Abstract

A molecular tool was adapted for practical identification of the world's first transgenic event (Embrapa 5.1) in common bean (Phaseolus vulgarisL.). Based on the technology of RNA interference and on the transformation of plants via the Biobalistic method, a transgenic bean was created with effective resistance to Bean golden mosaic virus, its principal virus. To support the monitoring of this technology, the development of new cultivars, and the process of producing, benefiting, and distributing of seeds and grains, a molecular detection tool based on loop‐mediated isothermal amplification (LAMP) was developed. This tool can facilitating diagnosis, making it fast, specific, low‐cost, and easily executed. Based on the gene Ahas(GenBank M88686) of the Embrapa 5.1 event, sets of initiators were planned (forward outer primer–backward outer primer and forward inner primer–backward inner primer) that are characteristic of the LAMP method. These were evaluated for their sensitivity and specificity in detecting their target. The LAMP method allows direct visual interpretation. The Neutral Red (NR) pH indicator was adopted to facilitate molecular diagnosis in moderately well equipped environments, thus reducing the risk of contamination and the need for skilled labor. This agility in diagnosis occurs because several phases commonly used in molecular techniques, such as polymerase chain reaction, can be excluded. The LAMP method, combined with a rapid DNA extraction method (modified NaOH) and the addition of NR, provides technological support for addressing issues of biosafety, traceability, and identification of the Embrapa 5.1 event in beans in the field and in industry.

Abstract

This work evaluated the effect of genotype by environment interaction in Andean common bean lines with white grains, in Central Southern Brazil, to identify lines with high agronomic performance, stability and adaptability, aiming to meet domestic demand and to increase the Brazilian participation in the foreign market of common bean. Nineteen trials with twelve Andean lines were conducted in 2007, 2008 and 2009, in Central Southern Brazil. Grain yield and other agronomic traits were evaluated. Data were subjected to analysis of variance and of adaptability/stability using Annicchiarico and modified AMMI methods. Significant differences were found between lines for all traits evaluated. Genotype by environment interaction was important for lines with Andean origin and white seed. The utilization of weighted mean of absolute scores and yield with the AMMI results enabled the identification of the most stable and adapted lines. Lines Poroto Alubia, CNFB 16211, Ouro Branco and WAF 160 were stable and adapted, using both methods. CNFB 16211 line presented high agronomic performance, stability and adaptability and therefore this line may be a new cultivar. USWA 70 and WAF 75 lines presented grain size similar to that required by the foreign market and superior to the Brazilian cultivars, besides favorable agronomic traits, and thus these lines may be indicated as new cultivars. Os objetivos deste trabalho foram avaliar o efeito da interação genótipos x ambientes em linhagens de feijoeiro-comum de origem andina com grãos brancos na Região Centro-Sul do Brasil e identificar linhagens com alto potencial agronômico, estabilidade e adaptabilidade, visando atender a demanda interna e possibilitar a participação brasileira no mercado externo. Dezenove ensaios constituídos por 12 linhagens foram conduzidos em campo na Região Centro-Sul do Brasil nos anos 2007, 2008 e 2009. Avaliaram-se a produtividade de grãos e outros caracteres de importância agronômica. Os dados foram submetidos a análises de variância e de adaptabilidade/estabilidade pelos métodos de Annicchiarico e AMMI com modificações. Houve diferenças entre as linhagens para todos os caracteres avaliados. A interação genótipos x ambientes foi importante na avaliação de linhagens de origem andina com grãos brancos. A utilização da média ponderada pelos escores absolutos e da produtividade com os resultados da análise AMMI facilitou a identificação de linhagens mais estáveis e adaptadas. As linhagens Poroto Alubia, CNFB 16211, Ouro Branco e WAF 160 são estáveis e adaptadas utilizando-se as duas metodologias. A linhagem CNFB 16211 apresenta alto potencial agronômico, alta estabilidade e adaptabilidade sendo, portanto, possível a sua indicação como nova cultivar. As linhagens USWA 70 e WAF 75 apresentam tamanho de grão semelhante ao exigido pelo mercado externo e superior ao das cultivares brasileiras, além de apresentarem caracteres agronômicos favoráveis, sendo portanto possível a sua indicação como novas cultivares.

BRS Esteio is a common bean cultivar with black grains of excellent cooking quality, suited for cultivation in 21 Brazilian States. In 77 trials of Value for Cultivation and Use, the cultivar had a normal cycle (90 days), an average yield of 2,529 kg ha-1, which is 8.1% higher than that of the control cultivars, a high yield potential (4,702 kg ha-1), upright growth, tolerance to lodging, and moderate resistance to anthracnose.

