3. LET-99-dependent spatial restriction of active force generators makes spindle's position robust

Abstract

During the asymmetric division of the Caenorhabditis elegans nematode zygote, the polarity cues distribution and daughter cell fates depend on the correct positioning of the mitotic spindle, which results from both centering and cortical pulling forces. Revealed by anaphase spindle rocking, these pulling forces are regulated by the force generator dynamics, which are in turn consequent of mitotic progression. We found a novel, additional, regulation of these forces by the spindle position. It controls astral microtubule availability at the cortex, on which the active force generators can pull. Importantly, this positional control relies on the polarity dependent LET-99 cortical band, which restricts or concentrates generators to a posterior crescent. We ascribed this control to the microtubule dynamics at the cortex. Indeed, in mapping the cortical contacts, we found a correlation between the centrosome-cortex distance and the microtubule contact density. In turn, it modulates pulling force generator activity. We modelled this control, predicting and experimentally validating that the posterior crescent extent controlled where the anaphase oscillations started, in addition to mitotic progression. Finally, we propose that spatially restricting force generator to a posterior crescent sets the spindle's final position, reflecting polarity through the LET-99 dependent restriction of force generators to a posterior crescent. This regulation superimposes that of force generator processivity. This novel control confers a low dependence on microtubule and active force generator exact numbers or dynamics, provided that they exceed the threshold needed for posterior displacement. Interestingly, this robustness originates in cell mechanics rather than biochemical networks.

Abstract

Finite topological quantum systems can undergo continuous metastable quantum phase transitions to change their topological nature. Here we show how to nucleate the transition between ring currents and dark soliton states in a toroidally trapped Bose-Einstein condensate. An adiabatic passage to wind and unwind its phase is achieved by explicit global breaking of the rotational symmetry. 14. Written Spelling to Dictation: Sound-To-Spelling Regularity Affects Both Writing Latencies and Durations

Abstract

The authors examined the effect of sound-to-spelling regularity on written spelling latencies and writing durations in a dictation task in which participants had to write each target word 3 times in succession. The authors found that irregular words (i.e., those containing low-probability phoneme-to-grapheme mappings) were slower both to initially produce and to execute in writing than were regular words. The regularity effect was found both when participants could and could not see their writing (Experiments 1 and 2) and was larger for low- than for high-frequency words (Experiment 3). These results suggest that central processing of the conflict generated by lexically specific and assembled spelling information for irregular words is not entirely resolved when the more peripheral processes controlling handwriting begin.

In order to investigate the persistence of the N=50 gap for the Nickel isotopes, an experiment has been performed at RIBF (RIKEN, Japan) to produce exotic nuclei around ⁷⁸Ni by in-flight fission of a ²³⁸U beam. The BigRIPS and ZeroDegree spectrometers were used to purify and select the nuclei of interest. Their decay was detected with the EURICA detector for γ-ray detection, coupled with the charged particle WAS3aBi detector. The analysis of the correlations between the ion implantation and their decay, as well as the γ decay spectra in simple and double coincidence allowed us to propose a level scheme of the excited states for the ⁷⁹Zn, and we assigned the spin and parity for most of the states. We also constrained the spin value for the fundamental state of the ⁸⁰Cu, between 3hbar and 5hbar. Comparison with a core-particle coupling model allowed us to interpret the level scheme and propose hypothesis about the nature of the observed states. Abstract

In Caenorhabditis elegans, two lateral blast cells called P(11/12)L and P(11/12)R are symmetric left--right homologs at hatching, migrate subsequently in opposite anteroposterior directions during the first larval stage, and adopt two different fates, thus breaking the symmetry between them. Our results show that, unlike most other cell fate decisions in C. elegans, the orientation of P(11/12)L/R migration is highly biased, but not fixed. The handedness of their migration is linked to whole body handedness and is randomized in lin-12/Notch mutants and by ablation of the Y cell. Migration handedness is independent of P11 and P12 fate determination, previously shown to require the LIN-44/Wnt and the LIN-3/EGF pathways (L. I. Jiang and P. W. Sternberg, 1998, Development 125, 2337-2347). We further show that several changes in P(11/12)L/R asymmetry have occurred during nematode evolution: loss of asymmetry or reversals in orientation of migration. Strikingly, for most species studied, handedness of migration is highly biased but not fixed. Thus, whereas the final cell fate pattern of P11/12 is invariant, the developmental route leading to it is subject both to developmental indeterminacy and to evolutionary variations. Abstract

Laser ablation of centrosomes in one-cell Caenorhabditis elegansembryos shows that chromatids can segregate independently of centrosomes and also independently of the activity of kinetochore microtubules during mitosis. CLASP and RanGTP are required to generate this centrosome-independent force, whereas SPD-1 and BMK-1 act as brakes to oppose it.
