1. A novel endocast technique providing a 3D quantitative analysis of the gastrovascular system in Rhizostoma pulmo: An unexpected through-gut in cnidaria

Abstract: The investigation of jellyfish gastrovascular systems mainly focused on stain injections and dissections, negatively affected by thickness and opacity of the mesoglea. Therefore, descriptions are incomplete and data about tridimensional structures are scarce. In this work, morphological and functional anatomy of the gastrovascular system of Rhizostoma pulmo (Macri 1778) was investigated in detail with innovative techniques: resin endocasts and 3D X-ray computed microtomography. The gastrovascular system consists of a series of branching canals ending with numerous openings within the frilled margins of the oral arms. Canals presented a peculiar double hemi-canal structure with a medial adhesion area which separates centrifugal and centripetal flows. The inward flow involves only the “mouth” openings on the internal wing of the oral arm and relative hemi-canals, while the outward flow involves only the two outermost wings’ hemi-canals and relative “anal” openings on the external oral arm. The openings differentiation recalls the functional characteristics of a through-gut apparatus. We cannot define the gastrovascular system in Rhizostoma pulmo as a traditional through-gut, rather an example of adaptive convergence, that partially invalidates the paradigm of a single oral opening with both the uptake and excrete function.

Abstract: The mammalian 20S catalytic core of the proteasome is made of 14 different subunits (α1-7 and β1-7) but exists as different subtypes depending on the cell type. In immune cells, for instance, constitutive catalytic proteasome subunits can be replaced by the so-called immuno-catalytic subunits, giving rise to the immunoproteasome. Proteasome activity is also altered by post-translational modifications (PTMs) and by genetic variants. Immunochemical methods are commonly used to investigate these PTMs whereby protein-tagging is necessary to monitor their effect on 20S assembly. Here, we present a new miniaturized workflow combining top-down and bottom-up mass spectrometry of immunopurified 20S proteasomes that analyze the proteasome assembly status as well as the full proteoform footprint, revealing PTMs, mutations, single nucleotide polymorphisms (SNPs) and induction of immune-subunits in different biological samples, including organoids, biopsies and B-lymphoblastoid cell lines derived from patients with proteasome-associated autoinflammatory syndromes (PRAAS). We emphasize the benefits of using top-down mass spectrometry in preserving the endogenous conformation of protein modifications, while enabling a rapid turnaround (1 h run) and ensuring high sensitivity (1–2 pmol) and demonstrate its capacity to semi-quantify constitutive and immune proteasome subunits.

4. Cytolethal Distending Toxin Promotes Replicative Stress Leading to Genetic Instability Transmitted to Daughter Cells

Abstract: The cytolethal distending toxin (CDT) is produced by several Gram-negative pathogenic bacteria. In addition to inflammation, experimental evidences are in favor of a protumoral role of CDT-harboring bacteria such as Escherichia coli, Campylobacter jejuni, or Helicobacter hepaticus. CDT may contribute to cell transformation in vitro and carcinogenesis in mice models, through the genotoxic action of CdtB catalytic subunit. Here, we investigate the mechanism of action by which CDT leads to genetic instability in human cell lines and colorectal organoids from healthy patients’ biopsies. We demonstrate that CDT holotoxin induces a replicative stress dependent on CdtB. The slowing down of DNA replication occurs mainly in late S phase, resulting in the expression of fragile sites and important chromosomic aberrations. These DNA abnormalities induced after CDT treatment are responsible for anaphase bridge formation in mitosis and interphase DNA bridge between daughter cells in G1 phase. Moreover, CDT-genotoxic potential preferentially affects human cycling cells compared to quiescent cells. Finally, the toxin induces nuclear distension associated to DNA damage in proliferating cells of human colorectal organoids, resulting in decreased growth. Our findings thus identify CDT as a bacterial virulence factor targeting proliferating cells, such as human colorectal progenitors or stem cells, inducing replicative stress and genetic instability transmitted to daughter cells that may therefore contribute to carcinogenesis. As some CDT-carrying bacterial strains were detected in patients with colorectal cancer, targeting these bacteria could be a promising therapeutic strategy.

5. Characterization of Human Colon Organoids From Inflammatory Bowel Disease Patients

Abstract: Inflammatory Bowel Diseases (IBD) are chronic inflammatory disorders, where epithelial defects drive, at least in part, some of the pathology. We reconstituted human intestinal epithelial organ, by using three-dimension culture of human colon organoids. Our aim was to characterize morphological and functional phenotypes of control (non-IBD) organoids, compared to inflamed organoids from IBD patients. The results generated describe the epithelial defects associated with IBD in primary organoid cultures, and evaluate the use of this model for pharmacological testing of anti-inflammatory approaches. Human colonic tissues were obtained from either surgical resections or biopsies, all harvested in non-inflammatory zones. Crypts were isolated from controls (non-IBD) and IBD patients and were cultured up to 12-days. Morphological (size, budding formation, polarization, luminal content), cell composition (proliferation, differentiation, immaturity markers expression), and functional (chemokine and tight junction protein expression) parameters were measured by immunohistochemistry, RT-qPCR or western-blot. The effects of inflammatory cocktail or anti-inflammatory treatments were studied in controls and IBD organoid cultures respectively. Organoid cultures from controls or IBD patients had the same cell composition after 10 to 12-days of culture, but IBD organoid cultures showed an inflammatory phenotype with decreased size and budding capacity, increased cell death, luminal debris, and inverted polarization. Tight junction proteins were also significantly decreased in IBD organoid cultures. Inflammatory cytokine cocktail reproduced this inflammatory phenotype in non-IBD organoids. Clinically used treatments (5-ASA, glucocorticoids, anti-TNF) reduced some, but not all parameters. Inflammatory phenotype is associated with IBD epithelium, and can be studied in organoid cultures. This model constitutes a reliable human pre-clinical model to investigate new strategies targeting epithelial repair.

6. Improvement of Microstomia in Scleroderma after Carbon Dioxide Laser Treatment

Abstract: Limited mouth opening (LMO) is a frequent complication of systemic sclerosis (SS). Its management is complex and there are limited treatment options. We report four patients with SS and severe LMO [interincisal distance (IID) 2) laser. Pulsed CO2 laser treatment of the white lips was performed after all patients had signed a written informed consent in the absence of alternative treatment. Treatment was carried out under locoregional anaesthesia using a Sharplan 30C CO2 laser in the Silk Touch® resurfacing mode. One to three laser sessions were performed at intervals of 8-12 months between sessions. Assessments were performed at 3 and 12 months with measurement of the IID using a ruler, calculation of the Mouth Handicap in Systemic Sclerosis (MHISS) scale and global evaluation by the patients. Adverse events were also reported. In all four patients, an improvement in IID occurred 3 months after the first session with a mean gain of +5 mm (range: 2-7). At 12 months, a mean gain of +8.5 mm (range: 7-10) in IID was observed. The MHISS score decreased by a mean of -14 (range: 11-17). All patients showed improvement of lip flexibility or mouth opening, allowing better phonation and mastication and easier dental care. Adverse effects were transient erythema and/or dyschromia. CO2 laser appears to be effective and well tolerated in the improvement of LMO in SS.

