BACKGROUND: Aedes albopictus is an important vector for pathogens such as dengue, Zika, and chikungunya viruses. While insecticides is the mainstay for mosquito control, their widespread and excessive use has led to the increased resistance in Ae. albopictus globally. Gut symbiotic bacteria are believed to play a potential role in insect physiology, potentially linking to mosquitoes metabolic resistance against insecticides. METHODS: We investigated the role of symbiotic bacteria in the development of resistance in Ae. albopictus by comparing gut symbiotic bacteria between deltamethrin-sensitive and deltamethrin-resistant populations. Adults were reared from field-collected larvae. Sensitive and resistant mosquitoes were screened using 0.03% and 0.09% deltamethrin, respectively, on the basis of the World Health Organization (WHO) tube bioassay. Sensitive and resistant field-collected larvae were screened using 5 × LC50 (lethal concentration at 50% mortality) and 20 × LC50 concentration of deltamethrin, respectively. Laboratory strain deltamethrin-sensitive adults and larvae were used as controls. The DNA of gut samples from these mosquitoes were extracted using the magnetic bead method. Bacterial 16S rDNA was sequenced using BGISEQ method. We isolated and cultured gut microorganisms from adult and larvae mosquitoes using four different media: Luria Bertani (LB), brain heart infusion (BHI), nutrient agar (NA), and salmonella shigella (SS). Background: Anopheles stephensi, a malaria‐transmitting mosquito species, has developed resistance to various insecticides such as DDT, Dieldrin, Malathion, and synthetic pyrethroids. To combat this issue, the World Health Organization (WHO) suggests using Actellic®300CS and Icon®10CS for Indoor Residual Spraying to tackle pyrethroid‐resistant mosquitoes. The aim of this research project was to evaluate the susceptibility of An. stephensi to certain insecticides at the diagnostic concentration + intensity 5x diagnostic concentration (5XDC) assays in Iran and to study the lasting effectiveness of Actellic®300CS and Icon®10CS against this particular malaria vector. Methods: This study assessed the susceptibility of An. stephensi populations in southern Iran to various insecticides, including deltamethrin 0.05%, DDT 4%, malathion 5%, bendiocarb 0.1%, a synergist assay with PBO 4% combined with deltamethrin 0.05%, and an intensity assay using 5x the diagnostic concentration of deltamethrin (0.25%) and bendiocarb 0.5%. Laboratory cone bioassay tests were conducted to determine the residual effectiveness of Actellic®300 and Icon®10CS insecticides on different surfaces commonly found in households, such as cement, mud, plaster, and wood. The tests were carried out following the WHO test kits and standard testing protocols. Results: The An. stephensi populations in Bandar Abbas were found to be susceptible to malathion 5% and deltamethrin 0.25% (5XDC), but exhibited resistance to DDT, standard concentration of deltamethrin, and both standard and intensity concentrations of bendiocarb. In laboratory cone bioassay tests, An. stephensi mortality rates when exposed to Actellic®300CS and Icon®10CS on different surfaces remained consistently more than 80%. Actellic®300CS achieved more than 80% mortality on all substrates for the entire 300‐day post‐spraying period. Conversely, Icon®10CS maintained mortality rates more than 80% on plaster and wood surfaces for 165 days and on mud and cement surfaces for 270 days post‐spraying. Both Actellic®300CS and Icon®10CS demonstrated 100% mortality within 72 h of each test on all surfaces throughout the entire 300‐day post‐spraying period. Conclusion: The study shows the varying levels of resistance of An. stephensi Bandar Abbas population to different insecticides and demonstrates the consistent performance of Actellic®300CS in controlling these mosquitoes on various surfaces. The findings suggest that long‐lasting CS formulations may be more effective for malaria vector control compared to the current options. Mosquito coil is commonly used in many African households for protection against mosquito bites. The coil usually has semi‐volatile pyrethroids as an active ingredient, which usually diffuse across open space, and the cloud either kills mosquitoes that are exposed, or mosquitoes can be exposed to sublethal doses of the insecticides. This study was conducted to assess the impact of sublethal doses of mosquito coil on the development of insecticide resistance in Aedes aegypti, a major vector for dengue fever and several other arboviral diseases. A laboratory colony of Ae. aegypti was exposed to sublethal doses of a meperfluthrin‐based mosquito coil in a Peet‐Grady chamber once per generation for 16 generations. The susceptibility of the exposed colony to a diagnostic dose of the mosquito coil as well as to three other insecticides was determined. Three different kdr mutations and five enzyme activities were evaluated in both the exposed and control colonies. After 16 generations of sublethal exposure to mosquito coils, the full diagnostic dose of the coil caused 68% mortality to the exposed colony compared to 100% mortality in the control colony. Mortality caused by deltamethrin (0.05%) was also significantly lower in the exposed colony. The frequency of 1016I kdr mutation as well as MFO and alpha esterase activities were higher in the exposed colony compared to the control colony. This study provides evidence of the development of pyrethroid resistance in an Ae. aegypti population due to sublethal exposure to mosquito coil for 16 generations. Given the large‐scale use of mosquito coils in many African households, its role as a pyrethroid resistance selection source should be taken into consideration when designing resistance management strategies. Aedes albopictus is highly prevalent in the northern part of West Bengal and is considered to be responsible for the recent dengue outbreaks in this region. Control of this vector is largely relied on the use of synthetic pyrethroids, which can lead to the development of resistance. In the present study, larvae of three wild Ae. albopictus populations from the dengue‐endemic regions were screened for deltamethrin resistance, and the role of cytochrome P450 monooxygenases (CYPs) was investigated in deltamethrin exposed and unexposed larvae. Two populations were incipient resistant, and one population was completely resistant against deltamethrin. Monooxygenase titration assay revealed the involvement of CYPs in deltamethrin resistance along with an induction effect of deltamethrin exposure. Gene expression studies revealed differential expression of five CYP6 family genes, CYP6A8, CYP6P12, CYP6A14, CYP6N3 and CYP6N6, with high constitutive expression of CYP6A8 and CYP6P12 in all the populations before and after deltamethrin exposure. The species Ceratitis capitata (Weidemann, 1824), known as Mediterranean fruit fly, is an important pest for fruit trees worldwide. The control of this pest is carried out mainly by the chemical method, using organophosphorus, pyrethroid, and spinosyn pesticides. The lethality of doses of malathion, deltamethrin, and spinetoram on immature and adult stages of C. capitata was evaluated under laboratory conditions. The egg, larval, pupal, and adult stages were exposed to doses of pesticides through contact and ingestion. The doses were obtained by decreasing the recommended doses (malathion: 200 mL L−1, deltamethrin: 50 mL L−1, and spinetoram: 120 g ha−1) with intervals of 20%, this being the minimum dose used. The variables evaluated were: viability of eggs and pupae and mortality of larvae and adults. Application of spinetoram and malathion through contact caused high mortality of adults, with LD90 at the sub‐doses of 43.74% (52.48 g c.p. ha−1) and 68.81% (137.62 mL c.p. 100 L−1), respectively. The mortality of adults by ingestion of deltamethrin and malathion allowed us to estimate only the LD50. Only the pesticide deltamethrin presented LD50 for the egg stage. The larval and pupal stages the effect of the pesticides on larvae treated through contact resulted in 52% mortality with spinetoram and 62% with malathion. The mortality of larvae and pupae treated through ingestion was below 20% for the three pesticides. Sub‐doses of spinetoram (43.74%; 52.48 g c.p. ha−1) and malathion (68.81%; 137.62 mL c.p. 100 L−1) can be used since they present a 90% efficiency on C. capitata adult when applied by contact under laboratory conditions. Honeybees are important insects, both economically and as pollinators. While foraging, bees can come in contact with environmental pollutants such as pesticides, possibly in combination with other xenobiotic compounds that may compromise bee health. Our current study investigated the acute exposure to nanosized titanium dioxide, a common additive in food, cosmetics, paints and agricultural products, along with deltamethrin (DLT), a well‐known pyrethroid pesticide. The effects of binary exposure to titanium dioxide nanoparticles (TiO2‐NPs) nanoparticles at concentrations of 25, 50, 75 and 100 μg/mL with DLT were noted on Apis mellifera brain histology along with their possible synergism. In DLT‐ and combined TiO2‐NP‐DLT treated bees, survival rates were lowered and several histological alterations were observed including an increased number of cells with pyknotic nuclei, along with cytoplasmic vacuolization, chromatin compaction and degeneration indicating autophagic activity and a decrease in the cross‐sectional area of nanoparticle‐treated mushroom body calyces. A synergistic relationship between TiO2‐NPs and DLT was revealed, while LD50 for combined TiO2‐NP‐DLT treatment was 0.101, 0.09 and 0.02 μg/bee at 24, 48 and 72 h, respectively. In summary, results demonstrate that TiO2‐NP and DLT co‐exposure can induce damage in bee brain structures at higher concentrations, which indicates an additional risk factor for bee health in the field. Bell peppers, a globally significant crop, face infestations from various pests. In a study, bell peppers were treated with deltamethrin, ethion, fenazaquin, and fenpropathrin at recommended and double the doses, repeated twice with a 10-day interval. The QuEChERS method underwent validation for linearity, matrix match, accuracy, and precision in bell pepper matrices for residue analysis. The limit of detection for the tested pesticides on bell peppers was 0.01 mg/L, with a quantification limit of 0.05 mg/L. Recovery studies showed a range of 94.80% to 102.80%. Initial deposits of deltamethrin, ethion, fenazaquin, and fenpropathrin on bell peppers at recommended doses were 0.371, 1.237, 0.617, and 0.640 mg/L, respectively, and at double doses were 0.712, 1.945, 1.221, and 1.189 mg/L, respectively. Safe waiting periods of 10, 11, 10, and 8 days were suggested for deltamethrin, ethion, fenazaquin, and fenpropathrin, respectively. The corresponding half-lives for the pesticides were 1.96, 1.79, 2.06, and 1.69 days, all following first-order dissipation kinetics. Dietary risk assessment indicated Hazard Quotients (HQ) below 1 and Theoretical Maximum Daily Intake (TMDI) below Acceptable Daily Intake (ADI) and Maximum Permissible Intake (MPI) levels. West Nile virus (WNV) is the leading mosquito-borne disease causing-pathogen in the United States. Concerningly, there are no prophylactics or drug treatments for WNV and public health programs rely heavily on vector control efforts