O objetivo deste trabalho foi realizar a estratificação ambiental nos Estado de Goiás e no Distrito Federal para a avaliação da produtividade de grãos de genótipos de feijoeiro-comum tipo Carioca, e comparar a eficiência de diferentes métodos de estratificação ambiental. Foram realizados 25 ensaios em blocos ao acaso, com três repetições, nas épocas de semeadura das águas e do inverno, nos anos de 2003 e 2004. Os ensaios foram compostos por 16 genótipos de grãos tipo Carioca. A estratificação ambiental foi realizada por meio de quatro metodologias: tradicional de Lin; percentagem simples da interação de genótipos com os locais dois a dois, de Cruz e Castoldi; correlação de Pearson para os pares de locais; e método da ecovalência de Wricke. Os locais identificados como similares em pelo menos três metodologias foram considerados pouco informativos, e eles foram os seguintes municípios do Estado de Goias: Anápolis, Santo Antônio de Goiás, Goiatuba e Urutaí, nas safras das águas/2003 e águas/2004, inverno/2003 e inverno/2004, respectivamente. Nenhum dos locais foi identificado como pouco informativo em pelo menos duas safras e, portanto, nenhum deles foi eliminado da rede de avaliação de genótipos de feijoeiro-comum tipo Carioca. Os locais utilizados em todas as safras (Rio Verde, GO, e Planaltina, DF) não foram identificados como pouco informativos em nenhuma delas e, portanto, são indicados para a avaliação de genótipos de feijoeiro-comum. Os métodos de Lin e da ecovalência foram os mais indicados para identificação de locais pouco informativos The objective of this study was to perform the environmental stratification in the states of Goiás and Distrito Federal for grain yield evaluation of Carioca common bean genotypes, and to compare the efficiency of different methods of environmental stratification. Twenty-five trials were carried out in a randomized block design with three replications, both in the rainy and in the winter sowing seasons, in 2003 and 2004. The trials were composed of 16 genotypes with Carioca grain type. The environmental stratification was performed through four methodologies: traditional Lin; Cruz & Castoldi's estimate of the simple percentage of the genotypes by location interactions; estimate of Pearson's correlation for pairs of locals; and the Wricke's ecovalence method. Locations identified as similar by at least three of the methodologies were considered poor-informative, and they were the following Goiás state municipalities: Anápolis, Santo Antônio de Goiás, Goiatuba and Urutaí, in the rainy/2003, rainy/2004, winter/2003 and winter/2004 growing seasons, respectively. None of the locations was identified as poor-informative in at least two seasons and, therefore, none of them was eliminated from the evaluation network of Carioca common bean genotypes. The locations used in all growing seasons (Rio Verde, GO, and Planaltina, DF) were not identified as poor-informative in none of them and, therefore, they were indicated for evaluation of Carioca common bean genotypes. Abstract

Common bean (Phaseolus vulgaris L.) is the most important legume crop for direct human consumption in Brazil. Overall patterns of genetic diversity of common bean in the country are not well understood. Hence, this study sought to integrate morphological and agronomic evaluations with prior molecular diversity data from a geo-referenced collection of Brazilian common bean. Our sample included 279 randomly chosen landrace accessions from the main growing regions of beans in Brazil. Eighteen morphological and three agronomic characters (yield and susceptibility to common bacterial blight and rust diseases) were evaluated in field conditions in Brazil or California. Results show that the sample was diverse morphologically but did not include all the morphological variability described in domesticated common bean outside Brazil. Several accessions showed clear morphological differences in spite of having the same name. The integration of the current morphological and agronomic evaluations with a prior 74-marker diversity assessment of the same landraces improved the identification of five subpopulations identified on the basis of microsatellite diversity. These subpopulations could be distinguished by their morphology, the type and frequency of the market classes they included, and the frequency of susceptibility to diseases. Abstract

The search for more productive varieties of beans, with lower production cost and adaptable to most technological conditions has been intensified. The objective of this study was, therefore, to evaluate the stability and adaptability of common bean lines of the carioca group in the final evaluation network in the Central Cerrado of Brazil, in 2005 and 2006. Twelve lines and four cultivars were evaluated in 23 environments. A completely randomized block design was used with three replications, with plots of four rows of four meters. Grain yield data were subjected to analysis of variance, stability and of adaptability by the methodologies of Lin and Binns, Annichiarico and visual assessment, respectively. Abstract

The purpose of this study was the environmental stratification of the states of Paraná and Santa Catarina for the evaluation of carioca common bean genotypes. The grain yield was evaluated in 20 trials, in a randomized block design with three replications testing 16 genotypes with carioca grain. The environments were stratified by five methodologies: traditional Lin; mean square of the interaction for paired locations, estimate of the percentage of the complex interaction; estimate of Pearson's correlation, and the Wricke method. Locations identified as uninformative by at least three of the methodologies were excluded. The locations considered uninformative in the respective growing seasons were Roncador (rainy/2003 and dry/2003) and Laranjeiras do Sul (rainy/2004) and no location in the growing season dry/2004. Only Roncador was identified as not very informative in at least two seasons and was therefore eliminated from the evaluation network of Carioca common bean genotypes. [ABSTRACT FROM AUTHOR]

Abstract

The mapping population consisted of 94 F2 generation plants derived from a cross between the CNFC 7812 and CNFC 8056 lines, with different protein contents, 24% and 19% respectively. Seven hundred and fifty-two molecular markers were tested among the parents and four individuals from the segregant population. A total of 101 loci were used to develop the genetic map. The polymorphism rate was 8.3% and 23.2% for the microsatellite and RAPD markers, respectively. The sizes of the linkage groups ranged from 6.7 to 139.0 cM , presenting a mean of 49.4 ± 36.8. The map length was 840.7 cM and the mean group length was 45.9 cM. The average distance between the framework loci was 16.1 cM. This map was compared with international reference bean maps and results were discussed.