7. Oyster Farming, Temperature, and Plankton Influence the Dynamics of Pathogenic Vibrios in the Thau Lagoon

Abstract: Vibrio species have been associated with recurrent mass mortalities of juvenile oysters Crassostrea gigas threatening oyster farming worldwide. However, knowledge of the ecology of pathogens in affected oyster farming areas remains scarce. Specifically, there are no data regarding (i) the environmental reservoirs of Vibrio populations pathogenic to oysters, (ii) the environmental factors favoring their transmission, and (iii) the influence of oyster farming on the persistence of those pathogens. This knowledge gap limits our capacity to predict and mitigate disease occurrence. To address these issues, we monitored Vibrio species potentially pathogenic to C. gigas in 2013 and 2014 in the Thau Lagoon, a major oyster farming region in the coastal French Mediterranean. Sampling stations were chosen inside and outside oyster farms. Abundance and composition of phyto-, microzoo-, and mesozooplankton communities were measured monthly. The spatial and temporal dynamics of plankton and Vibrio species were compared, and positive correlations between plankton species and vibrios were verified by qPCR on isolated specimens of plankton. Vibrio crassostreae was present in the water column over both years, whereas Vibrio tasmaniensis was mostly found in 2013 and Vibrio aestuarianus was never detected. Moreover, V. tasmaniensis and V. crassostreae were found both as free-living or plankton-attached vibrios 1 month after spring mortalities of the oyster juveniles. Overall, V. crassostreae was associated with temperature and plankton composition, whereas V. tasmaniensis correlated with plankton composition only. The abundance of Vibrio species in the water column was similar inside and outside oyster farms, suggesting important spatial dispersion of pathogens in surrounding areas. Remarkably, a major increase in V. tasmaniensis and V. crassostreae was measured in the sediment of oyster farms during cold months. Thus, a winter reservoir of pathogenic vibrios could contribute to their ecology in this Mediterranean shellfish farming ecosystem.

Abstract: Bevacizumab, an anti-VEGF monoclonal antibody, has recently emerged as a new option for severe forms of hereditary hemorrhagic telangiectasia (HHT). Its utilization in this orphan disease has rapidly spread despite the lack of randomized trials and international guidelines. The objective of this study is to report the main clinical data (baseline characteristics, dose schedule, efficacy, adverse events and deaths) of HHT patients treated by intravenous bevacizumab in France.Retrospective observational study of HHT patients treated with bevacizumab for a severe form of the disease in the 14 centers of the French HHT network.Forty-six patients (median age: 68 years) were treated between March 2009 and May 2015. Ten patients were treated for high output cardiac failure, 20 patients for severe hemorrhages and 16 for both indications. The standard protocol (6 infusions of 5mg/kg every 2 weeks) was initially used in 89% of the cases but diverse strategies were subsequently applied. A clinical improvement was noted by the referent physician for 74% of the patients with a median effect's duration of 6 months. Wound healing complications led to 2 amputations. Arthralgia/arthritis and arterial hypertension occurred in 5 patients each. One third of the patients were dead at the time of the final update, coherently with age and the poor prognosis of these highly symptomatic patients.Intravenous bevacizumab seems to provide a clinical benefice in severe HHT patients. Precautions concerning wound healing and vascular pathologies must be respected. Prospective double blinded versus placebo trials are needed.

9. Where to Forage in the Absence of Sea Ice? Bathymetry As a Key Factor for an Arctic Seabird.

Abstract: The earth is warming at an alarming rate, especially in the Arctic, where a marked decline in sea ice cover may have far-ranging consequences for endemic species. Little auks, endemic Arctic seabirds, are key bioindicators as they forage in the marginal ice zone and feed preferentially on lipid-rich Arctic copepods and ice-associated amphipods sensitive to the consequences of global warming. We tested how little auks cope with an ice-free foraging environment during the breeding season. To this end, we took advantage of natural variation in sea ice concentration along the east coast of Greenland. We compared foraging and diving behaviour, chick diet and growth and adult body condition between two years, in the presence versus nearby absence of sea ice in the vicinity of their breeding site. Moreover, we sampled zooplankton at sea when sea ice was absent to evaluate prey location and little auk dietary preferences. Little auks foraged in the same areas both years, irrespective of sea ice presence/concentration, and targeted the shelf break and the continental shelf. We confirmed that breeding little auks showed a clear preference for larger copepod species to feed their chick, but caught smaller copepods and nearly no ice-associated amphipod when sea ice was absent. Nevertheless, these dietary changes had no impact on chick growth and adult body condition. Our findings demonstrate the importance of bathymetry for profitable little auk foraging, whatever the sea-ice conditions. Our investigations, along with recent studies, also confirm more flexibility than previously predicted for this key species in a warming Arctic.

Abstract: Hepatitis C virus (HCV) infection is associated with lymphoproliferative disorders and B-cell non-Hodgkin lymphomas (B-NHLs). Evaluation of the efficacy and safety profiles of different antiviral therapies in HCV patients with B-NHL is warranted.First, we evaluated the sustained virologic response (SVR) and safety of Peg-interferon-alpha (Peg-IFN) + ribavirin +/- first protease inhibitors (PI1s) therapy in 61 HCV patients with B-NHL enrolled in a nationwide observational survey compared with 94 matched HCV-infected controls without B-NHL. In a second series, interferon-free regimens using a newly optimal combination therapy with direct-acting antiviral drugs (DAAs) were evaluated in 10 patients with HCV and B-NHL.The main lymphoma type was diffuse large B-cell lymphoma (38%) followed by marginal zone lymphoma (31%). 11. Sporadic early-onset colorectal cancer is a specific sub-type of cancer: a morphological, molecular and genetics study.

Abstract: Sporadic early onset colorectal carcinoma (EOCRC) which has by definition no identified hereditary predisposition is a growing problem that remains poorly understood. Molecular analysis could improve identification of distinct sub-types of colorectal cancers (CRC) with therapeutic implications and thus can help establish that sporadic EOCRC is a distinct entity. From 954 patients resected for CRC at our institution, 98 patients were selected. Patients aged 45-60 years were excluded to help define "young" and "old" groups. Thirty-nine cases of sporadic EOCRC (patients ≤ 45 years with microsatellite stable tumors) were compared to both microsatellite stable tumors from older patients (36 cases, patients>60 years) and to groups of patients with microsatellite instability. Each group was tested for TP53, KRAS, BRAF, PIK3CA mutations and the presence of a methylator phenotype. Gene expression profiles were also used for pathway analysis. Compared to microsatellite stable CRC from old patients, sporadic EOCRC were characterized by distal location, frequent synchronous metastases and infrequent synchronous adenomas but did not have specific morphological characteristics. A familial history of CRC was more common in sporadic EOCRC patients despite a lack of identified hereditary conditions (p = 0.013). Genetic studies also showed the absence of BRAF mutations (p = 0.022) and the methylator phenotype (p = 0.005) in sporadic EOCRC compared to older patients. Gene expression analysis implicated key pathways such as Wnt/beta catenin, MAP Kinase, growth factor signaling (EGFR, HGF, PDGF) and the TNFR1 pathway in sporadic EOCRC. Wnt/beta catenin signaling activation was confirmed by aberrant nuclear beta catenin immunostaining (p = 0.01). This study strongly suggests that sporadic EOCRC is a distinct clinico-molecular entity presenting as a distal and aggressive disease associated with chromosome instability. Furthermore, several signaling pathways including the TNFR1 pathway have been identified as potential biomarkers for both the diagnosis and treatment of this disease.