to lessen disease incidence. Insecticides can be effective in reducing vector numbers if implemented strategically, but can diminish in efficacy and promote insecticide resistance otherwise. Vector control programs which employ mass-fogging applications of insecticides, often conduct these methods during the late-night hours, when diel temperatures are coldest, and without a-priori knowledge on daily mosquito activity patterns. This study's aims were to 1) quantify the effect of temperature on the toxicity of two conventional insecticides used in fogging applications (malathion and deltamethrin) to Culex tarsalis, an important WNV vector, and 2) quantify the time of host-seeking of Cx. tarsalis and other local mosquito species in Maricopa County, Arizona. The temperature-toxicity relationship of insecticides was assessed using the WHO tube bioassay, and adult Cx. tarsalis, collected as larvae, were exposed to three different insecticide doses at three temperature regimes (15, 25, and 35°C; 80% RH). Time of host-seeking was assessed using collection bottle rotators with encephalitis vector survey traps baited with dry ice, first at 3h intervals during a full day, followed by 1h intervals during the night-time. Malathion became less toxic at cooler temperatures at all doses, while deltamethrin was less toxic at cooler temperatures at the low dose. Regarding time of host-seeking, Cx. tarsalis, Aedes vexans, and Culex quinquefasciatus were the most abundant vectors captured. During the 3-hour interval surveillance over a full day, Cx. tarsalis were most-active during post-midnight biting (00:00–6:00), accounting for 69.0% of all Cx. tarsalis, while pre-midnight biting (18:00–24:00) accounted for 30.0% of Cx. tarsalis. During the 1-hour interval surveillance overnight, Cx. tarsalis were most-active during pre-midnight hours (18:00–24:00), accounting for 50.2% of Cx. tarsalis captures, while post-midnight biting (00:00–6:00) accounted for 49.8% of Cx. tarsalis. Our results suggest that programs employing large-scale applications of insecticidal fogging should consider temperature-toxicity relationships coupled with time of host-seeking data to maximize the efficacy of vector control interventions in reducing mosquito-borne disease burden. Insecticidal fogging is often conducted during post-midnight hours when temperatures are coldest. The researchers aimed to test the effect of temperature on the toxicity of two common insecticides to Culex tarsalis, a primary mosquito vector of WNV, and to identify when local vectors were actively seeking a host to bite. The research location was a productive riparian (wetland) area adjacent to many urban centers in Arizona. The researchers found that both insecticides became less toxic at colder temperatures and that early evening accounted for a significant portion of captures, yet is outside of normal fogging hours. Human outdoor activities also increase during these hours, which may result in elevated risk of disease transmission. This research serves to improve our understanding and usage of insecticides to reduce disease burden. Deltamethrin is an insecticide used to control harmful agricultural insects that otherwise damage crops and to control vector‐borne diseases. Long‐term exposure to deltamethrin results in the inflammation of the lungs. The present study elucidates the molecular mechanism underlying the deltamethrin‐induced lung damage. The lung samples were extracted from the Swiss albino mice following the treatment of low (2.5 mg/kg) and high (5 mg/kg) doses of deltamethrin. The mRNA expression of TCR, IL‐4, and IL‐13 showed upregulation, while the expression of NFAT and FOS was downregulated following a low dose of deltamethrin. Moreover, the expression of TCR was downregulated with the exposure of a high dose of deltamethrin. Furthermore, the immunohistochemistry data confirmed the pattern of protein expression for TCR, FOS, IL‐4, and IL‐13 following a low dose of deltamethrin exposure. However, no change was seen in the TCR, NFAT, FOS, JUN, IL‐4, and IL‐13 immunopositive cells of the high‐dose treatment group. Also, ELISA results showed increased expression of IL‐13 in the BAL fluid of animals exposed to low doses of deltamethrin. Overall, the present study showed that deltamethrin exposure induces lung damage and immune dysregulation via dysregulating the NFAT signalling pathway. [ABSTRACT FROM AUTHOR]

Pyrethroid-chlorfenapyr nets have been recommended for malaria control by the World Health Organisation (WHO) after an alpha-cypermethrin-chlorfenapyr net showed improved impact in epidemiological trials. PermaNet® Dual is a new deltamethrin-chlorfenapyr net developed by Vestergaard Sàrl to expand options to control programmes. A series of laboratory studies were performed according to WHO guidelines to assess the regeneration time, efficacy and wash-resistance of PermaNet® Dual. Regeneration time was determined by subjecting net pieces to cone bioassays and tunnel tests before and 0, 1, 2, 3, 5 and 7 days after washing. The wash-resistance of PermaNet® Dual was evaluated compared to WHO-prequalified pyrethroid-only (PermaNet® 2.0) and pyrethroid-chlorfenapyr (Interceptor® G2) nets by testing net pieces washed 0, 1, 3, 5, 10, 15 and 20 times in cone bioassays and tunnel tests. Tests were performed with susceptible and pyrethroid-resistant strains of Anopheles gambiae to assess the pyrethroid and chlorfenapyr components separately. Net pieces were also analysed to determine insecticide content. In regeneration time studies, the biological activity of the deltamethrin and chlorfenapyr components of PermaNet® Dual regenerated within one day after washing and a 1-day washing interval was adopted for wash-resistance studies. PermaNet® Dual induced high mortality (98%) and blood-feeding inhibition (98%) of the susceptible strain after 20 washes fulfilling WHO efficacy criteria in tunnel tests (≥80% mortality, ≥90% blood-feeding inhibition). Similar results were obtained with PermaNet® 2.0 (99% mortality, 99% blood-feeding inhibition) and Interceptor® G2 (99% mortality, 98% blood-feeding inhibition) washed 20 times. In wash-resistance tunnel tests against the pyrethroid-resistant strain, PermaNet® Dual washed 20 times induced high mortality (91%) and blood-feeding inhibition (73%), which was similar to Interceptor® G2 (87% mortality, 79% blood-feeding inhibition) and superior to PermaNet® 2.0 (47% mortality, 68% blood-feeding inhibition). Background: Acaricide resistance in cattle ticks is a significant concern in (sub)tropical regions, particularly Brazil. The Larval Packet Test (LPT) is the standard laboratory bioassay for resistance diagnosis, which requires triplicates of seven acaricidal dilutions plus controls to cover larval mortalities ranging between 0 and 100%. The value of the LPT lies in providing resistance ratios based on the ratio between the LC50 calculated with potentially resistant and susceptible ticks. However, LC50 ratios are difficult to translate into practical advice for farmers. Moreover, LPT requires laboratory facilities to maintain susceptible tick colonies, and it takes 6 weeks to obtain the larvae to be tested by LPT derived from engorged female ticks collected from cattle in the field. Our novel approach was twofold: first, we upgraded the LPT to the Resistance Intensity Test (RIT) by adopting the latest WHO guidelines for resistance detection in mosquitoes, which combines a 1 × recommended dose with 5 × and 10 × concentrated doses to reveal low, moderate and high resistance intensity, respectively. This reduced the number of test papers and tick larvae and, more importantly, provided relevant information on the resistance level. Our second innovative step was to abolish testing larvae entirely and expose partly engorged adult ticks to the same acaricidal doses immediately after removing them from cattle in the field. This resulted in the Rapid Tick exposure Test (RaTexT®), wherein partly engorged adult ticks were exposed to an acaricide-impregnated, specially designed matrix providing test results within 24 h. This approach directly compared resistance detection in tick larvae in the RIT with resistance in adult ticks in RaTexT®. Methods: Laboratory validation was conducted in Brazil with resistant and susceptible colonies of Rhipicephalus microplus ticks. For field validation, adult R. microplus ticks collected from different cattle farms in Brazil were evaluated for resistance to RaTexT®, and the results regarding their larval progenies were compared with those for the RIT. Partly engorged adult ticks derived from cattle infested with laboratory and field strains of R. microplus were exposed to deltamethrin in RaTexT® containers, which contained six rows of four interconnected compartments, accommodating five to eight semi-engorged female ticks with a preferred size ranging between 5 and 8 mm. The corresponding larvae of each strain were exposed in the RIT to the same deltamethrin concentrations in filter papers. Results: In RaTexT®, mortality in adult ticks from a resistant strain of R. microplus from Seropédica in Brazil was 38.4%, 54.2% and 75.0% at the 1 ×, 5 × and 10 × doses of deltamethrin, respectively. In RIT, mortality of larvae from the same resistant strain was 2.0%, 4.9% and 19.5% at 1 ×, 5 × and 10 × doses, respectively. The results of RaTexT® and RIT agreed since both tests identified a high level of resistance based on a cut-off of 90% mortality. In RaTexT®, mortality of adult ticks from a susceptible strain originating from Porto Alegre was 73.8%, 92.9% and 97.6% at the 1 ×, 5 × and 10 × doses, respectively. In RIT, mortality of larvae from the susceptible strain was 95.2%, 95.2% and 96.8% at the 1 ×, 5 × and 10 × doses, respectively. Interestingly, both tests identified a low number of unexpected resistant individuals in the susceptible strain since the mortality of neither larvae nor adults reached 100%. This effect remained unnoticed in the LPT, wherein a resistance ratio of 159.5 was found based on the LC50 of the resistant strain divided by the LC50 of the susceptible strain. Next, RaTexT® was compared with RIT using adult and larval ticks derived from three field strains of R. microplus in Brazil. RaTexT® detected high levels of resistance to deltamethrin in adult ticks in all strains, which was confirmed in larvae tested by the RIT. Both tests agreed on the same resistance level with significantly lower mortality rates in larvae than in adult ticks. Conclusions: RaTexT® is a novel rapid pen-site test for detecting acaricide resistance in adult livestock ticks. Mosquito-borne viral diseases such as dengue fever, chikungunya, and yellow fever have been documented in Ethiopia since the 1960s. However, the efficacy of public health insecticides against Aedes aegypti that transmits these viruses remains poorly understood in the country, particularly in the Afar Region. Thus, the aim of the study was to assess the susceptibility status of Ae. aegypti to deltamethrin, permethrin, alpha-cypermethrin, pirimiphos-methyl, bendiocarb, and propoxur insecticides. Larvae and pupae of Aedes species were collected from Awash Arba, Awash Sebat, and Werer towns of the Afar Region of Ethiopia during July-October 2022, brought to the Aklilu Lemma Institute of Pathobiology, insectary and reared to adults. Non-blood-fed, 3–5 days-old females Ae. aegypti were exposed to pyrethroid, carbamate, and organophosphate insecticide impregnated papers in tube test following the standard guidelines. Knockdown rates were noted at 10 minutes interval until one hour. The mortality in mosquitoes was recorded 24 hours after 60 minutes of exposure. The mortality rates of Ae. aegypti exposed to propoxur were 87% in all the study towns. Similarly, 88% mortality in Ae. aegypti was recorded when tested with bendiocarb in Awash Sebat and Awash Arba towns. Suspected resistance of Ae. aegypti (95% mortality) to alpha-cypermethrin was observed in Awash Arba town. However, Ae. aegypti collected from all the three sites was observed to be susceptible to deltamethrin, permethrin, and pirimiphos-methyl. Ae. aegypti was resistant to 0.1% bendiocarb and 0.1% propoxur and possibly resistant to 0.05% alpha-cypermethrin. On the other hand, it was susceptible to 0.05% deltamethrin, 0.75% permethrin, and 0.25% pirimiphos-methyl. Thus, vector control products with deltamethrin, permethrin, and pirimiphos-methyl can be used in the control of adult Ae. aegypti in the Afar Region of Ethiopia. However, further studies should be carried out to evaluate the susceptibility status of Ae. aegypti to alpha-cypermethrin in the Awash Arba area. [ABSTRACT FROM AUTHOR]

