Estimated breeding value correlations between calf health traits and other traits evaluated nationally were generally low in magnitude. The strongest correlation estimates were with longevity, particularly between stayability and heifer livability (0.217) and between stayability and cow livability (0.288); respiratory disease resistance was also favorably correlated with heifer (0.190) and cow (0.178) livability. Correlations with cow health traits were generally low and unfavorable. Linear models including the random effect of herd-by-sire indicated that herd-by-sire accounted for approximately 2% of phenotypic variance for scours and stayability, which may indicate a genotype by environment interaction effect for these traits. Milk fat production is highly influenced by nutrition and rumen fermentation. Rumination is an essential part of the ruminant digestive process and can serve as an indicator of rumen fermentation. The objective of this research was to quantify variation in rumination time between and within dairy herds and test for relationships between rumination time and milk fat production and fatty acid (FA) profile as a proxy of rumen fermentation. Our hypothesis was that rumination may indicate disruptions to rumen fermentation and that cows that spent less time ruminating would have lower milk fat due to these rumen disruptions. Data were collected from 1,733 Holstein cows on 5 commercial dairy farms (4 in Pennsylvania and 1 in New York) of 200 to 700 head using 1 of 2 commercially-available rumination sensing systems, CowManager SensOor ear tags (Agis Automatisering BV) or SCR model HR-LDn neck collars (SCR Engineers). Rumination data were collected for 7 consecutive days leading up to a DHIA test, summed within day, then averaged to obtain mean daily minutes of rumination time. Milk samples from the DHIA test were analyzed for fat content by mid-infrared spectroscopy and for milk FA profile by gas chromatography. Rumination data were analyzed using multiple linear regression models. Rumination time was related to concentration of specific odd- and branched-chain and trans FA in milk but was not directly related to milk fat concentration. Rumination time also did not contribute to models predicting milk fat concentration after accounting for other cow-level variables. There was a linear relationship between trans-10 C18:1 and rumination time that was positive after accounting for the effect of farm (partial R 2 of 2.97% across all data, 4.24% in SCR data, and 2.22% in CowManager data). Although rumination time was not related directly to milk fat, it was associated with differences in trans and odd- and branched-chain FA that have been demonstrated to change during subacute ruminal acidosis or biohydrogenation-induced milk fat depression, which may affect milk fat and other production variables.