12. Impairment of visual function and retinal ER stress activation in Wfs1-deficient mice.

Abstract: Wolfram syndrome is an early onset genetic disease (1/180,000) featuring diabetes mellitus and optic neuropathy, associated to mutations in the WFS1 gene. Wfs1-/- mouse model shows pancreatic beta cell atrophy, but its visual performance has not been investigated, prompting us to study its visual function and histopathology of the retina and optic nerve. Electroretinogram and visual evoked potentials (VEPs) were performed in Wfs1-/- and Wfs1+/+ mice at 3, 6, 9 and 12 months of age. Fundi were pictured with Micron III apparatus. Retinal ganglion cell (RGC) abundance was determined from Brn3a immunolabeling of retinal sections. RGC axonal loss was quantified by electron microscopy in transversal optic nerve sections. Endoplasmic reticulum stress was assessed using immunoglobulin binding protein (BiP), protein disulfide isomerase (PDI) and inositol-requiring enzyme 1 alpha (Ire1α) markers. Electroretinograms amplitudes were slightly reduced and latencies increased with time in Wfs1-/- mice. Similarly, VEPs showed decreased N+P amplitudes and increased N-wave latency. Analysis of unfolded protein response signaling revealed an activation of endoplasmic reticulum stress in Wfs1-/- mutant mouse retinas. Altogether, progressive VEPs alterations with minimal neuronal cell loss suggest functional alteration of the action potential in the Wfs1-/- optic pathways.

Abstract: Immunohistochemistry (IHC) and/or MSI-PCR (microsatellite instability-polymerase chain reaction) tests are performed routinely to detect mismatch repair deficiency (MMR-D). Classical MMR-D tumors present a loss of MLH1/PMS2 or MSH2/MSH6 with MSI-High. Other profiles of MMR-D tumors have been described but have been rarely studied. In this study, we established a classification of unusual MMR-D tumors and determined their frequency and clinical impact. All MMR-D tumors identified between 2007 and 2017 were selected. Any profile besides the classical MMR-D phenotype was defined as unusual. For patients with unusual MMR-D tumors, IHC, and PCR data were reviewed, the tumor mutation burden (TMB) was evaluated and clinical and genetic features were collected. Of the 4948 cases of MMR testing, 3800 had both the available IHC and MSI-PCR results and 585 of these had MMR-D. After reviewing the IHC and PCR, 21% of the cases initially identified as unusual MMR-D were reclassified, which resulted in a final identification of 89 unusual MMR-D tumors (15%). Unusual MMR-D tumors were more often associated with non-CRC than classical MMR-D tumors. Unusual MMR-D tumors were classified into four sub-groups: i) isolated loss of PMS2 or MSH6, ii) classical loss of MLH1/PMS2 or MSH2/MSH6 without MSI, iii) four MMR proteins retained with MSI and, iv) complex loss of MMR proteins, with clinical characteristics for each sub-group. 14. Ontogenetic dietary shifts of the medusa Rhizostoma pulmo (Cnidaria: Scyphozoa)

Abstract: Identifying ontogenetic changes in jellyfish diet is fundamental to understand trophic interactions during their life cycle. Scyphomedusae blooms exert major predation pressure on plankton communities, although their role in ecosystems has long been misrepresented. This study assesses seasonal and ontogenetic changes in the diet of the scyphomedusa Rhizostoma pulmo, one of the largest yet overlooked Mediterranean jellyfish. Medusae gut contents (n = 127) were collected during one year in Bages Sigean lagoon, southern France. Results show that the diet composition differs from the availability of prey in the environment with contrasting preferences along ontogeny. Calanoid (70%) and harpacticoid (45.8%) copepods were the most frequent prey and the major carbon contributors for small medusae (bell diameter 15 cm), which obtain most of their carbon intake from ciliates and fish eggs (20.9%). The overall impact on micro and mesozooplankton showed that small medusae consume 5% of the copepods daily standing stock, while large medusae consumed 8% of ciliates daily standing stock. Our results stress that R. pulmo display different trophic pathways along its life cycle, firstly interacting with the classical food web, and shifting afterwards to a greater interaction with the microbial loop.

Abstract: BACKGROUND Whipple's disease is a very rare disease needing a long-term treatment. The most frequent symptoms are recurrent arthralgia or arthritis, chronic diarrhea, abdominal pain, and weight loss. OBJECTIVES In this article, we have highlighted the main clinical features and diagnostic procedures that lead to the diagnosis and comment on the clinical response, treatment, and the factors of relapse. METHODS Subjects were recruited from the Internal Medicine and Rheumatologic Departments of an University Hospital from November 1997 to January 2016. Overall, 12 subjects were finally diagnosed. RESULTS Mean age was 54.3 years (age range: 30-81), with more male patients (58.3%). Almost all patients had articular symptoms and impaired general condition (91.7%); and a majority had digestive symptoms (75%). Regardless of the symptoms, the most efficient diagnostic tools were the PCR screening on the gastrointestinal biopsies and saliva (83.3 and 72.7% positive results, respectively). More than half of the patients relapsed (55.6%). The relapsing patients were older [63.2 (44-81)] and mostly male with a majority (60%) of digestive symptoms and a delayed diagnosis. CONCLUSIONS In current practice, it is highly difficult to diagnose Whipple's disease. In order to decrease the delay between the first symptoms and the diagnosis, effective tools such as saliva and stools PCR should be used because higher delays of diagnosis lead to a higher number of relapses.

Abstract: International audience; Estrogens, such as the 17β-estradiol (E2) and the 17α-ethinylestradiol (EE2), have been regarded as a global threat to aquatic ecosystems due to their pseudo-persistence, their high estrogenic activity and their toxicity towards non-target species. Data regarding their ecotoxicological effects on marine calanoid copepods are very scarce. In this study, the calanoid copepod Acartia clausi was used as a model organism for estrogens exposure in marine pelagic ecosystems. Lethal effects of estrogens on A. clausi life-stages (Embryos, one day old nauplii: N1, three day old nauplii: N3, copepodites: C1-C3 and adults: C6) were investigated using 48 h acute tests. Copepods showed stage-specific responses against E2 and EE2 acute exposure. The most resistant life stage was N1 with LC50 values > 1500 μg L-1 and >5000 μg L-1, respectively for E2 and EE2. For N3, C1-C3, and C6, sensitivity to estrogens decreased with age and survival was affected at concentrations above those detected in the environment reflecting low estrogens acute toxicity for these life stages. In contrast, embryonic stage revealed high vulnerability to E2 and EE2 acute effects. Embryos showed non-monotonic dose-response and hatching success was significantly reduced at low realistic concentrations of E2 (0.005, 0.5, and 5 μg L-1) and EE2 (0.05 and 5 μg L-1).Survival, development and sex ratio of A. clausi to EE2 exposure at 1 and 100 μg L-1 were also determined during a life cycle experiment. Fitness of the females of the generation F0 was evaluated by measuring lifespan, prosome length and egg production. The main observed effects were the decrease of females’ prosome length, the feminization of the population and the reduction of the egg production for the generation F0 at 100 μg L-1 of EE2. This concentration is above those reported in the environment indicating the tolerance of A. clausi to EE2 at environmentally relevant concentrations.