Resistance to insecticides is one of the great challenges that vector control programs must face. The constant use of pyrethroid-type insecticides worldwide has caused selection pressure in populations of the Aedes aegypti vector, which has promoted the emergence of resistant populations. The resistance mechanism to pyrethroid insecticides most studied to date is target-site mutations that desensitize the voltage-gated sodium channel (VGSC) of the insect to the action of pyrethroids. In the present study, susceptibility to the pyrethroid insecticides permethrin, lambda-cyhalothrin, and deltamethrin was evaluated in fourteen populations from the department of Córdoba, Colombia. The CDC bottle bioassay and WHO tube methods were used. Additionally, the frequencies of the F1534C, V1016I, and V410L mutations were determined, and the association of resistance with the tri-locus haplotypes was examined. The results varied between the two techniques used, with resistance to permethrin observed in thirteen of the fourteen populations, resistance to lambda-cyhalothrin in two populations, and susceptibility to deltamethrin in all the populations under study with the CDC method. In contrast, the WHO method showed resistance to the three insecticides evaluated in all populations. The frequencies of the mutated alleles ranged from 0.05–0.43 for 1016I, 0.94–1.0 for 1534C, and 0.01–0.59 for 410L. The triple homozygous mutant CIL haplotype was associated with resistance to all three pyrethroids evaluated with the WHO bioassay, while with the CDC bioassay, it was only associated with resistance to permethrin. This study highlights the importance of implementing systematic monitoring of kdr mutations, allowing resistance management strategies to be dynamically adjusted to achieve effective control of Aedes aegypti. Introduction: Culex quinquefasciatus is one of the most important carriers of human pathogens. Using the insecticides is one of the most important methods of combating this vector. But the genetic resistance created in Culex quinquefasciatus led to disruption in the fight against this pest. Consequently, it is necessary to know the level of resistance to fight this vector. Based on this, the present study was conducted to investigate the prevalence of kdr resistance in Culex quinquefasciatus against organochlorine insecticides in the world. Methods: This study was conducted by systematic review, and meta-analysis on the prevalence of kdr resistance and mortality rate in Culex quinquefasciatus against organochlorine insecticides in the world. All pertinent articles were extracted and analyzed in accordance with this information during an unrestricted search of the scientific databases Web of Science, PubMed, Scopus, biooan.org, Embase, ProQuest, and Google Scholar until the end of November 2023. Statistical analysis of data was done using fixed and random effects model in meta-analysis, I2 index, Cochran's test, and meta-regression by STATA version 17 software. Results: Seventy articles were included in the meta-analysis process. Based on the findings, the prevalence of Kdr in Culex quinquefasciatus against organochlorine insecticide was estimated at 63.1%. Moreover, the mortality rate against the insecticide deltamethrin was 46%, DDT 18.5%, permethrin 42.6%, malathion 54.4% and lambdacyhalothrin 53%. Conclusion: More than half of Cx. quinquefasciatus had Kdr. This vector was relatively resistant to DDT and permethrin insecticides and sensitive to malathion, deltamethrin and lambdacyhalothrin. In order to prevent the development of resistance to alternative insecticides, it is consequently critical to combat this vector with efficacious insecticides. AbstractObjectiveMethodsResultsConclusionIn recent years, it has been known that the melatonin hormone, secreted from the pineal gland, possesses significant antioxidant activity. This study explores the therapeutic effect of melatonin on the deleterious effects of deltamethrin, a pyrethroid pesticide extensively used worldwide, including in Türkiye, on mouse liver cells.Hepatocytes from Balb/C mice were isolated using a two-stage perfusion method, resulting in over 85% live hepatocytes. The isolated cells were cultured with different doses of deltamethrin (1 and 10 µM) and melatonin (100 µM) for 24 and 48 hours. At the conclusion of the culture period, hepatocytes were extracted at the 24th and 48th hours, and Malondialdehyde (MDA), Total Antioxidant Capacity (TAC), Total Oxidation Status (TOS), and DNA damages (8-hydroxy-2’-deoxyguanosine (8-OHdG)) were examined.While an increase in MDA, TOS, and DNA damage was observed in the deltamethrin-administered groups of hepatocytes, a decrease in TAC level was noted. It was determined that the applied deltamethrin had no effect on cell viability throughout the application period.Furthermore, it was observed that melatonin, when administered concurrently with deltamethrin, reduced the toxic effect of deltamethrin. Brown planthopper (BPH), Nilaparvata lugens (Stal) is a monophagous and one of most devastating pest of rice. Application of insecticides is inevitable to control the infestation of BPH on rice crops. However, the indiscriminate use of various insecticides in rice is leading to pest resurgence. Several factors have been connected to the resurgence of brown planthoppers, especially the reduction in natural enemies and changes in plant nutrition after insecticide application. Hence, in the present study, we studied the resurgence of BPH post application of different insecticides in rice and assessed the impact of insecticides on natural enemy population during Rabi 2022-23. Among nine insecticides tested, two insecticides viz., deltamethrin 11 EC @ 15 g a.i/ha (44.70%) and imidacloprid 17.8 SL @ 25 g a.i/ha (23.01%) significantly increased the population of N. lugens over untreated check and consequently resulted in higher BPH resurgence. A greater reduction in predatory mirid bugs and spiders was observed in the deltamethrin and imidacloprid treated groups compared to the untreated check. In conclusion, we found that deltamethrin and imidacloprid induce the BPH resurgence in rice compared to other tested insecticides and the observed resurgence was partly due to the reduction of natural enemies. The Mediterranean Corn Borer (MCB), Sesamia nonagrioides, poses a significant threat to maize crops, necessitating effective pest management strategies. This study investigates the compatibility of two entomopathogenic nematode (EPN) isolates, Steinernema feltiae KV6 and Heterorhabditis bacteriophora EO7, with four registered insecticides for MCB control: deltamethrin, flubendiamide, spinetoram, and betacyfluthrin. The impact of these insecticides on EPN mortality, infectivity, and reproduction was assessed. Results indicate that deltamethrin exhibits the lowest toxicity to EPNs, with mortality rates of 1.3% for S. feltiae and 0.63% for H. bacteriophora at field dose (FD) after 24 h and 4.63% and 1.96%, respectively, after 48 h. In contrast, betacyfluthrin showed higher toxicity, with mortality rates of 38.04% and 14.17% for S. feltiae at 2FD and FD after 48 h. The infectivity assays demonstrated that deltamethrin-treated EPNs caused up to 100% mortality in MCB larvae, while the reproduction capacity varied significantly between the EPN species and insecticides. H. bacteriophora exhibited higher progeny production, especially in the presence of deltamethrin (87,900 IJs/larva). The findings suggest that integrating EPNs with selective insecticides like deltamethrin can enhance pest control efficacy and support sustainable agricultural practices. The versatility of cytochrome P450 reductase (CPR) in transferring electrons to P450s from other closely related species has been extensively exploited, e.g., by using An. gambiae CPR (AgCPR), as a homologous surrogate, to validate the role of An. funestus P450s in insecticide resistance. However, genomic variation between the AgCPR and An. funestus CPR (AfCPR) suggests that the full metabolism spectrum of An. funestus P450s might be missed when using AgCPR. To test this hypothesis, we expressed AgCPR and AfCPR side-by-side with CYP6P9a and CYP6P9b and functionally validated their role in the detoxification of insecticides from five different classes. Major variations were observed within the FAD- and NADP-binding domains of AgCPR and AfCPR, e.g., the coordinates of the second FAD stacking residue AfCPR-Y456 differ from that of AgCPR-His456. While no significant differences were observed in the cytochrome c reductase activities, when co-expressed with their endogenous AfCPR, the P450s significantly metabolized higher amounts of permethrin and deltamethrin, with CYP6P9b-AfCPR membrane metabolizing α-cypermethrin as well. Only the CYP6P9a-AfCPR membrane significantly metabolized DDT (producing dicofol), bendiocarb, clothianidin, and chlorfenapyr (bioactivation into tralopyril). This demonstrates the broad substrate specificity of An. funestus CYP6P9a/-b, capturing their role in conferring cross-resistance towards unrelated insecticide classes, which can complicate resistance management. Pyrethroids are synthetic organic insecticides. Deltamethrin, as one of the pyrethroids, has high insecticidal activity against pests and parasites and is less toxic to mammals, and is widely used in cities and urban areas worldwide. After entering the natural environment, deltamethrin circulates between solid, liquid and gas phases and enters organisms through the food chain, posing significant health risks. Increasing evidence has shown that deltamethrin has varying degrees of toxicity to a variety of organisms. This review summarized worldwide studies of deltamethrin residues in different media and found that deltamethrin is widely detected in a range of environments (including soil, water, sediment, and air) and organisms. In addition, the metabolism of deltamethrin, including metabolites and enzymes, was discussed. This review shed the mechanism of toxicity of deltamethrin and its metabolites, including neurotoxicity, immunotoxicity, endocrine disruption toxicity, reproductive toxicity, hepatorenal toxicity. Fall armyworm (FAW) Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae), is a global pest causing damage to several crops. However, its management using chemical control is a challenge due to its capacity to evolve resistance to insecticides. After 6 generations of selection with lambda-cyhalothrin, the LC50 for the insecticide-resistant strain (Lambda-Sel) was 486 ppm, higher than that of the field strain (FAW-MUL) (7.5 ppm), susceptible laboratory strain (Lab-PK) (0.46 ppm), and laboratory unselected strain (UNSEL) (5.26 ppm). Laboratory selection with lambda-cyhalothrin increased resistance from 16.3- to 1056.52-fold and 1.43- to 92.4-fold to lambda-cyhalothrin compared to Lab-PK and UNSEL strains, respectively. The selected strain of S. frugiperda (Lambda-Sel) presented low cross-resistance to chlorpyrifos, moderate to deltamethrin and indoxacarb, very low to spinosad, and no cross-resistance to emamectin benzoate. The realized heritability (h 2) of lambda-cyhalothrin resistance in the Lambda-Sel strain was very high (0.88). The reciprocal cross progenies of F1 (Lambda-Sel ♀ × Lab-PK ♂), F1ʹ (Lambda-Sel ♂ × Lab-PK ♀), BC1 (F1 ♀ × Lambda-Sel ♂), and BC2 (F1 ♀ × Lab-PK ♂) showed high resistance ratios of 545.64-, 396.52-, 181.18-, and 146.54-fold, respectively compared to Lab-PK. The degree of dominance values for lambda-cyhalothrin in F1 and F1ʹ indicates incompletely dominant resistance. The difference between observed and expected mortality in backcross populations (BC1 and BC2) revealed a polygenic resistance. In conclusion, the resistance to lambda-cyhalothrin was autosomal, incompletely dominant, and polygenic. Deltamethrin is used against plant pests (e.g., mites and ants) and, in farm animals, against biting insects because of its acaricidal and repellent effects against ticks, thus protecting the sheep and cattle from the transmission of pathogens. However, its impact on the environment still needs to be fully evaluated. This study evaluates the impact of this pyrethroid on the nitrogen-fixing nodules in Medicago truncatula, a model legume. This research compares nodular biomass and root weight between a deltamethrin-treated section and an untreated control section of this legume. Our results indicate a significant reduction in the biomass of nitrogen-fixing nodules in the treated grove, suggesting that deltamethrin negatively affects the symbiotic relationship between M. truncatula and nitrogen-fixing bacteria. This reduction in nodule formation can impair soil fertility and plant growth, highlighting an ecological risk associated with pyrethroid's use in livestock farming. These findings underscore the need for a shift towards Green Veterinary Pharmacology (GVP), which promotes environmentally sustainable practices in managing livestock health. Elevated resistance to pyrethroids in major malaria vectors has led to the introduction of novel insecticides including neonicotinoids. There is a fear that efficacy of these new insecticides could be impacted by cross-resistance mechanisms from metabolic resistance to pyrethroids. In this study, after evaluating the resistance to deltamethrin, clothianidin and mixture of clothianidin + deltamethrin in the lab using CDC bottle assays, the efficacy of the new IRS formulation Fludora® Fusion was tested in comparison to clothianidin and deltamethrin applied alone using experimental hut trials against wild free-flying pyrethroid-resistant Anopheles funestus from Elende and field An. gambiae collected from Nkolondom reared in the lab and released in the huts. Additionally, cone tests on the treated walls were performed each month for a period of twelve months to evaluate the residual efficacy of the sprayed products. Furthermore, the L1014F-kdr target-site mutation and the L119F-GSTe2 mediated metabolic resistance to pyrethroids were genotyped on a subset of mosquitoes from the EHT to assess the potential cross-resistance. All Anopheles species tested were fully susceptible to clothianidin and clothianidin + deltamethrin mixture in CDC bottle assay while resistance was noted to deltamethrin. Accordingly, Fludora® Fusion (62.83% vs 42.42%) and clothianidin (64.42% vs 42.42%) induced significantly higher mortality rates in EHT than deltamethrin (42.42%) against free flying An. funestus from Elende in month 1 (M1) and no significant difference in mortality was observed between the first (M1) and sixth (M6) months of the evaluation (P > 0.05). However, lower mortality rates were recorded against An. gambiae s.s from Nkolondom (mortality rates 50%, 45.56% and 26.68%). In-situ cone test on the wall showed a high residual efficacy of Fludora® Fusion and clothianidin on the susceptible strain KISUMU (> 12 months) and moderately on the highly pyrethroid-resistant An. gambiae strain from Nkolondom (6 months). Interestingly, no association was observed between the L119F-GSTe2 mutation and the ability of mosquitoes to survive exposure to Fludora® Fusion, whereas a trend was observed with the L1014F-kdr mutation. This study highlights that Fludora® Fusion, through its clothianidin component, has good potential of controlling pyrethroid-resistant mosquitoes with prolonged residual efficacy. Background: The widespread use of insecticide-treated nets (ITNs) has significantly contributed to the reduction in malaria cases and deaths observed across Africa. Unfortunately, this control strategy is threatened by the rapid spread of pyrethroid resistance in malaria vectors. Dual-active-ingredient insecticidal nets are now available to mitigate the impact of pyrethroid resistance. To facilitate evidence-based decisions regarding product selection in specific use settings, data are needed on the efficacy of these different nets against local mosquito populations. Methods: Two experimental hut trials were performed in Za-Kpota, southern Benin in 2021 to evaluate the performance of Interceptor G2 (BASF), Royal Guard (Disease Control Technologies) and PermaNet 3.0 (Vestergaard Frandsen), all dual-active-ingredient bednets, in comparison to untreated or standard pyrethroid-treated bednets, against free-flying wild Anopheles gambiae mosquitoes. The performance of some of these next-generation nets was compared to the same type of nets that have been in use for up to 2 years. Mosquitoes collected in the huts were followed up after exposure to assess the sublethal effects of treatments on certain life-history traits. Results: The predominant species in the study site was Anopheles gambiae sensu stricto (An. gambiae s.s.). Both Anopheles coluzzii and An. gambiae s.s. were resistant to pyrethroids (deltamethrin susceptibility was restored by piperonyl butoxide pre-exposure). In the experimental hut trials, the highest blood-feeding inhibition (5.56%) was recorded for the Royal Guard net, relative to the standard PermaNet 2.0 net (44.44% inhibition). The highest 72-h mortality rate (90.11%) was recorded for the Interceptor G2 net compared to the PermaNet 2.0 net (56.04%). After exposure, the risk of death of An. gambiae sensu lato (An. gambiae s.l.) was 6.5-fold higher with the Interceptor G2 net and 4.4-fold higher with the PermaNet 3.0 net compared to the respective untreated net. Lower mosquito mortality was recorded with an aged Interceptor G2 net compared to a new Interceptor G2 net. Oviposition rates were lower in mosquitoes collected from huts containing ITNs compared to those of untreated controls. None of the mosquitoes collected from huts equipped with Royal Guard nets laid any eggs. Conclusions: The Royal Guard and Interceptor G2 nets showed a potential to significantly improve the control of malaria-transmitting vectors. This experiment was conducted to determine the growth performance, blood profile and carcass characteristics of broilers fed yellow maize stored in Polypropylene (PP) and ZeroFly® Hermetic (ZFH) storage bags for six months. Two hundred unsexed day-old Cobb-500 broiler chicks were randomly allocated to 2 treatments (2 storage bags) with 100 chicks per treatment in a Completely Randomized Design experiment. Each treatment had four replications with 25 birds per replicate. The experimental diets were formulated using the same quantities of maize stored in the PP and ZFH bags. The study was in three phases, that is, starter, grower and finisher phases and each had 22%, 20% and 18% crude protein (CP), respectively. The experiment lasted for six weeks. Feed and water were offered ad libitum. Statistical analyses were performed with SAS Version 9.4. Effects of storage bags were assessed using a one-way analysis of variance (ANOVA). Tukey's Honestly Significant Difference test was used as an option in the Mixed Procedure to determine differences between means (p < 0.05). Broilers on the ZFH bag dietary treatment recorded significantly (p = 0.035) higher weight gain in grams (2395.11 ± 32.15), than their counterparts on the PP treatment (1980.30 ± 82.19). The feed conversion ratio, measured as feed: gain ratio, was better (p = 0.041) for the ZFH birds (1.74 ± 0.03) than the PP birds (2.24 ± 0.19). Birds fed the ZFH diet had higher (p < 0.05) dressing percentage (74.05 ± 0.24), percentage whole thigh (14.45 ± 0.10) and percentage drumstick (9.22 ± 0.16) than their PP counterparts. All the blood profile parameters measured were similar except total cholesterol and aspartate aminotransferase in which birds on the ZFH maize recorded higher values (p < 0.05). This study examined the pattern of resistance to widely applied synthetic pyrethroids, i.e., cypermethrin and deltamethrin, against larvae of Rhipicephalus microplus ticks sampled from Marathwada region in Maharashtra, India. The study also examined the role of α- and β-esterases and glutathione-S-transferase (GST) in resistance development. All eight R. microplus isolates tested were resistant to deltamethrin (RL IV), having RR50 values from 6.88 to 131.26. LPT analysis exhibited the resistance level II deltamethrin resistance in Beed and Hingoli, III in Dharashiv, and IV in Sambhajinagar, Parbhani, Latur, Jalna, and Nanded isolates. The LIT analysis showed that Dharashiv field isolates had the lowest LC50 value of 229.09 ppm against cypermethrin, while Sambhajinagar field isolates had the highest at 489.78 ppm. The RR50 ranged from 1145.45 to 2448.9. Seven isolates were level I resistant to cypermethrin while the Jalna isolate was level II resistant. In larvae treated with deltamethrin and cypermethrin, the activity of α- and β-esterase enzymes increased significantly compared to control groups. The enzyme ratios in treated larvae ranged from 0.7533 to 1.7023 for α-esterase and 0.7434 to 3.2054 for β-esterase. The Hingoli isolate treated with cypermethrin exhibited the highest α-esterase activity (903.261), whereas Sambhajinagar isolate had the highest GST enzyme ratio (2.8224) after deltamethrin exposure. When exposed to cypermethrin, the Hingoli isolate showed the highest GST enzyme ratio, 2.0832. The present study provides the current resistance status in tick populations from Marathwada region indicating deltamethrin and cypermethrin to be ineffective for tick control. BACKGROUND: The potential of copper nanoparticles (Cu‐NPs) to be used as an alternative control strategy against olive fruit flies (Bactrocera oleae) with reduced sensitivity to the pyrethroid deltamethrin and the impact of both nanosized and bulk copper hydroxide (Cu(OH)2) on the insect's reproductive and endosymbiotic parameters were