Abstract: BACKGROUND AND AIMS: There is a need to evaluate the benefit-risk ratio of current therapies in inflammatory bowel disease (IBD) patients to provide the best quality of care. The primary objective of I-CARE (IBD Cancer and serious infections in Europe) was to assess prospectively safety concerns in IBD, with specific focus on the risk of cancer/lymphoma and serious infections in patients treated with anti-tumor necrosis factor and other biologic monotherapy as well as in combination with immunomodulators.METHODS: I-CARE was designed as a European prospective longitudinal observational multicenter cohort study to include patients with a diagnosis of Crohn's disease, ulcerative colitis, or IBD unclassified established at least 3 months prior to enrollment.RESULTS: A total of 10,206 patients were enrolled between March 2016 and April 2019, including 6169 (60.4%) patients with Crohn's disease, 3853 (37.8%) with ulcerative colitis, and 184 (1.8%) with a diagnosis of IBD unclassified. Thirty-two percent of patients were receiving azathioprine/thiopurines, 4.6% 6-mercaptopurine, and 3.2% methotrexate at study entry. At inclusion, 47.3% of patients were treated with an anti-tumor necrosis factor agent, 8.8% with vedolizumab, and 3.4% with ustekinumab. Roughly one-quarter of patients (26.8%) underwent prior IBD-related surgery. Sixty-six percent of patients had been previously treated with systemic steroids. Three percent of patients had a medical history of cancer prior to inclusion and 1.1% had a history of colonic, esophageal, or uterine cervix high-grade dysplasia.CONCLUSIONS: I-CARE is an ongoing investigator-initiated observational European prospective cohort study that will provide unique information on the long-term benefits and risks of biological therapies in IBD patients. 19. High yield of surveillance in patients diagnosed with constitutional mismatch repair deficiency

Abstract: BackgroundConstitutional mismatch repair deficiency (CMMRD) is a rare autosomal recessively inherited syndrome that is caused by biallelic pathogenic variants of the mismatch repair genes. It is characterised by the development of multiple tumours in the first and second decade of life including brain, gastrointestinal and haematological tumours often resulting in early death. In order to improve the prognosis of these patients, the European collaborative group ‘care for CMMRD’ developed a surveillance programme in 2014 and established a registry of patients with CMMRD in Paris. The aim of the study was to evaluate the outcome of this programme.MethodsTwenty-two patients with a definitive diagnosis of CMMRD and with at least one follow-up study were selected from the registry. Medical data on the outcome of surveillance were collected from these patients.ResultsDuring a mean follow-up of 4 years, the programme detected eight malignant tumours including three brain tumours, three upper gastrointestinal cancers and two colorectal cancers. Most tumours could successfully be treated. In addition, many adenomas were detected in the duodenum, and colorectum and subsequently removed. Seven patients developed a symptomatic malignancy, including two brain tumours, one small bowel cancer and four haematological malignancies. At the end of the follow-up, 16 out of 22 patients (73%) who participated in the surveillance programme were still alive.ConclusionThe study suggests a beneficial effect of surveillance of the digestive tract and brains.

Abstract: This study aims at quantifying and characterising microplastics (MP) distribution in the water column of the NW Mediterranean Sea as well as MP ingestion by the 2 main planktivorous fish of the area, sardine and anchovy. Debris of similar sizes were found in all water column samples and in all but 2 fish guts (out of 169). MP were found in 93% of water column samples with an average concentration of 0.23 ± 0.20 MP·m−3, but in only 12% of sardines (0.20 ± 0.69 MP·ind−1) and 11% of anchovies (0.11 ± 0.31 MP·ind−1). Fibres were the only shape of MP encountered and polyethylene terephthalate was the main polymer identified in water columns (61%), sardines (71%) and anchovies (89%). This study confirms the ubiquity of MP in the Mediterranean Sea and imparts low occurrence in fish digestive tracts.

Abstract: Estrogens, such as the 17β-estradiol (E2) and the 17α-ethinylestradiol (EE2), have been regarded as a global threat to aquatic ecosystems due to their pseudo-persistence, their high estrogenic activity and their toxicity towards non-target species. Data regarding their ecotoxicological effects on marine calanoid copepods are very scarce. In this study, the calanoid copepod Acartia clausi was used as a model organism for estrogens exposure in marine pelagic ecosystems. Lethal effects of estrogens on A. clausi life-stages (Embryos, one day old nauplii: N1, three day old nauplii: N3, copepodites: C1-C3 and adults: C6) were investigated using 48 h acute tests. Abstract: International audience; Imbalance between proteases and their inhibitors plays a crucial role in the development of Inflammatory Bowel Diseases (IBD). Increased elastolytic activity is observed in the colon of patients suffering from IBD. Here, we aimed at identifying the players involved in elastolytic hyperactivity associated with IBD and their contribution to the disease. We revealed that epithelial cells are a major source of elastolytic activity in healthy human colonic tissues and this activity is greatly increased in IBD patients, both in diseased and distant sites of inflammation. This study identified a previously unrevealed production of elastase 2A (ELA2A) by colonic epithelial cells, which was enhanced in IBD patients. We demonstrated that ELA2A hyperactivity is sufficient to lead to a leaky epithelial barrier. Epithelial ELA2A hyperactivity also modified the cytokine gene expression profile with an increase of pro-inflammatory cytokine transcripts, while reducing the expression of pro-resolving and repair factor genes. ELA2A thus appears as a novel actor produced by intestinal epithelial cells, which can drive inflammation and loss of barrier function, two essentials pathophysiological hallmarks of IBD. Targeting ELA2A hyperactivity should thus be considered as a potential target for IBD treatment.

Abstract: Background and Aims Intestinal epithelial cells [IECs] from inflammatory bowel disease [IBD] patients exhibit an excessive induction of endoplasmic reticulum stress [ER stress] linked to altered intestinal barrier function and inflammation. Colonic tissues and the luminal content of IBD patients are also characterized by increased serine protease activity. The possible link between ER stress and serine protease activity in colitis-associated epithelial dysfunctions is unknown. We aimed to study the association between ER stress and serine protease activity in enterocytes and its impact on intestinal functions Methods The impact of ER stress induced by Thapsigargin on serine protease secretion was studied using either human intestinal cell lines or organoids. Moreover, treating human intestinal cells with protease-activated receptor antagonists allowed us to investigate ER stress-resulting molecular mechanisms that induce proteolytic activity and alter intestinal epithelial cell biology. Results Colonic biopsies from IBD patients exhibited increased epithelial trypsin-like activity associated with elevated ER stress. Induction of ER stress in human intestinal epithelial cells displayed enhanced apical trypsin-like activity. ER stress-induced increased trypsin activity destabilized intestinal barrier function by increasing permeability and by controlling inflammatory mediators such as C-X-C chemokine ligand 8 [CXCL8]. The deleterious impact of ER stress-associated trypsin activity was specifically dependent on the activation of protease-activated receptors 2 and 4. Abstract: Recurrent jellyfish blooms in the coastal zone call for understanding the impacts of jelly-falls on the functioning of benthic communities, especially in shallow enclosed ecosystems where their biomass can affect local carbon cycling and productivity. Each year, blooms of the jellyfish Aurelia coerulea appear and collapse in a semienclosed coastal Mediterranean lagoon (the Thau lagoon, south of France). Although the lagoon is shallow, large accumulations of dead jellyfish are never observed on its bottom, so it was hypothesized that decaying jellyfish were rapidly consumed by local macrobenthic organisms. The current work aimed to test this hypothesis, by estimating the impact of the presence of dead A. aurelia medusae on local macrobenthic community composition and assessing their biomass loss rates under different scenarios of accessibility by the macrobenthos. Unexpectedly, our results revealed a limited role of macrobenthic scavengers in the disappearance of dead medusae, although this later was particularly fast (19-78h). Only one taxon (Tritia sp., Nassariidae family) showed a significant response to the presence of dead A. coerulea medusae on the seabed. Thus, our results suggest that the fast disappearance of dead jellyfish biomass in Thau results from its rapid degradation and consumption by local microorganisms, likely due to the combined effects of high local temperatures and the small size of A. coerulea medusae. Thus, the important biomass produced during A. aurelia blooms in Thau might essentially boost its microbial food web. The potential role of jellyfish blooms in controlling biogeochemical cycles and food web functioning in shallow lagoons is discussed, underlying the need to include this process in ecosystem-based models. info:eu-repo/semantics/publishedVersion

Abstract: WOS:000572257800001; International audience; The current lack of knowledge on the trophic ecology of scyphozoans, particularly at the benthic stage, prevents a full understanding of the controls on many jellyfish blooms. The blooming scyphozoan (Aurelia coerulea) completes its entire life cycle in the Thau lagoon (southern France), where the annual population dynamics of both its benthic and pelagic stages have been described. This offered an exceptional framework to investigate the trophic processes regulating jellyfish populations over time. To this aim, stable isotopic signature analysis (δ13C and δ15N) was used to infer the diet of both A. coerulea scyphistomae and medusae over 1 year. These results were matched with medusae gut content analysis and with the monthly abundances of local plankton groups. Lastly, the isotopic signatures of A. coerulea scyphistomae and medusae were compared with those of the oysters (Crassostrea gigas) cultivated in the lagoon to evaluate the potential interspecific trophic competition. The results revealed two seasonal shifts in the trophic niche of A. coerulea and substantial overlap between the diets of its benthic and pelagic stages. Conversely, trophic niche overlaps with the oysters were restricted, suggesting a limited impact of the local jellyfish bloom on shellfish production. Phytoplankton, microzooplankton, mesozooplankton, and sedimentary organic matter were all important food sources during critical periods of A. coerulea life-cycle. However, microzooplankton abundance was found to be key for the production of buds by the scyphistomae and, therefore it is likely to control the benthic population size and, thereby, to modulate the intensity of its annual bloom in Thau.

Abstract: [Aim] Global anthropogenic changes have altered biogeography and phenology of marine populations, thereby promoting a spatial reconfiguration in the functioning of marine ecosystems. Among these changes, massive proliferations of jellyfish in temperate latitudes warn of potential alterations in biogeochemical fluxes, ecosystems’ structure and assets, and the services they provide to human welfare. Understanding driving factors shaping large-scale patterns of jellyfish proliferations is a pressing need in global ecology and sustainability science. Using a comprehensive dataset of the largest blooming scyphomedusae in southern European seas we test, over broad space–time scales, current hypotheses relating jellyfish dynamics to warming and eutrophication., [Location] Southern European seas., [Time period] 1875–2019., [Major taxa studied] Rhizostoma pulmo., [Methods] We have gathered historical and contemporaneous records of R. pulmo from the Mediterranean and Black Seas over the last two centuries (7,359 records). Generalized statistical models were used to assess the influence of thermal (latitudinal) and productivity (longitudinal) gradients on the biogeographical patterns, and the species’ phenology at large and regional scales., [Results] Rhizostoma pulmo abundance exhibited an enhanced magnitude and frequency in recent decades, concurrently with positive temperature anomalies. We found that the latitudinal temperature gradient, but not productivity, shaped long-term bloom intensity and biogeographical patterns of the species. Our analysis further uncovered a significant effect of the interannual variability of spring temperature on the species’ phenology over the period 2008–2018, with warmer springs favouring an earlier start (c. 3 months) and a longer duration (from 5 to 7 months) of jellyfish season., [Main conclusions] Among the current hypotheses linking jellyfish changes with anthropogenic disturbances, only the warming-based hypothesis gained support over wide space–time scales, while the eutrophication-based hypothesis mainly applied at local scales. Abstract: In recent decades, an increase in the abundance and frequency of bloom events has been reported for the scyphozoan Rhizostoma pulmo in the Mediterranean Sea. Understanding such events requires a thorough assessment of the species' population dynamics through environmental windows allowing species development. The semi-enclosed coastal lagoon of Bages Sigean, France (43 degrees 05 ' 12.72 '' N; 3 degrees 00 ' 35.3 '' E) offers an exceptional framework for investigating the population dynamics of the species, and how its growth rates and environmental niches vary over time. Three cohorts starting in April, May and June 2019 were identified, while the overall population growth reached the maximum biomass (10.2 g m(-3)) in July. Bell diameter and total length were identified as the best morphological proxies of biomass estimation. The abundances of the two most abundant copepods' species appear to drive R. pulmo's dynamics in the lagoon. Based on multinomial analysis and using the von Bertalanffy model, different growth rates for juveniles (4.7 and 2.4 mm day(-1)) and adults (1.8 and 0.9 mm day(-1)) were determined for the first two cohorts. Thermohaline niches varied during ontogeny, but also among populations in three coastal Mediterranean lagoons: Bages Sigean (France), Mar Menor (Spain) and Bizerte (Tunisia), shedding light on the metapopulation dynamics of R. pulmo inhabiting the Mediterranean Sea. The pressing need for understanding the dynamics of jellyfish abundances and their impacts on ecosystems, calls for increased efforts on monitoring these populations and their life history traits to parametrize and build reliable ecosystem models.

Abstract: International audience; Excessive use of nickel oxide nanoparticles (NiO NPs) in various industrial and commercial products can lead to various negative effects in human and environmental health due to their possible discharge into the environment. Nerveless, information about their eco-toxicological effects on marine organisms are lacking. Copepods are good eco-toxicological models because of their high sensitivity to environmental stress and their key role in the marine food webs. In this study, 48 h acute tests were conducted on the marine planktonic copepod Centropages ponticus to assess lethal and sublethal toxicities of NiO NPs. The results revealed LC 50 (48 h) of 4 mg/L for adult females. Aggregation and settling of NiO NPs were observed at concentrations ≥2 mg/L. Exposure to sublethal concentrations (≥0.02 mg/L for 48 h) had significant negative effects on reproductive success in C. ponticus. Egg production after 24 h and 48 h decreased by 32% and 46%, respectively at 0.02 mg/L and 70% and 82%, respectively, at 2 mg/L. Hatching success was reduced by 70% and 79% at 2 mg/L for eggs produced after 24 h and 48 h respectively. Antioxidant enzymatic activity increased significantly with NiO NP concentration and time, indicating that NiO NPs can cause oxidative stress in C. ponticus even under short-term exposure, while significant inhibition of acetylcholinesterase activity at 2 mg/L after 48 h suggests neurotoxic effects of NiO NPs.