investigated. RESULTS: The application of nanosized and bulk copper applied by feeding resulted in significant levels of adult mortality, comparable to or surpassing those achieved with deltamethrin at recommended doses. Combinations of Cu‐NPs or copper oxide nanoparticles (CuO‐NPs) with deltamethrin significantly enhanced the insecticide's efficacy against B. oleae adults. When combined with deltamethrin, Cu‐NPs significantly reduced the mean total number of offspring compared with the control, and the number of stings, pupae, female and total number of offspring compared with the insecticide alone. Both bulk and nanosized copper negatively affected the abundance of the endosymbiotic bacterium Candidatus Erwinia dacicola which is crucial for the survival of B. oleae larvae. CONCLUSION: The Cu‐NPs can aid the control of B. oleae both by reducing larval survival and by enhancing deltamethrin performance in terms of toxicity and reduced fecundity, providing an effective anti‐resistance tool and minimizing the environmental footprint of synthetic pesticides by reducing the required doses for the control of the pest. © 2024 The Authors. Tomato fruits are considerably affected by array of insect pests infesting at different stages of crop growth. Among the factors responsible for low yield of tomato, insect pests are major one. The present experiments entitled was carried out at Students' Instructional Farm, Acharya Deva University of Agriculture and Technology, Kumarganj, Ayodhya (UP) during rabi season 2020-21 to test the efficacy of certain combination insecticides against major insect pests. Among the efficacy of certain combination insecticides, the treatment Betacyfluthrin 8.49% + Imidacloprid 19.81% OD 400ml/ha was observed most effective against whitefly, Jassid and serpentine leaf miner, whereas Chlorpyriphos 50%+ Cypermethrin 5% EC 1000 ml/ha was least effective against whitefly, Profenofos 40%+Cypermethrin 4% EC 1250 ml/ha was found least effective against Jassid and Triazophos 35%+ Deltamethrin 1% EC 1000ml/ha was least effective against serpentine leaf miner. Chlorantraniliprole 10%+Lambda cyhalothrin 5% ZC 250 ml/ha was most effective and found Triazophos 35%+ Deltamehtrin 1% EC 100ml/ha least effective against larval population of H. armigera followed by Imidacloprid 40%+Fipronil 40% WG 500 g/ha, Imidacloprid 40%+Ethiprole 40% WG 500 g/ha. The maximum yield (203.41 q/ha) was recorded in Betacyfluthrin 8.49% +Imidacloprid 19.81% OD 400ml/ha and minimum yield was recorded in Triazophos 35%+ Deltramethrin 1% EC 1000ml/ha (163.17 q/ha). The benefit cost-ratio 1:19.31 was recorded highest in Betacyfluthrin 8.49%+Imidacloprid 19.81% OD 400 ml/ha and lowest 1:3.12 in Imidacloprid 40%+Ethiprole 40% WG 500 g/ha. The study was conducted at the Students' Instructional Farm, Acharya Narendra Deva University of Agriculture and Technology, Kumarganj, Ayodhya, Uttar Pradesh, India during the Kharif 2021. The investigation was aimed to evaluate specific combination insecticides against the Brown Plant Hopper and Rice Gundhi bug. Notably, the treatment Imidacloprid 40% + Fipronil 40% WG @ 500 g/ha followed by Imidacloprid 40% + Ethiprole 40% WG @ 500 g/ha demonstrated the highest effectiveness. Conversely, the least effective treatment was Chlorantraniliprole 10% + Lambda Cyhalothrin 5% ZC @ 250 ml/ha followed by Profenofos 40% + Cypermethrin 4% EC @ 1250 ml/ha when compared to the control treatment. All treatments resulted in significantly higher yields compared to the control treatment (18.17 q/ha). Methods We investigated the role of symbiotic bacteria in the development of resistance in Ae. albopictus by comparing gut symbiotic bacteria between deltamethrin-sensitive and deltamethrin-resistant populations. Adults were reared from field-collected larvae. Sensitive and resistant mosquitoes were screened using 0.03% and 0.09% deltamethrin, respectively, on the basis of the World Health Organization (WHO) tube bioassay. Sensitive and resistant field-collected larvae were screened using 5 × LC50 (lethal concentration at 50% mortality) and 20 × LC50 concentration of deltamethrin, respectively. Laboratory strain deltamethrin-sensitive adults and larvae were used as controls. The DNA of gut samples from these mosquitoes were extracted using the magnetic bead method. Bacterial 16S rDNA was sequenced using BGISEQ method. Case Report: A 42 Year old male presented with complaints of sudden painless diminution of vision in RE for 3days, after fall of ectoparasticide (DELTAMETHRIN1.25%) in Right Eye. Uncorrected visual acuity in Right eye: counting fingers at 1/2metre, Left Eye: 6/6. Anterior segment evaluation showed Right and Left eye within normal limits. Pupils were 3mm round, regular, reactive to light in OU. Dilated Fundus Examination using indirect ophthalmoscopy showed Right Eye C:D:0.5, A:V 2:3 with subretinal fluid accumulation in macula and Left Eye normal fundus picture. A diagnosis of RE Central serous Chorioretinopathy (CSCR) was made using Optical Coherence Tomography. Discussion: With an incidence rate of 9.9 per 100,000, or roughly six times higher than that of women, CSCR mostly affects men. Acute or subacute central vision loss or distortion is the most prevalent symptom, and it typically affects men between the ages of 20 and 50.With regard to illnesses like CSCR, this case study aims to provide clinicians with more insight into the diagnosis, treatment, and management of pesticide-induced ocular damage. Conclusion: This is rare case of retinal toxicity following exposure to Deltamethrin. For an accurate diagnosis and management of CSCR, it is imperative to comprehend its risk factors, particularly the exposure to pesticides. Emphasizing eye protection and preventative health measures will help us safeguard our vision from environmental risks such as pesticide exposure.

A Gas chromatography-electron capture detector-based analytical technique for the detection of deltamethrin residues in chicken meat samples was standardized and validated in the present study. Deltamethrin was extracted from the chicken meat matrix as per QuEChERS protocol. The method standardized was sensitive, reliable, accurate, and precise and allows the detection of deltamethrin residues in chicken meat > 0.08 µg/g. The standardized method was applied for the detection of deltamethrin residues in 33 chicken meat samples collected from Tamil Nadu, India. Two samples of chicken meat were positive for deltamethrin residues and the percent of samples positive for deltamethrin was 0.06%. However, the levels were less than MRL values of deletamethrin as indicated by FSSAI, India. Monitoring studies with a higher number of samples are required to assess the actual status of pesticide residues in animal food products. Hence, regular monitoring programs and creating awareness among livestock farmers training are also required to control pesticide residues in animal food products.

Black soldier fly larvae (Hermetia illucens) receive growing interest as a potential alternative animal feed source. These insects may be exposed to insecticide residues in the rearing substrate. This study aimed to investigate the effects of six different pyrethroid and organophosphate insecticides on this insect species' performance. The toxicity of two "model" substances for each of these classes (cypermethrin; pirimiphos‐methyl) was quantified, with and without the synergist piperonyl butoxide (PBO). Critical effect doses corresponding to 10% yield (CED10) for cypermethrin (0.4 mg/kg) and pirimiphos‐methyl (4.8 mg/kg) were determined. The addition of PBO to cypermethrin enhanced its relative potency with a factor 2.6. These data were compared against the relative toxicity of two analogue substances in each class (permethrin, deltamethrin; chlorpyrifos‐methyl, malathion). Results suggest that exposure to concentrations complying with legal limits can cause significant reductions in yield. Exposure to multiple substances at lower concentrations resulted in negative additive and synergistic effects. Of the tested substances, deltamethrin was most toxic, causing 94% yield at 0.5 mg/kg. Analytical results suggested that transfer of tested substances to the larval biomass was substance‐ and concentration‐specific, but appeared to be correlated to reduced yields and the presence of PBO. Transfer of organophosphates was overall low (<2%), but ranged from 8% to 75% for pyrethroids. Due to very low limits in insect biomass (∼0.01 mg/kg), high transfer may result in noncompliance. It is recommended that rearing companies implement lower contractual thresholds, and that policymakers consider adjusting legally allowed maximum residue levels in insect feed. We investigated the expression of 4 cytochrome P450 genes (CYP4G19, CYP6J1, CYP6K1, and CYP4C21) in 4 field-collected strains (WM, RG386, CDR, and Ryan) of the German cockroach, Blattella germanica (L.), collected from California. The UCR susceptible strain was used as a comparison. Topical assays using a diagnostic dose (3× LD95) of deltamethrin revealed decreased sensitivity in all field-collected strains with mortality ranging from 0% to 58%, and the addition of PBO before deltamethrin treatment increased mortality to 52.5%–87.5%. Using qPCR to investigate the expression levels of CYP4G19, CYP4C21, CYP6J1, and CYP6K1, we found that only CYP6K1 was significantly overexpressed (2.1–5.8× higher) in all field-collected strains when compared to the UCR strain. Next, we investigated the role of the CYP6K1 gene by performing gene knockdown using RNAi. After dsCYP6K1 treatment, the expression levels of CYP6K1 in WM and Ryan strains were significantly reduced (P  < 0.01) by 91%–94% vs. those treated with dsEGFP (control) on the third and sixth day posttreatment. RG386, CDR, and Ryan strains were more susceptible compared to their respective controls to topically applied deltamethrin 6 days after treatment with dsCYP6K1. Bactrocera dorsalis and Bactrocera carambolae are major pests of various commodities that could cause significant losses, if not controlled. Insecticide usage is a common control carried out by farmers. The test was conducted to determine the toxicity level of several types of insecticides on the mortality of two different fruit fly species. The fruit fly population used in the study came from the collection owned by Center for Forecasting Plant Pest Organisms (BBPOPT), Karawang. The research method used was an experimental method with a Randomized Block Design. The test was carried out using the dry film method. The concentration ranges used were 0.02-1 ml/L fipronil insecticide, 0.01-0.6 ml/L ethion insecticide, 0.02-0.6 ml/L deltamethrin insecticide, and 0.02-0.8 ml/L lambda-cyhalothrin insecticide. The control treatment used was acetone alone. Each treatment was replicated thrice. Mortality was observed at 1, 3, 24, and 48 hours after treatment. Cumulative mortality at 48 hours after treatment was analyzed using probit to determine the LC50 and LC95 values at 48 hours. The results showed that the insecticide deltamethrin was the most toxic insecticide to B. carambolae with an LC50 value of 0.025 ml/L and an LC95 of 0.646 ml/L. Ethion was the most toxic insecticide for B. dorsalis with LC50 values of 0.046 ml/L and LC95 of 0.746 ml/L. Deltamethrin (DM) has been widely used as a type II pyrethroid insecticide. Its high lipophilicity allows it to easily pass through the