Abstract: International audience; MUTYH-associated polyposis (MUTYH-associated polyposis, MAP) is an autosomal recessive inheritance disorder related to bi-allelic constitutional pathogenic variants of the MUTYH gene which was first described in 2002. In 2011, a group of French experts composed of clinicians and biologists, performed a summary of the available data on this condition and drew up recommendations concerning the indications and the modalities of molecular analysis of the MUTYH gene in index cases and their relatives, as well as the management of affected individuals. In view of recent developments, some recommendations have become obsolete, in particular with regard to the molecular analysis strategy since MUTYH gene has been recently included in a consensus panel of 14 genes predisposing to colorectal cancer. This led us to revise all the points of the previous expertise. We report here the revised version of this work which successively considers the phenotype and the tumor risks associated with this genotype, the differential diagnoses, the indication criteria and the strategy of the molecular analysis and the recommendations for the management of affected individuals. We also discuss the phenotype and the tumor risks associated with mono-allelic pathogenic variants of MUTYH gene.

Abstract: Background & Aims Reinforced hepatocellular carcinoma (HCC) surveillance using magnetic resonance imaging (MRI) could increase early tumour detection but faces cost-effectiveness issues. In this study, we aimed to evaluate the cost-effectiveness of MRI for the detection of very early HCC (Barcelona Clinic Liver Cancer [BCLC] 0) in patients with an annual HCC risk >3%. Methods French patients with compensated cirrhosis included in 4 multicentre prospective cohorts were considered. A scoring system was constructed to identify patients with an annual risk >3%. Using a Markov model, the economic evaluation estimated the costs and life years (LYs) gained with MRI vs. ultrasound (US) monitoring over a 20-year period. The incremental cost-effectiveness ratio (ICER) was calculated by dividing the incremental costs by the incremental LYs. Results Among 2,513 patients with non-viral causes of cirrhosis (n = 840) and/or cured HCV (n = 1,489)/controlled HBV infection (n = 184), 206 cases of HCC were detected after a 37-month follow-up. When applied to training (n = 1,658) and validation (n = 855) sets, the construction of a scoring system identified 33.4% and 37.5% of patients with an annual HCC risk >3% (3-year C-Indexes 75 and 76, respectively). In patients with a 3% annual risk, the incremental LY gained with MRI was 0.4 for an additional cost of €6,134, resulting in an ICER of €15,447 per LY. Compared to US monitoring, MRI detected 5x more BCLC 0 HCC. The deterministic sensitivity analysis confirmed the impact of HCC incidence. At a willingness to pay of €50,000/LY, MRI screening had a 100% probability of being cost-effective. Conclusions In the era of HCV eradication/HBV control, patients with annual HCC risk >3% represent one-third of French patients with cirrhosis. MRI is cost-effective in this population and could favour early HCC detection. Lay summary The early identification of hepatocellular carcinoma in patients with cirrhosis is important to improve patient outcomes. Magnetic resonance imaging could increase early tumour detection but is more expensive and less accessible than ultrasound (the standard modality for surveillance). Herein, using a simple score, we identified a subgroup of patients with cirrhosis (accounting for >one-third), who were at increased risk of hepatocellular carcinoma and for whom the increased expense of magnetic resonance imaging would be justified by the potential improvement in outcomes., Graphical abstract, Highlights • HCC risk stratification in patients with cirrhosis aims at improving performance of cancer surveillance. • Using a simple scoring system, one-third of patients with cirrhosis with an annual HCC risk >3% can be easily identified. • Semi-annual surveillance using MRI in this population could enable the cost-effective detection of 5x more BCLC 0 HCCs.

Abstract: Germline mutations of the POLE gene are responsible for polymerase proofreading-associated polyposis syndrome (PPAP). These mutations were hypothesised to predispose to extra-gastrointestinal tumours (ovary, endometrium, brain), but this association has not been confirmed so far. We report a family with an autosomal dominant inheritance of PPAP due to a c.1089C>A; p.Asn363Lys mutation in the proofreading exonuclease domain of POLE. Ten patients presenting a history of colorectal tumours and three patients with polyposis are indexed in this family. Three carriers (including siblings and a distant cousin at 30, 45 and 52 respectively) and another member (at 37 not tested) presented glioblastoma. This is the second family reported to carry this mutation. Among the four glioblastomas in the family that we report, both show similar pathology: giant cell glioblastoma. These cases suggest that the c.1089C>A germline POLE mutation may confer an increased risk of brain cancer [incidence 17.4% (4/23) in mutation carriers combining the two families]. More observations are needed to support this hypothesis. It seems that not all mutations of POLE are equally associated with extra-gastrointestinal tumours. Although carriers of a mutation responsible for PPAP should benefit from screening for colorectal and uterine cancer, due to the rapid evolution of glioblastoma the value of neurological follow-up and brain imaging screening remains questionable. Nevertheless, considering the limitations of standard therapy for glioblastoma, mutation status could be useful for targeting therapy. The biological mechanism linking POLE mutation to glioblastoma remains to be determined.

Abstract: The jellyfish Obelia belongs to the family Campanulariidae (Hydrozoa, Leptomedusae), an important and widely distributed family of hydrozoans. Obelia is a very popular and widespread medusa, however first report on its complete life cycle was only published in the late nineties in Northern Japan. This is probably due to its size, as the medusa umbrella diameter is lower than 1mm. This Hydrozoa presents a bentho-pelagic life cycle with both polyp and medusa stages. In Thau lagoon, Northwestern Mediterranean Sea, populations of Obelia occur each year. Some polyps colonies were identified and sampled in the lagoon on Zostera noltii leaves, indicating that the populations realise their full development cycle within the lagoon. Morphological and genetical (COI) approaches, on both polyps and medusae, have revealed that main populations are composed by O. dichotoma individuals (side branches typically irregular in length; hydroteca bell-shaped, usually not very deep, thin walled, often thrown into fine longitudinal folds; hydrotecal rim with smooth or with shallow cusps –crenate, slightly flared; diaphragm transverse to oblique) but that O. bidentata (lateral branches roughly in right angles pairs are given on both sides; slightly oblique diaphragm; hydrotecal rim with bimucronate cusps) is also present in lower abundances. An in situ pelagic monitoring every two weeks since 2008 have allowed understanding the populations dynamics of the genus and the environmental factors that seemed to be particularly appropriated for promoting optimum growth conditions. Baseline information regarding seasonal cycles and historical abundances were provided from a 8 years monitoring, which has allowed detecting blooms. Indeed, the medusae were found at very low densities during the study period with main abundances under 5 ind.m-3, in accordance to previous reports for Obelia spp. medusae around the world. Nevertheless, 2 exceptional events took place in June 2008 and May 2013 were blooms reached 1232 and 660 ind.m-3 respectively. Such data raise questions regarding the potential importance of this small and understudied hydrozoa in the pelagic community of Thau lagoon. Consequently, it is necessary to improve the available databases documenting medusae blooms, as this will provide baseline information about these little-studied events.

Abstract: International audience; In the context of increasing demand for environmental recovery, aquatic systems may face the challenge of evolving under oligotrophication. This is the case in Mediterranean lagoons, in particular the shellfish-farmed Thau lagoon in France, where we studied recruitment of the Pacific oyster Crassostrea gigas. Oyster spat and environmental parameters were monitored at several sampling sites for 3 yr (2012 to 2014) using an original method with a temporal overlap deployment of collectors to study pre- and post-settlement processes and to identify the best conditions for recruitment. Contrary to the ‘no Pacific oyster reproduction’ paradigm in Mediterranean lagoons, our study showed that recruitment of this introduced species is possible in the Thau lagoon at levels comparable to those in other traditional French breeding basins. We identified a favorable environmental window for recruitment characterized by high water temperature (>26.5°C) and high nanophytoplankton and Chaetoceros spp. abundances (>4.3 × 106 and 345 × 103 cells l-1, respectively). In these favorable conditions, we hypothesize that the ecosystem functions as an autotrophic system, in contrast to the heterotrophic system that characterizes unfavorable conditions. Under heterotrophic conditions, high abundances of mixotrophic and heterotrophic organisms (ciliates and dinoflagellates) limited the metamorphosis of C. gigas larvae, leading to poor recruitment. This study provides new knowledge on the reproduction of the Pacific oyster in a Mediterranean lagoon under warming and oligotrophication. The shellfish industry will profit from the discovery of spatfields to develop new nursery practices that are eco-friendly and limit risks of transfers with other spatfall areas.

Abstract: 23 pages, 10 figures, 5 tables, supplementary data https://doi.org/10.1016/j.marenvres.2019.104791, Harmful invader ctenophore Mnemiopsis leidyi's expansions in the Eurasian Seas, its spatio-temporal population dynamics depending on environmental conditions in recipient habitats have been synthesized. M. leidyi found suitable temperature, salinity and productivity conditions in the temperate and subtropical environments of the semi-enclosed seas, in the coastal areas of open basins and in closed water bodies, where it created autonomous populations. M. leidyi changes its phenology depending on seasonal temperature regime in different environments. We assessed ranges of sea surface temperature, sea surface salinity and sea surface chlorophyll values, sufficient for M. leidyi general occurrence and reproduction based on comprehensive long-term datasets, contributed by co-authors. This assessment revealed that there are at least two eco-types (Southern and Northern) in the recipient seas of Eurasia with features specific for their donor areas. Abstract: Until recently, jellyfish have been ignored as an important source of food, due to their low nutritional value. Here, quantitative PCR was used to detect and quantify the DNA of the jellyfish Aurelia coerulea in the gut contents of commercially important fish species from the Thau Lagoon. Individuals from five fish species were collected during two different periods: the bloom period, when the pelagic stages of A. coerulea are abundant, and the post-bloom period, when only the benthic stage – polyps – is present in the lagoon. The DNA of A. coerulea was detected in the guts of 41.9% of the fish analysed, belonging to four different species. The eel Anguilla anguilla and the seabream Sparus aurata were important jellyfish consumers during the bloom and post-bloom periods, respectively. These results provide new insights on the potential control of jellyfish populations and on jellyfish importance as a food source for exploited fishes.

Abstract: For many jellyfish, the magnitude and timing of medusae blooms are recognized to result from the benthic stage dynamics. However, information on the scyphistomae of jellyfish populations in the wild remains scarce. Here, bi-mensual underwater photoquadrat surveys were combined with scyphistomae sampling and observation to describe the annual (February 2017-January 2018) benthic stage dynamics and asexual reproduction strategy of Aurelia coerulea in the Thau lagoon (43 degrees 2531.1N; 03 degrees 420.9E). Our results revealed unexpected seasonal patterns of variation: scyphistoma coverage peaked in the spring (11.6 +/- 3.7% on 21stApril) and was minimal in the summer and autumn (1.4 +/- 1.3% on 10th October). The increase in scyphistoma coverage mainly resulted from an intense production of buds between February and April during the spring rise in water temperature (peak of 12,800 buds m(-2) on 21stApril), but scyphistoma coverage appeared to be negatively influenced by the interaction of high summer temperatures and salinities. Strobilation was observed from November to April. It peaked on 17th November, with 33.1% of the scyphistomae strobilating and an average production of 19,100 strobila disks m(-2). Abstract: International audience; The recent discovery of Pacific oyster Crassostrea gigas (also known as Magallana gigas) spatfields in a Mediterranean lagoon intensely exploited for shellfish farming (Thau lagoon) revealed significant contrasts in spatial patterns of recruitment. We evaluated the processes that drive spatial patterns in oyster recruitment by comparing observed recruitment, simulated hydrodynamic connectivity and ecological variables. We hypothesized that spatial variability of recruitment depends on (1) hydrodynamic connectivity and (2) the ecology of the larval supply, settlement, metamorphosis, survival and biotic environmental parameters. We assessed recruitment at 6-8 experimental sites by larval sampling and spat collection inside and outside oyster farming areas and on an east-west gradient, from 2012-2014. Hydrodynamic connectivity was simulated using a numerical 3D transport model assessed with a Eulerian indicator. The supply of large umbo larvae did not differ significantly inside and outside oyster farming areas, whereas the supply of pediveligers to sites outside shellfish farms was structured by hydrodynamic connectivity. Inside shellfish farming zones, unfavorable conditions due to trophic competition with filter-feeders jeopardized their settlement. In this case, our results suggest loss of settlement competence by oyster larvae. This confirms our hypothesis of top-down trophic control by the oysters inside farming zones of Thau lagoon in summer that fails to meet the ecological requirements of these areas as oyster nurseries. Knowledge of oyster dispersal, connectivity and recruitment in coastal lagoons will help local development of sustainable natural spat collection. On a global scale, our method could be transposed to other basins or used for other species such as mussels, clams or scallops, to better understand the spatial patterns of bivalve recruitment. Management of the oyster industry based on natural spat collection will help develop a sustainable activity, based on locally adapted oyster strains but also by reducing the risks of transferring pathogens between basins and the global carbon footprint of this industry.

Abstract: Japanese oysters spatfields were recently discovered in the French Mediterranean Thau lagoon farmed for shellfish. This discovery led to interesting issues concerning research for a highly seashell exploited ecosystem. The analyses of various environmental parameters characterized favorable sites and periods for spat collection. Spat collection in Thau lagoon presented high temporal and spatial heterogeneities related to hydrodynamic, predation, competition and trophic resource variability. Here, the results highlight various explicative factors of the Japanese oysters recruitment within Thau lagoon and underline the high ecological variability in the studied system under oligotrophication.

Abstract: Colonic macrophages are considered as major effectors of inflammatory bowel diseases (IBD), and the control of gut inflammation through C-type lectin receptors is an emerging concept. We show that during colitis the loss of Dectin-1 prevents intestinal inflammation, while the lack of MR exacerbates it. A marked increase in Dectin-1 expression in DSS-exposed MR-deficient mice, support the critical contribution of Dectin-1 in the development of colitis. Dectin-1 is crucial for Ly6ChighCCR2high monocyte population enrichment in the blood and their recruitment to inflamed colon as precursors of M1 inflammatory macrophages. Dectin-1 also promotes inflammasome-dependent IL-1β secretion through the leukotriene B4 production. Interestingly, the colonic inflammation is associated with a concomitant overexpression of Dectin-1/CCL2/LTA4H and a down-regulation of MR on macrophages from IBD patients. Thus, MR and Dectin-1 on macrophage are important mucosal inflammatory regulators during IBD. Finally, this study offers breakthroughs on the mechanism may contribute to the pathogenesis of IBD in humans.