blood-brain barrier and affect the nervous system. However, its impact on the early neural development of zebrafish and the mechanism of toxicity remains unclear. In this study, zebrafish (Danio rerio) embryos were exposed to DM with different concentration of 0,10, 20 and 50 µg⋅L-1 to analyze the development and movement ability. The neurotoxicity of DM to zebrafish larvae was evaluated on the development of the nerve cells of transgenic zebrafish line. The results showed that the frequency of spontaneous coiling decreased 41.2% (P<0.001) by 50 µg⋅L-1 DM exposure at 24 hpf. The hatching rate of all the DM exposure groups was higher than the control group and heart rate were significantly increased in 20 and 50 µg⋅L-1 groups (P<0.01, P< 0.0001), and the body length was significantly reduced (P<0.01) at 36 hpf. These results showed that the effect of DM on embryo development was dose-dependent. Through the analysis of the spontaneous swim, light-dark stimulus and open field behavior of zebrafish larvae, it was found that zebrafish larvae became sluggish in those three scenarios, indicating that DM exposure affected the mobility of zebrafish, and the ability to respond to external environmental stimuli and the ability to actively explore new environments was weakened. Further analysis of transgenic zebrafish strains with specific markers for neural cell systems found that DM exposure caused a significant decrease in the number of nerve stem/progenitor cells in zebrafish (P<0.01, P<0.001), neuronal cell activity was enhanced, and the number of apoptotic cells was significantly increased (P<0.0001). The results suggested that the neurotoxicity of zebrafish larvae induced by DM is related to the nerve cell viability. DM is toxic to the central nervous system of zebrafish by destroying the number of nerve stem / progenitor cells, over-activating neurons and promoting apoptosis of nerve cells, thus affecting the behavior of zebrafish embryos and larvae. Background: Ae. aegypti is the vector of important μ arboviruses, including dengue, Zika, chikungunya and yellow fever. Despite not being specifically targeted by insecticide-based control programs in West Africa, resistance to insecticides in Ae. aegypti has been reported in countries within this region. In this study, we investigated the status and mechanisms of Ae. aegypti resistance in Niamey, the capital of Niger. This research aims to provide baseline data necessary for arbovirus outbreak prevention and preparedness in the country. Methods: Ovitraps were used to collect Ae. aegypti eggs, which were subsequently hatched in the insectary for bioassay tests. The hatched larvae were then reared to 3–5-day-old adults for WHO tube and CDC bottle bioassays, including synergist tests. The kdr mutations F1534C, V1016I, and V410L were genotyped using allele-specific PCR and TaqMan qPCR methods. Results: Ae. aegypti from Niamey exhibited moderate resistance to pyrethroids but susceptibility to organophosphates and carbamates. The kdr mutations, F1534C, V1016I and V410L were detected with the resistant tri-locus haplotype 1534C+1016L+410L associated with both permethrin and deltamethrin resistance. Whereas the homozygote tri-locus resistant genotype 1534CC+1016LL+410LL was linked only to permethrin resistance. The involvement of oxidase and esterase enzymes in resistance mechanisms was suggested by partial restoration of mosquitoes' susceptibility to pyrethroids in synergist bioassays. Conclusion: This study is the first report of Ae. aegypti resistance to pyrethroid insecticides in Niamey. The resistance is underpinned by target site mutations and potentially involves metabolic enzymes. Background: Mexico has experienced a significant reduction in malaria cases over the past two decades. Certification of localities as malaria-free areas (MFAs) has been proposed as a steppingstone before elimination is achieved throughout the country. The Mexican state of Quintana Roo is a candidate for MFA certification. Monitoring the status of insecticide susceptibility of major vectors is crucial for MFA certification. This study describes the susceptibility status of Anopheles albimanus, main malaria vector, from historically important malaria foci in Quintana Roo, using both phenotypic and genotypic approaches. Methods: Adult mosquito collections were carried out at three localities: Palmar (Municipality of Othon P. Blanco), Buenavista (Bacalar) and Puerto Morelos (Puerto Morelos). Outdoor human-landing catches were performed by pairs of trained staff from 18:00 to 22:00 during 3-night periods at each locality during the rainy season of 2022. Wild-caught female mosquitoes were exposed to diagnostic doses of deltamethrin, permethrin, malathion, pirimiphos-methyl or bendiocarb using CDC bottle bioassays. Mortality was registered at the diagnostic time and recovery was assessed 24 h after exposure. Molecular analyses targeting the Voltage-Gated Sodium Channel (vgsc) gene and acetylcholinesterase (ace-1) gene were used to screen for target site polymorphisms. An SNP analysis was carried out to identify mutations at position 995 in the vgsc gene and at position 280 in the ace-1 gene. Results: A total of 2828 anophelines were collected. The main species identified were Anopheles albimanus (82%) and Anopheles vestitipennis (16%). Mortalities in the CDC bottle bioassay ranged from 99% to 100% for all the insecticides and mosquito species. Sequence analysis was performed on 35 An. albimanus across the three localities; of those, 25 were analysed for vgsc and 10 for ace-1 mutations. All individuals showed wild type alleles. Conclusion: The results demonstrated that An. albimanus populations from historical malaria foci in Quintana Roo are susceptible to the main insecticides used by the Ministry of Health. Background: Culex pipiens pallens is a well-known mosquito vector for several diseases. Deltamethrin, a commonly used pyrethroid insecticide, has been frequently applied to manage adult Cx. pipiens pallens. However, mosquitoes can develop resistance to these insecticides as a result of insecticide misuse and, therefore, it is crucial to identify novel methods to control insecticide resistance. The relationship between commensal bacteria and vector resistance has been recently recognized. Bacteriophages (= phages) are effective tools by which to control insect commensal bacteria, but there have as yet been no studies using phages on adult mosquitoes. In this study, we isolated an Aeromonas phage vB AhM-LH that specifically targets resistance-associated symbiotic bacteria in mosquitoes. We investigated the impact of Aeromonas phage vB AhM-LH in an abundance of Aeromonas hydrophila in the gut of Cx. pipiens pallens and its effect on the status of deltamethrin resistance. Methods: Phages were isolated on double-layer agar plates and their biological properties analyzed. Phage morphology was observed by transmission electron microscopy (TEM) after negative staining. The phage was then introduced into the mosquito intestines via oral feeding. The inhibitory effect of Aeromonas phage vB AhM-LH on Aeromonas hydrophila in mosquito intestines was assessed through quantitative real-time PCR analysis. Deltamethrin resistance of mosquitoes was assessed using WHO bottle bioassays. Results: An Aeromonas phage vB AhM-LH was isolated from sewage and identified as belonging to the Myoviridae family in the order Caudovirales using TEM. Based on biological characteristics analysis and in vitro antibacterial experiments, Aeromonas phage vB AhM-LH was observed to exhibit excellent stability and effective bactericidal activity. Sequencing revealed that the Aeromonas phage vB AhM-LH genome comprises 43,663 bp (51.6% CG content) with 81 predicted open reading frames. No integrase-related gene was detected in the vB AH-LH genome, which marked it as a potential biological antibacterial. Finally, we found that Aeromonas phage vB AhM-LH could significantly reduce deltamethrin resistance in Cx. pipiens pallens, in both the laboratory and field settings, by decreasing the abundance of Aeromonas hydrophila in their midgut. Conclusions: Our findings demonstrate that Aeromonas phage vB AhM-LH could effectively modulate commensal bacteria Aeromonas hydrophila in adult mosquitoes, thus representing a promising strategy to mitigate mosquito vector resistance. Insecticide resistance in mosquitoes is spreading worldwide and represents a growing threat to vector control. Insecticide resistance is caused by different mechanisms including higher metabolic detoxication, target-site modification, reduced penetration and behavioral changes that are not easily detectable with simple diagnostic methods. Indeed, most molecular resistance diagnostic tools are costly and labor intensive and then difficult to use for routine monitoring of insecticide resistance. The present study aims to determine whether mosquito susceptibility status against the pyrethroid insecticides (mostly used for mosquito control) could be established by the protein signatures of legs and/or thoraxes submitted to MALDI-TOF Mass Spectrometry (MS). The quality of MS spectra for both body parts was controlled to avoid any bias due to unconformity protein profiling. The comparison of MS profiles from three inbreeds Ae. aegypti lines from French Guiana (IRF, IR03, IR13), with distinct deltamethrin resistance genotype / phenotype and the susceptible reference laboratory line BORA (French Polynesia), showed different protein signatures. On both body parts, the analysis of whole protein profiles revealed a singularity of BORA line compared to the three inbreeding lines from French Guiana origin, suggesting that the first criteria of differentiation is the geographical origin and/or the breeding history rather than the insecticide susceptibility profile. However, a deeper analysis of the protein profiles allowed to identify 10 and 11 discriminating peaks from leg and thorax spectra, respectively. Among them, a specific peak around 4870 Da was detected in legs and thoraxes of pyrethroid resistant lines compared to the susceptible counterparts hence suggesting that MS profiling may be promising to rapidly distinguish resistant and susceptible phenotypes. Further work is needed to confirm the nature of this peak as a deltamethrin resistant marker and to validate the routine use of MS profiling to track insecticide resistance in Ae. aegypti field populations. Pesticides are used to control agricultural and industrial pests, and even combat diseases such as malaria, posing a serious threat to public health and many aspects of life. Therefore, identification, extraction, and preconcentration play a crucial role in the determination and measurement of these compounds. The cooled-heated device headspace solid-phase microextraction technique provides preconcentration of volatile and semi-volatile compounds. The aim of this research is to design and fabricate a new device for the simultaneous cooling of the fiber and evaporated sample and extracting pesticides. The innovative technique was evaluated by the extraction and gas chromatography mass spectrometry determination of chlorpyrifos, deltamethrin, fenvalerate and cypermethrin in soil samples. Optimization of the effective extraction parameters was carried out by the simplex method. Under the optimized conditions (i.e., cold and heat temperature of –10 and 70°C, water addition of 20 µL, and