Abstract: BACKGROUND AND OBJECTIVES To analyze overall survival (OS) rates for the three curative treatments of hepatocellular carcinoma (HCC) on an intention-to-treat (ITT) basis. METHODS Cohort study based on data from a multidisciplinary team meeting (MDT) dedicated to HCC. From 2006 to 2013, we included every patient with newly diagnosed HCC, for whom curative treatment (liver transplantation (LT), radiofrequency ablation (RFA), surgical resection (SR)) was decided upon. RESULTS We included 387 consecutive patients. LT was decided in 136 cases, RFA in 131 cases, SR in 120 cases. Sixty-six percent of patients received the planned treatment. Five-year OS on an ITT basis were: 35% for the LT-group, 32% for the RFA-group, 34% for the SR-group (P = 0.77). Abstract: International audience; Communication between the endoplasmic reticulum (ER) and mitochondria plays a pivotal role in Ca2+ signaling, energy metabolism, and cell survival. Dysfunction in this cross-talk leads to metabolic and neurodegenerative diseases. Wolfram syndrome is a fatal neurodegenerative disease caused by mutations in the ER-resident protein WFS1. Here, we showed that WFS1 formed a complex with neuronal calcium sensor 1 (NCS1) and inositol 1,4,5-trisphosphate receptor (IP3R) to promote Ca2+ transfer between the ER and mitochondria. In addition, we found that NCS1 abundance was reduced in WFS1-null patient fibroblasts, which showed reduced ER-mitochondria interactions and Ca2+ exchange. Moreover, in WFS1-deficient cells, NCS1 overexpression not only restored ER-mitochondria interactions and Ca2+ transfer but also rescued mitochondrial dysfunction. Our results describe a key role of NCS1 in ER-mitochondria cross-talk, uncover a pathogenic mechanism for Wolfram syndrome, and potentially reveal insights into the pathogenesis of other neurodegenerative diseases.

Abstract: International audience; Growing evidence has shown a profound modification of plankton communities of the North East Atlantic and adjacent seas over the past decades. This drastic change has been attributed to a modification of the environmental conditions that regulate the dynamics and the spatial distribution of ectothermic species in the ocean. Recently, several studies have highlighted modifications of the regional climate station L4 (50° 15.00′N, 4° 13.02′W) in the Western English Channel. We here focus on the modification of the plankton community by studying the long-term, annual and seasonal changes of five zooplankton groups and eight copepod genera. We detail the main composition and the phenology of the plankton communities during four climatic periods identified at the L4 station: 1988–1994, 1995–2000, 2001–2007 and 2008–2012. Our results show that long-term environmental changes underlined by Molinero et al. (2013) drive a profound restructuration of the plankton community modifying the phenology and the dominance of key planktonic groups including fish larvae. Consequently, the slow but deep modifications detected in the plankton community highlight a climate driven ecosystem shift in the Western English Channel.

Abstract: While proteases are essential in gastrointestinal physiology, accumulating evidence indicates that dysregulated proteolysis plays a pivotal role in the pathophysiology of inflammatory bowel disease (IBD). Nonetheless, the identity of overactive proteases released by human colonic mucosa remains largely unknown. Studies of protease abundance have primarily investigated expression profiles, not taking into account their enzymatic activity. Herein we have used serine protease-targeted activity-based probes (ABPs) coupled with mass spectral analysis to identify active forms of proteases secreted by the colonic mucosa of healthy controls and IBD patients. Profiling of (Pro-Lys)-ABP bound proteases revealed that most of hyperactive proteases from IBD secretome are clustered at 28-kDa. We identified seven active proteases: the serine proteases cathepsin G, plasma kallikrein, plasmin, tryptase, chymotrypsin-like elastase 3 A, and thrombin and the aminopeptidase B. Only cathepsin G and thrombin were overactive in supernatants from IBD patient tissues compared to healthy controls. Gene expression analysis highlighted the transcription of genes encoding these proteases into intestinal mucosae. Abstract: International audience; Rivers are known to be major contributors to eutrophication in marine coastal waters, but little is known on the short-term impact of freshwater surges on the structure and functioning of the marine plankton community. The effect of adding river water, reducing the salinity by 15 and 30%, on an autumn plankton community in a Mediterranean coastal lagoon (Thau Lagoon, France) was determined during a 6-day mesocosm experiment. Adding river water brought not only nutrients but also chlorophyceans that did not survive in the brackish mesocosm waters. The addition of water led to initial increases (days 1-2) in bacterial production as well as increases in the abundances of bacterioplankton and picoeukaryotes. After day 3, the increases were more significant for diatoms and dinoflagellates that were already present in the Thau Lagoon water (mainly Pseudo-nitzschia spp. group delicatissima and Prorocentrum triestinum) and other larger organisms (tintinnids, rotifers). At the same time, the abundances of bacterioplankton, cyanobacteria, and picoeukaryote fell, some nutrients (NH4 (+), SiO4 (3-)) returned to pre-input levels, and the plankton structure moved from a trophic food web based on secondary production to the accumulation of primary producers in the mesocosms with added river water. Our results also show that, after freshwater inputs, there is rapid emergence of plankton species that are potentially harmful to living organisms. This suggests that flash flood events may lead to sanitary issues, other than pathogens, in exploited marine areas.

Abstract: We provide evidence for a functional role of bioactive lipid mediators of the docosapentaenoic acid (DPA) metabolome in intestinal inflammation. Supported by changes in DPA-derived mediators in colon biopsies from inflammatory bowel diseases, we studied the pharmacological properties of two mediators. Exogenous administration of protectin (PD)1n-3 DPA or resolvin (Rv)D5n-3 DPA in mice reduced dextran sulfate sodium-induced colitis though a mechanism partly linked to decreased leukocyte–endothelial interaction and reduced granulocyte trafficking, as assessed by intravital microscopy. The translational impact of these data was determined by the ability of PD1n-3 DPA and RvD5n-3 DPA to reduce human neutrophil adhesion onto TNF-α–activated human endothelial monolayers. We propose that n-3 DPA-derived mediators could represent the basis for innovative therapeutic strategies in settings of intestinal inflammation.

Abstract: The resolution of inflammation is an active process orchestrated by specialized proresolving lipid mediators (SPM) that limit the host response within the affected tissue; failure of effective resolution may lead to tissue injury. Because persistence of inflammatory signals is a main feature of chronic inflammatory conditions, including inflammatory bowel diseases (IBDs), herein we investigate expression and functions of SPM in intestinal inflammation. Targeted liquid chromatography-tandem mass spectrometry-based metabololipidomics was used to identify SPMs from n-3 polyunsaturated fatty acids in human IBD colon biopsies, quantifying a significant up-regulation of the resolvin and protectin pathway compared with normal gut tissue. Systemic treatment with protectin (PD)1n-3 DPA or resolvin (Rv)D5n-3 DPA protected against colitis and intestinal ischemia/reperfusion-induced inflammation in mice. Inhibition of 15-lipoxygenase activity reduced PD1n-3 DPA and augmented intestinal inflammation in experimental colitis. Intravital microscopy of mouse mesenteric venules demonstrated that PD1n-3 DPA and RvD5n-3 DPA decreased the extent of leukocyte adhesion and emigration following ischemia-reperfusion. These data were translated by assessing human neutrophil–endothelial interactions under flow: PD1n-3 DPA and RvD5n-3 DPA reduced cell adhesion onto TNF-α–activated human endothelial monolayers. In conclusion, we propose that innovative therapies based on n-3 DPA-derived mediators could be developed to enable antiinflammatory and tissue protective effects in inflammatory pathologies of the gut.