extraction time of 15 min), relative standard deviations of 3.94 to 6.51% were obtained of the studied compounds in five replicated analyses. The aquatic environment encompasses a wide variety of pollutants, from plastics to drug residues, pesticides, food compounds, and other food by-products, and improper disposal of waste is the main cause of the accumulation of toxic substances in water. Monitoring, assessing, and attempting to control the effects of contaminants in the aquatic environment are necessary and essential to protect the environment and thus human and animal health, and the study of aquatic ecotoxicology has become topical. In this respect, zebrafish are used as model organisms to study the bioaccumulation, toxicity, and influence of environmental pollutants due to their structural, functional, and material advantages. There are many similarities between the metabolism and physiological structures of zebrafish and humans, and the nervous system structure, blood–brain barrier function, and social behavior of zebrafish are characteristics that make them an ideal animal model for studying neurotoxicity. The aim of the study was to highlight the neurotoxicity of nanoplastics, microplastics, fipronil, deltamethrin, and rotenone and to highlight the main behavioral, histological, and oxidative status changes produced in zebrafish exposed to them. Simple Summary: The tiger mosquito, Aedes albopictus, spreads the virus causing dengue fever, notably in Southeast Asian countries. To stop the spread of this disease, the Aedes mosquitoes are killed with chemicals known as pyrethroids. However, despite pyrethroids being highly effective insecticides, tiger mosquitoes are becoming resistant and surviving exposure to these chemicals. We have measured that tiger mosquitoes from two regions in Cambodia (the capital, Phnom Penh, and rural Pailin province) are highly resistant to pyrethroids. To determine what causes resistance, we have implicated unusually high activity of enzymes, known as P450 monooxygenases, which break down pyrethroids and thus render them harmless in the resistant mosquitoes. We have ruled out another common mechanism of pyrethroid resistance, which is changes in the mosquitoes' DNA sequence, which prevents pyrethroids from binding to a protein known as the voltage-gated sodium channel, thereby disrupting signals being sent throughout the nervous system. Overall, our findings describe an important survey monitoring the prevalence of insecticide resistance in disease-spreading mosquitoes in Cambodia, and with the increased understanding of what causes resistance, we are in a strong position to advise that mosquito control methods alternative to pyrethroids should be implemented as soon as possible to ensure continuing management of dengue fever. (1) Background: In Cambodia, Aedes albopictus is an important vector of the dengue virus. Vector control using insecticides is a major strategy implemented in managing mosquito-borne diseases. Resistance, however, threatens to undermine the use of insecticides. In this study, we present the levels of insecticide resistance of Ae. albopictus in Cambodia and the mechanisms involved. (2) Methods: Two Ae. albopictus populations were collected from the capital, Phnom Penh city, and from rural Pailin province. Adults were tested with diagnostic doses of malathion (0.8%), deltamethrin (0.03%), permethrin (0.25%), and DDT (4%) using WHO tube assays. Synergist assays using piperonyl butoxide (PBO) were implemented before the pyrethroid assays to detect the potential involvement of metabolic resistance mechanisms. Adult female mosquitoes collected from Phnom Penh and Pailin were tested for voltage-gated sodium channel (VGSC) kdr (knockdown resistance) mutations commonly found in Aedes sp.-resistant populations throughout Asia (S989P, V1016G, and F1534C), as well as for other mutations (V410L, L982W, A1007G, I1011M, T1520I, and D1763Y). (3) Results: The two populations showed resistance against all the insecticides tested (<90% mortality). The use of PBO (an inhibitor of P450s) strongly restored the efficacy of deltamethrin and permethrin against the two resistant populations. Sequences of regions of the vgsc gene showed a lack of kdr mutations known to be associated with pyrethroid resistance. However, four novel non-synonymous mutations (L412P/S, C983S, Q1554STOP, and R1718L) and twenty-nine synonymous mutations were detected. It remains to be determined whether these mutations contribute to pyrethroid resistance. (4) Conclusions: Pyrethroid resistance is occurring in two Ae. albopictus populations originating from urban and rural areas of Cambodia. The resistance is likely due to metabolic resistance specifically involving P450s monooxygenases. The levels of resistance against different insecticide classes are a cause for concern in Cambodia. Overdoses of pesticides lead to a decrease in the yield and quality of plants, such as beans. The unconscious use of deltamethrin, one of the synthetic insecticides, increases the amount of reactive oxygen species (ROS) by causing oxidative stress in plants. In this case, plants tolerate stress by activating the antioxidant defense mechanism and many genes. 5-Aminolevulinic acid (ALA) improves tolerance to stress by acting exogenously in low doses. There are many gene families that are effective in the regulation of this mechanism. In addition, one of the response mechanisms at the molecular level against environmental stressors in plants is retrotransposon movement. In this study, the expression levels of superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT), glutathione reductase (GR), and stress-associated protein (SAP) genes were determined by Q-PCR in deltamethrin (0.5 ppm) and various doses (20, 40, and 80 mg/l) of ALA-treated bean seedlings. In addition, one of the response mechanisms at the molecular level against environmental stressors in plants is retrotransposon movement. It was determined that deltamethrin increased the expression of SOD (1.8-fold), GPX (1.4-fold), CAT (2.7-fold), and SAP (2.5-fold) genes, while 20 and 40 mg/l ALA gradually increased the expression of these genes at levels close to control, but 80 mg/l ALA increased the expression of these genes almost to the same level as deltamethrin (2.1-fold, 1.4-fold, 2.6-fold, and 2.6-fold in SOD, GPX, CAT, and SAP genes, respectively). In addition, retrotransposon-microsatellite amplified polymorphism (REMAP) was performed to determine the polymorphism caused by retrotransposon movements. While deltamethrin treatment has caused a decrease in genomic template stability (GTS) (27%), ALA treatments have prevented this decline. At doses of 20, 40, and 80 mg/L of ALA treatments, the GTS ratios were determined to be 96.8%, 74.6%, and 58.7%, respectively. To study the acaricide resistance status and possible mechanisms of action in conferring resistance to commonly used acaricides (deltamethrin and coumaphos), Hyalomma anatolicum ticks were collected from 6 dairy farms of Hisar and Charkhi Dadri districts of Haryana. By using standard larval packet test, H. anatolicum tick larvae of Charkhi Dadri isolates were found to be susceptible (100% mortality) to both the acaricides. Level-I resistance against coumaphos was recorded from four isolates, whereas, level-II was observed in only one isolate, collected from Hisar. One isolates (Kaimri) from Hisar also showed level-I resistance against deltamethrin. Biochemically, the ticks having higher values of resistance factor (RF) against coumaphos were found to possess increased enzymatic activity of α-esterase, β-esterase, glutathione-S-transferase (GST) and mono-oxygenase enzymes, whereas, the monoamine oxidase did not show any constant trend. However, the RF showed a statistical significant correlation with GST only. Native PAGE analysis of H. anatolicum ticks revealed the presence of nine types of esterases (EST-1 h to EST-9 h) by using napthyl acetate as substrate. In the inhibitory assay, esterases were found to be inhibited by PMSF, indicating the presence of serine residue at catalytic triad. The partial cds of carboxylesterase and domain II of sodium channel genes were sequenced to determine any proposed mutations in resistant isolates of H. anatolicum ticks, however, no mutations were observed in either gene, indicating that increased expression of detoxification enzymes as a possible mechanism for resistance development, in the current study. Background: This study investigates the effectiveness of new-generation mosquito nets, like Olyset® Plus and PermaNet® 3.0, and dual-action nets such as Interceptor® G2, against pyrethroid-resistant Anopheles gambiae mosquitoes following the 2023 mass distribution of long-lasting insecticidal nets in Benin. Methods: We tested wild mosquito populations from six communes in Benin against various pyrethroid (permethrin 0.75%, alphacypermethrin 0.05%, and deltamethrin 0.05%) using WHO tube tests. Additionally, we exposed mosquitoes to chlorfenapyr 100 µg/ml using the CDC bottle bioassay method. A subset of mosquitoes underwent biochemical and PCR tests to check the overexpression of metabolic enzymes and the Kdr L1014F mutation. We evaluated the effectiveness of Olyset® Plus, PermaNet® 3.0, and Interceptor® G2 nets using cone and tunnel tests on both laboratory and field populations of An. gambiae. Results: Overall, the highest mortality rate was 60% with pyrethroid and 98 to100% with chlorfenapyr. In cone tests, all three types of nets induced mortality rates above 80% in the susceptible laboratory strain of An. gambiae. Notably, Olyset® Plus showed the highest mortality rates for pyrethroid-resistant mosquitoes in cone tests, ranging from 81.03% (95% CI: 68.59–90.13) in Djougou to 96.08% (95% CI: 86.54–99.52) in Akpro-Missérété. PermaNet® 3.0 had variable rates, from 42.5% (95% CI: 27.04–59.11) in Djougou to 58.54% (95% CI: 42.11–73.68) in Porto-Novo. However, revealed good results for Interceptor® G2, with 94% (95% CI: 87.40–97.77) mortality and 89.09% blood sampling inhibition in local populations of An. gambiae. In comparison, Interceptor® had lower rates of 17% (95% CI: 10.23–25.82) and 60%, respectively. Conclusion: These results suggest that tunnel tests are effective for evaluating dual-active ingredient nets. Background: The residual activity of a clothianidin + deltamethrin mixture and clothianidin alone in IRS covered more than the period of malaria transmission in northern Benin. The aim of this study was to show whether the prolonged residual efficacy of clothianidin-based products resulted in a greater reduction in vector populations and subsequent malaria transmission compared with the shorter residual efficacy of pirimiphos-methyl. Methods: Human bait mosquito collections by local volunteers and pyrethrum spray collections were used in 6 communes under IRS monitoring and evaluation from 2019 to 2021. ELISA/CSP and species PCR tests were performed on Anopheles gambiae sensu lato (s.l.) to determine the infectivity rate and subspecies by commune and year. The decrease in biting rate, entomological inoculation rate, incidence, inhibition of blood feeding, resting density of An. gambiae s.l. were studied and compared between insecticides per commune. Results: The An. gambiae complex was the major vector throughout the study area, acounting for 98.71% (19,660/19,917) of all Anopheles mosquitoes collected. Anopheles gambiae s.l. collected was lower inside treated houses (45.19%: 4,630/10,245) than outside (54.73%: 5,607/10,245) after IRS (p < 0.001). A significant decrease (p < 0.001) in the biting rate was observed after IRS in all departments except Donga in 2021 after IRS with clothianidin 50 WG. The impact of insecticides on EIR reduction was most noticeable with pirimiphos-methyl 300 CS, followed by the clothianidin + deltamethrin mixture and finally clothianidin 50 WG. A reduction in new cases of malaria was observed in 2020, the year of mass distribution of LLINs and IRS, as well as individual and collective protection measures linked to COVID-19. Anopheles gambiae s.l. blood-feeding rates and parous were high and similar for all insecticides in treated houses. Conclusion: To achieve the goal of zero malaria, the optimal choice of vector control tools plays an important role. Pesticide formulations are typically applied as mixtures, and their synergistic effects can increase toxicity to the organisms in the environment. Despite pesticide mixtures being the leading cause of pesticide exposure incidents, little attention has been given to assessing their combined toxicity and interactions. This survey purposed to reveal the cumulative toxic effects of deltamethrin (DEL) and cyazofamid (CYA) on earthworms (Eisenia fetida) by examining multiple endpoints. Our findings revealed that the LC50 values of DEL for E. fetida, following 7- and 14-day exposures, ranged from 887.7 (728–1095) to 1552 (1226–2298) mg kg−1, while those of CYA ranged from 316.8 (246.2–489.4) to 483.2 (326.1–1202) mg kg−1. The combinations of DEL and CYA induced synergistic influences on the organisms. The contents of Cu/Zn-SOD and CarE showed significant variations when exposed to DEL, CYA, and their combinations compared to the untreated group. Furthermore, the mixture administration resulted in more pronounced alterations in the expression of five genes (hsp70, tctp, gst, mt, and crt) associated with cellular stress, carcinogenesis, detoxification, and endoplasmic reticulum compared to single exposures. In conclusion, our comprehensive findings provided detailed insights into the cumulative toxic effects of chemical mixtures across miscellaneous endpoints and concentration ranges. Background: Clothianidin-based indoor residual spraying (IRS) formulations have become available for malaria control as either solo formulations of clothianidin or a mixture of clothianidin with the pyrethroid deltamethrin. While both formulations have been successfully used for malaria control, studies investigating the effect of the pyrethroid in IRS mixtures may help improve our understanding for development of future IRS products. It has been speculated that the irritant effect of the pyrethroid in the mixture formulation may result in shorter mosquito contact times with the treated walls potentially leading to a lower impact. Methods: We compared contact irritancy expressed as the number of mosquito take-offs from cement surfaces treated with an IRS formulation containing clothianidin alone (SumiShield® 50WG) to clothianidin-deltamethrin mixture IRS formulations against pyrethroid-resistant Anopheles gambiae sensu lato under controlled laboratory conditions using a modified version of the World Health Organisation cone bioassay. To control for the pyrethroid, comparison was made with a deltamethrin-only formulation. Both commercial and generic non-commercial mixture formulations of clothianidin and deltamethrin were tested. Results: The clothianidin solo formulation did not show significant contact irritancy relative to the untreated control (3.5 take-offs vs. 3.1 take-offs, p = 0.614) while all deltamethrin-containing IRS induced significant irritant effects. The number of take-offs compared to the clothianidin solo formulation (3.5) was significantly higher with the commercial clothianidin-deltamethrin mixture (6.1, p = 0.001), generic clothianidin-deltamethrin mixture (7.0, p < 0.001), and deltamethrin-only (8.2, p < 0.001) formulations. The commercial clothianidin-deltamethrin mixture induced similar contact irritancy as the generic clothianidin-deltamethrin mixture (6.1 take-offs vs. 7.0 take-offs, p = 0.263) and deltamethrin-only IRS (6.1 take-offs vs. 8.2, p = 0.071), showing that the irritant effect in the mixture was attributable to its deltamethrin component. Conclusions: This study provides evidence that the enhanced contact irritancy of the pyrethroid in clothianidin-deltamethrin IRS mixtures can shorten mosquito contact times with treated walls compared to the clothianidin solo formulation. Further trials are needed to directly compare the efficacy of these formulation types under field conditions and establish the impact of this enhanced contact irritancy on the performance of IRS mixture formulations containing pyrethroids. Background and Objectives Wheat is a critical crop in Iran and pivotal in food security, economic stability, and political landscapes. Wheat fields are the habitat of many insects, including both beneficial and pest species. Therefore, various insecticides are employed in wheat fields to safeguard crop yields. However, pursuing pest control often entails unintended consequences for non-target organisms, particularly pollinators and natural enemies. This study aimed to evaluate the effect of both chemical and herbal insecticides on non-target insect populations within Iranian wheat fields. Although plant-derived insecticides may offer ecological benefits, the study emphasizes the need to evaluate their efficacy against pests and their impact on beneficial insects. Therefore, it is necessary to consider the possible effects of herbal and chemical insecticides on natural enemies. Materials and Methods This study evaluated the adverse effects of some recommended insecticides for wheat pests on some natural enemies. A completely randomized block design with five treatments and four replicates was employed. The experiment was conducted in Ardabil, Tehran, and Golestan, Iran. The size of each plot was 100m² (10 x 10), with a distance of 2m between blocks and plots. A total of 30 wheat stalks were randomly selected for sampling in each plot, and all larvae on these stalks were counted within the wheat field. Sampling was performed on four occasions: one day before and 3, 7, and 14 days after spraying. The treatments included matrine (at concentrations of 1.5 and 2ml/L), deltamethrin (0.3ml/L), fenitrothion (1ml/L), and a control treatment (water). The potential effects of the insecticides were examined on the larvae of natural enemies, including the seven-spotted ladybird, flower fly, and green lacewing. Tehran and Ardabil provinces had low flower fly and green lacewing populations across all treatments, including control. However, the ladybird population was sufficient for evaluation. Therefore, the effects of the insecticides on ladybirds were investigated in these provinces. Results The reduction in the population of seven-spotted ladybirds in Ardabil caused by the insecticides was insignificant on days 3 and 7 and significant on day 14. Deltamethrin caused the most negative effect on days 3 and 7, while matrine 2ml/L had the most negative effect on day 14. Deltamethrin consistently caused the largest ladybird population decline in Tehran, with a significant difference from other treatments on all days except day 3. Fenitrothion exhibited the least adverse effect throughout the experiment. Golestan had the healthiest natural enemy populations. In this province, a suitable population of seven-spotted ladybirds, flower flies, and green lacewings was observed. The study confirmed insecticide-induced declines in these populations, with deltamethrin causing the most significant reductions for ladybirds and flower flies, while fenitrothion had the least impact. All insecticides severely reduced green lacewing populations. Based on the results of the 7th day, the reduction of the population of seven-spotted ladybirds in different provinces caused by matrine 1.5 and 2ml/L, fenitrothion, and deltamethrin was 31.04% - 57.91%, 54.19% - 77.58%, 25.01% - 70.16% and 72.02% - 89.35%, respectively. In addition, population reduction caused by matrine 1.5 and 2 ml/L, fenitrothion, and deltamethrin was 31.61%, 67.01%, 24.58%, and 79.06% for flower fly, and 73.24%, 82.79%, 73.24% and 95.94% for green lacewing, respectively. In all cases, the reduction caused by matrine 2ml/L was higher than matrine 1.5ml/L. On day 7, most insecticides were slightly harmful to seven-spotted ladybirds in all provinces except Tehran and Golestan, where fenitrothion was harmless and deltamartin moderately harmful. On this day, matrine, fenitrothion, and deltamethrin were slightly harmful, harmless, and moderately harmful to flower flies, respectively. Moreover, matrine 1.5ml/L and fenitrothion were slightly harmful to green lacewing, while matrine 2ml/L and deltamethrin were moderately harmful. Discussion Deltamethrin consistently exhibited the most pronounced adverse effects across all treatments and natural enemy populations, while fenitrothion had the least severe impact. Green lacewing populations displayed the highest insecticide sensitivity among the studied natural enemies. Based on the results, the effect of insecticides on non-target beneficial organisms was essential when developing integrated pest management (IPM) strategies. Avoiding the application of insecticides, particularly deltamethrin, during peak natural enemy populations was crucial to minimize unintended ecological consequences and promote a balanced ecosystem within wheat fields. The sweet potato/cotton whitefly Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) is one of the most destructive vegetable pests in greenhouses in Türkiye. While the preferred method of control primarily relies on insecticides such as neonicotinoids, organophosphates and pyrethroids, their intensive use has caused whiteflies to rapidly become resistant. Samples were collected from 13 Turkish B. tabaci greenhouse populations. Bioassays from the Marmara Region identified as high as a 53-fold increase in resistance to chlorpyrifos-ethyl, a 303-fold increase to imidacloprid and a 282-fold increase to acetamiprid. Bioassays in the Central Anatolia Region reported a 76-fold increase in resistance to deltamethrin and—15-fold increase to thiamethoxam. Most of the populations showed cross-resistance for neonicotinoids. All collected B. tabaci populations were determined to be MEAM 1 (also referred to as biotype B) for diagnostic band E0.14. The E0.14 esterase variant that helps to diagnose biotype B insects was found in all individuals from field populations. Almost all populations showed higher non-specific esterase, glutathione-S-transferase (GST) and monooxygenase activity when compared to susceptible SUD-S population. AChE insensitivity assays for individual B. tabaci suggest a target-site modification as a mechanism of resistance to chlorpyrifos-ethyl. This is the first report of AChE variants identified based on their sensitivities to chlorpyrifos ethyl-oxon and pirimicarb in Turkish B. tabaci populations. Fixed ace mutations in the target-site of organophosphates and pyrethroids were identified in six populations. Resistance to organophosphates and neonicotinoids were at least partially related to both ace mutations and insensitive AChE and monooxygenase activities, respectively.
